This diploma thesis was focused on the preparation of magnetic and fluorescently labelled spinel type nanoparticles, specifically nanoparticles of zinc-doped cobalt ferrite intended for application in magnetic resonance imaging and fluorescence microscopy in experimental practice. In previous studies, various ferrite nanoparticles exhibited relatively high transverse relaxation times and strong negative T2 contrast. These properties were also supposed for the zinc-doped cobalt ferrite nanoparticles. This assumption was confirmed by studying their magnetic and contrasting properties. Fluorescence properties of the prepared nanoparticles were also successfully studied. With respect to the intended applications of these particles, the issue of suitable surface modification of magnetic cores, their colloidal stabilization in an aqueous suspension and toxicity in biological systems were studied. RNDr. Jan Kotek, Ph.D. Supervisor's email: modrej@natur.cuni.cz Abstract: The aim of this thesis was to synthesize macrocyclic ligands (based on the DO3A and DO2A framework) with exchangeable protons on the aminic coordinating groups that could be (after the complexation with suitable paramagnetic metal ions) potential contrast agents based on saturation transfer, so called CEST contrast agents (Chemical Exchange Saturation Transfer). Two ligands H3L1 and H2L2 with one and two aminoethyl groups have been synthesized. The structurally similar ligands H3L1 Me and H3L1 2Me with one exchangeable proton and without it have been also prepared. The protonation constants of the ligands H3L1 and H2L2 have been determined by pH 1 H NMR and potentiometric titrations. The stability constants of metal ion complexes with H3L1 and H2L2 have been also determined by potentiometry. The CEST effect of Eu3+ and Yb3+ complexes with H3L1 , H3L1 Me and H3L1 2Me in solution have been studied. The dissertation deals with the psychological training of swimmers from the Jihlava junior swimming club, who regularly take part in top swimming events. The theoretical part describes the mental processes in sport, personality traits, sport training, optimizing of psychophysiological conditions of performance, psychological training of swimmers. The research part searched the initial state of psychological readiness of the group and designed the intervention program, which was subsequently processed. Research participants neither were engaged in psychological training, nor had any information about it. Some of them suffer from pre-start nervousness and they reveal a high level of activation together with physiological manifestations that can affect the results of the race. The intervention program was designed so as to compensate these effects and to optimally tune, concentrate and motivate the swimmer. The knowledge of psychological training was improved and its importance was increased.

Title: The effect of yogic relaxation on muscle tone Objectives: The aim of this thesis is to present a technique of yogic relaxation, yoga nidra, as one of the possible methods for releasing mental and physical tension. The aim of the practical part of this thesis is to evaluate the effect of yoga nidra on muscle tone. Methods: The practical part of this thesis is a pilot study with six participants. For objective assessment of muscle tone the method of myotonometry was used. M. soleus was selected as the muscle to be measured. First, the original muscle tone of m. soleus on the dominant lower limb of the participants was measured. Then the group of participants was instructed about the practice of yoga nidra and was asked to practice it every day in the following 14 days at home using a given audio recording. After the two-week training, the muscle tone of m. soleus was measured again in order to determine the long-term effect of yoga nidra on muscle tone. Finally, the participants participated in a group lesson of yoga nidra guided by a yoga teacher. Then, the muscle tone of m. soleus was measured again in order to determine the instant effect of yoga nidra on muscle tone. Results: After the two-week training of yoga nidra the research found a decrease in muscle tone of m. soleus in 5 out of 6...
