Studies On The Functional Roles Of Peptidase N, A M1 Family Member, During Stress And Infection

The cytosolic protein degradation pathway, performed by ATP-dependent proteases and ATP-independent peptidases, plays important roles in several cellular activities, e.g. cell division, cell cycle progression, intracellular signaling, MHC class I antigen presentation, host-pathogen interactions, etc. The roles of ATP-dependent proteases during stress and infection have been studied in great detail but the functional roles of ATP-independent peptidases are not clearly understood. In this study, the functional roles of E. coli or S. typhimurium encoded Peptidase N (PepN), an ATP-independent enzyme belonging to theM1 family of metallopeptidases, were investigated. The thesis will address four different aspects. (i) In the first part, the utility of using E coli ∆pepN to identify and characterize novel peptidases will be shown. It is known that deletion of pepN leads to inability to cleave the majority of in vitro peptidase substrates in E. coli and S. typhimurium. To study the differences between two closely related paralogs of the M17 family, E. coli encoded pepA and pepB were cloned in pBAD24 vector and introduced in E. coli ∆pepN. Peptidase A (PepA) and Peptidase B (PepB) expression increases the cleavage of several aminopeptidase substrates and partially rescues growth of ∆pepN during nutritional downshift and high temperature stress (NDHT), a dual stress involving growth in minimal media at 42°C. Purified PepA and PepB enzymes display broad substrate specificity; however, distinct differences are observed between these two paralogs: PepA is more stable at high temperature whereas PepB displays broader substrate specificity as it cleaves Asp and Insulin B chain peptide. The strategy utilized in this study, i.e. overexpression of peptidases in ∆pepN followed by screening for substrate specificities in total cell extracts, may be used to rapidly identify the substrate preferences of novel peptidases encoded in genomes of different organisms. (ii) The second aspect investigates the functional roles of PepN during stress and infection in S. typhimurium. PepN has two conserved signature motifs of the M1 family, GAMEN and HEXXH, which play roles in substrate recognition and catalysis. To address the roles of catalytic activity of PepN, the residue E-298, which is present in the HEXXH motif and acts as a general base during catalysis, was mutated to A-298 by site-specific mutagenesis and introduced into ∆pepN (pBR322/pepNE298A). Biochemical and biophysical analysis of purified PepN (WT and E298A) revealed loss of catalytic activity of E298A but no major structural changes were observed in comparison to the WT protein. The functional roles of this mutation using ∆pepN expressing pBR322/pepN or pBR322/pepNE298A were investigated using two conditions: (i) Nutritional downshift high temperature (NDHT)stress and (ii) systemic infection in mice. Monitoring growth profiles of different strains demonstrated the requirement of the enzymatic activity of PepN for adaptation and growth to NDHT stress. Earlier studies have shown that S. typhimurium ∆pepN hyper proliferates in peripheral organs during systemic infection in mice. However, expression of wild type (WT)or E298A PepN led to lower colony forming units (CFU), demonstrating that the decrease in CFU is independent of catalytic activity. These observations are consistent with lower serum amounts of inflammatory cytokines, lower tissue damage and increase in survival of mice infected with S. typhimurium expressing WT or E298A PepN. (iii) Although pathogen encoded peptidases are known to be important during infection, their roles in modulating host responses in immunocompromised individuals are not well studied. In the third part of this thesis, the roles of S. typhimurium encoded PepN were studied in mice lacking Interferon-γ (Ifnγ), a cytokine important for immunity. S. typhimurium lacking pepN displays enhanced CFU compared to WT in peripheral organs during systemic infection in C57BL/6 mice. However, Ifnγ-/-mice show higher CFU compared to C57BL/6 mice, resulting in lower fold differences between WT and ∆pepN. Concomitantly, reintroduction of pepN in ∆pepN reduces CFU, demonstrating pepN dependence. In addition, three distinct differences were observed between infection ofC57BL/6 and Ifnγ-/-mice upon infection with different S. typhimurium strains: (i) cytokine profiles, (ii) histological analysis and (iii) mice survival. Overall, the roles of the host encoded Ifnγ during infection with S. typhimurium strains with varying degrees of virulence will be highlighted. (iv) The final aspect of this study reveals differences in gene expression between S. typhimurium grown in rich medium (Luria-Bertani) versus NDHT stress. This adaptation affects several pathways and the gene expression of secretory proteins that are important for virulence in S. typhimurium are greatly reduced during NDHT stress. Also, analysis of secretory protein amounts in different media conditions shows reduction during growth in minimal media plus high temperature stress. The functional consequences of this reduction in secretory protein amounts lead to lower bacterial replication after infection of RAW cells or mice infected via the oral route. In addition, the differences in gene expression between WT and ∆pepN during these conditions were studied. Interestingly, there is reduction in expression of flagellar genes whereas the genes involved in nitrogen metabolism are upregulated in ∆pepN upon exposure to NDHT stress. Further studies were performed by quantifying the motility of different S. typhimurium strains grown in a variety of culture conditions. Overall, this part of the study attempts to compare and contrast the possible adaptive responses of WT and ∆pepN to NDHT stress. Together, this thesis addresses multiple aspects of the biochemistry and roles of the enigmatic PepN during stress and infection.

Atividade mutagênica em solos sob a influência de rejeitos de carvão

O solo é um compartimento ambiental altamente complexo e, em razão do crescimento populacional acelerado e da intensa atividade industrial e agrícola, tem sofrido com a contaminação de substâncias de origens diversas. Entre as principais atividades que contribuem para a perturbação da qualidade dos solos estão as atividades de geração de energia pela queima de combustíveis fósseis, como o carvão mineral. Dentre as substâncias presentes nos rejeitos do carvão queimado em usinas termelétricas estão compostos orgânicos e metais pesados que interagem com o material genético, produzindo mutações e acarretando prejuízos em nível de organismo (como as neoplasias), mas também em níveis maiores de organização biológica (como a perda de diversidade genética em populações). Estudos de avaliação do potencial mutagênico em amostras de solo são escassos, principalmente investigando a ação de mutágenos de origem inorgânica. Além disso, estudos de mutagênese em solos sob a influência de rejeitos de carvão têm sido pouco abordados na literatura. Desta forma, os objetivos deste trabalho foram: (i) testar um protocolo para avaliação de atividade mutagênica em extratos inorgânicos e orgânicos de solo; (ii) avaliar a presença e o perfil dos compostos mutagênicos em solos sob a influência de cinzas de carvão e (iii) investigar as rotas ambientais para dispersão dos compostos mutagênicos na área de estudo. Para atingir estes objetivos, processos de extração de compostos orgânicos e inorgânicos de solo foram testados quanto a sua eficiência em extrair da amostra compostos potencialmente mutagênicos frente a linhagem de Salmonella typhimurium que detecta erro no quadro de leitura (TA98). Os protocolos mais eficientes foram utilizados para os testes com diferentes amostras de solo sob a influência de rejeitos de carvão utilizando diversas linhagens, na ausência e presença de ativação metabólica (fração S9 mix). Os resultados de mutagênese associados aos resultados da caracterização química de compostos orgânicos e metais indicaram que os contaminantes presentes nas cinzas de carvão podem se dispersar com facilidade para áreas adjacentes. Essa abordagem de estudo permitiu relacionar a presença de determinadas classes de compostos com diferentes danos no DNA e inferir distintas rotas ambientais de dispersão para esses compostos. A realização deste trabalho ressalta a importância de estudos em matrizes ambientais complexas, em especial solos contaminados, que buscam integrar os dados de mutagênese e utilizá-los sob uma perspectiva ecológica. / Soil is a highly complex environmental compartment that has suffered contamination by substances from multiple sources mainly due to fast population growth and intense industrial and agricultural activity. Among the main activities that affect soil quality are power generation activities that use fossil fuels, such as mineral coal. Among the several compounds present in coal ashes that are employed in coal-fired power plants are substances that interact with the genetic material, causing mutations and/or damage at the individual level (like neoplasias) but also at higher levels of biological organization (like loss of genetic diversity in populations). There are few studies on the mutagenic potential of soil samples, especially those that investigate the action of mutagens from inorganic sources. Moreover, studies of mutagenesis in soils under the influence of coal-fired power plants have rarely been reported in literature. Thus, this work aimed at: (i) testing a protocol for the evaluation of mutagenic activity in inorganic and organic extracts from soil samples; (ii) evaluating the presence and the profile of mutagenic compounds in soils under the influence of coal ashes and (iii) investigating environmental routes of dispersion of mutagenic compounds in the study area. In order to achieve these aims, extraction processes for inorganic and organic compounds were tested concerning their efficiency to extract compounds potentially mutagenic to the specific Salmonella typhimurium strain that detects frameshift mutagens (TA98). The most efficient protocols were further utilized for the tests with different soil samples under the influence of coal-fired power plant using several strains, in absence and presence of metabolic activation (S9 mix fraction). Mutagenesis results associated with the chemical characterization of the organic compounds and metals indicated that contaminants present in coal ashes can be easily dispersed to adjacent areas. However, the implementation of industrial processes to obtain these products depends on the development of strategies for this microorganism high-cell density cultures (HCDC). One of the HCDC’s greatest challenges is overcoming Salmonella’s metabolic limitations, as it presents a high organic acids production (mainly acetic acid) that inhibits biomass formation. In this context, this work proposes two approaches to deal with this problem and implement Salmonella’s HCDC: studying the cultivation of S. typhimurium using glycerol as carbon source to reduce the generation of organic acids; studying the cultivation of a recombinant strain of S. typhimurium expressing the enzyme acetyl-CoA synthetase (ACS) from E. coli, for improved acetate assimilation). Initially, cultures were grown in agitated flasks in minimal media for two carbon sources (glucose or glycerol) for the wild-type and the recombinant strain. After the preliminary experiments, the recombinant strain was cultivated in bioreactor operated in batch mode with minimal medium formulated with glycerol, glucose or acetic acid as carbon source, to evaluate acetate production and assimilation. The wild-type strain was cultivated in a continuous-mode bioreactor on minimal medium with glycerol at D=0.10; 0.17 and 0.22 h-1 to evaluate the S. typhimurium glycerol metabolism. During the cultivations, samples were collected and analyzed by high-performance liquid chromatography to quantify the production of organic acids and substrate consumption. To quantify the concentration of biomass, optical density (600 nm) readings of culture broth and dry cell weight measurements were performed. Agitated flasks and batch cultivations results indicated that the acetate production is reduced in medium with glycerol for both strains, and that the genetically modified cells present a lower acetate accumulation phenotype compared to the wild-type. Continuous cultures of the wild-type strain showed no acetate accumulation for a 0.1 h-1 dilution rate. At rates of 0.17 h-1 and 0.22 h-1 acetate accumulation was observed, but acetate flux was 2-fold lower than the flux reached in chemostat with glucose-formulated medium. Simulations were performed with the STM_v1.0 model using as input data the glycerol and oxygen fluxes estimated from the experimental results. Good predictions were obtained for the biomass, CO2 and acetate fluxes at the higher dilution rates. The results suggest that fed-batch culture using glycerol as carbon source along with an exponential feed to maintain a 0.1 h-1 specific growth rate as a promising strategy to obtain high cellular concentrations of wild-type S. typhimurium. The efficient acetate uptake observed for the recombinant S. typhimurium cells may allow higher values of specific growth rate to be used for this strain, resulting in a higher productivity of biomass. / Nos últimos anos, a aplicação de linhagens atenuadas de Salmonella spp. vem sendo amplamente investigada para o desenvolvimento de diversos produtos biotecnológicos, principalmente vacinas. No entanto, a implementação de processos industriais para a obtenção destes produtos depende do desenvolvimento de estratégias para o cultivo em alta densidade celular (CADC) deste microrganismo. Para isso, um dos grandes desafios a ser superado se refere às limitações metabólicas da Salmonella, uma vez que esta apresenta elevada produção de ácidos orgânicos (principalmente ácido acético) que inibem a formação de biomassa. Neste contexto, este trabalho propõe duas abordagens para lidar com este problema e implementar CADC de Salmonella: estudar o crescimento de Salmonella typhimurium em glicerol avaliando seu metabolismo como uma fonte de carbono menos propensa à geração de ácidos orgânicos; e estudar a produção destes ácidos por uma cepa de S. typhimurium geneticamente modificada para diminuir o acúmulo de acetato (super-expressão do gene acs, de Escherichia coli, que codifica a enzima acetil-CoA sintetase (ACS) responsável pela assimilação de acetato). Inicialmente foram realizados cultivos em frascos agitados em meio mínimo para duas fontes de carbono (glicose ou glicerol), para a cepa selvagem e para a cepa recombinante. Após os experimentos preliminares, foram realizados cultivos em biorreator operado em modo batelada com a cepa modificada em meio mínimo formulado com glicerol, glicose ou ácido acético, como fonte de carbono, a fim de avaliar a produção e a assimilação de acetato. Foram realizados cultivos em biorreator em modo contínuo com a cepa selvagem em meio mínimo com glicerol com D=0,10; 0,17 e 0,22 h-1 para avaliar o metabolismo do glicerol pela S. typhimurium. Ao longo dos cultivos foram coletadas amostras do caldo e analisadas por cromatografia líquida de alta eficiência para quantificar a produção de ácidos orgânicos e o consumo de substrato. Para quantificar a biomassa produzida, foram realizadas medidas de densidade ótica (600 nm) e de massa seca. Os resultados dos cultivos em frascos agitados e das bateladas indicam que, para ambas as cepas, a produção de acetato é reduzida em meio formulado com glicerol, e que as células modificadas geneticamente apresentam um fenótipo de menor acúmulo de acetato comparadas à linhagem selvagem. Os cultivos contínuos realizados com a cepa selvagem mostraram que não houve acúmulo de acetato para a taxa de diluição de 0,1 h-1. Já nas taxas de 0,17 h-1 e 0,22 h-1, apesar de haver acúmulo, o fluxo de produção de acetato foi cerca de 2 vezes menor que o observado em quimiostato em meio formulado com glicose. Foram realizadas simulações com o modelo STM_v1.0, tendo como dados de entrada os fluxos de glicerol e de oxigênio estimados a partir dos dados experimentais. O modelo descreveu bem os fluxos de biomassa, CO2 e acetato para as taxas de diluição mais altas. Os resultados obtidos sugerem o cultivo em batelada alimentada com glicerol como fonte de carbono, e alimentação exponencial definida para manter a velocidade específica de crescimento em 0,1 h-1, como uma estratégia promissora para obter altas concentrações celulares de S. typhimurium selvagem. Para a S. typhimurium recombinante, devido à sua eficiente assimilação de acetato, valores ainda maiores de velocidade específica de crescimento poderiam ser impostos, com elevado aumento de produtividade em biomassa.

Thus, this work aimed at: (i) testing a protocol for the evaluation of mutagenic activity in inorganic and organic extracts from soil samples; (ii) evaluating the presence and the profile of mutagenic compounds in soils under the influence of coal ashes and (iii) investigating environmental routes of dispersion of mutagenic compounds in the study area. In order to achieve these aims, extraction processes for inorganic and organic compounds were tested concerning their efficiency to extract compounds potentially mutagenic to the specific Salmonella typhimurium strain that detects frameshift mutagens (TA98). The most efficient protocols were further utilized for the tests with different soil samples under the influence of coal-fired power plant using several strains, in absence and presence of metabolic activation (S9 mix fraction). Mutagenesis results associated with the chemical characterization of the organic compounds and metals indicated that contaminants present in coal ashes can be easily dispersed to adjacent areas. This approach allowed relating the presence of certain classes of compounds to specific damages in DNA and inferring distinct dispersion routes for these compounds. This work highlights the importance of studies concerning complex environmental matrices, specially contaminated soils, which seek the integration of mutagenesis data and their use from an ecological perspective.

O presente trabalho objetivou identificar os fatores de risco para a presença de S. Enteritidis no sistema de produção de frangos de corte, avaliar, qualificar e quantificar as variáveis encontradas e elaborar um modelo teórico de estimativa de risco deste sorovar em frangos criados em sistema de integração. Os dados foram obtidos de trabalhos recentes realizados por alguns autores e deram subsídios à realização de uma análise de riscos microbiológicos. Para caracterização molecular foram utilizadas 42 cepas de Salmonella isoladas de frangos e rações inoculados experimentalmente com uma cepa de S. Typhimurium. A inoculação da bactéria foi realizada na ração e a mesma tratada com diferentes concentrações dos ácidos propílico, fórmico e acético sendo, então, fornecida para consumo ad libitum até os 21 dias de idade, quando as aves foram sacrificadas. Foram obtidos diferentes perfis genéticos com o uso do ERIC e BOX-PCR, que se mostraram eficientes para discriminação das cepas em estudo. / The aim of this work was identify the risk factors for S. Enteritidis in the production system of broiler chickens, to evaluate, qualify and quantify the variables studied and to make a theoretical model of risk assessment of this serovar in broilers in integration system. Therefore, the data was obtain from works of some authors and supported the proposed model of microbiological risk analysis. For molecular characterization were included 42 Salmonella spp. strains isolated from chicks and feed experimentally inoculated with S. Tiphimurium. After inoculation of feed with the specific dose of strain, it was submitted to treatment with propilic, formic and acetic acids in several concentrations and it was given to birds ad libitum until 21 days old, when they were sacrificed. It was obtained different patterns through the ERIC and BOX-PCR techniques, which showed good discrimination power for the strains analyzed.

Phytochemical analysis and bioactivity of Garcinia Kola (Heckel) seeds on selected bacterial pathogens

Garcinia kola is one of the plants used in folklore remedies for the treatment of microbial infections. Bacterial resistance to commonly used antibiotics has necessitated the search for newer and alternative compounds for the treatment of drug resistant microbial infections. This study focuses on the bioactivity of G. kola seeds on Streptococcus pyogenes (ATCC 49399), Staphylococcus aureus (NCTC 6571), Plesiomonas Shigelloides (ATCC 51903) and Salmonella typhimurium (ATCC 13311), organisms which can cause illnesses from mild to severe with potentially fatal outcomes. The crude ethyl acetate, ethanol, methanol, acetone and aqueous extracts were screened by agar-well diffusion method and the activities of the extract were further determined by Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) assays. The inhibition zones ranged from 0 - 24 mm, while MIC and MBC of the extract ranged between 0.04 - 1.25 mg/mL and 0.081 - 2.5 mg/mL respectively. Chloroform/ Ethyl Acetate/ Formic acid (CEF) solvent system separated more active compounds followed by Ethyl Acetate/ Methanol/ Water (EMW) and Benzene/ Ethanol/ Ammonium Hydroxide (BEA). The extracts were fractionated by Thin Layer Chromatography (TLC). Bioautography was used to assess the activity of the possible classes of compounds present in the more active extracts. Column chromatography was used to purify the active compounds from the mixture while Gas Chromatography-Mass Spectrometry (GC-MS) was used to identify the phyto components of the fractions. The MIC of the fractions ranged between 0.0006 - 2.5 mg/mL. CEF 3 (F3), CEF 11 (F11) and CEF 12 (F12) revealed the presence of high levels fatty acids Linoleic acid, 1, 2-Benzenedicarboxylic acid and 2, 3-Dihydro-3, 5-dihydroxy-6-methyl, respectively. The results obtained from this study justify the use of this plant in traditional medicine and provide leads which could be further exploited for the development of new and potent antimicrobials.

Insights Into The Contribution Of Hfq In Salmonella Pathogenesis : Possible Role In Immune Evasion And Vaccine Development

Chapter I Introduction Salmonellae are facultative Gram-negative intracellular pathogens. Different serovars of it causes a variety of diseases in multiple hosts with different disease outcomes. Salmonella enterica serovar Enteritidis and Typhimurium (STM) can infect domestic animals causing gastroenteritis or typhoid like fever. Typhoid fever in humans which is actually caused by Salmonella enterica serovar Typhi still remains a significant health problem in many parts of the world with an estimated annual incidence of nearly 16 million cases and about 600,000 deaths. The infection begins via the orofecal route following which it invades the intestinal mucosa through several ways, namely by antigen sampling M cells, CD18 macrophages present in the intestinal lumen or via a forced entry in the non-phagocytic enterocytes. Upon entry, Salmonella resides in an intracellular phagosomal compartment called Salmonella containing vacuole (SCV) and has several strategies to counteract the host defense mechanisms. Following phagocytosis and its compartmentalization into Salmonella containing vacuole (SCV), a series of defense mechanisms are initiated. These include toxic reactive oxygen species or super oxide production, nitric oxide production, phagosomal acidification and release of hydrolases and defensins through fusion of phagosome with lysosomes generating highly bactericidal environment. The SCV transiently acquires endocytic markers like TfnR, EEA1, Rab4, Rab5, Rab11 and Rab7 and resist killing by avoiding phagosomal maturation and vesicular trafficking of iNOS and NADPH oxidase vesicles. Moreover, Salmonella also uses acidic pH of the SCV (~ pH 4.5) to assemble the Salmonella Pathogenecity Island 2 (SPI-2) type three secretion system (TTSS) which is essential for survival inside the macrophages. Salmonella uses these hostile conditions inside the host as cues for regulating their virulence factors using global regulatory factors. Hfq is one such global regulator playing an important role in many physiological processes and stress responses. Understanding the importance of Hfq regulated genes which impart Salmonella survival advantage under hostile conditions for successful infection will be of particular significance. The host too recognizes pathogen using innate immune receptors present either on the cell surface like TLRs (Toll Like Receptors) or inside the cells like NLRs (Nod Like Receptors). Innate immune receptors recognize pathogen associated molecular patterns (PAMPs) such as Lipopolysacharide (LPS), peptidoglycon (PGN), or hypomethylated DNA or RNA. Recognition of PAMPs by innate receptors leads, via activation of transcription factors (NF-κB and IRF3), to the generation of pro and anti-inflammatory cytokines, chemokines. Vaccination has been practiced for many years and it is one of the most effective methods of controlling infectious diseases like typhoid. At present two licensed vaccines against Salmonella are in use globally namely, Vi polysaccharide subunit vaccine (Typhim Vi™) and live attenuated typhoid vaccine (Vivotif Berma™). Lack of immunological memory, low efficacy (55-75 % protection) and requirement of higher number of doses are the important practical shortcomings associated with the currently used vaccines. So there is a need for a safer and immunogenic vaccine to combat Salmonella infection. Chapter II Salmonella Typhimurium lacking hfq gene induces long term memory response and confers protective immunity Currently available vaccines for typhoid have less-than-desired efficacy and certain unacceptable side effects, making it pertinent to search for new improved ones. Of the various strategies used for the generation of vaccine strains, focus is on manipulation of virulence regulator genes for bacterial attenuation. Hfq is a RNA chaperon which mediates the binding of small RNA to the mRNA and assists in post-transcriptional gene regulation in bacteria. Salmonella hfq deletion mutant is highly attenuated in vitro as well as in vivo implying its role in bacterial virulence. In this study, we have evaluated the efficacy of the Salmonella Typhimurium hfq deletion mutant as a candidate for live oral vaccine against Salmonella infection in murine salmonellosis model. The hfq deletion mutant is not only able to confer protection when administered orally to the mice against oral challenge with serovar Typhimurium virulent strain, but also elicits cross protective immune responses to other Salmonella serovars. The vaccine candidate appears to be safe for use in pregnant mice. This protection is partially mediated by the increase in the number of CD4+ T lymphocytes upon vaccination. STM hfq deletion mutant further exhibited significant increase in the lipopolysaccharide as well as outer membrane protein specific IgG in the serum as well as secretory S-IgA in the intestinal washes. In addition, vaccination led to an increased serum IFN-γ and IL-6. Taken together, our results suggest that the Salmonella Typhimurium hfq deletion mutant can be an excellent live oral vaccine candidate. Chapter III Acidic pH induced STM1485 gene governs intracellular replication and pathogenesis in Salmonella During the course of infection, Salmonella has to face several potentially lethal environmental conditions such as low pH both inside and outside the host. The ability to sense and respond to the acidic pH is crucial for survival and replication of Salmonella. Exposure to acidic pH results in the expression of large pool of virulence genes. One such gene highly up regulated inside the macrophage is STM 1485. In order to understand physiological role of STM 1485 in Salmonella pathogenesis, STM 1485 gene was deleted chromosomally and characterized in vitro and in vivo. In vitro the mutant did not show any growth defects at pH 4.5 and no difference in acid tolerance response. The 1485 deletion mutant was compromised in its capacity to proliferate inside the cells and is further lowered inside activated macrophages. We further showed that surface translocation of SPI-2 encoded translocon protein SseB was reduced at low pH in vitro in STM 1485 mutant and the mutant was found to colocalize with lysosomes higher than the wild type. In addition, the STM 1485 deletion mutant displayed decreased virulence in murine typhoid model when infected intragastrically. Based on our results, we hypothesize that the acid shock protein encoded by the STM 1485 might be involved in the formation of SPI-2 translocon at low pH and there by contributing to the virulence of Salmonella. Chapter IV Role of Nod1 in sensing vacuolar pathogen Salmonella in epithelial cells Nod1 and Nod2 are the archetypal members of the Nod like receptor family (NLR) and they recognize distinct peptidoglycan motifs of Gram-negative and Gram-positive bacteria respectively. Role of Nod1 and Nod2 in sensing bacterial pathogens have been elucidated. However, the role of Nod1 in sensing vacuolar pathogen Salmonella in epithelial cells is not understood. So in this study we investiged the role of Nod1 in the innate immune response against Salmonella in epithelial cells. We demonstrate that the recognition of Salmonella by Nod1 leads to NF-κB activation and this activation is diminished in epithelial cells expressing a dominant-negative Nod1 construct or Nod1 shRNA. Using a set of Salmonella mutants we show that the availability of ligand is higher when the bacteria were in cytosol rather than in vacuole. Further we also observed that the Nod1 mediated killing of Salmonella is mediated through the defensins. Based on our results we hypothesize that Salmonella uses its vacuolar niche to evade Nod1 mediated innate immune response.

Zur Wirksamkeit der postpartalen Ferkel-Impfung mit der attenuierten Lebendvakzine Salmoporc® bei der Infektion von Absatzferkeln mit Salmonella Typhimurium

Das Ziel dieser Arbeit war es, anhand von klinischen, kulturellen, serologischen sowie hämatologischen Parametern den frühen Einsatz des Salmonella-Lebendimpfstoffs Salmoporc® bei Saugferkeln in der ersten Lebenswoche zu prüfen. Dabei ergaben sich im Wesentlichen zwei zentrale Fragestellungen. Zum einen waren die Wirksamkeit sowie die Verträglichkeit der Vakzine bei der Anwendung bei Saugferkeln am dritten Lebenstag sowie im Alter von vier Wochen zu prüfen. Zum anderen war zu hinterfragen, ob der Antikörperstatus bzw. die präpartale Impfung von Muttersauen einen Einfluss auf den Impferfolg bei deren Ferkeln haben. Zu diesem Zweck wurden im Rahmen dieser Arbeit Impf- und Infektionsversuche mit Sauen und deren Ferkeln nach einem bereits etablierten Modell durchgeführt. Es wurden neun tragende und Salmonellen-freie Sauen in drei Gruppen zu je drei Sauen eingeteilt. Eine Gruppe Sauen wurde sechs sowie drei Wochen ante partum parenteral mit Salmoporc® geimpft. Deren Ferkel sowie die Ferkel von drei ungeimpften Sauen wurden am dritten und am 28. Lebenstag oral immunisiert. Die Ferkel der drei verbleibenden ungeimpften Sauen wurden nicht vakziniert und dienten als Kontrollgruppe. Die Versuche zeigten, dass die Impfung der Ferkel am dritten Lebenstag und in der 4. Lebenswoche sowohl bei den Ferkeln der nicht immunisierten Sauen wie auch bei den Ferkeln der vor der Geburt geimpften Sauen zu keinerlei klinischen Symptomen post vaccinationem führte, was für eine sehr gute Verträglichkeit der untersuchten Vakzine beim Saugferkel spricht. Die bakteriologische Untersuchung ausgewählter immunologisch und fleischhygienisch relevanter Organe von einem Teil der geimpften Ferkel am zehnten Lebenstag offenbarte zudem die noch erhaltene Invasivität des Impfstammes, welche einen entscheidenden Einfluss auf die Ausbildung einer belastbaren Immunität der Tiere post vaccinationem hat. So waren extraintestinale Nachweise des Impfstamms in den Tonsillen und Mandibularlymphknoten aller Tiere und bei einem Großteil der Tiere auch in den darmassoziierten Lymphknoten und teilweise in der Milz möglich. Am 49. Lebenstag wurden alle verbliebenen Ferkel via Magenschlundsonde intragastral mit je 1 x 1010 KbE eines Salmonella Typhimurium DT104-Wildstammes infiziert und anschließend über sieben Tage klinisch und bakteriologisch untersucht. Eine Woche post infectionem wurden schließlich ausgewählte Organe der Tiere kulturell auf den Infektionsstamm hin untersucht. Nach der Belastungsinfektion offenbarten sich deutliche klinische Effekte der Impfung. Bei den Tieren der nicht geimpften Kontrollgruppe wurden deutliche Symptome einer Salmonelleninfektion beobachtet, wohingegen bei den geimpften Ferkeln, unabhängig vom Impfstatus der Muttersauen, keinerlei Salmonelloseanzeichen feststellbar waren. Auch bei den kulturellen Untersuchungen zeigten sich deutliche Effekte der Impfung auf die Salmonellen-Ausscheidungskinetik. So schieden die geimpften Tiere beider Impfgruppen den vollvirulenten Challenge Stamm in signifikant niedrigerer Menge mit den Fäzes aus als die Kontrolltiere, weshalb die Saugferkelvakzinierung gut geeignet erscheint, die Salmonellenverbreitung durch die Fäzes infizierter Tiere zu reduzieren. Die bakteriologischen Organuntersuchungen zeigten in den lymphatischen Geweben, aber vor allem auch in den fleischhygienisch relevanten Organen deutliche Unterschiede zwischen den geimpften Ferkeln und den Kontrolltieren, die den Infektionsstamm in diesem frühen Infektionsstadium zu teilweise hohen Prozentsätzen in Leber, Milz und Unterarm-Muskulatur aufwiesen. Somit konnte gezeigt werden, dass der untersuchte Impfstoff auch bei der Anwendung beim neugeborenen Saugferkel gut verträglich ist und zudem eine belastbare Immunität induziert. Der Vakzinierungsstatus der Muttertiere hat hierbei keinen negativen Einfluss auf den Impfschutz der Ferkel. Die Anwesenheit maternaler Immunglobuline scheint sogar geeignet, die zelluläre Immunantwort der Ferkel in besonders hohem Maße zu stimulieren. Auch die Anforderungen nach § 5 der Schweine-Salmonellen-Verordnung vom 13. März 2007 an eine Salmonellenimpfung, die serologische Untersuchung auf Salmonellen-Antikörper nicht zu beeinträchtigen, werden durch Salmoporc® erfüllt.
