Temporal trends in West Antarctic accumulation rates: evidence from observed and simulated records

Reconstructed snow accumulation rate observations from five new firn cores show a statistically significant negative trend in accumulation rates over the past four decades across the central West Antarctic ice sheet. A negative temporal trend in accumulation is unexpected in light of rising surface temperatures and simulations run by GCMs. Both the magnitude of the mean accumulation rates and the range of interannual variability observed in the new records compares favorably to older records, suggesting that the new accumulation rate records may serve as a regional proxy for recent temporal trends in West Antarctic accumulation rates. The observed negative trend is likely the result of Southern Hemisphere high-latitude internal atmospheric dynamics, dominated by changes in the austral fall season. The well-documented positive trend in the Southern Annular Mode causes a low pressure center to form over the Amundsen Sea, which in turn produces lower accumulation rates across the western portion of the West Antarctic ice sheet. The new accumulation rate records are compared to several models/reanalyses to test the skill of simulated accumulation rate predictions. While the models/reanalyses and the new observations agree well in both mean and variability, the simulated records do not capture the full negative trend observed in the reconstructed records.

Samuel Brannan has almost become a legend in the history of the American West. Having led a group of Mormons around the Horn to California in 1846, he became a business and political leader in early California. He gained great wealth in speculation and merchandising, and is noted for his activities in the Vigilance Committee of 1851, his part in heralding the gold rush, and his developments in Calistoga and elsewhere.Although much has been written about Brannan, very little of it deals with the last years of his life. Very little factual information has been available on his activities during this period. These last ten years of his life are the focal point of this thesis. A large number of letters written by Brannan during this period have recently been found and with various secondary sources have shed light on his activities during these years.

New Hope was a small Mormon agricultural community in Central California. It was founded in 1846, by Samuel Brannan, on the hope that Brigham Young would make it the center stake of The Church of Jesus Christ of Latter-day Saints. The approximately twenty farmers at New Hope labored diligently under the illusion that thousands of their brethren would shortly join them. When President Young decided to settle in the Great Basin, the New Hope settlement was terminated that same year: 1847.

The play, Samuel Brannan, is the final result of a careful study of the history of Samuel Brannan. The information which has been selected to serve as the base for the plot was gleaned from the works of historians, letters written by Samuel, and individual journals. Although license has been taken with historical facts by the playwright in order to make this play theatrical, the play is based upon authentic happenings. The play is concerned with the events of Brannan's life, his unusual successes, and subsequent failures. It also treats his character, his attitudes, his beliefs, and the factors which motivated him to actions which caused his disintegration.

Sexuality And Courtships Of 19th Century Vienna Vs 21st Century America Through The Roles Of Alfred And Sam In Romance, Romance

For my thesis, I plan to research the sexuality and the process of courtship, and the differences between 19th century Vienna and modern America, specifically as to how it relates to my characters in Romance, Romance. My interest in the topic springs directly from the script. What I mean is, my initial idea of sexuality of the 19th century is very Victorian and straight-laced, where as I think of modern day as very sexually liberated. However, in the script, the first act, set in Vienna, is much more sexually explicit and active. In fact, the second act is all building up to an affair that does not even occur. Working as these characters, I believe that a historical view of what sexuality was actually like outside of Victorian England will lead me to a much better understanding of my character, and his particular past and desires for both sexual partners and potential mates. I also believe this research will assist me in constructing the modern character of Sam who can all at once love his wife, desperately want an affair, and cower away from said affair when the opportunity finally arises. Another interesting impact this research will create for my work is the juxtaposition of these two time periods and their individual codes of etiquette for behavior in this play. While the play is divided into two separate one-acts, they are presented together as one collective work, so the combination of each piece clearly leads to comparisons and further understanding of the other play. I believe this research will not only enlighten me about these two eras but also enlighten my performance, and assist me in effectively carrying out this complex pair of roles with a richer character developed. It also will provide me with a better understanding of the juxtaposition of these two plays, why they are paired together in the way they have been, and how this juxtaposition is effective in this particular work of musical theatre.

Mechanically Scanned Interference Pattern Structured Illumination Imaging

A method of lensless, single pixel imaging is presented. This method, referred to as MAS-IPSII, is theoretically capable of resolutions as small as one quarter of the wavelength of the imaging light. The resolution is not limited by the aperture of any optic, making high resolutions (including subwavelength) feasible even at very large (greater than a meter) distances. Imaging requires only flat optics and a coherent source, making it a good candidate for imaging with extreme wavelengths in the UV and x-ray regimes. The method is demonstrated by the imaging of various test targets. Both real and complex imaging (i.e. holography) is demonstrated.

A Step into Structural Biology: Structural Determination of TNK1-UBA and Computational Design of a Radical SAM Cyclase

Structural biology uncovers life's secrets by studying protein structures via techniques like X-ray crystallography. This knowledge drives advancements in protein engineering for the improvement of human lives. Yet, obtaining high-quality crystals in X-ray crystallography is challenging. To overcome this, we used Translocation ETS Leukemia protein Sterile Alpha Motif domain (TELSAM), a promising polymer-forming crystallization chaperone (PFCC), to enhance protein crystallization. Human thirty-eight-negative kinase-1 (TNK1), a key player in cancer progression, possess a ubiquitin association (UBA) domain that binds polyubiquitin and regulates TNK1 activity and stability. Although sequence analysis hints at an unconventional TNK1 UBA domain architecture, its molecular structure lacks experimental validation. To gain insight into TNK1 regulation, we fused the UBA domain to the 1TEL crystallization chaperone and obtained crystals diffracting as far as 1.53 Ã…. 1TEL enabled solution of the X-ray phases. GG and GSGG linkers allowed the UBA to reproducibly find a productive binding mode against its 1TEL polymer and to crystallize at protein concentrations as low as 0.1 mg/mL. Our findings support a TELSAM fusion crystallization mechanism, highlighting fewer crystal contacts compared to traditional crystals. Both modeling and experimental validation indicate that the UBA domain exhibits selectivity towards polyubiquitin chain length and linkages. Radical S-adenosylmethionine (SAM) enzymes catalyze various radical-mediated substrate transformations. Despite the growing interest of computational enzyme design in industrial small molecule synthesis, radical SAM enzymes remain relatively unexplored. We used PyRosetta to leverage hydrogen bonding design (hbDes) and hydrophobic interaction design (hpDes) to enable a radical cyclization reaction on our selected substrate. Although the purified enzymes demonstrated activation potential with a reducing agent, enzymatic assays failed to exhibit activity against the reactant. To obtain successful results, addressing additional questions and issues is required, which may involve the implementation of machine learning.
