Caractérisations biophysiques et structurales du complexe de réplication des Rhabdoviridae

Le virus de la stomatite vésiculaire (VSV) sert de modèle pour l'étude de la multiplication des virus (Mononegavirales) alors que la rage(RV) reste un sérieux problème de santé publique. Le génome de VSV et RV code notamment la nucléoprotéine (N) et la phosphoprotéine (P). N s'associe étroitement à l'ARN viral. Ce complexe N-ARN sert de matrice pour la réplication et la transcription virale. P est le cofacteur de la polymérase virale (L) et chaperonne N. En interagissant avec N-ARN (domaine C-terminal) et avec L (domaine N-terminal), P assure le lien physique entre l'ARN viral et L. La stœchiométrie de P, sa structure et son rôle exact pendant la transcription et la réplication restent incertains. Mon travail a consisté à une caractérisation biophysique et structurale de P et des complexes N-ARN-P pour mieux comprendre la dynamique du complexe de réplication de ces virus.<br />L'analyse biophysique montre que P RV & VSV existent sous forme de dimère allongé en solution. L'analyse bioinformatique a révélé une organisation modulaire, confirmé par des études biochimiques et biophysiques de mutants de P RV. La structure du domaine C-terminal de P VSV a été résolue par RMN et montre une homologie celle du C-ter de P RV. La caractérisation de l'interaction entre P et les anneaux N-ARN a révélé l'existence de deux types de complexes N-ARN-P (contenant un et 2 dimères de P par anneau). L'étude par ME des complexes nucléocapsides-P a permis de mettre en évidence un changement de conformation important.<br />Pour devenir accessible à L, l'ARN viral doit se dissocier localement de N. L'interaction N-ARN-P représente potentiellement une nouvelle cible pour le développement d'antiviraux.

Avaliação da potência do hormônio da paratireóide humano recombinante por bioensaio, métodos cromatográficos e eletroforético / Recombinant human parathyroid hormone potency evaluation by bioassay, chromatographic and electrophoretic methods

Fundação de Amparo à Pesquisa do Estado do Rio Grande do Sul - FAPERGS / The human parathyroid hormone (hPTH) is a polypeptide secreted by the parathyroid glands that is essential for the maintenance of the calcium ion homeostasis in the blood. The recombinant DNA technology has enabled the expression of hPTH gene in Escherichia coli, and thus the large-scale production of recombinant human parathyroid hormone (rhPTH 1-34), teriparatide, which contain the active amino-terminal fragment of the full length hPTH. The rhPTH is clinically used to treat osteoporosis at high risk of fractures in postmenopausal women, men with osteoporosis primary or hypogonadal and adults with glucocorticoid-induced osteoporosis (GIO). Cappilary zone electrophoresis (CZE) method was developed and validated for the assessment of rhPTH in biopharmaceutical formulations. The analysis for CZE method was performed on a fused-silica capillary (effective length, 40 cm; 50 μm i.d.), using electrolyte solution consisted of 50 mM dihydrogen phosphate solution at pH 3.0. The capillary was maintained at 25º C, the applied voltage was 20 kV. Injections were performed using a pressure mode at 50 mbar for 45 s, with detection by photodiode array detector set at 200 nm. Separation was obtained with a migration time of 5.3 min, and was linear over the concentration range of 0.25-250 μg mL-1 (r2 = 0.9992). The limits of detection and quantitation were 0.12 and 0.40 μg/mL, respectively. Specificity and stability-indicating capability were established in degradation studies, which also showed that there was no interference of the excipients. The accuracy was 100.28% with bias lower than 0.85%. Moreover, the in vitro cytotoxicity test of acidic, photolytic and thermal degradated forms showed significant differences (p<0.05) compared to intact molecule. The cell proliferation and alkaline phosphatase activity bioassays in UMR-106 cells were developed and applied to assess the biological activity of rhPTH in biopharmaceutical formulations The results of content/potency were correlated to those of the validated reversed-phase liquid chromatography (RP-LC), size-exclusion liquid chromatography (SE-LC) and CZE methods, showing significant correlation (p> 0.05) Thus, the application of the validated physico-chemical methods together with in vitro bioassays, was suggested to improve quality control of rhPTH biotechnology-derived product and to support studies of biosimilars. / O hormônio da paratireóide humano (hPTH) é um polipeptídeo produzido e secretado pelas glândulas paratireóides, e é fundamental para a manutenção da homeostase dos íons cálcio no sangue. A tecnologia do DNA recombinante possibilitou a expressão do gene do hPTH em Escherichia coli, e a produção em grande escala do hormônio da paratireóide humano recombinante (rhPTH 1-34), também denominado Teriparatida, o qual apresenta a sequência de aminoácidos responsável pela porção biologicamente ativa do paratôrmonio natural. O rhPTH é clinicamente indicado para o tratamento da osteoporose de alto risco de fraturas em mulheres pós-menopausa, de homens com osteoporose primária ou hipogonadal, e da osteoporose associada à terapia sistêmica com glicocorticóides. Neste trabalho foi desenvolvido e validado método por eletroforese capilar de zona (ECZ) para a avaliação de rhPTH em produtos biofarmacêuticos. No método por ECZ, utilizou-se capilar de sílica fundida (40 cm de comprimento efetivo x 50 μm d.i.) e solução eletrolítica composta de fosfato de sódio dihidrogenado 50 mM, pH 3,0. O capilar foi mantido a temperatura de 25ºC, e a tensão aplicada foi de 20 kV. O tempo de injeção foi de 45 s, com pressão de 50 mBar, e detecção por arranjo de diodos (DAD), em 200 nm. A separação eletroforética foi obtida com tempo de migração de 5,3 min, sendo linear na faixa de concentração de 0,25-250 μg/mL (r2 = 0,9992). Os limites de detecção e quantificação foram de 0,12 e 0,40 μg/mL, respectivamente. A especificidade foi avaliada através de análises com os excipientes da formulação biofarmacêutica e estudos de degradação, demonstrando a seletividade do método. A exatidão foi 100,28% com bias inferior a 0,85%. Além disso, realizou-se o teste de citotoxicidade in vitro das formas degradadas, apresentando, para as amostras submetidas às condições ácida, fotolítica e térmica, diferença significativa (p< 0,05) em relação à molécula íntegra. Os bioensaios de proliferação celular e da atividade da fosfatase alcalina em células UMR-106 foram desenvolvidos e aplicados para avaliação da atividade biológica de rhPTH em formulações biofarmacêuticas. Os resultados de teor/potência foram correlacionados com os métodos já validados por cromatografia líquida em fase reversa (CL-FR), cromatografia líquida por exclusão molecular (CL-EM) e ECZ, apresentando correlação significativa (p> 0,05). Assim, sugere-se que o métodos físico-químicos validados sejam aplicados paralelamente aos bioensaios in vitro para aprimorar o controle da qualidade do produto biotecnológico de rhPTH, e para avaliação da biossimilaridade de rhPTH.

Etude comparative des matériaux de garnissage dans les réacteurs de filtration pour l’assainissement non collectif / Comparative study of packing materials of filtration reactors for on-site wastewater treatment

L'assainissement non collectif concerne 12 à 15 millions de personnes en France. La filière classique de ce mode d’assainissement se compose généralement d'un prétraitement anaérobie par une fosse septique recevant l’ensemble des eaux usées domestiques suivi d’un système d’infiltration dans le sol ou d’un filtre à sable. Le filtre à sable vertical drainé met à profit le pourvoir épuratoire qui est principalement lié à la présence d’une biomasse sous forme d’un biofilm. Cette dynamique de la croissance de la biomasse ou du biofilm est soumise à l’impact de la nature de matériaux filtrants. L’écoulement insaturé dans ces systèmes conditionne également cette croissance du biofilm. Dans ce contexte, l'objectif du travail de la thèse est d’appréhender les mécanismes mis en jeu et particulièrement l’impact des matériaux dans le fonctionnement des filtres en comparant notamment deux types de matériaux: les sables de rivière et les agrégats concassés. Pour cela, une étude expérimental sur une unité pilote composé des réacteurs de filtration du diamètre de 30cm et différents épaisseurs de garnissage (15, 30 et 70cm) a été construite. Les réacteurs garnis de deux sables roulés et deux agrégats concassés, sont alimentés en effluent septique avec une charge volumique 12cm/jour par 10 bâchés par jour. Suite des matériaux, une étude de la performance épuratoire avec le suivi des composants biochimiques de la biomasse totale et de la matrice extracellulaire du biofilm est réalisée en comparant notamment les deux types de matériaux filtrants. / The onsite wastewater treatment systems concern 12 to 15 million of people in France. The treatment plant is generally composed by a septic tank as pretreatment, followed by soil infiltration field or sand filtration bed. The vertical drained sand filter provides the purification capacity thanks to the presence of a biomass in form of the biofilm. The dynamic of the biomass growth or the biofilm development is under the impact of filter materials’ nature. In this context, the objective of this work is to understand the mechanisms involved and especially the impact of medium in the functioning of the filtration reactor by comparing two types of packing materials: river sands and crushed aggregates. For this purpose, an experimental study is conducted with pilot unity composed by filtration reactors of 30cm of diameter and different packing thicknesses (15, 30 and 70cm). The reactors packed with two river sands and two crushed aggregates are fed with septic effluent with a volumic hydraulic charge of 12cm/day by 10 batches per day. Based on a characterization of materials, a study of purification performance with biochemical components monitoring of the total biomass and the extracellular matrix of the biofilm is realized by comparing two types of filter materials. The purification performance has presented similar efficiencies of particulates and organic matters removals by fine river sand and fine crushed aggregate. The nitrogen pollutants removals are more effective in the fine river sand which presents the finest granulometry with an alternative of aerobic and anoxic phases along the reactor depth and with a biomass more abundant. The distribution and the quality of the total biomass and the extracellular matrix differentiate between the river sand and the crushed aggregates. As the reference material, the fine river sand presented an earlier stabilization of total biomass growth with a less important production of extracellular exudates compared to the crushed aggregates. The origin of impacts brought by the crushed aggregates might be due to the higher fine particles content which created microenvironments poor in substrates or in oxygen and also due to a more heterogeneous mineralogy. The extracellular components of highest percentage in the biofilm of crushed aggregate are polysaccharides type substances.

Size Separation Techniques for the Characterisation of Cross-Linked Casein: A Review of Methods and Their Applications

Casein is the major protein fraction in milk, and its cross-linking has been a topic of scientific interest for many years. Enzymatic cross-linking has huge potential to modify relevant techno-functional properties of casein, whereas non-enzymatic cross-linking occurs naturally during the storage and processing of milk and dairy products. Two size separation techniques were applied for characterisation of these reactions: gel electrophoresis and size exclusion chromatography. This review summarises their separation principles and discusses the outcome of studies on cross-linked casein from the last ~20 years. Both methods, however, show limitations concerning separation range and are applied mainly under denaturing and reducing conditions. In contrast, field flow fractionation has a broad separation range and can be easily applied under native conditions. Although this method has become a powerful tool in polymer and nanoparticle analysis and was used in few studies on casein micelles, it has not yet been applied to investigate cross-linked casein. Finally, the principles and requirements for absolute molar mass determination are reviewed, which will be of increased interest in the future since suitable calibration substances for casein polymers are scarce.

L’objectif de ce travail de thèse était d’étudier l’effet de la stérilisation par électrons accélérés sur les copolymères d’oléfines cycliques (COC), utilisés comme conditionnement de produits pharmaceutiques, ainsi que son impact sur les interactions avec des formulations pharmaceutiques. Grâce à la méthodologie analytique adoptée qui a fait appel à différentes techniques de caractérisation, telles que la chromatographie d’exclusion stérique, la chromatographie liquide haute performance à polarité de phases inversée, la spectroscopie infra rouge à transformée de Fourier, la microscopie à force atomique et les mesures d’angles de contact, nous avons pu mettre en évidence différents types de modifications dans le volume et sur la surface du matériau après stérilisation ainsi qu’après vieillissement. La modification principale du dans la masse du matériau, observée à la dose réglementaire de stérilisation (25 kGy), est la scission des chaînes du polymère, qui s’accompagne de la création de composés de faible masse molaire, donc de migrants potentiels risquant d’influencer la sécurité d’emploi des COC. En effet, certains de ces composés ont été retrouvés avec une concentration relativement importante dans les solutions de mise en contact avec les COC stérilisés, et notamment en solution aqueuse. Toutefois, l’étude préliminaire de toxicité a montré l’absence de cytotoxicité des extractibles obtenus à la dose de la stérilisation.Les modifications relatives à la surface des COC radio-stérilisés sont, quant à elles, de deux natures : physique avec une augmentation de la rugosité de surface et chimique avec la formation de produits d’oxydation polaires ; ces deux types de modifications conduisent à l’augmentation de la mouillabilité de surface. Cependant dans certains cas, notamment après vieillissement, ces modifications sont relativement faibles, même à des doses supérieures à celle préconisée pour la stérilisation, ce qui peut être corrélé à l’absence de l’effet de l’irradiation sur le comportement des COC vis-à-vis des solutions médicamenteuses. En effet, aucune variation de la sorption des principes actifs choisis n’a été montrée entre les COC irradiés et non irradiés vieillis. / The aim of this work was to study the effect of electron beam radio-sterilization on cyclo olefins copolymers (COC) used as pharmaceutical storage materials, as well as to investigate its impact on the interaction with pharmaceuticals formulations. Due to the analytical methodology used which dealt with different techniques of characterization such as size exclusion chromatography, reversed phase high performance liquid chromatography, Fourier transformed infrared spectroscopy, atomic force microscopy and contact angle measurements, we have been able to put into evidence different kinds of modifications both in the bulk and on the surface of the sterilized material and also after ageing.The principal modification of material’s bulk, observed at the recommended dose for sterilization (25 kGy), was polymer chains scissions, accompanied with creation of low molecular weight compounds, that are potentials migrants that risk to affect the safe use of COC. Indeed, some of these compounds have been found with a relatively important concentration in the solutions where sterilized COC was stored, especially in aqueous solutions. However, the preliminary study of toxicity has shown the absence of cytotoxicity of the extractables obtained at the sterilization dose.Surface modifications of radio-sterilized COC are of two types: a physical one, with an increase of the surface’s roughness and a chemical one with the formation of polar oxidation products; these two modifications result in an increase of surface’s wettability that may be important. However, in some cases such as for aged samples, these modifications are relatively weak even at doses higher than the one recommended for sterilization, which can explain the absence of the effect of radiation on the behavior of COC towards drug solutions. Indeed, no variation of drug sorption has been observed between aged COC irradiated and none irradiated.

Ausgewählte Eigenschaften des Sporopollenins der Kiefer

Gegenstand der Arbeit sind Zusammenhänge zwischen physikochemischen Eigenschaften und Funktionen der Exine bei Ausbreitung, Bestäubung und Befruchtung. Dabei bewährte sich der Einsatz der 3-kammrigen Sporopolleninkapseln (Zentralkapsel und Sacci) in der Permeationschromatographie. Sowohl kinetisch bedingte chromatographische Dispersion kleiner Moleküle als auch Konzentrationsänderungen von Zuckern und Dextranmolekülen im Medium wurden zur Bestimmung von Permeabilitätskoeffizienten der Nexine genutzt. Die Wasserabsorptionskapazität von Exinefragmenten und die hydraulische Leitfähigkeit der Nexine wurden anhand von Konzentrationsänderungen ausgeschlossener Dextranmoleküle ermittelt. Das Tectum der saccalen Sexine ist eine Mikrofiltermembran mit scharfer Trenngrenze im Submikrometerbereich; daher werden an den Sacci nur Hydrokolloide mit Stokes''schen Radius über 100 nm (z.B. aus nativem Dextran) ausgeschlossen. Die Nexine ist eine nicht-ideale Umkehrosmose-Membran, die in Zucker- und Salzlösungen hohe Reflexionskoeffizienten zeigt; zusätzlich besitzt sie wenige große Poren, die den Austausch von Zuckern und selbst kleinen Polymermolekülen ermöglichen. Die hydraulische Leitfähigkeit der Nexine liegt im Größenbereich derjenigen von Plasmamembranen (0,39-0,48 µm s-1 MPa-1); die Ergebnisse zeigen, dass die Exine weder die Nährstoffaufnahme des Sporoplasten aus der lokulären Flüssigkeit noch dessen rasche Rehydratation in der Mikropyle behindert. Die Einfaltungen der distalen Nexine (oberhalb der Sacci) und die Omega-Faltung der Exine zwischen den Sacci (Leptom) bieten beim Quellvorgang Schutz vor zu schneller Flächenausdehnung der Plasmamembran. Der Corpus kann mit konzentrierten Elektrolytlösungen beladen werden. Beim anschließenden osmotischen Schwellen in Wasser reißt die Exine, und der Sporoplast wird mit anhaftender Intine ausgeschleudert. Wasser und andere polare Flüssigkeiten adhärieren stärker als hydrophobe Flüssigkeiten an Sporopollenin. Die Sporopolleninmatrix weist eine hohe Feststoffdichte auf, ist wenig quellfähig (0,18 mL g-1 TM) und deformationsstabil. Dies ermöglicht die Pulverbildung beim Trocknen. / Subject of this thesis are relationships between physicochemical properties and functions of the exine concerning propagation, pollination and fecundation. Here the application of the 3-chambered sporopollenin-microcapsules (central capsule and sacci) in permeation chromatography proved of value. Both the kinetically dependent dispersion of small molecules and changes in concentration of sugars and dextran molecules in the medium were analysed to determine permeability coefficients of the nexine. The water absorption capacity of exine fragments and the hydraulic conductance of the nexine were calculated by means of changes in concentrations of excluded dextran molecules. The tectum of the saccal sexine is a microfiltration membrane with a sharp cut off in the submicrometer range; thus hydrocolloids with Stokes´radii over 100 nm (e.g. from native dextran) are excluded from the sacci. The nexine is a non-ideal reverse osmosis membrane having high reflexion coefficients in sugar and salt solutions; in addition few large pores allow the exchange of sugars and even of small polymers. The hydraulic conductance of the nexine is in the range typically for plasmamembranes (0.39-0.48 µm s-1 MPa-1); the results indicate that the exine does neither obstruct the uptake of nutrients by the sporoplast from the locular fluid nor hinder the rapid rehydration in the micropyle. When rehydrating, the distal foldings of the nexine (above the sacci) and the omega-like folding of the exine between the sacci (leptom), provide protection for the plasmamembrane when its surface area has to increase too rapidly. The corpus can be loaded with a concentrated electrolyte solution. When subsequently transferred into water the exine rupture and the sporoplast along with the intact intine is ejected. Water and other polar liquids adhere stronger to sporopollenin than hydrophobic ones. The matrix of sporopollenin show a high density in its solid content, water absorption capacity is low (0.18 mL g-1 DM) and it is resistant to deformation. This enable the formation of powder while dehydrating.
