Synthesis, Spectroscopic Studies, and Computational Analysis of a Solvatochromic Phthalocyanine Derivative

A near-IR absorbing phthalocyanine containing alkylthio and alyklamino substituents along the periphery was determined to have optical properties that vary depending on surrounding solvent environment. Solvatochromic behavior of this novel chromophore has been characterized through UV-Vis absorbance and fluorescence spectroscopy. Conformational analysis using the B3LYP/6-31G* model indicates the lowest energy conformer with hydrogen bond like interactions between the sulfur lone pair and the hydrogen on the alkylamino substituent. TD-DFT analysis of the solvatochromic phthalocyanine and its parent molecule confirms the derivative exhibits tunable optical properties based on various solvent parameters. Chapter 1 reviews the energy crisis and the potential biological implications of near-IR absorbing phthalocyanines. In addition, a brief introduction of TD-DFT is given to further describe the computational analysis utilized herein. Chapter 2 describes the synthesis, spectroscopic studies, and computational analysis of a near-IR absorbing solvatochromic phthalocyanine derivative. Chapter 3 provides a summary of the work described in this thesis and provides future directions based on the completed research.

Espectro de Absorção da Orto-betaína em Água: um Estudo Teórico usando o Método Seqüencial Monte Carlo/Mecânica Quântica / Absorption spectrum of the ortho-betaine in Water: a Study Theoretical Method using Sequential Monte Carlo/Mechanics Quantum

Statistically uncorrelated configurations have been obtained from MC simulations with unpolarized and in-solution polarized solute. To account for the solute electronic polarization, we have used an iterative procedure based on the S-QM/MM methodology to determine the in-solution dipole moment of orto-betaine in presence of water molecules treated as point charges. MP2/cc-pVDZ calculations on the configurations bearing the orto-betaine molecule electrostatically embedded in the 320 water molecules show that the solute polarization increases the dipole moment of orto-betaine, compared to gas phase, by 49%, giving the in-solvent dipole values of 12.2 D. MC simulation results for the Helmholtz free energy as function of the torsion angle indicate that the mosts table conformer of the orto-betaine in water is that with an angle of 60o, after including solute polarization. The average number of solute-solvent hydrogen bonds was analyzed and it was found a large increase of 3,25 in polarized situation to only 1,82 in the unpolarized situation. Modeling the water molecules by point charges (electrostatic embedding), the results show that solvent effects on the absorption spectrum have a significant contribution of the electrostatic interactions. In comparison with the result of gas-phase, TD-BHandHLYP/6311+G(2d,p) calculations give for the π → π∗ electronic transition solvatochromic shifts of 0.42 eV and 0.90 eV for the unpolarized and polarized orto-betaine, respectively. The experimental spectral shift value of 0.90 eV is very well reproduced by the present theoretical results after including the solute polarization. The TD-B3LYP/6311+G(2d,p) model gives the corresponding value for the polarized orto-betaine of 0.52 eV which is an underestimated result in qualitative agreement with experiment. This study indicates that the inclusion of the solute polarization is important for a reliable description of the spectral shifts considered here. / Usando seqüencialmente simulações Monte Carlo e cálculos de mecânica quântica (S-QM/MM) baseados no método TD-DFT nós estudamos o deslocamento solvatocrômico da orto-betaína em água. Configurações estatisticamente descorrelacionadas foram obtidas de simulações Monte Carlo com o soluto polarizado e não polarizado em solução. Para levar em conta a polarização eletrônica do soluto, nós usamos um processo iterativo baseado na metodologia S-MC/QM para determinar o momento de dipolo da orto-betaína em solução na presença de moléculas de água tratadas como cargas pontuais. Cálculos MP2/cc-pVDZ nas configurações contendo a molécula de orto-betaína embebida eletrostaticamente em 320 moléculas de água mostram que a polarização do soluto aumenta o momento de dipolo da orto-betaína, comparado com a fase gasosa, em 49% fornecendo o valor de 12,2 D em solução. Os resultados das simulações Monte Carlo para a energia livre de Helmholtz como função do ângulo de torção indicam que o confôrmero mais estável da orto-betaína em água é aquele cujo ângulo é 60◦, após a inclusão da polarização do soluto. O número médio de ligações de hidrogênio soluto-solvente foi analisado tendo sido encontrado um decréscimo substancial de 3,25 ligações na situação polarizada, para somente 1,82 ligações na situação não polarizada. Modelando as moléculas de água por cargas pontuais (embebidas eletrostaticamente) os resultados mostram que os efeitos de solvente no espectro de absorção têm uma contribuição importante de interações eletrostáticas. Em comparação com o resultado de fase gasosa, cálculos TD-BHandHLYP/6-311+G(2d,p) fornecem para a transição eletrônica π → π∗ um deslocamento solvatocrômico de 0,42 eV e 0,90 eV para a orto-betaína não polarizada e polarizada, respectivamente. O modelo TD-B3LYP/6-311+G(2d,p) fornece para a orto- betaína polarizada o correspondente valor de 0,52 eV, que é um resultado subestimado, em acordo apenas qualitativo com o experimento. Este estudo indica que a inclusão da polarização do soluto é importante para uma descrição realista do deslocamento no espectro considerado.

Solvatação por solventes puros e suas misturas: relevância para química e química verde / Pure solvents and their mixtures solvation: relevance to chemistry and creen chemistry

Neste trabalho, sondas solvatocrômicas foram empregadas no estudo de solventes puros, misturas aquosas de solventes próticos e apróticos, líquidos iônicos e suas misturas aquosas. As sondas estudadas foram agrupadas em duas séries, cujas mudanças relevantes foram: (i) série RPMBr2: R de metila a 1-octila permitiu a variação da hidrofobicidade da sonda enquanto o pKa se manteve constante; (ii) série de merocianinas derivadas de piridina, quinolina e acridina. Isso resultou em sondas de hidrofobicidades diferentes com valores parecidos de pKa. Desta forma foi possível isolar os efeitos sobre solvatação provocados pela hidrofobicidade das sondas. As duas séries de sondas mostraram comportamentos solvatocrômicos diferentes. Uma equação modificada de Taft-Kamlet-Abboud permitiu as quantificações individuais de cada propriedade do solvente, tais como acidez, dipolaridade/polarizabilidade e hidrofobicidade para a resposta de cada sonda. O modelo de solvatação preferencial que considera explicitamente a presença de três espécies na mistura binária de solvente foi aplicado, este considera que a água, o solvente orgânico e a espécie solvente orgânico-água competem pela camada de solvatação da sonda. Os resultados deste tratamento ajudaram no entendimento das forças atuantes na solvatação, especialmente com relação aos efeitos das propriedades de ambos a sonda e o solvente, e do aumento da temperatura. Estudos preliminares de misturas aquosas de líquidos iônicos mostraram as semelhanças e as diferenças entre a solvatação por estas misturas \"verdes\" e a por misturas aquosas de álcoois. Organokatalysierte Kaskadenreaktionen ungeschützer Kohlenhydrate und Chromophorsynthese zur Untersuchung von Wasser- und Protonierungsdynamiken

In der vorliegenden Dissertation konnten erfolgreich neben der Entwicklung und Optimierung neuer Methoden in der Kettenverlängerung von Aldosen und Ketosen auch neue Farbstoffe zur Untersuchung von Protonierungs- und Wasserdynamiken designt und synthetisiert werden. Die Erweiterung des Verständnisses im Zusammenspiel von Aminosäuren und Kohlenhydraten hat einen großen Einfluss in der wissenschaftlichen Gesellschaft der Kohlenhydratchemie. Die Kontrolle im Aufbau von Stereotetraden und -pentaden führt zu einer Ausweitung der bekannten Möglichkeiten in der stereoselektiven Synthese von Naturstoffen und Biomimetika. Außerdem konnten durch eine einfache Methode die C-Glykoside seltener Kohlenhydrate dargestellt werden. Einbau von Aminosäuren in Kohlenhydratstrukturen konnten in hohen Stereoselektivitäten und der Möglichkeit der Stereomanipulation durch Wahl des Isocyanides erreicht werden. Die Synthese des zellgängigen Farbstoffes PAc-SNARF, sowie des Biolinkers IA-SNARF ermöglicht eine bildgebenden ratiometrischen pH-Untersuchungen in Zellen und auf Proteinoberflächen. Die Verallgemeinerung der Synthese der Farbstoff-Precursor für die SNARF-Derivate über eine Friedel-Crafts-Acylierung erlaubt eine kostengünstige Darstellung einer großen Bandbreite von Farbstoffen. Mithilfe der neu synthetisierten sterisch anspruchsvollen N-Methyl-6-oxychinoliniumbetain-Derivate verbunden mit der Fluoreszenzaufkonvertierungs-Spektroskopie konnte eine Verlangsamung von Wasser an hydrophoben Oberflächen bewiesen werden. Die gesammelten Ergebnisse und Erkenntnisse in diesen verschiedenen Themengebieten werden in Zukunft einen großen Einfluss in der Wissenschaftswelt haben. / In the presented dissertation new methods in the chain elongation of carbohydrates could be established and optimized. Besides that, new probes for the investigation of protonation and water dynamics could be designed and synthesized. The extension of comprehension in the interaction between amino acids and carbohydrates through hydrogen bonds has a great impact in the scientific community of carbohydrate research. The stereochemical control in the construction of stereotetrads and –pentads leads to a considerable extension of known methods in the synthesis of natural compounds and biomimetics. Additionally the C-glycosides of rare carbohydrates could be synthesized through simple methods. Installation of amino acids into carbohydrate structures could be achieved with very high stereoselectivity and the potential of manipulating the stereochemical course through the choice of different isocyanides. The synthesis of the cell permeable PAc-SNARF and the cysteine-bioapplicable IA-SNARF allow the ratiometric pH-imaging of cells and protein surfaces. The generalization of the synthesis of dye-precursors for SNARF-derivatives through friedel-crafts-acylation allow an inexpensive approach in synthesizing a broad spectrum of dyes. Through deployment of newly developed sterical demanding N-Methyl-6-oxyquinolinium betaine-derivates together with the fluorescence upconversion spectroscopy a deceleration of water reorientation near hydrophobic surfaces could be proven. The here summarized results and insights in the different topics will have a considerable influence in academic sciences.

Ein weiteres Augenmerk lag auf der Untersuchung der Wechselwirkung zwischen den ILs und silikatischen Oberflächen. Neben der Studie der anionenvermittelten starken Physisorption von 1-Methylimidazoliumchlorid an Aerosil®300 konnte auch eine neuartige Methode zur gezielten Chemisorption von Imidazolium- und Phosphoniumkationen an Siliziumdioxidoberflächen entwickelt werden. Dabei wurden verschieden Carbene und Ylide als basische IL-typische Kationenprecursoren eingesetzt. Die Analyse der erhaltenen Materialien erfolgte mit Hilfe verschiedener Methoden der Festkörper-NMR-Spektroskopie.
