Distyly, pollen flow and seed set in Menyanthes trifoliata (Menyanthaceae)

The influence of variation in style length on pollen flow and seed set was examined in six populations of Menyanthes trifoliata in southwestern British Columbia to evaluate Ganders' hypothesis that morphological distyly increases the fecundity of a diallelic self-incompatible plant. In five populations, Menyanthes was distylous and self-incompatible. The sixth population consisted of pins and morphological homostyles (thrums with unusually long styles). In each population morph frequency, pollen frequency, the composition of stigmatic pollen loads and seed set were estimated. Results from the six populations demonstrate that the size and composition of stigmatic pollen loads fluctuates erratically during the flowering season. Pins and thrums experienced disassortative pollination, assortative pollination and random pollination at different times during the season. Homostyles were always assortatively pollinated. Among populations there was a high correlation between morph frequency (pollen frequency) and the composition of stigmatic pollen loads. However, in anisoplethic populations of Menyanthes morphological distyly seems to compensate for the rarity of one floral form, by increasing the proportion of compatible pollen received by the opposite floral form. Comparison of the composition of stigmatic pollen loads of homostyles with those of thrums revealed that the separation of stigmas and anthers in distylous flowers of Menyanthes reduces the number of incompatible pollen grains received, and the reciprocal placement of stigmas and anthers appears to increase the number of compatible pollen grains received. Seed set in the six populations of Menyanthes was always below the potential maximum. Among populations there was a high correlation between the number of compatible pollen grains received and seed set. In at least one population, pollen availability was a major factor limiting seed set, but other factors are probably influencing seed set in Menyanthes as well. Thrums set significantly more seeds per capsule than homostyles. In populations of Menyanthes, the reciprocal placement of stigmas and anthers in distylous flowers (compared to homostylous flowers) increases the amount of compatible pollen deposited on stigmas, and this increase is associated with greater fecundity. / Science, Faculty of / Botany, Department of / Graduate

O Brasil é o maior produtor de laranja do mundo, liderando também a produção e exportação de suco concentrado congelado. Um dos fatores que ameaçam o setor é o grande número de doenças, principalmente, as bacterianas como o Huanglongbing (HLB). O HLB é considerado uma das mais graves doenças dos citros, que compromete seu desenvolvimento e produção, podendo levar à morte das plantas afetadas. Os agentes associados a esta doença são bactérias endógenas restritas ao floema (Candidatus Liberibacter spp.). No gênero Citrus, ainda não foram encontradas espécies resistentes ao HLB, o que tem estimulado a utilização de ferramentas biotecnológicas, como a transformação genética, para auxiliar os programas de melhoramento genético, nos quais genes de interesse agronômico são introduzidos visando resistência a doenças. Na busca por uma planta transgênica resistente ao HLB, é desejável obter construções gênicas nas quais o gene de interesse se expresse, preferencialmente, na região em que a bactéria coloniza a planta, ou seja, no caso do HLB, no floema. Este trabalho objetivou avaliar plantas transgênicas de citrange \'Carrizo\' [Poncirus trifoliata (L.) Raf. x Citrus sinensis (L.) Osbeck] com gene uidA (β-glucuronidase), dirigido pelos promotores dos genes CsPP2.1 (Phloem protein 2) e CsVTE2 (homogentisato fitiltransferase) obtidos de Citrus sinensis. Foram analisadas as especificidades de cada promotor, e estes foram comparados com a ação do promotor constitutivo CaMV35S. Análises moleculares, como Southern blot e qPCR, foram realizadas a fim de verificar o número de cópias do transgene em cada planta avaliada, e quantificar a expressão relativa do gene uidA dirigido pelos diferentes promotores, respectivamente. Análises histológicas foram feitas para verificar a localização da expressão do gene repórter GUS (uidA). A expressão relativa do gene uidA dirigido pelos promotores específicos foi mais baixa quando comparada com o promotor constitutivo CaMV35S, uma vez que estes promotores direcionam a expressão apenas para tecidos específicos. O promotor CsPP2.1 direcionou a expressão do gene uidA para o tecido floemático, mais especificamente junto às células companheiras e elementos de tubo crivado. O promotor CsVTE2 direcionou a expressão do gene repórter preferencialmente para a periferia do parênquima cortical e para as células guarda, o que corresponde às áreas com presença de cloroplastos, além de também direcionar a expressão para o tecido floemático. Apesar de não ser específico de floema, o promotor CsVTE2 constitui-se em uma boa alternativa para construções visando o combate a doenças vasculares por estar envolvido em importantes processos fisiológicos da planta, como o combate a estresses oxidativos e o carregamento de fotoassimilados no floema. Os resultados sugerem que CsPP2.1 e CsVTE2 são promotores potenciais para serem utilizados em construções gênicas visando o controle de doenças vasculares tais como o HLB. / Brazil is the world\'s largest sweet orange producer, leading also the export of frozen concentrated orange juice. However, the citrus industry is constantly threatened by diseases, particularly those caused by bacteria, such as Huanglongbing (HLB). HLB is considered one of the most harmful citrus diseases, leading the plants to their death. The associated agents of this disease are endogenous bacteria restricted to the phloem of the plants (Candidatus Liberibacter spp.). No resistance has been found in Citrus spp. that could be used in breeding programs. Therefore, biotechnological strategies, such as genetic transformation, could help such programs to introduce genes for disease resistance. It is desirable the use of gene constructs that are preferentially expressed where the bacterium colonizes the plant, i.e., the phloem in plants affected by HLB. This study aimed to evaluate transgenic plants of \'Carrizo\' citrange [Poncirus trifoliata (L.) Raf. x Citrus sinensis (L.) Osbeck] with uidA gene (β-glucuronidase), driven by the promoters of CsPP2.1 (Phloem protein 2) and CsVTE2 (homogentisato fitiltransferase) genes obtained from Citrus sinensis plants. Each promoter specificity was analyzed and gene expression was compared with that induced by the constitutive promoter CaMV35S. Molecular analyses such as Southern blot and qPCR were performed in order to verify the number of transgenic copies in plants and to quantify relative expression of the uidA gene driven by different promoters, respectively. Histological analyses were done in order to identify the exact location of the GUS (uidA) reporter gene expression. Relative expressions of the uidA gene driven by specific promotors were lower than that driven by the CaMV35S constitutive promoter, since the expression was limited to specific tissues. The CsPP2.1 promoter drove the uidA gene expression to the phloem tissue, specifically to the companion cells and sieve elements. On the other hand, CsVTE2 promoter drove the reporter gene expression mainly to the periphery of the cortical parenchyma and to the guard cells, where high number of chloroplasts were present, and also drove the expression to the phloem. Although the CsVTE2 promoter is not phloem-specific, it might be used in gene constructs aiming vascular disease control, since it is involved in physiological processes such as mitigating oxidative stress and photoassimilate phloem loading. The results suggest that CsPP2.1 e CsVTE2 are promising promoters to be used in gene constructs in order to control vascular diseases, such as HLB.

Antiplasmodial and larvicidal flavonoids from Derris trifoliata

From the dichloromethane-methanol (1:1) extract of the seed pods of Derris trifoliata, a new flavanone derivative (S)-lupinifolin 4´-methyl ether was isolated. In addition, the known flavonoids lupinifolin and rotenone were identified. The structures were determined on the basis of spectroscopic evidence. Lupinfolin showed moderate in vitro antiplasmodial activity against the D6 (chloroquine-sensitive) and W2 (chloroquineresistant) strains of Plasmodium falciparum. The different parts of this plant showed larvicidal activities against Aedes aegypti and rotenoids were identified as the active principles.

The leves it was collected in Volta Grande of Xingú, Altamira, PA (collect 1) and Lake of Catalão, Manaus, AM (collect 2 e 3). The dried specimen’s it was deposited in the herbarium of the Federal Institute of Amazonas (IFAM) with record numbers 10653 and 10654. The collected material was dried and pulverized. The collect 1, was extracted by dichloromethane (DCM), methanol (MeOH) and water (H2O). The collect 2 and 3 was extracted by hexane, methanol and water. The preliminary phytochemical analysis of leaves and fungi showed that dichlorometane and hexane extracts contain terpenes and methanolic extracts contain phenolic compounds. The broths from fungi showed that diclorometânica phase (DCM) contain terpenes, and phase of Ethyl Acetate (AcOEt) contain terpene and phenolic compounds. The phytochemical study of the leves led to the isolation of: Friedelan-3-one (I), 30-hydroxy-friedelan-3-one (II), Lupeol (III), the mixture of β -Sitosterol and Stigmasterol steroids (IV and V), hydroxycoriatin (VI), the mixture of α and β-amyrin (VII and VIII ), 7- hydroxy-6-methoxy-coumarin (IX), 7,4 ''' dymethylamentoflavone (X), methyllonyflavna 3'-O-(XI) and purification of the 28-hydroxy-friedelan-3-one (XIV). The phytochemical study of the Diaporthe hongkongensis, fungi, led to the isolation of (3-oxociclobutil) 2-methyl-1- hydroxi metilciclopenta -2,4- diene carboxylate (XII) and 9-amino-7- (2,5,7-triazabicyclo [2.2.1] hept-3-yl)-2,3,6- trioxi-1-azabicyclo [3.2.2] nona-4,8-diene-4- carbaldehyde (XIII) both isollated of the first time. All compound were identified by 1H, 13C NMR, 2D and mass espectra data. The antioxidant activity, shoed the MeOH extract of leaves from collection 2, was equivalent with 1,15 and 1,13, the H2O phase showed 2,8 and 4.,51 (DPPH• and Fe3+) were considered significant. The antibacterial activity the DCM phase of leaves of collectes 2 and 3 and the EtOAc phase of collection 1, showed CIM and CMB found was 500/mL against Aeromonas hydrophila and Flavobacterium columnare. The aqueous phase of collection 1 showed CIM and CMB found was 1000 μg/ml against Flavobacterium columnare and Pseudomonas fluorescens. The mycelial extracts of fungi 4 and 6, tested with methodology to inhibition halo, showed medium activity, 1 cm, against Aeromonas hydrophila and Klebsiella pneumoniae. The DCM and EtOAc phases of metabolite broth of the fungu 4, showed activity with halo of 1 and 1,2 cm. The antimalical activity. The antimalarial activity showed that hexane and MeOH extracts of leves of the collection 3, showed IC50 of 1,51 and 1,77 mg/mL when compared to the standard. The MeOH extract of fungi 4, showed 6,89 of parasitaemia being significant. The activity by isolated compound sowed CIM of 73,58% to the compound X and 71,75% to the compound XI against Klebsiella pneumoniae . / O presente trabalho teve como objetivo realizar o estudo químico e biológico das folhas de Piranhea trifoliata e de seus fungos endofíticos (Picrodendraceae) e realizar a avaliação do seu potencial frente aos ensaios antimicrobiano, antioxidante e antimalárico. As folhas foram coletadas na Volta Grande do Xingú, Altamira, PA (coleta 1) e no lago do Catalão, Manaus, AM (coletas 2 e 3). A coleta 1 foi extraída com diclorometano, metanol e Água, e as coletas 2 e 3 foram extraídas com hexano, metanol e água. Parte do material das coletas 2 e 3 foi também processada para a obtenção dos fungos endofíticos e estes foram extraídos com diclorometano e acetato de etila. A análise fitoquímica preliminar das folhas e dos fungos mostrou que os extratos diclorometânicos e hexânico possuem terpenos e os extratos metanólicos apresentam substânciastos fenólicas. Os caldos metabólitos dos fungos mostraram que a fase diclorometânica (DCM) possui terpenos, e a fase de acetato de etila (AcOEt) possui indícios de terpenos e substâncias fenólicas. O estudo fitoquímico das folhas levou ao isolamento da Friedela-3-ona, 30-hidroxi-friedelan-3-ona, Lupeol, a mistura dos esteróides β-sitosterol e Estigmasterol, hidroxicoriatina, a mistura de α e β–amirina, 7-hidroxi-6-metoxi-cumarina, 7,4’’’ dimetilamentoflavona, 3’-O- metilloniflavna e a 28-hidroxi-friedelan-3-ona. Já o estudo fitoquímico do fungo Diaporthe hongkongensis, levou ao isolamento de (3-oxociclobutil) metil1-hydroxi-2 metilciclopenta-2,4-dieno carboxilato e 9-amino-7-(2,5,7-triazabiciclo[2.2.1]hept-3-il)-2,3,6-trioxi-1-azabiciclo [3.2.2] nona-4,8-dieno-4-carbaldeido ambas inéditas. Todas as substânciais foram identificadas por RMN de 1H e de 13C, mono e bidimensionais e espectrômetro de massas. Quanto à atividade antioxidante, o extrato MeOH das folhas da coleta 2, apresentou equivalência de 1,15 e 1,13, a fase H2O apresentou 2,8 e 4,51 (DPPH e Fe3+) sendo considerados significativos. A atividade antibacteriana mostrou que a fase DCM das folhas das coletas 2 e 3 e a fase AcOEt da coleta 1, apresentaram CIM e CMB de 500 μg/mL frente às bactérias Aeromonas hydrophila e Flavobacterium columnare. A fase aquosa da coleta 1, apresentou CIM e CMB de 1000 μg/mL frente às bactérias Flavobacterium columnare e Pseudomonas fluorescens. Os extratos miceliais dos fungos 4 e 6, testados pela a metodologia do halo de inibição, apresentaram média atividade, 1 cm frente às bactérias Aeromonas hydrophila e Klebsiella pneumoniae. As fases DCM e AcOEt do caldo metabólito do fungo 4 mostraram-se ativas com 1 e 1,2 cm de halo. A atividade antimalárica mostrou que os extratos hexânicos e MeOH das folhas da coleta 3 apresentaram IC50 de 1,51 e 1,77 μg/mL quando comparado com o padrão. O extrato MeOH do fungo 4 apresentou 6,89 de parasitemias sendo significativo. Ao testar as substâncias isoladas frente as bactérias, observou-se a CIM de 73,58% em X e 71,75% em XI frente a bactéria K. pneumoniae.

Até o momento não há medidas curativas eficientes no controle do HLB, portanto a verificação de maior tolerância de combinações copa/porta-enxerto frente ao HLB poderá contribuir para o avanço no manejo da doença. Neste trabalho avaliou-se o desempenho horticultural de laranjeira ‘Valência’ enxertada em 16 porta-enxertos de citros e a tolerância das combinações ao HLB, com o objetivo de selecionar combinações que tolerem o progresso da enfermidade. O experimento foi instalado em 2011 em Bebedouro-SP, no espaçamento de 6,0 x 2,5 m em regime de sequeiro. O delineamento experimental adotado foi o inteiramente casualizado com 16 tratamentos (porta-enxertos) e 30 repetições. Realizaram-se estudos de análise de sobrevivência feitos através de avaliações de sintomas do HLB. O desempenho horticultural das diferentes combinações copa/porta-enxerto foi analisado por meio de variáveis de crescimento das plantas, produção, eficiência produtiva, qualidade dos frutos e tolerância à seca. Foram avaliadas a intensidade de brotação das plantas para correlacionar com a variável intensidade de plantas com sintomas de HLB. Os resultados mostraram que a produção de frutos e crescimento vegetativo para os porta-enxertos citrange ‘Carrizo’ tetraploide, ‘Cleópatra’ x ‘Christian’ foi equivalente aos porta-enxertos limoeiro ‘Cravo’ e citrumelo ‘Swingle’. O porta-enxerto trifoliata ‘Flying Dragon’ apresentou boa aptidão para o cultivo adensado, por ter proporcionado pouco vigor e maior valor de eficiência produtiva. Os híbridos com parental P. trifoliata conferiram as copas de laranjeira ‘Valência’ produção de frutos de maior qualidade. Os porta-enxertos ‘Sunki’ x ‘Benecke’, tangerineira ‘Sunki e ‘Carrizo’ tetraploide induziram média tolerância à seca inferior à ‘Cravo’ tido como padrão de tolerância. O trifoliata ‘Flying Dragon’ induziu baixa intensidade de brotação, menor incidência da doença e alta proporção acumulativa de sobrevivência. Porta-enxertos menos vigorosos que conferiram menor volume às copas apresentaram menor incidência de HLB, resultando em menor exposição ao vetor da bactéria causadora da doença. A capacidade das plantas enxertadas sobre o trifoliata ‘Flying Dragon’ apresentarem sintomas de HLB tardiamente em relação aos demais porta-enxertos estudados, pode constituir um indicativo de maior capacidade desse material tolerar o progresso da doença. / Brazil is the largest producer of oranges in the world, and the huanglongbing (HLB) is the most important and devastating disease of the world citriculture. It is present in the country, more specifically in the States of São Paulo, Paraná, and Minas Gerais and has been causing severe damage to the citriculture. Until now there are no valid curative measures in HLB control, hence verification of greater tolerance of combinations scion/rootstock front HLB may contribute to advancement in the management of the disease. This work evaluated the horticultural performance of 'Valencia' orange tree grafted on 16 citrus rootstocks and tolerance of combinations to HLB, with the objective of selecting combinations that tolerate the progress of the disease. The experiment was installed in 2011 in Bebedouro-SP, at a spacing of 6.0 x 2.5 m in the rainy season. It has a completely randomized design with 16 treatments (rootstock) and 30 replications. Survival analysis studies were performed using HLB symptom assessments The horticultural performance of the different combinations scion/rootstocks was analyzed using variables of plant growth, production, productive efficiency, fruit quality and drought tolerance. The intensity of sprouting of the plants was evaluated to correlate with the variable intensity of plants with HLB symptoms. The results showed that the superiority found for fruit production and vegetative growth for 'Carrizo' tetraploid, ‘Cleopatra’ x ‘Christian’ rootstocks was equivalent to 'Cravo' and 'Swingle' rootstocks. The rootstock trifoliate ‘Flying Dragon’ showed a good aptitude for dense cultivation, introducing low vigor and higher value production efficiency plants. Hybrids with parental P. trifoliata conferred to 'Valencia' sweet orange scion production of higher quality fruits. The ‘Sunki’ x ‘Benecke’, ‘Sunki’ and 'Carrizo tretraploid' rootstocks induced medium tolerance to drought, getting behind only 'Cravo' considered as a tolerance standard. ‘Flying Dragon’ trifoliate obtained low sprouting intensity, lower incidence of the disease and high cumulative proportion of survival. Less vigorous rootstocks that confer lower crown volume had a lower incidence of HLB, resulting in less exposure to the vector of the disease-causing bacteria. The ability of plants grafted on ‘Flying Dragon’ trifoliata to present late HLB symptoms late compared to the other rootstocks studied, may be an indication of the more significant capacity of this material to tolerate the disease progression. / FAPESP: 2016/17480-3
