This diploma thesis deals with lithium-ion batteries. It is focused on negative electrode on grafit based. The goal of this thesis is to show the problematics of lithium-ion batteries together with possibilities for improvement of their basic parametres as capacity and current elektricity loadability. The first part is focused on the description of functionality of lithiumion battery. The second part is practical and it is focused on production of negative electrodes from natural graphite 280H which has different thickness and compression pressure. The third part describes preparation of negative electrodes from natural graphite 280H and also includes results of measurement. In the last part are different types of negative electrodes from natural graphite 280H compared and evaluated including the determination of conclusions. This comparison and evaluation based on obtained data.

This thesis consists of one chapter accepted for publication in a book and four papers published in international journals with impact factors. All of the contributions deal with the role of wood ants in energy and nutrient fluxes in forest ecosystems. Wood ant nests are known as hot spots of carbon dioxide (CO2) production and are also thought to affect methane (CH4) flux. Stable high temperatures are maintained in ant nests even in cold environments. This study is focused on quantification of CO2 and CH4 flux in wood ant nests, contribution of ants and microbes to CO2 production, properties of nest material that affect CO2 production and the role of ants and microbes in the maintenance of nest temperature. The research was conducted in temperate and boreal forests inhabited by wood ants (Formica s. str.). Gas fluxes were measured either by an infrared gas analyser or a static chamber technique. Ants and nest materials were also incubated in a laboratory. Material properties potentially influencing CO2 flux, such as moisture, nutrient content or temperature were determined. According to the results, CH4 oxidation was lower in wood ant nests than in the surrounding forest soil suggesting that some characteristics of ant nests hinder CH4 oxidation or promote CH4 production. With growing customer demands for product quality and product characteristics, increasing technology speed and increasing product purity, there is growing demand for analyzes of chemical composition. These demands are very often against each other, especially the increasing demands on the speed and accuracy of analyzes. Analytical requirements may not only apply to the test technique, but also to the quality of the sample, which is inseparable from it. What is recently seen as the biggest drawback, the new technology is trying to use as its main priority. This is undoubtedly true of atomic emission spectrometry.

Selektivní růst gallium-nitridových tenkých vrstev na substráty pokryté maskou z pyrolyzovaného rezistu / Selective gallium nitride thin-film growth on substrates covered by pyrolyzed resist mask

This thesis deals with deposition of GaN thin films and GaN selective growth utilizing pyrolyzed resist masks. Carbon masks were prepared on silicon substrates by electron-beam litography and resist pyrolysis. As a further step, Ga and GaN were deposited on the masked substrates by Moleculer Beam Epitaxy (MBE) method. A selective growth of Ga droplets was achieved. These results were used for preparation of GaN crystallites by pulse deposition. It is also shown that direct MBE deposition of GaN on the masked substrates leads to a selective growth of GaN thin films with GaN film growing only on the areas which are not covered by the carbon mask. The results are explained by enhanced surface diffusion of gallium atoms on the surface of the carbon mask.

Vývoj a použití elektrochemického průtokového detektoru s obnovitelnou náplní / Development and application of electrochemical flow-through detector with renewable working material

The aim of this thesis was development and initial testing of new porous electrochemical detector for flow-through arrangements, especially for flow injection analysis (FIA) and liquid chromatography. One of the most advantageous properties of the detector is simple renewal of working material and thus its suitability for determination of strongly passivating substances. Glassy carbon microbeads and copper microparticles were tested as a working material within this study. Initially, basic electrochemical properties of the glassy carbon-based detector were examined by FIA using hydroquinone and potassium ferrocyanide as model substances. For both model substances high degree of conversion was achieved (around 100 %), and thus it was concluded that glassy carbon-based detector can be considered as coulometric. Hereafter, practical application of the carbon-based detector are presented on five electrochemically active substances closely related to the human medicine and pharmacy - thymol, tyrosine, sulfamethizole and vanillylmandelic acid (VMA) simultaneously determinined with homovanillic acid (HVA). Dynamics of carbon and phosphorus flows in arbuscular mycorrhizal symbiosis Mgr. Tereza Konvalinková (doctoral thesis) Abstract Arbuscular mycorrhizal fungi (AMF) are widespread and highly specialized root symbionts, which gain all of their carbon (C) from the hosts, supplying plants with mineral nutrients (particularly with phosphorus, P) in return. This thesis focuses on the size and flexibility of C and P flows in arbuscular mycorrhiza in relation to environmental conditions, in particular to light and P availability. The indications that the symbiotic flows are regulated actively by both partners are discussed. The main findings are presented as a compilation of separate scientific works (two research articles, one review and one book section). A glasshouse experiment has shown that both mycorrhizal benefits and mycorrhizal colonization of medic (Medicago truncatula) by an AMF species (R. irregularis) decline along the gradient of decreasing light intensity. Interestingly, morphological adaptation of medic to the long-term light deprivation was boosted by mycorrhiza, probably because of C demand of AMF and due to the improved nutrition of the mycorrhizal plants. This study investigated characteristics of algal organic matter (AOM) derived from four species (cyanobacteria Microcystis aeruginosa and Merismopedia tenuissima, diatom Fragilaria crotonensis and green alga Chlamydomonas geitleri) dominating phytoplanktonic populations in the reservoirs supplying drinking water treatment plants - the water reservoirs Švihov (the Želivka water treatment plant) and Josefův Důl (the Jizerské Mountains), Czech Republic. The four mentioned microorganisms were cultivated under laboratory conditions and the algal growth was quantified by a number of cells, chrorophyll-a and optical density. Production of AOM was evaluated by dissolved organic carbon concentration measurements, by specific UV absorbance, the amount of peptide/protein and non-peptide (carbohydrate) contents, water affinity and molecular weight (MW). AOM of all considered microorganisms are formed predominantly by hydrophilic substances with low values of specific UV absorbance (< 2 L/(m.mg)). Considerable portions of both types of AOM, i.e. extracellular and cellular organic matters are represented by substances with MW < 1 kDa or with MW > 100 kDa.
