Ketolátky a volné mastné kyseliny v mléce vysokoužitkových krav

The aim of this diploma thesis was to evaluate the occurrence of ketones and free fatty acids in milk of high-yielding cows. Ketosis is a very common metabolic disorder in dairy cows resulting in reduced milk production, fertility disturbance and more frequent occurrences of other diseases. Ketosis is not only a serious health problem, but also an economic problem, that can be avoided by balancing the feeding dose respecting the physiological needs of energy delivery after birth and at the beginning of lactation of high-yielding cows. The monitoring was carried out in 2016 at Chyšná, which is one of the centers of Agribusiness Košetice a.s. Owerall were 1407 cows monitored during the 6 utility tests (from January to June 2016). The occurrence of subclinical ketoses was defined by the content of ketones in milk (acetone and beta-hydroxybutyric acid). Critical was the period of the first 12 days of lactation, when the occurence of subclinical ketoses was the highest, the occurence of this disease was lower than in other breeds. At Chyšná breeding, the low incidence of subclinical ketoses is mainly influenced by zootechnical work and by working with the feed consultant and the company for the production of compound feeds. As a precautionary process, regular feed analysis and examination of free fatty acids and ketones in milk can be recommended.

This thesis provides an overview of extra-contractual legal institutions of the use of copyrighted work. The main emphasis is on the institution of free use and related topic of private copying and fair compensation. Statutory licenses, both chargeable and free of charge, are not omitted. The thesis is divided into three chapters. Brief introduction is followed by chapter providing definition of the fundamental terms of copyright from the perspective of Czech copyright law. The second chapter contains summary of sources of copyright law. Czech law, as well as European and international law is included. The third chapter contains crutial part of the thesis, the analysis of the extra-contractual legal institutions of the use of copyrighted work. After brief comment on free works follows analysis of the Three-Step Test, which is of great significance for all examined legal institutions. Relevant case law - WTO Panel report WT/DS160/R - is examined and the Max Planck Institute's Declaration on a balanced Interpretation of the Three-Step Test is also considered. Subsequent subchapters are dedicated to the institutions of free use and private copying under both, Czech and European copyright law. - ENGLISH The content of this thesis is aimed towards the interpretation of the so-called non- contractual legal institutions of the use of copyrighted work. These institutions represent such legal tools, that exceptionally allow individuals different from the author to exploit copyrighted work with no dependence on the author's permission or will. The text of the work is divided into four main chapters. For introduction into the matter, the opening part is dedicated to the explanation of the term copyright law, its position in the Czech legal system and the main principles it lies upon. This broad explanation is subsequently followed by definition of its fundamental terms - author's work and its use. The crucial part of the work lies in the fourth chapter, which bears the same title as the work itself. The structure of this chapter follows the organisation of the Czech Copyright Act. After description of the free work, the paper gets to the interpretation of individual exemptions and limitations to copyright, thus the institutes of free use and statutory licences. Lipidy v půdě vykazují poměrně vysokou resistenci k biodegradaci, mohou proto tak zajistit informace o zdrojích organické hmoty a diagenetických procesech. Půdní organická hmota je vysoce heterogenní a vyskytují se v ní různé dynamické systémy. Vztahy mezi těmito systémy a molekulární strukturou ještě nejsou úplně známy. Analýza lipidů a bitumenu byla provedena u dvou vzroků (rašelina a lignit). Množství skupin sloučenin (volné uhlovodíky a volné ketony, vázané alkoholy, vázané mastné kyseliny, vázané -hydroxy kyseliny, volné dikyseliny a polycyklické sloučeniny) bylo idetifikováno pro oba vzorky a jejich distribuce byly určeny. GC/MS analýza volných a vázaných lipidů zjistila jejich různé zdroje během chemické degradace. Některé podobné znaky mohou být znakem uchovávání části vosků a suberinu z vyšších rostlin (dlouhé řetězce vázaných mastných kyselin). Velké rozdíly v molekulárním složení lipidů byly pozorovány mezi vzorky, dokládající význam studia lipidů z mladého sedimentu (rašelina) a starého (lignit).

Background: Oxidative damage is one of the most frequent types of cell components damage leading to oxidation of lipids, proteins and the molecule of DNA. As a consequence, there is a higher occurrence of several pathologies such as atherosclerosis, neurodegenerative diseases, cancer; or diabetes. In our study, influence of whole body anesthesia during minor surgery on the level of DNA damage was examined using comet assay technique. Methods: The basic principle of this method is fixing the cells (lymphocytes) in agarose, their lysis for the removal of membranes, incubation with the specific enzymes and electrophoresis of the released cell nuclei. During the electrophoresis, free low-molecular weight and negatively charged fragments of DNA move towards anode which causes the formation of the typical comet cell shape. Finally, the gels are stained by ethidium bromide (DNA intercalating dye) and visualized. Results: We have observed single strand breakages (SSBs) and, with the use of modified assay using specific enzymes for detection of specific lesions, also oxidized purines and pyrimidines. The extent of DNA damage as determined by the intensity of the tail of the comet was quantified using LUCIA Comet Assay (Laboratory Imaging, Czech Republic) software for image analysis. This graduation theses deals with the cultivation, processing, preparation of tea and the chemical composition of tea leaves in the theoretical part. Furthermore deals with the synthesis of biologically active substances in plant materials and their conversion from the preparation of tea to the transformation in the human organism after its consumption. It also briefly summarizes and defines the issues of free radicals and antioxidants as part of their functioning in the human body, and then attempts to objectively summarize their main pharmacological effects. The practical part, focused on the determination of phenolic substances and the total antioxidant capacity of selected green and black teas, is supplemented by their sensory analysis.

This thesis examined the predictive power of different strategies for future stock returns. The analysis was conducted using a data sample of 3976 firms traded on the New York Stock Exchange (NYSE) and NASDAQ during a 29 year time horizon, from July 1986 to June 2015. Predictive powers of different strategies were also tested during three sub-periods and during bull and bear markets using both long-only and long/short portfolios to check whether the predictive power is robust. It was found that the FCF yield is a better indicator of future stock returns than the gross profitability. The difference between average monthly returns was significant during all tested time periods and market situations using both long-only and long/short portfolios. The newly introduced FCF profitable value strategy proved to be a better predictor of future stock returns than the profitable value strategy. The FCF profitable value strategy presents also an improvement over the FCF yield strategy. It was found that the FCF profitable value has a better predictive power for future stock returns than the FCF yield at least during some time periods or market situations.
