Antioxidační látky v potravinách a jejich funkce při obraně organismu před účinky volných radikálů / Antioxidants in Food and Their Function in the Process of Defence Against the Free Radicals Effects

The thesis deals with antioxidants in food and their function in the process of defence against the effects of free radicals. At first, antioxidants are defined and their clasification is described in the theoretical part. Then this part deals with antioxidants in food, their effects on people´s health and antioxidants recommended daily doses. Possitive and negative effects of free radicals on people´s health and their relation to antioxidants are mentioned next. In the practical part, I concentrate on the survey focused on the investigation of the population awareness of antioxidants and their effects on health. One hundred and sixty respondents took part in the survey. They were devided into two groups; working people and pensioners. The questionnaire contained twelve questions which investigated the population awareness of the topic. The data collection took place in January 2013 and it was evaluated in February 2013. The collected data were evaluated according to basic statistical methods. The ascertained answers were put into unified tables and then they were compared. Especially, the general awareness was investigated and then the effect of education on the knoledge of the topic was considered.

Metody zjišťování látek antioxidačního charakteru / The Methods for the Detection of Antioxidant Substances

Antioxidants and free radicals are widely discussed in the clinical and nutritional literature. Various reactive oxygen species (ROS) may be produced from normal biochemical, essential metabolic processes or from external sources as exposure to a variety of agents presented in the environment. Lipids, proteins, carbohydrates and DNA are all capable of reacting with ROS and can be implicated in etiology of various human disorders. The antioxidant system protects the organism from the effects of free radicals by scavenging them and thus preventing structural damage of cells. Measuring the individual components of the antioxidant system has provided important information about the defences against free radical effect. During the last decade, many analytical methods have been developed to determine the antioxidant activity of natural compounds and their mixtures in vitro. Some procedures involve the methods based on the generation of various radical species and their elimination by treatment with potential antioxidant compounds. The tests used for the evalution of total antioxidant activity are: Trolox equivalent antioxidant capacity-TEAC, diphenylpicrylhydrazil - DPPH, oxygen radical absorbance capacity-ORAC and the methods testing the ability to prevent lipid peroxidation. The other group includes the procedures that are based on measurement of redox properties of compouds (ferric reducing antioxidant potential assay-FRAP, cyclic voltammetry, HPLC).

Free radicals are highly reactive species with one unpaired electron in orbital. Reactive oxygen and nitrogen species count among important biologic molecules of radical nature. It is very important to fix a concentration of free radicals in cell on non-toxic limits. Products of radical damage are cumulated extra or intracellulary and they are main components of lipofuscin-like pigments. Lipofuscin-like pigments contains in their molecular structure fluorofores, so they are good substrates for fluorescent analysis. Alzheimer's disease is a very actual social and economical problem. Etiology of Alzheimer's disease is still unknown. Histologically, the characteristic presence of Alzheimer's disease is a senil plaques of amyloide ?. ROS and RNS diffuse through hematoencephalic barrier in vessel's lumen and attacks red blood cells. Radical damage of erythrocytes is associated with an increase of concentration of oxidative stress products in cytosol. Sample for fluorescent analysis has been prepared from a red-blood cell extract from 30 patients and 8 healthy controls. Fluorescent spectra of healthy controls have emission maxima in area 327-343nm. In compare with controls, spectra of pacients were more heterogenous in area upon 380nm. d-Framy jako algebraické duály bitopologických prostorů / d-Frames as algebraic duals of bitopological spaces

Achim Jung and Drew Moshier developed a Stone-type duality theory for bitopological spaces, amongst others, as a practical tool for solving a particular problem in the theory of stably compact spaces. By doing so they discovered that the duality of bitopological spaces and their algebraic counterparts, called d-frames, covers several of the known dualities. In this thesis we aim to take Jung's and Moshier's work as a starting point and fill in some of the missing aspects of the theory. In particular, we investigate basic categorical properties of d-frames, we give a Vietoris construction for d-frames which generalises the corresponding known Vietoris constructions for other categories, and we investigate the connection between bispaces and a paraconsistent logic and then develop a suitable (geometric) logic for d-frames.

The thesis is divided into two parts: theoretical and practical. The theoretical part summarizes elemental knowledge of writing skills, its classification and evaluation. Special attention is given to the concept of free writing. The practical part compares theoretical knowledge with realized research. The research analyses the attitude of the target group to free writing and its correction.

The diploma thesis aims to prepare a study of the cooling system of a multifunctional building with regard to climate change predicted until the middle of the 21st century. The work includes determining the heat load of refrigerated rooms and designing measures to reduce heat gains in the summer. Diploma thesis includes design of a cooling system with fan-coil units as end appliances with the necessary hydraulic calculations. A single-stage absorption cooling unit is used as the considered cooling source, to which a free cooling heat exchanger is assigned. The design of the cooling source includes the necessary hydraulic calculations and dimensioning of the system ensuring its functional operation. Part of the diploma thesis is drawing documentation of cooling technology and connection of fan-coil units.

Antioxidační aktivita vybraných druhů drobného ovoce / The antioxidant activity of selected species of small fruit

This diploma´s thesis deals with the determination of total antioxidant activity of red and white gooseberries and red, white and black currants. There are available information about small fruits, an overview of natural and synthetic antioxidants and a summary of the methods to determine the total antioxidant activity in the theoretical part. Frozen fruit samples were provided by Research and Breeding Institute of Pomology Holovousy Ltd. in the project QI111A141 – Research of new technologies in the cultivation of gooseberries and currants with a focus on quality and use of fetuses (Ministry of Agriculture). The total antioxidant activity of red and white gooseberries and currants was measured by spectrophotometric methods using free radicals ABTS•+ and DPPH and was expressed as a Trolox equivalent. The total antioxidant activity of black currants was measured by electron paramagnetic resonance using free radicals ABTS•+ and DPPH and expressed as a TEACABTS and a TEACDPPH. There were measured 8 species of red gooseberries, 6 species of white gooseberries, 12 species of red currants, 6 species of white currants and 8 species of black currants in this thesis. The highest values of total antioxidant activity showed species of red and white gooseberries Alan, Karmen, Pax and Invicta and species of red and white currants Rovada, Orion and Olin. Among black currants, species Ometa and Ruben had the highest values of total antioxidant activity.

RNA secondary structure is very important in many biological processes. Efficient structure prediction can give information for experimental investigations of these processes. Many available programs for secondary structure prediction exist. Some of them use single sequence, the others use more related sequences. Pseudoknots are still problematic for most methods. This work presents several methods and publicly available software and the implementation of minimum free energy method is described.

Diploma thesis is concerned with problems of parking places in VUT FSI area, Technická 2, Brno. In introduction there are explained methods of parking, sorting and orientation proportions of vehicles. Then there are explained basic requirements for parking areas according to valid norms and rules. In this task there is analyzed original and actual status of parking. Before separating suggestions is finished calculation of required number of parking places. Solution of whole problems around parking places is in three projects. Results are compared in conclusion.
