Influence of Methionine on Growth and Nitrogen Balance in Weanling Quarter Horses

Twenty-four Quarter horse weanlings (120 d +/- 10 d) were blocked by age into 4 groups (n = 6) for a 56 d trial to evaluate the influence of methionine on growth and nitrogen retention. Weanlings were housed by block and individually fed 1 of 4 concentrate diets twice daily at 1.5% BW (as fed). Weanlings were randomly assigned to 1 of 4 treatments: basal (0.20 MET), basal + 0.03% methionine (0.23 MET), basal + 0.07% methionine (0.27 MET), and basal + 0.11% methionine (0.31 MET). Diets were formulated to be isonitrogenous, isocaloric, and contain equal amounts of LYS and THR. Coastal bermudagrass hay was individually fed at 0.75% BW (as fed). Growth measurements, body weight, rump fat, and plasma were obtained every 7 d. The final 4 days consisted of total collection of urine and feces. Feed, fecal, and urine samples were analyzed for nitrogen content and nitrogen balance was calculated. Urine was analyzed for urea and ammonia concentration. Plasma was analyzed for urea concentration. Grain, hay, and fecal samples were analyzed for nutrient composition. Data were analyzed using the PROC MIX procedure of SAS. Linear, quadratic, and cubic effects were tested in the form of contrasts. There was no influence of treatment on growth measurements, nitrogen balance, or urinary urea or ammonia. Intake of LYS and THR (g/d) did not differ among treatments (P = 0.78 and P = 0.38 respectively). Plasma urea nitrogen (PUN) was influenced by treatment (P = 0.005) exhibiting quadratic (P = 0.04) and cubic (P = 0.002) effects. An unexpected peak in PUN was observed with 0.27 MET. Upon analysis, 0.20 MET contained more lysine than formulated, and 0.27 MET contained the least lysine. Treatments 0.20 MET and 0.31 MET contained more threonine compared to formulations. These differences may explain unexpected values of PUN concentration. Results suggest future studies that more closely isolate methionine as the only dietary variable are necessary to better explain the methionine requirements of weanling horses.

Foram realizados dois experimentos para determinar a composição química, os coeficientes de digestibilidade de nutrientes e os valores de energia e proteína digestíveis do ovo em pó (OP) e avaliar o desempenho e os componentes sanguíneos e plasmáticos de leitões recém-desmamados alimentados com dietas contendo níveis crescentes de proteína do OP em substituição à proteína do plasma sanguíneo. O Experimento I consistiu de um ensaio de digestibilidade, sendo utilizados oito leitões (quatro machos castrados e quatro fêmeas), mestiços Landrace x Large White com 15,9 kg de peso médio. Os tratamentos consistiram de uma dieta basal e uma dieta teste (70% dieta basal + 30% OP) com quatro repetições cada. Utilizou-se a metodologia da coleta parcial de fezes com o uso de 0,5% de óxido crômico (Cr2O3) adicionado às dietas como indicador. Os coeficientes de digestibilidade aparente do OP foram de 87,18, 80,76, 81,99 e 70, 54%, respectivamente, para a matéria seca, energia bruta, proteína bruta e extrato etéreo. A partir dos coeficientes de digestibilidade determinados e os valores de proteína bruta (56,53%) e energia bruta (5.897 kcal/kg) foram calculados os valores de 43,91% de proteína digestível e 5.139 kcal/kg de energia digestível do OP para leitões em fase de creche. No Experimento II, foram utilizados 90 animais (machos castrados e fêmeas), mestiços Landrace x Large White, desmamados com 24 dias e 5,6 kg de peso médio. O período experimental compreendeu duas fases, a pré-inicial (1 a 14 dias) e inicial (15 a 28 dias pós-desmame), onde os tratamentos consistiram de cinco rações isonutritivas com níveis crescentes (0, 25, 50, 75 e 100%) de substituição protéica do plasma sanguíneo da dieta pelo OP. O delineamento experimental foi o de blocos casualizados, com nove repetições por tratamento e dois animais por unidade experimental (baia). Os animais e a ração foram pesados para o cálculo do ganho diário de peso (GDP), consumo diário de ração (CDR) e conversão alimentar (CA) em cada fase e período total. Ao final do ensaio de desempenho foi retirada uma amostra de sangue de cada animal para a determinação dos componentes plasmáticos (uréia, proteína total, albumina, globulina, relação albumina/globulina, triglicérides e colesterol) e sanguíneos (hematócrito e hemoglobina). Não houve diferença entre os tratamentos (P > 0,05) para as variáveis de desempenho na fase de 1 a 14 dias pós-desmame. Para a segunda fase experimental e para o período total, foi observada uma redução linear (P < 0,01) do GDP e CDR, à medida que aumentava o nível de substituição do PS pelo OP. Para as variáveis dos componentes plasmáticos e sanguíneos não foram observadas diferenças significativas (P > 0,05) entre os tratamentos. Concluiu-se que o ovo em pó pode substituir completamente o plasma sanguíneo sem afetar o desempenho dos animais na fase de 1 a 14 dias pós-desmame, enquanto que na fase de 15 a 28 dias essa substituição prejudicou o desempenho dos animais, refletindo assim, em um pior desempenho no período total (1 a 28 dias). The apparent digestibility coefficients of dry matter, gross energy, crude protein and crude fat were, respectivily, 87.18, 80.76, 81.99 and 70.54%. From these results of apparent digestibility coeficients and the values of crude protein (56.53%) and gross energy (5,897 kcal/kg) resulted in 43.91% of digestible protein and 5,139 kcal/kg of digestible energy for OP. In Experiment II, ninety Landrace x Large White crossbred pigs (barrows and gilts) were weaned at 24 days of age with 5,6 kg live weight. The experimental period was divided in two phases, 1 to 14 days and 15 to 28 days post-weaning. The treatments consisted of five dietary levels of 0, 25, 50, 75 and 100 % of substitution of plasma protein for OP protein. Pigs were alloted in a randomized block design with nine replications per treatment and two pigs per experimental unit (pen). Pigs were weighed and the data of feed intake per pen were registered every week. Pig performance data such as average daily gain (GDP), daily feed intake (CDR) and feed conversion (CA) were analyzed by polynomial regression. No treatment effects (P > 0.05) were observed on performance for 1-14 day post-weaning phase. For the 15-28 phase and for the total period, linear reductions (P < 0.01) of GDP and CDR, were observed with increased dietary OP levels. No treatment effects (P > 0.05) were observed on plasma and blood components. Therefore, it can be conclued that the spray-dried egg can completely replace plasma without affecting the performance of pigs during 1-14 day post-weaning phase. For the 15-28 day phase and total period, added OP levels showed a depressive effect on pig performance. The plasma and blood components were not affected by the inclusion of spray-dried egg.

Acidificantes como alternativas aos antimicrobianos melhoradores do desempenho de leitões na fase de creche / Acidifiers as alternatives to antimicrobial growth promoter of weanling pigs

O objetivo deste trabalho foi avaliar misturas (blends) de acidificantes e seus sais como alternativas aos antimicrobianos melhoradores do desempenho de leitões na fase de creche. Foi realizado um experimento em blocos casualizados, com 34 dias de duração para testar cinco tratamentos. Para o período de 1 a 14 dias, os tratamentos foram: Am (antimicrobiano) - dieta pré-inicial com 0,004% de sulfato de colistina (40 ppm); A1 (acidificante 1) - pré-inicial com 0,5% do blend 1 (contendo ácido fórmico, 145.000 ppm; ácido fosfórico, 85.000 ppm); A2 (acidificante 2) - pré-inicial com 0,15% do blend 2 (butirato de sódio, 64.000 ppm) e 0,4% do blend 3 (ácido láctico, 620.000 ppm; ácido fórmico, 40.000 ppm); A3 (acidificante 3) - pré-inicial com 0,8% do blend 4 (ácido propiônico, 198.000 ppm; ácido acético, 196.000 ppm; ácido fórmico, 196.000 ppm; ácido fosfórico, 21.000 ppm; ácido cítrico, 8.500 ppm); A4 (acidificante 4) - dieta basal com 0,6% do blend 4 e 0,15% do blend 5 (ácido benzóico, 590.000 ppm; ácido fórmico, 70.000 ppm; ácido fosfórico, 50.000 ppm; ácido cítrico, 40.000 ppm). Para o período de 14 a 34 dias, os tratamentos foram: Am - dieta inicial com 0,004% de sulfato de colistina; A1 - inicial com 0,3% do blend 1; A2 - inicial com 0,1% do blend 2 e 0,3% do blend 3; A3 - inicial com 0,6% do blend 4; A4 - inicial com 0,5% do blend 4 e 0,1% do blend 5. Foram utilizados 160 leitões Topigs recém-desmamados, com idade média em torno de 24 dias e peso inicial de 6,69±1,82 kg. Foram alocados quatro leitões (dois machos castrados e duas fêmeas) em cada baia (unidade experimental). Para os dados de desempenho e freqüência de diarréia, foram utilizadas 8 repetições (blocos) por tratamento. Ao final do experimento, um animal de cada baia, dos 4 primeiros blocos, foi abatido para análise da morfologia intestinal, pH estomacal e cecal e morfometria de órgãos. Durante a fase pré-inicial, o tratamento A2 (combinação dos blends 2 e 3) proporcionou melhor peso aos 14 dias (P14) (P=0,03) e ganho diário de peso (GDP) (P=0,04) que o tratamento A3 (blend 4), e melhor conversão alimentar (CA) (P=0,004) que o tratamento Am (antimicrobiano). Para o período total, o tratamento A4 (blends 4 e 5) apresentou melhor CA (P=0,0006) que o tratamento Am (antimicrobiano). Os tratamentos não afetaram (P>0,05) a freqüência de diarréia e o pH estomacal. O tratamento A4 (blends 4 e 5) apresentou menor valor de pH cecal (P=0,015) que o tratamento Am e menor valor de peso relativo do pâncreas (P=0,013), comparado aos tratamentos Am e A2. Para morfologia intestinal, o tratamento A2 proporcionou menores valores de profundidade de cripta (PC) (P=0,03) do jejuno que os tratamentos A3 e Am e maior relação altura de vilosidade:profundidade de cripta (P=0,04) do jejuno que os tratamentos A1 e A3. A análise econômica, no período de 1 a 14 dias, mostrou que os acidificantes propiciaram custos por kg de GDP, no mínimo, 4,09% inferiores ao antimicrobiano (Am). No período de 1 a 34 dias, os tratamentos A1 e A4 apresentaram custos por kg de GDP até 1,08% menores que o Am. Assim, o presente estudo mostrou que misturas de ácidos orgânicos, inorgânicos e seus sais podem ser uma alternativa viável aos antimicrobianos melhoradores do desempenho de leitões na fase de creche. / The purpose of this work was to evaluate several acidifier blends as alternatives to antimicrobial growth promoters of weanling pigs. A 34-d randomized complete block design experiment was carried out to compare five treatments. For 1-14 d experimental period, the treatments were: Am (antimicrobial) - pre-starter diet with 0.004% of colistin sulfate (40 ppm); A1 (acidifier 1) - pre-starter diet with 0.5% of blend 1 (containing formic acid, 145,000 ppm; phosphoric acid, 85,000 ppm); A2 (acidifier 2) - pre-starter diet with 0.15% of blend 2 (butyric acid, 64,000 ppm) and 0.4% of blend 3 (lactic acid, 620,000 ppm; formic acid, 40,000 ppm); A3 (acidifier 3) - pre-starter diet with 0.8% of blend 4 (propionic acid, 198,000 ppm; acetic acid, 196,000 ppm; formic acid, 196,000 ppm; phosphoric acid, 21,000 ppm; citric acid, 8,500 ppm); and A4 (acidifier 4) - pre-starter diet with 0.6% of blend 4 and 0.15% of blend 5 (benzoic acid, 590,000 ppm; formic acid, 70,000 ppm; phosphoric acid, 50,000 ppm; citric acid, 40,000 ppm). For 14-34 d experimental period, the treatments were: Am - starter diet with 0.004% of colistin sulfate; A1 - starter diet with 0.3% of blend 1; A2 - starter diet with 0.1% of blend 2 and 0.3% of blend 3; A3 - starter diet with 0.6% of blend 4; and A4 - starter diet with 0.5% of blend 4 and 0.1% of blend 5. One hundred and sixty Topigs 24-d-weaned pigs, with 6.69±1.82 kg live weight were allotted to 20 suspended pens, with four pigs (two castrated male and two female) per pen (experimental unit). For performance and diarrhea incidence data, 8 replications per treatment were used. On 34th day of experimental period, an animal of each pen of first 4 blocks was slaughtered for small intestine morphology, stomach and caecum pH and organs morphometry. For 1-14 d experimental period, treatment A2 (combination of blends 2 and 3) gave better body weight at 14th day (BW14) (P=.03) and average daily gain (ADG) (P=.04) than treatment A3 (blend 4), and better feed conversion (FC) (P=.004) than treatment Am (antimicrobial). For total experimental period (1-34 d), treatment A4 (blends 4 and 5) gave better FC (P=.0006) than treatment Am (antimicrobial). Treatments did not affect (P>.05) diarrhea frequency and stomach pH. Treatment A4 (blends 4 and 5) gave lower pH value (P=.015) than treatment Am and smaller relative pancreas weight (P=.013) than treatments Am and A2. For intestinal morphology, treatment A2 provided smaller (P=.003) jejunum crypt depth (CD) than treatments A3 and Am, and bigger (P=.04) ratio of jejunum villus height:crypt depth than treatments A1 and A3. Economical analysis showed, for 1-14 d experimental period, that the acidifiers provided cost per kg of ADG at least 4.09% lower than antimicrobial (Am). For 1-34 d experimental period, treatments A1 and A4 showed cost per kg of ADG until 1.08% lower than Am. Therefore, this study showed that blends of organic and inorganic acids and their salts can be viable alternatives to antimicrobial growth promoters of weanling pigs.

Prebiotics and Beta-Glucan in Modulation of Growth Performance, Nutrient Utilization and Alkaline Phosphatase Kinetics in the Weanling Pig

This thesis examined effects of dietary supplementations (0.75%) of the prebiotics of retrograded resistant cornstarch, fibersol-2 and inulin, and oat β-glucan in replacing feed antibiotics on growth performance, plasma urea concentrations, total tract dry matter (DM) and lactose digestibility, fecal scores, proximal jejunal and serum alkaline phosphatase (AP) kinetics and large intestinal fermentation in weanling pigs fed corn and soybean meal-based diets. There were no differences (P > 0.05) in the growth performance, plasma urea concentrations, DM and lactose digestibility and the volatile short-chain fatty acid concentrations in the cecal and fecal samples among the treatment diets. Dietary lactose was completely digested in the weanling pigs. Supplementations of fibersol-2, inulin and β-glucan significantly affected some of the jejunal and serum AP kinetics. In conclusion, lactose was a highly digestible carbohydrate and dietary supplementations of the three prebiotic and β-glucan at 0.75% had little effects on growth performance and plasma urea concentration but might affect gut and the whole body health status via influencing the AP detoxification kinetics in the weanling pigs. / Ontario Pork, Ontario Ministry of Agriculture, Agriculture Adaptation Council

THE EFFECTS OF ACTIGEN® AND THREONINE SUPPLEMENTATION ON GROWTH PARAMETERS, IMMUNE FUNCTION, AND INTESTINAL HEALTH IN MONOGASTRICS

The objectives of these experiments were to evaluate the main effects and interactive effects between dietary supplementation of a mannan oligosaccharide based product, Actigen® (ACT), and different levels of threonine in monogastrics, as evidenced by growth parameters, immune function, and intestinal health characteristics. In nursery piglets, ACT supplementation decreased average daily feed intake (P=0.04), but had no effect on body weight or feed conversion ratio. There were no noted differences between threonine levels on performance traits. There were no consistent differences in complete blood count or cytokine gene expression profiles in the blood. The highest level of threonine, 77% true ileal digestible threonine:lysine (Thr:Lys), increased villus height (P=0.007) and villus height:crypt depth (P=0.01). The lowest level, 57% Thr:Lys, decreased villus surface area (P=0.04) and goblet cell density (P=0.04). Supplementation with ACT increased total goblet cell area (P=0.02) and density (P=0.05). There were no interactions observed between ACT and Thr:Lys levels. In broiler chicks, feeding a diet containing 0.56% threonine decreased body weight (PPPP=0.07). On d 7, birds supplemented with ACT had heavier spleens as a percentage of body weight (P=0.01) compared to no ACT. When adjusted for body weight, the 0.56% threonine fed birds had smaller spleens (P=0.05) on d 7 when compared to the other threonine levels. Humerus (PPP=0.04) and birds fed 0.56% threonine had a higher concentration of potassium (PP=0.03) and few goblet cells (P=0.04) on d 7 when compared to the other threonine levels. In d 21 jejunum, supplementation with ACT reduced apical width (P=0.03) and surface area (P=0.02). An interaction was observed between ACT and threonine level in the jejunum on d 21 on basal width (P=0.03) and surface area (P=0.02), indicating that in diets lacking ACT, excess threonine increased villus size. Overall, ACT and threonine acted primarily independently to modulate the intestinal architecture of both nursery piglets and broiler chicks. However, in broiler chicks ACT and threonine interacted to alter villus size. These results indicate that ACT and threonine have direct effects on the intestines of monogastrics.

Para o período de 1 a 14 dias, os tratamentos foram: Am (antimicrobiano) - dieta pré-inicial com 0,004% de sulfato de colistina (40 ppm); A1 (acidificante 1) - pré-inicial com 0,5% do blend 1 (contendo ácido fórmico, 145.000 ppm; ácido fosfórico, 85.000 ppm); A2 (acidificante 2) - pré-inicial com 0,15% do blend 2 (butirato de sódio, 64.000 ppm) e 0,4% do blend 3 (ácido láctico, 620.000 ppm; ácido fórmico, 40.000 ppm); A3 (acidificante 3) - pré-inicial com 0,8% do blend 4 (ácido propiônico, 198.000 ppm; ácido acético, 196.000 ppm; ácido fórmico, 196.000 ppm; ácido fosfórico, 21.000 ppm; ácido cítrico, 8.500 ppm); A4 (acidificante 4) - dieta basal com 0,6% do blend 4 e 0,15% do blend 5 (ácido benzóico, 590.000 ppm; ácido fórmico, 70.000 ppm; ácido fosfórico, 50.000 ppm; ácido cítrico, 40.000 ppm). Para o período de 14 a 34 dias, os tratamentos foram: Am - dieta inicial com 0,004% de sulfato de colistina; A1 - inicial com 0,3% do blend 1; A2 - inicial com 0,1% do blend 2 e 0,3% do blend 3; A3 - inicial com 0,6% do blend 4; A4 - inicial com 0,5% do blend 4 e 0,1% do blend 5. Foram utilizados 160 leitões Topigs recém-desmamados, com idade média em torno de 24 dias e peso inicial de 6,69±1,82 kg. Foram alocados quatro leitões (dois machos castrados e duas fêmeas) em cada baia (unidade experimental). Para os dados de desempenho e freqüência de diarréia, foram utilizadas 8 repetições (blocos) por tratamento. Ao final do experimento, um animal de cada baia, dos 4 primeiros blocos, foi abatido para análise da morfologia intestinal, pH estomacal e cecal e morfometria de órgãos. Durante a fase pré-inicial, o tratamento A2 (combinação dos blends 2 e 3) proporcionou melhor peso aos 14 dias (P14) (P=0,03) e ganho diário de peso (GDP) (P=0,04) que o tratamento A3 (blend 4), e melhor conversão alimentar (CA) (P=0,004) que o tratamento Am (antimicrobiano). Para o período total, o tratamento A4 (blends 4 e 5) apresentou melhor CA (P=0,0006) que o tratamento Am (antimicrobiano). Os tratamentos não afetaram (P>0,05) a freqüência de diarréia e o pH estomacal. O tratamento A4 (blends 4 e 5) apresentou menor valor de pH cecal (P=0,015) que o tratamento Am e menor valor de peso relativo do pâncreas (P=0,013), comparado aos tratamentos Am e A2. Para morfologia intestinal, o tratamento A2 proporcionou menores valores de profundidade de cripta (PC) (P=0,03) do jejuno que os tratamentos A3 e Am e maior relação altura de vilosidade:profundidade de cripta (P=0,04) do jejuno que os tratamentos A1 e A3. A análise econômica, no período de 1 a 14 dias, mostrou que os acidificantes propiciaram custos por kg de GDP, no mínimo, 4,09% inferiores ao antimicrobiano (Am). No período de 1 a 34 dias, os tratamentos A1 e A4 apresentaram custos por kg de GDP até 1,08% menores que o Am. Assim, o presente estudo mostrou que misturas de ácidos orgânicos, inorgânicos e seus sais podem ser uma alternativa viável aos antimicrobianos melhoradores do desempenho de leitões na fase de creche. / The purpose of this work was to evaluate several acidifier blends as alternatives to antimicrobial growth promoters of weanling pigs. A 34-d randomized complete block design experiment was carried out to compare five treatments. For 1-14 d experimental period, the treatments were: Am (antimicrobial) - pre-starter diet with 0.004% of colistin sulfate (40 ppm); A1 (acidifier 1) - pre-starter diet with 0.5% of blend 1 (containing formic acid, 145,000 ppm; phosphoric acid, 85,000 ppm); A2 (acidifier 2) - pre-starter diet with 0.15% of blend 2 (butyric acid, 64,000 ppm) and 0.4% of blend 3 (lactic acid, 620,000 ppm; formic acid, 40,000 ppm); A3 (acidifier 3) - pre-starter diet with 0.8% of blend 4 (propionic acid, 198,000 ppm; acetic acid, 196,000 ppm; formic acid, 196,000 ppm; phosphoric acid, 21,000 ppm; citric acid, 8,500 ppm); and A4 (acidifier 4) - pre-starter diet with 0.6% of blend 4 and 0.15% of blend 5 (benzoic acid, 590,000 ppm; formic acid, 70,000 ppm; phosphoric acid, 50,000 ppm; citric acid, 40,000 ppm). For 14-34 d experimental period, the treatments were: Am - starter diet with 0.004% of colistin sulfate; A1 - starter diet with 0.3% of blend 1; A2 - starter diet with 0.1% of blend 2 and 0.3% of blend 3; A3 - starter diet with 0.6% of blend 4; and A4 - starter diet with 0.5% of blend 4 and 0.1% of blend 5. One hundred and sixty Topigs 24-d-weaned pigs, with 6.69±1.82 kg live weight were allotted to 20 suspended pens, with four pigs (two castrated male and two female) per pen (experimental unit). For performance and diarrhea incidence data, 8 replications per treatment were used. On 34th day of experimental period, an animal of each pen of first 4 blocks was slaughtered for small intestine morphology, stomach and caecum pH and organs morphometry. For 1-14 d experimental period, treatment A2 (combination of blends 2 and 3) gave better body weight at 14th day (BW14) (P=.03) and average daily gain (ADG) (P=.04) than treatment A3 (blend 4), and better feed conversion (FC) (P=.004) than treatment Am (antimicrobial). For total experimental period (1-34 d), treatment A4 (blends 4 and 5) gave better FC (P=.0006) than treatment Am (antimicrobial). Treatments did not affect (P>.05) diarrhea frequency and stomach pH. Treatment A4 (blends 4 and 5) gave lower pH value (P=.015) than treatment Am and smaller relative pancreas weight (P=.013) than treatments Am and A2. For intestinal morphology, treatment A2 provided smaller (P=.003) jejunum crypt depth (CD) than treatments A3 and Am, and bigger (P=.04) ratio of jejunum villus height:crypt depth than treatments A1 and A3. Economical analysis showed, for 1-14 d experimental period, that the acidifiers provided cost per kg of ADG at least 4.09% lower than antimicrobial (Am). For 1-34 d experimental period, treatments A1 and A4 showed cost per kg of ADG until 1.08% lower than Am. Therefore, this study showed that blends of organic and inorganic acids and their salts can be viable alternatives to antimicrobial growth promoters of weanling pigs.

EFFECTS OF DIETARY ENZYMATICALLY TREATED YEAST IN WEANLING PIGS AND COCCIDIA-CHALLENGED BROILER CHICKENS

<p>The objective of this thesis was to investigate the effect of dietary enzymatically treated yeast (ETY) in weanling pigs and evaluate the effect of ETY in broiler chickens during a coccidia challenge. Two studies were carried out to investigate this objective.</p> <p>The first experiment examined the effect of ETY on the growth performance, nutrient digestibility, immune response, and gut health of weanling pigs. A total of 192 weanling pigs (6.0 ± 1.04 kg) were allocated to 4 corn, soybean-based diets with increasing concentrations of ETY (0, 1, 2, or 4 g/kg) for a 43-d trial. There were 8 replicate pens (4 replicate pens per sex) and 6 pigs per replicate for each dietary treatment. The experiment was set up as a randomized complete block design. Dietary ETY supplementation did not affect the growth performance of weanling pigs. The ATTD of neutral detergent fiber (NDF) linearly increased (P < 0.05) at d 28. The concentrations of serum antioxidant and antibody markers increased (P < 0.05) from d 14 to d 43; a linear increase (P < 0.05) in catalase was observed on d 14 with increasing ETY supplementation in the diets. The inclusion of ETY in the diet did not affect the mRNA abundance of anti-inflammatory markers in the ileal mucosa of pigs but increased (P < 0.01) glutathione peroxidase 4. Ileal villus height (VH) and villus height to crypt depth (VH:CD) ratio were greater (P < 0.05) in pigs fed ETY supplemented diets relative to control pigs. However, jejunal VH and the VH:CD ratio was not affected by ETY inclusion in the diet. Pigs fed diets with increasing ETY levels had higher (P < 0.05) ileal digesta butyrate concentration relative to control pigs, but not propionate or acetate. </p> <p>The second experiment examined the effect of ETY on the growth performance, nutrient digestibility, and intestinal health of broiler chickens during a coccidia challenge. From d 1 to 14 post hatching, 480 broiler chickens (49.9 ± 3.95 g) were allocated to 3 corn-soybean meal-based diets with increasing concentrations of ETY (0, 1, or 2 g/kg). There were 16 replicate cages and 10 birds per cage. The experiment was designed as a randomized complete block design with body weight (BW) used as a blocking factor. On d 14 post hatching, the birds were combined and re-randomized within each of the 0, 1, or 2 ETY g/kg experimental diets. Following this, the number of birds was reduced to 8 birds per cage with 8 replicate cages. Each of the 3 diet groups was split into a challenge or no-challenge group. This resulted in a 3 × 2 factorial arrangement of treatments with 3 experimental diets and 2 challenge states. The BW ratio before re-randomization was maintained across all treatments during the reallotment. On d 15 post hatching, the birds in the challenge group were orally gavaged with 1 mL solution containing 25,000, 25,000, and 125,000 oocysts of E. maxima, E. tenella, and E. acervulina, respectively. The birds belonging to the no-challenge group were orally gavaged with 1 mL phosphate buffered saline. Data were analyzed using the MIXED procedure of SAS, and polynomial contrasts were used to estimate the linear and quadratic effects of ETY. The coccidia challenge (CC) decreased (P < 0.01) the BW gain, feed intake, and G:F of broiler chickens from d 14 to 21. Increasing supplementation of dietary ETY improved (P < 0.05) the G:F of birds. Also, the CC reduced (P < 0.01) the apparent total tract utilization of dry matter (DM), nitrogen (N) and gross energy (GE). The CC reduced (P < 0.01) the apparent metabolizable energy (AME) and the nitrogen corrected apparent metabolizable energy (AMEn) of diets fed to broiler chickens. On d 21, dietary ETY linearly increased (P < 0.01) the apparent ileal digestibility of DM, N, and GE in broiler chickens. The CC increased (P < 0.01) the mRNA gene expression of TNFα, IL-1β, IL-10, and IL-6. There was a tendency (P < 0.1) for ETY to reduce IL-1β expression in broiler chickens on d 21. Additionally, ETY supplementation increased (P < 0.05) the gene expression of occludin in the ceca of broiler chickens but not claudin 1. Serum catalase increased (P < 0.05) with increasing supplementation of dietary ETY in broiler chickens on d 21. Dietary ETY linearly increased (P < 0.05) the ileal villus height to crypt depth ratio and ileal goblet cell count and density in broiler chickens. The ileal and excreta oocyst counts decreased (P < 0.01) with increasing supplementation of dietary ETY in coccidia-challenged broiler chickens on d 21. </p> <p>In summary, ETY inclusion in diets of weanling pigs partially reduced reactive oxygen species damage and enhanced intestinal health without negatively affecting growth performance. Hence, ETY could be favorable in attenuating some of the deleterious effects of post-weaning stress in weanling pigs. Dietary ETY also reduced oxidative damage, improved growth performance, enhanced nutrient utilization, and augmented intestinal development in broiler chickens. However, the inclusion of ETY did not mitigate the adverse effects of a coccidia challenge in broiler chickens. Therefore, further studies may be necessary to investigate the prospect of ETY as a dietary strategy for combating coccidiosis.</p> <p><br></p>

Lawsonia intracellularis and the disease it causes in horses, equine proliferative enteropathy (EPE), is an emerging pathogen of increasing importance to the horse industry from both an economic and welfare standpoint. Long recognized as an economically important disease of swine, the hallmark of EPE is a protein-losing enteropathy, where affected horses suffer weight loss and some ultimately succumb to the disease despite aggressive treatment. There are currently no known EPE preventative measures and the epidemiology of the disease remains poorly defined. While EPE is a sporadic disease affecting less than 25% of exposed horses, some farms experience clinical cases year after year. Further, weanlings are uniquely susceptible to this disease, although no conclusive reason for this predisposition has been identified. The overall hypothesis is that the host immune response plays a significant role in the susceptibility of weanlings to L. intracellularis infection and the occurrence of clinical equine proliferative enteropathy. To test this hypothesis, four individual hypotheses were proposed: (H1) previous farm history of EPE does not have an effect on weanling seroprevalence, (H2) passively-acquired antibodies do not have an effect on susceptibility to L. intracellularis and the occurrence of EPE, (H3) the serological status of mares can be used to determine the role they play in the epidemiology of EPE on endemic farms, and (H4) L. intracellularis-specific IFN-g expression is not associated with increased resistance to EPE.

Extratos vegetais como alternativas aos antimicrobianos promotores do crescimento de leitões recém-desmamados / Herbal extracts as alternatives to antimicrobial growth promoters of weanling pigs

O uso de antimicrobianos como promotores do crescimento na alimentação animal tem proporcionado uma melhora considerável no desempenho animal. Porém, seu uso vem sendo proibido em diversos países e, face a esta restrição, tem-se buscado alternativas aos antimicrobianos promotores do crescimento. Assim, o objetivo do trabalho foi avaliar extratos vegetais como alternativas aos antimicrobianos promotores do crescimento de leitões recémdesmamados, por meio do desempenho e da morfometria de órgãos dos animais. Um experimento em blocos casualizados, com 35 dias de duração, foi realizado para testar cinco tratamentos: controle - ração basal; antimicrobiano - basal com colistina + tiamutin (75 ppm de cada); extrato de cravo (Ec) - basal com 420 ppm de extrato vegetal (210 ppm de óleo essencial de cravo + 210 ppm do princípio ativo eugenol); extrato de orégano (Eo) - basal com 420 ppm de extrato vegetal (210 ppm de óleo essencial de orégano + 210 ppm do princípio ativo carvacrol) e extrato de cravo + orégano (E c+ Eo) - basal com 420 ppm de extrato vegetal (105 ppm de óleo essencial de cravo + 105 ppm de eugenol e 105 ppm de óleo essencial de orégano + 105 ppm de carvacrol), sendo todos os extratos vegetais encapsulados. Para o desempenho, foram utilizados 80 leitões, com idade média em torno de 24 dias e peso inicial de 7,19 ± 1,55, oito repetições por tratamento e dois animais por unidade experimental. Ao final do período experimental, um animal de cada baia, dos quatro primeiros blocos, foi abatido para morfometria de órgãos. Foram testados contrastes específicos de importância prática. No período de 1 a 14 dias, animais do tratamento antimicrobiano apresentaram melhor conversão alimentar (CA) que a média dos animais dos tratamentos com extratos (P=0,02), enquanto que animais do tratamento Ec apresentaram melhor CA que os do tratamento Eo (P=0,03). No período total, os animais do tratamento antimicrobiano apresentaram maior peso aos 35 dias (P35) (P=0,05) e ganho diário de peso (GDP) (P=0,06) que a média dos animais dos tratamentos com extratos. Os animais do tratamento Ec + Eo apresentaram maior P35 (P=0,02) e GDP (P=0,02) em relação à média dos animais dos outros tratamentos com extratos. Para a morfometria de órgãos os antimicrobianos proporcionaram maior peso relativo dos rins (P=0,01) em relação aos outros tratamentos de extratos. De modo geral, os antimicrobianos proporcionaram os melhores desempenhos de leitões em fase de creche. Quanto aos extratos vegetais, a combinação de Ec + Eo acarretou um desempenho muito próximo àquele obtido com os antimicrobianos, demonstrando ser uma alternativa promissora como promotor do crescimento de leitões recém-desmamados. / The use of antimicrobial as growth promoters in the animal feed has been related to an increase on animal performance. However, due to the restriction of many countries to the use of antimicrobial as growth promoters, alternatives are being studied. So, the purpose of this work was to evaluate herbal extracts as alternatives to antimicrobial growth promoters of weanling pigs based on performance and organ weights. A 35-d randomized complete block design experiment was carried out to compare five treatments: control - basal diet; antimicrobial - basal diet with colistin + tiamutin (75 ppm of each); herbal extract of clove (Ec) - basal diet with 420 ppm of herbal extract (210 ppm of essential oil of clove + 210 ppm of active compound eugenol); herbal extract of oregano (Eo) - basal diet with 420 ppm of herbal extract (210 ppm of essential oil of oregano + 210 ppm of active compound carvacrol); and herbal extract of clove + oregano (Ec + Eo) - basal diet with 420 ppm of herbal extract (105 ppm of essential oil of clove + 105 ppm of eugenol and 105 ppm of essential oil of oregano + 105 ppm of carvacrol ), being all the herbal extracts encapsulated. Eighty pigs (average age around 24 d and initial live weight of 7.19 ± 1.55 kg), eight replications per treatment, and two animals per experimental unit were used for performance data. At the end of experimental period, an animal per pen of first four blocks was slaughtered for organ morphometry data. Specific contrasts of practical importance were tested. For 1-14 d period, pigs fed antimicrobial showed better fed conversion (CA) than the mean of those fed herbal extract treatments (P=.02), while pigs fed Ec gave better CA than those fed Eo (P=.03). For the total period, pigs fed antimicrobial showed higher body weight at 35 days (P35) (P=.05) and average daily gain (GDP) (P=.06) than the mean of those fed herbal extracts. Pigs fed Ec + Eo gave higher P35 (P=.02) and GDP (P=.02) than the mean of those fed other herbal extract treatments. For the organ morphometry data, the antimicrobials provided a higher relative weight of kidneys (P=.01) than the mean of the other treatments with herbal extracts. Overall, antimicrobial agents provided the best performance of weanling pigs. Concerning to herbal extracts, the combination of clove and oregano provided performance close to that of pigs fed antimicrobials, showing that this combination can be a potential alternative as growth promoter of weanling pigs.
