Ilustra aplicação destas técnicas através da apresentação de casos, desenvolve simplificações do modelo adotado e cria tabelas (GA) para facilitar a implementação do sistema proposto.

In the case of injury to the CNS, different proteins act to repair and protect cells in the brain and spinal cord. In the present study, we looked at dorsal root injury and hypoglossal nerve avulsion and transection. Here we studied for the first time the expression of Parkin in these types of injuries. However the antibodies against Parkin used here have not been able to detect Parkin in the injuries examined, neither with fluorescence or using DAB. The roles of Mts1, GFAP, and clusterin after injury have been investigated earlier, but their co-localization in the same cells was first shown in this study in the hypoglossal nucleus with immunohisto-chemical methods. These results may also be of value in the process of finding an effective treatment for neurodegenerative disorders such as ALS.

Mytilus edulis est un mollusque bivalve de grand intérêt économique et écotoxicologique. Cette espèce sentinelle est connue pour sa résistance aux contaminants chimiques et biologiques. Néanmoins, depuis quelques années la moule bleue est touchée par des mortalités dans les élevages des Pertuis Charentais ayant pour dénominateur commun la présence de bactéries virulentes de type Vibrio. Le premier axe de cette thèse décrit les interactions des isolats de V. Splendidus avec la moule bleue au niveau cellulaire et physiologique. Les infections expérimentales ont permis la sélection de deux isolats bactériens affiliés à V. splendidus/V. hemicentroti : une souche virulente codée 10/068 1T1 et une souche inoffensive codée 12/056 M24T1. Ces deux bactéries ont été marquées à la GFP et validées en tant que modèles authentiques d’exposition à travers leurs caractéristiques de croissance et de virulence. Par ailleurs, V. hemicentroti 10/068 1T1 est capable d’altérer différentes fonctions hémocytaires incluant la motilité, l’adhésion, l’internalisation, la production de ROS, la maturation du phagosome et la viabilité contrairement à la bactérie non virulente. Les produits extracellulaires bactériens semblent être toxiques et inhibent certaines réponses cellulaires (internalisation et production de ROS). Enfin, nous avons reproduis avec succès l’infection des animaux par le pathogène via un modèle expérimental de cohabitation. Le suivi de l’infection montre que V. hemicentroti 10/068 1T1 a pour cible principale les branchies. Le deuxième axe explore le fonctionnement du système MXR (MultiXenobiotic Resistance) chez la moule bleue. La séquence codante complète d’un nouveau transporteur ABCG2 a été établie et la protéine résultante a été identifiée. La caractérisation moléculaire montre la présence du transcrit dans les hémocytes ainsi que dans les branchies et son homologie avec les autres protéines appartenant à diverses espèces. L’utilisation des sondes fluorescentes bodipy prazosin et pheophorbide A, combinées avec des bloqueurs spécifiques, démontre l’activité d’efflux de ce transporteur et son hétérogénéité dans les tissus et cellules. Par ailleurs, il est également démontré que l’expression des trois transporteurs ABC (abcb, abcc, abcg2) identifiés chez la moule bleue est modulée par les contaminants chimiques. Les animaux exposés au BaP au laboratoire ou prélevés sur un terrain contaminé montrent une surexpression des transcrits abc dans les branchies et une sous expression dans les hémocytes. La saisonnalité, sur le terrain, a également un effet sur les niveaux des transcrits et interfère avec les réponses liées aux contaminants. Seul le transporteur abcb exprimé dans les branchies n’est pas affecté par des variations saisonnières et montre une surexpression dans le site contaminé tout au long de l’année. En conclusion, nos résultats démontrent la vulnérabilité de la moule bleue face un pathogène. L'impact immunotoxique des xénobiotiques et le rôle que peuvent jouer les transporteurs ABC dans le fonctionnement du système immunitaire des moules reste à explorer. / Mytilus edulis is a bivalve mollusc representing an economic and ecotoxicological interest. This sentinel species is known for its resistance to chemical and biological contaminants. However, for few years, abnormal mortality events have been reported for farmed blue mussels in France where different Vibrio strains were isolated. The first section of this thesis describes cellular and physiological interactions of V. Splendidus isolates with the blue mussel. Experimental infections allowed the selection of two isolates affiliated to V. splendidus/V. hemicentroti type strains: a virulent 10/068 1T1 and an innocuous 12/056 M24T1. These two strains were GFP-tagged and validated for their growth characteristics and virulence as genuine models for exposure. V. splendidus 10/068 1T1 is capable to alter different functions of hemocytes including motility, adhesion, internalization, ROS production, phagosome maturation and viability, unlike the avirulent strain. Furthermore, bacterial extracellular products appeared toxic and inhibit cellular responses (internalization and ROS production). Finally, we successfully reproduced experimental infection by water tank cohabitation assays with septic animals. Infection monitoring shows the targeting of gills by bacteria. The second section explores the MXR (MultiXenobiotic Resistance) system functioning in the blue mussel. For the first time, a complete ABCG2 amino acid sequence was established. Molecular characterization shows the presence of the abcg2 transcript in hemocytes and gills and its homology with other proteins from various species. The combination of the fluorescent probes bodipy prazosin and pheophorbide A with specific blockers demonstrate the transporter efflux activity and its heterogeneity in tissues and cells. Moreover, the expression of three ABC transporters (abcb, abcc, abcg2) identified in the blue mussel has been shown to be modulated by chemical contaminants. Mussels exposed to BaP in the laboratory or collected from contaminated mussel beds in the field show upregulated abc transcripts in gills whereas these mRNA undergone a downregulation in hemocytes. Season had also an effect on mRNA levels and interacted with site effects. Only the abcb gene displayed a more abundant mRNA level in gills dissected from animals collected in the more polluted area all over the diachronic study. In conclusion, our results demonstrate the vulnerability of the blue mussel towards a pathogen. The immunotoxic impact of environmental xenobiotics and the precise role of ABC efflux pumps in the immune system of the mussel has yet to be explored.

Les principaux transporteurs ABC et cytochromes P450 exprimés au niveau des microvaisseaux cérébraux chez l’homme sont l’ABCB1/P-gp, l’ABCG2/BCRP et le CYP1B1. Au niveau des organes périphériques, la régulation de l’expression de ces trois marqueurs fait intervenir des facteurs de transcription et notamment le récepteur aryl hydrocarbon (AhR). Or les transcrits d’AhR sont très exprimés au niveau des microvaisseaux cérébraux humains. Les travaux de cette thèse ont donc dans un premier temps été consacrés à l’étude de l’implication de la voie du récepteur Ah dans la régulation de l’expression de l’ABCB1, de l’ABCG2 et du CYP1B1 au niveau de la barrière hémato-encéphalique. In vivo, nous avons mis en évidence qu’un traitement aigu par la dioxine (ou TCDD), ligand puissant d’AhR, induisait l’expression génique et protéique du Cyp1b1 au niveau des microvaisseaux cérébraux de rats adultes Srague-Dawley. De même, in vitro, l’exposition au TCDD a fortement induit l’expression du CYP1B1 dans la lignée hCMEC/D3, un modèle in vitro de l’endothélium cérébral humain et le recours à la technique de l’ARN interférence nous a permis de démontrer que le récepteur Ah était impliqué dans les effets observés. En revanche, que ce soit in vivo ou in vitro, l’exposition au TCDD n’a entrainé aucune modification significative de l’expression de l’ABCB1 ou de l’ABCG2. Dans un second temps, nous avons étudié l’interrelation entre la voie AhR et les voies de réponse à l’hypoxie cellulaire. Les différentes expériences réalisées sur la lignée cellulaire hCMEC/D3 ont mis en évidence une interaction non réciproque entre ces deux voies de signalisation : en cas d’activation simultanée, la réponse à l’hypoxie abolit la réponse AhR tandis que l’activation de la voie AhR ne modifie pas la réponse adaptative à l’hypoxie. / ABCB1 (P-gp), ABCG2 (BCRP) and CYP1B1 are the main ABC transporters and cytochrome P450 enzymes expressed at the human blood-brain barrier (BBB). In peripheral tissue, expression of these proteins is regulated by transcription factors such as the aryl hydrocarbon receptor (AhR). Interestingly, high levels of AhR mRNA are detected in human brain microvessels. We therefore investigated the potential implication of AhR in the regulation of ABCB1, ABCG2 and CYP1B1 expression. In vivo, a single dose of TCDD, a highly potent AhR ligand, increased Cyp1b1 transcripts and protein expression in rat brain microvessels. Similarly, exposing hCMEC/D3 cells, an in vitro promising model of human BBB, to TCDD induced CYP1B1 expression. Using small interfering RNA, we established AhR involvement in TCDD effects. However, either in vivo or in vitro TCDD treatment had no effect on ABCB1 or ABCG2 expression. Next, we investigated the crosstalk between AhR and hypoxia signalling pathways in case of simultaneous activation. Our experiments revealed that a crosstalk between these two pathways effectively occurred in hCMEC/D3 cells: hypoxia inhibited AhR response but not the reverse.

The diploma thesis deals with the theme of Modeling business processes with orientation to the metric process interface. The aim of the thesis is to design the architecture of metric, which enabled to construct type metrics of the process interface. Firstly, the reader is informed about the basic terms related to process modeling, process management and connection of strategy with metrics. The next part describes the architecture of metrics created, which is oriented to supporting the process management of the organization. The set of type metrics was chosen with consideration to the possibility of their placement on process interface. The set created in this manner should help to connect strategy with processes when implementing into organization. The last part of the thesis describes the practical application of knowledge obtained in the PARMA project. This project deals with the proposal and implementation of the process management into the Bureau of the Municipality of Prague 10. The thesis deals with two processes and the conclusions obtained in the theoretic part of the thesis are applied herein.

Essa investigação é orientada por epistemologias decorrentes da terceira onda do feminismo, bem como mescla considerações teóricas de diferentes áreas de saber. Devido ao seu caráter empírico, essa pesquisa não se vale somente de conhecimentos institucionalizados que possam subsidia-la, mas também se vale dos conhecimentos dos sujeitos envolvidos no campo que são, por excelência aqueles que vivenciam a problemática apresentada e que, portanto, possuem propriedade para tratar sobre esta. Nesse sentido, esse trabalho se direciona as dinâmicas da produção literária local, com vistas a compreender como a expressão da experiência feminina através da literatura e a atuação dessas mulheres na disputa pelos meios em que se produz a literatura tem sido relevante para os novos horizontes esboçados pela produção cultural alternativa à cultura dominante. / This dissertation draws on female authorship. It is focused on the identification of the place that the writer woman occupy on Brazilian society, taking as a geographic reference the region nominated as ABC Paulista. This investigation is guided by epistemologies from the feminist third wave, as well as mixes considerations from different knowledge areas. Due to its empirical character, this research is not only based on the institutionalized knowledge that can subsidize it, but it also uses the knowledge of the subjects involved in the field, that are, par excellence, those who experience the question presented and, therefore, are those who have property to deal with this. In this sense, this work addresses the dynamics of local literary production to understanding how the expression of the female experience through literature, and the performance of these women in the dispute over the means in which literature is produced has been relevant to the new horizons outlined by a cultural production alternative to the dominant culture.

El proyecto de rediseño del sistema se ha realizado para mantener la operación activa y el flujo de medicamentos en la empresa. Si bien la farmacia o el almacén principal representa el 10% de los ingresos netos de la organización, es la principal área de apoyo para los principales servicios ofrecidos por la clínica a los clientes, ya que es responsable del suministro médico al consumidor interno y externo donde se interactúa con las actividades de la empresa. La implementación del proyecto consistió en calcular los lotes óptimos para cada línea de productos que se agruparon para un mejor tratamiento de los medicamentos: 700 artículos a 50 familias. Las líneas de productos para aquellos con características similares se definieron y se pudieron comprar a un proveedor en particular. Además, se establecieron descuentos para reducir los niveles de precios en la farmacia. En relación con el control de stock, se estableció el Plan Cíclico basado en el análisis Multicriterio ABC. Como parte del alcance del proyecto, se tomó una muestra con un nivel de confianza del 90% de la implementación del tipo de producto "A" total para aplicar durante 3 meses. En efecto, los pedidos de productos elegidos en el Plan Piloto cayeron del 80% al 12.5% de las ventas no atendidas por la falta de disponibilidad de existencias y los altos precios. Los resultados se lograron con la implementación de nuevos procedimientos de POES que involucran a los responsables de las áreas de la Clínica. / The System redesign project has been done in order to maintain active operation and flow of medicines in the company. Although Pharmacy - or main warehouse - accounts for 10% of net income in the organization, it is the main area of support for the main services offered by the clinic to the customers, because it is responsible for the medical supply to consumer internal and external interacting with the activities of the company. The implementation of the project was to calculate the optimal batch order for each product line, which were grouped for better treatment of medicines - 700 items to 50 families. Product lines for those with similar characteristics were defined and could be purchased from a particular vendor. Also, discounts were established to reduce the levels of prices in the pharmacy. In relation to stock control, counting Cyclical Plan based on analysis Multicriteria ABC was established. As part of the project scope, a sample at 90% confidence level of total product type “A” implementation to apply for 3 months was taken. In effect, orders for products chosen in the Pilot Plan fell from 80% to 12.5% of sales not served by non-availability of stock and high prices. The results were achieved with the implementation of new POES procedures involving those responsible for the areas in the Clinic. / Tesis

This diploma´s thesis is focused on design of the logistics concept of selected production section in manufacturing firm with using logistics method Just In Time. In analytical part is analysed current situation of managing material flows for selected production sections, ways of supplying production units and amount of stock which is stored in production areas. In proposal part are suggested some changes in various spheres along with new design of the logistics concept and possible economical contributions in case of applying this logistics concept.

The subject of this thesis are the costs of a specific company, their monitoring and ma-nagement. The theoretical part summarizes the information that served as the default resources. These findings have been applied in the practical part, for the selected com-pany. In the final section are formulated suggestions, which should lead to better ma-nagement of the costs.

The aim of the thesis, which is divided into a theoretical and a practical part, is to describe and analyze the current tendency in a company’s cost management, and suggest a precaution leading to an improvement. The theoretical part compiles the classification of costs, calculations, and tools and processes of cost management. The practical part then describes cost management in the analyzed company. Both vertical and horizontal financial analysis are rendered – from the analyses, precautions are defined, as well as cost items, which are suggested to be paid attention to.
