Sleep patterns of naval aviation personnel conducting mine hunting operations

Solberg, Bennett Jon. 09 1900 (has links)
Human Systems Integration Report / Detailed research conducted over the past forty years has conclusively determined that varying degrees of sleep loss, shifts in sleep cycle, increased stress and even changes in time zone with respect to daylight transition result in a myriad of physiological and psychological degradations (Helmreich, 2000). Fatigue affects human performance, resulting in predictable changes not only on the individual level but also on the system as a whole. This descriptive study investigates the amount and quality of sleep received by aviation personnel assigned to an operational squadron conducting mine hunting operations. Wrist activity monitors (actigraphs) were used to determine objective assessments of sleep quantity and quantity. Demographic variables and additional measures such as reported sleepiness, fatigue ratings, caffeine and alcohol use, were also collected. Despite a number of factors which altered the original study design, significant differences in amount of sleep, sleep quality and predicted effectiveness of personnel by officer-enlisted status were identified.

Exploring the Molecular Behavior of Carbohydrates by NMR Spectroscopy : Shapes, motions and interactions

Engström, Olof January 2015 (has links)
Carbohydrates are essential biomolecules that decorate cell membranes and proteins in organisms. They are important both as structural elements and as identification markers. Many biological and pathogenic processes rely on the identification of carbohydrates by proteins, thereby making them attractive as molecular blueprints for drugs. This thesis describes how NMR spectroscopy can be utilized to study carbohydrates in solution at a molecular level. This versatile technique facilitates for investigations of (i) shapes, (ii) motions and (iii) interactions. A conformational study of an E. coli O-antigen was performed by calculating atomic distances from NMR NOESY experiments. The acquired data was utilized to validate MD simulations of the LPS embedded in a membrane. The agreement between experimental and calculated data was good and deviations were proven to arise from spin-diffusion. In another study presented herein, both the conformation and the dynamic behavior of amide side-chains linked to derivatives of D-Fucp3N, a sugar found in the O-antigen of bacteria, were investigated. J-couplings facilitated a conformational analysis and 13C saturation transfer NMR experiments were utilized to measure rate constants of amide cis-trans isomerizations. 13C NMR relaxation and 1H PFG diffusion measurements were carried out to explore and describe the molecular motion of mannofullerenes. The dominating motions of the mannofullerene spectral density were found to be related to pulsating motions of the linkers rather than global rotational diffusion. The promising inhibition of Ebola viruses identified for a larger mannofullerene can thus be explained by an efficient rebinding mechanism that arises from the observed flexibility in the linker. Molecular interactions between sugars and caffeine in water were studied by monitoring chemical shift displacements in titrations. The magnitude of the chemical shift displacements indicate that the binding occurs by a face to face stacking of the aromatic plane of caffeine to the ring plane of the sugar, and that the interaction is at least partly driven by solvation effects. Also, the binding of a Shigella flexneri serotype Y octasaccharide to a bacteriophage Sf6 tail spike protein was investigated. This interaction was studied by 1H STD NMR and trNOESY experiments. A quantitative analysis of the STD data was performed employing a newly developed method, CORCEMA-ST-CSD, that is able to simulate STD data more accurately since the line broadening of protein resonances are accounted for in the calculations. / <p>At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 2: Manuscript. Longo, Daniela Paula. January 2006 (has links)
Abstract: Oil, water and surfactant mixture may form a variety of structures and phases accordingly to the characteristics and the ratio of the used components. Microcompartmented systems as liquid crystals and microemulsions are used to encapsulated drugs and active substances in pharmaceutical and cosmetic products. The objective of this research was the development of microcompartmented systems for incorporation of caffeine with the purpose of topical use on treatment of the gynoid lipodystrophy. The direct mixture of PPG-5-Ceteth-20 (surfactant), dybutyl adipate (oil phase) and water in differents proportions made possible to obtain formulations with different structuring forms as microemulsions, emulsions and liquid crystalline mesophases. The formulations were characterized through the polarization light microscopy, refractive index, relative density, conductivity and behavior rheological. Solubility studies were accomplished showed that the systems were capable to solubilize about 1,5% of caffeine. In vitro release studies using the cell of diffusion model in static system of Franz's type adapted to the dissolution equipment indicated formulations with characteristics of reservoirs systems. The permeability study in vivo used male pig (Landrace x Large White) and they were submitted to the treatment. Formulations with or without caffeine were applied. Skin's fragments and hypoderm of them were removed for histology analysis after the treatment. The formulation with liquid crystal of hexagonal phase characteristics was efficient in increasing skin permeability to the caffeine, thus facilitating its drug action. The application of caffeine this way led to a significant reduction of the hypoderms thickness. The developed system is adjusted for topical use, is skin penetration enhancers and it guarantees the effectiveness of the caffeine in the treatment of the cellulite. / Mestre

Abstract: Currently, equine exercise physiology is in a considerable development in Brazil. The use of scientific methods for the evaluation of the exercise and training protocols is fundamental for the maximization of equine performance. It also can be applied to evaluate the effects of legal (ergogenics) and/or illegal substances during competitions on athletic horses. Some metabolic, endocrine, cardiac and hematologic responses of trained Arab horses submitted to intense exercise and to the administration of caffeine, aminophylline and clenbuterol, a single dose given at 30 minutes before the effort. Doses were, respectively, 5 mg.kg-1, 10 mg.kg-1 and 0,8 Êg.kg-1. A treadmill was employed for the evaluation of exercise and drug effects, with 10% slope and speed increments. Blood samples were withdrawn 15 seconds before the end of each exercise bout. Hematocrit, erythrocytes and leucocytes count, hemoglobin, glucose, lactate, insulin and cortisol concentration, were determined. Heart rate was also monitored at the same time points. A minimum criterion of P.0,05 was adopted for statistical significance. Our results revealed significant variations in all physiologic parameters studied. Caffeine impaired the aerobic capacity but improved the aerobic potency. Aminophylline interfered in glycemic curve during intense exercise, but improved anaerobic performance. Finally, clenbuterol did not improve the parameters association with aerobic metabolism, but, markedly, impaired cardiac response, and increased insulin concentration compromising glycemic control during intense exercise. / Doutor

Efeito tóxico da cafeína sobre o ciclo de vida de Chironomus sancticaroli (Chironomidae, Diptera) e Daphnia magna (Daphniidae, Cladocera) / Caffeine toxic effect on Chironomus sancticaroli (Diptera: Chironomidae) life cycle and Daphnia magna (Cladocera: Daphnidae)

Bernegossi, Aline Christine 02 April 2019 (has links)
Caffeine is present in human daily life either as traditional coffee or constituent of medicines, stimulants, teas, chocolate, among other products. This substance is an emerging organic contaminant from untreated wastewater. Its presence in aquatic systems has been documented at low concentrations, from &eta;g.L-1 to &#181;g.L-1 and the effects of these concentrations on aquatic organisms have not yet been effectively investigated. In order to evaluate the toxic effects of caffeine, at concentrations usually found in aquatic systems, acute and chronic ecotoxicological tests were performed with Chironomus sancticaroli and Daphnia magna and, an intergenerational with C. sancticaroli, focusing on biological responses of mortality, immobility, growth, development and reproduction. As a result, caffeine did not present acute toxicity in the shor-term test performed for D. magna and C. sancticaroli. Chronic toxicity of caffeine was observed, with interference in the Chironomus larvae development, mainly in the concentration of 200 &#181;g.L-1, and in the growth and reproduction of D. magna, in which it was observed presence of induction in the production of neonates and deceleration to the development of the organism (delay in the process of ecdysis). In intergeneration test, changes in the larval developmental patterns were observed. In intergenerational testing, changes in larval developmental patterns were observed. The results demonstrate that caffeine, in concentrations detected environmentally, can cause small influences in the organism\'s life cycle of the, for example, changes in the size of the wings of the females and in the length of the body of the larvae of C. sancticaroli. The use of biomolecular techniques to evaluate the metabolic responses of organisms when exposed to caffeine at low concentrations is recommended.

Derivados da cafeína como moderadores do estresse oxidativo provocado por sobrecarga de ferro / Caffeine derivatives as moderators of oxidative stress induced by iron overload

Pastrana Alta, Elizabeth Carmen Pastrana 30 May 2014 (has links)
Iron is an abundant and important chemical element for organisms. In human beings, redox reactions of iron is essential to the vast majority of their enzymatic functions. However, outside suited biochemical compartments, iron may behave as a toxin, since it may catalyze the formation of reactive oxygen species (ROS). Iron overload may affect the central nervous system. Several evidences point to the role of iron in neurodegenerative conditions such as Alzheimer Disease (AD). AD is characterized by the accumulation of insoluble aggregates of the &#946;-amiloid protein (A&#946;), and iron may both actively induce the aggregation of A&#946; and generate free radicals. Chelation therapy is the frontline treatment for iron overload, employing ligands such as desferrioxamine (DFO). Caffeine is a xanthine with antioxidant activity. Studies with animal models of AD indicate that caffeine may decrease the rate of formation of A&#946;, as well as improve learning and memory. In this work, a ligand was synthesized incorporating the chelator moiety of DFO and the caffeine environment (DFCAF), which could help to maintain normal cerebral iron homeostasis. DFCAF was characterized by elemental analysis, thermal analysis and spectroscopic techniques. Binding of DFCAF with metal ions (Fe(III), Cu(II) and Mn(II)) occurred with a 1:1 metal:ligand stoichiometry. Antioxidant activity of DFCAF was assayed in the iron/ascorbate system, modified ORAC and reduction of DPPH radical. Whenever oxidation was iron-dependent, DFCAF displayed identical antioxidant activity as the parent DFO. Both free DFCAF and Fe(DFCAF) were permeable to HeLa cells. as opposed to the observed by DFO or Fe(DFO). It was possible to show that conjugation of caffeine to DFO lead to a new molecule which preserves both DFO binding properties and caffeine structure. The presence of the xanthine environment in DFCAF assists on its cell permeability, which can be of interest as an adjuvant on the therapy of AD.

Quantificação do potencial osteogênico do osso autógeno + células osteoblásticas implantados em defeito ósseo no rato tratado com cafeína / Quantification of the osteogenic potential of autogenous bone + osteoblastic cells implanted in bone defect in rats treated with caffeine

Macedo, Rander Moreira 25 September 2009 (has links)
Studies suggest that caffeine acts on the bone for increasing the excretion of calcium and inhibition of osteoblasts proliferation, increasing the risk of fractures, osteoporosis and periodontal disease. The effects of caffeine on bone difficult the application of dental implants due to large bone defects and insufficient bone volume. Several methods are proposed for the regeneration of bone defects, including the use of different types of grafts, which show ability to promote bone formation. Despite the disadvantages, the autogenous bone is still considered the gold standard as bone graft because the potential osteogenic, osteoinductive and osteoconductive. The bone tissue engineering has been used as a strategy for bone regeneration. The mesenchymal stem cells are considered multipotent and can replicate as undifferentiated cells, with potential to differentiate into lineages of bone, cartilage, fat and cartilage. This study aimed to quantify histomorphometrycally bone repair by grafts of a combination of autogenous bone obtained from the skull and osteoblastic cells in bone defects produced by dental extraction in rats subjected to daily administration of caffeine. The animals were divided into: Control (c), autogenous bone (ab) and autogenous bone + osteoblastic cells (ab + oc) and received daily injections intraperitoneally of 30 mg/kg/day of caffeine for thirty days, the counterparts received saline solution. The rats were sacrificed at times of 7th, 21st and 42nd days post-surgery and tissue samples were processed to obtain thin sections (5 m) and stained with HE. Through an image analysis system was estimated the fraction of volume of bone, collagen and blood clot in the bone defect. The histological and histometric results showed that in animals under treatment with caffeine had a lower bone formation statistically significant at 1%, and a delay in the resorption of blood clots when compared to the alveoli of animals under treatment with saline. The qualitative analysis of the fragment of autogenous bone alone or associated with osteoblastic cells showed a progressive osteointegration and no foreign body reaction in animals treated with saline, and implanted the cells not provided immunogenic reactions or tumor formation, allowing an increase (25%) on bone repair in animals treated with caffeine. It was concluded that the graft/implant of osteoblastic cells associated with autogenous bone from the skull was able to compensate in later periods, the deleterious effects of caffeine on alveolar boné repair.
