Production, purification et caractérisation d’une gonadotropine chorionique équine recombinante à usage vétérinaire / Production, purification and characterization of recombinant equine chorionic gonadotropin for veterinary use

Des hormones gonadotropiques sont utilisées pour la maîtrise de la reproduction dans le domaine vétérinaire. Ces hormones sont actuellement extraites de tissus ou de fluides animaux. L’entreprise CEVA Santé Animale a récemment fait le choix stratégique de produire ces hormones par voie recombinante. L’objectif de cette étude était d’obtenir une gonadotropine chorionique équine recombinante, reCG, pure et biologiquement active, à partir d’une lignée de cellules mammifères CHO. Les étapes de production, de purification et de caractérisation de l’hormone recombinante ont été développées. Les cellules CHO ont été cultivées en fiole d’Erlenmeyer dans différents milieux de culture. Le suivi de la croissance cellulaire et de la quantité d’hormone produite a permis de sélectionner deux milieux. Le procédé de production, avec ces deux milieux, a été optimisé en bioréacteur en contrôlant les paramètres de culture (température, pH). Les protéines produites dans le surnageant, de ces deux cultures, ont été nommées reCG 1 et reCG 2. Un procédé de purification en 3 étapes a été mis au point pour la reCG 1. Plusieurs résines et conditions chromatographiques ont été criblées en microplaques. Les résines multimodales utilisées ont permis d’éliminer des contaminants majeurs grâce à leur sélectivité. Les agrégats de la reCG ont été éliminés grâce à une résine anionique. Le procédé de purification global a été validé pour la reCG 1 et la reCG 2. Il a permis d’obtenir une pureté de 98 % avec un rendement de 80 %. L’activité biologique de la reCG 1 et la reCG 2, in vitro et in vivo, est comparable à celle de la protéine naturelle. L’activité biologique in vivo des reCG est cohérente avec l’étude réalisée sur les glycosylations des hormones et notamment avec leur degré de sialylation. / The gonadotrophic hormones are used for reproduction control in farming animals. Up to now, these hormones were extracted from animal fluids or tissues. The company CEVA Santé Animal has recently decided to produce recombinant versions of these hormones. The objective of this study was to obtain a pure and biologically active recombinant equine chorionic gonadotropin (reCG) after expression in CHO mammalian cells. The production, purification and characterization steps have been developed. CHO cells were grown in Erlenmeyer flasks with different culture media. Two media were selected based on their cell growth potency and of the amount of reCG produced. By using a bioreactor to control key parameters (temperature, pH), the production process was then optimized. The recombinant proteins that accumulated in the supernatant of the two conditions were called reCG 1 and reCG 2. A 3-steps purification process was then developed using reCG 1. Several resins and chromatographic conditions were screened in microplates. Multimodal resins were used to eliminate the main contaminants thanks to their selectivity. reCG aggregates were efficiently eliminated by a chromatographic step with an anionic resin. The overall purification process was finally validated for reCG 1 and reCG 2. Purity and yield were respectively, 98 % and 80 % for the two reCG. We verified that the in vitro and in vivo activities of reCG 1 and reCG 2 were comparable to those of the CG extracted from natural sources. The in vivo assays also confirmed previous studies showing that the degree of glycosylation of an hormone, and most notably their level of sialytation, is important for their biological activity.

The theme of the thesis is the use of a questionnaire GPOP (Golden Profiler of Personality) in international research, specifically in the Czech Republic and Switzerland. The theoretical part is devoted to the three basic thematic areas, which are interconnected through the influence of C. G. Jung: cross-cultural comparison of the Czech Republic and Switzerland, Jung's typology, from which GPOP questionnaire derives from and mid-life issues described by Jung as individuation process. The emphasis is placed mainly on various forms of cross- cultural research and cultural standards in both countries on the basis of thereof the results are interpreted. The empirical part includes a detailed description of the questionnaire method and a sample of studied people. It is followed by a quantitative study in which there are statistically compared results of the four basic dimensions GPOP test pro-bands in the Czech Republic and Switzerland. An overview of personality types in individual countries and comparison of personality differences on cross-cultural background comes after. Finally, it assesses the work with GPOP method and its application.

[pt] Esta dissertação tem por objetivo investigar o aspecto transgressivo que caracteriza o pensamento da psicanalista francesa Nathalie Zaltzman ao revisitar a metapsicologia freudiana. Com a acepção de ultrapassar, violar, não cumprir, a transgressão revela-se uma ação psíquica vital para o sujeito, sobretudo quando esse se encontra em perigo de vida devido a circunstâncias opressoras e asfixiantes. Iniciamos nossa investigação por meio da articulação teórica do viés anarquista da pulsão de morte, manifestação em forma de luta e resistência contra forças que diminuem ou anulam a existência de um ser humano. A partir dessa perspectiva, percorremos proposições acerca da clínica. Destacamos a importância dada à ação de desligamento da pulsão de morte e a urgência da escuta, durante o tratamento, de suas manifestações. Expandindo a discussão com a abordagem do processo civilizatório, demonstramos como o trabalho de cultura poderia ser considerado o seu viés transgressor. Por meio desse, poder-se-ia lidar de maneira mais lúcida com a dimensão maléfica do humano, evitada a qualquer custo pela perspectiva civilizatória. Afirma-se a necessidade de fomentar vias que façam frente a posturas censoras e moralizantes. Essas impediriam o mal, dimensão inelutável e inerente à condição humana, de ocupar lugar nas representações psíquicas conscientes de cada indivíduo e no patrimônio simbólico da humanidade. Uma vez impedido, só lhe restaria abrir caminhos violentos de satisfação, reascendendo a barbárie. / [en] The present dissertation aims to investigate the transgressive aspect which characterizes the thoughts of the French psychoanalyst Nathalie Zaltzman in review of freudian metapsychology. With the sense of overtaking, violating, unaccomplishing, transgression is revealed as a vital psychic way out, specially when one s life is endangered due to oppressive and suffocating circumstances. We start our investigation with the theoretical articulation of the anarchist aspect of death drive, a manifestation characterized by the struggle and resistance against forces which dominate, diminish or nullify the human being existence. Based upon such perspective, we go along propositions about the clinic. We outline the importance given to the act of unattachment from death drive and the urgency of its listening, throughout the treatment, its manifestations. In order to take part in the discussion with the civilizing process, we demonstrate how culture work may be considered its transgressive way. By taking such approach for granted one could handle with a more lucid way in facing the maleficent dimension of the human, which is avoided by all costs in the civilizing perspective. It is argued the need to promote ways of confronting censorship and moralizing positions. Such act would prevent evil, ineluctable and insisting dimension of the human, from taking place in the conscious psychic representations of each single person and in the symbolic patrimony of humanity. Once there is an imposition, the only way left is to open violent paths of satisfaction, reigniting barbarism.

Procédés de cultures de cellules VERO en milieu sans sérum : contributions au développement d'une stratégie PAT / Vero cell culture processes in serum-free medium : contributions to the development of the PAT strategy

Ce travail apporte une contribution au développement de la stratégie PAT pour les procédés de culture de cellules animales. Il propose l'amélioration de la compréhension et de la maîtrise de la culture de cellules Vero, dédiées à la production de vaccins viraux, et cultivées sur microporteurs dans un milieu sans sérum. Une première partie a permis de cribler les effets de certains groupes de composés du milieu de culture par le suivi de la croissance en microplaques. Puis, des études cinétiques et métaboliques plus approfondies, réalisées en spinners, ont montré que le métabolisme carboné des cellules Vero est saturé par l'accumulation intracellulaire de pyruvate et qu’il est peu orienté vers la croissance. Alors que le renouvellement du milieu ou l'ajout ponctuel de glutamine amélioraient la croissance cellulaire sans ré-équilibrer le métabolisme, la substitution du glucose et de la glutamine a permis de réduire l’apoptose et d'améliorer les performances métaboliques et de croissance. Par ailleurs, les spectroscopies diélectrique et proche-infrarouge ont été évaluées pour le contrôle en-ligne du procédé, en prenant en compte les particularités des cellules adhérentes. Nous avons montré leur capacité à évaluer les concentrations de cellules, de composés du milieu, et à détecter l'apoptose. Enfin, les principales améliorations par substitution de la glutamine ont été appliquées en bioréacteurs, à la production d'un vaccin prototype contre la dengue, dans des conditions proches de celles d'un procédé industriel. Ceci a permis de limiter les renouvellements de milieu pendant l’expansion cellulaire, sans compromettre la production de particules virales infectieuses / This work contributes to the development of the PAT strategy for animal cell culture processes. The aim of this study was to improve the understanding and the control of Vero cell culture, dedicated to the production of viral vaccines, and grown on microcarriers in serum-free medium. An initial study was performed to screen the effects of certain groups of compounds of the culture medium, by the cell growth monitoring in microplates. Then, kinetic and metabolic studies conducted in spinners flasks allowed to go further and to show that the Vero cell metabolism is saturated through the pyruvate intracellular accumulation and that it is not oriented toward growth. While media renewal or punctual addition of glutamine improve the cell growth without improving the metabolism balance, the substitution of glucose and glutamine allowed to reduce apoptosis and to improve growth and metabolic performances.Furthermore, dielectric and near-infrared spectroscopies have been evaluated for the in-line process monitoring, taking into account the particularities of adherent cells. We have demonstrated their ability to quantify cell concentrations, medium component concentrations, and to detect apoptosis. Finally, major improvements by substitution of glutamine have been applied to bioreactor culture to produce a dengue vaccine prototype, with culture conditions close to industrial process. In these cases, medium renewal during the cell expansion was removed without compromising the production of infectious viral particles
