A Novel Mechanism Underlies Pathological, β-amyloid-induced Neuronal Hyperexcitation

abstract: Patients with Alzheimer's disease (AD) exhibit a significantly higher incidence of unprovoked seizures compared to age-matched non-AD controls, and animal models of AD (i.e., transgenic human amyloid precursor protein, hAPP mice) display neural hyper-excitation and epileptic seizures. Hyperexcitation is particularly important because it contributes to the high incidence of epilepsy in AD patients as well as AD-related synaptic deficits and neurodegeneration. Given that there is significant amyloid-β (Aβ) accumulation and deposition in AD brain, Aβ exposure ultimately may be responsible for neural hyper-excitation in both AD patients and animal models. Emerging evidence indicates that α7 nicotinic acetylcholine receptors (α7-nAChR) are involved in AD pathology, because synaptic impairment and learning and memory deficits in a hAPPα7-/- mouse model are decreased by nAChR α7 subunit gene deletion. Given that Aβ potently modulates α7-nAChR function, that α7-nAChR expression is significantly enhanced in both AD patients and animal models, and that α7-nAChR play an important role in regulating neuronal excitability, it is reasonable that α7-nAChRs may contribute to Aβ-induced neural hyperexcitation. We hypothesize that increased α7-nAChR expression and function as a consequence of Aβ exposure is important in Aβ-induced neural hyperexcitation. In this project, we found that exposure of Aβ aggregates at a nanomolar range induces neuronal hyperexcitation and toxicity via an upregulation of α7-nAChR in cultured hippocampus pyramidal neurons. Aβ up-regulates α7-nAChRs function and expression through a post translational mechanism. α7-nAChR up-regulation occurs prior to Aβ-induced neuronal hyperexcitation and toxicity. Moreover, inhibition of α7-nAChR or deletion of α7-nAChR prevented Aβ induced neuronal hyperexcitation and toxicity, which suggests that α7-nAChRs are required for Aβ induced neuronal hyperexcitation and toxicity. These results reveal a profound role for α7-nAChR in mediating Aβ-induced neuronal hyperexcitation and toxicity and predict that Aβ-induced up-regulation of α7-nAChR could be an early and critical event in AD etiopathogenesis. Continues characteristics of active and passive parts of the network where it connects certification, ending a general guide for network architects, which can be used to understand the theory of progress in the design and implementation of data networks with regard to its functionality and reliability. This section explaining how the proposal looks like a LAN, which is one of the most widespread types of networks, and most standards it is just related. The practical part is focused on the demonstration of a possible case design, including its economic parameters. The various alternatives are worked out with regard to the choice of materials offered different levels of quality and possible types of treatment for the future. The alternatives are as follows economically analyzed and evaluated.

The nature and origins of beat-to-beat variability in the heart : in vivo to single cells

Introduction: Beat-to-beat variability in cycle length exists in spontaneously beating cardiac preparations of varying complexities from the level of the isolated whole heart to the single sinoatrial nodal cell (SANC). The nature of this variability is poorly characterised as are its fundamental physiological origins. Methods: Recordings of spontaneous electrical activity were made from hearts in vivo, during Langendorff-perfusion, and from single SANC. Heart rate variability (HRV) was calculated in the time- and frequency-domains at baseline and in response to pharmacological mediators that interfered with critical processes involved in automaticity (catecholamines, carbachol, ivabradine, zatebradine, ryanodine and thapsigargin). In addition, a novel 2D technique for imaging Ca2+ fluorescence in spontaneously beating, fluo4-AM loaded, patched single sinoatrial nodal cells was developed to investigate the biophysical behaviour of Ca2+ during pacemaking to see if variability in this was responsible for SANC HRV. Results: Under baseline, temperature-stable conditions, levels of HRV were greatest in vivo (human > rat). SANC exhibited slightly lower levels of HRV, whereas HRV levels expressed by Langendorff-perfused hearts were the least (rabbit > rat), although still comprised a significant proportion of the variability witnessed in vivo. Anaesthetising in vivo rabbits decreased HRV to levels similar to those seen in the Langendorff-perfused heart. HRV was decreased by catecholamines and by ryanodine/thapsigargin in the Langendorff heart. Conversely, HRV was increased by carbachol, ivabradine, zatebradine and ryanodine in SANC. Heart rate changes had a marked effect on levels of HRV. 2D Ca2+ imaging of SANC showed that diastolic local Ca2+ releases (LCRs) occurred earlier than previously thought, with early LCRs having characteristics that were distinct from later LCRs. Mean time of occurrence of all the LCRs within a given diastole closely predicted the duration of the cycle. The rate of restitution of the whole cell Ca2+ transient (used as a surrogate for the pumping function of SERCA) in turn closely predicted the mean time of occurrence of LCRs. Tight synchronisation of the electrical activity of the cell with the biophysical behaviour of Ca2+ appeared to predict shorter cycle lengths. Isoprenaline increased LCR amplitude, though did not increase LCR number, size or duration. Isoprenaline caused LCRs to occur earlier, and synchronised their occurrence and the rate of pumping of Ca2+ back into the sarcoplasmic reticulum. Finally, LCRs were found to preferentially recur in certain regions of the cell, dubbed hotspots. Isoprenaline favoured hotspot production. Conclusion: Whilst greatest in vivo, significant HRV exists in spontaneously beating cardiac preparations devoid of a functioning autonomic nervous system. Studies in SANC indicate that the origin of this is likely to be variability in release of LCRs from the SR via ryanodine receptors. This in turn is controlled by SR refilling kinetics via SR Ca2+ pumping. The coupled system of membrane- and Ca2+-pacemaker clocks are so heavily intertwined that myriad factors will come to bear on generating such variability, including the amount of Ca2+ available for pumping and the phosphorylation state of key proteins, to the extent that variability in no one process can take the credit for generating such HRV.

O papel de sistemas agroflorestais para a conectividade em paisagem fragmentada do nordeste do Rio Grande do Sul

Esse estudo teve como objetivo comparar duas formas de manejo de cultivos de banana em relação ao seu potencial de abrigar espécies florestais. Nós assumimos que o registro de espécies florestais utilizando um elemento da matriz mostra que a espécie não evita a matriz totalmente e pode ser capaz de cruzá-la, indicando a importância dessas estruturas para a manutenção de conectividade funcional entre as áreas de hábitat. Foram amostrados 30 cultivos incluindo áreas sob manejo convencional e agroflorestal. Em cada unidade amostral foram realizadas transecções para acessar a proporção de aves florestais, generalistas e de áreas abertas. Variáveis descritoras da estrutura interna e da estrutura da paisagem do entorno dos cultivos foram consideradas para verificar se a avifauna responde à características intrínsecas dos cultivos ou ao contexto de paisagem em que os cultivos estão inseridos. A proporção de espécies florestais foi maior em cultivos agroflorestais (F28;0,001=19,29; p<0,0004), enquanto a proporção de generalistas não apresentou diferença significativa (F28;0,001=0,22; p<0,22) e a de espécies de áreas abertas foi maior em cultivos convencionais (F28;0,001=8,92; p<0,006). Os dois conjuntos de variáveis considerados, isto é, a estrutura interna do cultivo e do entorno deste foram importantes para predizer a proporção de espécies florestais. Embora tenhamos observado o efeito da paisagem na proporção de espécies florestais, salientamos aqui que sistemas convencionais, mesmo em contextos de paisagem similares à sistemas agroflorestais, não apresentaram a mesma proporção de espécies florestais. Esse resultado indica um diferencial de cultivos agroflorestais de banana para a ocorrência de espécies florestais. Em nível de paisagem, a proporção de florestas e consequentemente a diminuição do isolamento entre manchas é o responsável pela presença potencial de espécies em fragmentos. No entanto, em nível de mancha, mesmo esta apresentando um contexto de paisagem favorável para que seja ocupada por espécies de áreas fonte, o mecanismo que influencia a presença das espécies é a elevada estrutura interna dos bananais. / This study aimed to compare two management of banana crops in relation to its potential to host forest bird species. We assume that the record of forest bird species using a matrix element shows that the species does not entirely avoid this area and may be able to cross it, indicating the importance of these structures for the maintenance of functional connectivity between areas of habitat. We sampled 30 areas including crops under conventional and agroforestry management. In each sampling unit we used transects to access the composition and abundance of bird species. Descriptive variables of the internal structure and the structure of the surrounding landscape of crops were considered to determine whether the bird responds to the intrinsic characteristics of the crops or the landscape context in which crops are inserted. The proportion of forest species was higher in agroforestry crops (F28, 0,001 = 19.29, p <0.0004), while the proportion of generalists showed no significant difference (F28, 0,001 = 0.22, p <0.22) and species of open areas was greater in conventional crops (F28, 0,001 = 8.92, p <0.006). The two sets of variables considered, i.e. the internal structure of the crop and of surroundings, were important to predict the proportion of forest birds species. Although we observed the effect of landscape in the proportion of forest species, we note here that conventional systems, even in contexts similar to the landscape agroforestry systems, did not show the same proportion of forest species. This result indicates a upper value of agroforestry crops for the occurrence of forest bird species. At landscape level, the proportion of forests and consequently the lower isolation between patches is responsible for the potential presence of species in fragments. Traumatic Brain Injury Induces Rapid Enhancement of Cortical Excitability in Juvenile Rats

abstract: Following a traumatic brain injury (TBI) 5-50% of patients will develop post traumatic epilepsy (PTE). Pediatric patients are most susceptible with the highest incidence of PTE. Currently, we cannot prevent the development of PTE and knowledge of basic mechanisms are unknown. This has led to several shortcomings to the treatment of PTE, one of which is the use of anticonvulsant medication to the population of TBI patients that are not likely to develop PTE. The complication of identifying the two populations has been hindered by the ability to find a marker to the pathogenesis of PTE. The central hypothesis of this dissertation is that following TBI, the cortex undergoes distinct cellular and synaptic reorganization that facilitates cortical excitability and promotes seizure development. Chapter 2 of this dissertation details excitatory and inhibitory changes in the rat cortex after severe TBI. This dissertation aims to identify cortical changes to a single cell level after severe TBI using whole cell patch clamp and electroencephalogram electrophysiology. The work of this dissertation concluded that excitatory and inhibitory synaptic activity in cortical controlled impact (CCI) animals showed the development of distinct burst discharges that were not present in control animals. The results suggest that CCI induces early "silent" seizures that are detectable on EEG and correlate with changes to the synaptic excitability in the cortex. The synaptic changes and development of burst discharges may play an important role in synchronizing the network and promoting the development of PTE. / Dissertation/Thesis / Masters Thesis Biology 2014

Development and modelling of new wideband microstrip patch antennas with capacitive feed probes

The principal contributions of this study include the development of a new capacitive feeding mechanism for wideband probe-fed microstrip patch antennas as well as the implementation of a spectral-domain moment-method formulation for the efficient analysis of large, but finite arrays of these elements. Such antenna configurations are very useful in the wireless communications industry, but extremely difficult to analyse with commercially available software. Probe-fed microstrip patch antennas have always been a popular candidate for a variety of antenna systems. Due to their many salient features, they are well suited for modern wireless communication systems. However, these systems often require antennas with wideband properties, while an inherent limitation of probe-fed microstrip patch antennas is its narrow impedance bandwidth. This can be overcome by manufacturing the antenna on a thick low-loss substrate, but at the same time it also complicates things by rendering the input impedance of the antenna very inductive. In this thesis, a new capacitive feeding mechanism is introduced that can be used for probe-fed microstrip patch antennas on thick substrates. It consists of a small probe-fed capacitor patch that is situated next to the resonant patch. The benefits of this configuration include the fact that only one substrate layer is required to support the antenna. It is also very easy to design and optimise. The use of full-wave methods for an accurate analysis of microstrip antennas, has basically become standard practice. These methods can become very demanding in terms of computational resources, especially when large antenna arrays have to be analysed. As such, this thesis includes a spectral-domain moment-method formulation, which was developed for the analysis of probe-fed microstrip patch antennas or antenna arrays that comprise of the new capacitive feeding mechanism. Here, entire-domain and subdomain basis functions are combined in a unique way so as to minimise the computational requirements, most notably computer memory. It is shown that, for general antenna array configurations, memory savings of more than 2500 times can be achieved when compared with typical commercial software packages where only subdomain basis functions are used. Some of the numerical complexities that are dealt with, include various methods to evaluate the spectral integrals as well as special algorithms to eliminate the recalculation of duplicate interactions. The thesis also contains a quantitative comparison of various attachment modes that are often used in the moment-method modelling of probe-to-patch transitions. Various numerical and experimental results are included in order to verify the spectral-domain moment-method formulation, to characterise the new feeding mechanism and to illustrate its use for various applications. These results show that, in terms of accuracy, the spectral-domain moment-method formulation compares well with commercial codes, while by comparison, it demands very little computer memory. The characterisation results show that the input impedance of the antenna can be fully controlled by only adjusting the size of the capacitor patch as well as the width of the gap between the capacitor patch and the resonant patch. In terms of applications, it is shown how the new antenna element can effectively be employed in linear arrays with vertical polarisation, horizontal polarisation or dual slant-polarisation. Inward rectifier potassium channel Kir2.1 is located in muscular, heart and nerve cells and its dysfunction causes various diseases. Practical part of this stage is focused on cultivation of the HEK293T cell line that is used to transfection of the plasmid Kir2.1 and subsequent measurement of the ionic current through the electrophysiological method patch-clamp in whole-cell mode.
