The used methodological base was a Bibliographical Revision that it allowed a dialogue of the main themes and, a Research of Field, where we took place open interviews and collection of information in municipal and federal government institutions. The field research had as last end the characterization of the area and the rebuilding of the process of creation of APA - Alter of the Ground through oral report and of participant observation. It was looked for, to leave of those methods, to reach the objectives ends. Such objectives are related with the understanding attempt of as the process of creation of an Area of Environmental Protection - APA, in the case APA - Alter do Ch?o, can be or no seen as inspiration for the maintainable development with freedom in a given area, taking into account aspects political, economical, social and environmental that you/they permeate such process. That process that, in all of the levels, it is sustained starting from the relationship society-nature and starting from the faith that Unit of Conservation - UC of Maintainable Use, as APA's and the development can be configured as possible relationships since there is a perception that the society has freedom and she needs to acquire capacities to do choices, making possible to the members of this to reach a landing in that you/they recognize that his/her survival and of other generations they depend on these choices. In this sense, they are two the main results of this study: the first is related to the perception that APA's while strategies of environmental public politics can be seen as means of making possible the relationship society-nature starting from a democratic participation process in that the main involved actors agree and stimulated her use the same ones in the search of the development. And, according to the positive vision of the development, harnessing it to processes that unchain creation of UC's of Maintainable Use, like APA - Alter do Ch?o. / Esta disserta??o teve como fundamento o estudo de caso: a cria??o APA Alter do Ch?o, situada na regi?o do Eixo Forte, no distrito de Alter do Ch?o, em Santar?m, Par?, Amaz?nia. A base metodol?gica utilizada foi uma Revis?o Bibliogr?fica que permitiu um di?logo das principais tem?ticas referenciadas e, uma Pesquisa de Campo, onde se realizou entrevistas abertas e coleta de informa??es em institui??es governamentais municipais e federais. A pesquisa de campo teve como fim ?ltimo a caracteriza??o da ?rea e a reconstitui??o do processo de cria??o da APA Alter do Ch?o atrav?s de relato oral e de observa??o participante. Five soil profiles were selecred in two toposequences: profile (CAM1), occurring on the relief’s high part (top); on backslope (CAM2 and CAM4) and on foot slope (CAM3 and CAM5). Profile CAM6 is located on a different area from the others, namely the sandy region. The soils were described morphologically and submited to granulometric, chemical, and mineralogical analyses. The profiles presented A-B and A-C horizon sequences. CAM1 with granular structure and very small blocks; and CAM2 with medium-sized blocks structure, reveal water dynamics influence in the transformation from Bw to Bt. Profile CAM1 (top) presents a very clayey texture, just as CAM2 e CAM4 profiles related with a colluvium mantle, present médium- clayey texture. Profile CAM3, from mixed colluvium and alluvium sediments, dysplay glerization colors and medium/sandy texture, similar to that of CAM5 profile. CAM6 profile testure is typical to the sandy domain. Soils were moderately to highly acidic, allic and dystrofic, with ki ratio around 2.0, indicating a high extent of weathering. The mineralogical examination recorded kaolinite with a high crystallinity degree in every soil. Profiles were classified as Yellow Latosol (CAM1), Yellow Argisol (CAM2 and CAM4), Fluvic Neosol (CAM3), Cambisol (CAM5), and Quarzipsammentic Neosol (CAM6). / Caracterizações morfológicas, físicas, químicas e mineralógicas foram realizadas em solos derivados de sedimentos da Formação Alter do Chão que ocupam o campus da Universidade Federal do Amazonas (UFAM), com o objetivo de subsidiar o entendimento dos processos de formação e de classificação desses solos, Foram selecionados em duas toposseqüências, seis perfis de solo: o perfil (CAM1), ocorrendo na parte alta do relevo (topo); na encosta (CAM2 e CAM4) e na baixada (CAM3, CAM5 e CAM6). Os solos foram descritos morfologicamente e submetidos às análises granulométricas, químicas e mineralógicas. Os perfis apresentaram seqüências A-B e A-C CAM1 com estrutura granular e muito pequenos blocos; e CAM2, com estrutura de médios blocos, evidenciam influência da dinâmica da água na transformação Bw para Bt. O perfil CAM1 (topo) tem textura muito argilosa, bem como, os perfis CAM2 e CAM4 relacionados com uma rampa de colúvio, têm texturas média/argilosas. O perfil CAM3, originado de sedimentos colúvio-aluvionais, mostra cores de gleização e textura média/arenosa, similar à textura do perfil CAM5. A textura do perfil CAM6 é típica do domínio arenoso. Os solos foram moderados e fortemente ácidos, álicos e distróficos, com Ki em torno de 2,0, indicando alto grau de intemperismo. O exame mineralógico registrou caulinita com elevado grau de cristalinidade, em todos os solos. Os perfis foram classificados como Latossolo Amarelo (CAM1), Argissolo Amarelo (CAM2 e CAM4), Neossolo Flúvico (CAM3), Cambissolo (CAM5), Neossolo Quartzarênico (CAM6).

Differential Effects of Gram-positive and Gram-negative Inflammatory Stimuli on the Expression and Function of Energy Substrate Transporters in Human Mammary Epithelial cells

Mastitis is often bacterial in origin. Lipoteichoic acid (LTA) and lipopolysaccharide (LPS), endotoxins from gram-positive and gram-negative bacteria, respectively, are potent inducers of mammary gland inflammation. Inflammation can alter expression of transporters responsible for transport of substrates important in synthesis of milk constituents and cellular metabolic energy. Since, gram-positive and gram-negative bacterial infections cause a different clinical course of mastitis, I investigated whether LTA and LPS differentially alter proton-coupled (MCT1) and sodium-coupled monocarboxylate transporter (SMCT1, SMCT2) expression and functional outcomes of altered expression. Human mammary epithelial cells (MCF-12A) were incubated with 1 microgram/mL LPS or LTA for 6, 12 and 24 hours and mRNA expression of TNF-alpha, IL-1β, IL-6, MCT1, SMCT1, and SMCT2 were measured using Quantitative RT-PCR. LPS decreased SMCT1, but increased SMCT2 expression after 6 h, while LTA increased MCT1 expression at 6 h, followed by gradual decrease in expression until 24 h. To know whether such differential changes in transporter expression by LPS and LTA could cause changes in cellular energy production, I quantified creatine (Cr) and high-energy phosphate substrates (CrP, ATP, ADP, AMP) and oxygen consumption rates using HPLC and Hansatech oxygen electrode, respectively. At 12 h, LPS increased concentrations of Cr, CrP, ATP and ADP, whereas LTA caused changes in CrP and ADP concentrations relative to control. Both LPS and LTA decreased oxygen consumption rates after 12 h. Furthermore, to know whether changes in transporter expression lead to differences in substrate availability, I performed uptake studies for carnitine using radiolabelled tritium L-carnitine. LPS and LTA challenge did not affect the affinity, but caused a 2-3-fold increase in maximal activity (Vmax) of carnitine transport. Although increases in Vmax were not significant, the increase in Vmax after 12 h exposure by LPS and LTA corresponds to changes in mRNA expression of the OCTN2 transporter (previously reported in the laboratory). In conclusion, LPS and LTA differentially alter mRNA expression of transporters, which leads to changes in cellular energy levels and oxygen consumption rates and possibly to changes in the functional activity of transporters. Whether such differences contribute to the different clinical course of mastitis warrants further investigation.

The Confidant as the Alter-Ego of the Protagonist in the Principal Tragedies of Racine

The thesis states that the confidant in the tragedies of Jean Baptist Racine evolves from the traditional servant figure to a sophisticated intimate of the principal character. The confidant's identity becomes synonymous with that of the principal character: he appears as his alter ego. The sources used are six of Racine's secular tragedies, in addition to critical works and essays of his writings. The tragedies included in this study are La Thebaide, whose secondary characters serve as a comparison to the more developed confidants as found in Andromaque, B /r/nice, Mithridate, Britannicus, and Phedre. Racine presents a variety of tragic characters whose multifaceted personality emerges through the intervention of their confidant. Representing one side of the protagonist's character, or his "other self, " the confidant becomes Racine's dramatic tool to portray the internal struggle in all its aspects. Racine's preoccupation with moral issues and his desire to instruct his audience pervade his writings. It is thus possible to trace the development of the confidant from his part as self-effacing messenger to his role as alter ego to the principal figure where he dramatically demonstrates the tragic, inner division of man.
