Efeito de Bacillus thuringiensis e produtos fitossanitários sintéticos na longevidade de operárias Apis mellifera L. (Hymenoptera: Apidae)

As abelhas africanizadas (Apis mellifera L.) estão desaparecendo e acredita-se que seja devido ao intenso uso de inseticidas, poluição e alterações climáticas, no entanto muitas culturas dependem da polinização realizada por essas abelhas. Desta forma, o objetivo deste trabalho foi avaliar o efeito de três linhagens de Bacillus thuringiensis (Bt) (IPS 82, BR 81 e BR 147), entomopatógeno utilizado para o controle de lagartas desfolhadoras, e de três produtos fitossanitários sintéticos (PFS 1: Acetamiprido + Alfa-cipermetrina; PFS 2: Imidacloporido + Beta-Ciflutina e PFS 3: Fenitrotiona + Esfenvalerato), sobre a longevidade de operárias adultas de A. mellifera africanizada. Foram realizados três bioensaios com as linhagens de Bt e dos PFSs: A) Pulverização sobre A. mellifera; B) Contato de A. mellifera com superfície pulverizada e C) Pasta cândi incorporada com os produtos e análise histológica do intestino médio das abelhas. Nos três bioensaios a testemunha foi composta por água deionizada esterilizada. Cada tratamento foi composto por 60 abelhas, sendo cada abelha considerada uma repetição. Os bioensaios foram mantidos em câmara climatizada tipo BOD (34 ± 2ºC, U.R. de 60 ± 5%) e a longevidade das operárias avaliada a uma; duas; três; quatro; cinco; seis; nove; 12; 15; 18; 21; 24; 30; 36; 42; 48; 60; 72; 96; e 120 horas.Na pulverização de Bt sobre A. mellifera verificou-se que apenas a linhagem Bt 1 (IPS 82) causou redução da longevidade de operárias (88,7 horas), enquanto no bioensaio de contato a linhagem Bt 3 - BR147 provocou redução (101,2 horas). Todas as linhagens de Bt avaliadas, quando incorporadas à alimentação de A. mellifera reduziram a longevidade das operárias (Bt 1 – IPS 82: 64,50 horas; Bt 2 – BR 81: 64,56 horas e Bt 3 – BR 147: 60,00horas) e com a análise histológica pôde-se perceber a desintegração das células epiteliais do mesêntero das abelhas, causada pelas bactérias que elas ingeriram. Os três produtos fitossanitários sintéticos reduziram a longevidade das operárias de A. mellifera, no bioensaio de pulverização direta (PFS 1 e PFS 2 – 1,0 hora; PFS 3 – 2,21 horas). No contato de A. mellifera com superfície pulverizada com PFS, as abelhas ficaram duas horas em contato com os produtos na placa de Petri, e ao final dessas duas horas, 100% abelhas que entraram em contato com os três produtos fitossanitários sintéticos estavam mortas. Quando incorporados os PSFs na alimentação, ambos reduziram a longevidade das operárias (PFS 1 – 22 horas, PFS 2 – 21,72 horas e PFS 3 – 43,93 horas) e na análise histológica foi possível observar diferenças na estruturação do intestino médio das operárias de A. mellifera alimentadas com os três PFSs. Apesar das linhagens de Bt reduzirem a longevidade das operárias de A. mellifera não é possível afirmar que não sejam seletivas, pois a interferência é mínima quando comparada à interferência provocada pelos PFSs, os quais provocaram drástica redução na longevidade de A. mellifera. / Africanized bees (Apis mellifera L.) are disappearing and are believed to be due to the intense use of insecticides, pollution and climate change, however many crops depend on the pollination performed by these bees. The objective of this work was to evaluate the effect of three strains of Bacillus thuringiensis (Bt) (IPS 82, BR 81 and BR 147), entomopathogen used for the control of Acetamiprid + Alpha-cypermethrin, PFS 2: Imidacloporide + Beta-Cyflutin and PFS 3: Fenitrothione + Esfenvalerate), on the longevity of African female A. mellifera workers. Three bioassays were performed with the Bt and PFS lines: A) Spraying on A. mellifera; B) A. mellifera contact with pulverized surface and C) Candi paste incorporated with the products and histological analysis of the medium intestine of the bees. In the three bioassays the control was composed of sterilized deionized water. Each treatment consisted of 60 bees, each bee being considered a repetition. The bioassays were maintained in a BOD type chamber (34 ± 2ºC, 60 ± 5% U.R) and the workers longevity was evaluated at one; two; three; four; five; six; nine; 12; 13; 18; 21; 24; 30; 36; 42; 48; 60; 72; 96; And 120 hours. In Bt spraying on A. mellifera, it was verified that only the Bt 1 strain (IPS 82) caused a reduction in worker longevity (88.7 hours), while in the contact bioassay the Bt 3 - BR147 strain caused a reduction (101, 2 hours). All strains of Bt evaluated when incorporated into A. mellifera feed reduced workers longevity (Bt 1 - IPS 82: 64.50 hours; Bt 2 - BR 81: 64.56 hours and Bt 3 - BR 147: 60). And the histological analysis showed the disintegration of the epithelial cells of the bees mesentery, caused by the bacteria they ingested. The three synthetic phytosanitary products reduced the longevity of A. mellifera workers in the direct spray bioassay (PFS 1 and PFS 2 - 1.0 hour; PFS 3 - 2.21 hours). At the contact of A. mellifera with a PFS-sprayed surface, the bees stayed two hours in contact with the products on the petri dish, and at the end of these two hours, 100% bees that came in contact with the three synthetic phytosanitary products were dead. When the PSFs were incorporated in the diet, they reduced the longevity of the workers (PFS 1 - 22 hours, PFS 2 - 21.72 hours and PFS 3 - 43.93 hours) and in the histological analysis it was possible to observe differences in the workers from A. mellifera fed with the three PFSs. although the Bt lineages reduce the longevity of A. mellifera workers, it is not possible to assert that they are not selective, since the interference is minimal when compared to interference caused by PFSs, which caused a drastic reduction in the longevity of A. mellifera.

RESUMO As abelhas africanizadas (Apis mellifera L.) apresentam um comportamento higiênico mais intenso comparado a outras subespécies de abelhas europeias. Embora o comportamento higiênico das abelhas já seja bem conhecido, ainda existem estudos a serem feitos em torno desse complexo comportamento. Assim, no presente trabalho pretendeu-se obter subsídios para uma melhor compreensão desse comportamento em relação ao comportamento de forrageamento e ao desenvolvimento interno das colônias, através da comparação do desempenho entre colônias de linhagens higiênicas e não higiênicas de abelhas africanizadas (Apis mellifera L.). Portanto, quarenta e uma colônias de abelhas Africanizadas Apis mellifera L. foram testadas quanto ao comportamento higiênico (CH). O presente trabalho foi desenvolvido no Laboratório Apilab do Departamento de Genética da FMRP-USP. Foram constituídos dois grupos de colônias: um grupo de 3 colônias higiênicas (H) com CH igual ou superior a 90% e 3 colônias não higiênicas (NH) com valor igual ou inferior a 55%, com o objetivo de se estudar, mediante monitoramento mensal durante 13 meses (julho de 2011 a julho de 2012), as variáveis relacionadas ao desenvolvimento interno das colônias (atividade de postura, área de cria aberta e de cria operculada), atividades forrageiras (área de pólen, área de néctar aberto e de néctar operculado) e ganho de peso. Para o ganho de peso foram utilizadas balanças eletrônicas adaptadas. Os mapeamentos dos dados climáticos foram registrados por um período na Estação Climatológica do Apilab na USP, porém devido a um acidente com queda de energia a fonte foi queimada, não sendo possível o uso programado dos dados climáticos para estudos de correlação com as variáveis das colônias. Os dados das variáveis internas da colônia foram obtidos utilizando-se de um suporte de madeira com tela de arame dividindo a área total em 36 quadrantes idênticos dentro do qual se colocava o quadro da colmeia para se estimar as áreas respectivas em % (néctar, pólen, crias, oviposição etc.). Os dados foram transformados em arco-seno para aplicação dos testes estatísticos. As comparações estatísticas foram realizadas usando-se o teste t-Student e análises de correlação pelo método de Spearman, (Software Statistica 8). Como principais resultados obtivemos os seguintes: as colônias H tiveram melhor desempenho que as NH quanto as atividades de coleta de pólen (P = <0,001), área de cria aberta (P = <0,001), área de cria operculada (P = 0,050) e oviposição das rainhas (P = 0,015). As colônias H apresentaram maior taxa de remoção de crias doentes e mortas que as NH. As abelhas das colônias H são melhores coletoras de pólen que as NH. Não houve diferença entre as linhagens quanto ao ganho de peso. Encontramos correlação positiva significante nas colônias (H e NH) entre áreas de pólen com as áreas com cria aberta H (r = 0,599; P = 0,029), NH (r = 0,791; P = 0,000) e entre as áreas de pólen com cria operculada H (r = 0,659; P = 0,013), NH (r = 0,731; P = 0,004), confirmando que o feromônio das larvas estimula a coleta de pólen. Houve também correlação positiva significante nas colônias H entre área de néctar operculado e cria operculada (r = 0,714; P = 0,005), e néctar operculado e pólen (r = 0,659; P = 0,013). Concluiu-se que o CH pode também ser utilizado como uma característica para critério de seleção para produção de pólen, sendo o CH considerado como uma das melhores alternativas para os programas de melhoramento de abelhas. / The africanized bees (Apis mellifera L.) present a more intensive hygienic behavior in comparison with another European subspecies bees. Although the hygienic behavior of these bees is already very known, there are studies being conducted on this complex behavior. Then, the present study aimed to obtain data to comprehend better the hygienic behavior in relation to the forager behavior and to the internal development of colonies through the comparison of development between colonies of A. mellifera bees from lineage hygienic and not hygienic. Thus, forty-one colonies of Africanized honey bee (Apis mellifera L.) were tested for hygienic behavior. This work was developed in the Apilab of the Department of Genetics, FMRP-USP in Ribeirão Preto-SP. There were two groups of colonies: a group of 3 hygienic colonies (H) ( H test => 90% ) and 3 non-hygienic colonies (NH) ( H test =< 55%) , with the objective of studying, through monitoring monthly from July/ 2011 to July/ 2012, variables related to the internal development of the colonies (oviposition area, open brood area and capped brood area) foraging activities (pollen area, open nectar area, capped nectar area) and weight gain. For weight gain determination adapted electronic scales were used. Mappings of climatic data were recorded for a period at the Apilab-USP, but due to an accident the Climatological Station was damaged, not being possible the correlation studies. Data of internal variables of the colonies were obtained using a wooden support mesh wire dividing the total area under 36 identical quadrants within which is placed the frame of the hive to estimate the respective areas in % (nectar, pollen, brood, eggs etc.). Data were transformed to arcsine in statistical tests. Statistical comparisons were performed using the Student t test and correlation analysis by the method of Spearman (Software Statistica 8). As main results we achieved the following: H colonies outperformed the NH in the following variables: pollen area (P = <0.001), open brood area (P = <0.001), capped brood area (P = 0.050) and oviposition (P = 0.015). Hygienic colonies (H) showed higher removal rate of sick and dead brood than Non hygienic colonies (NH). The Africanized Honey Bees of the Hygienic colonies ( H ) are better pollen-collecting than the Honey Bees of the Non-hygienic colonies (NH). There was no difference between strains regarding weight gain. Significant positive correlation was found in both groups of colonies (H and NH) between area of pollen with open brood H (r = 0.599; P = 0.029), NH (r = 0.791; P = 0.000) and between areas of pollen with capped brood H (r = 0.659; P = 0.013), NH (r = 0.731; P = 0.004), confirming that the larval pheromone stimulates pollen collection. There were also significant positive correlation in the Hygienic colonies (H) between area of capped brood and capped nectar area (r = 0.714; P = 0.005) as well as between pollen area and capped nectar area (r = 0.659; P = 0.013). It was also concluded that the Hygienic Behavior of the Honey Bees can also be used as a feature selection for pollen production. The Hygienic Behavior feature is being considered today as one of the best alternatives for Honey Bee breeding programs.

Avaliação de formulações alimentares no desenvolvimento de colônias de abelhas africanizadas (Apis mellifera Linnaeus, 1758) na savana amazônica de Roraima

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / As abelhas africanizadas encontram nas flores suas principais fontes de alimento, sendo possível observar que em condições ambientais desfavoráveis há uma diminuição na população e conseqüentemente uma menor produção de mel nas épocas de florada. Em Roraima a produção de mel esta concentrada após o período de chuvas, motivo pelo qual é importante o fornecimento de uma suplementação alimentar durante as chuvas. O objetivo deste trabalho foi analisar o desenvolvimento de colônias de abelhas africanizadas A. mellifera com diferentes suplementações alimentares, para manter e/ou melhorar a produção apícola. A pesquisa foi realizada de 12 de abril a 5 de agosto de 2011 no Apiário Experimental do departamento de Zootecnia da Universidade Federal de Roraima. Foram utilizadas vinte colônias em caixas modelo Langstroth, suplementadas com alimentação energética e escolhidas aleatoriamente para receberem três diferentes rações isoprotéicas com 23% de proteína bruta. Os quatro tratamentos foram: T1 = testemunha (sem suplementação alimentar protéica); T2 = farelo de milho + farelo de soja tostada + farelo de arroz (em quantidades iguais); T3 = 30% de farelo de soja tostada + 19% de leite de soja desidratado + 20% farelo de milho + 31% açúcar cristal; T4 = 45% de farelo de soja + 40% farinha de pupunha + 15% açúcar cristal. O desempenho das dietas foi determinado pelo consumo do alimento protéico e o desenvolvimento das colônias quanto ao ganho de peso, áreas de cria e áreas de alimento, sendo que foram analisadas separadamente áreas abertas e fechadas de operárias e zangões, além de áreas com alimento (cm). O consumo de alimento e o ganho de peso foram obtidos por meio de pesagens; as áreas ocupadas nos favos foram analisadas por meio de fotografias digitais e processadas com ajuda do programa computacional Adobe Photoshop CS2. Foram encontradas diferenças significativas para o consumo de ração e o ganho de peso, sendo que, o maior consumo de ração foi observado nas colônias do tratamento 4 e o maior ganho de peso nas colônias do tratamento 1, que receberam unicamente alimentação energética e apresentaram as maiores áreas de cria e alimento. Em relação às variáveis ambientais houve correlação negativa da temperatura e umidade com o ganho de peso das colônias do tratamento 2. / Africanized bees find in the flowers their main source of food; as a result, it has been shown a reduction in population under unfavourable environmental conditions and, consequently, a lower honey production during the early spring. In Roraima, the highest honey production rates occur after the rainy season, the reason for which is important the bees be given supplement during the rainy period. This research was carried out to analyze the development of Africanized honeybee A. mellifera colonies according to different kinds of food supplements offered in order to maintain and/or increase the apicultural production rates. The research was conducted from April 12th to August 5th, 2011, in the Experimental Apiary of Zootechny Department of Federal University. It has been used twenty Langstroth beehives, provided with energetic food and chosen at random to be given three different isoproteinaceous feed containing 23% of raw protein. The four treatments were: T1 = witness (without any proteinaceous supplement); T2 = corn flour + toasted soybean flour + rice flour (in equal parts); T3 = 30% of toasted soybean flour + 19% of soybean dry milk + 20% corn flour + 31% crystal sugar; T4 = 45% of soybean flour + 40% of peach palm fruit flour+ 15% of crystal sugar. The diets performances were determined by the consumption of the proteinaceous feed; the development of the colonies, in turn, as regards weight gain, brood area and food area, being analyzed separately the uncapped and capped worker and drone areas, as well as the food area (cm). The food consumption and the weight gain were obtained by weighing; the honeycomb area was analyzed by taking digital photographs of it and processing the data using the software Adobe Photoshop CS2. It has been found significant differences for feed consumption and weight gain, showing the colonies of T4 the highest feed consumption, and the colonies of T1, which were only given energetic food, the highest weight gain; it has also been observed in the latter the biggest brood and food areas. It has occurred a negative correlation of temperature and humidity with the weight gain in one of the colonies of T2.

O princípio básico era ofertar ninhos alternativos para os apicultores que trabalham com abelhas africanizadas (Apis mellífera). Para o cálculo do custo unitário de cada ninho foram considerados os preços atuais das matérias primas, bem como as suas depreciações anuais. Constatou-se uma economia de até 30% na construção dos ninhos alternativos quando comparados aos custos dos ninhos tradicionais de madeira, mostrando-se uma opção viável e rentável ao apicultor. Em relação ao comportamento das abelhas, concluiu-se que o tipo de material (concreto ou madeira) utilizado na construção dos ninhos não teve influência significativa na agressividade das mesmas. A defensividade, fortemente atrelada a fatores genéticos, pode ser em função do grau de africanização e do local de instalação dos apiários. / The objective of this work is to analyze the economic feasibility and acceptability of nests Langstroth hives made of concrete-based construction wastes. The basic principle was to offer alternative nests beekeepers working with Africanized honeybees (Apis mellifera). In order to calculate the unit cost of each nest, it was considered the current commodity prices, as well as its annual depreciation. It was found that there were savings of up to 30% in the construction of alternative nests when compared to the costs of traditional wooden nests, which can be seen as a viable and profitable option to the beekeeper. As for the behavior of the bees, it was concluded that the type of material (concrete or wood) used in the nest construction had no significant influence on the bee aggressiveness. Defensiveness, strongly linked to genetic factors, may be a function of the Africanization level and location of installed apiaries.

Although this mite is unlikely to, by itself, cause the mortality of hives, it plays an important role as a vector for many viral diseases. These diseases are identified as some of the most important reasons for the Colony Collapse Disorder. The effects of V.destructor infestation are disparate in different parts of the world. Greater morbidity - in the form of colony losses - has been reported in colonies of European honey bees (EHB) in countries of Europe, Asia and North America. However, this mite has been in Brasil for many years and there are no reports of losses of Africanized honey bees (AHB) colonies. Studies carried out in Mexico showed that some resistance behaviors to the Varroa mite - especially grooming and hygienic behavior - appear to be different in each subspecies. Could those mechanisms explain why the AHB are less susceptible to Colony Collapse Disorder? In order to answer this question, we propose a mathematical model based on differential equations, to analyze the role of these resistance mechanisms in the overall health of the colony, and it’s ability to face environmental challenges. / Os ácaros ectoparasitas Varroa destructor, que parasitam as abelhas tornaram-se um problema global. Embora seja pouco provável que estes ácaros, por si só, provoquem a mortalidade das colmeias, eles desempenham um importante papel como vetor de muitas doenças virais. E estas doenças são identificados como algumas das mais importantes razões para a Desordem do Colapso das Colônias. Os efeitos da infestação do V.destructor são distintas em diferentes partes do mundo. Maiores mortalidades de colônias têm sido relatadas em colônias de abelhas européias (AE) em países da Europa, Ásia e América do Norte. No entanto, este ácaro está presente no Brasil já por muitos anos e não existem relatos de perdas em colônias das abelhas africanizadas (AA). Estudos realizados no México mostraram que alguns comportamentos de resistência ao ácaro Varroa - especialmente o grooming e o comportamento higiênico - são diferentes em cada uma das subespécie. Poderiam então esses mecanismos explicar por que as abelhas africanizadas são menos suscetíveis à Desordem do Colapso das Colônias? A fim de responder a esta pergunta, propomos um modelo matemático baseado em equações diferenciais, com o objetivo de analisar o papel desses mecanismos de resistência na saúde geral da colônia e na capacidade da colônia para enfrentar desafios ambientais.
