High-Frequency Ultrasound Drug Delivery and Cavitation

The viability of a drug delivery system which encapsulates chemotherapeutic drugs (Doxorubicin) in the hydrophobic core of polymeric micelles and triggers release by ultrasound application was investigated at an applied frequency of 500 kHz. The investigation also included elucidating the mechanism of drug release at 70 kHz, a frequency which had previously been shown to induce drug release. A fluorescence detection chamber was used to measure in vitro drug release from both Pluronic and stabilized micelles and a hydrophone was used to monitor bubble activity during the experiments. A threshold for release between 0.35 and 0.40 in mechanical index was found at 70 kHz and shown to correspond with the appearance of the subharmonic signal in the acoustic spectrum. Additionally, drug release was found to correlate with increase in subharmonic emission. No evidence of drug release or of the subharmonic signal was detected at 500 kHz. These findings confirmed the role of cavitation in ultrasonic drug release from micelles. A mathematical model of a bubble oscillator was solved to explore the differences in the behavior of a single 10 um bubble under 70 and 500 kHz ultrasound. The dynamics were found to be fundamentally different; the bubble follows a period-doubling route to chaos at 500 kHz and an intermittent route to chaos at 70 kHz. It was concluded that this type of "intermittent subharmonic" oscillation is associated with the apparent drug release. This research confirmed the central role of cavitation in ultrasonically-triggered drug delivery from micelles, established the importance of subharmonic bubble oscillations as an indicator, and expounded the key dynamic differences between 70 and 500 kHz ultrasonic cavitation.

Korrelation von Elastizitätsmodul und Ermüdungsschädigung von Straßenbeton

Gegenstand dieser Dissertation ist die Etablierung des Elastizitätsmoduls als Parameter, der qualitative Aussagen über den Schädigungszustand des Baustoffs Straßenbeton ermöglicht. Zu diesem Zweck erfolgte eine systematische Ermüdung von labormaßstäblichen Betonprobekörpern bei zeitgleicher Messung des Elastizitätsmoduls mit Hilfe von unterschiedlichen Verfahren. Im ersten Schritt wurde ein Versuchsprogramm entwickelt, mit dem Probekörper mittels des Spaltzug-Schwellversuchs gezielt in einen definierten Ermüdungszustand versetzt werden können. Hierfür wurde der Parameter des Grenz-Elastizitätsmoduls definiert, welcher, wenn er unterschritten wird, zum Pausieren des Versuchs führt. In diesen systematisch eingehaltenen Lastpausen erfolgten begleitende Untersuchungen der Ultraschalllaufzeit und der Eigenfrequenz zur Bestimmung des Elastizitätsmoduls der Probekörper während des Ermüdungsvorganges. Es zeigt sich zwischen den Ergebnissen aller untersuchten Verfahren eine sehr gute Synchronität hinsichtlich des qualitativen ermüdungsbedingten Verlaufs des Elastizitätsmoduls. Die vier angewandten Verfahren, die sich voneinander unabhängiger physikalischer Phänomene bedienen, ermöglichen neben einer qualitativen Aussage über die Schädigung des Materials die Bestimmung von Absolutwerten des Elastizitätsmoduls. Je nach verwendetem Verfahren weichen die absoluten Elastizitätsmoduln leicht voneinander ab. Der Elastizitätsmodul bestätigt sich als geeigneter Parameter zur Beschreibung der Degradation des Baustoffs Straßenbeton im Zuge des Ermüdungsprozesses. Im zweiten Schritt wurden der Einfluss längerer Lastpausen sowie die Verminderung der Betonfestigkeit im Zuge der Materialermüdung tiefergehend untersucht. Es wurde festgestellt, dass längere Lastpausen in der zyklischen Belastung einen signifikanten Einfluss auf den ermüdungsbedingten Verlauf des Elastizitätsmoduls haben können. Weiterhin legen die Untersuchungen nahe, dass es im Zuge einer starken Ermüdung zu einer zum Teil signifikanten Verminderung der Festigkeit kommt. Als Resultat der Untersuchungen konnten sowohl ein Verfahren zur Bestimmung charakteristischer Verläufe für die Verminderung des Elastizitätsmoduls als auch ein Verfahren zur Abschätzung der materialspezifischen Verminderung der Festigkeit im Zuge der Ermüdung entwickelt werden. Diese Verfahren könnten zukünftig den Regelwerken der RSO Beton und der RDO Beton zugeführt werden, um die ermüdungsbedingte zeitliche Entwicklung des Elastizitätsmoduls und der Festigkeit in den durch die Regelwerke festgeschriebenen Prognose- und Dimensionierungsprozessen zu berücksichtigen. Durch die Einbeziehung zeitlich veränderlicher Werte im Prognoseverfahren, welches als Grundlage für die RSO Beton dienen soll, wird in dieser Dissertation exemplarisch gezeigt, dass die Berücksichtigung der Auswirkungen der Betonermüdung auf diese für die Prognose und die Dimensionierung von Betonfahrbahnbefestigungen sehr relevanten Parameter zur signifikanten Erhöhung der Ausfallrate gegenüber der Verwendung konstanter Werte führen kann. / The subject of this doctoral dissertation is the establishment of the elastic modulus as a parameter that enables qualitative statements about the state of damage of the concrete pavement building material. For this purpose, a systematic fatigue of laboratory-scale concrete specimens was carried out with simultaneous measurement of the elastic modulus by means of different methods. In the first step, a test program was developed for the targeted and systematic fatigue of concrete specimens by means of the cyclic indirect tensile test. For this purpose, a limit value for the elastic modulus was defined. When the elastic modulus fell below the limit, the test was paused to perform accompanying investigations of the ultrasonic transit time and the natural frequency in order to determine the change of the elastic modulus of the specimens during the fatigue process. There is a very good synchronicity between the results of all investigated methods for the qualitative determination of the elastic modulus. The four methods applied, which make use of physical phenomena that are independent of each other, allow, in addition to a qualitative statement about the damage to the material, the determination of absolute values of the elastic modulus. Although there are slight differences between the methods for the determination of the elastic modulus, it is confirmed as a suitable parameter for describing the degradation of the concrete pavement building material in the course of the fatigue process. In the second step, the influence of longer loading pauses and the reduction of concrete strength due to material fatigue were investigated in more detail. It was found that longer loading pauses during cyclic loading can have a significant influence on the fatigue-related course of the elastic modulus. Furthermore, the investigations suggest that in the course of severe fatigue there is a sometimes significant reduction in strength. As a result of the investigations, both a method for determining characteristic curves for the reduction of the elastic modulus and a method for estimating the material-specific reduction of strength in the course of fatigue could be developed. In the future, these methods could be implemented into the RSO Beton and RDO Beton regulations in order to take into account the fatigue-related temporal development of the elastic modulus and the strength in the forecasting and dimensioning processes specified by the regulations. By including time-varying values in the forecasting procedure, which is to serve as the basis for the RSO Beton, this doctoral dissertation exemplifies that the consideration of the effects of concrete fatigue on these very relevant parameters for the forecasting and dimensioning of concrete pavements, can lead to the significant increase of the failure rate compared to the use of constant values.

Depuis 2005, il existe un intérêt important dans la communauté internationale de métrologiepour de nouvelles déterminations précises de la constante de Boltzmann kB ; lebut étant de redéfinir en 2015 l’unité internationale de température, le kelvin. Actuellement,cinq techniques sont utilisées pour déterminer kB avec comme objectif d’atteindreune incertitude relative inférieure à 1 × 10−6. La méthode retenue pour cette thèse est latechnique acoustique.La constante de Boltzmann est liée à la vitesse du son u dans un gaz parfait par l’équationdu viriel acoustique. La méthode décrite dans cette thèse consiste à mesurer u en utilisantun résonateur de forme quasi sphérique et de volume intérieur de 0,5 L, rempli d’argon.Ces mesures sont effectuées lors d’un isotherme à la température du point triple de l’eau,T = 273,16 K, pour des pressions statiques P allant de 0,05 MPa à 0,7 MPa. La constantede Boltzmann est ensuite déterminée en estimant u à pression nulle par une régressionpolynomiale.Dans cette thèse, un modèle de propagation des ondes acoustiques dans un résonateur quasisphérique est défini. Aussi, les moyens techniques utilisés pour contrôler soigneusement lesparamètres de l’expérience qui ont un effet sur les mesures de u (comme la température,la pression statique, la composition du gaz, etc) sont présentés. De nouvelles techniquesexpérimentales et des nouveaux moyens d’analyse des données sont proposés, comme lamesure du rayon du résonateur par spectroscopie électromagnétique, mais aussi l’utilisationde l’écart-type d’Allan comme un outil efficace pour étudier la présence d’impuretélors d’une expérience de longue durée. Les effets systématiques sont analysés puis corrigés.Pour certains, la correction est estimée grâce à un modèle analytique, comme l’effet lié auxcouches limites thermiques. Pour d’autres, des corrections basées sur des fonctions empiriquessont proposées ; c’est le cas pour l’effet du débit de gaz continu sur les mesures deu, effet qui est caractérisé expérimentalement dans cette thèse.Enfin, l’analyse des données acquises en 2009 au LCM/LNE-CNAM lors de deux isothermeseffectuées avec de l’argon est présentée. Celle-ci a permis d’obtenir la valeur kB =1, 3806475 (16) × 10−23 J · K−1, c’est à dire avec une incertitude relative de 1, 14 × 10−6. / Since 2005, there is an important interest in the international metrology community fornew accurate determinations of the Boltzmann constant kB ; the purpose is to redefine in2015 the unit of thermodynamic temperature, the kelvin. The method used in the present work is based on acoustic measurements.The Boltzmann constant is linked to the speed of sound u in a noble gas by the virial acousticalequation. The method described here consists in measuring u inside a quasi-sphericalacoustic resonator of inner volume of 0.5 L filled with argon. Measurements are performedduring an isotherm process at the temperature of the triple point of water, T = 273.16 K,at static pressures P from 0.05 MPa to 0.7 MPa. The Boltzmann constant is then determinedby estimating u at zero pressure limit with a polynomial regression.In the present work an acoustic wave propagation model within a quasi-spherical resonatoris defined. Also, the technical means used to carefully control the parameters of theexperiment with an effect on the measurement of u (like temperature, static pressure, gascomposition, etc.) are presented. New exprimental methods and data analyses are described,like the measurement of the radius of the resonator by electromagnetic spectroscopy,as well as the use of the Allan deviation as an efficient tool to study the gas impuritypresence during a long-term experience. Systematic effects are analyzed and corrected. Insome cases the corrections are based on analytical models like the thermal layer boundaryeffect. In other cases, empirical correction functions are proposed, as for the case of changesin the measurements of u related to the continuous gas flow, which was experimentally characterizedin the present work.Finally, the analysis of the data acquiered in 2009 at LCM/LNE-CNAM during two isothermprocesses using argon is presented. This leads to the value kB = 1.3806475 (16) ×10−23 J · K−1, i.e. with a relative uncertainty of 1.14 × 10−6.

Monitorování a analýza koroze výztužné oceli v železobetonových prvcích a konstrukcích akustickými metodami / Monitoring and Analysis of Corrosion of Reinforcing Steel in Reinforced Concrete Elements and Structures Using the Acoustic Methods

The dissertation thesis deals with the study of non-destructive acoustic methods as instruments for monitoring and analysing corrosion of reinforcing steel in reinforced concrete elements. Four acoustic methods were selected for this task - the impact-echo method, the nonlinear acoustic spectroscopy method, the acoustic emission method, and the ultrasonic pulse velocity method. To verify the functionality of these methods, testing was carried out on three sets of reinforced concrete samples that had been exposed to the effects of sodium chloride, which corroded the embedded steel reinforcement in these samples. Suitable parameters were proposed for individual acoustic methods to monitor corrosion of the reinforcements. In addition, experiments were designed to demonstrate the ability of the selected acoustic methods to reveal the corrosion of steel reinforcement and its influence on the concrete matrix and to assess the condition of the degraded elements and structures. The analysis of the measurement results based on their comparison shows the advantages and disadvantages of the individual methods and of their practical applications. To verify the results, correlation with common methods that are currently used for the study of corrosion was carried out and included for example the electrical resistivity measurement of the reinforcement and simultaneous monitoring of the sample surface using a confocal microscope to record the development of microcracks during the degradation.
