Desenvolvimento e avaliação de subprodutos de uva e sua utilização como ingrediente alimentício

Entre as frutas, a uva é uma das maiores fontes de compostos fenólicos, sendo que a indústria de derivados de uva gera uma grande quantidade de resíduos que, na maioria das vezes, são desprezados. Estes resíduos concentram quantidade significativa de compostos bioativos, quedespertam o interesse devido a suas propriedades antioxidantes e seus efeitos benéficos para a saúde humana. No intuito de aproveitamento deste subproduto, o presente estudo objetivou elaborar uma farinha utilizando resíduo de uva e analisar suas propriedades bioativas. Foram utilizadas duas técnicas de secagem do resíduo de uva (estufa e liofilizador) para a obtenção de dois tipos de farinhas, uma obtida pelo resíduo proveniente do sistema convencional de cultivo e outra proveniente do sistema orgânico de cultivo. Além disso, parte das uvas havia sido tratada com radiação UV-C. A farinha foi utilizada para elaboração de biscoitos tipo cookies, para avaliar o potencial como ingrediente alimentício e agregação de compostos bioativos no produto. Teores de antocianinas totais, compostos fenólicos totais e atividade antioxidante, expressa em EC50 e em TEAC foram determinados para o resíduo, farinha e biscoitos. Também foram determinados o pH, o teor de cinzas e cor para o resíduo, a farinha e o biscoito Em geral pode-se observar que o tratamento com UV-C apresentou efeito negativo ou nulo nos resultados. A liofilização demonstrou manter mais as propriedades bioativas. O sistema de cultivo convencional demonstrou uma maior manutenção das propriedades bioativas em parte das análises. / Among the fruits, the grape is one of the largest sources of phenolic compounds. the industry of grape spinoffs produces a large amount of waste that most often are underrated. These residues concentrate significant amount of bioactive compounds that arouse interest due to its antioxidant properties and its beneficial effects on human health. In order to take advantage of this by-product, the present study aimed to elaborate flour using residual grape and analyze their bioactive properties. Was used two techniques of drying the residue grape (dryer and freeze dryer) to obtain two kinds of flour, one obtained from the residue from the conventional cultivation and another derived from the organic cultivation. Furthermore, the grapes had been treated with UV-C radiation. The flour was used to prepare cookies, to evaluate its potential as a food ingredient and aggregation of bioactive compounds in the product. The contents of total anthocyanins, phenolic compounds and antioxidant activity, expressed as EC50 and TEAC were determined in the residue, fluor and cookies. Overall treatment with UV-C showed negative or null results. Lyophilization demonstrated longer maintain bioactive properties, as phenolic compounds and anthocyanins. Regarding the type of cultivation, the conventional system in some analyzes showed better maintenance bioactive properties.

Os teores de compostos biativos, caracteristicas fisico-quimicas de quailidade pós-colheita e produtividade, podem diferir entre genótipos e idade da planta. O Vale do Ribeira é uma região de comunidades tradicionais que mantêm diferentes acessos de batata-doce. Este estudo foi realizado de fevereiro a outubro de 2015, o objetivo principal foi realizar a caracterização físico-química e de produtividade de três acessos de batata-doce pertencentes ao Banco de Germplasma da Universidade Estadual Paulista.Registro. As análises do experimento foram feitas nos acessos VR 13-01 (casca roxa e polpa roxa fortemente pigmntada), VR13-11 (casca rosa e polpa branca) e VR13-50 (casca creme e polpa branca). O delineamento experimental foi em blocos casualizados com seis repetições por tratamento e cada parcela tinha 5 plantas.O espaçamento de plantio entre plantas foi de 0,25 m e 0,80 m entre linhas. As avaliações das analises físico-químicas, compostos bioativos e produtividade foram feitas aos 120,150 e 180 dias após o plantio. Foram determinados a produçao total de matéria fresca da parte aérea, produçao total de matária seca da raiz, produtividade total de raizes, produtividade comercial de raízes, produção de raizes não comercias, teor de sólidos solúveis, teor de amido, comteúdo de composto... The starch and sugar content may differ genotypes and harvest season. The Vale do Ribeira region has traditional communities keep different accessions of sweet potato. This study was conducted from February to October 2015, and main objetive was the physico-chemical characterization, bioactive compounds and productivity of three accessions of sweet potato belonging to the Germplasn Bank of Universidad Estadual Paulista. Accession were VR 13-01 (purple skin and strongly purple pulp).VR13-11( lightly purple skin and white pulp) and VR13-50 ( cream skin and white pulp).The experimental design was randomized blocks with six replications per treatment and each plot had 5 plants. The planting distance between plants was 0.25 m and 0.80 m between rows. The biochemical, physico chemical characterization and production yields was made 120,150 and 180 days after planting. The total productions yield, soluble solids content, and starch content were determined and significant differences among accessions obtained by ANOVA and means separated by a Scott-Knott test. Aplicação de cloreto de cálcio em pré-colheita nos frutos de amoreira-preta 'Tupy' /

Objetivou-se avaliar o efeito da aplicação pré-colheita de cloreto de cálcio (CaCl2) na qualidade de frutos da amoreira preta 'Tupy'. Foram avaliadas plantas, cujas mudas foram plantadas em 23/06/2009. Conduziram-se seis hastes mais vigorosas, numa área experimental não irrigada, no sistema de espaldeira em T, com dois arames e com 1,2 metros de altura. O espaçamento foi de 0,6 m entre plantas e 4,0 m entrelinhas e a densidade de plantio de 4.166 plantas ha-1. No experimento o delineamento experimental adotado foi de blocos casualizados em fatorial 4 x 3, sendo o primeiro fator correspondente às concentrações de cloreto de cálcio (0; 1,5; 3 e 4,5 %) e o segundo aos números de aplicações (1, 2 e 3 aplicações). A fase fenológica adotada para a primeira aplicação do cloreto de cálcio foi dos frutos verdes em início de inchamento e ainda aderidos aos restos florais. Quando realizada mais de uma aplicação, havia um intervalo de cinco dias entre uma e outra. Foram realizadas avaliações físicas, físico-químicas, bioquímicas e enzimáticas nos frutos. As the fruits of the black mulberry 'Tupy' plants were evaluated, whose seedlings were planted on 06/23/2009. Development and characterization of peptide antioxidants from sorghum proteins

In recent years, growing interests in developing safe and efficient antioxidants from natural sources due to the health-related risks associated with synthetic antioxidants. Recently, peptide antioxidants have drawn growing interests as since proteins are a macronutrient with various functionalities and high consumer acceptability. A lot of dietary proteins have been validated for their antioxidant potentials especially those obtained from animal proteins, nuts and pulses. Relatively less information is available on characterizing the antioxidant profile of cereal protein, and even less for sorghum protein. Sorghum is the fifth largest crop worldwide and is the third in United States. U.S. is leading in global sorghum production and distribution, and the state of Kansas is producing nearly half of U.S. sorghum. Currently, about one third of the U.S. sorghum is being used for ethanol production, resulting in more than 450 kilotons of by-products (e.g., DDGS) annually, which were often discarded or underutilized. DDGS is a premium protein source (~ 30% protein) that could be potentially modified into value-added products such as peptide antioxidants. In this study, relevant literatures detailing the extraction of cereal proteins, enzymatic hydrolysis of proteins, purification and characterization of hydrolysates, and evaluation of antioxidant profiles were extensively reviewed in Chapter 1. As preliminary experiments, sorghum kafirin protein was extracted from defatted sorghum white flour and hydrolyzed by 10 different types of enzymes from microbial, plant and animal sources. Hydrolysates prepared with Neutrase, Alcalase, and Papain displayed the most promising antioxidant activities as well as total protein recovery were primarily selected and investigated in depth described in Chapter 2, Chapter 3, and Chapter 4. The reaction conditions including substrate content, enzyme-to-substrate ratio, and hydrolysis time are critical parameters in producing peptides with desired activity and consistency, were therefore examined and optimized for each case of kafirin hydrolysates. The antioxidant capacity of the resulting hydrolysates was measured for antioxidant capacity through in vitro assays (DPPH, ABTS, ORAC, reducing power, and metal chelating) and then demonstrated in model systems (oil-in-water emulsion and ground meat). The fractions of hydrolysates possessing strongest activities were further fractionated by gel filtration and HPLC. Peaks representing the largest areas from HPLC were identified for major sequences by MALDI-TOF-MS. The experiment results indicated that all the three selected fractions of kafirin hydrolysates revealed excellent inhibition effects against oil and fat oxidations, which could be employed as tools to predict their performances in real food products. In addition, the structure studies showed that medium-sized hydrolysates of Neutrase (3 – 10 kDa) and Alcalase (5 – 10 kDa), and small-sized hydrolysates of Papain (1 – 3 kDa) exhibited relatively stronger activities. This study provided a workable processing method and critical reaction parameters for the production of peptide antioxidants from sorghum protein. The experiment results revealed that the sorghum peptide antioxidant could act through multiple mechanisms including free radical scavenging, metal ion chelation, hydrogen donating, and forming physical barriers to minimize the contact of oxidative agents to targets. These antioxidative peptides are a promising ingredient that can be potentially incorporated to food and feed products as alternatives to synthetic antioxidants or synergetic elements to nonpeptic antioxidants for protection of susceptible food ingredients. This study also made a positive impact to sorghum ethanol industry by guiding the conversion of sorghum protein-rich by-products into value-added antioxidant products as an additional revenue stream.

Cyanobacterial secondary metabolites are a rich source of bioactive compounds with potential utilization in pharmacology. The aim of this study was to evaluate the effect of the novel compound nostatin A isolated from the cyanobacterium Desmonostoc muscorum. Project was focused on the extraction, purification and characterization of the cytostatic effect caused by this novel compound and its naturally occurred structural analogues. The cytostatic activity of nostatin A and its analogs was evaluated in HeLa cell line. Experiments based on microscopy, flow cytometry and HPLC-HRMS techniques were performed in order to clarify the cytostatic effect of nostatin A in HeLa cells and its mechanism of the action.

Aplicação de um vidro bioativo em tíbias de coelhos. / Bioactive glass application in rabbit tibiae.

Foi estudada a ação de preenchimento e de osteocondução em perfurações em tíbias de coelhos, que receberam a aplicação de um biovidro em pó cuja composição é (teor % m/m): 45,9 SiO2, 23,9 Na2O, 21,6 CaO e 7,54 P2O5. Foram realizadas perfurações nas tuberosidades das tíbias direitas de 24 coelhos, machos, adultos da raça Nova Zelândia, com peso entre 2,6 e 4,0 quilogramas. Foi utilizado sangue do próprio coelho misturado ao vidro para facilitar a colocação do material nas perfurações. Foram realizadas radiografias, microscopia de luz comum, microscopia eletrônica de varredura (MEV) e espectroscopia dispersiva de raios-X buscando revelar a interação do vidro com o osso. A caracterização do material foi realizada por meio das técnicas de análise térmica diferencial, difração de raios-X, MEV e avaliação do potencial de citotoxicidade in vitro do material. Os resultados indicam que o material não sofreu rejeição pelo organismo dos coelhos e que houve formação de osso na região em que foi implantado o material. Foi observada a presença de células ósseas e de tecido osteóide bem irrigados. / In this research were studied both filling and osteoconduction action in rabbit tibiae, which received a powder bioactive glass of de following composition (45,9% SiO2, 23,9% Na2O, 21,6% CaO, 7,54% P2O5, all in wt%). It was carried out in right tibiae tuberosity perfurations of 24 New Zealand white, male, mature rabbits weighing between 2.6 e 4.0 Kg. There was used animal’s autologous blood mixed with bioactive glass to facilitate its placed in tibia. It were observed in tibia tuberosity perfurations findings by means of radiography, light microscopy, SEM and EDS to show bone tissue and bioactive glass interaction. Material characterization was made by chemical composition analysis, SEM, DTA, XRD analysis and evaluation of cytotoxicity potential in vitro. Results indicated that material was biocompatible and acted as a osteoconductive material. Both bone cells and osteid tissue were found with satisfactory blood irrigation.

Diante disso, este trabalho teve o objetivo de determinar o potencial biotecnológico de actinobactérias da coleção UFPEDA contra Candida spp. Para avaliação da atividade biotecnológica, 32 actinobactérias foram testadas contra sete isolados clínicos de Candida spp. utilizando diferentes meios de cultura para o crescimento (ALA, ISP-4, AY, ISP-3) e temperaturas (37 ºC ou 45 ºC). No ensaio primário, foi evidenciada atividade apenas quando as actinobactérias foram cultivadas em meio ISP-3 a temperatura de 37 ºC. Dessas, apenas 2 linhagens (6,25 %) apresentaram atividade antifúngica com halo de inibição variando entre 11 mm e 18 mm. As duas linhagens produtoras de metabólitos secundários com atividades antifúngicas não apresentaram diferença estatística, por isso foi selecionada a linhagem G24 por apresentar um pigmento na cor vermelha para dar seguimento às análises. A fermentação da linhagem (G24) foi realizada para avaliar atividade antifúngica, biomassa e pH, durante 5 dias, utilizando diferentes meios (MPE, M1 e ISP-3),sendo tais parâmetros monitorados a cada 24 horas. O melhor meio para produção de metabólitos secundários foi ISP-3 fermentado durante 48 h em pH 7.0, sendo evidenciados halos de inibição de 13 a 18 mm e biomassa de 0,1 g/mL de meio. Estabelecidas às condições, foi realizada a extração do princípio bioativo, sendo evidenciada atividade apenas para biomassa quando extraído em acetato de etila. A concentração mínima inibitória (CMI) do extrato bruto variou de 125 μg/mL a 62,5 μg/mL para Candida spp testadas. Análise Cromatográfica do Extrato Bruto as frações semi-purificadas foram analisadas por cromatografia em camada delgada (CCD) sendo evidenciadas duas frações, uma com atividade antifúngica e outra um corante natural. As características morfológicas do isolado G24 foram analisadas por microscopia óptica, sendo observados esporos verticilados pertencente ao gênero Streptomyces. O gene 16S rDNA foi amplificado e enviado para sequenciamento, sendo identificada como Streptomyces sp. Diante destes resultados, podemos concluir que a linhagem (Streptomyces sp), isolado da rizosfera de Caesalpinia pyramidalis tul, do bioma Caatinga, apresenta uma significativa atividade antifúngica e necessita de estudos espectroscópios para caracterização do composto bioativo e do corante. / Actinobacteria are forming Gram-positive bacteria of branched filaments that stand out for the production of secondary metabolites such as antibiotics, antitumor, phytohormones and naturias dyes. Given this, this study aimed to determine the biotechnological potential of actinomycetes of UFPEDA collection against Candida spp. To evaluate the biotechnological activity, 32 actinomycetes were tested against seven clinical isolates of Candida spp., Using different culture media (ALA, ISP-4, AY, ISP-3) and temperatures (37 ° C and 45 ° C). In the primary test, activity was detected only when the actinomycetes were grown in ISP-medium 3 to a temperature of 37 ° C. Of these, only 2 strains (6.25%) showed antifungal activity with inhibition zone ranging between 11 mm and 18 mm. Fermentation of strain (G24) was used to evaluate antifungal activity, biomass and pH for 5 days, using different media (MPE M1 and ISP-3), these parameters being monitored every 24 hours. The best medium for production of secondary metabolites ISP-3 was fermented for 48 h at pH 7.0, being evidenced inhibition zones 13 to 18 mm and biomass of 0.1 g / ml medium. Established conditions, the extraction of bioactive principle has been performed and demonstrated activity only when biomass extracted into ethyl acetate. The minimum inhibitory concentration (MIC) of the crude extract ranged from 125 mg / mL to 62.5 mg / mL for Candida spp tested. In chemical prospecting, semi-purified fractions were analyzed by thin layer chromatography (TLC) was evidenced two fractions, one with antifungal activity and one with a natural dye. The morphological characteristics of isolated G24 were analyzed by optical microscopy, observed verticilados spores belonging to the genus Streptomyces. The 16S rDNA gene was amplified and sent to sequencing, being identified as Streptomyces sp. Given these results, we can conclude that the line (G24) (Streptomyces sp), isolated from the rhizosphere of Caesalpinia pyramidalis tul, the Caatinga biome, presents a significant antifungal activity and requires spectroscopes studies to characterize the bioactive compound and the dye.

Novos compostos antifúngicos e antitumorais têm sido propostos para tratamento destas doenças. Assim, o propósito desta pesquisa foi diagnosticar e traçar dados epidemiológicos de fungemia, assim como avaliar a ação antifúngica e antitumoral de isoflavonas da soja (daidzeína e genisteína) e hidroxipiridonas (ciclopirox olamina e octopirox olamina). Foram analisadas amostras de sangue de pacientes internados em hospital universitário de Recife-PE, Brasil e realizado antifungigrama dos agentes etiológicos com fluconazol, anfotericina B, isoflavonas da soja e hidroxipiridonas. Ainda, as isoflavonas e hidroxipiridonas foram avaliadas quanto à citotoxicidade às células neoplásicas (pulmão, laringe e cólon) e a ação da octopirox olamina contra candidemia e tumor sarcoma 180 em análises experimentais. Oito casos de fungemia foram diagnosticados tendo Candida, Histoplasma, Trichosporon, Cryptococcus e um fungo demáceo como agentes etiológicos. A maioria dos pacientes pertencia a idade adulta, sexo masculino e eram portadores da síndrome da imunodeficiência adquirida. Nenhuma das isoflavonas apresentou atividade antifúngica, contudo, a genisteína foi efetiva para câncer de laringe. Para tratamento de neoplasias associadas à candidemia, octopirox olamina pode ser uma alternativa promissora. / Fungemia, presence of fungi in the blood, has a rising incidence in recent decades, causing serious problem in hospitalized patients, especially cancer. New antifungal and antitumor compounds have been proposed for treating these diseases. Thus, the purpose of this research was to diagnose and trace epidemiological data fungemia as well as evaluate the antifungal and antitumour of soy isoflavones (daidzein and genistein) and hydroxypyridones (ciclopirox olamine and octopirox olamine). We analyzed blood samples from patients admitted to a university hospital in Recife-PE, Brazil and performed in vitro the antifungal susceptibility of the etiologic agent fluconazole, amphotericin B, and soy isoflavones hydroxypyridones. Still, isoflavones and hydroxypyridones were evaluated for cytotoxicity to tumor cells (lung, larynx and colon) and the action of candidemia and octopirox olamine against sarcoma 180 tumor in experimental analyzes. Were diagnosed eight cases of fungemia with Candida, Histoplasma, Trichosporon, Cryptococcus and a dematiaceous fungus as etiological agents. Most patients belonged to adulthood, males and were carriers of acquired immunodeficiency syndrome. None of isoflavones showed antifungal activity, however, genistein was effective for larynx cancer. In contrast, hydroxypyridones especially octopirox olamine in already at low concentrations has antifungal and antitumor in vitro and in vivo. For treatment of malignancies associated with candidemia, octopirox olamine may be a promising alternative.
