Caractérisation des agents infectieux responsables de deux maladies à prion : l'ESB atypique de type H chez les bovins et la tremblante de type "CH1641" chez les petits ruminants / Characterization of infectious agents responsible of two different prion disease : the CH1641 scrapie for small ruminants and the atypical H-type BSE for bovine

Nous avons entrepris la caractérisation de la protéine prion pathologique par Western blot à partir d’isolats de petits ruminants et de bovins atteints d’Encéphalopathie Spongiforme Subaiguë Transmissible (ESST) afin d’identifier des signatures moléculaires divergeant de la signature associée à l’ESST affectant habituellement ces animaux ; la tremblante classique pour les petits ruminants et l’ESB classique (ESB-C) pour les bovins. Cette étude a permis d’évaluer la fréquence de phénotypes inhabituels et a contribué à envisager les formes d’ESB atypiques comme sporadiques. Ensuite la transmission des isolats de petits ruminants aux souris C57Bl/6 et TgOvPrP4 a permis d’une part de confirmer l’absence de contamination par l’agent de l’ESB, tout du moins pour les ovins, et d’autre part d’approfondir les connaissances sur la diversité des souches dans les isolats de tremblante. Les travaux de transmission aux souris C57Bl/6 des isolats bovins atteints d’ESB-H ont quant à eux conduit à la description de l’émergence d’une souche présentant les caractéristiques de l’ESB-C suggérant que la forme sporadique d’ESB-H puisse être à l’origine de la contamination initiale des bovins ayant conduit après recyclage par le biais des farines de viande et d’os à l’épizootie d’ESB-C / Molecular characterization using Western blot method has been carried out to identify unusual PrPres pattern from small ruminants and bovines TSE affected. Such analyse allow us to define the frequency of these unusual signature and contribute to consider BSE-H as sporadic form of ruminant TSE. Transmission studies from unusual sheep isolates in C57Bl/6 and TgovPrP4 mice firstly contribute to confirm that none of these sheep isolates was infected by BSE transmission. Secondly, strain characteristics observed through experimental transmission of unusual isolates and classical scrapie affected isolates show results that give some element about strain diversity in both scrapie types. Serial passages of H-BSE in C57Bl/6 mice show the emergence of a prion strain with features similar to classical BSE. Such findings might help to explain the origin of the classical BSE epizootic disease, which could have originated from this kind of sporadic form of BSE

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Este estudo teve como objetivo avaliar o papel da administração crônica de cafeína durante a gestação de camundongos C57BL/6 sobre o remodelamento cardíaco e a expressão de componentes do sistema renina-angiotensina (SRA) na prole macho adulta. Fêmeas C57BL/6 grávidas foram divididas em dois grupos (n = 10): grupo controle (C), progenitoras foram injetadas apenas com o veículo (solução salina NaCl 0,9%), e grupo cafeína (CF), progenitoras receberam diariamente uma injecção subcutânea contendo 20 mg/kg de cafeína (1 mg/ml de solução salina). Após o desmame, os filhotes tiveram livre acesso à ração padrão até 90 dias de idade quando foram sacrificados. Rim e ventrículo esquerdo (VE) foram coletados para análise estrutural e western blotting. O grupo cafeína mostrou uma redução significativa no ganho de massa corporal (MC) (-18%; P <0,0001). O grupo cafeína apresentou ainda um aumento da pressão arterial sistólica (+ 48%; P <0,0001) e freqüência cardíaca (+10%; P <0,01) em relação ao grupo controle. A massa do VE corrigida pela MC no grupo da cafeína foi maior que no grupo C (+10%; P <0,01). O grupo cafeína apresentou um aumento na área de cardiomiócitos (+40%; P <0,05), e reduzida densidade capilar (-25%; P <0,05). No rim, as expressões de renina (128%; P <0,05) e dos receptores 1 da angiotensina II (AT1R) (88%; P <0,05) foram significativamente maiores nos animais do grupo cafeína. No VE, o grupo cafeína demonstrou aumento da expressão de ECA (+30%; P <0,05), angiotensina II (+60%; P <0,01), e AT1R (+77%; P <0,01) e diminuição da expressão do receptor 2 de angiotensina II (-46%; P <0,05). Em conclusão, a administração crônica de cafeína durante a gestação, possivelmente programa a expressão de componentes de sistema renina-angiotensina, levando à ativação persistente do SRA renal e cardíaco local, que por sua vez promove o aumento da pressão sanguínea, remodelação e efeitos cardíacos adversos. / This study aimed to evaluate the role of caffeine chronic administration during gestation of C57BL/6 mice on cardiac remodeling and the expression of components of the renin-angiotensin system (RAS) in male offspring as adults. Pregnant C57BL/6 female mice were divided into two groups (n=10): Control group (C), dams were injected with the vehicle only (saline 0.9% NaCl), and Caffeine group (CF), dams received daily a subcutaneous injection containing 20 mg/kg of caffeine/day (1 mg/ml saline). After weaning, pups had free access to the standard chow until 90 days of age when they were killed. Kidney and left ventricle (LV) were collected for structural analysis and Western blot. Caffeine group showed a significant reduction in body mass (BM) gain (-18%;P<0.0001). Caffeine group had increased systolic blood pressure (+ 48%;P<0.0001) and higher heart rate (+10%;P<0.01) than control group. LV mass adjusted by BM in caffeine group was greater than in C group (+10%;P<0.01). Caffeine group had increase in the area of cardiomyocytes (+40%;P<0.05), and reduced capillary density (-25%;P<0.05). In the kidney, the expressions of renin (+128%; P<0.05) and angiotensin II receptor 1(AT1R) (+88%;P<0.05) were significantly greater in caffeine mice. In the LV, caffeine group showed increased expression of ACE (+30%; P <0.05), angiotensin II (+60%;P<0.01), and AT1R (+77%;P<0.01), and decreased expression of angiotensin II receptor 2 (-46%;P<0.05). In conclusion, chronic administration of caffeine during gestation possibly programs the expression of renin-angiotensin system components, leading to persistent activation of local renal and cardiac RAS, which in turn promotes increased BP and adverse cardiac remodeling.

Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro / O objetivo deste trabalho foi estudar a ação do fenofibrato, um agonista do receptor ativador da proliferação peroxissomal alfa, no remodelamento cardíaco e na expressão de componentes do sistema renina-angiotensina (SRA) em um modelo de obesidade induzida por dieta. Camundongos machos C57Bl/6 com três meses de idade foram alimentados durante 11 semanas com dieta controle (grupo C, 3,57 kcal/g de dieta) ou dieta hiperlipídica (grupo HL, 5,40 kcal/g de dieta), em seguida foram separados em quatro grupos e estudados durante cinco semanas: C; HL; C-L (C mais fenofibrato) e HL-F (HL mais fenofibrato). Os animais HL foram mais pesados e apresentaram maior pressão arterial (PA) comparados aos animais C, mas HL-F foram mais leves e tiveram PA menor que HL. A resistência insulínica vista nos camundongos HL foi melhorada com fenofibrato nos camundongos HL-F. Fenofibrato reduziu colesterol total, triglicerídeos e aumentou HDL-c. Os animais HL apresentou um ventrículo esquerdo (VE) mais pesado e com espessura da parede maior, como também cardiomiócitos maiores e uma menor razão cardiomiócito/capilares que os animais C. Fenofibrato foi eficiente em melhorar estas alterações. As expressões cardíacas de Angiotensina II (ANG II) e de seu receptor tipo 1 (AT1R) foram maiores, enquanto que a expressão de seu receptor tipo 2 (AT2R) foi menor nos animais HL que nos animais C, e fenofibrato foi eficiente em atenuar estas diferenças. Como conclusão, a dieta HL lidera para a obesidade, elevação da PA, hipertrofia cardíaca, alterações metabólicas e expressão proteica alterada do SRA em camundongos, sugerindo a participação do SRA nestas alterações. HF had thicker and heavier left ventricle (LV) with bigger LV cardiomyocyte and smaller cardiomyocyte-to-capillaries ratio than SC, and fenofibrate was efficient in treating these alterations. Cardiac expressions of angiotensin II (ANG II) and ANG II receptor 1 were higher, and ANG II receptor 2 was lower in HF than in SC, and fenofibrate was efficient in attenuating these differences. In conclusion, HF diet leads to obesity, BP elevation, cardiac hypertrophy, metabolic changes and altered RAS protein expression in mice, suggesting that RAS is involved. Fenofibrate is efficient in decreasing BP and in controlling RAS protein expressions, and treats the insulin resistance and the adverse cardiac remodeling decreasing the cardiomyocyte hypertrophy and improving the myocardial vascularization, therefore, decreasing important cardiovascular risk factors

Dieta hiperlipídica materna e pós-natal promove remodelamento adverso do fígado, pâncreas e tecido adiposo na prole / Maternal and postnatal high fat diet provoke adverse liver, pancreas and adipose tissue remodelling in offspring

A dieta hiperlipídica (high-fat, HF) materna durante a gestação e/ou lactação aumenta a susceptibilidade da prole para o desenvolvimento de doenças crônicas na fase adulta. Verificar a hipótese que a ingestão materna de dieta HF nos períodos críticos de desenvolvimento (gestação e/ou lactação) predispõe à doença não alcoólica do fígado gorduroso e alterações pancreáticas e no tecido adiposo de camundongos machos adultos. Camundongos C57BL/6 fêmeas receberam durante a gestação e/ou lactação dieta padrão (standard chow, SC) ou HF. Filhotes machos foram divididos em cinco grupos: SC provenientes de mães SC; G provenientes de mães HF durante a gestação; L provenientes de mães HF durante a lactação; GL/HF provenientes de mães HF durante a gestação/lactação, mantendo a mesma dieta HF no período pós-natal (do desmame aos 3 meses deidade); GL provenientes de mães HF durante a gestação/lactação trocando a dieta para SC no período pós-natal (do desmame aos 3 meses deidade). Foi analisada ao longo do experimento a massa corporal da prole. No sacrifício (3 meses), o fígado, o pâncreas e a gordura epididimária foram removidos, pesados e processados e o sangue foi coletado para análise bioquímica. Ao nascimento e ao desmame, filhotes GL/HF foram mais pesados (+6% e +44%, p<0,05, respectivamente) que os filhotes SC. Os filhotes G apresentaram resistência à insulina e menor expressão do transportador de glicose no fígado (GLUT-2). A esteatose hepática foi observada nos grupos G, L, GL e principalmente nos filhotes do grupo GL/HF. A expressão hepática da proteína ligante de elementos regulatórios de esteróis (SREBP-1c) estava aumentada nos filhotes G, GL e GL/HF. Os filhotes G, GL e GL/HF apresentaram hipertrofia da ilhota pancreática e dos adipócitos quando comparados com o grupo SC. O consumo de dieta HF durante a gestação mostra-se ser o período mais prejudicial para os filhotes adultos de camundongos. A programação metabólica por dieta HF leva ao remodelamento adverso do fígado, do pâncreas e do tecido adiposo / Maternal high-fat diet (HF) during gestation and/or lactation period increases the susceptibility to development of chronic disease in offspring adult life. This work aimed to verify the hypothesis that maternal intake of high-fat diet in critical periods of pregnancy and/or suckling period predisposes to non alcoholic fatty liver disease, pancreatic and adipose tissue alterations in adulthood mice offspring. C57BL/6 female mice were fed, during gestation and/or lactation phases, with standard chow (SC) or HF diet. Male pups were divided into 5 groups: SC- from SC fed dam; G- from HF fed dam during gestation period; L- from HF fed dam during lactation period; GL- from HF fed dam during gestation and lactation periods and GL/HF- from HF fed dam during gestation and lactation, maintaining HF diet from post-weaning to adulthood. We analyzed body mass in all experiment, and at the euthanasia (3 mo-old), liver, pancreas and adipose tissue were removed, weighted and embedded. Blood was collected to biochemical analyses. At birth and at weaning, GL/HF pups were heavier than SC pups (+6% and +44%, p<0.05, respectively). G offspring showed insulin resistance and lower glucose transporter-2 expression (GLUT-2). Hepatic steatosis was present in G, L, GL and mainly in GL/HF offspring. Sterol regulatory element-binding protein-1c (SREBP-1c) expression was higher in G, GL and GL/HF offspring. It is important to mention that pancreatic islet hypertrophy and adipocyte hypertrophy were affected in G, GL and GL/HF offspring in comparison to SC. HF diet administration during gestation period is worse than lactation period. Furthermore, this type of programming by HF predisposes to adverse remodeling in liver, pancreas and adipose tissue in adult mice offspring

Ao nascimento e ao desmame, filhotes GL/HF foram mais pesados (+6% e +44%, p<0,05, respectivamente) que os filhotes SC. Os filhotes G apresentaram resistência à insulina e menor expressão do transportador de glicose no fígado (GLUT-2). A esteatose hepática foi observada nos grupos G, L, GL e principalmente nos filhotes do grupo GL/HF. A expressão hepática da proteína ligante de elementos regulatórios de esteróis (SREBP-1c) estava aumentada nos filhotes G, GL e GL/HF. Os filhotes G, GL e GL/HF apresentaram hipertrofia da ilhota pancreática e dos adipócitos quando comparados com o grupo SC. O consumo de dieta HF durante a gestação mostra-se ser o período mais prejudicial para os filhotes adultos de camundongos. A programação metabólica por dieta HF leva ao remodelamento adverso do fígado, do pâncreas e do tecido adiposo / Maternal high-fat diet (HF) during gestation and/or lactation period increases the susceptibility to development of chronic disease in offspring adult life. This work aimed to verify the hypothesis that maternal intake of high-fat diet in critical periods of pregnancy and/or suckling period predisposes to non alcoholic fatty liver disease, pancreatic and adipose tissue alterations in adulthood mice offspring. C57BL/6 female mice were fed, during gestation and/or lactation phases, with standard chow (SC) or HF diet. Male pups were divided into 5 groups: SC- from SC fed dam; G- from HF fed dam during gestation period; L- from HF fed dam during lactation period; GL- from HF fed dam during gestation and lactation periods and GL/HF- from HF fed dam during gestation and lactation, maintaining HF diet from post-weaning to adulthood. We analyzed body mass in all experiment, and at the euthanasia (3 mo-old), liver, pancreas and adipose tissue were removed, weighted and embedded. Blood was collected to biochemical analyses. At birth and at weaning, GL/HF pups were heavier than SC pups (+6% and +44%, p<0.05, respectively). G offspring showed insulin resistance and lower glucose transporter-2 expression (GLUT-2). Hepatic steatosis was present in G, L, GL and mainly in GL/HF offspring. Sterol regulatory element-binding protein-1c (SREBP-1c) expression was higher in G, GL and GL/HF offspring. It is important to mention that pancreatic islet hypertrophy and adipocyte hypertrophy were affected in G, GL and GL/HF offspring in comparison to SC. HF diet administration during gestation period is worse than lactation period. Furthermore, this type of programming by HF predisposes to adverse remodeling in liver, pancreas and adipose tissue in adult mice offspring

Modeling diarrheagenic E. coli infections and co-infections: specific roles of diet and pathogen

Enteric pathogens such as Enteroaggregative E. coli (EAEC), Enteropathogenic E. coli (EPEC) and Enterotoxigenic E. coli (ETEC) have been reported to cause diarrhoea in children under the age of 5 years. The incidences of these pathogens are due to factors such as poor water quality, sanitation and hygiene practices. Infections with these pathogens result in diarrhoea and have been reported to result in severe disease outcomes more especially in children under 2 years of age. EPEC infections have been well studied using in vitro analyses, with studies highlighting the adherence traits, proteins and virulence genes involved in pathogenesis and inflammatory responses. EPEC is characterized by localized adherence with microcolony formation at the site of infection. In vivo studies have reported on human EPEC infection. However, the current animal models have not been able to replicate clinical outcomes (such as diarrhoea and weigh loss) of EPEC infection similar to humans. Therefore, there is still a need for a suitable small animal model that mimic clinical outcomes of human EPEC infections in vivo. Children living in poor environmental conditions are more susceptible to diarrhoeal pathogens. Furthermore, the incidences of children being exposed to co-infections (more than one pathogen at the same time) is relatively high. The EAEC/EPEC (A/P) and EPEC/ETEC (P/T) co-infections have been increasingly detected in children with and without diarrhoea. It has been suggested that patients infected with these co-infections might result in severe disease outcome than those infected with single pathogens. Pathogens are constantly evolving and the microbe-microbe interaction in the host can result in these pathogens competing for the same niche and thus result in increased virulence. Interaction of co-infections can lead to increased inflammatory responses thus affecting the infected host. The first objective of this study was to develop an EPEC murine model using weaned C57BL/6 mice that have been pretreated with antibiotic cocktail. Mice were orally infected with wild-type (WT) typical EPEC, bfp- and escN mutant strains. The WT had transient weight loss and wet stools with mucous; and the bfp- infected mice also had transient weight loss and bloody stool appearance. Increase in inflammatory biomarkers MPO, LCN-2, CRP, IL-6 and SAA were observed in the WT and bfp- infected mice. The mice infected with escN mutant did not exhibit any weight changes and the stools were similar to the uninfected mice. Furthermore, no inflammatory biomarkers were observed in mice infected with the escN mutant. Metabolic perturbations were observed in WT EPEC infected mice at day 3 post infection with the TCA cycle metabolites (reduced succinate, citrate, fumarate, cis-aconitate) being excreted at lower quantities indicating that the energy production in the infected mice was greatly affected. The second objective of this study was to determine the interaction between the P/T coinfections using in vitro and in vivo analyses. In vitro, human colorectal tumour 8 (HCT-8) cells were infected with single strains of ETEC, EPEC and both the pathogens and incubated for 3 hours. After infection the cells were analysed for bacterial adherence using real-time PCR. The single strains adhered at the same rate similar to the P/T coinfected cells. IL-8, as a marker of inflammatory response, was measured using ELISA. The results indicated that the P/T co-infected cells had a significant increase in IL-8 response higher than the single infections. The P/T co-infections were further analysed in vivo using the EPEC murine model developed in this study. Interestingly, mice infected with P/T co-infections developed severe diarrhoea accompanied with significant increased weight loss and some mice died during the 3-day infection period. The inflammatory responses MPO, LCN-2 and SAA were higher in the co-infected mice indicating a synergistic effect. The bfp and eltA virulence genes were significantly increased in the P/T co-infections. The third objective of this study was to determine the interaction between A/P coinfections using in vitro and in vivo analyses. HeLa cells and HCT-8 cells were infected with EAEC, EPEC and both the pathogens at the same time in order to determine adherence and inflammatory responses. EAEC adherence was higher than EPEC and A/P co-infections adherence. A/P co-infections did not have increased IL-8 response in HCT-8 cells when compared to EAEC alone. The virulence genes involved in EPEC adherence and Type 3 Secretion System (bfp, eae, tir, ler, per, espB and espA) were significantly reduced in A/P co-infected cells. An interesting adherence trait was observed between the A/P co-infections in HeLA cells, EAEC was found to adhere around EPEC altering the localized adherence pattern. The A/P co-infections were further analysed using the EPEC murine model developed in this study. The A/P infected mice had diminished weight changes and EAEC shedding was enhanced when EPEC was present. Faecal inflammatory biomarkers MPO and LCN-2 in A/P infected mice did not have any additive effect. The findings of this study contributed significantly to the knowledge of human EPEC infection in weaned C57BL/6 mice, highlighting clinical outcomes, inflammatory responses and metabolic perturbations. Furthermore, this study also highlighted the interaction of P/T and A/P co-infections using in vitro and in vivo analyses in order to determine the disease severity and outcomes. It was observed in this study that coinfections can result in either synergistic or antagonistic effects. Further studies are therefore, required in order to understand the underlying mechanisms that are involved during co-infections and this can further assist in the development of therapeutic interventions. / NRF

Antiobesity and antidiabetic activity of P. balsamifera, its active Salicortin, and L. laricina, medicinal plants from the traditional pharmacopoeia of the James Bay Cree

La prévalence de l’obésité, du diabète de type 2, et du syndrome métabolique, sont à la hausse chez les Cris d’Eeyou Istchee (CEI-Nord du Québec). Ces problèmes sont aggravés par leur diète non traditionnelle, leur sédentarité, ainsi que par une résistance culturelle aux produits pharmaceutiques. Afin de développer des traitements antidiabétiques culturellement adaptés, notre équipe a effectué une enquête ethnobotanique qui a identifié 17 plantes provenant de la pharmacopée traditionnelle des CEI. À partir des études de criblage effectuées in vitro, deux plantes parmi les 17 ont attiré notre attention. Populus balsamifera L. (Salicaceae) pour ses propriétés anti-obésité et Larix laricina K. Koch (Pinaceae) pour ses propriétés antidiabétiques. P. balsamifera et son composé actif salicortin ont inhibé l’accumulation de triglycérides durant l’adipogénèse dans les adipocytes 3T3-L1. L. laricina a augmenté le transport de glucose et l’activation de l’AMPK dans les cellules musculaires C2C12, l’adipogénèse dans les 3T3-L1 et a démontré un fort potentiel découpleur (propriété anti-obésité). Les objectifs de cette thèse sont d'évaluer les potentiels anti-obésité et antidiabétique et d’élucider les mécanismes d'action de P. balsamifera, salicortin, et L. laricina chez la souris C57BL/6 rendue obèse par une diète riche en gras (HFD). Les souris ont été soumises pendant huit (étude préventive) ou seize semaines (étude traitement) à une HFD, ou à une HFD dans laquelle P. balsamifera, salicortin, ou L. laricina a été incorporé soit dès le départ (prévention), ou dans les 8 dernières des 16 semaines d'administration de HFD (traitement). iv Les résultats démontrent que P. balsamifera (dans les deux études) et salicortin (évalué dans l’étude traitement) diminuent: le poids corporel, le gras rétropéritonéal, la sévérité de la stéatose et l’accumulation de triglycérides hépatique (ERK impliqué), les niveaux de glycémie et d'insuline, et le ratio leptine/adiponectine. Dans les deux études, P. balsamifera a significativement réduit la consommation de nourriture mais cet effet coupe-faim nécessite d’être approfondi. Dans l'étude préventive, P. balsamifera a augmenté la dépense énergétique (hausse de la température à la surface de la peau et de l’activation de la protéine découplante-1; UCP-1). Les voies de signalisation activées par P. balsamifera et par salicortin (de façon plus modeste) sont impliquées dans: la production de glucose hépatique (Akt), l’expression de Glut4 dans le muscle squelettique, la captation du glucose et du métabolisme des lipides (Akt dans le tissu adipeux), la différenciation des adipocytes (ERK et PPARg), l’inflammation dans le foie (IKKαβ), et l'oxydation des acides gras dans le muscle, le foie, ou le tissu adipeux (PPARa et CPT-1). D’autre part, L. laricina a également diminué les niveaux de glycémie et d’insuline, le ratio leptine/adiponectine, le gras rétropéritonéal et le poids corporel. Ces effets ont été observés en conjonction avec une augmentation de la dépense énergétique: hausse de température à la surface de la peau (prévention) et amélioration de la fonction mitochondriale et de la synthèse d'ATP (traitement). En conclusion, l’utilisation de P. balsamifera, salicortin et L. laricina comme des traitements alternatifs et culturellement adaptés aux CEI représente une contribution importante dans la prévention et le traitement de l’obésité et du diabète. / The prevalence of obesity, insulin resistance, and the metabolic syndrome is increasing among the Cree of Eeyou Istchee (CEI - Northern Quebec). Non-traditional diet and sedentary lifestyle along with cultural disconnect of modern type 2 diabetes (T2D) therapies are involved. In order to establish culturally adapted antidiabetic treatments, our research team conducted an ethnobotanical survey, where 17 plants were identified from the CEI traditional pharmacopoeia. Based on data obtained from in vitro screening studies, two plant species out of 17 were of particular interest for their properties as antiobesity, namely Populus balsamifera L. (Salicaceae), and antidiabetic agents, namely Larix laricina K. Koch (Pinaceae). P. balsamifera and its active salicortin inhibited triglyceride accumulation during adipogenesis in 3T3-L1 adipocytes. L. laricina increased glucose uptake and AMPK activation in C2C12 myotubes, adipogenesis in the 3T3-L1 adipocyte cell line, and was observed as one of the strongest uncouplers, severely disrupting mitochondrial function (increasing fuel consumption/metabolic rate; antiobesity property). The purpose of this PhD thesis is to evaluate the antiobesity and antidiabetic potential of P. balsamifera, salicortin, and L. laricina, in an in vivo model of diet-induced obese (DIO) C57BL/6 mice, as well as to investigate their possible mechanisms of action. Mice were subjected for eight (prevention study) or sixteen weeks (treatment study) to a high fat diet (HFD), or HFD to which P. balsamifera, salicortin, or L. laricina were incorporated either at onset (prevention), or in the last 8 of the 16 weeks of administration of the HFD (treatment). The results showed that P. balsamifera (in either study) and salicortin (incorporated in HFD only in treatment study) decreased the weight of whole vii body, retroperitoneal fat pad, reduced the severity of hepatic macrovesicular steatosis and triglyceride accumulation (ERK pathway implicated). They also decreased glycemia and improved insulin sensitivity by diminishing insulin levels, and altering adipokine secretion whereby reducing the leptin/adiponectin ratio. In both studies, P. balsamifera significantly reduced food intake. This appetite-reducing effect needs to be investigated further. In the prevention study this was accompanied by an increase in energy expenditure (increase in skin temperature and tends to increase expression of uncoupling protein-1; UCP-1). The signaling pathways activated by P. balsamifera and slightly by salicortin are implicated in either controlling hepatic glucose output (Akt), skeletal muscle Glut4 expression, glucose uptake and lipid metabolism in adipose tissue (Akt), adipocyte differentiation (ERK pathway and PPARg), decreasing the hepatic inflammatory state (IKKab), and increasing muscular, hepatic, or adipose tissue fatty acid oxidation (PPARa, CPT-1). As for L. laricina, it effectively decreased glycemia levels, insulin levels and the leptin/adiponectin ratio, improved insulin sensitivity and slightly decreased abdominal fat pad and body weights. This occurred in conjunction with increased energy expenditure as demonstrated by elevated skin temperature in the prevention study, and tendency to improve mitochondrial function and ATP synthesis in the treatment protocol. In conclusion, these results represent a major contribution, identifying P. balsamifera, salicortin, and L. laricina, as promising alternative, and culturally adapted therapies for the prevention and treatment care of obesity and diabetes among the CEI.
