Production et caractérisation de farine de patate douce (Ipomoeabatatas.Lam) : optimisation de la technologie de panification / Production and characterisation of sweet potato (Ipomoea batatas Lam) flours : optimisation of the technology of breadmaking

La patate douce est un tubercule appartenant à l'ordre des polémoniales et à la famille des convolvulacées. Il contient de l’amidon, qui est l’élément principal, des dextrines, du ß-carotène, puis d’autres éléments nutritifs en quantités variables. L’objectif de la thèse a été de valoriser la patate douce sous forme de farines et de mettre en évidence les potentialités technologiques de ces farines. Deux études complémentaires ont été réalisées au cours de ce travail de thèse, la première concerne l’étude de l’impact des prétraitements thermiques et chimiques sur les propriétés physicochimiques, thermiques, rhéologiques et fonctionnelles des farines issues de patates douces ayant : TPD (tranche de patate douce non prétraitée), TPDCaCl2 (tranche de patate douce prétraitée au CaCl2) et enfin TPDB (tranche de patate douce prétraitée par blanchiment). Il en résulte une légère augmentation de la teneur en sucres réducteurs dans les farines TPDCaCl2 par rapport aux farines TPDB et TPD. Cependant, la valeur de L* (luminance) dans la farine TPDB était inférieure à celles des farines TPDCaCl2 et TPD. La teneur en calcium était relativement élevée dans la farine TPDCaCl2 (120,6 mg/100g) par rapport aux farines TPD et TPDB (respectivement, 77,8 et 67,6 mg/100g). Les valeurs d’enthalpie de gélatinisation et de module élastique (G’) étaient supérieures dans la farine TPDCaCl2. La capacité de gonflement était élevée dans les farines blanchies. Il a également été constaté une légère augmentation de la constante d'énergie de liaison d'eau du modèle BET dans les farines traitées au CaCl2 et une augmentation marquée pour les farines blanchies. De cette étude, on déduit que les farines traitées au CaCl2 possèdent les propriétés physicochimiques les plus intéressantes pour une application en panification et dans la formulation alimentaire. Dans une seconde étude, les essais de panification ont été réalisés avec différents taux d’incorporation à la farine de blé (0, 10, 15, 20, 25 et 30 %) des farines de patate douce traitées au CaCl2 afin de déterminer le taux d’incorporation optimal permettant de réaliser un compromis entre l’amélioration des propriétés nutritionnelles et la diminution des propriétés rhéologiques de la pâte à pain, suite à l’incorporation de farine de patate douce. Les paramètres texturaux des pâtes ont été déterminés. Il en résulte une diminution de la cohésion, la valeur de a* (échelle de couleurs rouge-vert) a augmenté avec l’augmentation du taux de substitution, contrairement aux valeurs de la luminance L* qui ont diminué. Finalement, la farine de patate douce pouvait être incorporée dans une formulation de panification jusqu’à 25 % sans dépréciation notable des qualités organosensorielles des pains / Sweet potato is an edible tuber belonging to the order of Solanales and Convolvulaceae family. It contains starch, which is the main component, dextrins, ß-carotene, and other nutrients in varying amounts. The thesis aimed promoting the use of sweet potato as flour by highlighting its nutritional potential. Two complementary studies were carried out in this thesis work, the first concerned the study of the impact of thermal and chemical pretreatments on physicochemical, thermal, rheological, and functional properties of sweet potatoes flours obtained after different pretreatments : TPD (unpretreated sweet potato slices), TPDCaCl2 (CaCl2- pretreated sweet potato slices) and finally TPDB (bleached sweet potato slices). These results show a slight increase in reducing sugar content in TPDCaCl2flour compared to TPDB and TPD flours. However, the value of L* (luminance) in the TPDB flour was below the ones ofTPDCaCl2 and TPD flours. The calcium content was relatively high in TPDCaCl2 flour (120.6 mg / 100 g) compared to TPD and TPDB flours (respectively, 77.8 and 67.6 mg / 100g). The gelatinization enthalpy and elastic modulus (G’) were higher in the TPDCaCl2 flour. The swelling capacity was high in bleached flour. It was also noticed a slight increase in the water binding energy constant of the BET model in CaCl2-pretreated and blanched. In this study, it was deduced that the flours pretreated with CaCl2 have physicochemical properties interesting for bakery and food formulation applications. In the second part of the thesis study, baking tests were carried out with different levels of incorporation to wheat flour (0, 10, 15, 20, 25 and 30 %) of the CaCl2-pretreated sweet potato flour in order to determine the optimal incorporation level allowing the compromise between improved nutritional properties and the decreased rheological properties of the dough, due to the sweet potato flour incorporation. Textural parameters of doughs were determined. This resulted in a decrease in cohesion and an increase in the value of a* (red - green color scale) with increasing the degree of substitution, in contrast to the luminance L * values which felt. XYLEM FLOW IN CUT ACACIA HOLOSERICEA STEMS

Acacia holosericea A. Cunn. Ex G. Don (Velvet Leaf Wattle, Family Mimosaceae) is indigenous to Australia and holds promise as a novel cut foliage crop due to its silvery green silky phyllodes. Insufficient water uptake, possibly due to low stem hydraulic conductivity (Kh), is potentially responsible for early wilting and desiccation of phyllodes and limiting vase life. This study aimed to characterize the anatomy of stem xylem conduits and determine cation (K+ and Ca2+) mediated stem Kh. Differential localization of Ca2+ in xylem vessels and the effects of KCl and CaCl2 salts as cation contributors in vase solutions were also evaluated for their effects on cut foliage longevity. Anatomical characteristics of stem xylem conduits were studied using light, scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Tracheids and vessels with simple perforation plates were the principal water conducting cells. SEM and TEM revealed bordered vestured intervessel pits in xylem conduits. Vestures were branched and coralloid-like structures. Xylem conduit lengths were assessed by ink perfusion. The majority of conduits (89%) were short (1 to 5 cm), and only ~ 2% were long (15 to 16 cm). Mean conduit diameter was 77 ± 0.9 µm, with 29% of conduits in the range 70 to 80 µm. Effects of S-carvone (0, 0.318, and 0.636 mM), a monoterpene inhibitor of wound-healing enzyme inhibitor found in caraway (Carum carvi) and dill (Anethum graveoleus) seeds, on several native Australian, non-proteaceous cut flower and foliage species including A. holosericea (Mimosaceae), Baeckea frutescens (Myrtaceae), Chamelaucium uncinatum cv. ‘Mullering Brook’ (Myrtaceae), and the non-native Chrysanthemum sp. cv. ‘Dark Splendid Reagan’ (Asteraceae) were examined. As comparator treatments regular recutting of stem ends and use of standard tap water (STW: 0.7 mM CaCl2, 1.5 mM NaHCO3, and 0.05 mM CuSO4 in vases) were tested. S-Carvone treatments significantly (P≤0.05) extended the vase life of B. frutescens and C. uncinatum, constituting the first report of positive S-carvone effects on the vase life of Myrtaceous species. S-Carvone at 0.318 and 0.636 mM did not have antibacterial effects against Bacillus cereus (the main vase solution microbe) either in vitro or in the vase solution. Regular recutting of stem ends consistently improved all vase life parameters [viz. relative fresh weight (RFW), solution uptake, and vase life] in the three native species examined. STW had a positive effect on RFW and solution uptake only for A. holosericea cut foliage. Effects of di- and monovalent cations (Ca2+ as CaCl2 and K+ as KCl) on stem Kh of cut stem segments were studied. Abundance of Ca2+ on pit membranes versus xylem lumen wall surfaces was investigated using a novel low vacuum (LV) SEM plus energy dispersive X-ray (EDX) microanalysis technique. Both salts (0.1, 1, 10, and 100 mM KCl or CaCl2) did not significantly increase stem Kh compared to the corresponding deionised (DI) water controls (experiment 1). Highest increase in Kh was with KCl and CaCl2 at 10 and 1 mM, respectively. Increases in Kh with 100 mM KCl and CaCl2 were significant over DI water (experiment 2) for long (10 and 20 cm) and short (2 and 5 cm) stem segments, respectively. Increases in Kh of 1.2- and 2.4-fold for 100 mM KCl over DI water were found with increasing stem length from 2 to 20 cm. Kh decreased as stem segment length increased from 5 to 20 cm. However, contrasting results were found with 100 mM CaCl2, where ΔKh was larger in shorter (2 cm) than longer (20 cm) segments. To prevent dislocation of ions and distortion damage to the specimens, stem pieces were first LV freeze-dried, and then carbon-coated, viewed under SEM, and analysed for elemental composition and distribution by EDX. However, the method could not identify specific calcium peaks in xylem vessels perhaps because background signals were too high, and tissue topography interfered with signal detection. Effects of KCl and CaCl2 on vase life were also tested. RFW, solution uptake, and vase life were higher with 10 mM KCl and CaCl2 in the vase solution than with 0, 1, and 100 mM. STW had a significant (P≤0.05) positive effect on RFW and solution uptake rate when tested against deionised water, 10 mM KCl and 10 mM CaCl2. Different combinations of 0.05 mM CuSO4, 10 mM CaCl2, and 10 mM KCl were also tested as vase solutions. A significant positive effect on RFW and vase life was obtained with CuSO4 alone, CaCl2 alone, and CuSO4 plus KCl. Only CuSO4 and CaCl2 gave a significant positive effect on solution uptake rate. None of the eight treatments tested showed a consistent effect on stomatal conductance or stem Kh. Overall, the research revealed that tracheids and vessels with simple perforation plates and bordered vestured intervessel pits are the principal water conducting cells in A. holosericea. LV-SEM-EDX technique was unable to assess the spatial distribution of Ca2+ on xylem vessels, but this was the first attempt to apply this technique. Also, this is the first report on the extension of vase life of B. frutescens and C. uncinatum (Myrtaceous species) using S-carvone. In vase solutions, 0.05 mM CuSO4, 10 mM KCl plus 0.05 mM CuSO4 and 10 mM CaCl2 should have positive influences on the water balance of A. holosericea cut foliage stems.

Les micelles de caséines, composées de caséines, minéraux et eau, sont en équilibres dynamiques, elles échangent en permanence de leur matière avec leur environnement. Les micelles de caséines possèdent d’intéressantes fonctionnalités pour la fabrication de produits laitiers, telles que leur capacité à stabiliser des émulsions et à former des gels sous l’action de la présure. Des changements environnementaux, variations de pH, additions de sels ou d’agents chélatants, affectent les équilibres des micelles et induisent des modifications de leurs compositions et propriétés colloïdales. Ces changements modifient également leurs propriétés fonctionnelles, bien que cet aspect soit peu décri. Le but de ce projet était de comprendre les relations liant l’environnement, l’équilibre minéral, les propriétés colloïdales et fonctionnelles des micelles de caséines. L’impact de cinq facteurs (pH, Na3Cit, NaCl, CaCl2, MgCl2) modifiant les micelles fut étudié en focalisant sur leurs propriétés émulsifL’acidification et l’addition de Na3Cit ont causé les modifications minérales les plus importantes en solubilisant le phosphate de calcium micellaire. Le premier conduisit au « gonflement » des micelles alors que l’agent chélatant causa leur dissociation en petits agrégats. L’ajout de NaCl n’eut aucun impact sur le contenu minéral des micelles mais provoqua aussi la libération d’agrégats, révélée par cryo microscopie électronique et diffusion de rayons-X aux petits angles. Le rôle du phosphate de calcium sur les fonctionnalités fut confirmé et l’étude révéla l’importance de contrôler la libé / Caseins micelles, composed of caseins, minerals and water, are under dynamic equilibria, they constantly exchange materials with their surrounding environments. In addition, casein micelles possess valuable functionalities in regards to the formation of dairy products, such as the ability to stabilize emulsions or to form rennet gels. Environmental changes, such as variations in pH, additions of salts or chelating agents, affect the casein micelles equilibria and lead to modifications in their compositional and colloidal properties. Such changes also modify their functional properties, although this aspect is poorly described in the literature. This project aimed to understand the relationships that link the environmental modifications, the mineral balance, the colloidal and functional properties of the casein micelles. The impact of five modifying factors (pH, Na3Cit, NaCl, CaCl2, MgCl2) of the casein micelles were studied and the focus was placed on emulsion and rennet coagulation fuThe former only induce the swelling of casein micelles while the latter led to their disruption into smaller aggregates. NaCl had no impact on the micellar mineral content but also caused the release of small aggregates, as revealed by electronic microscopy and small angle X-ray scattering analyses. The decisive role of micellar calcium phosphate on the functionalities was confirmed and this study highlighted the importance of monitoring the release of small aggregates, as they strongly affected emulsions stability and gels firmness. Evaluation of Test Methods for De-icer Scaling Resistance of Concrete

The standard ASTM C672 de-icer salt scaling resistance test has been found to be overly aggressive to concretes containing slag cement. It was compared to the newly proposed ASTM WK9367 method, based on the Quebec BNQ test, as well as several modifications, including use of an accelerated curing regime developed in Virginia (VADOT). Sixteen concrete mixtures were studied using high-alkali cement, low-alkali cement, grade 100 slag and grade 120 slag with slag contents of 0%, 20%, 35% and 50%. Vinsol resin air-entraining admixture was compared to Micro Air®. Reducing the water cement ratio from 0.42 to 0.38 had the biggest impact on improving scaling resistance of slag concretes. In general, increased slag contents increased scaling regardless of the test method used. The Micro Air® admixture provided a lower air void actor and higher hardened air content compared to Vinsol resin.

Solvent–Solute Interaction : Studied by Synchrotron Radiation Based Photo and Auger Electron Spectroscopies

Aqueous solutions were studied using photoelectron and Auger spectroscopy, based on synchrotron radiation and a liquid micro-jet setup. By varying the photon energy in photoelectron spectra, we depth profiled an aqueous tetrabutylammonium iodide (TBAI) solution. Assuming uniform angular emission from the core levels, we found that the TBA+ ions were oriented at the surface with the hydrophobic butyl arms sticking into the liquid. We investigated the association between ions and their neighbors in aqueous solutions by studying the electronic decay after core ionization. The (2p)−1 decay of solvated K+ and Ca2+ ions was studied. The main features in the investigated decay spectra corresponded to two-hole final states localized on the ions. The spectra also showed additional features, related to delocalized two-hole final states with vacancies on a cation and a neighboring water molecule. These two processes compete, and by comparing relative intensities and using the known rate for the localized decay, we determined the time-scale for the delocalized process for the two ions. We compared to delocalized electronic decay processes in Na+, Mg2+, and Al3+, and found that they were slower in K+ and Ca2+, due to different internal decay mechanisms of the ions, as well as external differences in the ion-solute distances and interactions. In the O 1s Auger spectra of aqueous metal halide solutions, we observed features related to delocalized two-hole final states with vacancies on a water molecule and a neighboring solvated anion. The relative intensity of these feature indicated that the strength of the interaction between the halide ions and water correlated with ionic size.
