Optimization of culture medium for the cultivation of Actinoplanes sp. mutant strains and purification of acarbose: Research article

In order to improve the production of acarbose, the fermentation medium of acarbose-producing strain Actinoplanes sp. KCTC 9161 – L14 mutant was optimized in this internship. Fractional factorial design was employ to investigate the influences of glucose, maltose and corn power on acarbose production (by a-glucosidase inhibitory ability). Two significant factors: glucose and maltose have significant and positive effects on acarbose amount. In addition, a model was obtained from the regression results of fractional factorial experiment. Other success, we demonstrated that chromatography by active charcoal column can used to purify acarbose from fermentation broth. Acarbose amount in purification solution was 191.5 g/L and an acarbose - purification process was inducted. / Nhằm mục đích nâng cao khả năng sinh tổng hợp hoạt chất acarbose từ chủng đột biến Actinoplanes sp. KCTC 9161-L14, môi trường lên men của chủng dùng để sản xuất acarbose đã được tối ưu hóa. Một phần mềm thiết kế đã được thiết lập để khảo sát ảnh hưởng của glucose, maltose và bột ngô đến khả năng sản xuất acarbose (thông qua hoạt tính ức chế a-glucosidase). Kết quả đã cho thấy, hai yếu tố glucose và maltose có ý nghĩa quan trọng và ảnh hưởng trực tiếp đến khả năng sinh tổng hợp acarbose. Một phương trình đã được hình thành từ kết quả tối ưu. Bên cạnh đó, chúng tôi đã chứng minh được cột sắc ký sử dụng than hoạt tính có thể tinh sạch acarbose từ dịch lên men. Hàm lượng acarbose trong dung dịch tinh sạch đạt 191,5 g/l và một quy trình tinh sạch acarbose được đề xuất.

Climate change mitigation: an analysis of liquefied petroleum gas (LPG) as an alternative domestic energy source to charcoal in Zambia

This study analyses the possibility of introducing liquefied petroleum gas (LPG) as an alternative domestic energy source to charcoal in Zambia to mitigate climate change. The study, conducted in Lusaka, used questionnaires, focus group discussions, interviews and literature study to collect data. The study revealed that the introduction of LPG is possible. However, some challenges need to be addressed in order to actualise the change. Lack of knowledge about LPG; non-availability of LPG in most parts of the country; perceived danger of LPG; high prices of LPG and accessories; changing people‟s mind-sets about charcoal; social inequity and lack of government initiative, were the main challenges identified. Social-Ecology and sustainable livelihoods were used as theoretical frameworks to ground the study and analyse the results. The study recommends that government takes the lead to make the change possible and that further research be done on sustainable livelihoods for charcoal producers and traders. / Development Studies / M. Sediment reservoir dynamics on steepland valley floors : influence of network structure and effects of inherited ages

Sediment deposit ages inferred from radiocarbon dating of stream bank material were used to estimate residence times of valley-floor deposits in headwater valleys of the Oregon Coast Range, USA. Inherited ages of radiocarbon-dated material, i.e., time between carbon fixation in wood and its incorporation in a sediment deposit, can result in over-estimation of the ages of those deposits and, hence, the residence times of sediment within those units. Calibrated radiocarbon dates of 126 charcoal pieces sampled from Knowles Creek were used to estimate the distribution of inherited ages in fourteen depositional units representing three deposit types: fluvial fines, fluvial gravels, and debris flows. Within a depositional unit, the inherited age distribution of a piece of charcoal was estimated by convolving its calibrated age distribution with that of the piece of charcoal with the smallest weighted-mean calibrated age (i.e., an approximation of a unit's date of deposition) within that unit. All inherited age distributions for a particular deposit type were then added and normalized to provide a probability distribution of inherited ages for that deposit type. Probability distributions of inherited ages average 688, 1506, and 666 yr for fluvial fines, fluvial gravels, and debris flow units, respectively. Curves were fit to inherited age distributions for each deposit type. These curve fits were then convolved with deposit age distributions (i.e., equal to calibrated age distributions of woody material sampled from stream banks) of samples from Bear Creek (Lancaster and Casebeer, 2007) to correct these deposit ages for inherited age. This convolution gives a corrected deposit age. In cases in which means of corrected deposit age distributions for an upper unit were older than those of a lower unit within a stratigraphic column, the upper sample’s corrected deposit age distribution was set to that of the youngest lower in the stratigraphic section. Convolution shifted individual deposit age distributions towards zero and increased their standard deviation by an average of 365%. However, convolution decreased the standard deviations of normalized probability distribution functions of deposit ages inferred from many samples from 1340 to 1197 yr, and from 471 to 416 yr for lower and upper reaches, respectively, of the Bear Creek valley in the Oregon Coast Range. Convolution decreased estimates of mean deposit ages from 1296 to 1051 yr, and from 308 to 245 yr for lower and upper reaches, respectively, of the Bear Creek. Estimates of percentages of basin denudation passing through each reach's deposit ("trapping efficiency") increased from 11.6% to 14.4%, and from 25.4% to 31.9% for lower and upper Bear Creek, respectively. However, basic shapes of residence time distributions and, thus, inferences regarding removal of sediment from the reaches did not change after deposit dates were corrected. Sediment residence times in the lower Bear Creek valley are exponentially distributed, which implies that all sediment has a uniform probability of evacuation from deposits, whereas the power-law-distributed residence times in upper Bear imply preferential evacuation of younger deposits and preservation of older deposits. Much of the sediment transported onto valley floors via debris flows is deposited, and then is evacuated over longer times. Volumes and residence times of stored sediment in these deposits at the transition from debris flow to fluvial evacuation, and their associated width of valley floors, vary throughout a network. Export volumes and frequencies from tributaries are controls on deposit volumes and may control valley widening of mainstem valley floors. In addition, closely spaced tributaries may exert composite effects on valley floor landforms. It is hypothesized that the volumes of sediment stored at confluences increases with contributing watershed area of tributaries to the point where tributary slopes are low enough to cause most debris flows to be deposited within tributary valleys instead of in the mainstem valley. In four ~1 km reaches with contributing watershed areas of 0.3 to 5.0 km², field surveys provided measures of width of valley floors and volume of deposits, and radiocarbon dating of charcoal provided residence times of sediment in these deposits. Mean residence times of reaches vary between 1.1 and 2.5 kyr. Exponential distributions fit to residence times within two of the reaches imply evacuation of sediment independent of deposit ages. Power-law fits to residence times of the other two reaches imply age-dependent evacuation of deposits. Distribution shapes of residence times, and their means, do not vary systematically with contributing watershed area of mainstems. Mean width of mainstem valley floors increases with contributing watershed areas of both mainstems and their respective tributaries. Volumes of sediment stored on the valley floor increase with contributing areas of mainstems, and these volumes at tributary junctions peaked at tributary contributing areas of ~0.1 km². Percentage of basin denudation entering storage decreases with contributing area of mainstem. This decrease may be due to increasing percentages of sediment supply via fluvial transport for larger watersheds, and much, if not most, of this supply routes through the system quickly. / Graduation date: 2012

Remoção de alquilbenzeno linear sulfonado (LAS) e caracterização microbiana em reator anaeróbio de leito fluidificado / Removal of linear alkylbenzene sulfonate (LAS) and microbial characterization in anaerobic fluidized bed reactor

Nesse trabalho foi estudado a degradação anaeróbia do alquilbenzeno linear sulfonado (LAS), um surfactante amplamente utilizado na fabricação de detergentes e presente em esgoto doméstico e águas residuárias industriais. Para isso foi utilizado reator anaeróbio de leito fluidificado em escala de bancada (1,2 L) preenchido com material suporte para imobilização da biomassa. Quatro diferentes suportes foram testados previamente em reatores de leito fluidificado em menor escala (350 ml): carvão ativado (R1), argila expandida (R2), pérolas de vidro (R3) e areia (R4). Todos os reatores foram inoculados com lodo proveniente de reator UASB utilizado no tratamento de dejetos de suinocultura e alimentados com substrato sintético acrescido de LAS. Os reatores foram mantidos a 30°C e operados com tempo de detenção hidráulica (TDH) de 18 horas. Foi possível constatar que os quatro reatores foram aptos na remoção de matéria orgânica (acima de 84%) e LAS (acima de 81%), respectivamente para concentração inicial média de 550 mg/L e 16,5 mg/L. No entanto, carvão ativado e argila expandida sofreram processo de fragmentação durante a operação do reator. Assim, areia foi o material escolhido para preencher o reator em escala de bancada devido aos bons resultados de remoção do LAS (99%), menor custo e facilidade de aquisição, quando comparada a pérola de vidro. Após 270 dias de operação, com concentrações crescentes de LAS e média de 32,3 mg/L, constatou-se que o reator em escala de bancada removeu 93% de LAS. Todos os suportes adsorveram pouco LAS (máximo de 0,43 mgLAS/gargila) não interferindo significativamente no processo de remoção biológica. Exames microscópicos realizados durante a operação dos reatores mostraram que estiveram presentes microrganismos com morfologias semelhantes a espiroquetas, bacilos, bactérias filamentosas e cocos, entre outros. A biomassa presente no final da operação do reator em escala de bancada e nos reatores menores preenchidos com pérolas de vidro (R3) e areia (R4) foi submetida à técnica de clonagem e sequenciamento do gene 16S RNAr para o Domínio Bacteria. Observou-se que os reatores apresentaram clones relacionados a diversos Filos como Bacteroidetes, Proteobacteria, Verrucomicrobia e Firmicutes, entre outros. / This work presents the anaerobic degradation of linear alkylbenzene sulphonate (LAS), a surfactant widely used in the production of detergents and present in domestic and industrial wastewaters. It was used an anaerobic fluidized bed reactor in bench scale (1,2L) filled with support material for biomass immobilization. Four different supports were previously tested in small scale fluidized bed reactors (350 ml): activated charcoal (R1), expanded clay (R2), glass beads (R3) and sand (R4). All reactors were inoculated with sludge from a UASB reactor treating swine wastewater and were fed with a synthetic substrate supplemented with LAS. The reactors were kept at 30ºC and operated with a hydraulic retention time (HRT) of 18 h. It was possible to note that the four tested reactors were able to remove organic matter (higher than 84%) and LAS (higher than 81%), respectively, to initial mean value of 550 mg/L and 16.5 mg/L. However, activated charcoal and expanded clay both produced shearing during reactor operation. Thus, sand was the chosen material to fill the bench scale reacto because of good results of LAS removal (99%) and smaller cost and affordability compared to glass beads. After 270 days of operation, with crescent LAS concentrations and average of 32,3 mg/L, it was found that the bench scale reactor was able to remove LAS in 93%. O estudo do metabolismo de poliaminas (PAS), envolvendo parâmetros fisiológicos, bioquímicos e moleculares, pode gerar uma melhor compreensão do processo de maturação, e criar estratégias importantes para a otimização da embriogênese somática (ES) em cana-de-açúcar. O objetivo deste trabalho foi estudar a relação entre os conteúdos endógenos de PAs, associados à determinação da competência embriogenética e a maturação progressiva de embriões somáticos em cana-de-açúcar. Calos embriogênicos (E) e não-embriogênicos (NE) da variedade SP-803280, foram submetidos a diferentes tratamentos, utilizando agentes de maturação adicionados ao meio MS. Observou-se que as culturas NE não foram capazes de promover a diferenciação de embriões somáticos, devido principalmente, ao alto grau de oxidação das culturas. Os tratamentos suplementados com 0,75 e 1,5 g.L-1 de carvão ativado (CA), foram os que apresentaram as maiores freqüências de formação de embriões somáticos. A partir dos resultados obtidos, foi possível associar a ES em calos de cana-de-açúcar ao aumento do conteúdo endógeno de Spd (Espermidina) e Spm (Espermina), principalmente Spd, acompanhado por uma redução nos níveis de Put (Putrescina). / Somatic embryogenesis (SE) is a highly potential supplier of the material involved in the regeneration and genetic transformation of transgenic plants. The aim was to study correlations between endogenous PA contents, associated to defining embryogenetic competence and the progressive maturation of somatic embryos. Embryogenic (E) and non-embryogenic (NE) calli from the SP-803280 variety, were submitted to different maturation treatments. It was found that the NE cultures submitted to the two maturation experiments, were incapable of promoting somatic embryo differentiation, mainly due to browning. It was noted that in the first experiment, total PA endogenous content of the E callus was higher than in the NE, although, in the control, no significant differences between both callus types were encountered. We can associate the progression of sugarcane somatic embryogenesis to an increase in endogenous content of Spd e Spm, mainly Spd, accompanied for a reduction in Put levels. Thus, it was shown that somatic embryo maturation in sugarcane can be related with PA biosynthesis, thereby indicating the importance of their metabolism in the competence of sugarcane embryogenic cultures.

Bioenergy, development and food security in Sub-Saharan Africa

Die vorliegende Dissertation beinhaltet eine detaillierte Analyse traditioneller und moderner Bioenergie, sowie deren möglicher Entwicklungspfade und Auswirkung auf Ernährungssicherheit im ländlichen Tansania. Die Arbeit bereichert die wissenschaftlichen Debatten mittels Aufzeigen von Optionen der nachhaltigen Produktion und Verbrauch von Bioenergie. Der Komplex wird in vier separaten und begutachteten Fachartikeln (drei publiziert, einer under review) bearbeitet. Alle Publikationen erzielen Aussagen für vier Einkommensgruppen auf lokaler Ebene. Für eine optimale Analyse ist die Arbeit thematisch in die Forschungsgebiete traditionelle und moderne Bioenergie unterteilt, die sich auch in den Kapiteln bzgl. des aktuellen Forschungsstandes sowie der Abfolge der Fachartikel widerspiegeln. Die erste Publikation beschäftigt sich mit traditionellem Bioenergiekonsum und der postulierten Einführung eines feuerholz-spezifischen, energetisch optimierten Kochers in dem Hauptuntersuchungsdorf Laela. Die Effekte einer Einführung von technologisch optimierten Holzkohlemeilern werden in der zweiten Publikation widergegeben. Nach diesem Fokus auf traditionelle Bioenergie analysiert der dritte Artikel die Effekte auf Nahrungsmittelsicherheit, die sich aus der angenommenen Verstromung von Pflanzenölen ergeben. Der vierte Artikel analysiert die Möglichkeiten einer erhöhten Palmölproduktion und leitet Ergebnisse für die tansanische Region Kigoma ab. Eine finale Empfehlung dieser Dissertation ist, dass die Verbreitung und Nutzung von verbesserten Kochern stark erhöht werden muss. Gleichzeitig sollten politischen Maßnahmen ergriffen werden, die Zugang und Verfügbarkeit von verlässlicher, erschwinglicher und nachhaltiger traditioneller wie moderner Bioenergie garantieren. Zudem muss die Etablierung einer effizienteren wie nachhaltigeren Holzkohle-Wertschöpfungskette vorangetrieben werden. Pflanzenölbasierte Elektrizitätsproduktion dagegen wirkt sich negativ auf die Ernährungssicherheit aus. / This PhD thesis provides a detailed analysis of the traditional and modern bioenergy situation as well as their development pathways and their respective influence on food security in case study villages in rural Tanzania. It adds to the current literature as it provides a profound understanding of how to enhance and sustain bioenergy production and consumption in terms of resource capacity and overall sustainability. For this, it comprises four peer-reviewed papers (3 published, one under review). All papers derive results for four income groups at local level. For an optimized analysis, the dissertation is thematically subdivided into the research areas of traditional and modern bioenergy which is displayed in the state of art research sections as well as the clustering of the papers. The first paper focuses on traditional bioenergy consumption and the potential introduction of a firewood-efficient stove in the major case-study village Laela. The effects of the introduction of more efficient kiln technology on resource consumption in the same village are mirrored in the second publication. After this focus on traditional bioenergy, the third paper investigates food security effects resulting out of the assumed use of vegetable oil for electrification purposes. The fourth paper analyses options for increased palm oil production and derives results for the Tanzanian region of Kigoma. The final recommendation of the thesis is that the prevalence and use of improved stoves needs to be increased substantially. Simultaneously, policy measures that foster the access to and availability of reliable, affordable and sustainable traditional as well as modern energy should be implemented. Furthermore, the establishment of a more efficient and sustainable charcoal value chain needs to be promoted. Vegetable oil based electrification, however, does not contribute towards food security.

Prospects for sustained harvesting of mopane (Colophospermum mopane) on the Venetia Limpopo Nature Reserve and its implications for browsing ungulates

Thesis (MSc) -- Stellenbosch University, 1996. / ENGLISH ABSTRACT:Principle objectives of this study were to investigate the ecological feasibility of sustained utilization of mopane, using Venetia Limpopo Nature Reserve as a pilot study area. The study was limited to three different mopane dominated vegetation types on the reserve with most research being conducted in C.mopane woodland. Results can be summarized as follows: 1. Colophospermum mopane is ubiquitous on VLNR. 2. C.mopane woodland has the deusest mopane growth of the three vegetation types and highest densities are found on soils (OaB, VaB, Du, SwB & VaA) in association with C.mopane woodland. There seems to be a correlation between mopane density and soil type. 3. Largest mopane trees according to height and circumference were found in C.mopane/C.apiculatum open woodland. There is no significant difference between height and circumference on different soil types. 4. Most senility occurs in C.mopane shrubland and there is a significant difference in senility between mopane associated with different soils on which this vegetation type occurs. 5. Oldest average ages per tree were fo•.md in C.mopane woodland while growth rates were fastest in C.moparse!C.apiculatum open woodland. 6. There is a significant positive correlation between total and charcoal weights for mopane. Total and charcoul weights per tree were highest for C.mopane/C.apiculatum open woodland, while weights per hectare were highest for C.mopane woodland. Soils could not positively be associated with this trend. 7. Mopar.e lends itself to harvesting, especially C.mopane ••10odland in the Endora, Lizzulea and Hilda areas. Winter months seem to be most desirable for harvesting activities. Hand felling in elongated patches (increases ecotone boundary) is recommended where labour is cheap. A harvesting strategy of between 25% and 50% would make the operation economically viable and ecologically acceptable. An optimum harvesting rate of 27% is suggested by a harvesting model developed specifically for this mopane data. 8. Total herbivore densities, distribution and species proportions can be expected to change following bush clearing. Habitat for grazers should increase due to an increased grass production. Habitat for browsers should not be influenced much. Effect of noise and human disturbance during harvesting, especially for elephants is negligible, as hunting activities already take place on VLNR. 9. Grass production would increase but quality should decrease after harvesting, due to an increase in low quality opportunistic species, especially if soil disturbance takes place. Grass quality is best in association with herbaceous species such as Salvadora angustifolia. Carrying capacity for grazers should increase after harvesting as a result of habitat change and grass biomass increase. 10. Greatest advantages for browsers after harvesting, is the fact that leaves become senescent later in autumn and that spring leaf flush takes place earlier thus prolonging bro\'se availability. / AFRIKAANSE OPSOMMING:Die hoof doelwitte van hierdie studie was om te bepaal wat die invloed van die oes van mop nie op die ekologie sou wees as dit op 'n volgehoue basis plaasvind en tot w•atter mate mopanie geskik is vir hou skoolvervaardiging. Die navorsing is op die Venetia Limpopo Natuureservaat in slegs drie van die belangrikste mopanie veldtipes uitgevoer met die oorgrote meerderheid van die navorsing in die C.mopane bosveld. Die resultate van die studie kan soos volg opgesom word: 1. Colophospermum mopane is alomteenwoordig op die VLNR. 2. Die C.mopane bosveld het die hoogste digtheid van al drie veldtipes en word ook geassosieer met sekere grondtipes (OaB, VaB, Du, SwB & VaA) wat eie is aan hierdie spesifieke veldtipe. Daar blyk 'n korrelasie te wees tussen die digthede van mopanie en grondtipes. 3. Die grootste mopanie borne, ten opsigte van hnogte en omtrek, word in die C.mopane/C.apiculatum oop bosveld aangetref. Daar is nie 'n beduidende verskil tussen die hoogte en omtrek van mopanie op die verskillende grondtipes nie. 4. Die C.mopane struikveld toon die meeste seniliteit ten opsigte van die persentasie kruin dood. Daar is 'n definitiewe aanduiding dat seniliteit ooreenstem met sekere grondtipes veral met betrekking tot die mopanie struikveld. 5. Die C.mopane bosveld het gemiddeld die oudste borne terwyl die C.mopane/C.apiculatum oop bosveld die vinnigste groeikoers toon. Die bogenoemde feite het 'n ver0and met die grondtipes. 6. Daar is 'n posltiewe korrelasie tussen die totale gewig en die houtskoolgewig van mopanle. Die S\vaarste borne is in die C.mopane!C.apiculatum oop bosveld aangetref, terwyl die swaarste gewig per hektaar in die C.mopane bosveld aangetref is. Gewig kon nie met grondt pes gekorreleer word nie. 7. Die mopanie in die C.mopane bosveld is geskik vir die oes vir houtskool, veral in die Endora, Lizzulea en Hilda gebiede. Dit is raadsaam om die borne in verlengde stroke per hand af te kap (vergroot die ekotoongebied), veral waar arbeid goedkoop is. 'n Oesstrategie van tussen 25% en 50% vir mopanie word aanbeveel om dit ekonomies en ekologies aanvaarbaar te maak. 'n Optimum oesstrategie van 27% word aanbeveel deur 'n oes model wat spesifiak vir hierdie mopane data opgestel is. 8. Daar kan verwag word dat na die oes van mopanie die getalle asook verspreidingspatrone van alle hoefdierspesies kan verander. 'n Voordeel van lae mopanie digthede is dat die verspreiding van die blaarbiomassa ewerediger oor die seisoene sal geskied. Grasvreters word bevoordeel vanwee die verandering in habitat. Die effek van geraas en ander oesbedrywighede op olifante behoort klein te wees aangesien jagaktiwiteite alreeds op diereservaat toegepas word en hulle redelik gewoond daaraan behoort te wees. 9. Grasproduksie behoort toe te neem terwyl die kwaliteit daarvan afneem as gevolg van opportunistieslae kwaliteit grasspcsies wat toeneem na die oes van mopanie, veral as die grand tydens oesaktiwiteite versteur word. Die beste kwaliteit gras word in assosiasie met kruidagtige spesies, veral Salvadora angustifolia, gevi.nd. As gevolg van die verandering in habitat asook die toename in grasbiomassa neem die drakrag vir grasvreters na die oes an mopanietoe. 10. Di.e totale blaarbi.)massa behoort na die oes van mopanie af te neem, maar behoort terselfdertyd op die oorblywende borne as gevolg van die afname in kompetisie vi= watr en anrler natuurlike hulpbronne toe te neem. Hoer persentasies blomvorming en saadproduksie vind by laer boomdigthede plaas. Die grootste enkele voordeel vir blaarvreters, na die oes van mopanie, is die feit dat die blare later in die herfs afval en vroeer in die lente uitbot en dus die voedselbeskikbaarheid vir blaarvreters verleng.

Análise da viabilidade de produção de painéis de cimento reforçados com biomassa vegetal e escória de alto-forno / Analysis of the viability of production of cement panels reinforced with vegetal biomass and blast furnace slag

Fundação Araucária / O objetivo dessa pesquisa foi caracterizar painéis de cimento reforçados com biomassa vegetal de bambu mossô (Phyllostachys pubescens) e biomassa do bagaço de cana-de-açúcar (Saccharum officinarum) com adição de escória de altoforno à carvão vegetal. Realizou-se a caracterização física e química dos materiais. Após, foi realizado ensaio de inibição para análise da compatibilidade da biomassa de bambu mossô e de cana-de-açúcar com o cimento Portland. Os tratamentos adotados para as biomassas foram: imersão em água fria, água quente, hidróxido de sódio, silicato de sódio, silano e aditivo acelerador de pega à base de cloreto de cálcio. Foi analisado a resistência à compressão dos compósitos com a adição de diferentes teores de escória de alto-forno à carvão vegetal. Finalmente foram determinadas as propriedades físicas e mecânicas dos painéis compostos de biomassa vegetal, cimento Portland e escória de alto-forno. Os resultados do ensaio de inibição feitos com a biomassa vegetal indicaram que a adição de cloreto de cálcio à mistura é o tratamento mais eficiente para reduzir o índice de inibição da biomassa de bambu mossô e de cana-de-açúcar. Para os compósitos com escória de alto-forno, o ensaio de inibição demonstrou que a adição de 50% de escória de alto-forno se mostrou eficaz no aumento da compatibilidade. O teor de 40% de escória apresentou resistência superior aos demais traços. Para os painéis cimentobiomassa vegetal verificou-se que a biomassa de bambo mossô apresenta total viabilidade para a confecção de painéis enquanto que a biomassa de cana-deaçúcar é inviável. A relevância desse estudo está no emprego de resíduos de atividades industriais, como biomassa vegetal e escória, para a produção de novos materiais de construção. / This study aims at characterizing cement panels reinforced with mosso bamboo biomass (Phyllostachys pubescens) and biomass of sugarcane bagasse (Saccharum officinarum) with addition of blast furnace charcoal slag to the mix. The physical and chemical characterization of the materials were carried out. Afterwards, an inhibition test was performed to analyze the compatibility of the powders with Portland cement. The treatments adopted were: immersion in cold water, immersion in hot water, sodium hydroxide, sodium silicate, silane and calcium chloride. The composites compressive strength was analyzed with the addition of different mixes of blast furnace charcoal slag. Finally, the physical and mechanical properties of the panels made of vegetal biomass, Portland cement and blast furnace slag were determined. The results of the inhibition test, made with plant biomass shows that the addition of calcium chloride to the blend is the most efficient treatment to reduce the inhibition rate of the biomass of mosso bamboo and biomass of sugarcane. For composites with blast furnace slag, the inhibition test showed that the addition of 50% blast furnace slag proved effective in increasing compatibility, thus improving compressive strength. In this case, the treatment with 40% slag content presented superior resistance to the other traces. For the cement-vegetal biomass panels the study found out that the mosso bamboo biomass presents total feasibility for the preparation of panels whereas the sugar cane biomass is not feasible. The relevance of this study is the use of industrial waste, such as vegetal biomass and slag, for the production of new construction materials.

Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2016. / The FZV nanoparticles immobilized onto the surface of the activated charcoal (nFZV-CAG) were synthesized and characterized for the removal of the pharmaceutical Nimesulide (NMS) in aqueous systems. The studies were performed in batch mode with stirring speeds of 250 rpm during 120 minutes, so that it was possible to evaluate efficiency, concentration (20, 25 and 30% of the composite) and different dosages (0,1 to 10g) of nFZV-CAG. The results showed that the dosage of 10g of 20%nFZV-CAG removed approximately 80% of NMS 50 mg L-1 in just 30 minutes of reaction, and reaching 100% in 120 minutes. It was still observed a removal of 80% of the COD (Chemical Oxygen Demand) at the end of the reaction. The reaction rates increased with the dosage of the composite, which was expected. Though, increasing the concentration (%) of nFZV did not result in higher reaction rates. The reactions followed a pseudo-first order kinetics for the removal of NMS. After the samples were collected, they were submitted to chemical analysis, employing the techniques of UV-VIS spectroscopy, high performance liquid chromatography (HPLC) and chemical oxygen demand (COD). In order to characterize the composite, the following techniques were used: Scanning Electron Microscopy (SEM) coupled with Energy Dispersive x-Ray spectroscopy (EDS), which showed clearly the presence of nanoparticles onto the charcoal surface. Analysis of the surface of nFZV-CAG were also performed. / The composite nFZV-CAG was used for the catalytic ozonation of NMS in aqueous media. The ozone was generated by an ozone generator IPABRAS, fuelled with air. Different catalytic processes were evaluated such as O3, CAG, O3-CAG, O3-nFZV-CAG, and the variation of the nFZV concentration for the removal of NMS. The results showed that the combination of O3- nFZV-CAG was very efficient, leading to the mineralization of approximately 70% of NMS in 120 minutes of reaction. Such efficiency can be attributed to the electrolysis process, in which the Fe0 generates Fe2+ which generates hydroxyl radicals that are highly oxidant, leading to the destruction of the contaminant. The reactions followed the pseudo-first order kinetics for the removal of the pharmaceutical. After the samples were collected, they were submitted to chemical analysis such as, UV-VIS spectroscopy, high performance liquid chromatography (HPLC) and chemical oxygen demand (COD).
