Physiological and genetic manipulation of adventitious rooting in Prunus spp

Many species from economically important genera remain rooting recalcitrant, prohibiting the commercialisation of many species in forestry and horticulture, and hindering genetic improvement by conventional breeding or recombinant DNA technology, where vegetative propagation is often used to preserve the genetic fidelity of elite progeny. Two cherry species (Prunus avium and P. padus) were used as models in this study to investigate the physiological and genetic manipulation of adventitious rooting. Mature trees are typically more difficult to propagate vegetatively than their juvenile counterparts. For some trees, micropropagation can circumvent certain effects of ageing and maturation, restoring shoot vigour and rooting, but the mechanism(s) involved have not been elucidated. During micropropagation, subculture interval was found not to be the predominant factor promoting the 'apparent rejuvenation' of mature P. avium tissue. 'Apparently rejuvenated' ex vitro and hedged (putatively) mature P. avium trees were treated with gibberellins predicted to have a range of structural related activities. GA, improved the rooting of cuttings from hedged (putatively) mature cherry, but not from ex vitro trees. Methodology to regenerate adventitious shoots from P. avium leaf explants was developed, (putative) transgenic P. padus plants were produced by an Agrobacterium tumefaciens mediated strategy. Auxin redistribution in planta is postulated to require a component of active transport; inhibition of the predominantly basipetal transport has profound effects on rooting. The putative function of the Arabidopsis thaliana AtAUX1 gene is that of a cellular auxin influx carrier, possibly, as described by the chemiosmotic hypothesis. This thesis examined the hypothesis that transformation with the AtAUX1 gene would enhance the delivery of the root-inducing signal to improve rooting of P. padus, a species which is rooting recalcitrant and more or less obligate on exogenous auxin for this process. However, all six, constitutively expressed, Cauliflower Mosaic Virus 35S promoter driven, 35S::AtAUX1, transgenic shoot lines had reduced rooting.

Para avaliação fenológica, em cada época foram marcados 40 botões florais em 10 plantas, totalizando 400 botões. O ciclo foi dividido em estádios fenológicos, sendo avaliada a duração dos estádios, taxa de frutificação e crescimento dos frutos. Na avaliação da produção os frutos maduros foram colhidos e pesados. Para análise qualitativa foram coletadas amostras de frutos e avaliados o peso médio de fruto, cor de fruto e as características químicas da polpa: teor de sólidos solúveis, pH, acidez titulável, açúcares redutores, ‘ratio’ e teor de ácido ascórbico. Os resultados obtidos permitiram concluir que a duração dos estádios é influenciada pela temperatura ambiente, sendo mais curto em condições de altas temperaturas. A elevada incidência de precipitações interfere negativamente na fixação dos frutos da aceroleira, prejudicando a produção. Também pode-se observar que o maior índice de abortamento ocorre no estádio de perda de pétala. Com relação ao desenvolvimento dos frutos, apesar da diferença na velocidade de crescimento e tamanho dos frutos, em ambas as avaliações pode-se observar que houve um mais acentuado nos primeiros 8 dias, seguindo depois um padrão linear. Para os dados de produção e qualidade dos frutos pode-se concluir que houve influência da época... / The barbados cherry tree is a tropical plant that produces fruits rich in vitamin C. Collaboration for Environmental Sustainability on Gotland, Sweden. Nature’s Blueprint: Biomimicry as a Potential Strategy.

This master's thesis explores the potential for collaboration among diverse actors on the island of Gotland to achieve environmental sustainability. The study employs the concept of biomimicry, utilising selected natural phenomena as a source of inspiration. A theoretical framework and qualitative research methodology are employed to guide the process, with the aim of developing a strategy to enhance the collaborative state for environmental sustainability on Gotland. Drawing on principles derived from nature, the study investigates how biomimicry can provide solutions and inspire collaborative efforts for environmental sustainability. Central to the research is the exploration of trust-building mechanisms among diverse actors. Recognising that trust is a fundamental ingredient for effective collaboration, the study examines strategies that can foster trust within the context of environmental sustainability. By identifying barriers and enablers of trust, the thesis proposes actionable recommendations to enhance the trust-building process on Gotland, thereby fostering a conducive environment for collaboration. The proposed strategy aims to create a collaborative framework that fosters long-term partnerships, harnessing the diverse expertise and resources of each actor to address the multifaceted challenges of environmental sustainability on Gotland. In conclusion, this master's thesis underscores the value of applying biomimicry principles to foster collaboration for environmental sustainability on Gotland.

In her doctoral thesis the author would like to present the play Yoshitsune and the Thousand Cherry trees (Yoshitsune senbonzakura) as a unique piece of Japanese drama which has a great complexity and an outstanding place within the corpus of Japanese literature. Even if the play can be compared to Chushingura mono in its importance, Yoshitsune senbonzakura has not been widely translated to western languages. There are two exceptions, however: the English translation (Jones, Jr. 1993) and the German translation (Klopfenstein 1982). While comparing the two translations and using the most full original texts available (as preserved in Takeda Izumo and Namiki Sosuke Joruri Collection and Yuda Yoshio's Bunraku Joruri Collection), the author's goal is to present the play to Czech readers and its interpretation to Czech scholars. As for the flow and structure of the interpretation of Yoshitsune senbonzakura the author begins with the historical background of the legend of Minamoto Yoshitsune, capturing the life of Yoshitsune from the time of Heiji rebellion (1159) when he was born to his death in 1189. Next the author concentrates on the legend itself as it evolved within the course of Japanese literature. With a shift from the court literature of Heian period towards the battlefield stories and...
