Diferenças moleculares entre citótipos de Mazama americana (Artiodactyla: Cervidae) /

Resumo: O veado-mateiro (Mazama americana) possui uma ampla distribuição geográfica na região neotropical. Estudos citogenéticos com a espécie revelam variações cromossômicas (citótipos) que apontam sua divisão em outras espécies. Neste trabalho, foram examinadas as relações filogenéticas desta espécie, analisando parte dos genes mitocondriais (citocromo-b e região controladora D-loop), dos genes nucleares (Beta e Kapa caseínas e do exon I do gene IRBP) e um fragmento do gene, presente no cromossomo Y, chamado SRY, para amostras de 19 indivíduos provenientes de diferentes regiões do Brasil. Os genes nucleares da kapa e beta caseína e do SRY, mostraram-se monomórficos, não sendo possível a obtenção de sequências para o gene IRBP. As inferências filogenéticas pelos genes mitocondriais revelam duas linhagens evolutivas, a dos indivíduos das populações da Bacia do Rio Paraná e a dos indivíduos do oeste da Bacia do Rio Amazonas. Também houve uma correlação entre os diferentes cariótipos e as distintas linhagens moleculares encontradas. Além disso, pode-se sugerir a ocorrência de convergência evolutiva entre estes grupos, bem como um possível caso de simpatria ou de retenção de polimorfismo ancestral nos indivíduos do leste da Amazônia. / Abstract: The red brocket deer (Mazama americana) has a wide distribution in Neotropics. In this regard, cytogenetic studies in this species revealed chromosomic variations (cytotypes) which strongly suggest that red brockets can be divided into other species. In the present study, we examined phylogenetic relationships of 19 samples of individuals from different areas of Brazil through mitochondrial (cytochrome b and control region D-loop), nuclear (b-casein, k-casein, and first exon of the IRBP) and SRY gene analysis. The sequence analysis showed that b- and k-caseins as well as SRY nuclear genes were monomorphic, whereas IBRP gene sequencing was not possible. Phylogenetic inferences concerning mitochondrial gene analysis demonstrated two evolutionary lineages, one from Parana River Basin (southeast Brazil) and other from west of Amazon River Basin (northwest Brazil). Animais originários do Estado de Rondônia têm apresentado importantes diferenças citogenéticas em relação ao padrão de outras populações, o que suscita necessidade de estudos mais aprofundados para definição da sua posição filogenética. O presente estudo objetivou identificar as diferentes populações de veado-mateiro desta região, verificando a existência de mais de uma espécie no local. Para tanto, foram obtidos 51 fragmentos de tecido de animais caçados por indígenas e pela população local em todas as regiões do Estado dos quais 33 tiveram seu DNA extraídos, amplificados (região de 480pb do citocromo b) e seqüenciados de forma satisfatória. Estas seqüências foram alinhadas e comparadas, gerando 21 haplótipos que se encontram distribuídos de forma aleatória pelas diversas regiões de coleta. Estes haplótipos serviram de base para a elaboração de redes de distância e árvores filogenéticas que quando analisadas sugeriram a existência de espécies crípticas dentro do que hoje se denomina Mazama americana no Estado de Rondônia. / Abstract: The red brocket deer is the largest species of Mazama genus and it is distributed in almost all Neotropical regions. Individuals originated from Rondônia state in Brazil have been presented important cytogenetic differences when compared with populations of other regions of country; however more studies are necessary to define correct phylogenetic position of species. The objective of present study was performed the identification of different populations of red brocket deer from Rondônia state by verification of occurrence of more than one species on mentioned region. For this, 51 fragments of tissues from hunted animals were obtained with Indians and local people of all regions of Rondônia state. In 31 fragments of tissues the DNA was successful extract, amplified (480 bp region of cytochrome b) and sequenced. These sequences were aligned and compared creating 21 haplotypes, which are distributed in a randomly way thru the different regions of sampling. The haplotypes were used to elaborate distance nets and phylogenetic trees, which when analyzed suggested the existence of cryptic species on Mazama americana species that occurs in Rondônia state. / Mestre

En augmentant la distribution des ressources dans l'espace, la fragmentation du paysage contraint le mouvement des organismes. Deux types de stratégies adaptatives antagonistes ont été mises en évidence en réponse à cette pression : une diminution ou une augmentation de la propension et de la capacité des organismes à se déplacer. La majorité de la littérature traite de la diminution des mouvements en réponse à la fragmentation du paysage. En étudiant un organisme contraint de traverser la matrice pour achever son cycle de vie (i.e. Bufo bufo), je me suis intéressée à la stratégie d'augmentation et d'optimisation des mouvements en réponse à la fragmentation. Peu d'études permettent de conclure sur l'existence de plasticité phénotypique, d'effets maternels ou d'évolution des traits liés au mouvement en réponse à la fragmentation. J'ai ainsi inclus dans ma démarche expérimentale des élevages en conditions standardisées afin d'étudier l'évolution des caractéristiques du mouvement en réponse à la fragmentation. Plus précisément, j'ai étudié l'évolution des capacités de navigation en m'intéressant à la vitesse dévolution, à la base magnétique et à l'influence de l'orientation vectorielle sur la dispersion. Je me suis également intéressée à l'évolution du comportement exploratoire en réponse à la fragmentation / Landscape fragmentation increases resource distribution and constrains animal movements. Two kinds of adaptive strategy have been revealed in response to this selective pressure: an increase or a decrease of animal capacity and propensity to move. I studied the resistance strategy (i.e. increase of movements) which is poorly documented in the literature. To do so I focused on an organism facing the need to cross the matrix to achieve its life cycle (Bufo bufo). Only a few studies enable to disentangle phenotypic plasticity, maternal effect and evolution of movement characteristics. I used common garden rearing and cross-breeding experiments in order to focus on the evolutionary dimension of movement characteristics changes induced by landscape fragmentation. More precisely, I studied the evolution of navigation capacities by focusing on vector orientation, its evolutionary rate, its genetic basis, its magnetic basis and its influence on dispersal. I also studied the evolution of the exploratory behaviour in response to landscape fragmentation and searched for convergent evolution of this behaviour

The mechanisms underlying convergent evolution in the plumage patterns of birds

Convergent evolution is a central theme in biology. Birds are an ideal system to examine the mechanisms underlying convergent evolution. Although bird patterning is diverse, within-feather patterns have repeatedly converged on the same four types: mottled patterns, scales, bars and spots. Other avian patterns occur, e.g. stripes, but are rare. In my thesis I examine the four main mechanisms underlying convergent evolution in plumage patterns: evolutionary genetics, evolutionary development, natural selection for signaling and camouflage. Japanese quail (Coturnix japonica) is a model system in developmental biology. Examining the developmental basis of pattern formation using molecular techniques, the dorsal patterning of embryonic quail is likely due to activation of the melanocortin-1 receptor, which is a highly conserved pathway in vertebrates. I examined whether a reaction-diffusion based theoretical model of pattern formation may predict developmental constraint in two groups that have different lifestyles and spectacular patterns: waterfowl (Anseriformes) and gamebirds (Galliformes). Tracing the evolutionary trajectory of pattern evolution with Bayesian comparative modeling there was evidence for developmental constraint in pattern evolution. Adaptive explanations may also result in convergence. Cuckoo-hawk mimicry has been demonstrated in the common cuckoo (Cuculus canorus) and the Eurasian sparrowhawk (Accipiter nisus), but may be prevalent in Old World cuckoos. Randomly selecting a parasitic cuckoo from each genera of Old World cuckoos and <8 sympatric raptors, I quantified their barred patterns using digital image analysis and found that parasitism can explain convergent evolution in the patterns of parasitic cuckoos and raptors. Patterns may have evolved due to ecological selection. Examining the patterns of 80% of all avian species worldwide, I found that habitat does not predict patterning, and that all four patterns are found in all habitats. These results demonstrate that the mechanisms of convergent evolution are diverse, and that development and natural selection have contributed to pattern evolution.

Evolution of the Neckeraceae (Bryopsida)

The group of pleurocarpous mosses comprises approximately 5000 species, which corresponds to about half of all mosses. The pleurocarpous mosses (i.e. “the Core Pleurocarps”) form a monophylum, which consists typically of perennial mosses with creeping stems and abundant lateral branches. In pleurocarpous mosses the archegonium and thus also sporophyte development is restricted to the apices of short, specialized lateral branches, in contrast to most other mosses, where archegonia and sporophytes develop terminally on the main axis (acrocarpous) or on major branches (cladocarpous). Traditionally, pleurocarpous mosses have been divided into three orders based mainly on their sporophytic characters. Brotherus described the Neckeraceae in 1925 and placed it into the Leucodontales, later the family has alternatively been divided into two or three separate families: the Thamnobryaceae, the Neckeraceae and the Leptodontaceae. These families have been placed even in different orders (Neckeraceae and Leptodontaceae among the leucodontalean mosses and Thamnobryaceae among hypnalean mosses) according to their peristome structure and the grade of peristome reduction. A growing amount of evidence indicates that a grouping based on sporophytic characters is artificial and based on convergent evolution. According to the latest phylogenetic studies of pleurocarpous mosses, based on molecular data, the Neckeraceae belong to the order Hypnales and share a sister group relationship with the Lembophyllaceae. In the most recent comprehensive classification 28 genera were included in the Neckeraceae family. This classification was based on both morphological and molecular data, but done with limited taxon sampling that did not cover all species of the family. Some previous studies based on molecular data have challenged the family concept of the Neckeraceae, indicating the need for a revision of the family. Here the family concept of the Neckeraceae is revisited, the closest relatives of the family are resolved and its position within pleurocarpous mosses is shown. In addition, new insights into the morphological evolution of the family are provided. Previous phylogenetic studies indicated that branch lengths among pleurocarpous mosses are usually extremely short. Therefore we chose to use mainly non-coding DNA sequences from rapidly evolving DNA regions. The phylogenetic reconstructions are based on extensive sequence data from all genomes: plastid trnS-trnF and rpl16, nuclear ITS1 &amp;amp; 2 and mitochondrial nad5. Both parsimony (PAUP and PRAP2) and Bayesian statistics (MrBayes) were employed for phylogenetic reconstructions. In order to use the information provided by length mutations indels were included in the analyses as binary data using a simple indel coding approach. No severe conflicts appeared between the different methods used, but the indel coding affected the support values of the inferred topologies. Therefore, all support values resulting from different methods are shown along the phylogenetic trees. The morphological features are studied and synapomorphies for each clade formed in the phylogenetic analyses are interpreted. A new delimitation of the family makes it necessary to reconsider the relevance of the morphological description and the morphological features characteristic of the family need to be reconsidered. Due to new groupings, some changes in the morphological circumscriptions of the genera are necessary, resulting in two new genera and several new combinations. Chapter 1 gives a broad overview of the relationships of the pleurocarpous mosses and shows the need for changes in the definition of genera, families and the corresponding nomenclature in this group. Chapter 2 is a population genetic study on the genus Thamnobryum. The main aim of this chapter is to test the species concept in Thamnobryum that are endemic to strictly restricted regions showing only minor differences in the morphological features in comparison to some more common species. In Chapter 3 the monophyly of the Neckeraceae is tested. In addition, in this chapter the ancestral character states of some morphological characters within the Neckeraceae are reconstructed. Chapters 4 and 5 resolve the genus composition and the relationships within the family in more detail. The results of this thesis show that the Neckeraceae need re-circumscription; this includes changes in the genus composition. The Lembophyllaceae is confirmed to be the sister group of the Neckeraceae. In addition to the new phylogeny, the potential evolution of several characters as a result of environmental selection pressures is analyzed. From the ancestral state reconstructions made (using BayesTraits) for both the habitat and a selection of morphological characters, character state distributions and habitat shift appear congruent, peristome reduction being a good example. However, some character states do not correlate with the habitat, suggesting very complex evolutionary patterns underlying these morphological characters. Many widely distributed genera that are composed of several species and seem to be morphologically coherent (Echinodium, Homalia, Thamnobryum, partly Neckera), are shown in this thesis to be polyphyletic. They are replaced by smaller, geographically more restricted genera that at least in some cases (e.g. Thamnomalia, Homalia s.str., Neckera s.str.) seem to form morphologically heterogeneous genera. In other words, morphology can be misleading in the family Neckeraceae even at the genus level and convergent evolution in both morphological and sequence level characters are common within the family. Special habitat conditions have been shown to result in similar morphological structures also in several other moss groups. This kind of convergent evolution occurs in aquatic mosses, and seems to have occurred among the neckeraceous species Thamnobryum alopecurum and its allies. However, similar morphological structure in similar aquatic habitats can also be due to true phylogenetic relationships as is the case within the Neckeraceae for Handeliobryum sikkimense and Hydrocryphae wardii, or the members of Touwia. The geographical grouping seems to be more strongly correlated with the phylogenetic grouping than thought before.

Convergent evolution of humeral and femoral functional morphology in slow arboreal mammals

Ökomorphologische Konvergenz tritt auf, wenn Arten mit demselben Lebensstil unabhängig voneinander ähnliche morphologische Merkmale evolvieren. Neue Fallstudien können dabei helfen, die diesem Prozess zugrunde liegenden Mechanismen aufzuklären. Diese Arbeit befasst sich mit einigen konvergent evolvierten, langsamen, baumbewohnenden Säugetieren, d. h. zwei Linien baumlebender Faultieren, dem Zwergameisenbär, den Lorisiden, zwei Kladen ausgestorbener Lemuren, d. h. Paläopropithekiden und Megaladapis, und dem Koala. Es werden funktionsmorphologische Konvergenzen in diesen Taxa erforscht, indem ihr Humerus und ihr Femur sowie jene ihrer nah verwandten, aber ökologisch unterschiedlichen Taxa untersucht werden. Erstmals werden Knochen mittels phylogenetisch vergleichender Methoden auf vier anatomischen Ebenen, d.h. äußere Form, diaphysäre Mikrostruktur und Anatomie sowie epiphysäre Trabekelarchitektur, analysiert. Viele langsame, baumbewohnende Säugetiere teilen eine geringe kortikale Kompaktheit, was wahrscheinlich mit ihrer extrem niedrigen Stoffwechselrate und ihren biomechanischen Anforderungen zusammenhängt. Langsame, arboreale Xenarthra, d. h. baumbewohnende Faultiere und der Zwergameisenbär, weisen ein Muster unvollständiger Konvergenz für eine Reihe äußerer und innerer anatomischer Merkmale auf, was möglicherweise durch die relativ unterschiedliche Ökologie des Zwergameisenbären erklärt wird. Auf einer breiteren Säugetierskala konvergieren andere Merkmale, die möglicherweise mit der Ökologie der langsamen baumbewohnenden Lebensweise in einigen der untersuchten Taxa zusammenhängen, obwohl komplexe Muster auch durch andere evolutionäre Prozesse erklärt werden können. Nur suspensorisch lebende Taxa tragen signifikant zur Konvergenz bei. Diese Arbeit hebt die stärkere Konvergenz hervor, die sich in der inneren Struktur des Knochens widerspiegelt. / Ecomorphological convergence occurs when similar morphological traits are independently evolved by species with the same lifestyle. Novel case studies can help to elucidate the underlying mechanisms of this process. This work addresses some convergent slow arboreal mammals, i.e. two lineages of ‘tree sloths’, the silky anteater, ‘Lorisidae’, two clades of extinct lemurs, i.e. palaeopropithecids and Megaladapis, and the koala. Functional morphological convergences are searched in these taxa, studying their humerus and femur as well as those of their closely related ecologically distinct taxa. For the first time, bones are analyzed at four anatomical levels, i.e. external shape, diaphyseal microstructure and anatomy and epiphyseal trabecular architecture, through phylogenetic comparative methods. Many slow arboreal mammals share a low cortical compactness, probably related to their extremely low metabolic rate and biomechanical demands. Slow arboreal xenarthrans, i.e. ‘tree sloths’ and the silky anteater, exhibit a pattern of incomplete convergence for a set of external and internal anatomical features, possibly explained by the relatively distinct ecology of the silky anteater. On a wider mammalian scale, other traits possibly related to slow arboreal ecology converge in some of the studied taxa, although with complex patterns also explained by other evolutionary processes. Only suspensory taxa significantly contribute to convergence. This thesis highlights the stronger convergence reflected by bone internal structure. By providing potential explanations for convergence in slow arboreal mammals, the inherent complexity of this process is here emphasized.

An interdisciplinary approach to describing biological diversity

The concept of biodiversity – the phenotypic and genotypic variation among organisms – is central to conservation biology. There is growing recognition that biodiversity does not exist in isolation, but rather is intrinsically and evolutionarily linked to cultural diversity and indigenous knowledge systems. In Canada, caribou (Rangifer tarandus) occupy a central place in the livelihoods and identities of indigenous people and display substantial variation across their distribution. However, quantifying caribou intraspecific variation has proven challenging. Interdisciplinary approaches are necessary to produce effective species characterizations and conservation strategies that acknowledge the interdependent relationships between people and nature in complex social-ecological systems. In this dissertation I use multiple disciplinary traditions to develop comprehensive and united representations of caribou variation through an exploration of population genetics, phylogenetics, traditional knowledge, language, and visual approaches in the Sahtú region of the Northwest Territories, Canada. First, I examine caribou variation through analysis of population genetics and the relationships Dene and Métis people establish with animals within bioculturally diverse systems. Next, I focus on how the Pleistocene glacial-interglacial cycles have shaped the current patterns of caribou phylogeographic lineage diversification. Finally, I explore how art can be used to facilitate cross-cultural collaboration and externalize the unique heterogeneity of biocultural diversity. The results demonstrate a broad scale understanding of the distribution, spatial organization, and the degree of differentiation of caribou populations in the region. I found evidence for caribou population differentiation that corresponds to the caribou types recognized by Dene people: tǫdzı “boreal woodland caribou,” ɂekwę́ “barren-ground caribou,” and shúhta ɂepę́ “mountain caribou.” Phylogenetic results reveal that in their northern margin the boreal ecotype of woodland caribou evolved independently from the northern Beringian lineage in contrast with southern boreal caribou which belong to the sub-Laurentide refugia lineage. In addition, I demonstrate how art can be used improve communication, participation, and knowledge production among interdisciplinary research collaborations and across language and knowledge systems. A collaborative process of research that facilitates łeghágots'enetę “learning together” has the potential to produce sustainable conservation solutions, develop efficient and effective wildlife management policies, and ensure caribou remain an important part of the landscape. / February 2017

Développement d’outils moléculaires et cellulaires pour générer des variétés de Pomelo « Star Ruby » ne produisant pas de Furocoumarines / Development of molecular and cellular tools to generate Star Ruby grapefruit varieties non producing furanocoumarins

Les furocoumarines sont des composés phénoliques impliqués dans la défense contre les herbivores. Ces molécules sont majoritairement décrites dans quatre familles botaniques, notamment les Rutaceae, dont font partie les agrumes. Ces molécules sont phototoxiques ce qui peut poser des problèmes pour leur utilisation comme par exemple en cosmétique ou en phytothérapie. D’autre part, en cas d’ingestion par exemple via la consommation de jus de certains agrumes, elles ont responsables de l’inhibition d’enzymes de détoxication comme le CYP3A4 humain. Cela peut conduire à des surdosages médicamenteux connus sous le nom d’Effet Pomelo. Ce travail de thèse a consisté à réfléchir et à développer, des outils qui permettront de générer de manière ciblée des variétés de pomelo qui ne produisent plus de furocoumarines. Nous avons abordé l’ensemble des étapes essentielles pour la mise en place d’une stratégie global : i) des méthodes reproductibles ont été développées pour la production de protoplastes et de cultures cellulaires de pomelo Star Ruby ; ii) des conditions de transformation de protoplastes par électroporation ont également été mises au point ; iii) finalement, pour inhiber de manière spécifique la voie de biosynthèse des furocoumarines, nous avons choisi de mettre en œuvre une approche d’édition de génome en utilisant une méthodologie CRISPR/Cas9. La mise au point de la méthode a été réalisée avec un gène codant pour une umbelliferone 6-dimethylallyl transférase. Les résultats obtenus indiquent que la stratégie est envisageable. Pour renforcer la stratégie CRISPR/Cas9, nous avons mis en œuvre une démarche d’identification de gènes cibles additionnels. En utilisant une approche de data mining de bases de données génomiques et transcriptomiques nous avons identifié 18 séquences candidates, potentiellement impliquées dans la voie de biosynthèse des furocoumarines. L’expression hétérologue des protéines correspondantes et leur caractérisation fonctionnelle a permis de montrer que CYP706J12 est en mesure de métaboliser l’hérniarine, une coumarine. Ce résultat apporte des éléments pour émettre des hypothèses sur l’évolution convergente de la synthèse des coumarines et des furocoumarines chez les végétaux supérieurs. / Furanocoumarins are phenolic compounds involved in defense against herbivores. These molecules are mainly described in four botanical families. Rutaceae, one of those families, includes Citrus species. Furanocoumarins are phototoxic compounds, which can be problematic for their use in cosmetics or in phytotherapy. Furanocoumarin ingestion via citrus juice consumption, may inhibit human enzymes of detoxification, such as human CYP3A4. This can lead to drug overdoses known as the “Grapefruit Juice Effect”. This work consisted in the development of tools that will allow to generate new varieties of pomelo that no longer produce furanocoumarins by targeted genome edition. We have covered the essential steps for the implementation of a global strategy: i) reproducible methods have been developed for the production of protoplasts and cell cultures of Star Ruby grapefruit; ii) conditions for protoplast transformation by electroporation have also been developed; iii) finally, to specifically inhibit the furanocoumarin biosynthetic pathway, we chose to implement a genome editing approach using a CRISPR / Cas9 methodology. The development of the method was carried out with a gene encoding umbelliferon 6-dimethylallyltransferase. The results obtained indicate that the strategy is feasible. To strengthen the CRISPR / Cas9 strategy, we implemented a method to identify additional target genes. Using a data mining approach of available genomic and transcriptomic databases we identified 18 candidate sequences potentially involved in the furanocoumarin biosynthetic pathway. Heterologous expression of the corresponding proteins and their functional characterization made it possible to show that CYP706J12 is able to metabolize herniarin (a coumarin). This result provides elements to hypothesize about the convergent evolution of coumarin and furanocoumarin synthesis in higher plants.

L'enzyme bactérienne β-lactamase hydrolyse l'antibiotique β-lactame en cassant la structure de base "anneau β-lactame". Pour identifier les nouvelles β-lactamases, une étude complète a été réalisée dans diverses bases de données biologiques telles que Human Microbiome Project, env_nr et NCBI nr. L'analyse a révélé que les séquences ancestrales putatives et les recherches de profil HMM jouaient un rôle important dans l'identification de la base de données homologue et métagénomique à distance dans l'enzyme β-lactamase existante comme matière noire. Les larges analyses phylogénétiques des β-lactamases existantes et nouvellement identifiées représentent les nouveaux clades dans les arbres. En outre, l'activité d'hydrolyse des antibiotiques β-lactamines de séquences nouvellement identifiées (provenant d'archées et d'humains) a été étudiée en laboratoire, ce qui montre l'activité de la β-lactamase. La deuxième phase de l'étude a été entreprise pour examiner l'évolution fonctionnelle des β-lactamases. Premièrement, des séquences de protéines ß-lactamase 1155 ont été extraites de la base de données ARG-ANNOT et des valeurs CMI la littérature correspondante. Les résultats ont révélé que l'activité fonctionnelle de la β-lactamase évoluait de manière convergente au sein de la classe moléculaire. La troisième phase de cette thèse représente le développement d'une base de données intégrative de β-lactamases. La base de données publique actuelle de β-lactamases a des informations limitées, par conséquence, une base de données intégrative a été développée. / The β-lactam antibiotics are one of the oldest and widely used antimicrobial drugs. The bacterial enzyme β-lactamase hydrolyzes the β-lactam antibiotic by breaking the core structure “β-lactam ring”. To identify the novel β-lactamases a comprehensive investigation was performed in different biological databases such as Human Microbiome Project, env_nr, and NCBI nr. The analysis revealed that putative ancestral sequences and HMM profile searches played a significant role in the identification of remote homologous and uncovered the existing β-lactamase enzyme in the metagenomic database as dark-matter. The comprehensive phylogenetic analyses of extant and newly identified β-lactamase represent the novel clades in the trees. Further, the β-lactam antibiotic hydrolysis activity of newly identified sequences (from archaea and human) was investigated in laboratory, which shows β-lactamase activity.The second phase of the investigation was undertaken to examine the functional evolution of β-lactamases. First, 1155 β-lactamase protein sequences were retrieved from ARG-ANNOT database and MIC values from the corresponding literature. The results revealed that the functional activity of β-lactamase evolved convergently within the molecular class.The third phase of this thesis presents development of an integrative β-lactamase database. The existing public database of β-lactamase has limited information, therefore, an integrative database was developed.

Fundamental and applied studies of Ixodes ricinus salivary proteins / Etudes fondamentales et appliquées de protéines salivaires de la tique Ixodes ricinus

De nombreuses études suggèrent que linhibition de la voie alterne du complément de lhôte est nécessaire aux parasites hématophages pour leur permettre daccomplir leur repas sanguin. Une revue décrivant ces études a été publiée dans Developmental and Comparative Immunology (Schroeder et al., (2009) Dev. Comp. Immunol. 33, 5-13). Plusieurs études suggèrent que linhibition de la voie alterne de lhôte par les protéines salivaires de tiques est importante pour lacquisition du repas sanguin et la transmission subséquente de pathogènes à lhôte infesté. Confortant cette hypothèse, une protéine salivaire capable dinhiber la voie alterne du complément a été clonée chez la tique américaine Ixodes scapularis (Valenzuela et al., (2000) J. Biol. Chem. 275, 18717-18723). Cette protéine, appelée Isac, ne présente aucune homologie avec les autres molécules anti-complément connues à ce jour, suggérant que cette protéine a été acquise au cours de lévolution par un mécanisme dévolution convergente. En plus de cet aspect fondamental, Isac représente un candidat antigénique prometteur pour le développement dun vaccin anti-tique potentiellement capable dinduire le rejet de la tique et/ou de prévenir la transmission des pathogènes. Le but initial de cette thèse était didentifier le ou les homologue(s) de la protéine Isac de la tique américaine Ixodes scapularis chez la tique européenne Ixodes ricinus. De façon intéressante, deux séquences différentes ont été isolées du transcriptome des glandes salivaires de la tique I. ricinus (Daix et al., (2007) Insect Mol. Biol. 16 (2), 155-166). Lexpression de ces séquences a révélé quelles codent pour des protéines secrétées capables dinhiber la voie alterne du complément. Ces protéines ont été appelées IRAC I et II pour « Ixodes ricinus anti-complement protein I and II ». La caractérisation des IRACs à laide danticorps monoclonaux a permis de révéler que ces deux protéines sont exprimées de façon constitutive au sein des glandes salivaires de la tique Ixodes ricinus et sont sur-exprimées au cours du repas sanguin. Létude de lexpression des protéines IRAC I et IRAC II au sein de la population dIxodes ricinus a révélé que ces deux protéines sont des paralogues codés par des gènes différents et non par des allèles dun même locus. Enfin, des analyses phylogéniques portant sur les différentes séquences codant pour les protéines homologues à Isac isolées chez les tiques Ixodes scapularis, Ixodes ricinus et Ixodes pacificus ont révélé que les tiques appartenant au complexe Ixodes ricinus codent pour une famille encore non décrite de molécules anti complément qui se sont diversifiées au cours de lévolution par un processus de sélection darwinienne positive. Les analyses phylogéniques des IRACs suggèrent que ces séquences ont subi une diversification par un processus de pression de sélection darwinienne positive, menant probablement à des molécules possédant des propriétés biologiques différentes. Dans la seconde étude, nous avons testé lhypothèse de travail selon laquelle chaque paralogue pourrait posséder des activités inhibitrices différentes à lencontre du complément de différentes espèces dhôtes naturels, contribuant ainsi à élargir le spectre dhôte des tiques I. ricinus. Les résultats obtenus démontrent que cette hypothèse est correcte (Schroeder et al., (2007) Microbes Infect. 9 (2), 247-250). Dans la troisième et dernière étude de ce manuscrit, nous avons testé le potentiel des protéines IRACs comme candidat antigénique pour le développement dun vaccin anti tique. Des recombinants de lherpèsvirus bovin 4 (BoHV-4) exprimant IRAC I ou IRAC II ont été produits. De façon intéressante, nous avons observé que bien que les recombinants BoHV-4 expriment de hauts taux de protéines IRACs fonctionnelles in vitro, les lapins immunisés à laide des recombinants BoHV-4 exprimant les IRACs ne développent pas de réponse humorale détectable à lencontre des transgènes. Dans le but daugmenter limmunogénicité des IRACs exprimés comme transgène, une seconde génération de recombinants BoHV-4 a été produite. Ceux-ci induisent lexpression des IRACs sous la forme de protéines de fusion transmembranaires à la surface des cellules infectées. Linoculation de ces recombinants à des lapins a engendré le développement dune forte réponse humorale à lencontre des IRACs. Néanmoins, cette réponse immune na pas engendré deffet majeur sur le repas sanguin de femelles Ixodes ricinus placées sur les lapins immunisés. / An increasing number of studies demonstrate that inhibition of host complement activation is crucial for completion of the blood feeding process of hematophagous parasites. A review of these studies has been published in Developmental and Comparative Immunology (Schroeder et al., (2009) Dev. Comp. Immunol. 33, 5-13). Several observations suggest that inhibition of the host complement alternative pathway by tick salivary proteins is crucial for the achievement of blood feeding and efficient transmission of the pathogens transmitted by the parasite. Strongly supporting this conclusion, a salivary protein able to inhibit the alternative pathway was cloned from the American tick Ixodes scapularis (Valenzuela et al., (2000) J. Biol. Chem. 275, 18717-18723). Interestingly, this molecule, termed Isac, has no similarity to any previously reported anti-complement molecules suggesting that it has been acquired through a mechanism of convergent evolution. In addition to this fundamental aspect, Isac is also a promising candidate antigen for the development of an anti-tick vaccine potentially able to induce the reject of the tick and/or to prevent the transmission of the pathogens. The initial goal of the present work was to clone the orthologue of Isac from the European tick Ixodes ricinus. Interestingly, two different sequences were isolated from the transcriptome of I. ricinus salivary glands (Daix et al., (2007) Insect Mol. Biol. 16 (2), 155-166). Expression of these sequences revealed that they both encode secreted proteins able to inhibit the complement alternative pathway. These proteins were called I. ricinus anticomplement (IRAC) protein I and II. Further characterization of IRACs using monoclonal antibodies revealed that both proteins are expressed constitutively in I. ricinus salivary glands and are up-regulated during blood feeding. Analysis of a series of individual ticks revealed that all ticks tested express both IRAC I and IRAC II, demonstrating that they are the products of different genes and not of alleles of the same locus. Finally, phylogenetic analyses of the I. ricinus IRAC I and II sequences together with homologues from I. scapularis and I. pacificus demonstrates that ticks belonging to the Ixodes ricinus complex encode a new family of relatively small anti-complement molecules undergoing diversification by positive Darwinian selection. Phylogenetic analyses of IRACs suggested that these sequences were diversifying by a process of positive Darwinian selection, possibly leading to molecules with different biological properties. In the second study, we tested the hypothesis that each paralogue may have different inhibitory activities against the complement of different natural host species, thereby contributing to broaden the host range of I. ricinus ticks. The data obtained demonstrated that this working hypothesis is correct (Schroeder et al., (2007) Microbes Infect. 9 (2), 247-250). In the third and last chapter of the present manuscript, we addressed the potential of IRAC I and II as candidate antigens for the development of an anti-tick vaccine. Bovine herpesvirus 4 (BoHV-4) recombinants expressing IRAC I or II were produced. Interestingly, we observed that although both recombinants expressed high levels of functional IRAC proteins in vitro, our attempts to immunize rabbits against IRACs via infection with these viruses invariably failed. In order to improve the immunogenicity of IRACs expressed as transgene, a second generation of BoHV-4 recombinants was produced. The latter expressed IRACs as transmembrane fusion proteins on cell surface. Comparison of the vaccine potential of BoHV-4 recombinant viruses expressing either secreted or transmembrane IRAC proteins revealed that while the former did not induce a detectable immune response against IRACs, the latter led to high titers of anti-IRAC antibodies. However, the immune response induced against IRACs did not lead to the reject of the tick but only slightly increased the duration of the blood feeding process.
