Development of a user interface design tool : Bombardier Transportation HMI Designer 3.0 ConceptStröm, Per, Gidlund, Elisabet January 2012 (has links)
This report aims to describe the process of developing a prototype for the new version of Hmi Designer developed by Bombardier Transportation. This designer tool is used to create the graphical user interface on a train control computer. A new version of the design tool is necessary because the remaining part of Bombardier's suite of application development has changed both the programming environment and programming language. The prototype developed should illustrate the possibilities at a later stage, performing a full-scale project. The prototype is implemented as a plugin in Qt Creator, hence the described approach for developing plugins in the programming environment and the advantages and disadvantages that are given, with a switch to Qt Creator. This work has resulted in the prototype with the capability to load existing project files, view and modify the design and save the project in its native format. The work has reached the expected result; it is possible to build a design tool based on Qt Creator with the requested functionality. The evaluation at Bombardier showed that the prospect of this project is very positive which gives Bombardier excellent opportunities to launch a full scale project.

Qualitative Detection of Selected Designer Drugs and Relevant Metabolites in Environmental Water SamplesPruyn, Marley 14 July 2016 (has links)
Designer drugs are compounds which have been synthetically derived from illicit drugs. After consumption, drugs and their metabolites are introduced into the sewage water which is treated and disposed into the environment. A combined target, suspect and non-target workflow was created to detect designer drugs in environmental water samples. Multiple water samples were spiked with an unknown mixture of drugs and metabolites to assess the efficiency of the method. Samples were collected from sewage influent and effluent pipes, downstream from a sewage outfall and reclaimed water. Analysis was conducted with high resolution MS using the QExactive Orbitrap. Screening was performed using a database compiled in-house using TraceFinder EFS. Structure confirmation was achieved using MassFrontier. Target drugs and their metabolites were detected in sewage influent but not in sewage effluent, downstream of the effluent pipe, or in reclaimed water. The workflow was adequate to detect designer drugs in multiple water matrices at concentrations as low as 20ppt.

Synthetic Cannabinoid Usage among College Students: The Example of K2 and SpiceStephens, Jason L. 08 1900 (has links)
The primary goal of this study was to investigate the awareness and prevalence of Spice and K2 usage among a population of college students, as well as the demographics of such users. The study also sought to determine whether or not students prefer these products over natural cannabis, in addition to examining the most popular methods of obtainment and the most commonly reported side effects of K2 and Spice usage. Participants consisted of 643 undergraduate students enrolled at the University of North Texas during the fall 2011 semester. Findings indicate that while students exhibit a relatively high awareness of K2 and Spice, usage of these products is not a prevalent occurrence. Implications of the findings are discussed.

A Navigation Mesh-Based Pathfinding Implmentation in CET Designer : An Alternative to a Waypoint Graph-Based SolutionKarlsson, Markus January 2021 (has links)
This thesis details the implementation of a navigation mesh within the interior space planning software CET Designer. The motivation for this is to investigate whether a navigation mesh is a suitable alternative to its current path finding solution which is based on a waypoint graph. An empirical analysis between the navigation mesh and the waypoint graph was also conducted. A navigation mesh is successfully implemented and an alternative triangulation method is also tested. The alternative method which is referred to as "narrowest first-ear clipping" seems to create a navigation mesh that produces slightly shorter paths than a navigation mesh that utilizes an arbitrary triangulation method. It is argued that the navigation mesh alleviates much work from the user since it requires less user input. The mesh is also able to generate paths between any two given points within it. This makes it much more dynamic than the waypoint graph. The conducted analysis shows that the navigation mesh seems to generate paths that are generally shorter than the waypoint graph. The path generation time for the navigation mesh is on average slower than the waypoint graph. We failed to provide an environment where the navigation mesh was faster on average than the waypoint graph. However, the time difference between the navigation mesh and the waypoint graph seemed to reduce as the test environment size increased.

Elucidating Mechanisms of Chromatin Crosstalk Using ‘Designer’ NucleosomesYerkesh, Zhadyra 04 1900 (has links)
The molecular target of epigenetic signaling is chromatin. Histones are extensively post-translationally modified (PTM), and many of these individual modifications have been studied in depth. As PTMs occur at multiple positions within histones, the degree to which these modifications might influence each other remains one of the major challenges of chromatin biology. Although major discoveries in understanding the complex repertoire of histone modifications were achieved using reductionist experimental systems with synthetic histone peptides, they do not explain the role of putative PTM cross-talks in a chromatin context. However, generating chromatin substrates of defined modification status has proved to be a technically challenging task.
In this thesis, I first demonstrate our work on establishing a novel approach to produce libraries of modified nucleosomes. We employed protein trans-splicing and sortase-mediated ligation strategies to incorporate chemical modifications on histone tails of ‘ligation-ready’ nucleosomes. Subsequently, the ‘designer’ nucleosome libraries were used for testing the binding of heterochromatin protein 1 (HP1) and elucidated the previously uncharacterized crosstalk of H3K9me2 and S28ph marks. Further investigations explained the mechanism of this crosstalk and highlighted the importance of developing chemical biology tools for elucidating complex chromatin signaling.
Second, I describe our reconstitution systems for the assembly of semisynthetic recombinant chromatin carrying methylation marks on DNA and distinct modifications on histones, e.g. H3K9me3. I aimed to understand the mechanisms of the interplay between chromatin and one of the DNA maintenance methylation factors, UHRF1. I showed that UHRF1 strongly interacts with nucleosomes containing linker DNA. However, it exerts only residual enzymatic activity in this context. Based on functional H3 ubiquitylation assays in vitro, I found that hemi-methylated nucleosomes stimulate enzymatic activity of UHRF1, suggesting that the protein’s chromatin targeting and activation are a two-step process. The positioning of hemi-methylated CpG on nucleosome regulates UHRF1 target selectivity. Further, mutational analysis revealed that the PHD domain of the factor is indispensable for H3 binding and that its SRA domain is required for catalytic activation. Overall, our work adds a new layer of positional complexity to the me½CpG-dependent regulation of UHRF1 and expands the current model of DNA methylation maintenance.
