Process Planning for Hybrid Manufacturing with Directed Energy Deposition and Machining Processing

This thesis details the creation and application of a generalized process plan for the hybrid manufacturing of AISI 316L stainless steel, using direct energy deposition (DED) and ball-nose end-mill machining, that includes the inspection and measurement of objects created by that hybrid manufacturing process plan. The proposed process plan progresses through the selection of substrate thickness, single-track, multi-track, and multi-layer depositions, then on to machining processing. A manufacturers' recommended set and range of DED parameters were used to create a designed experiment that aided in the analysis of objects created in each of the DED process planning steps; those objects were then machined in the same enclosure using a set of machining parameters screened from industry recommendations for ball-nose milling of stainless steel, after which measurements were taken for surface roughness, some material characteristics, and for tool deterioration. The results, analyses, and discussions collected herein show that the proposed process plan can provide models for geometrical outputs for each step in the plan, some improvements in substrate stability, surface roughness, tool deterioration, and material porosity due to voids. Current research in hybrid manufacturing does not show generalized process planning influences. The process plan as demonstrated by the work in this thesis will help operators, designers, and researchers in the future by defining a generalized workflow that can be applied to other materials used in hybrid manufacturing.

Ecology of northern long-eared bats (Myotis septentrionalis) in a coastal setting after the introduction of White-nose Syndrome

Northern long-eared bat (Myotis septentrionalis) populations have declined sharply in recent years due to the introduction and rapid spread of White-nose Syndrome. This has prompted an urgent need for a greater understanding of their natural history in order to support the conservation of extant populations, particularly where forested day-roost and foraging habitats are being fragmented by development. Prior to 2006, with other Myotis species on the endangered species list, northern long-eared bats were understudied. In recent years, with the pressing concern to document the ecology of all cave bats affected by White-nose Syndrome on the landscape, researchers have now prioritized northern long-eared bat habitat needs, day-roost use, social dynamics, and barriers to gene flow. A relatively new discovery has been the numerous coastal populations that occur in smaller forest patches often surrounded by anthropogenic development. The goal of my research is to aid in informing future monitoring and management protocols that are specific to northern long-eared bats, particularly as they may shift from Threatened to Endangered under the Endangered Species Act. The chapters of this dissertation explore (1) similarities and differences between coastal and more documented upland habitat associations for populations of northern long-eared bats throughout the summer months through the use of acoustic detectors; (2) relationships between acoustic activity patterns and weather on an hourly basis for several bat species on Long Island, New York; and (3–4) day-roost characteristics and social network dynamics of a coastal northern long-eared bat maternity colony on Long Island, New York. I found that northern long-eared bat colonies have larger spatial footprints in – and a moderate tolerance for – forests in anthropogenically-developed areas than previously believed, though many natural history aspects of the bat were similar among coastal and interior populations. Installation of artificial day-roost structures such as bat boxes would likely be used and highly beneficial to colonies in these conditions, particularly to offset the deleterious effects of stochastic disturbance events on day-roost availability and to support social cohesion (and, by extension, reproductive success) for coastal maternity colonies. Additionally, I found that acoustic activity increased in riparian areas, or at sites closer to water features, and as proportion of forest coverage increased at a broad scale. However, immediately around the detector this relationship was reversed. Taken together, this demonstrates that riparian corridors, water features, and forest structural heterogeneity should explicitly be included in management guidelines. / Doctor of Philosophy / White-nose Syndrome is a fungal disease that has killed millions of cave bats in North America, including the northern long-eared bat (Myotis septentrionalis) that during the spring through fall uses forests for roosting and foraging habitat. As the disease continues to spread, and as forests are increasingly being lost to urbanization, it is imperative for researchers and land managers to understand the landscape needs of this species so that they can retain those forests that are used by this species. In addition to broader landscape needs, it is important to understand what specific forest stand to tree features the bats are using, how their social dynamics support their reproductive success, and how to best support habitat conditions foster mating in the fall swarm to avoid inbreeding. In recent years, more northern long-eared bats have been documented in coastal landscapes, suggesting that these areas might be more important to the conservation of the species than previously believed. In this dissertation, my aim was to explore how or why the landscapes in these coastal areas were being used by these bats both during nightly foraging and during the day as resting areas (day-roosts). Additionally, I explored how multiple species of bats were responding to weather conditions on the coast across seasons. Overall, I found that complex forests continue to be an important resource for northern long-eared bats, as well as water features whereby the latter should be more explicitly included in conservation management plans. Northern long-eared bats also show an ability to successfully use small forest patches within a larger urbanized landscape, including using human structures for day-roosts. Human structures might provide reliable, multi-year day-roosts in areas where extreme coastal weather events (hurricanes) occur or increase or as natural processes reduce the suitability of these forests to provide day-roosts. These structures, particularly artificial roost boxes could also support the social network of reproductive northern long-eared bats and thus maintain population stability to some extent.

La pollution atmosphérique est omniprésente du fait de nombreuses sources émettrices de composés chimiques. Dans ce contexte, les Hydrocarbures Aromatiques Polycycliques (HAP) sont largement diffusés dans l’air et ont déjà montré des effets délétères sur la santé.Ce travail a consisté en l’évaluation de la neurotoxicité du fluorène, composé choisi comme molécule représentative de la pollution liée aux HAP, chez le rat adulte exposé par voie i.p., par voie orale ou par inhalation. Le modèle d’exposition par inhalation a ensuite été appliqué à l’étude des effets du polluant sur le développement sensori-moteur et l’activité comportementale de l’animal exposé in utero ou au cours de la lactation.Ces études ont montré que le fluorène était susceptible d’affecter le niveau d’anxiété et l’activité locomotrice du rat adulte exposé directement et indirectement au polluant, et n’avait aucun effet sur les capacités d’apprentissage. En revanche, aucune atteinte majeure de la maturation des fonctions sensori-motrices n’a été mise en évidence. L’analyse de la présence du composé et de trois de ses métabolites a par ailleurs montré que le polluant était capable de traverser la barrière hémato-encéphalique et d’être métabolisé au niveau du cerveau. Les réponses physiologiques et comportementales étant variables entre les études, les effets induits par le fluorène dépendent donc de la voie d’entrée, du niveau de contamination et du moment d’exposition.Ces résultats ont ainsi révélé chez l’animal la toxicité comportementale du fluorène à des niveaux de contamination réalistes, confirmant ainsi le risque sanitaire de l’exposition aux HAP tout au long de la vie des individus / Actually air pollution is ubiquitous due to the emission of chemical compound from many sources. In this context, Polycyclic Aromaric Hydrocarbons (PAH)related compounds are widely distributed in the air and have shown deleterious health effects.Fluorene was chosen as a representative compound of PAHs pollution. This work consisted in the evaluation of its neurotoxic effects in adult animals, exposed i.p., orally or by exposure nose-only. The inhalation model of exposure was then applied to the study of its effects on sensorimotor development and on behavioral activity of animals exposed in utero or during lactation.The results showed that fluorene is able to induce behavioral changes in adult animals exposed directly or indirectly on the level of anxiety and the locomotor activity, whereas no effect on learning and memory abilities has been observed. However, no defect on the development of motor and sensory functions was demonstrated. Furthermore, the analysis of the presence of the compound and three of its metabolites showed that the pollutant was able to cross the blood brain barrier and can be metabolized in the brain. Moreover, variations have been observed concerning behavioral and physiological responses between studies, showing that effects induced by fluorene are dependent on the way of administration, the level of contamination and the time of exposure.In conclusion, these results demonstrate in animals the behavioral toxicity of fluorene at levels of contamination corresponding to human cases of exposure, confirming so the risk of PAH exposure throughout life

Impacto da composição química de microambientes em bens culturais: reatividade e monitoramento / Impact of microenvironmental chemical composition on heritage objects: reactivity and monitoring

Esta Tese de doutorado teve por objetivo aumentar a compreensão dos aspectos sinérgicos na corrosão de bens culturais feitos com chumbo (Pb) e avaliar a agressividade ambiental sobre bens culturais em igrejas e museus brasileiros para a proposição de estratégias de conservação. Neste contexto, foram investigados, em microambientes simulados, os efeitos sinérgicos entre formaldeído e ácido acético, ácido fórmico ou dióxido de carbono na corrosão de Pb; foi feita a síntese e caracterização de diferentes formiatos gerados nos processos de corrosão de Pb por formaldeído ou ácido fórmico; foi avaliada a agressividade ambiental sobre bens culturais da Igreja de São Francisco (São Paulo, ambiente urbano industrial), da Igreja dos Freis Carmelitas (Santos, ambiente urbano industrial costeiro), do Museu de Arte Contemporânea da USP (MAC, São Paulo, reserva técnica da nova sede) e do Museu de Arqueologia e Etnologia da USP (MAE, São Paulo, reserva técnica), através de cupons metálicos (chumbo e cobre) expostos nos ambientes interno e externo dessas instituições e através do uso de monitores miniaturizados; foi feita a otimização destes monitores e, finalmente, foi considerado o desenvolvimento de um nariz eletrônico (EN) baseado em microbalanças de quartzo (QCMs) para o uso em conservação preventiva. Nestas investigações microscopia Raman foi a principal técnica empregada, sendo que as demais foram FTIR-ATR, µ-FTIR-ATR, µ-FTIR, nano-FTIR (SINS), s-SNOM, AFM, SEM, SEM-EDX, XRD e SR-XRD. Os resultados apontaram para a necessidade de individualização dos objetos artísticos tanto para a proposição de estratégias de conservação quanto para o entendimento de sua reatividade, mostraram a agressividade ambiental de todas as instituições investigadas, indicaram a potencialidade de ENs em conservação preventiva e demonstraram a utilidade de técnicas de alta resolução lateral, como nano-FTIR, em investigações de corrosão metálica. Mais especificamente, as investigações de sinergia entre poluentes aumentaram o entendimento da reatividade de Pb e permitiram a proposição de esquemas mais completos de sua reatividade, levando a discussões acerca da conservação desses bens. A agressividade química ambiental das igrejas e museus a bens culturais foi inferida pela caracterização de cupons metálicos expostos em seus interiores e exteriores, enquanto que a agressividade relacionada a fatores físicos, como temperatura e umidade relativa, foi inferida através do uso de monitores miniaturizados expostos somente nas igrejas. Com esse monitoramento, estratégias de conservação foram propostas. Para a devida caracterização dos cupons metálicos, expostos nas igrejas e museus ou nos microambientes simulados, a síntese e a caracterização de diferentes formiatos de Pb foi essencial. Por fim, visando melhorar o desempenho dos monitores miniaturizados e ampliar sua aplicabilidade (com seu futuro uso como EN), um novo dispositivo foi construído e um estágio na Universidade de Aveiro, em Portugal, foi realizado para o desenvolvimento de um EN baseado em QCMs. / The objective of this doctoral thesis was to increase the understanding of synergistic aspects in the corrosion of cultural objects made with lead (Pb) and to assess the environmental aggressiveness on cultural objects of Brazilian churches and museums in order to propose conservation strategies. In this context, in simulated micro-environments, the synergistic effects between formaldehyde and acetic acid, formic acid or carbon dioxide in Pb corrosion were investigated; the syntheses and characterization of different formates generated by formaldehyde or formic acid in Pb corrosion processes was conducted; the environmental aggressiveness on cultural objects of the church of São Francisco (São Paulo, urban industrial environment), of the church of the Carmelite Friars (Santos, urban industrial littoral environment), of the Museum of Contemporary Art of the USP (MAC, São Paulo, new site) and of the Museum of Archaeology and Ethnology of the USP (MAE, São Paulo) was assessed exposing metal coupons (lead and copper) in their internal and external environments and using miniaturized monitors; these monitors were optimized and finally, the development of an \'electronic nose\' (EN) based on quartz crystal microbalances (QCMs) was considered for preventive conservation. In these investigations the principal technique employed was Raman microscopy, and FTIR-ATR, µ-FTIR-ATR, µ-FTIR, nano-FTIR (SINS), s-SNOM, AFM, SEM, SEM-EDX, XRD and SR-XRD were also used. The results pointed out to the necessity of individualization of the artistic objects when investigating their reactivity or when proposing conservation strategies, showed the environmental aggressiveness of all the investigated institutions, indicated the potential of ENs in preventive conservation and demonstrated the utility of techniques of high lateral resolution such as nano-FTIR for the investigation of metal corrosion. More specifically, the investigations of synergistic effects between gaseous pollutants increased the understanding of Pb reactivity and allowed to propose more completed schemes of its reactivity, which allowed discussions on the conservation of these objects. The aggressiveness related to the environmental chemical composition of the churches and museums was inferred through the characterization of metal coupons exposed in their indoor and outdoor environments, whilst the aggressiveness related to physical factors such as temperature and relative humidity was inferred through the use of miniaturized monitors exposed only in the churches. With this monitoring, conservation strategies were proposed. For the well characterization of the metal coupons, exposed in the churches and museums or in simulated microenvironments, the syntheses and characterization of different Pb formates were essential. Finally, aiming at still improving the performance of the miniaturized monitors and increasing their applicability (in their future use as EN), a new device was constructed, and an internship was considered in the University of Aveiro, Portugal, for the development of an EN based on QCMs.

Efeitos da inalação crônica de cocaína \"crack\" no aparelho respiratório de camundongos / Chronic inhalation effects of crack cocaine in the respiratory system of mice

INTRODUÇÃO: Uma das formas de uso (ilegal) da cocaína é a forma fumada, através da queima de uma base de cocaína na forma de \"pedras\" (cocaína \"crack\"). O uso da cocaína \"crack\" pode causar alterações de caráter crônico ou agudo no trato respiratório. Neste estudo foram investigados os efeitos da inalação crônica de cocaína \"crack\" no aparelho respiratório de camundongos. MÉTODOS: Quarenta camundongos Balb/c machos com 60 dias de idade (10 animais controles e 30 expostos) foram usados no estudo. Dez animais foram usados para determinar o nível de cocaína sérica após uma única exposição. Vinte animais foram expostos à fumaça de \"crack\" em uma câmara de inalação, 5 dias por semana, durante 2 meses. Os dez animais controles foram mantidos no biotério. Análises morfométricas quantitativas de cortes histológicos de nariz e pulmão foram realizadas através da microscopia óptica. Foram quantificados a espessura e quantidade de muco no epitélio nasal e brônquico, a hemossiderina pulmonar, a espessura da parede e o índice de vasoconstrição de artérias pulmonares de pequeno calibre e a densidade de macrófagos e neutrófilos nas regiões alveolares e peribrônquicas. O grupo exposto e controle foram comparados entre si e as diferenças foram consideradas significativas quando p<0,05. RESULTADOS: No epitélio nasal observou-se um aumento de muco (p<0,001) e diminuição de sua espessura (p<0,001) no grupo exposto. No epitélio brônquico não houve alteração da quantidade de muco (p=0,075) entre os grupos, mas observou-se significante diminuição da espessura epitelial (p<0,001) no grupo exposto. Houve aumento da hemossiderina pulmonar (p<0,001) no grupo exposto. Nas artérias pulmonares de pequeno calibre não houve aumento da espessura da parede (p=0,782) entre os grupos, mas o índice de vasoconstrição foi significativamente maior (p=0,034) no grupo exposto. A densidade celular dos macrófagos foi maior tanto na região alveolar (p=0,001) como na região peribrônquica (p=0,001). A densidade celular de neutrófilos foi maior na região alveolar (p=0,002) do grupo exposto sendo que na região peribrônquica a diferença não foi significante (p=0,419) entre os grupos. CONCLUSÕES: Nossos resultados mostram que a inalação do \"crack\" causa alterações no aparelho respiratório, afetando desde a mucosa nasal até o parênquima pulmonar. Este estudo contribui para uma melhor compreensão dos efeitos adversos do uso crônico da cocaína \"crack\" no trato respiratório. / INTRODUCTION: One of the illegal ways of using cocaine is smoking its rock through a burning cocaine basis shaped resin (crack cocaine). The use of crack cocaine may cause chronic and severe alterations in the respiratory system. In this research, the chronic inhalation effects of \"crack\" cocaine in mice respiratory systems were studied. METHODS: Forty male Balb/c mice, 60-days-old (10 control animals and 30 exposed) were utilized in this study. Ten of them were chosen to determine the level of silky cocaine after one single exposition. Twenty animals were exposed to crack smoke in an inhalation chamber, 5 days a week, during 2 months. All the control animals were kept in the housing unit. Quantitative morphometric analysis of histological cuts in the nose and lung were realized across by the optical microscopy. The following were quantified: the thickness, likewise the amount of mucus in nasal and bronchial epitheliums, lung hemosiderin, ply of the sheet and the rate of vasoconstriction in lung arteries of small diameter, in addition to the density of macrophages and neutrophiles inside of alveolar and peribronchial regions. A comparison between the high-risk and control group was established. The differences were considered significant when p<0,05. RESULTS: In the nasal epithelium, an increase of mucus was observed (p<0,001), as well as a decrease of its thickness (p<0,001) in the high-risk group. In the bronchial epithelium, there was no alteration in the amount of mucus (p=0,075) between the groups, but there was a significant decrease in the epithelial ply (p<0,001) inside of the exposed group. There was an increase of lung hemosiderin (p<0,001) in the exposed group. In arteries of small diameter, there was no expansion of the thickness in the sheet (p=0,782) between the groups, although the level of vasoconstriction was significantly higher (p=0,034) in the exposed group. The cellular density of macrophages was higher in the alveolar region (p=0,001), as in the peribronchial one (p=0,001). The cellular density observed in neutrophiles was higher in the alveolar site (p=0,002) of the exposed group, since that in the peribronchial region the difference was not significant (p=0,419) between the groups. CONCLUSIONS: Our results show that crack inhalation may cause alterations in the respiratory system, affecting from the nasal mucus until the lung parenchyma. This research contributes for a better comprehension of the adverse effects originating from a chronic use of crack cocaine in the respiratory tract.

O desenvolvimento de sensores em sistemas para controle ambiental tem-se mostrado uma área de elevado interesse científico e técnico. Os principais desafios nesta área estão relacionados ao desenvolvimento de sensores com capacidade de detecção de várias substâncias. Neste contexto, os capacitores MOS apresentam-se como dispositivos versáteis para a geração de imagens químicas com potencial de detecção e classificação de diferentes substâncias a partir de apenas um único sensor. No presente trabalho, foi proposto um sensor MOS com um perfil geométrico de porta em forma de cata-vento composta por Pd, Au e Pt. A resposta do sensor mostrou ter alta sensibilidade a moléculas ricas em átomos de H, como os gases H2 e NH3. As medidas de capacitância mostraram que o sensor tem uma resposta não linear para H2 e NH3 obedecendo à lei da isoterma de Langmuir. O sensor MOS mostrou-se eficiente na geração de imagens químicas através da técnica de escaneamento por luz pulsada. As imagens químicas correspondentes aos gases H2 e NH3 mostraram diferentes padrões quando o N2 foi utilizado como gás transportador. A diferença entre os padrões aconteceu principalmente devido ao perfil geométrico da porta metálica. A sensibilidade do sensor mostrou dependência com o potencial de polarização. Nas medidas de capacitância, a maior sensibilidade foi observada para potenciais próximos da tensão de banda plana. Já para as imagens químicas, a maior sensibilidade foi observada para potenciais inteiramente na região de depleção. A sensibilidade do sensor também se mostrou dependente do gás transporta- dor. O sensor mostrou ser mais sensível com N2 como gás transportador do que com ar seco. No entanto, o processo de dessorção dos íons H+ resultou ser mais eficiente em ar seco. Os resultados obtidos no presente trabalho sugerem a possibilidade de fabricação de um nariz optoeletrônico utilizando apenas um único sensor MOS. / The development of sensors and systems for environmental control has been shown to be an area of high scientific and technical interest. The main challenges in this area are related to the development of sensors capable of detecting many different substances. In this context, the MOS devices present themselves as versatile devices for chemical imaging with potential for detection and classification of different substances only using one single sensor. In the present work, was proposed a MOS sensor with a wing-vane geometric profile of its gate constituted of Pd, Au and Pt metals. The sensor\'s response showed to have high sensitivity to molecules rich on H atoms, such as H2 and NH3 gases. Capacitance measurements showed that the sensor has a nonlinear response for H2 and NH3 obeying the Langmuir isotherm law. The MOS sensor proved to be efficient in Chemical Imaging generation through the scanned light pulse technique. The chemical images of the H2 and NH3 gases showed different patterns when the N2 was used as carrier gas. The different patterns responses happened mainly due to geometric profile of the metallic gate. The sensor sensitivity showed dependence on the bias potential. In the capacitance measures, greater sensitivity was observed for potential near the flat-band voltage. In the chemical images, the greater sensitivity was observed for bias potential within depletion region. The sensor sensitivity was also dependent on the carrier gas. The sensor showed to be more sensitive with N2 as carrier gas than to dry air. However the desorption process of H+ have been more efficient in dry air. The results obtained in the present work suggest the possibility of manufacturing an optoelectronic nose using only a single MOS sensor.

In this context, the MOS devices present themselves as versatile devices for chemical imaging with potential for detection and classification of different substances only using one single sensor. In the present work, was proposed a MOS sensor with a wing-vane geometric profile of its gate constituted of Pd, Au and Pt metals. The sensor\'s response showed to have high sensitivity to molecules rich on H atoms, such as H2 and NH3 gases. Capacitance measurements showed that the sensor has a nonlinear response for H2 and NH3 obeying the Langmuir isotherm law. The MOS sensor proved to be efficient in Chemical Imaging generation through the scanned light pulse technique. The chemical images of the H2 and NH3 gases showed different patterns when the N2 was used as carrier gas. The different patterns responses happened mainly due to geometric profile of the metallic gate. The sensor sensitivity showed dependence on the bias potential. In the capacitance measures, greater sensitivity was observed for potential near the flat-band voltage. In the chemical images, the greater sensitivity was observed for bias potential within depletion region. The sensor sensitivity was also dependent on the carrier gas. The sensor showed to be more sensitive with N2 as carrier gas than to dry air. However the desorption process of H+ have been more efficient in dry air. The results obtained in the present work suggest the possibility of manufacturing an optoelectronic nose using only a single MOS sensor.

Efeito subcrônico do diesel no epitélio nasal e na via aérea em modelo murino / Subchronic effects of diesel on nasal and airway epithelium in a murine model

A combustão do diesel (DEP) é a principal fonte de partículas ultrafinas (PUFs) relacionadas à poluição causada pelo tráfego. Indivíduos com doenças respiratórias crônicas estão propensos a exacerbações durante a exposição à poluição ambiente. Este estudo avaliou os efeitos de exposição subcrônica a uma baixa dose de partículas de combustão de diesel (DEP). 90 camundongos machos BALB/c foram divididos em 3 grupos: (a) Controle: instilação nasal de solução salina (n = 30); (b) DEP15: 15?g de DEP/10?l de solução salina (n = 30); e (c) DEP30: 30?g de DEP/10?l de solução salina (n = 30) durante cinco dias por semana, por 30 e 60 dias. Os animais foram anestesiados com pentobarbital de sódio (50mg/kg ip) e sacrificados por exanguinação. A contagem de células inflamatórias e as concentrações de interleucinas (IL) -4, -10, -13 e -17 no lavado broncoalveolar (LBA) foram avaliadas por ensaio imunoenzimático (Elisa). mRNA da MUC5ac foi avaliado por PCR em tempo real. A análise histológica do septo nasal e bronquíolos foi realizada para avaliar: (a) a espessura do epitélio brônquico e nasal, (b) o conteúdo de muco neutro e ácido na mucosa nasal. Nossos resultados mostraram que a instilação de DEP30 após 30 dias aumentou o número de células inflamatórias totais em relação ao controle (p=0,033). Ao comparar os resultados de DEP30 com o grupo Controle após 60 dias observamos os seguintes aumentos: (a) na expressão de MUC5AC nos pulmões (p = 0,016), no conteúdo de muco ácido no septo nasal (p = 0,017), nas células inflamatórias totais no LBA (p<0,001), no número de macrófagos no LBA (p=0,035) e na espessura do epitélio nasal (p=0,042). Nossos dados sugerem que dose baixa de DEP induz inflamação do trato respiratório com padrão tempo-dependente. / Diesel exhaust is the major source of ultrafine particles (UFPs) in trafficrelated pollution. Subjects with chronic respiratory diseases have great risk of exacerbations during exposure to air pollution. This study evaluated the effects of sub-chronic exposure to a low dose of diesel exhaust particles (DEPs). Ninety male BALB/c mice were divided into 3 groups: (a) Control: nasal saline instillation (n=30); (b) DEP15: nasal instillation of 15?g of DEP/10?l of saline (n=30); and (c) DEP30: nasal instillation of 30?g of DEP/10?l of saline (n=30). Nasal instillations were performed five-days a week, during 30 e 60 days. Animals were anesthetized with pentobarbital sodium (50mg/kg i.p), and sacrificed by exsanguination. Bronchoalveolar lavage (BAL) was performed to assess inflammatory cell count and concetrations of interleukin (IL)-4, -10, -13 and -17 by enzyme-linked immunosorbent assay (ELISA). The RNAm MUC5ac gene expression was evaluated by real-time PCR. Histological analysis in nasal septum and bronchioles assessed: (a) bronchial and nasal epithelium thickness (b) acidic and neutral nasal mucous content. Our results showed that the instillation of DEP30 after 30 days increased the number of total inflammatory cells, as compared to the Control group (p = 0.033). The results of DEP30 after 60 days showed increases in: (a) the expression of MUC5AC in the lungs (p = 0.016); (b) acidic mucus production in the nasal septum (p = 0.017); (c) total inflammatory cells in the BAL fluid (p <0.001); (d) the number of macrophages in BALF (p = 0.035); and (d) nasal epithelium thickness (p = 0.042), as compared with control after 60 days. Our data suggest that a low dose of DEP induces inflammation of the respiratory tract in a time- dependent manner.

Impacto da composição química de microambientes em bens culturais: reatividade e monitoramento / Impact of microenvironmental chemical composition on heritage objects: reactivity and monitoring

Esta Tese de doutorado teve por objetivo aumentar a compreensão dos aspectos sinérgicos na corrosão de bens culturais feitos com chumbo (Pb) e avaliar a agressividade ambiental sobre bens culturais em igrejas e museus brasileiros para a proposição de estratégias de conservação. Neste contexto, foram investigados, em microambientes simulados, os efeitos sinérgicos entre formaldeído e ácido acético, ácido fórmico ou dióxido de carbono na corrosão de Pb; foi feita a síntese e caracterização de diferentes formiatos gerados nos processos de corrosão de Pb por formaldeído ou ácido fórmico; foi avaliada a agressividade ambiental sobre bens culturais da Igreja de São Francisco (São Paulo, ambiente urbano industrial), da Igreja dos Freis Carmelitas (Santos, ambiente urbano industrial costeiro), do Museu de Arte Contemporânea da USP (MAC, São Paulo, reserva técnica da nova sede) e do Museu de Arqueologia e Etnologia da USP (MAE, São Paulo, reserva técnica), através de cupons metálicos (chumbo e cobre) expostos nos ambientes interno e externo dessas instituições e através do uso de monitores miniaturizados; foi feita a otimização destes monitores e, finalmente, foi considerado o desenvolvimento de um nariz eletrônico (EN) baseado em microbalanças de quartzo (QCMs) para o uso em conservação preventiva. Nestas investigações microscopia Raman foi a principal técnica empregada, sendo que as demais foram FTIR-ATR, &#181;-FTIR-ATR, &#181;-FTIR, nano-FTIR (SINS), s-SNOM, AFM, SEM, SEM-EDX, XRD e SR-XRD. Os resultados apontaram para a necessidade de individualização dos objetos artísticos tanto para a proposição de estratégias de conservação quanto para o entendimento de sua reatividade, mostraram a agressividade ambiental de todas as instituições investigadas, indicaram a potencialidade de ENs em conservação preventiva e demonstraram a utilidade de técnicas de alta resolução lateral, como nano-FTIR, em investigações de corrosão metálica. Mais especificamente, as investigações de sinergia entre poluentes aumentaram o entendimento da reatividade de Pb e permitiram a proposição de esquemas mais completos de sua reatividade, levando a discussões acerca da conservação desses bens. A agressividade química ambiental das igrejas e museus a bens culturais foi inferida pela caracterização de cupons metálicos expostos em seus interiores e exteriores, enquanto que a agressividade relacionada a fatores físicos, como temperatura e umidade relativa, foi inferida através do uso de monitores miniaturizados expostos somente nas igrejas. Com esse monitoramento, estratégias de conservação foram propostas. Para a devida caracterização dos cupons metálicos, expostos nas igrejas e museus ou nos microambientes simulados, a síntese e a caracterização de diferentes formiatos de Pb foi essencial. Por fim, visando melhorar o desempenho dos monitores miniaturizados e ampliar sua aplicabilidade (com seu futuro uso como EN), um novo dispositivo foi construído e um estágio na Universidade de Aveiro, em Portugal, foi realizado para o desenvolvimento de um EN baseado em QCMs. / The objective of this doctoral thesis was to increase the understanding of synergistic aspects in the corrosion of cultural objects made with lead (Pb) and to assess the environmental aggressiveness on cultural objects of Brazilian churches and museums in order to propose conservation strategies. In this context, in simulated micro-environments, the synergistic effects between formaldehyde and acetic acid, formic acid or carbon dioxide in Pb corrosion were investigated; the syntheses and characterization of different formates generated by formaldehyde or formic acid in Pb corrosion processes was conducted; the environmental aggressiveness on cultural objects of the church of São Francisco (São Paulo, urban industrial environment), of the church of the Carmelite Friars (Santos, urban industrial littoral environment), of the Museum of Contemporary Art of the USP (MAC, São Paulo, new site) and of the Museum of Archaeology and Ethnology of the USP (MAE, São Paulo) was assessed exposing metal coupons (lead and copper) in their internal and external environments and using miniaturized monitors; these monitors were optimized and finally, the development of an \'electronic nose\' (EN) based on quartz crystal microbalances (QCMs) was considered for preventive conservation. In these investigations the principal technique employed was Raman microscopy, and FTIR-ATR, &#181;-FTIR-ATR, &#181;-FTIR, nano-FTIR (SINS), s-SNOM, AFM, SEM, SEM-EDX, XRD and SR-XRD were also used. The results pointed out to the necessity of individualization of the artistic objects when investigating their reactivity or when proposing conservation strategies, showed the environmental aggressiveness of all the investigated institutions, indicated the potential of ENs in preventive conservation and demonstrated the utility of techniques of high lateral resolution such as nano-FTIR for the investigation of metal corrosion. More specifically, the investigations of synergistic effects between gaseous pollutants increased the understanding of Pb reactivity and allowed to propose more completed schemes of its reactivity, which allowed discussions on the conservation of these objects. The aggressiveness related to the environmental chemical composition of the churches and museums was inferred through the characterization of metal coupons exposed in their indoor and outdoor environments, whilst the aggressiveness related to physical factors such as temperature and relative humidity was inferred through the use of miniaturized monitors exposed only in the churches. With this monitoring, conservation strategies were proposed. For the well characterization of the metal coupons, exposed in the churches and museums or in simulated microenvironments, the syntheses and characterization of different Pb formates were essential. Finally, aiming at still improving the performance of the miniaturized monitors and increasing their applicability (in their future use as EN), a new device was constructed, and an internship was considered in the University of Aveiro, Portugal, for the development of an EN based on QCMs.
