Characterisation of Xanthomonas campestris pv. campestris isolates from South Africa using genomic DNA fingerprinting and pathogenicity tests / by Lizyben Chidamba

Black rot caused by Xanthomonas campestris pv. campestris (X. c pv. campestris) is a major disease constraint to cabbage production. The control of black rot is difficult and resistant cultivars could play an important role in reducing the losses due to the disease. Information on the distribution and diversity of X. c pv. campestris is critical before any meaningful disease resistance screening can be done. However, little is known about the diversity and international significance of South African X. c pv. campestris strains. To assess the genetic diversity and international significance of X. c pv. campestris strains in South Africa, strains of the pathogen were obtained from cabbage growing districts in Gauteng, Mpumalanga and North West Provinces of South Africa in 2010. International strains were obtained from international culture collections. Isolates from South Africa were purified and race typed using differential sets of Brassica spp according to Nickerson–Zwaan protocols. Four races, race 1(14%), race 3 (7%), race 4 (68%) and race 6 (10%) of the pathogen were identified. Repetitive DNA polymerase chain reaction–based fingerprinting using Eric– and Box–primers were used to assess the genetic diversity. Polyacrylamide gel electrophoresis allowed clear and reproducible differentiation of the PCR products. Of the amplified loci for South African isolates 5 loci were present in at least 90 % of the isolates for Eric–profiles and 6 in at least 80% of the isolates for Box–profiles. Of these prominent loci, none had corresponding high presence in international isolates. While no loci had a presence greater than 51% and 61% for Eric– and Box– profiles in international isolates, respectively, several loci among South African isolates were unique to isolates from specific geographic origin. Generated fingerprints of X. c pv. campestris were similar for the South African isolates and distinguishable from those of X. c pv. armoraciae and X. c pv. raphani reference strains. However, when international X. c pv. campestris were considered, no profile pattern was observed to be unique to international X. c pv. campestris isolates as was the case with South African isolates. Eric– and Box–PCR profiles of international isolates varied widely with some isolates having profile patterns similar to those of reference strains. Cluster analysis divided X. c pv. campestris into two major groups, the South African group and the international isolates group. The South African group could be divided into subgroups, which clustered according to the geographical origin of the isolates. The same was observed for international isolates, which generally clustered isolates according to country of origin. However, isolates from different countries also clustered together. A few X. c pv. campestris strains of international origin clustered with the South African isolates group. Furthermore, a few South African isolates were clustered in the international isolate group. Although X. c pv. campestris distribution may be unique to its geographical origin, our findings, based on the present data set, suggest wide spread of the pathogen both at national and international level. The existence of different races, genetic variability and international distribution of the pathogen should be considered when resistant crucifer cultivars are bred to control black rot of crucifers / Thesis (M.Sc. As amostras de água foram filtradas em filtro Millipore® e a membrana colocada sobre o centro de uma placa de Petri contendo agar cetrimida, As colônias típicas de bactérias do gênero Pseudomonas foram identificadas pelo método de Gram, inoculação em TSI agar (Triple Sugar Iron), crescimento a 42ºC, produção de pigmento, produção de alginato, oxidase, motilidade e alcalinização da acetamida. O teste utilizado para análise genômica foi o ERIC-PCR. Dos 76 (42,2%) isolados de Pseudomonas aeruginosa, 15 eram provenientes das amostras de torneira de lavagem das mãos, 18 de reservatório de garrafa pet, 23 de seringa tríplice e 20 do motor de alta rotação. Todos os isolados de Pseudomonas aeruginosa foram submetidos ao teste de suscetibilidade segundo a técnica de Kirby- Bauer. Dos antibióticos testados o que apresentou melhor resultado quanto à sensibilidade (65,8%) foi a ciprofloxacina. Quanto à similaridade genética dos isolados dos diferentes pontos analisados, foram encontrados nove “clusters” de 100% de similaridade. / The biofilm found in water supplies and lines of hospitals and dental units is extremely important because it presents a large number of bacteria, leading to risk of infection in immunocompromised patients vulnerable to opportunistic pathogens such as the Pseudomonas aeruginosa. One hundred eighty water samples from dental units of the city of Barretos-SP were evaluated. The water samples filtered in Millipore® filter were incubated in plates containing Cetrimide ágar. The bacteria colonies were identified through the gram-staining test, inoculation in agar T.S.I (triple sugar Iron), growth at 42 ° C, pigment production, production of alginate, oxidase acetamide alkalization and motility observation. All Pseudomonas aeruginosa bacteria were submitted to the susceptibility test according to the Kirby-Bauer technique. The test used for genomic analysis was the ERIC-PCR. From the total microorganisms studied, 76 (42,2%) were positive for Pseudomonas aeruginosa, isolates from 180 water samples, where 15 strains were from hand washing incoming local water supplies, 18 from PET bottled water, 23 from 3-in-1 syringes and 20 from the high speed handpiece. In relation to the antibiotics tested, the one presenting the best result with regard to sensibility was ciprofloxacin with 65.8%. The genetic similarity of isolates from different points analyzed, there were nine clusters of 100% similarity.

The aim of my thesis is a complete analysis of child characters in Eric-Emmanuel Schmitt´s tetralogy Le cycle de l´invisible. The child characters are the main protagonists of all the cycle. Therefore they the have been given the main focus in my work. In the theoretical part of my work I'm introducing the basis of the analysis of the characters in general. A treatise of the character with it´s different aspects and functions in a literary text is a part of the theoretical basis. In the subsequent analysis of individual texts I have been analyzing child characters appearing in the cycle. I have been dealing with their characteristics, their relation to the storyline and to the narrative structure of the text, to the other characters, especially adults. During my work I have been trying to reach a more general characteristic of Schmitt´s child fables and also to answer the question why the author chose children as his prose protagonists.

The purpose of this work is try to understand what the terms violence and politic action means in the philosophy of Weil, and at the same time, the meaning of philosophy itself. We start using the relationship between philosophy and violence, related to the negative act of the reason and, that´s why, the main problem for the philosophical speech in its pretension of absolute understanding. One of the Veil´s characteristics is consider what represent the problem of the violence after Hegel´s perspective of an absolutely coherent speech, so that, considering the lack of the absolute speech in order to contain the reality of the violence. As a result, we have the reflection about the relationship between violence and reasonable action in the politic area, in order to think the politics through the philosophy. Weil takes the politics as consideration of the human being living in common according to his essential structures getting back a positive relationship between moral and politics, in order to raise a philosophy that is capable of understand the modern politics and consider the politic action as a fighting against the violence, referring to nature, the relationship between individual and society and the solution in the modern state, as the owner of the monopoly of the use of violence. The state appears as a condition for the conciliation between the sensible life of the individual in the concrete moral and the imposing demand through the rationality of the modern society. Finally, a reasonable life, with the possibilities of the modern society, must find a meaning in the politic life, participating of the decisions of common interest. It´s the transposition of the problems solution of the using of violence for the debating area, through the education of the politic people, social task of the philosopher, and it´s the aim of the politic action. This reflection intends to be coherent, according to its time, must recognize that history, politics, economy, and philosophy, force the philosopher to think about the common destiny of the human kind. That´s what Weil reflects about a world state, concrete category that gather the moral categories, of society and the state, and where all these politic categories are together in the purpose of understanding the reality / A filosofia, para Eric Weil, não é apenas uma visão sobre o mundo, ou um saber dogmático ou relativista, mas um comprometimento com a razão e com a liberdade enquanto tentativa de compreensão do homem, do seu discurso e da sua ação. Deste modo, o objetivo deste trabalho é compreender o que os termos violência e ação política significam na filosofia de Weil, e, ao mesmo tempo, o significado da própria filosofia. Parte-se da relação entre filosofia e violência, enquanto ato negador da razão e, por isso, problema fundamental para o discurso filosófico na sua pretensão de compreensão absoluta. O específico de Weil é considerar o que o problema da violência representa depois da perspectiva hegeliana de um discurso absolutamente coerente, isto é, a consideração da insuficiência do discurso Absoluto para dar conta da realidade da violência. Decorre daí a reflexão acerca da relação entre violência e ação razoável no campo político, propondo-se a pensar a política a partir da filosofia. Weil parte da política como consideração da vida em comum dos homens segundo as suas estruturas essenciais, recuperando uma relação positiva entre moral e política, para erguer, a partir destas indicações, uma filosofia capaz de dar conta da política moderna e lança mão da consideração da ação política como luta contra a violência, seja como violência da natureza exterior, seja na relação violenta entre indivíduo e sociedade, seja na solução apontada no Estado moderno como detentor do monopólio do uso da violência. O Estado aparece como condição para a conciliação entre a vida sensata do indivíduo na moral concreta da sua comunidade e as exigências impostas pelo tipo de racionalidade da sociedade moderna. Por fim, uma vida razoável e dona das possibilidades oferecidas pela sociedade, deve encontrar tradução na vida política pela possibilidade de, vencidas as violências da natureza, do indivíduo e da sociedade, participar das decisões que tocam os interesses coletivos. É a transposição da resolução dos problemas do uso da violência ao plano da discussão através da educação do povo político, tarefa social do filósofo e finalidade por excelência da ação política. Tal reflexão, para ser coerente com seu tempo, deve reconhecer que história, política, economia e filosofia obrigam o filósofo a refletir sobre o destino comum da humanidade, o que Weil faz ao pensar um Estado mundial, categoria concreta na qual reúne as categorias da moral, da sociedade e do Estado, e onde todas estas categorias políticas se articulam em vista da compreensão da realidade.

Specialty coffee expansion in traditional retail: lessons from non-traditional retailers

Amanor-Boadu / Despite at least three waves of transformations in the US coffee retail market, traditional retailers have not altered their merchandizing approaches for decades. This may be due primarily to the fact that there are still margins being made in selling canned coffee, the initial coffee wave in this research. Yet, because of their significant role in the retail segment, traditional retailers cannot be ignored by coffee suppliers. This implies that with each shift in the coffee industry, it is important for the participants to find ways of enabling the traditional retailer to make the necessary transformation – at least with their products – so that they can secure their market share and their continuing success. The research develops a number of case studies of different coffee retailers who are developing innovative processes for merchandizing new coffee formats, such as Keurig K-Cups and Ready to Drink (RTD) products. The research shows that coffee merchandizers can learn from these retailers to develop support programs for their traditional retail customers to leverage their importance in the coffee market to enhance their own sales and profitability. We identify a number of value innovation strategies that may be used to achieve this objective of enhancing performance in traditional coffee merchandizing. For example, we identify a store-within-a store strategy that is already in operation for a number of product categories in traditional retail, and suggest that it be expanded to include coffee. This approach will elevate purchases across the segment and help enhance overall competitiveness. The approach is not unlike Kroger’s treatment of its natural and organics as a separate department or Roche Brothers’ creation of a gourmet specialty, which is prominently displayed near the store’s entry. It has become a prime location where new and exciting entrants to the specialty assortment are presented to shoppers in an elevated way. In this location, they are typically expected to sell-through initial quantities rapidly. The research presents innovative ideas to help coffee purveyors help their customers reposition emerging “waves” of coffee products in their traditional retail systems. It hopes that traditional retailers will benefit from the case studies of lessons from other categories and initiatives so that they can improve their own performance, and in so doing help coffee purveyors enhance their own performance.

O centro da pesquisa são os Contos morais, um ciclo de seis filmes composto por A Padeira do bairro (1963), A Carreira de Suzanne (1963), A Colecionadora (1967), Minha noite com ela (1969), O Joelho de Claire (1970) e Amor à tarde (1972). Esta é a primeira fase de destaque do cineasta e o período de consolidação do seu estilo. É apresentado um panorama histórico da passagem da profissão como crítico na revista Cahiers du Cinéma à realização dos Contos morais. A partir da análise fílmica, atendo-se à narrativa, à encenação e aos modos de produção, são identificadas as linhas de força da poética do cineasta. Constata-se que seu particular modo de produção reflete esteticamente em seus filmes. O diretor emprega a construção de uma rigorosa estrutura narrativa ficcional, isso combinado com uma abordagem realista do espaço, da trama e das atuações; finalmente, destaca a valorização da verborragia de seus personagens / Abstract: This paper has as object of research movies of the French director Éric Rohmer, who settled with the French new wave, in the context of the 1950s and 1960s. The center of the research are the Moral tales, a cycle of six movies composed of The Bakery girl of Monceau (1963), Suzanne's Career (1963), The Collector (1967), My Night at Maud¿s (1969), Claire's Knee (1970) and Love in the afternoon (1972). This is the first prominent phase of the filmmaker and the period of consolidation of his style. A historical overview of the transition of his profession as film critic in Cahiers du Cinéma to the directing of the Moral tales is presented. From film analysis, focusing the narrative, the mise en scène and the production methods, the power lines of the poetic of the filmmaker are identified. It is verified that his particular way of producing reflects aesthetically in his films. Rohmer focuses on moral issues and sentimental relations of the characters, showing a strongly objectified and transparent film. The director employs a strict construction of fictional narrative structure, combined with a realistic approach of the space, plot and performances; finally, he highlights the appreciation of verbiage of his characters / Mestrado / Multimeios / Mestre em Multimeios

”Jag har erfarit Influxus ...” : en komparativ analys av Eric Hermelins litterära förhållningssätt till Emanuel Swedenborg / Influxus by experience – a comparative study of Eric Hermelin and his approach to the teachings of Emanuel Swedenborg

Eric Hermelin, the translator and mystic who, while being incarcerated at a mental institution outside Lund in Sweden, managed to write himself into the history of literature by translating the Great Persian sufi poets of the Middle Ages, Rumi, Attar and Sadi, to name a few. Amongst his other translations we find the native Emanuel Swedenborg who will be the focus of this study.The purpose for this study is to look upon how the teachings of Swedenborg affected Hermelin through a comparative analysis where the translations of Hermelin will play a key role. By comparing them to the original texts of both Swedenborg and the Persian poets and by putting Hermelin in context with his times we hope to find out in what way Hermelin related to Swedenborg.Analysis is centered around three concepts; the Personality, the Correspondence and Influxus, which mirrors the Hermelin focus regarding Swedenborg. The study shows that Swedenborg is central to Hermelin in helping him finding strength for his massive work. Hermelin knows Influxus by experience and he seems well aware of the mystic and esoteric context in which Swedenborg belongs where the Persian poets also plays an important part. In a way Hermelin relate Swedenborg to the Persian poets using the teachings of Swedenborg as an instrument to point out the similarities of content.
