O comportamento natatório de larvas de peixes na fase da primeira alimentação de Rachycentron canadum e Epinephelus marginatus foi descrito usando imagens de câmeras de alta frequência (60 a 2000 quadros por segundo) pelas técnicas de imageamento: microscopia de campo claro, sistema de filtros pareados e holografia. Oito métricas comportamentais foram registradas na presença de náuplios de copépodes; rotíferos Brachionus sp. e; sem alimento: natação rotineira (1 a 2,9 vezes o comprimento do corpo por segundo, CP s-¹); natação explosiva (3 a 40 CP s-¹); recuo; distâncias percorridas; batimentos do complexo caudal na natação e manutenção da posição (40 b s-¹); contrações rápidas do corpo em formato de C (de 0,16 a 0,40 s) e S (0,009 a 0,17 s); repouso; e comportamento alimentar (visualização das presas e tentativa de captura). As larvas alteraram a velocidade de natação na presença de presas e aumentaram em função da idade. Os maiores valores alcançados do número de Reynolds foram na presença de presas, para natação rotineira foi 200. As contrações da musculatura e mudanças na velocidade de natação indicam relação com aspectos ecológicos dos organismos, como busca por alimento e interações intra e interespecíficas. / The swimming behavior of first feeding fish larvae Rachycentron canadum and Epinephelus marginatus was described using images acquired with high-speed cameras (60 to 2000 frames per second) by the imaging techniques: bright field microscopy, matched filters system and holography. A inversão do sexo em peixes hermafroditas sequenciais (protândricos ou protogínicos) ocorre em decorrência de diversos fatores, dentre eles, fisiológicos, genéticos, podendo ser ainda decorrência do comportamento social. Nos peixes hermafroditas marinhos as alterações sexuais são acompanhadas por alterações anátomo-funcionais das gônadas, coloração e comportamento dos animais. A espécie Epinephelus marginatus (garoupa verdadeira), serranídeo da fauna nativa e amplamente difundida no litoral brasileiro, é um hermafrodita protogínico que apresenta dificuldade na manutenção de indivíduos do sexo masculino em seus cardumes e, além disso, encontra-se na lista de espécies sobreexplotadas. Utilizando-se técnicas de inversão sexual induzida, em cativeiro, para a obtenção de machos, este trabalho teve como objetivo estudar as alterações de alguns tecidos endócrinos durante o processo de inversão sexual, utilizando tratamentos com inibidores de aromatase (AI) e metiltestosterona (MT) em animais jovens. Ao longo de 90 dias de experimento a utilização do AI e do MT promoveu uma aparente desorganização da arquitetura gonadal padrão, massiva degeneração das células germinativas femininas, surgimento de centros melanomacrofágicos, proliferação de estruturas associadas ao sexo masculino (tecido intersticial e células germinativas masculinas) e a maturação das células germinativas masculinas. A utilização do andrógeno sintético MT (sozinho ou combinado com AI) possibilitou a obtenção de gônadas em fases mais avançadas de espermatogênese, inclusive a fase de espermiogênese inicial, com redução significativa dos valores do índice gonadossomático (IGS). Já com a utilização do AI foram encontradas gônadas em processos de intersexo mais clássicos após 90 dias experimentais. As análises de imunomarcação das células gonadotrópicas hipofisárias foram viáveis utilizando-se anticorpos de salmão e os resultados mostraram que as células produtoras de FSH (hormônio folículo-estimulante) e LH (hormônio luteinizante) distribuem-se pela proximal pars distalis e pars intermedia da adenohipófise. Os resultados obtidos com as análises imunohistoquímicas da hipófise demonstraram uma marcação de pouca intensidade utilizando-se o anticorpo anti-&#946;FSH no grupo tratado com AI após 90 dias. A análise dos esteróides gonadais mostrou que nos animais tratados com AI, não houve diminuição na concentração do estradiol aos 90 dias, desta forma a alça de feedback negativo provavelmente foi acionada, e a produção de FSH diminuiu, causando o aumento da concentração plasmática de 11 cetotestosterona (11 KT) (via ação do LH). Já nos grupos onde o MT foi administrado, houve uma diminuição na concentração de estradiol após o tratamento, causando possivelmente uma menor intensidade na alça de feedback negativo na hipófise, sendo a ação do andrógeno sintético mais efetiva na espermiogênese nestes animais. As análises das gônadas e da hipófise sugerem a atuação do FSH no início da transição do sexo, sendo proposta a liberação desta glicoproteína como o gatilho para a inversão sexual em hermafroditas protogínicos. Estudos mais detalhados ao longo dos 90 dias de transição gonadal em E. marginatus precisam ser realizados para constatar o real envolvimento do FSH na inversão do sexo e verificar se a liberação do LH estaria envolvida com a produção de 11-ceto-testosterona, a espermiogênese completa e espermiação / The sex inversion in sequential hermaphrodite fish (protandrous or protogynous) occurs due to several factors, including physiological, genetics, and also can be associated with the social behavior. In marine hermaphrodite fish, the sex alterations are followed by anatomic and functional alterations in the gonads, color patterns and animal behavior. The species Epinephelus marginatus (dusky grouper), a native serranid and broadly distributed in the Brazilian coast, is a protogynous with problems in maintaining males in the shoal and, additionally, this species is part of the overexploited marine fish red list. Induction of sex change, in captivity, using aromatase inhibitors (AI) and 17&#945;-methyltestosterone (MT) has been conducted with the purpose to study the morphophysiological alteration in some endocrine tissues in juveniles. During 90 experimental days, the use of AI and MT promoted an apparent disorganization of the standard gonad architecture, a massive degeneration of the female germ cells, the appearance of melanomacrophagic centers, a proliferation of the structures associated with the male sex (interstitial tissue and male germ cells) and the maturation of male germ cells. The use of the synthetic androgen, MT (alone or combined with AI) allowed the obtaining of gonads in a more advanced spermatogenesis phase, including an initial spermiogenesis phase, with a significant reduction of gonadossomatic index (GSI). With the use of AI, the gonads were found in a more classic intersex process after 90 days of experiment. The immunostaining analysis of the gonadotropic cells were viable using salmon antibodies and the results showed that FSH and LH cells were distributed in proximal pars distalis and pars intermedia in adenohypophysis. The immunohistochemistry analysis of the pituitary showed a low intensity of staining with the anti-&#946;FSH in the AI group, after 90 experimental days. The analysis of the gonad steroids showed that in the animals from the AI group, there was no decrease in estradiol levels after 90 days, so the negative feedback loop probably was activated, the FSH secretion decreased, allowing the increase of plasma 11- ketotestosterone (11KT) (through LH action). In the MT groups there was a decrease in plasma estradiol, what probably decreased the intensity of the negative feedback loop in the pituitary, and in this case, the action of the synthetic androgen was predominant in spermiogenesis. The analysis of the gonads and pituitary suggest the action of FSH in the beginning of the sex transition, with the proposal that this glycoprotein acts triggering the sex inversion in protogynous hermaphrodites. We suggest that more detailed studies during the 90 days of gonad transition in E. marginatus must be done in order to find the real involvement of FSH in sex inversion and to elucidate the actual role of LH with 11KT production, the full spermiogenesis and spermiation

A plasticidade do desenvolvimento gonadal em peixes, que contrasta com os padrões mais estáveis encontrados nos demais vertebrados, deu origem a várias questões intrigantes relativas tanto ao seu significado adaptativo quanto aos fatores genéticos e fisiológicos que modulam o processo. A inversão do sexo em peixes hermafroditas seqüenciais (protândricos ou protogínicos) ocorre em decorrência de diversos fatores, dentre eles, fisiológicos, genéticos e comportamentais podendo ser ainda decorrente do comportamento social. A garoupa verdadeira, Epinephelus marginatus é um peixe teleósteo, hermafrodita protogínico característico do litoral rochoso, maturando primeiramente como fêmeas, e posteriormente, na vida adulta, os ovários são substituídos por testículos, transformando-os em machos reprodutivamente ativos. No presente trabalho foram realizados três experimentos de inversão sexual em condições assistidas, em diferentes estações do ano, utilizando a mesma dose do inibidor de aromatase (IA) letrozole (100&mu;g/Kg) para promover a inversão sexual de E. marginatus visando contribuir e aprimorar o conhecimento dos mecanismos fisiológicos envolvidos nesse processo. O primeiro e o terceiro experimentos foram iniciados na primavera e início da primavera respectivamente e promoveram a inversão sexual com uma diminuição no índice gonadossomático (IGS) nos animais do grupo experimental que apresentaram uma desorganização gonadal peculiar, com lamelas características de ovário, no entanto em seu interior observou-se o compartimento intersticial desenvolvido com cistos de células germinativas masculinas em estágios avançados da espermatogênese, (espermatócitos, espermátides) células de Sertoli e grande quantidade de cistos rompidos com a liberação de espermatozoides no lúmen. Após trinta dias do início do experimento obteve-se um aumento significativo de 11 ceto-testosterona (11KT) e a concentração de estradiol (E2) permaneceu inalterada; com setenta e sete dias após o início do experimento constatou-se uma elevação na concentração de testosterona (T), uma queda nos níveis de 11KT, persistindo a manutenção na concentração de E2. Neste cenário, principalmente as concentrações de andrógenos podem ter ativado a alça de feedback negativo e juntamente com a ativação do sistema da kisspeptina podem ter promovido a ação do GnRH desviando a produção de FSH para LH. Nesse momento o número de cópias obtidas de &beta;FSH na hipófise foi significativamente menor que o número encontrado no grupo controle. Ao observarmos o aumento significativo da concentração plasmática de 17-alfaOHprogesterona podemos sugerir que a ação do GnRH tenha aumentado a secreção do LH, principal gonadotropina que estimula a produção de progestágenos. Os machos invertidos produziram um volume médio de sêmen de 118,20 ± 24,83 &mu;L com motilidade de 90% e densidade espermática de 8,94 ± 4,34 x 109 células mL-1. Não foram obtidas diferenças nas concentrações de proteínas totais plasmática, hepática e musculares. No experimento implantado no verão de 2010 o tratamento não promoveu a inversão sexual, o IGS não apresentou diferença significativa entre os animais tratados com inibidor de aromatase e os animais do grupo controle. Nas gônadas dos animais do grupo experimental foi observada uma ligeira desorganização da arquitetura gonadal com cistos em formação e um número grande de ovócitos em degeneração demonstrando que ao longo do processo de inversão sexual pode haver uma fase de intersexo. Após trinta dias do início do experimento obteve-se um aumento significativo de T e a concentração de E2 permaneceu inalterada. Não foram observadas diferenças significativas entre o número de cópias de &beta;FSH e &beta;LH ao longo do experimento. O resultado obtido para a concentração de proteínas totais do músculo dos animais do grupo experimental apresentou um aumento significativo em relação à concentração encontrada nos animais do grupo controle. Não ocorreu espermiação e a inversão se deu de forma incompleta promovendo indivíduos intersexo. De uma forma geral podemos concluir que o uso de inibidor de aromatase é um método eficaz para promover a inversão sexual em hermafroditas protogínicos como a garoupa verdadeira. O aumento dos andrógenos pode ser o gatilho da inversão sexual com papel fundamental na reestruturação gonadal e formação do tecido germinativo masculino neste processo / The plasticity of gonadal development in fish, which contrasts with the more stable patterns found in other vertebrates, has given rise to several thrilling questions concerning both, its adaptive significance regarding the genetic and physiological factors that modulate the process. The sex inversion in sequential hermaphrodites fish (protandrous or protogynuos) occurs due to several factors, including, physiological, genetic and behavioral, and may also be due to social behavior. The dusky grouper, Epinephelus marginatus is a teleost fish, protogynous hermaphrodite characteristic of rocky bottoms, maturing first as females and later in adult life, the ovaries are replaced by the testes, turning them into reproductively males. In this work three sex inversion experiments were performed in captivity in different seasons of the year, using the same dose of aromatase inhibitor (AI) letrozole (100&mu;g/Kg) to promote sexual inversion in E. marginatus, to contribute and improve the knowledge of the physiological mechanisms involved in this process. The first and third experiments started in the spring and early spring, respectively and promoted sexual inversion with a decrease in the gonadosomatic index (GSI) in the animals from the experimental group, that showed a peculiar disorganized gonadal, with lamellae, characteristic from ovaries, however with an interstitial compartment developed with cysts of male germ cells in advanced stages of spermatogenesis (spermatocytes, spermatids), Sertoli cells and a large amount of cysts ruptured with the release of sperm in the lumen. After thirty days from the beginning of the experiment, there was an increase in 11- ketotestosterone (11KT) levels and an unchanged concentration of estradiol (E2); after seventy-seven days, there was a significant increase in testosterone (T) levels, a decrease in the 11KT levels and the maintenance of E2 plasma concentration. In this point of view, mainly the concentration of androgens may have activated the negative feedback loop and with the activation of the kisspeptin system may have promoted the action of GnRH diverting the production of FSH to LH. At this moment the number of copies of pituitary &beta;FSH produced was ignificantly lower than the number of control group copies. The significant increase in plasma 17- alfaOHprogesterona concentration observed could suggest that the action of GnRH has increased the secretion of LH, the main gonadotropin that stimulates the production of progestogen. Inverted males produced an average volume of semen of 118.20 ± 24.83 &mu;l with 90% motility and sperm density of 8.94 ± 4.34 x 109 cells.ml-1. No differences were obtained at the concentrations of total protein in plasma, liver and muscle. In the experiment established in the summer of 2010, the treatment did not promote sexual inversion, the GSI was not significantly different between animals treated with aromatase inhibitor and the control group. In the gonads of the animals from the experimental group there was a slight disruption of gonadal architecture with cysts in formation and a large number of degenerating oocytes showing that during the process of sex inversion may be a phase of intersex. Thirty days after the beginning of the experiment there was a significant increase of T and E2 concentration remained unchanged. No significant differences were observed between the number of copies of &beta;FSH and &beta;LH throughout this experiment. The result obtained for the total protein concentration of the muscle in experimental group showed a significant increase compared with the control animals. There was no spermiation and the sex inversion occurred incompletely, with the presence of intersex individuals. In general we could conclude that the use of an aromatase inhibitor is an effective method to promote the sex inversion in hermaphroditic protogynous as the groupers. The increase of androgens could trigger the sex inversion exerting an important role in gonadal restructuring and the development of male germ tissue in the process

Exploration analytique de la microchimie fine de l'otolithe pour l'écologie et l'étude de la gestion des stocks des poissons coralliens de l'île de la Réunion / No English title available

La gestion des ressources alimentaires doit passer par un équilibre entre activités humaines et une exploitation raisonnée de ces ressources. Sur l'île de La Réunion, la réserve naturelle marine œuvre pour le maintien des populations de poissons traditionnellement exploitées comme le mérou macabit (Epinephelus merra, Serranidae) ou d'intérêt écologique (Acanthurus triostegus, Acanthuridae, et Plectroglyphidodon imparipennis, Pomacentridae). Pour mieux appréhender leur phase de vie larvaire souvent méconnue, la microchimie des otolithes a été utilisée pour retracer la vie des poissons dans un objectif appliqué à la gestion. Nos résultats ont permis de différencier la population de macabit à La Réunion en 3 groupes d'histoires de vie larvaires différentes sur un gradient nord-sud, à partir de fortes variations en baryum, un élément trace provenant de l'environnement. Ces variations sont retrouvées également chez A. triostegus et P. imparipennis, principalement issues d'évènements cycloniques. Des sources de cet élément ont été identifiées dans des eaux du socle basaltique de l'île de La Réunion, mais aussi à l'échelle de l'océan Indien, aux Maldives, Juan De Nova et Europa. Le baryum s'est ainsi révélé être un élément trace à privilégier pour l'analyse de l'histoire de vie des poissons récifaux et pour révéler les caractéristiques spatiales et temporelles de la structure de leurs populations à l'échelle de La Réunion et dans l'océan Indien. Des études complémentaires biologiques, chimiques et génétiques sont néanmoins nécessaires afin d'aboutir à des connaissances permettant d'adapter des mesures de gestion aux cycles biologiques des espèces. / Ood resources must managed between human activities and sustainable exploitation. On Reunion Island, the marine park works more specifically to maintain fish populations with high commercial value, regularly exploited by traditional fishermen, such as the grouper Epinephelus merra (Serranidae), Acanthurus triostegus (Acanthuridae) and Plectroglyphidodon imparipennis (Pomacentridae) which pelagic larval phase remains unknown. To better understand this phase, the otolith microchemistry was used to trace the life histories for management purposes. Our results allowed differentiating grouper population in Reunion in 3 different groups of larval life histories, using barium concentration in their otoliths which can be enriched in the environment. These chemical changes are also found in A. triostegus and P. imparipennis. In the northern reef area (Saint-Gilles and Saint-Paul), these variations are observed significantly mainly during cyclonic events. Natural sources of this element could be identified locally in groundwater collected in the porous basaltic basement of the Reunion, but also at the regional level, where other individuals were collected in the Indian Ocean (Maldives Juan De Nova, Europa). Therefore barium has proven to be an interesting trace element for the analysis of the life story of reef fish, and a tool to reveal the spatial and temporal characteristics of their populations across Reunion structure but also in the Western Indian Ocean. Nevertheless biological, chemical and genetic studies are necessary to complete the knowledge and adapt management measures to the biological cycles of the species.

Effects of dietary Bacillus subtilis spores on utilization of crystalline methionine in juvenile grouper, Epinephelus coioides, fed high plant-protein diets

With the aim to enhance the efficiency of utilization of crystalline methionine supplemented in the high plant-protein diet for grouper (Epinephelus coioides), this study used Bacillus subtitlis spore as a probiotic additive in the diet to shorten the absorption time difference between protein-bound amino acid and crystalline methionine. The study was conducted in two parts. In the first part, juvenile groupers were fed for 14 weeks with 5 experimental diets: fish meal diet, high plant-protein diet with/without crystalline methionine, as well as with/without B. subtitlis spore separately. Growth performance, PER, protein digestibility, amino acid digestibility, non-specific immune responses, and free amino acid concentration in both muscle and serum were assessed. The second part was a time-series study on serum free amino acids concentration after a force-feeding experiment. The results showed that crystalline methionine supplementation in the high plant-protein diet effectively improved the growth of E. coioides (P<0.05). However, B. subtitlis spore supplementation did not affect fish growth performance significantly (P>0.05). A delay in the appearance of peak serum amino acid concentration was observed when fishmeal was partially replaced by soy protein. On the other hand, the force-feeding experiment showed that serum essential amino acid (include methionine) concentrations droped drastically after they reached the peak concentrations from being forced-fed with the B. subtilis containing diet. Supplementation of crystalline methionine seemed to ease the drop of serum methionine concentration. Based on these results, it is concluded that addition of B. subtitlis spore in high plant-protein diet for the grouper does not enhance the utilization of crystalline methionine, but supplementation of crystalline methionine significantly improve the growth performance of the grouper.

Effects of dietary supplemental nucleotides on the health of juvenile grouper Epinephelus coioides

Dietary effects of a nucleotide mixture that contained AMP¡BGMP¡BCMP¡BUMP and IMP in equivalent amount were evaluated in 4 supplemental levels of 0, 0.05, 0.2, and 0.4 g/100g diet. Weight gain (519-616%) of juvenile groupers fed for 14 weeks with the test diets was not significantly affected by the supplements. Serum concentrations of total protein, albumin, and cholesterol, A/G ratio and activities of AST were also not significantly affected. Serum total bilirubin concentration in the 0.4 g/100g group was significantly higher than the 0 or 0.05 g/100g group. Head-kidney macrophage phagocytosis and intracellular superoxide anion production as well as serum lysozyme activity were also not different among treatments. Histological sections of fish gut indicated that villous heights of intestinal fold were significantly greater in the 0.2g/100g group than the 0.4 g/100g group, muscle layer thickness, in contrast, was not affected. When challenged with Photobacterium damsela subsp. damselae after fed the test for 3 weeks, relative percent survivals of the groupers were 87.8, 91.1, 90.0 and 94.4% (0, 0.05, 0.2, and 0.4 g/100g diet, respectively). The survivals were 74.5, 82.4, 86.3, and 85.3%, respectively when the feeding trial was extended to 18 weeks. The results show that a diet containg 38% protein and asupplemented with nucleotides at the tested levels was not effective in promoting growth, but was moderately effective in enhancing immune responses and pathogen resistance of juvenile grouper.

Evaluation of the Efficacy of the Minimum Size Rule in the Red Grouper and Red Snapper Fisheries With Respect to J and Circle Hook Mortality and Barotrauma and the Consequences for Survival and Movement

Although closed seasons, bag limits and quotas are used to manage fishes within the Grouper/Snapper Complex off the southeastern United States, size limits are the cornerstone of fisheries management. Because fishers must release all undersized fishes despite fish condition, this regulation has created a mandatory catch and release system. Inherent in this management strategy is the supposition that these undersized fish survive in sufficient numbers so as to justify this regulation. To satisfy this criteria fish mortality must be low and released fish must also experience minimal sub-lethal effects. Determination of sublethal effects and evaluation of their potential impairment and duration of injury are required to develop effective physiology-based criteria to evaluate the efficacy of the minimum size rule. The goal of this research was to evaluate some aspects of the efficacy of the minimum size rule in the red grouper and red snapper fisheries off Florida by collecting traditional fisheries data and analyzing it in light of fish physiology, ecomorphology and behavior. Study objectives included 1) determination of the causes for the differences of hook mortality for red grouper and red snapper in the recreational and recreational-for-hire fisheries by necropsy of acute and latent mortalities, analysis of tag and recapture data for both J and circle hooks, determination of fish dentition and any differences in feeding behavior, 2) examination of the effects of rapid depression from depth on fish survival by inspection and comparison of the red grouper and red snapper swim bladders in both healthy and swim bladder ruptured fish from various water depths, comparison of tag and recapture data, investigation of the effects of fish venting, and laboratory simulations using fish hyperbaric chambers to determine healing and survival from rapid depression trauma, 3) analysis of movement patterns of tagged fish and 4) evaluation of some of the consequences imposed by the minimum size limit based on study results.

Hormonal correlates of coloration and sexual change in the hermaphroditic grouper, Epinephelus adscensionis

Hermaphroditism, associated with territoriality and dominance behavior, is common in the marine environment. Male sex-specific coloration patterns and behavior are particularly evident in species where males are territorial and guard harems of females such as wrasses and groupers. Protogynous hermaphrodites that change sex from female to male are good models to study sexual behavior and related changes in the brain due to their abilities to reorganize their sexual phenotype as adults. Two hormones produced in the brain and implicated in the process of sex-specific behavior and reproductive development are arginine vasotocin (AVT) and gonadotropin releasing hormone (GnRH). While a wealth of data exists regarding these hormone systems separately, little is known about linkage between these two systems. Especially there is no data tracking these two systems together in any protogynous fish. This study was conducted to test the hypothesis that coordinated interactions between AVT and GnRH facilitate the process of behavioral and gonadal sex change in the rock hind Epinephelus adscensionis. Four topics were addressed to investigate the relationship between behavior and reproduction: i) rock hind sex change, sexual characteristics and conditions causing sex change to occur in captivity were detailed as a basis for examining the AVT system and GnRH during this process, ii) the distribution of a vasotocin V1a type receptor identified in rock hind brain was examined for the first time in a fish species using a custom designed antibody then the receptor protein was co-localized with GnRH producing cells within the brain to confirm that a pathway exists for AVT action on GnRH, iii) levels of AVT, AVT receptors, and GnRH messenger RNA (mRNA) were compared between male and female rock hind phenotypes, and iv) female rock hind at early stages of sex change were compared for brain mRNA expression of AVT, AVT receptors, and GnRH to determine the order of hormonal change during the process of sexual inversion in this species. This study provides a better understanding of the relationship between sex-specific behavior and reproductive development via AVT and GnRH systems that are conserved in all vertebrates. / text

Shelf-scale Mapping of Fish Distribution Using Active and Passive Acoustics

Fish sound production has been associated with courtship and spawning behavior. Acoustic recordings of fish sounds can be used to identify distribution and behavior. Passive acoustic monitoring (PAM) can record large amounts of acoustic data in a specific area for days to years. These data can be collected in remote locations under potentially unsafe seas throughout a 24-hour period providing datasets unattainable using observer-based methods. However, the instruments must withstand the caustic ocean environment and be retrieved to obtain the recorded data. This can prove difficult due to the risk of PAMs being lost, stolen or damaged, especially in highly active areas. In addition, point-source sound recordings are only one aspect of fish biogeography. Passive acoustic platforms that produce low self-generated noise, have high retrieval rates, and are equipped with a suite of environmental sensors are needed to relate patterns in fish sound production to concurrently collected oceanographic conditions on large, synoptic scales. The association of sound with reproduction further invokes the need for such non-invasive, near-real time datasets that can be used to enhance current management methods limited by survey bias, inaccurate fisher reports, and extensive delays between fisheries data collection and population assessment. Red grouper (Epinephelus morio) exhibit the distinctive behavior of digging holes and producing a unique sound during courtship. These behaviors can be used to identify red grouper distribution and potential spawning habitat over large spatial scales. The goal of this research was to provide a greater understanding of the temporal and spatial distribution of red grouper sound production and holes on the central West Florida Shelf (WFS) using active sonar and passive acoustic recorders. The technology demonstrated here establishes the necessary methods to map shelf-scale fish sound production. The results of this work could aid resource managers in determining critical spawning times and areas. Over 403,000 acoustic recordings were made across an approximately 39,000 km2 area on the WFS during periods throughout 2008 to 2011 using stationary passive acoustic recorders and hydrophone-integrated gliders. A custom MySQL database with a portal to MATLAB was developed to catalogue and process the large acoustic dataset stored on a server. Analyses of these data determined the daily, seasonal and spatial patterns of red grouper as well as toadfish and several unconfirmed fish species termed: 100 Hz Pulsing, 6 kHz Sound, 300 Hz FM Harmonic, and 365 Hz Harmonic. Red grouper sound production was correlated to sunrise and sunset, and was primarily recorded in water 15 to 93 m deep, with increased calling within known hard bottom areas and in Steamboat Lumps Marine Reserve. Analyses of high-resolution multibeam bathymetry collected in a portion of the reserve in 2006 and 2009 allowed detailed documentation and characterization of holes excavated by red grouper. Comparisons of the spatially overlapping datasets suggested holes are constructed and maintained over time, and provided evidence towards an increase in spawning habitat usage. High rates of sound production recorded from stationary recorders and a glider deployment were correlated to high hole density in Steamboat Lumps. This research demonstrates the utility of coupling passive acoustic data with high-resolution bathymetric data to verify the occupation of suspected male territory (holes) and to provide a more complete understanding of effective spawning habitat. Annual peaks in calling (July and August, and November and December) did not correspond to spawning peaks (March - May); however, passive acoustic monitoring was established as an effective tool to identify areas of potential spawning activity by recording the presence of red grouper. Sounds produced by other species of fish were recorded in the passive acoustic dataset. The distribution of toadfish calls suggests two species (Opsanus beta and O. pardus) were recorded; the latter had not been previously described. The call characteristics and spatial distribution of the four unknown fish-related sounds can be used to help confirm the sources. Long-term PAM studies that provide systematic monitoring can be a valuable assessment tool for all soniferous species. Glider technology, due to a high rate of successful retrieval and low self-generated noise, was proven to be a reliable and relatively inexpensive method to collect fisheries acoustic data in the field. The implementation of regular deployments of hydrophone-integrated gliders and fixed location passive acoustic monitoring stations is suggested to enhance fisheries management.
