Molekulární podstata autosomálně dominantní polycytémie / Molecular mechanism of autosomal dominant polycythemia

All red blood cells, erythrocytes, originate in bone marrow. The process of their differentiation and maturation is called erythopoiesis and is regulated through hormone erythropoietin (EPO), which functions as a stimulatory factor for erythropoiesis. EPO is produced in kidney and its production is regulated by oxygen supplementation. EPO is transported to bone marrow via blood vessels. Chronic overproduction of erythrocytes leads to disease called polycythemia. Polycythemia may be diagnosed for example by measurement of haematocrit or haemoglobin concentration in blood. EPO level may or may not be increased. Patients suffering from polycythemia may or may not have any symptoms. It depends on manifestation level of the disease. The most common symptoms are higher blood pressure, headaches, dizziness, swelling and epistaxis. Recently, the most common treatment of polycythemia is phlebothomy. The aim of this master thesis is to unravel the role of a newly described mutation, which was found among members of one family suffering from polycythemia with increased EPO level. Algumas glicoproteínas contém ácido siálico, que é o principal responsável pelas cargas negativas na superfície da hemácia que quando em solução cria um potencial elétrico (Ç) repulsivo. A carga elétrica negativa da superfície eritrocitária influencia na distribuição dos íons da solução ao redor da célula formando uma dupla camada de íons. A primeira, conhecida como camada compacta de cargas ou "Stern" é formada por íons rigidamente ligados à hemácia e a segunda camada é composta por íons distribuídos difusamente e conhecida como camada difusa. Sialylated glycoproteins of the RBC membrane are responsible for a negatively charged surface, which creates a repulsive electric zeta potential (Z) between the cells.The compact layer of charge or Stern consists of ions rigidly bonded to the cell and the double layer includes ions diffusely distributed around the cell. The aim of this study was to measure the RBC double layer thickness of the charge (DLC) around the cell, zeta potential (Z) and cell aggregation force in agglutination potentiator solutions, using optical tweezers. Amplitude de distribuição do diâmetro dos eritrócitos (RDW), volume corpuscular médio e reticulócitos em gato doméstico hígido (Felis catus Linnaeus, 1758) / Red cell distribution width (RDW), mean cell volume and reticulocytes in healthy domestic cat (Felis catus - Linnaeus, 1758)

The use of modern equipment that make the blood cell count used to calculate the RDW (red cell distribution width) and its fractions RDW-CV and RDW-SD, which provide a quantitative assessment of heterogeneity of each erythrocyte. Given the limited information available in the literature about the RDW for the domestic cat, this study aims to compare the values of RDW, with MCV and reticulocytes from 40 (n=14males, n=26 females) healthy mongrel cats, with age ranging from 1.2 to 5.5 years (3.0 ± 1.2) were used. The animals were kept in individual cages all over the experiment. Blood samples were taken directly from the external jugular vein and separated into two tubes: one containing anticoagulant EDTA (ethylene diamine tetra acetic acid) to 10% for the complete blood count and the second without anticoagulant for biochemical tests. The results showed that there is a direct relationship between values of RDW-SD and RDW-CV and an inverse correlation between RDW-CV with MCV. There was no relation between RDW and reticulocyte aggregates or punctate. There was no relation between sex and RDW as well. RDW values determined in this experiment can be used as a resource for future research and also as reference values for the feline species. / As alterações nos resultados dos padrões eritrocitários demandam decisões e norteiam a conduta do clínico quanto ao prognóstico e a terapêutica de eventuais patologias. A utilização de equipamentos modernos que fazem a contagem celular sanguínea permite calcular a amplitude de distribuição dos eritrócitos ou RDW (red cell distribution width) e suas frações RDW-CV e RDW-SD, que provêem uma avaliação quantitativa da heterogenicidade de cada eritrócito. Diante da escassa informação disponível na literatura científica sobre os valores de RDW para o gato doméstico, este estudo se propõe a comparar os valores de RDW com o VCM e reticulócitos de 40 gatos domésticos hígidos, sem raça definida, com idade variando de 1,2 a 5,5 anos (3,0±1,2), sendo 14 machos e 26 fêmeas provenientes do gatil da instituição. O sangue foi colhido diretamente da veia jugular externa e separado em dois tubos: um contendo anticoagulante EDTA (ácido etileno diamino tetra acético) a 10%, para a realização do hemograma e o segundo, sem anticoagulante, para as provas bioquímicas de triagem. Os resultados encontrados mostraram que existe uma relação direta significativa entre os valores de RDW-SD e RDW-CV e uma correlação inversa entre os valores de RDW-CV o VCM. Não houve correlação entre os valores de RDW e reticulócitos agregados ou ponteados. O sexo não interfere no diâmetro das hemácias. Os valores de RDW determinados neste experimento podem ser usados como fonte de consulta para futuras pesquisas e também como valores de referência da espécie felina.

Sezónní dynamika vybraných krevních parametrů u vybraných masných plemen ovcí chovaných v podhorských podmínkách / Seasonal dynamics of selected blood parametres of selected flesh breeds of sheep bred in foothills conditions

Sheep breeding is nowadays a developing branch of agriculture again. There is a lot of different breed and they react distinctly on the same conditions of the enviroment. Sheep of breeds charollais, suffolk, šumavská ovce and valaška bred in similar conditions were observed in spring and autumn of years 2007 and 2008. In these seasons were taking blood samples (from {$\pm$}7{--}24) ewes and lambs and were analyzed in hematology laboratory. There were determined haemoglobin level, haematocrit indicator, erytrocytes and leucocytes levels, glucose, cholesterol, triglycerides levels, urea and plasmatic proteins, activity of ALP and GMT enzymes, phosphor, calcium, magnesium, zinc and copper levels. The main objective of this project was determine seasonal changes in observed parametres. Next objectives were determine different changes in blood parametres in different breeds and different aimes of breeds. There were recognized that all the observed breeds don`t react the same way on similar conditions. There were not provably determined seasonal changes in observed parametres, but average Hb level was higher in autumn than in spring. Urea level was conversely higher in spring season than in autumn. The demostrable fact is, that the similar conditions induce different answers not only in different breeds but also in different aimes of breeds.
