Evalution of a Model for Experimentally-Induced Osteoarthritis in the Hip Joint of the Dog

Twelve normal mixed-breed dogs were selected and assigned to acetabular rotation or sham-operated groups. Rotation group dogs had pelvic osteotomies followed by application of an ilial bone plate, causing rotation of the acetabulum to reduce dorsal coverage of the femoral head. Sham group dogs received identical osteotomies but were plated in normal orientation. All dogs had force plate and subjective lameness evaluations pre-operatively and post-operatively. Pelvic radiographs were evaluated pre-operatively and at four-week intervals post-operatively. The dogs were killed at the conclusion of the study, and samples were taken for evaluation. Analysis of vertical ground reaction forces indicated a significant difference between treatment groups, however no difference was noted based on subjective scores. No difference was observed between groups based on radiographic estimations of degenerative changes or Norberg angle, however the coverage of the femoral head was less in rotated dogs. Mild inflammatory changes were discernible in the joint capsule of some dogs in both groups. A trend toward more severe change was present in the left hip of the treatment group, but statistically significant difference was noted only when comparing the right and left legs of the acetabular rotation group. Although evidence of osteoarthritis was noted histologically, only minor differences were detected in the dogs by other means. Because of the minimal changes noted over the duration of the study, we conclude that the model does result in histopathologic change consistent with osteoarthritis, but that force plate analysis, radiographic evaluation, and clinical lameness evaluation are insensitive measures of this change. / Master of Science

Experimental model for predicting cutting forces in machining carbon fiber reinforced polymer composites

The demand for materials with high mechanical performances such as Carbon Fiber Reinforced Plastics (CFRP) is increasing. However, there are major challenges in machining CFRP as it involves delamination, fiber pullouts, and extreme cutting tool wear. Analysis of chip formation mechanisms and prediction of associated cutting forces in CFRP machining enables one to address these challenges. This study proposes a mechanistic cutting force model for milling operations of the CFRP workpiece, considering its non-homogeneity and anisotropy, by taking into account variations of fiber cutting angle during machining. A mechanistic model of cutting force constants is obtained from a number of experimentally measured unidirectional CFRP milling forces. The obtained mechanistic force model predictions are verified against experimentally measured milling forces with arbitrary tool path indicating the accuracy of the proposed mechanistic model in predicting cutting forces. The proposed mechanistic cutting force model is capable of being integrated into the manufacturing process to allow optimized machining of quality certified CFRP work-pieces. / Graduate

Estudo teórico e experimental de um controlador para supressão de Flutter / Theoretical and experimental study on a Flutter suppression controller

Flutter é uma instabilidade aeroelástica dinâmica que envolve a interação de forças aerodinâmicas, elásticas e inerciais. Esta instabilidade pode ocorrer em superfícies de aeronaves, como asas, que apresentarão um comportamento oscilatório auto-sustentado e possíveis problemas estruturais se o mesmo não for suprimido. Um dos tipos clássicos de flutter envolve o acoplamento dos modos de vibrar de flexão e torção. Este tipo de flutter binário é conhecido como flutter flexo-torcional. Um dispositivo flexível é desenvolvido para testes de flutter flexo-torcional com asas rígidas em túnel de vento. O procedimento de projeto deste dispositivo flexível é baseado em simulações realizadas com um modelo em elementos finitos cujos resultados são testados em simulações realizadas com um modelo aeroelástico formulado para simular o comportamento aeroelástico do sistema experimental. Então, para se verificar os resultados analíticos, uma análise modal experimental é realizada e as freqüências e a forma dos modos são identificadas utilizando-se o Eigensystem Realization Algorithm. Depois disso, alguns testes em túnel de vento são realizados para a verificação da obtenção do flutter, para a caracterização do flutter e para a identificação do flutter. O desenvolvimento deste sistema experimental permite o estudo e a aplicação de leis de controle para a supressão ativa do flutter, que é o objetivo principal deste trabalho. Um controlador ativo para supressão de flutter através de realimentação de estados é projetado a partir do modelo Aeroelástico previamente desenvolvido. Este controlador é inicialmente testado em simulação e, então, são realizados experimentos em túnel de vento. O objetivo é suprimir o flutter e manter a estabilidade do sistema em malha fechada. O modelo para testes no túnel de vento é uma asa rígida retangular com perfil NACA 0012 com uma superfície de controle no bordo de fuga utilizada como atuador. / Flutter is a dynamic aeroelastic instability that involves the interaction of aerodynamic, elastic and inertial forces. This instability may occur in aircraft surfaces, like wings, which will present a self-sustained oscillatory behaviour and possible structural problems if not suppressed. One of the classical types of flutter involves the coupling of bending and torsion vibration modes. This binary type of flutter is known as flexural-torsional flutter. A flexible mount system is developed for flexural-torsional flutter tests with rigid wings in wind tunnels. The design procedure of this mount system is based in simulations performed with a finite element model which results are tested in simulations performed with an Aeroelastic model formulated to simulate the aeroelastic behaviour of the experimental system. Then, to verify the analytical results, an experimental modal analysis is performed and mode shapes and frequencies are identified using the Eigensystem Realization Algorithm. After this, some wind tunnel tests are performed to verify flutter achievement, for flutter characterization and for flutter identification. The development of this experimental system allows the study and application of active control laws for active flutter suppression, which is the main goal of this work. A state feedback controller for active flutter suppression is designed using the aeroelastic model previously developed. This controller is initially tested in simulations and, then, wind-tunnel experiments are performed. The goal is to suppress flutter and to maintain the stability of the closed loop system. The wind tunnel model is a rigid rectangular wing with a NACA 0012 airfoil section with a trailing edge control surface used as actuator.

Exequibilidade e definição de intervalos de referência para variáveis ecocardiográficas convencionais e de deformação miocárdica pelo método de rastreamento de pontos em hamsters sírios adultos / Feasibility and reference intervals for conventional echocardiography measures and myocardial strain by Speckle tracking in adult Syrians hamsters

O hamster sírio é utilizado como modelo experimental que permite explorar aspectos fisiopatológicos e avaliar a resposta terapêutica em diversas cardiopatias, algumas delas não tão bem reproduzidas em ratos. A avaliação cardiológica inclui a observação de aspectos fisiológicos e hemodinâmicos, assim como a utilização de exames complementares para análise estrutural e funcional cardíaca in vivo. Dentre esses exames destaca-se a ecocardiografia, que possibilita a avaliação da estrutura e função com elevada acurácia. Uma das formas de análise da função sistólica é por meio do cálculo da fração de ejeção do ventrículo esquerdo (FEVE), que leva em consideração os volumes estimados diastólico final e sistólico final do ventrículo esquerdo. Outro importante parâmetro de análise de função sistólica é a avaliação da deformação miocárdica. Os índices de deformação miocárdica podem ser obtidos utilizando o método de rastreamento de pontos. No entanto, até o presente momento, não existem dados a respeito de valores de referência a serem utilizados para a deformação miocárdica em hamsters. Diante disso, o presente estudo teve como objetivo definir a exequibilidade, os intervalos de referência e a variabilidade de parâmetros derivados da ecocardiografia convencional e da técnica de rastreamento de pontos em hamsters sírios adultos. Métodos: Foram incluídos no estudo 135 hamsters sírios fêmeas, anestesiados com ketamina/xilazina e submetidos à ecocardiografia em aparelho dedicado a pequenos animais, Vevo® 2100 utilizando transdutor linear de 30 MHz. Resultados: A exequibilidade da realização das medidas de ecocardiografia convencional foi superior a 93%. A análise da deformação endocárdica apresentou exequibilidade de 84% no eixo longo e, de 80% no eixo curto. A avaliação da deformação epicárdica nos eixos longo e curto apresentou baixa exequibilidade (42% e 62%, respectivamente). A FEVE pelo modo M, obtida no eixo longo, foi significativamente superior à obtida pelos métodos de área comprimento e de rastreamento de pontos (59,0±5,8; 53,8±4,7; 46,3±5,7; p<0,0001). Houve razoável correlação positiva da FEVE obtida pelos métodos de área comprimento e de rastreamento de pontos (r= +0,43; p<0,0001). A deformação longitudinal endocárdica global foi de -13,6(-7,5; -20,4)%, a deformação circunferencial endocárdica global foi de -20,5±3,1% e a deformação radial endocárdica global foi de +34,7±7,0%. A análise de regressão linear evidenciou influência da frequência cardíaca nos valores de deformação longitudinal e circunferencial, mas não da deformação radial (p=0,0003; p<0,0001 e p=0,83, respectivamente). Conclusões: Demonstrou-se que as medidas de ecocardiografia convencional e pelo método de rastreamento de pontos são exequíveis em hamsters. Foram definidos intervalos de referência para variáveis de ecocardiografia convencional e da deformação endocárdica global longitudinal, circunferencial e radial neste modelo experimental animal. / The Syrian hamster is an experimental model to study pathophysiology and to assess therapeutic response in several heart diseases, some of them not well reproduced in rats. The cardiologic evaluation includes observation of physiological and hemodynamic patterns, and complementary tests are used for in vivo cardiac structural and functional analysis. Among these, echocardiography allows the evaluation of structure and function with high accuracy. Analyzing systolic function can be done by calculating the left ventricular ejection fraction. Another important parameter of systolic function analysis is the assessment of the myocardial strain. Myocardial strain indices can be obtained using speckle tracking. However, there are no data regarding reference values for myocardial strain in hamsters. The present study aimed to determine feasibility, reference intervals and variability of parameters derived from conventional and speckle tracking echocardiography in adult Syrian hamsters. Methods: The study included 135 female Syrian hamsters anesthetized with ketamine/xylazine and submitted to a small animal dedicated echocardiography, Vevo® 2100 using a 30 MHz linear transducer. Results: The analysis of the endocardial strain presented a feasibility of 84% in the long axis and 80% in the short axis. The evaluation of the epicardial strain in the long and short axes presented low feasibility (42% and 62%, respectively). The M-mode left ventricular ejection fraction obtained on the long axis was significantly higher than that obtained by the area-length and Speckle tracking methods (59.0±5.8, 53.8±4.7, 46.3±5.7, p<0.0001). There was a reasonable positive correlation of left ventricular ejection fraction obtained by the area length method and the Speckle tracking method (r=+0.43; p<0.0001). The endocardial longitudinal strain was -13.6(-7.5;-20.4)%, the endocardial circumferential strain was -20.5±3.1% and the endocardial radial strain was +34,7±7.0%. Linear regression analysis showed influence of heart rate on the values of longitudinal and circumferential strain, but not on radial strain (p=0.0003, p<0.0001 and p=0.83, respectively). Conclusions: it has been shown that measurements of conventional echocardiography and speckle tracking are feasible in hamsters. Reference intervals for conventional echocardiographic variables and longitudinal, circumferential and radial global endocardial strain were obtained in this experimental animal model.

Safety Analysis of the Baihetan Dam : By Investigating the Pressure Distribution on the Plunge Pool Floor

Baihetan Dam is sited on the lower reaches of the Jinsha River in the southwest of China between the boarders of Sichuan and Yunnan province. The dam is scheduled to be taken into operation in the year of 2020 with an installed generation capacity of 14 GW which will put Baihetan Dam on the map as the third largest hydropower station in the world considering installed power output. In comparison, the world’s biggest dam Three Gorges has an installed generation capacity of 22,5 GW. To ensure a sufficient safety evaluation in terms of erosion (scour) formation at the bottom of the plunge pool, pressure simulations in the plunge pool floor in an experimental model at the Department of Hydraulic Engineering in Tsinghua University, Beijing, China has been performed. Data from two experiments with two different outflow configurations has been obtained and analyzed together with three earlier performed experiments on the same experimental model. The results from outflow configuration one had an incomplete data set and could not be compared to the other experiments. The results retrieved from the other experiments however showed the importance of a spillway design with nappe splitters and nappe blocks implemented and the value of a sufficient water cushion in the plunge pool. All four outflow configurations with nappe splitters or nappe blocks implemented held a hydrodynamic pressure below the recommended maximum pressure value of 15 cm water head (experimental model scale) stated by the East Asian Investigation and Design Institute, whilst the one configuration with no nappe splitters or nappe blocks exceeded the value. The design of outflow configuration two is four nappe splitters implemented in two spillways and two nappe blocks in two spillways. In this thesis, this configuration has proven to be the most suitable one in terms of maximum pressure minimization and pressure distribution at the plunge pool floor.

Além das suas propriedades nutricionais, há evidências de sua participação na redução das concentrações lipídicas do soro. Por outro lado, o fármaco rosuvastatina tem suas ações demonstradas no metabolismo lipídico, por reduzir a fração LDL-colesterol, bloqueando a chave do processo de produção de colesterol hepático. Embora a eficiência do medicamento seja comprovada, é importante considerar a alimentação como uma possível ação somatória, tendo em vista os graves problemas ocasionados pelas alterações lipídicas no organismo. À vista do exposto, a presente pesquisa teve por objetivo estudar o efeito da glicinina isolada da soja em comparação ao fármaco rosuvastatina em animais submetidos à dieta hipercolesterolêmica. Foram elaboradas duas dietas, uma padrão, contendo caseína como proteína (AIN-93M), e outra hipercolesterolêmica, composta pela dieta padrão adicionada de 1% de colesterol e 0.5% de ácido cólico. A proteína glicinina de soja (300 mg/kg de peso corporal) e o fármaco rosuvastatina (10 mg/Kg de peso corporal), ambos solubilizados em solução salina, foram administrados por gavagem. Foram utilizados ratos machos Wistar mantidos em gaiolas individuais sob condições adequadas. Os animais foram separados em cinco grupos (n = 9): 1) grupo padrão (STD) recebeu a dieta padrão; 2) grupo hipercolesterolêmico (HC) recebeu apenas a dieta hipercolesterolêmica; 3) grupo (HC+11S) recebeu a dieta e a proteína glicinina da soja; 4) grupo (HC+ROS) recebeu a dieta e o fármaco; 5) grupo (HC+ROS+11S) recebeu a dieta, a proteína e o fármaco. Ao final de 28 dias os animais foram sacrificados e o sangue removido para análises bioquímicas de colesterol total (CT), HDL-colesterol (HDL-C) e triglicérides (TG) plasmático, CT e TG hepático... / The 11S globulin (glycinin) is the predominant protein of soybeans and has been extensively characterized. In addition to its nutritional properties there is evidence of its action in reducing serum lipid concentrations. Moreover, the drug rosuvastatin has demonstrated its effects on lipid metabolism, by reducing LDL cholesterol and blocking the key enzyme in the process of cholesterol production in the liver. Although the efficiency of the drug is proven, it is important to consider the sum of its action and that of food, in view of the serious problems caused by changes in body fat. In view of the foregoing, the aim of the present study was to investigate the effects of isolated soy glycinin, as compared to the drug rosuvastatin, in rats subjected to a hypercholesterolemic diet. Two diets were prepared, a standard diet, containing casein as protein (AIN-93M), and a hypercholesterolemic, consisting of the standard diet plus 1% cholesterol and 0.5% cholic acid. The glycinin (300 mg/kg body weight) and the rosuvastatin (10 mg/kg body weight), both dissolved in saline, were administered by gavage. Male Wistar rats were kept in individual cages under appropriate conditions. The animals were divided into five groups (n=9): 1) standard group (STD) received the standard diet; 2) hypercholesterolemic group (HC) received the hypercholesterolemic diet alone; 3) group HC+11S received the HC diet and the glycinin; 4) group HC+ROS received the HC diet and the drug; 5) group HC+ROS+11S received the HC diet, the protein and the drug. After 28 days, the animals were sacrificed and the blood removed for biochemical analysis of total plasma cholesterol (TC), HDL-cholesterol (HDL-C) and triglycerides (TG), and hepatic TC and TG. É causada pela ingestão de ovos de Taenia solium, cujas larvas se instalam nos diversos espaços intracranianos e no parênquima encefálico. Uma de suas principais complicações é a hidrocefalia, que pode estar presente em até 30% dos casos. O tratamento da hidrocefalia na neurocisticercose é mais complexo que nas outras formas de hidrocefalia. As taxas de disfunção de derivação ventricular para o peritônio são altas, bem como as taxas de morbidade e mortalidade associadas. Recentes modelos experimentais de neurocisticercose têm sido estudados com foco na relação entre parasita e hospedeiro. Por outro lado, modelos experimentais de hidrocefalia existem há muito tempo e têm contribuído para o entendimento das lesões no tecido encefálico causadas pelo desbalanço entre produção, circulação e absorção do líquor. O objetivo deste trabalho foi desenvolver um modelo experimental de hidrocefalia induzida por neurocisticercose. Inoculamos cistos de Taenia crassiceps (cestodo análogo ao T. solium) no espaço subaracnóideo de ratos através de punção suboccipital da cisterna magna. Por ressonância magnética observamos dilatação do sistema ventricular e das cisternas basais. Exame microscópico demonstrou lesões estruturais no encéfalo induzidas pela hidrocefalia, bem como infiltrados inflamatórios nos compartimentos liquóricos. Assim, desenvolvemos um modelo que reproduz a hidrocefalia por neurocisticercose humana, em que há um componente mecânico de obstrução da circulação do líquor pelo parasita, bem como por inflamação desencadeada pela resposta do hospedeiro contra o parasita.

O papel dos nucleotídeos da adenina na função vascular e plaquetária já está bem estabelecido. Apirase (CD39) faz parte de uma família de ecto-enzimas capazes de hidrolisar nucleosídeos di- e trifosfatados da adenosina e sua participação no sistema tromborregulador tem sido estudada. Nós utilizamos um modelo experimental in vivo de trombose arterial aguda para testar a hipótese de que a administração de apirase solúvel pode prevenir a formação de trombos. Vinte e cinco coelhos brancos Nova Zelândia foram submetidos à lesão arterial com cateter balão e, após 15 dias, a um protocolo indutor de trombose. Treze animais receberam duas administrações intravenosas de apirase solúvel (com 90 minutos de intervalo) e 12 animais foram utilizados como controle. Após 3 horas do protocolo indutor de trombose, os animais foram mortos e a taxa e área de trombose foram avaliadas. A taxa de trombose no grupo apirase foi significativamente menor que no grupo controle (69% vs. 16,7%, respectivamente, P=0,015) assim como a área de trombose (1,7 mm2 ± 4,3 vs. 21,7 mm2 ± 37,4, respectivamente, P=0,008). Nossos resultados confirmam a participação da apirase na homeostasia através de um potente efeito antitrombótico. / The role of adenine nucleotides on vascular and platelet functions has long been established. Apyrase (CD39) takes part of a family of ecto-enzymes that hydrolyze adenosine di and triphosphate and its participation on thromboregulatory system is under study. We used an in vivo experimental model of acute arterial thrombosis to test the hypothesis that administering soluble form of potato apyrase could prevent thrombus formation. Twenty five white New Zealand male rabbits suffer balloon aortic endothelium denudation and fifteen days after were submitted to a thrombosis triggering protocol with a procoagulant (Russel’s viper venom) and epinephrine. After the thrombosis triggering protocol 13 animals received two soluble apyrase administration (with 90 minutes interval) and 12 animals that received no treatment were used as controls. Three hours after the triggering protocol, the animals were killed and the rate and area of arterial thrombosis were analyzed. The rate of thrombosis in the apyrase group was significantly lower than the control group (69% vs. 16,7%, respectively, P= 0,015) as well as the area of thrombosis (1,7 mm2 ± 4,3 vs. 21,7 mm2 ± 37,4, respectively, P=0,008). Our results confirm that apyrase do participate in homeostasis through a potent antithrombotic effect.
