Rôle de l’autophagie dans la biogenèse des vésicules membranaires apoptotiques

L’ischémie/reperfusion (I/R) occurrente à toutes transplantations d’organe solide, constitue un stimulus pro-autophagique/pro-apoptotique pour les cellules endothéliales. Nous avons récemment démontré que les cellules endothéliales apoptotiques (CEapo) sécrètent des vésicules apoptotiques exosome like (ApoExo) qui induisent une réponse auto-immune et accélèrent le rejet vasculaire dans un modèle de greffe aortique chez la souris. Ces ApoExo qui diffèrent des corps apoptotiques classiques par leur structure et leur contenu protéique contiennent le fragment C-terminal du perlécan, LG3. Le LG3, un auto-antigène d’importance en transplantation, favorise le remodelage vasculaire et est augmenté en circulation chez les patients greffés rénaux atteints d’un rejet vasculaire. De plus, la présence d’anticorps anti-LG3 avant la transplantation est associée à un risque plus élevé de développer un rejet vasculaire chez des patients greffés du rein et à la perte du greffon à long terme. Il est connu que la génération du fragment LG3 implique la protéolyse du perlécan par la cathepsine L, une protéase lysosomiale, mais le mécanisme de transport de ce fragment au sein des ApoExo est encore inconnu. Nous avons émis l’hypothèse que l’activité lysosomiale et l’autophagie jouent un rôle important dans la maturation et le chargement du LG3 dans les vésicules ApoExo sécrétées par les CEapo. Une étude longitudinale de microscopie électronique chez les cellules endothéliales humaines carencées en sérum pendant 1h, 2h et 3h, nous a révélé la présence du perlécan / fragment LG3 au sein de compartiments autophagiques (autophagosomes et autophagolysosomes) à différentes étapes du processus autophagique. Après 3 heures de carence en sérum, nous avons identifié le fragment LG3 dans des vésicules membranaires situées au sein de grands réseaux vacuolaires s’apparentant à des autophagolysosomes. L’inhibition de la cathepsine L, de l’acidification du lysosome et de l’autophagie diminuent la présence du fragment LG3 dans les vésicules ApoExo sans affecter la sécrétion de vésicules démontrant donc le rôle de l’autophagie dans l’intégration du fragment LG3 au sein des ApoExo. Toutefois, l’injection de vésicules ApoExo LG3-, provenant de cellules murines aortiques traitées à la bafilomycine, dans un modèle murin de greffe aortique induit une réponse auto-immune anti-LG3 ainsi qu’un remodelage vasculaire à des niveaux similaires aux souris ayant reçu des ApoExo véhicule. Une analyse protéomique de ces vésicules ApoExo LG3-, nous a révélé que la bafilomycine modifie le contenu protéique des ApoExo en induisant une augmentation de la présence de protéines lysosomiales et de la matrice extracellulaire dont le perlécan. Ceci suggère que la présence du motif LG3 au sein du perlécan natif non clivé dans les ApoExo LG3- pourrait être responsable de la mise en place de la réponse anti-LG3 observées chez les souris. Les travaux de ce doctorat ont permis de mettre en évidence que l’autophagie dans les cellules endothéliales productrices d’ApoExo ne régule pas l’immunogénicité des ApoExo. Cependant, elle régule au sein des cellules endothéliales apoptotiques le transport et le clivage du perlécan qui lui est immunogène. En effet, l’autophagie module les différentes formes de perlécan natif et clivé sécrétées dans les ApoExo. L’étude chez la souris greffée nous a donc permis de considérer non plus l’implication du fragment LG3 mais l’implication du motif LG3 présent au sein du perlécan natif non clivé et de ses différentes formes intermédiaires dans la réponse auto-immune anti-LG3 et le rejet vasculaire. La modulation de la sécrétion du perlécan et du LG3 dans les ApoExo constitue une cible thérapeutique potentielle afin de diminuer la réponse auto-immune pouvant augmenter le dommage vasculaire après la greffe / Ischemia/reperfusion (I/R) occurring in all solid organ transplantation, constitute a proautophagic / pro-apoptotic stimulus on endothelial cells. We recently demonstrated that apoptotic endothelial cells (CEapo) secrete apoptotic exosome-like vesicles (ApoExo) that induce autoimmune response and accelerate vascular rejection in a mouse aortic transplant model. These ApoExo, that differ from classical apoptotic bodies in structure and protein content, contain the C-terminal fragment of perlecan, LG3. LG3, an autoantigen of importance in transplantation, promotes vascular remodeling and is increased in circulation in renal transplant patients undergoing vascular rejection. In addition, the presence of anti-LG3 antibodies prior to transplantation is associated with a higher risk of developing vascular rejection in kidney transplant patients and long-term graft loss. It is known that the generation of LG3 fragment involves the proteolysis of perlecan by cathepsin-L, a lysosomal protease, but the mechanism of export of this fragment within ApoExo is still unknown. We hypothesized that lysosomal activity and autophagy play an important role in the maturation and the secretion of LG3 in ApoExo vesicles secreted by CEapo. Longitudinal electron microscopy study after 1h, 2h and 3h in serum starved endothelial human cells revealed the presence of perlecan / LG3 fragment within autophagic compartments (autophagosomes and autophagolysosomes) at different stages of the autophagic process. After 3 hours of serum starvation, we identified LG3 fragment in membrane vesicles located within large vacuolar networks reminiscent of autophagolysosomes. Inhibition of cathepsin-L, of lysosomal acidification and of autophagy decrease the presence of LG3 fragment in ApoExo vesicles without affecting vesicle secretion thus demonstrating the role of autophagy in the secretion of LG3 fragment within ApoExo. However, Injection of ApoExo LG3- vesicles from bafilomycin-treated aortic murine cells into a murine aortic transplant model induces autoimmune anti-LG3 response and vascular remodeling at levels similar to the ApoExo vehicle mice control group. Proteomic analysis of ApoExo from bafilomycin-treated aortic murine cells has demonstrated that bafilomycin modifies ApoExo protein content by inducing an increase of the presence of lysosomal proteins and the extracellular matrix, including perlecan. This suggests that the presence of LG3 motif in uncleaved native perlecan in ApoExo LG3- could be responsible for the establishment of the anti-LG3 response as well as the vascular remodeling observed in mice. Collectively, these results demonstrate that autophagy in endothelial cells producing ApoExo does not regulate the immuogenicity of ApoExo. However, it regulates within apoptotic endothelial cells the processing and cleavage of perlecan who is immunogenic. Indeed, autophagy modulates the different forms of native and cleaved perlecan secreted in ApoExo. Grafted mice study thus allowed us to consider neither the involvement of the LG3 fragment but the implication of the LG3 motif present in the uncleaved native perlecan and its intermediate forms in the anti-LG3 autoimmune response and vascular rejection. Modulating perlecan and LG3 secretion in ApoExo vesicles is a potential therapeutic target to reduce the autoimmune response that can increase vascular damage after transplantation.

Transmissive spongiform encephalopathies (TSEs) are neurodegenerative diseases characterized by depositions of abnormally folded prion protein (PrPTSE ) in brain. PrPTSE is at present the only specific biochemical marker of human and animal TSEs. Diagnostic tests are based on the detection of PrPres after proteinase K digestion of brain homogenate using Western blot or on the immunohistochemistry of fixed brain tissue, which are both difficult and time consuming. In this work we focused on development of a new type of tests based on PrP detection without need of proteinase K digestion. As deposits of PrPTSE remain in the body for a long time, there is a substantial chance of them being nonenzymatically modified by glycation. The detection of glycated PrPTSE may have a potential to serve as a diagnostic marker. We prepared monoclonal antibodies specific for carboxymethyl lysine/arginine modified prion protein. Bacterially expressed and purified recombinant human prion protein (rhPrP) was modified by glyoxylic acid that introduces carboxymethyl groups on lysine and arginine residues present within the molecule of the protein. Il frammento nominale nell’italiano digitato colloquiale. Proposta di classificazione sintattica, prospettive di analisi e applicazioni sul campo

Questo studio si concentra sull’analisi di un fenomeno assai comune nell’italiano e ben attestato da oltre un secolo in diverse altre lingue, antiche e moderne: le costruzioni prive di un verbo in forma finita nel loro nucleo sintattico principale, che evidentemente non sono state oggetto di una ellissi e che non sempre possono essere definite frasi. Dopo le analisi su questo fenomeno fatte da Mortara Garavelli (1971) sullo scritto letterario e da Cresti (1998) sul parlato colloquiale, in questa ricerca si vuole indagare la natura delle costruzioni senza verbo in una nuova varietà di italiano, ossia lo scritto informale e dialogico prodotto sul web, che sarà definito in questa ricerca come italiano digitato colloquiale (IDC). Pertanto, questo studio adotta un approccio corpus-based, ricercando le costruzioni senza verbo in una raccolta di testi di IDC realmente prodotti, ossia nel corpus COSMIANU (Corpus Of Social Media Italian Annotated with Nominal Utterances) (Comandini et al., 2018). Si è dunque deciso di individuare il fenomeno sulla base della definizione di enunciato nominale di Ferrari (2011; 2014), ma adottando due prospettive sintattiche ancora mai applicate in ambito italiano: la teoria sentenzialista di Merchant (2004; 2006; 2010) e quella non-sentenzialista di Barton & Progovac (2005), entrambe applicate in inglese a strutture ellittiche definite frammenti senza antecedente esplicito. Pertanto, si è deciso di definire le strutture senza verbo studiate come frammenti nominali, nell’ottica tanto di inquadrare un fenomeno che, nella nuova varietà di lingua studiata, assume forme diverse rispetto allo scritto letterario e al parlato colloquiale, quanto di unire simbolicamente due tradizioni di studio delle costruzioni senza verbo che non si sono mai incontrate, ossia quella italo-francese, risalente a Meillet (1906), e quella anglo-americana, risalente a Sweet (1900). Grazie all’analisi dei frammenti nominali nell’italiano digitato colloquiale in ottica non-sentenzialista, si so-no individuate undici classi di frammenti nominali, alcuni dei quali possono essere considerati delle frasi, poi-ché contengono o un rapporto predicativo tra due costituenti, o una Tense Phrase al proprio interno. Sul fronte dell’analisi sentenzialista, invece, si è ipotizzata l’esistenza di una nuova categoria di frammenti nominali, nei quali è stato eliso un elemento pro e un verbo essere. Grazie al contributo tanto della teoria sentenzialista, quanto di quella non-sentenzialista, è stato possibile notare come l’ IDC abbia come uno dei tanti tratti diagnostici proprio la presenza di frammenti nominali che ne incarnano le caratteristiche principali, ossia: a) l’estrema natura dialogica, che quindi spiega l’alta presenza di formule di saluto e di ringraziamento (es.: CIAO A TUTTE LE FANS; grazie 1000000000000) e di interiezioni (es.: bleah!); b) la forte aderenza al contesto comunicativo, con frammenti nominali che hanno come nodo iniziale un NP, un DP o un AP che fa direttamente riferimento a un elemento precedentemente reso rilevante nel contesto (es.: Bellissimoooooooooooo !!!!!!!!!!!!), oppure a un elemento immediatamente successivo, di cui si specifica la natura (es.: una domanda... perché é all'inverso?). Successivamente, si è testato come l’individuazione e l’analisi sintattica dei frammenti nominali possa aiutare a comprendere e a riconoscere meglio l’hate speech. Analizzando i frammenti nominali portatori d’odio nel corpus di tweet razzisti POP-HS-IT (Comandini & Patti, 2019), si è notato come l’ IDC d’odio presenti le medesime classi di frammenti nominali individuate in COSMIANU, ma in percentuali diverse, con una partico-lare rilevanza dei frammenti nominali che hanno come nodo iniziale un FocP (es.: FUORI QUESTE MERDE UMANE DALL'ITALIA). Inoltre, si è trovata una notevole presenza di frammenti nominali di classe FocP (es.: pezzi di merda loro e tutto l’islam) corrispondenti alle frasi esclamative studiate da Munaro (2006) (es.: Noioso, il tuo amico!), in cui l’elemento focalizzato a sinistra (pezzi di merda) è sempre una caratteristica intrinseca e non temporanea del soggetto (loro e tutto l’islam). Questa tipologia di frammenti nominali esclamativi e focalizzati veicola alcune delle caratteristiche più universali dell’hate speech, ossia l’espressione di un odio generalizzato e non dibattibile verso una categoria di persone vista come un gruppo monolitico. L’individuazione dei frammenti nominali più caratteristici dell’hate speech potrebbe aiutare i tool automatici ad annotare i testi d’odio in maniera più accurata.

Fiabilité des installations industrielles sous impact de fragments de structures - Effet domino

La plupart des sites industriels abritent des équipements et des réservoirs sous pression. Pour des raisons diverses (suppression, impact mécanique, surchauffe ou autre), ils peuvent être endommagés et même éclater. Cette explosion peut engendrer de nombreux projectiles. Au cours de leur vol, ces derniers peuvent impacter d'autres équipements, tels que des réservoirs sous pression ou d'autres installations sensibles (poste de commande, par exemple). Si une des cibles impactées subit une ruine mécanique, elle peut exploser et générer une nouvelle série de projectiles. Ces projectiles menacent, à leur tour, d'autres installations et ainsi de suite. Ce type d'enchaînement accidentel catastrophique est connu sous le nom d'effet domino ou de suraccident. Dans ce document, l'effet domino pouvant se produire sur des sites industriels est analysé au travers des projections produites par l'accident initial. Une approche probabiliste globale est ainsi développée dans laquelle le calcul de la probabilité d'occurrence du phénomène requiert le passage par quatre étapes : - Analyse des termes sources : les projectiles générés par l'explosion d'un réservoir ont différentes caractéristiques, à savoir le nombre de projectiles, la forme, la masse, la vitesse de départ et les angles de départ. Toutes ces grandeurs sont modélisées par des variables aléatoires. A l'aide du principe du maximum d'entropie et des données existantes, des distributions probabilistes sont développées pour toutes ces variables. On se limite, cependant, au cas de l'explosion d'un réservoir cylindrique ou sphérique. - Analyse de l'impact ou analyse du mouvement : la trajectoire d'un projectile (ou fragment de structure), en fonction de ses caractéristiques de départ, est décrite par une combinaison des effets d'inertie, de gravitation et d'aérodynamique. Une approche simplifiée faisant l'hypothèse de constance des coefficients aérodynamiques permet d'identifier analytiquement la trajectoire du projectile tandis qu'une solution numérique est obtenue par une approche complète où toutes les valeurs de ces coefficients sont prises en compte.Les mouvements de translation et de rotation sont également étudiés. A l'aide de l'analyse complète et des conditions d'impact, la probabilité d'impact est déterminée. L'étude est restreinte à des projectiles en forme de fond de réservoir, fond oblong de réservoir et plaque. Les formes des cibles de l'étude sont restreintes au cas ellipsoïdal, cylindrique et cubique. - Analyse de l'état des cibles impactées : dans un premier temps, des modèles simplifiés d'impact sont utilisés afin d'étudier l'interaction mécanique entre les projectiles et les réservoirs impactés. Un modèle mécanique complet comprenant une loi de comportement élasto-plastique et un modèle de rupture est également proposé. Ce modèle est ensuite implémenté dans un code de calcul sans maillage de type SPH, i.e. Smoothed Particle Hydrodynamics. Afin d'estimer la probabilité de rupture des réservoirs impactés, les modèles simplifiés sont mis en œuvre, ce qui permet de réduire le coût de calcul. - Occurrence du sur-accident : selon l'état mécanique résiduel de la cible et son état physique (conditions thermodynamiques, niveau de remplissage, etc.), l'impact de projectiles peut conduire à la poursuite du pnénomène. Genomic differentiation of big bluestem (Andropogon gerardii) along the Great Plains’ environmental gradient

Johnson / Big bluestem (Andropogon gerardii Vitman) is an ecologically dominant grass of the North American grasslands with precipitation-dependent productivity. However, climatic predictions for big bluestem’s dominant range in the Great Plains include increased periods of drought. The main objectives of this research were to determine the extent of neutral and non-neutral genetic differentiation and diversity among putative big bluestem ecotypes using amplified fragment length polymorphism (AFLP) markers. This is the first study of both neutral and non-neutral genetic diversity of big bluestem which also includes source populations of well-described ecotypes studied in reciprocal common gardens. A total of 378 plants were genotyped from 11 source prairies, originating from one of three ecoregions (Central Kansas, Eastern Kansas, and Illinois). Using two AFLP primer sets, 387 polymorphic markers (error rate 9.18%) were found. Un-rooted neighbor joining tree and principle-component analyses showed continuous genetic differentiation between Kansas and Illinois putative ecotypes, with genetic overlap occurring between Kansas ecotypes. Analysis of molecular variance showed high diversity within-prairie sites (80%) relative to across-prairies (11%), and across- ecoregions (9%) (p<0.001). Within-prairie genetic diversity levels were similar among ecoregions (84-92%), with the highest genetic variation maintained in Illinois prairies (92%). Population structure analyses supported K=6 genetic clusters across the environmental gradient, with Kansas prairies belonging to three main genetic groups, and Illinois prairies having largely divergent allele frequencies from Kansas prairies. Interestingly, BAYESCAN analysis of the three putative ecotypes identified eight F[subscript]ST-outlier AFLP loci under potential diversifying selection. Frequency patterns of loci under diversifying selection were further linked to geo-environmental descriptors including precipitation, temperature severity, diurnal temperature variation, prairie location, and elevation. The observed allele frequency divergence between Kansas and Illinois ecotypes suggests tallgrass restorations should consider possible maladaptation of non-local ecotypes and genetic swamping. However, high within-prairie genetic variation may help individual big bluestem populations withstand climatic variability.

Determination of binary fission-fragment yields in the reaction 251Cf(nth, f) and Verification of nuclear reaction theory predictions of fission-fragment distributions in the reaction 238U(n, f)

Neutron-induced fission has been studied at different excitation energies of the compound nucleus by measurements on the two fissioning systems, 252Cf* and 239U*. For the first time, the light fission fragment yields from the reaction 251Cf(nth, f) have been measured with high resolution. This experiment was performed with the recoil mass spectrometer LOHENGRIN at ILL in Grenoble, France. When the results from this work, where the compound nucleus is at thermal excitation, are compared to the spontaneous fission of 252Cf, enhanced emission yields as well as an increased mean kinetic energy is observed around A = 115. This suggests the existence of an additional super-deformed fission mode in 252Cf. The reaction 238U(n, f) was studied using the 2E-technique with a double Frisch grid ionization chamber. Fission fragment mass, energy and angular distributions were determined for incident neutron energies between 0.9 and 2.0 MeV. The experiments were performed at the Van de Graaff accelerator of IRMM in Geel, Belgium. This is the first measurement of the mass distribution for incident neutron energies around 0.9 MeV. The motivation for studying 238U(n, f) was to verify theoretical predictions of the mass distribution at the vibrational resonance in the fission cross section at 0.9 MeV. However, the predicted changes in fission fragment distributions could not be confirmed. A precise modelling of the fission process for the minor actinides becomes very important for future generation IV and accelerator driven nuclear reactors. Since fission fragment distributions depend on the excitation of the fissioning system, so does the number of delayed neutrons, which are one of the safety parameters in a reactor.

Comunidade de mamíferos terrestres de médio e grande porte da Escola do Meio Ambiente em Botucatu, São Paulo

Orientador: Elizabeth Moreira dos Santos Schmidt / Resumo: O Brasil possui a maior diversidade de espécies do mundo, grande parte está nos biomas Mata Atlântica e Cerrado, considerados hotspots para conservação. Os mamíferos de médio e grande porte, grupo altamente afetados pela antropização, desempenham papéis importantes na dinâmica de ecossistemas, influenciam no ciclo do carbono, agem como predadores e dispersores de sementes, polinizadores, predadores de topo de cadeia alimentar. O presente estudo teve como objetivo realizar o levantamento da mastofauna terrestre de médio e grande porte presente na área da Escola do Meio Ambiente (EMA) no município de Botucatu, São Paulo e gerar dados de riqueza, ocorrência e frequência de ocorrência de espécies encontradas no local. A área da escola é de aproximadamente 12 hectares, apresentando cobertura vegetal de remanescente de Cerrado e fragmento com fitofisionomias de Mata Atlântica. As metodologias utilizadas foram: transectos, parcelas de areia e, como método complementar, o uso de armadilha fotográfica por dois meses. Adicionalmente, entrevista com funcionários para obtenção de dados. Foram registradas vinte espécies de mamíferos. A ordem carnívora foi a que apresentou mais espécies, oito no total. A espécie de maior ocorrência foi Hydrochoerus hydrochaeris, seguida por Canis lupus familiaris. Nas entrevistas, os funcionários citaram duas espécies que não foram registradas no estudo, Cavia aperea, mamífero de pequeno porte, e Lontra longicaudis. A riqueza constatada pelo estudo enfatiz... Medium and large mammals are one of the groups most affected by anthropization, play important roles in the nature of ecosystems, influence the carbon cycle, act as seed predators and seed dispersers, pollinators, top food chain predators. The present study had the objective of surveying the medium and large size terrestrial mastofauna present in Environmental School (EMA) area in the city of Botucatu, São Paulo, and to generate data on the richness, occurrence and frequency of occurrence of species found on the spot. The school area is approximately 12 hectares, presenting vegetation cover of Brazilian Savana remnant and fragment with phytophysiognomies of Atlantic Forest. Using the methodologies used were trawling for traces, sand plots and as a complementary method the use of photographic trap for two months, in addition, interview with employees to obtain additional data. Twenty species of mammals were recorded. The carnivorous order was the one that presented the most species, eight in total. The species with the highest occurrence was Hydrochoerus hydrochaeris, followed by Canis lupus familiaris. In the interviews, officials cited two species that were not recorded in the study, Cavia aperea, a small mammal, and Lontra longicaudis. Partindo das relações estabelecidas por Novalis e Schlegel - principais representantes do Primeiro Romantismo Alemão - entre teoria, pensamento filosófico e poética, bem como da elaboração consciente de uma estética do fragmento, buscaremos compreender como a idéia do fragmentário engendra, na modernidade e no pós-modernismo, uma problemática da crise: das possibilidades de representação do real, de criação artística, de alcance e fixação da verdade, do discurso como instância ou como meio através do qual o mundo, o real e os indivíduos são compreendidos, tomados, discutidos e representados. Busca-se entender a crise da representação e a forma como esta se vincula, no plano da criação artística, a outras noções igualmente importantes discutidas ao longo do século passado: o estilhaçamento e a crise da noção de sujeito, a legitimidade dos discursos, a busca incessante pela originalidade radical como única forma de surgimento e manifestação do novo no domínio estético, a ruptura e o choque entre certo relativismo do qual se acusa a contemporaneidade em relação a uma alta tradição, uma alta cultura, que teriam produzido os últimos grandes modelos de discursos teleológicos, com seus conceitos de verdade absoluta, de revolução possível, de transformação plena da ordem estabelecida. Desse modo, além dos fragmentos literários dos românticos, o trabalho em questão passa pela análise dos romances Nadja, de André Breton e Vício... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The project presented now tries to situate - historically, critically and theoretically - some force lines of the philosophical, aesthetic and artistic thought developed since the Early German Romanticism, which during centuries were rejected, denied, reviewed or reconfigured, originating central issues within the critical and theoretical discourse of the modernism and postmodernism. From the relation established by Novalis and Schlegel - the main representatives of the Early German Romanticism - between theory, philosophical and poetic thought, besides the conscious elaboration of an aesthetic of the fragment, we seek out to understand how the idea of the fragment brings forth in modernism and postmodernism a problem of crisis: the possibilities of the representation of real; the artistic creation; the acquisition and fixation of the truth; the discourse as instance or as a way in which the world, the real and the individuals are understood, taken, discussed and represented. We seek to comprehend the representation crisis and the way it is linked, on the artistic creation area, to other notions equally important discussed throughout last century: the chipping and the notion of subject crisis, the legitimacy of discourses, the incessant search or the radical originality as the only way of appearance and manifestation of the new aesthetic domain, the disruption and the impact between a kind of relativism in which the contemporary is accused with regard to a high tradition, a high culture, which would have produced the last important models of teleological discourses, bringing its concepts of absolute truth, possible revolution, full transformation of the established order. Esta pesquisa discute a obra de Jorge Guinle (1947-1987) e o diálogo que o artista travou com a pintura moderna e contemporânea brasileira e internacional. Argumenta que a pintura de Jorge Guinle, embora concentrada apenas nos últimos sete anos da vida do artista, estabeleceu um diálogo profundo com a arte do século XX, sem, com isso, deixar de se lançar a uma indagação de seu próprio tempo. / This research discusses the work of Jorge Guinle (1947-1987) and the dialogue the artist sustained with the Brazilian and international modern and contemporary painting. It argues that Jorge Guinle\'s painting, although concentrated only in the last seven years of the artist\'s life, established a profound dialogue with the art of the twentieth century, without failing, however, to undertake an investigation of its own time.
