In the Southern Hemisphere, glaciations were milder than in the Northern Hemisphere, however, there were temperature reductions, and the climate became drier. Thus, climate-related modifications occurred in the Atlantic forest vegetation, which contracted its distribution being substituted towards other types, such as Cerrado and Caatinga . The original Atlantic forest vegetation became restricted to moist locations with milder temperatures, called refuges, which sheltered most part of the biodiversity at this time. It is known that such paleoclimatic changes affected the population dynamics of many species, especially in the Northern Hemisphere. However, it is still not clear how much these impacts influenced the species of the Neotropical region. The two species of this study, Drosophila maculifrons and Drosophila griseolineata, belong to the guaramunu group of Drosophila, and are considered sister-species that diverged about four million years ago. However, disregarding their close phylogenetic relationships, they present some distinct ecological patterns, the first species being more generalist, and the second more restricted to forest environments. Due to this ecological heterogeneity, these two species are potential indicators of the genetic consequences caused by the climatic fluctuations of the Quaternary, especially in face of a comparative perspective. The aims of this study were to evaluate the intraspecific diversity of different D. maculifrons and D. griseolineata populations, analyze the structure of individuals and populations in these two species of the guaramunu group and identify the ecological and evolutionary forces that modeled their distribution in Southern/Southeastern Brazil. In order to do so, 114 individuals were collected along the South, Southeast and Center-west regions of Brazil and Medellin, Colombia. Modeling analysis was performed, together with phylogenetic and phylogeographic analyses, with the last based in sequences of COI (Cytochrome Oxidase Subunit I) and COII (Cytochrome Oxidase Subunit II) genes. In general, the results that inferred the distribution of the most suitable habitat for each species indicate that the two species occur in sympatry at several points, although D. maculifrons is more widely distributed than D. griseolineata, at least in the brazilian territory. According to our phylogeographic analysis, D. maculifrons presents low levels of diversity and structure for mtDNA, which could be explained by a recent populational expansion event, dated for about 20 to 30 thousand years ago. On the other hand, D. griseolineata shows moderate levels of diversity and population structure, and its populations seem to have remained stables along time, showing a pattern of isolation by distance. So, it is interesting to evaluate the ecological and/or evolutionary factors which are responsible for all this difference, and this work represents a first step towards this understanding. / O período do Quaternário foi marcado por alterações consideráveis no clima e vegetação. No Hemisfério Sul, as glaciações foram mais amenas, entretanto, houve a redução da temperatura e o clima tornou-se mais seco. Dessa forma, ocorreram contrações na distribuição da Mata Atlântica e sua substituição por outros tipos, condizentes ao clima, como o Cerrado e a Caatinga. Assim, a vegetação de Mata Atlântica ficou restrita a locais úmidos e com temperaturas mais amenas, conhecidos como refúgios, os quais abrigaram grande parte da biodiversidade neste período. Sabe-se que essas alterações paleoclimáticas influenciaram a dinâmica populacional de muitas espécies, especialmente no Hemisfério Norte, porém ainda não está claro o quanto esses impactos influenciaram as espécies de distribuição Neotropical. As duas espécies em questão, Drosophila maculifrons e D. griseolineata, pertencem ao grupo guaramunu de Drosophila e são consideradas espécies irmãs, que divergiram aproximadamente há 4 milhões de anos. Porém, apesar do grau de parentesco, elas apresentam alguns padrões ecológicos distintos, sendo a primeira mais generalista e a segunda mais restrita a ambientes florestais. Devido a essa heterogeneidade ecológica, essas duas espécies são potenciais indicadoras das conseqüências genéticas ocasionadas pelas flutuações climáticas do Quaternário, principalmente em face de uma perspectiva comparativa. Os objetivos deste trabalho foram avaliar a diversidade intra-específica de diferentes populações de D. maculifrons e D. griseolineata, analisar a estruturação de indivíduos e populações nestas duas espécies do grupo guaramunu e inferir as forças ecológicas e evolutivas que modelaram sua distribuição ao longo do sul e sudeste brasileiros. Para isso, nossa amostragem conta com 114 indivíduos distribuídos ao longo das regiões Sul, Sudeste, Centro-Oeste do Brasil e Medellin na Colômbia. Foram realizadas análises de modelagem, bem como análises filogenéticas e filogeográficas, sendo que essas duas últimas basearam-se nas sequências dos genes COI (Citocromo Oxidase Subunidade I) e COII (Citocromo Oxidase Subunidade II). De uma forma geral, os resultados que inferiram a distribuição dos habitats mais adequados para cada espécie indicam que as duas espécies apresentam diversos pontos de simpatria, embora D. maculifrons seja mais amplamente distribuída em território brasileiro. De acordo com as análises filogeográficas, D. maculifrons apresenta baixos níveis de diversidade e estruturação em nível de DNA mitocondrial, o que pode ser explicado por um evento de expansão populacional recente, datado para aproximadamente 20 a 30 mil anos. Por outro lado, D. griseolineata apresenta níveis moderados de diversidade e estruturação populacional e suas populações parecem ter se mantido estáveis ao longo do tempo, apresentando um padrão de isolamento por distância. É, pois, interessante avaliar os fatores ecológicos e/ou evolutivos responsáveis por toda essa diferença, e esta dissertação representa um primeiro passo rumo a esse entendimento.

As análises indicaram que os indivíduos dos EUA e do Brasil não diferem entre si a ponto de inferir que compõem um complexos de espécies. Neste caso, a ampla distância geográfica não representou uma elevada divergência genética e morfológica.

Estudo do complexo de espécies Scinax hayii (Barbour, 1909) e Scinax dolloi (Werner, 1903) (Anura, Hylidae) ao longo da Mata Atlântica, Brasil /

Orientador: Itamar Alves Martins / Resumo: A escassez de informações, juntamente com a morfologia conservada, dificulta a caracterização e identificação de complexo de espécies do gênero Scinax como em Scinax hayii e Scinax dolloi. Este estudo utilizou a taxonomia integrativa, usando como base a morfologia, morfometria, bioacústica e análises moleculares do gene COI (DNA Barcoding) para investigar e caracterizar variações populacionais entre populações de S. hayii e S. dolloi amostradas na região sudeste do Brasil, nos domínios da Mata Atlântica, buscando elucidar possíveis problemas taxonômicos e determinar a distribuição de cada espécie. Análises do canto de anúncio em populações de S. hayii revela uma variação gradativa entre as populações da Serra do Mar e litoral tendo as populações do litoral paulista, canto distinto dos topótipos e de outras populações da Serra do Mar. As análises morfológicas e morfométricas aliadas ao gene COI, revelam que S. hayii possui estruturação geográfica e, apesar de possuir populações polimórficas, esse gene ao longo da distribuição de S. hayii tem características conservadas. Semelhanças nos parâmetros bioacústicos e moleculares mostram que a população de Castelo no Espírito Santo é similar a topótipos de S. dolloi. Diferenças no canto de anúncio, comprimento rostro-cloacal e no gene COI sugerem que populações localizadas no bloco sul da Serra da Mantiqueira representam uma nova espécie. / Abstract: Scarce information combined with conserved morphology hinder the characterization and identification of Scinax complex species, such as S. hayii and S. dolloi. We used integrative taxonomy, based on morphology, morphometry, bioacoustics and molecular analyzes (COI gene) in order to investigate and characterize population variations between S. hayii and S. dolloi populations, sampled in Southeastern Brazil, in the Forest Atlantic domains, seeking to elucidate possible taxonomic problems and also to determine the distribution of both species. Advertisement call analysis in S. hayii reveals a gradual variation between Serra do Mar mountais and litoral populations. Coastal populations have different calls compared to topotypes and other populations distributed in the mountains. Morphometric and morpholical analyzes combined with COI gene reveal that S. hayii has a geographic structure, even having polymorphic populations, COI gene throughout the distribution of S. hayii shows conserved characteristics. Similarities in bioacoustic and molecular parameters show that Castelo’s population in Espírito Santo state is similar to topotypes of S. dolloi. Differences in advertisement call, snout vent lenght and COI gene suggest that populations in the Southern Serra da Mantiqueira represent a new specie. / Doutor

Drosophila meridionalis é uma espécie endêmica da América do Sul, sendo amplamente distribuída na Costa Atlântica do Brasil. Com o objetivo de elaborar uma hipótese filogeográfica para esta espécie foram obtidas sequências do gene nuclear period e do gene mitocondrial COI. Foram calculados os índices de diversidade nucleotídica e realizados os testes: AMOVA, testes de neutralidade, a Mismatch Distribution, Bayesian Skyline Plot, NCPA. Foram obtidas três redes pelo gene COI, denominadas A (populações do interior), B (populações do litoral sul) e C (populações do litoral sudeste e oriental) e uma única rede obtida para o gene period, esta rede divide as populações em dois grupos sendo o primeiro congruente com a rede A e o segundo compreendendo as redes B e C, do gene COI. A AMOVA mostrou uma estruturação alta e significativa entre as populações do interior e o litoral para os dois genes (ct=0,72 gene COI; ct=0,70 gene period), que pode ser explicada pela presença de barreiras geográficas, como a Serra do Mar. Eventos de expansão populacional e de fluxo gênico restrito com isolamento por distância foram detectados nas populações do litoral e o interior respectivamente. A expansão da área de ocorrência de D. meridionalis provavelmente teve inicio com as populações do litoral do Rio Grande de Sul, em direção ao litoral de Santa Catarina com posterior colonização a longa distância dos estados de São Paulo, Rio de Janeiro e Bahia. Migrações assincrônicas de indivíduos de populações litorâneas de São Paulo e Santa Catarina provavelmente colonizaram o interior de São Paulo, e a partir destas populações, se iniciara uma expansão populacional em direção ao sul pelo interior, colonizando o Paraná e Rio Grande do Sul. A análise bayesiana (MCCT) indicou que o tempo do ancestral comum mais recente (TMRCA) para todos os haplótipos de D. meridionalis é de 81.700 anos atrás, data que marca a separação das populações do interior e do litoral aproximadamente no final do Pleistoceno. Eventos similares têm sido sugeridos para explicar a distribuição geográfica de espécies do cluster D. buzzatii, que ocorrem em simpatria em grande parte com populações de D. meridionalis. As Rotas de Dispersão de Drosophila buzzatii na América do Sul / The Dispersion Routes of Drosophila buzzatii in South America

Drosophila buzzatii é uma espécie cactófila associada a diferentes espécies de cactos e distribuída nos diferentes Domínios fitogeográficos da América do Sul. Baseado na diversidade de inversões cromossômicas e densidade populacional, o Chaco foi considerado por alguns autores como o centro de origem da espécie. Entretanto, trabalhos recentes, utilizando aloenzimas e DNA mitocondrial, apontaram para uma possível origem na Caatinga. Os objetivos deste trabalho foram delinear rotas de dispersão da espécie para explicar sua distribuição atual e possível distribuição durante o último período glacial, na América do Sul. Foram obtidas seqüências de 714 pb da COI do mtDNA de 132 indivíduos em 44 localidades, gerando 36 haplótipos. Foram calculados os índices de diversidade nucleotídica, o teste AMOVA, testes de neutralidade, a Mismatch Distribution, Baesyan Skyline Plot, NCPA e o sentido dos movimentos migratórios (Migrate N) a fim de determinar parte da história evolutiva da espécie. As diversidades nucleotídicas encontradas por Domínio foram de 0,0030 Caatinga; 0,0019 - Mata Atlântica; 0,0020 Cerrado; 0,0011 - Pampas e 0,0004 - Chaco. A AMOVA mostrou que 68,33% da variação é intra-populacional e que uma porção significativa da variação é devido a diferenças inter-regionais (ct = 0,07124 p = 0,00196). Os testes de Neutralidade (D de Tajima = -2,4150, p = 0,0317 e Fs de Fu = -28,6719, p = 0,00001), a forma em estrela da rede e haplótipos, e a Mismatch Distribution confirmam um evento de expansão populacional estimado em aproximadamente 494.257,3 anos atrás, segundo modelo de Rogers e Harpending (1992). Entretanto, a Baesyan Skiline Plot demonstrou que esse movimento de expansão parece ser mais antigo, cerca de 550.000 650.000 anos atrás. A NCPA demonstrou que há fluxo gênico restrito com isolamento por distância, confirmado pelo teste de Mantel e alguma dispersão a longa distância em alguns dos clados analisados. O resultado do programa Migrate N indicou um padrão complexo migratório entre os domínios, porém um padrão norte/sul pôde ser verificado. A estruturação genética pode ser explicada devido à grande área de distribuição da espécie, gerando isolamento por distância e pela presença de barreiras geográficas e climáticas (entre o Cerrado e a Caatinga) e no estado do Rio de Janeiro e Espírito Santo (ao longo da Mata Atlântica) onde há pouco ou nenhum indivíduo da espécie. Os eventos de expansão ocorreram no Quaternário durante o período glacial conhecido como Ilinioian e suas subdivisões. A partir dos resultados deste trabalho foi possível traçar diversas rotas de migração possíveis entre os domínios utilizados, sendo que o movimento mais antigo partiu da Caatinga o que vai contra a hipótese de que o Centro de Origem seja o Chaco. / The fruit-fly Drosophila buzzatii is a cactophilic species in association with cactus species distributed along the Phytogeographic Domains of Caatinga, Cerrado, Atlantic Forest, Pampas and Chaco. Based in the diversity of chromosomal inversion and populational density of the species, the Chaco Domain was considered the Center of Origin of the D. buzzatii. However, recent works, using allozymes and DNAmt, showed a possible origin of the D. buzzatii in the Caatinga. The objectives of this work were trace historical dispersion routes of D. buzzatii, current and ancient areas of distribution in the South America. We obtained DNA sequences in 132 samples in 44 localities with 714 bp length from the COI mtDNA gene, generating 36 haplotypes. The diversity indexes, AMOVA, neutrality tests, Mismatch Distribution, Baesyan Skyline Plot, NCPA and sense of migration movements was calculated, to describe part of the evolutionary history of the species. The nucleotide diversity was 0,0030 - Caatinga, 0,0019 - Mata Atlântica, 0,0020 - Cerrado, 0,0011 - Pampas and 0,0004 - Chaco. The AMOVA results, grouped by Domain showed that 68,33% of the variation is intra-population and a significant portion of the variation is due to inter-regional differences (ct = 0,07124 p = 0,00196). The Neutrality tests (Tajimas D = -2,4150, p = 0,0317 and Fus Fs = -28,6719, p = 0,00001), the star-shape of the haplotype network, and Mismatch Distribution showed population expansion signs, estimated in 494.257,3 ybp, according Rogers and Harpending model (1992). However, the BSP showed that the movement is ancient, estimated in 550.000 650.000 ybp. The NCPA showed restricted gene flow with isolation by distance, confirmed by the Mantel Test and some long distance dispersion. The results of the program Migrate N showed a complex pattern of migration between the domains, but a north/south pattern could be identified. The genetic structure can be explained to the widespread distribution of the species, that could generate isolation by distance and by the presence of geographic and climatic barriers (between Cerrado and Caatinga domains) and in the States of Rio de Janeiro and Espírito Santo (along the Atlantic Forest) when there is none or few individuals of the species D. buzzatii. The expansion movements occurred in the Quaternary Period during glaciations events in the Illinoian and their subdivisions, due to the decrease of the global moisture that generated favorable conditions to the expansion of the dry vegetation and associated species. Based on the results of this work it was possible to delineate many migration routes between the phytogeographic domains, and the more ancient movement started in the Caatinga and this result not support the hypothesis that Chaco was the Center of Origin from D. buzzatii.

As Rotas de Dispersão de Drosophila buzzatii na América do Sul / The Dispersion Routes of Drosophila buzzatii in South America

Drosophila buzzatii é uma espécie cactófila associada a diferentes espécies de cactos e distribuída nos diferentes Domínios fitogeográficos da América do Sul. Baseado na diversidade de inversões cromossômicas e densidade populacional, o Chaco foi considerado por alguns autores como o centro de origem da espécie. Entretanto, trabalhos recentes, utilizando aloenzimas e DNA mitocondrial, apontaram para uma possível origem na Caatinga. Os objetivos deste trabalho foram delinear rotas de dispersão da espécie para explicar sua distribuição atual e possível distribuição durante o último período glacial, na América do Sul. Foram obtidas seqüências de 714 pb da COI do mtDNA de 132 indivíduos em 44 localidades, gerando 36 haplótipos. Foram calculados os índices de diversidade nucleotídica, o teste AMOVA, testes de neutralidade, a Mismatch Distribution, Baesyan Skyline Plot, NCPA e o sentido dos movimentos migratórios (Migrate N) a fim de determinar parte da história evolutiva da espécie. As diversidades nucleotídicas encontradas por Domínio foram de 0,0030 Caatinga; 0,0019 - Mata Atlântica; 0,0020 Cerrado; 0,0011 - Pampas e 0,0004 - Chaco. A AMOVA mostrou que 68,33% da variação é intra-populacional e que uma porção significativa da variação é devido a diferenças inter-regionais (ct = 0,07124 p = 0,00196). Os testes de Neutralidade (D de Tajima = -2,4150, p = 0,0317 e Fs de Fu = -28,6719, p = 0,00001), a forma em estrela da rede e haplótipos, e a Mismatch Distribution confirmam um evento de expansão populacional estimado em aproximadamente 494.257,3 anos atrás, segundo modelo de Rogers e Harpending (1992). Entretanto, a Baesyan Skiline Plot demonstrou que esse movimento de expansão parece ser mais antigo, cerca de 550.000 650.000 anos atrás. A NCPA demonstrou que há fluxo gênico restrito com isolamento por distância, confirmado pelo teste de Mantel e alguma dispersão a longa distância em alguns dos clados analisados. O resultado do programa Migrate N indicou um padrão complexo migratório entre os domínios, porém um padrão norte/sul pôde ser verificado. A estruturação genética pode ser explicada devido à grande área de distribuição da espécie, gerando isolamento por distância e pela presença de barreiras geográficas e climáticas (entre o Cerrado e a Caatinga) e no estado do Rio de Janeiro e Espírito Santo (ao longo da Mata Atlântica) onde há pouco ou nenhum indivíduo da espécie. Os eventos de expansão ocorreram no Quaternário durante o período glacial conhecido como Ilinioian e suas subdivisões. A partir dos resultados deste trabalho foi possível traçar diversas rotas de migração possíveis entre os domínios utilizados, sendo que o movimento mais antigo partiu da Caatinga o que vai contra a hipótese de que o Centro de Origem seja o Chaco. / The fruit-fly Drosophila buzzatii is a cactophilic species in association with cactus species distributed along the Phytogeographic Domains of Caatinga, Cerrado, Atlantic Forest, Pampas and Chaco. Based in the diversity of chromosomal inversion and populational density of the species, the Chaco Domain was considered the Center of Origin of the D. buzzatii. However, recent works, using allozymes and DNAmt, showed a possible origin of the D. buzzatii in the Caatinga. The objectives of this work were trace historical dispersion routes of D. buzzatii, current and ancient areas of distribution in the South America. We obtained DNA sequences in 132 samples in 44 localities with 714 bp length from the COI mtDNA gene, generating 36 haplotypes. The diversity indexes, AMOVA, neutrality tests, Mismatch Distribution, Baesyan Skyline Plot, NCPA and sense of migration movements was calculated, to describe part of the evolutionary history of the species. The nucleotide diversity was 0,0030 - Caatinga, 0,0019 - Mata Atlântica, 0,0020 - Cerrado, 0,0011 - Pampas and 0,0004 - Chaco. The AMOVA results, grouped by Domain showed that 68,33% of the variation is intra-population and a significant portion of the variation is due to inter-regional differences (ct = 0,07124 p = 0,00196). The Neutrality tests (Tajimas D = -2,4150, p = 0,0317 and Fus Fs = -28,6719, p = 0,00001), the star-shape of the haplotype network, and Mismatch Distribution showed population expansion signs, estimated in 494.257,3 ybp, according Rogers and Harpending model (1992). However, the BSP showed that the movement is ancient, estimated in 550.000 650.000 ybp. The NCPA showed restricted gene flow with isolation by distance, confirmed by the Mantel Test and some long distance dispersion. The results of the program Migrate N showed a complex pattern of migration between the domains, but a north/south pattern could be identified. The genetic structure can be explained to the widespread distribution of the species, that could generate isolation by distance and by the presence of geographic and climatic barriers (between Cerrado and Caatinga domains) and in the States of Rio de Janeiro and Espírito Santo (along the Atlantic Forest) when there is none or few individuals of the species D. buzzatii. Ferramentas auxiliares na identificação de espécies de abelhas Meliponini, com ênfase no gênero Schwarziana (Lepeletier, 1836) / Auxiliar tools for Meliponini bees identification, emphasizing the genus Schwarziana

As abelhas sem ferrão estão entre os animais mais importantes para o equilíbrio do meio ambiente, isso devido a fatores como sua enorme diversidade e principalmente por serem importantes polinizadores tanto de ecossistemas naturais como de agroecossistemas. A necessidade de identificação dos animais amostrados por especialistas é fundamental para estudos ecológicos. Neste trabalho foram feitas análises interespecíficas e intra-populacionais sobre os indivíduos do gênero Schwarziana a fim de aperfeiçoar a utilização da técnica de morfometria geométrica, que nos permite identificações baseadas apenas nos padrões de venação das asas desses insetos. Não obstante, foram realizadas também análises de sequenciamento de dois genes do DNA mitocondrial, o COI e o 16S. O gênero estudado foi Schwarziana, com duas espécies válidas, S. quadripunctata e S. mourei. Nas análises morfométricas, os testes realizados por indivíduos os testes de validação cruzada identificaram de forma correta 70% das amostras de um total de 10 localidades diferentes. Esta acurácia aumenta ainda mais à medida que novos grupos são formados, alcançando próximo de 85% quando separadas por regiões. Em todos os ensaios realizados com a morfometria geométrica (partial warps e coordenadas alinhadas) atingiu-se uma taxa de 100% de identificação correta entre as duas espécies e nos ensaios feitos com as médias das colônias esta taxa também foi atingida para a identificação de todas as populações amostradas. Os testes feitos com os partial warps mostraram-se mais eficazes em relação às coordenadas alinhadas, já que nestes últimos a tolerância mínima para a separação dos grupos não foi atingida em alguns ensaios. As análises moleculares apontaram 53 sítios polimórficos para o gene COI, com um índice de diversidade nucleotídica de 0,02180 e de diversidade haplotípica de 0,8854, separando as amostras em 9 haplótipos, porém a rede de haplótipos não foi suficientemente conclusiva. A diferenciação genética total medida pelo parâmetro Fst somente para as populações de S. quadripunctata foi de 0.9453 (P<0,05). Já para o gene 16S foram encontrados 14 sítios polimórfico e um índice de diversidade nucleotídica de 0,00649 e de diversidade haplotípica de 0,8419, com 7 haplótipos gerados. O parâmetro Fst para todas amostras foi de 0.9552 (P<0,05) e somente para as de S. quadripunctata foi de 0.8736 (P<0,05). Para ambos os genes os testes Fst par-a-par mostraram uma maior variação entre as populações dos estados, mostrando que estas populações estão estruturadas. Os testes de Mantel correlacionaram positivamente os dados morfométricos, geográficos e moleculares. Das duas metodologias aplicadas em nossa pesquisa, podemos afirmar que a morfometria mostrou-se extremamente eficiente na diferenciação das populações amostradas. As análises moleculares indicaram que estas populações estão estruturadas mesmo analisando poucas amostras. No entanto ao unirmos estas duas metodologias, combinamos a simplicidade e a rapidez da morfometria geométrica com a capacidade sempre inovadora de estudos moleculares, obtendo desta forma ferramentas eficazes para acessar a biodiversidade em Meliponini, inclusive para rastrear geograficamente espécimes a partir de estudos com indivíduos de sua área de distibuição natural. / Stingless bees are among the most important animals to the environmental balance, that due to their diversity and mainly because they are important pollinators of both natural and agro-ecosystems. The need for identification of sampled animals by experts is essential for ecological studies. In this work, intra and interspecific population analysis was performed in order to improve the technique of geometric morphometry, which allows us to access this biodiversity through identifications based on patterns of wing venation of these insects. We also sequenced two mitochondrial genes, the COI and 16S. Schwarziana Lepeletier 1836 was the gender studied with two valid species, S. quadripunctata and S. mourei. In morphometric analysis, cross-validation tests carried out by individuals correctly identified 70% of samples from a total of 10 different locations. This accuracy increases further as new groups are formed, according to geographic proximity, reaching around 85% when separated by regions. In all tests with geometric morphometry (partial warps and aligned coordinates) reached a rate of 100% correct identification between the two species and the tests made with the averages of the colonies that rate achieved the perfect identification of all populations sampled. Partial warps tests were more effective in relation to aligned coordinates ones, as these last a minimum tolerance for the separation of the groups was not achieved in some tests. Molecular analysis showed 53 polymorphic sites for the COI gene, with nucleotide diversity of 0.02180 and haplotype diversity of 0.8854, separating the samples into 9 haplotypes, but their network of haplotypes was not sufficiently conclusive. The total genetic differentiation measured by Fst parameter only for the populations of S. quadripunctata was 0.9453 (P <0.05). As for the gene 16S, we found 14 polymorphic sites and a nucleotide diversity of 0.00649 and haplotype diversity of 0.8419, with seven haplotypes generated. The parameter Fst for all samples was 0.9552 (P <0.05) and only for S.quadripunctata was 0.8736 (P <0.05). For both genes Fst pairwise tests showed greater variation among populations of the states, showing these groups as a genetic structure. Mantel tests were positively correlated for morphometric, geographical and molecular data. Of the two methodologies in our research, we can affirm that the morphometry proved to be extremely efficient in the differentiation of populations. Molecular analysis showed that populations are consistent, even that a low sample size is available. However combining these two methodologies, we have joined the simplicity and speed of geometric morphometric with the always innovative ability of molecular studies, thus achieving effective tools for accessing biodiversity in Meliponini, including a geographically trace for specimens by studying individuals from their natural range.

Populações de Chrysoperla externa apresentam ampla distribuição geográfica, abrangendo desde o sul dos Estados Unidos até o sul da América do Sul, ocorrendo em diferentes ambientes. Contudo, há relativamente poucos estudos buscando compreender a estrutura genética de agentes de controle biológico, especialmente insetos predadores. Desta forma, os principais objetivos deste trabalho foram caracterizar geneticamente as populações de C. externa por meio de sequências do gene mitocondrial COI e compreender sua estrutura populacional nos municípios amostrados no Estado de São Paulo. Para tanto, indivíduos adultos foram coletados em pomares de citros, e da região torácica foi extraído o DNA total. O gene COI foi amplificado por meio da técnica de PCR e as amostras foram purificadas e sequenciadas. As populações de C. externa analisadas apresentaram elevada diversidade genética, bem distribuída entre os municípios amostrados. Esta homogeneização pode ser decorrência de fluxo gênico, ação antrópica, correntes de ar, proximidade das fazendas com matas nativas e elevado potencial reprodutivo de C. externa. A partir dos resultados obtidos é possível inferir que o agroecossistema, por ser um ambiente homogêneo, esteja contribuindo para a perda de estruturação que havia entre estas populações quando elas viviam em ecossistemas nativos, sendo assim, é fundamental que as populações de C. externa sejam estudas neste ambiente, para que possam ser compreendidas em seu habitat de origem e sem a influência da ação antrópica / The green lacewings are insects with great potential for use in programs of biological control of agricultural pests. Populations of Chrysoperla externa are widely distributed geographically, being found from the southern United States until the southern South America, occurring in different environments. However, there are relatively few studies trying to understand the genetic structure of biological control agents, especially predatory insects. Thus, the main objectives of this work were to characterize genetically the populations of C. externa through COI mitochondrial gene sequences and to understand its population structure in sampled municipalities in the State of São Paulo. For this purpose, adult individuals were collected in citrus orchards, and from its torax were extracted the total DNA. The COI gene was amplified by PCR and the samples were purified and sequenced. The populations showed high genetic diversity, well distributed among the municipalities. This homogenization may be due to gene flow, human action, action of winds, proximity of farms to native forests, and high reproductive potential of C. externa. From the results, is possible to infer that the agroecosystem, a homogeneous environment, may be contributing to the loss of structure that existed among the populations when they lived in native ecosystems, and therefore, the populations of C. externa must be studied in this environment, so they could be understood in its natural habitat and without the influence of the human action
