Prediction of antimicrobial peptides using hyperparameter optimized support vector machines

Philosophiae Doctor - PhD / Antimicrobial peptides (AMPs) play a key role in the innate immune response. They can be ubiquitously found in a wide range of eukaryotes including mammals, amphibians, insects, plants, and protozoa. In lower organisms, AMPs function merely as antibiotics by permeabilizing cell membranes and lysing invading microbes. Prediction of antimicrobial peptides is important because experimental methods used in characterizing AMPs are costly, time consuming and resource intensive and identification of AMPs in insects can serve as a template for the design of novel antibiotic. In order to fulfil this, firstly, data on antimicrobial peptides is extracted from UniProt, manually curated and stored into a centralized database called dragon antimicrobial peptide database (DAMPD). Secondly, based on the curated data, models to predict antimicrobial peptides are created using support vector machine with optimized hyperparameters. In particular, global optimization methods such as grid search, pattern search and derivative-free methods are utilised to optimize the SVM hyperparameters. These models are useful in characterizing unknown antimicrobial peptides. Finally, a webserver is created that will be used to predict antimicrobial peptides in haemotophagous insects such as Glossina morsitan and Anopheles gambiae. / South Africa

Évaluation du risque de Trypanosomose Animale au Ghana, et suivi de l’impact de l’intervention d’éradication de la maladie et du vecteur dans la region Ouest du Ghana / EVALUATION OF RISK FACTORS OF ANIMAL TRYPANOSOMOSIS IN GHANA, AND THE MONITORING OF THE IMPACT OF DISEASE AND VECTOR ERADICATION INTERVENTION IN THE UPPER WEST REGION OF GHANA

Les Trypanosomoses Animales Africaines (TAA) sont une contrainte majeure à la viabilité et à la durabilité des systèmes de production de bétail au Ghana. Sous la tutelle de la Campagne Pan-Africaine d'Eradication des Tsé-tsé et des Trypanosomoses (PATTEC), le Ghana collabore avec le Burkina Faso au sein d'un projet sous régional pour créer une zone libérée de glossines à leur frontière commune. Les objectifs de cette thèse sont de i) déterminer la situation initiale avant intervention en ce qui concerne le vecteur et la prévalence de la maladie dans la zone de lutte, ii) déterminer la structure des populations de glossines et ses conséquences sur la durabilité des efforts de lutte anti-vectorielle, iii) évaluer l'efficacité de la Pulvérisation Séquentielle d'Aérosols insecticides (SAT) pour contrôler les glossines riveraines et iv) évaluer les risques environnementaux associés à cette stratégie de lutte. Les résultats de l'enquête de base conduite dans la région nord-ouest du Ghana (zone d'étude) ont montré une large distribution de Glossina tachinoides alors que Glossina palpalis gambiensis était limitée à la limite sud de la zone d'étude. La prévalence parasitaire moyenne chez les bovins était de 2.5% (IC 95%: 1.06-5.77) et la prévalence sérologique de 19% (IC: 14.03-25.35). La densité apparente par piège et par jour (DAP) des glossines était de 8.7, 1.9 et 1.3 respectivement le long des rivières Volta noire, Kulpawn et Sissili. Une structuration génétique importante des populations de G. tachinoides a été observée entre sites d'étude d'un même bassin versant et entre bassins versants. Une densité locale de 0.48-0.61 glossines/m² a été inférée, ainsi qu'une distance de dispersion d'environ 11m par génération [IC 9 - 17]. Aucun biais de dispersion sexe-spécifique n'a été détecté. La dispersion observée était suffisante pour qu'une zone libérée de G. tachinoides puisse être ré-envahie par les populations mitoyennes des bassins versants adjacents.L'efficacité de la SAT à éliminer les espèces de glossines riveraines dans une section particulièrement difficile (canopée très dense et fortes densités de glossines) et l'efficacité ultérieure, un an après la SAT, d'une stratégie de lutte intégrée, ont également été testées. Les résultats montrent l'échec de l'éradication, attribué à une pénétration insuffisante des aérosols insecticides dans les galeries forestières denses. Toutefois, le taux de réduction global obtenu par la SAT fut important (98%) et la stratégie intégrée parvint à maintenir un niveau important de suppression des glossines. Enfin, une mesure de l'impact environnemental du projet a montré un impact non significatif de la deltaméthrine en aérosols sur les arthropodes aquatiques et terrestres non-ciblés. / African animal trypanosomosis (AAT) is a major constraint to viable and sustainable livestock production systems in Ghana. Under the umbrella of the Pan African Tsetse and Trypanosomosis Eradication Campaign (PATTEC), Ghana is collaborating with Burkina Faso in a sub-regional initiative aiming at creating tsetse-free areas across their common borders. The objective of this thesis was to conduct research to guide project implementation and specifically seeks to i) determine the pre-intervention vector and disease situation of the intervention area, ii) determine tsetse population structuring and the consequences on sustainable tsetse control efforts, iii) evaluate SAT for the control of riverine tsetse species in Ghana and iv) evaluate the environmental risk of the intervention programmes. Results of a baseline survey conducted in the Upper West Region (study area) indicated a wide-spread prevalence of Glossina tachinoides but Glossina palpalis gambiensis was limited to the southern edge of the study area. Average parasitological prevalence in cattle was estimated at 2.5% (95% CI: 1.06–5.77) and serological prevalence measured at 19% (95% CI: 14.03–25.35). The mean Index of Apparent Abundance (IAA) of tsetse was 8.7, 1.9 and 1.3 for samples taken along the Black Volta, Kulpawn and Sissili Rivers, respectively. Investigations of the G. tachinoides populations confirmed significant structuring within and between the three main river-basins of the study area, and indicated a local density of 0.48-0.61 flies/m² and dispersal distance that approximated 11 m per generation [CI 9 - 17]. No significant sex-biased dispersal was detected. However, the observed dispersal was deemed sufficient for a G. tachinoides-cleared area to be reinvaded from neighbouring populations in adjacent river basins. The potential of Sequential Aerosol Technique (SAT) to eliminate riverine tsetse species in a challenging subsection (dense tree canopy and high tsetse densities) and the subsequent efficacy of an integrated strategy, one year after the SAT operations, were also investigated. Results indicated failure to achieve elimination, attributed to insufficient penetration of insecticide aerosols in thick riverine forest galleries. However the overall reduction rate due to SAT was important (98%) and the subsequent integrated strategy maintained high levels of tsetse suppression. Finally an environmental impact assessment revealed no significant impact of deltamethrin aerosols on non-targeted aquatic and terrestrial arthropods.

A comparative genomics approach towards classifying immunity-related proteins in the tsetse fly

Research suggests that tsetse fly immunity factors are key determinants in the success and failure of infection and the maturation process of parasites. An analysis of tsetse fly immunity factors is limited by the paucity of genomic data for Glossina spp. Nevertheless, completely sequenced and assembled genomes of Drosophila melanogaster, Anopheles gambiae and Aedes aegypti provide an opportunity to characterize protein families in species such as Glossina by using a comparative genomics approach. In this study we characterize thioester-containing proteins (TEPs), a sub-family of immunity-related proteins, in Glossina by leveraging the EST data for G.morsitans and the genomic resources of D. melanogaster, A. gambiae as well as A.aegypti.A total of 17 TEPs corresponding to Drosophila (four TEPs), Anopheles (eleven TEPs) and Aedes aegypti (two TEPs) were collected from published data supplemented with Genbank searches. In the absence of genome data for G. morsitans, 124 000 G.morsitans ESTs were clustered and assembled into 18 413 transcripts (contigs and singletons). Five Glossina contigs (Gmcn1115, Gmcn1116, Gmcn2398, Gmcn2281 and Gmcn4297) were identified as putative TEPs by BLAST searches. Phylogenetic analyses were conducted to determine the relationship of collected TEP proteins.Gmcn1115 clustered with DmtepI and DmtepII while Gmcn2398 is placed in a separate branch, suggesting that it is specific to G. morsitans.The TEPs are highly conserved within D. melanogaster as reflected in the conservation of the thioester domain, while only two and one TEPs in A. gambiae and A. aegypti thioester domain show conservation of the thioester domain suggesting that these proteins are subjected to high levels of selection. Despite the absence of a sequenced genome for G. morsitans, at least two putative TEPs where identified from EST data.

Coding of tsetse repellents by olfactory sensory neurons: towards the improvement and the development of novel tsetse repellents

Philosophiae Doctor - PhD / Tsetse flies are the biological vectors of human and animal trypanosomiasis and hence representant medical and veterinary importance. The sense of smell plays a significant role in tsetse and its ecological interaction, such as finding blood meal source, resting, and larvicidal sites and for mating. Tsetse olfactory behaviour can be exploited for their management; however, olfactory studies in tsetse flies are still fragmentary. Here in my PhD thesis, using scanning electron microscopy, electrophysiology, behaviour, bioinformatics and molecular biology techniques, I have investigated tsetse flies (Glossina fuscipes fuscipes) olfaction using behaviourally well studied odorants, tsetse repellent by comparing with attractant odour. Insect olfaction is mediated by olfactory sensory neurons (OSNs), located in olfactory sensilla, which are cuticular structures exposed to the environment through pore and create a platform for chemical communication. In the sensilla shaft the dendrite of OSNs are housed, which are protected by called the sensillum lymph produced by support cells and contains a variety of olfactory proteins, including the odorant binding protein (OBP) and chemosensory proteins (CSP). While on the dendrite of OSNs are expressed olfactory receptors. In my PhD, studies I tried to decipher the sense of smell in tsetse fly. In the second chapter, I demonstrated that G. f. fuscipes is equipped with diverse olfactory sensilla, that various from basiconic, trichoid and coeloconic. I also demonstrated, there is shape, length, number difference between sensilla types and sexual dimorphism. There is a major difference between male and female, while male has the unique basiconic sensilla, club shaped found in the pits, which is absent from female pits. In my third chapter, I investigated the odorant receptors which are expressed on the dendrite of the olfactory sensory neurons (OSNs). G. f. fuscipes has 42 ORs, which were not functionally characterised. I used behaviourally well studied odorants, tsetse repellents, composed of four components blend. I demonstrated that tsetse repellent is also a strong antifeedant for both G. pallidipes and G. f. fuscipes using feeding bioassays as compared to the attractant odour, adding the value of tsetse repellent. However, the attractant odour enhanced the feeding index. Using DREAM (deorphanization of receptors based on expression alterations of mRNA levels). I found that in G. f. fuscipes, following a short in vivo exposure to the individual tsetse repellent component as well as an attractant volatile chemical, OSNs that respond to these compounds altered their mRNA expression in two opposite direction, significant downregulation and upregulation in their number of transcripts corresponding to the OR that they expressed and interacted with odorant. Also, I found that the odorants with opposite valence already segregate distinctly at the cellular and molecular target at the periphery, which is the reception of odorants by OSNs, which is the basis of sophisticated olfactory behaviour. Deorphanization of ORs in none model insect is a challenge, here by combining DREAM with molecular dynamics, as docking score, physiology and homology modelling with Drosophila a well-studied model insects, I was able to predict putative receptors of the tsetse repellent components and an attractant odour. However, many ORs were neutral, showing they were not activated by the odorants, demonstrating the selectivity of the technique as well as the receptors. In my fourth chapter, I investigated the OBPs structures and their interaction with odorants molecules. I demonstrated that OBPs are expressed both in the antenna, as well as in other tissues, such as legs. I also demonstrated that there are variations in the expression of OBPs between tissues as well as sexes. I also demonstrated that odorants induced a fast alteration in OBP mRNA expression, some odorants induced a decrease in the transcription of genes corresponding to the activated OBP and others increased the expression by many fold in OBPs in live insect, others were neutral after 5 hours of exposure. Moreover, with subsequent behavioural data showed that the behavioural response of G. f. fuscipes toward 1-octen-3-ol decreased significantly when 1-octen-3-ol putative OBPs were silenced with feeding of double-stranded RNA (dsRNA). In summary, our finding whereby odorant exposure affects the OBPs mRNA, their physiochemical properties and the silencing of these OBPs affected the behavioural response demonstrate that the OBPs are involved in odour detection that affect the percept of the given odorant. The expression of OBPs in olfactory tissues, antenna and their interaction with odorant and their effect on behavioural response when silenced shows their direct involvement in odour detection and reception. Furthermore, their expression in other tissues such as legs indicates they might also have role in other physiological functions, such as taste.

The susceptibility of Trypanosoma congolense isolated in Zambézia Province (Mozambique) to isometamidium chloride, homidium chloride and diminazene aceturate

Bovine trypanosomosis is a serious constraint to livestock development in large parts of Mozambique. In most areas where tsetse flies are present, the disease in livestock is controlled using curative and prophylactic trypanocidal drugs. Those drugs have been used for many years and new drugs are unlikely to become available in the near future. As a result, trypanosomes have developed resistance against the currently available trypanocidal compounds. Drug resistance has been detected in various African countries and is a serious impediment to the control of livestock trypanosomosis. A study was initiated to determine whether drug resistant trypanosome strains are present in Zambézia Province of Mozambique. The aim of this study was to determine the sensitivity of Trypanosoma congolense isolates from Chinde, Nicoadala and Maganja da Costa Districts to diminazene aceturate, isometamidium chloride and homidium chloride. To assess the effect of the farming system and the intensity of drug regimens on the development of drug resistance, trypanosome isolates were collected from cattle from subsistence, semisubsistence and commercial livestock production systems. Drug-use practices in each of the production systems were determined using a questionnaire. The methodology used to assess the level of drugs resistance in the trypanosome isolates was the standardized method described by Eisler et al. (2001). Seven isolates were selected for resistance testing. For each of the seven isolates, five different doses varying between 0.01-20 mg/kg body weight for isometamidium chloride, 0.01-10 mg/kg body weight for homidium chloride and 1-30 mg/kg body weight for diminazene aceturate were used. For each dose rate six mice were treated intraperitoneally with the appropriate quantity of the drug dissolved in 0.2 ml of sterile distilled water 24 hours after the inoculation of the blood containing the trypanosomes. The control mice (six mice per trypanocidal drug) received the same amount of water without the drug. In four of the seven isolates high levels of multiple drug resistance (diminazene aceturate and isometamidium chloride) were detected. One isolate had a low level of multiple (diminazene aceturate and isometamidium chloride) drug resistance. Two isolates were susceptible to both diminazene aceturate and isometamidium chloride. One of those was highly susceptible to isometamidium chloride even at the lowest dose rate. The observed levels of drug resistance could in most cases be correlated to the drug-use practices in the particular livestock production system. The results obtained from homidium chloride treatment are not conclusive, because most the mice cured after receiving 10 mg/kg body weight of the drug. Hence more research is required to establish the homidium threshold in mice. Les insectes sont soumis aux fluctuations thermiques de leur environnement mais disposent d’un panel varié de réponses comportementales, physiologiques et biochimiques pour en minimiser les effets délétères et maintenir leur intégrité physiologique. Ainsi certaines espèces régulent activement leur température interne indépendamment de la température de l’environnement. Si ces insectes peuvent s’affranchir des contraintes thermiques imposées par leur environnement, ceux qui se nourrissent du sang chaud d’hôtes vertébrés endothermes n’ont pas d’autres choix que de se confronter à une situation de stress thermique à chaque prise alimentaire. Le principal objectif de ce travail de thèse est de comprendre comment des insectes hématophages, employant des stratégies alimentaires différentes, gèrent le stress thermique associé au flux massif de chaleur engendré par l’ingestion du repas de sang. Nos résultats montrent que ces insectes ont su s’adapter en développant différentes stratégies de thermorégulation. / Insects are submitted to thermal fluctuations of their environment and have developed a wide ranged panel of behavioral, physiological and biochemical responses, to minimize the subsequent deleterious effects and maintain their physiological integrity. Some species actively regulate their internal temperature independently of the temperature of the environment. If these insects can overcome the constraints imposed by their thermal environment, those that feed on warm-blooded vertebrate hosts have no choice but to confront a situation of thermal stress at each feeding event. The main objective of this work is to understand how bloodsucking insects manage heat stress associated with the massive flow of heat generated by the ingestion of the blood meal. Our results show these insects have developed different strategies of thermoregulation to protect themselves from overheating.

Χαρακτηρισμός των συμβιωτικών σχέσεων του βακτηρίου Wolbachia με έντομα αγροτικής, δασικής και ιατρικής σημασίας

Το βακτήριο Wolbachia είναι ένα ενδοκυττάριο και μητρικά κληρονομούμενο συμβιωτικό βακτήριο. Ανήκει στην ομοταξία των Alphaproteobacteria και την τάξη των Rickettsiales. Αποτελεί ίσως τον πιο διαδεδομένο ενδοκυττάριο συμβιωτικό οργανισμό στον πλανήτη, καθώς έχει εντοπιστεί μέχρι στιγμής σε πληθώρα αρθροπόδων και νηματωδών της φιλαρίασης. Πρόσφατες μελέτες εκτιμούν ότι πάνω από το 40% των ειδών αρθροπόδων είναι μολυσμένα με το βακτήριο Wolbachia. Το συμβιωτικό αυτό βακτήριο επηρεάζει τις βιολογικές λειτουργίες και ιδιότητες των ξενιστών του και είναι υπεύθυνο για μια σειρά αναπαραγωγικών ανωμαλιών, όπως η κυτταροπλασματική ασυμβατότητα, η παρθενογένεση, η θανάτωση των αρσενικών εμβρύων και η θηλυκοποίηση. Τα μοναδικά αυτά βιολογικά χαρακτηριστικά του βακτηρίου Wolbachia προσελκύουν όλο και περισσότερο το ενδιαφέρον διαφόρων ερευνητών τόσο για το ρόλο του βακτηρίου σε εξελικτικές διαδικασίες (κυρίως ειδογένεση) όσο και για τη χρησιμοποίησή του σε περιβαλλοντικά φιλικές εφαρμογές καταπολέμησης οργανισμών που είναι επιβλαβείς στους τομείς του γεωργικού και δασικού περιβάλλοντος, και της υγείας. Προς την κατεύθυνση αυτή, και στα πλαίσια της παρούσας διατριβής, πραγματοποιήθηκε η ανίχνευση του συμβιωτικού βακτηρίου Wolbachia σε περισσότερα από 5300 άτομα από φυσικούς και εργαστηριακούς πληθυσμούς 11 διαφορετικών ειδών μύγας τσε-τσε από 13 Αφρικανικές χώρες. Τα αποτελέσματα έδειξαν τεράστια απόκλιση της παρουσίας του βακτηρίου τόσο μεταξύ ειδών όσο και μεταξύ πληθυσμών του ίδιου είδους. Επίσης, πραγματοποιήθηκε ο γενετικός χαρακτηρισμός των στελεχών Wolbachia από συνολικά 29 αντιπροσωπευτικά δείγματα διαφόρων πληθυσμών και ειδών μύγας τσε-τσε, ενώ σε αρκετά από αυτά παρατηρήθηκαν πολλαπλά στελέχη του βακτηρίου. Διαπιστώθηκε εντυπωσιακή γενετική ποικιλότητα στελεχών Wolbachia που απαντούν στα διάφορα είδη μύγας τσε-τσε καθώς και ασυμφωνία μεταξύ των φυλογενειών των στελεχών Wolbachia και των μυγών τσε-τσε ξενιστών της, γεγονός που σημαίνει οριζόντια μετακίνηση του συμβιωτικού βακτηρίου κατά την εξέλιξη. Επιπρόσθετα, εντοπίστηκαν για πρώτη φορά εκτεταμένα γεγονότα οριζόντιας μεταφοράς βακτηριακών γονιδίων στο γονιδίωμα τριών ειδών μύγας τσε-τσε: στο Glossina morsitans morsitans, Glossina pallidipes και Glossina austeni. Από εξελικτικής σκοπιάς, κρίσιμα ερωτήματα προκύπτουν από τα παραπάνω ευρήματα, και πιο συγκεκριμένα σχετικά με: την προέλευση-μηχανισμό αυτών των γεγονότων οριζόντιας μεταφοράς, τον χρονικό προσδιορισμό τους, τον πιθανό ρόλο τους σε διαδικασίες ειδογένεσης και την επιλεκτική εμφάνισή τους σε ορισμένα μόνο είδη Glossina π.χ. στo υποείδos Glossina morsitans centralis που είναι πολύ συγγενικό του Glossina morsitans morsitans δεν παρατηρήθηκε το φαινόμενο. Εξίσου σημαντική και επιβεβλημένη κρίνεται η διεξοδική διερεύνηση του ενδεχομένου τα βακτηριακά γονίδια που ενσωματώθηκαν στο ευκαρυωτικό γονιδίωμα της μύγας τσε-τσε να ευθύνονται για την έκφραση νέων λειτουργιών-ιδιοτήτων (ή να μεταβάλλουν τις ήδη υπάρχουσες), ιδίως μάλιστα εάν αυτές συνδέονται με την αποδοτικότητα μετάδοσης της νόσου της τρυπανοσωμίασης μέσω του φορέα της, δηλαδή της μύγας τσε-τσε. Τέλος, διαπιστώθηκε πιθανή αρνητική συσχέτιση της παρουσίας του βακτηρίου Wolbachia με τον παθογόνο ιό Salivary Gland hypertrophy Virus (SGHV), γεγονός που συζητείται στα πλαίσια βιολογικών εφαρμογών καταπολέμησης του εντόμου-φορέα και της τρυπανοσωμίασης. Παράλληλα, μεγάλο ενδιαφέρον παρουσιάζει η προοπτική χρησιμοποίησης του βακτηρίου Wolbachia για τη βιολογική καταπολέμηση εντόμων αγροτικής ή /και περιβαλλοντικής σημασίας, όπως είναι οι αφίδες και η καρπόκαψα καστανιάς. Το γεγονός αυτό προϋποθέτει την ανίχνευση και τη γενετική ταυτοποίηση του βακτηρίου σε φυσικούς πληθυσμούς εντόμων. Στα πλαίσια της παρούσας διατριβής πραγματοποιήθηκε ανίχνευση και χαρακτηρισμός του βακτηρίου Wolbachia σε 78 συνολικά άτομα από 22 είδη αφίδων, από 26 φυσικούς πληθυσμούς από την Ελλάδα. Από αυτούς τους 26 πληθυσμούς, μόλις οι 4 βρέθηκαν να είναι μολυσμένοι με το βακτήριο Wolbachia και συγκεκριμένα πληθυσμοί των ειδών: Aphis fabae, Aphis hederae, Metopolophium dirhodum και Baizongia pistaciae. Τα αποτελέσματα αυτά δείχνουν για πρώτη φορά ότι η παρουσία του βακτηρίου Wolbachia στις αφίδες είναι πιθανά πιο διαδεδομένη από ότι προέκυπτε από προηγούμενες μελέτες. Επίσης, μελετήθηκε η ανίχνευση και ο χαρακτηρισμός του βακτηρίου Wolbachia στα είδη Cydia splendana, Cydia fagiglandana και Pammene fasciana. Το βακτήριο Wolbachia ανιχνεύθηκε για πρώτη φορά στα συγκεκριμένα είδη και μάλιστα διαπιστώθηκε ότι η συχνότητα εμφάνισής του ποικίλει τόσο μεταξύ των δύο ειδών Cydia όσο και μεταξύ των πληθυσμών του κάθε είδους. Στο είδος Pammene fasciana, το βακτήριο ανιχνεύθηκε σε όλα τα άτομα που μελετήθηκαν. Τα αποτελέσματα της παρούσας διατριβής συζητούνται από τη σκοπιά τόσο της οικολογικής και εξελικτικής σημασίας τους όσο και της προοπτικής χρησιμοποίησης του συμβιωτικού βακτηρίου Wolbachia για τον πληθυσμιακό έλεγχο επιβλαβών εντόμων όπως οι μύγες τσε-τσε, οι αφίδες και η καρπόκαψα καστανιάς. / Wolbachia is an intracellular and maternally inherited symbiotic bacterium that belongs to the class of Alphaproteobacteria and the order of Rickettsiales. It is the most ubiquitous intracellular symbiotic organism of the planet, since it has been estimated that over 40% of insect species, in addition to filarial nematodes, crustaceans, and arachnids are infected with Wolbachia. In arthropods Wolbachia affects the biological functions and properties of its hosts and it is responsible for a number of reproductive abnormalities, such as cytoplasmic incompatibility (CI), thelytokous parthenogenesis, feminization of genetic males and male killing. These unique biological characteristics of Wolbachia are attracting the interest of various researchers for: (a) decyphering the role of Wolbachia in evolutionary processes (mainly speciation), and (b) for its use in environmentally friendly applications for the control of agricultural pests and disease vectors. The species of genus Glossina (Diptera: Glossinidae) known as tsetse flies, have been found to be infected with Wolbachia. Tsetse flies are the sole vectors of pathogenic trypanosomes in tropical Africa, causing the “sleeping sickness” in humans and the “nagana” in animals. The potential use of Wolbachia for the control of tsetse flies, prerequisite a thorough knowledge of its genetic identity and the interactions with the host. To further characterize the prevalence of Wolbachia in tsetse flies an extensive screen of more than 5300 specimens from natural and laboratory populations of 11 different Glossina species originating from 13 African countries was carried out. Our results indicated a huge divergence in the prevalence of Wolbachia, both among the species and among populations of the same species. Further characterization by MLST and wsp genotyping was carried out for the Wolbachia strains of 29 representative populations and species of tsetse flies. An impressive genetic diversity of Wolbachia strains in tsetse flies was revealed. Interestingly, disconcordance between the phylogeny of Wolbachia and that of the tsetse flies was observed, suggesting horizontal transmission of Wolbachia during the evolution. Moreover, extended horizontal gene transfer events were detected for first time in Glossina morsitans morsitans, Glossina pallidipes και Glossina austeni. These results raise critical questions concerning: (a) the origin/mechanism of these horizontal gene transfer events, (b) their temporal determination, (c) their potential role as agents of speciation and (d) their selective appearance in only some Glossina species e.g in the subspecies Glossina morsitans centralis which is closely related with Glossina morsitans morsitans the phenomenon was not observed. Equally important will be to examine if genes from the chromosomal insertions were potentially expressed and examine if these genes are associated with the vectorial capacity of tsetse flies for the trypanosoma transmission. Finally, a negative correlation between the presence of Wolbachia with the Salivary Gland Hypertrophy Virus (SGHV) was identified. This is further discussed in the context of biological applications for control of tsetse fly-vector and trypanosomiasis. Finally in this thesis, the detection and characterization of Wolbachia in 78 specimens of 22 aphids species, from 26 natural populations, from Greece was examined. Only 4 out of 26 populations were found to be infected with Wolbachia, and specifically the species: Aphis fabae, Aphis hederae, Metopolophium dirhodum και Baizongia pistaciae. These results indicated that the presence of Wolbachia in aphids is probably more prevalent than it was derived from previous studies. Also, detection and characterization of Wolbachia in the Cydia splendana, Cydia fagiglandana and Pammene fasciana was carried out. Wolbachia was detected for first time in these species, and it was found that the prevalence of Wolbachia varies between the two species of Cydia and among populations of each species, with the infection in Pammene fasciana being fixed. At the end the ecological and evolutionary importance of Wolbachia, together with the use of the bacterium for the population control of harmful insects like tsetse flies, aphids and moths is further discussed.
