Estudo de agentes polinizadores em cultura de morango (Fragaria x ananassa Duchesne - Rosaceae) / Pollinators in strawberry crop(Fragaria x ananassa Duchesne - Rosaceae)

Os objetivos deste estudo foram: avaliar a fauna de polinizadores potenciais do morangueiro em campos de produçaõ comercial; verificar a contribuição de diversos agentes de polinização na produtividade do morangueiro, conhecendo a biologia floral e reprodutiva dos cultivares mais utilizados para o plantio; testar, em cultivo fechado (estufa), diversas espécies de Meliponini (abelhas sem ferrão)como polinizador do morangueiro, avaliando a sua adaptação ao cultivo e sua efetividade nesta cultura. Os resultados obtidos revelaram que as flores do dos cultivares "Sweet Charlie" e "Oso Grande" necessitam de um polinizador para a produção de frutos sem deformação pois a morfologia de suas flores e o amadurecimento de suas estruturas reprodutivas não favorecem a ocorrência da autopolinização, nem a distribuição homogênea do pólen pelos estigmas da flor, resultando em frutos deformados que não alcançam o seu potencial máximo de desenvolvimento. A abelha jataí (Tetragonisca angustula) adaptou-se bem ás condições do cultivo fechado (estufa) e mostrou-se efetiva na polinização do morangueiro, resultando em frutos mais pesados e em uma porcentagem mais elevada de frutos bem formados. / The aims of this study were: to investigate potencial pollinators of strawberry crops in commercial fields; to verify the contribution of several pollination agents on strawberry productivity, studying the floral and reproductive biology of some commercial cultivars; to evaluate the possibility of the rational use of stingless bees in strawberry pollination inside greenhouses. The results showed that the floral morphology of "Sweet Charlie" and Oso Grande" cultivars and the maturation pattern of their stamens and pistils nor favour the occurrence of self pollination neither the regular pollen distribuction among stigmata, yielding deformed fruits that don´t complete their development. Therefore these cultivars need pollinators to commercial production. Tetragonisca angustula (Apinae, Meliponini)was remarkable for its relatively quick adaptation inside greenhouses and for its effectivity as strawberry pollinator, resulting in heavier berries and high percentage of marketable fruits.

Impacts of greenhouse gases from coal power stations on climatic trends in Witbank areas, South Africa

MESHWR / Department of Hydrology and Water Resources / Greenhouse gases (GHGs) from coal power station affect the behaviour of climatic parameters such as the temperature, rainfall and evaporation, over a long period of time, hence causing climatic trends. This study focused on investigating the impacts of Greenhouse gases (GHGs) from coal power stations on climatic and hydrological trends in Witbank area. To accomplish this, linear regression (LR) and Mann-Kendall (MK) trend test were used to detect the hydro-climatic trends and their significance. GHG emissions were obtained from Eskom’s sustainability report on the Eskom website. Temperature data for the years 1950- 2000 and 1993-2016 and rainfall data for the years 1925-2000 and 1993-2016 were used. Double Mass Analysis (DMA) was used to check the homogeneity and consistency of temperature and rainfall data from South African Weather Services (SAWS) station with the Lynch database and Water Research Commission (WRC) data. Data was patched and extended using LR where necessary. Trends in temperature, precipitation and flow were assessed using MK trend test and LR based on monthly, seasonal, and annual scales. GHG emissions were compared with the hydro-climatic data over time in order to detect the impacts of GHG emissions on temperature, rainfall and streamflow. The MK results indicated that GHG emissions had some impacts on temperature with statistically significant increase in annual, monthly and seasonal time scales for the period 1950-2016. LR also produced the same results for annual temperature. Monthly and seasonal temperature could not be produced with the LR method because of data gaps. The MK and LR models produced similar results, indicating that there was a non-significant increase in temperature before coal power stations were introduced (1950-1974) and a significant increase in temperature after the commissioning of coal power stations (1975-2016). MK and LR also produced the same results for annual rainfall data, indicating that there was a significant increase in rainfall before coal power stations were introduced (1925-1974) and a non-significant increase after the commissioning of coal power stations (1975-2016). For monthly time scales MK and LR indicated increasing and decreasing trends before and after coal power stations were introduced. MK and LR results for streamflow stations B2H004 and B2H007 showed similar results indicating non-significant increase in annual and seasonal streamflow, but differed in monthly streamflow where MK showed significant increases whilst LR showed non-significant trends. The study concluded that GHGs from coal power stations had significant impacts on the hydro-climatic trends in Witbank area. GHGs from coal power stations caused significant increase in temperature as temperature increased by 3.7°C after coal power stations were introduced, whereas temperature had increased by 1.7 °C. It is recommened that more research should be done on alternative sources of energy such as wind and solar energy to check their suitability and applicability in South Africa. / NRF

Agrivoltaic Implementation in Greenhouses : A Techno-Economic Analysis of Agrivoltaic Installations for Greenhouses in Sweden

Due to the growing population and climate change, the world will see an increase in demand for food, freshwater and renewable energy supply. Agrivoltaics has the possibility to address all these problems, by producing food and renewable energy but also by reducing water usage in agriculture. This thesis aims to study if agrivoltaics including storage has the potential to enable sustainable greenhouses in Stockholm, Sweden by trying to create a near net zero energy consumption for greenhouses with Agrivoltaics (AV) implemented. Furthermore a techno-economic assessment will be made for the AV-systems where Key Performance Indicator (KPI)’s are compared to economic parameters. The selected KPI’s were a near net zero energy consumption and irradiance underneath the Photovoltaics (PV) technology. The selected PV-technology was standard PV-modules, Semi-Transparent Module (STM) and Organic Solar Cell (OSC) PV. These technologies were paired with li-ion batteries between 0-100 kWh and simulated in the software System Advisor Model (SAM) over a 25 year period. The AV system was applied to two load profiles, one for indoor plants and one for tomatoes. The economic parameters calculated was Net Present Value (NPV), Net Capital Cost (NCC), and Levelised Cost of Electricity (LCOE).  The results showed that the system is efficient in summertime where the PV reached maximum capacity in summer and the battery works as a complement. In wintertime, the AV-system is not very efficient and most of the electricity comes from the grid. It was not possible to create a near net zero energy consumption including storage in Stockholm Sweden. The irradiance beneath the panels were at a maximum for OSC, it was slightly reduced for the STM, and below 50% for the standard PV-module, depending on the size of the AV-system. Depending on the shade tolerance of the plant, the PV-technology should be selected.

O estado de São Paulo é responsável por 70% da produção nacional de flores e plantas ornamentais. Esses cultivos ocupam 7,5 mil hectares e mais da metade desta área é cultivada em estufas. Atualmente existem aproximadamente 8 mil produtores de pequeno, médio e grande porte em todo Brasil e este setor gera em torno de 200 mil empregos diretos, abrangendo produção e comércio. As estufas são microcosmos planejados para propiciar as condições ambientais adequadas ao crescimento de plantas. Qualquer infestação é controlada usando praguicidas que, devido ao enclausuramento e ajustes das condições climáticas, podem prolongar e intensificar a exposição ocupacional aos mesmos. Alguns estudos sugerem que os trabalhadores de estufas de flores e plantas ornamentais estão expostos a níveis mais elevados de praguicidas durante o carregamento, mistura e aplicação dos praguicidas, bem como pelo contato contínuo com as flores e plantas ornamentais, quando comparados a outros trabalhadores que entram em contato com essa classe de compostos. No presente trabalho, objetivou-se conhecer o processo de trabalho, práticas de saúde, higiene e segurança e localizar possíveis fontes de exposição ocupacional a praguicidas nas estufas de flores da região do Alto Tietê, e também otimizar métodos analíticos para a detecção e quantificação de praguicidas organofosforados (diclorvós, metil-paration e metidation) em patches de algodão e em tubos coletores XAD-2, com o intuito de analisar a exposição ocupacional dérmica e inalatória. O processo de avaliação incluiu o acompanhamento da jornada de trabalho, entrevistas e aplicação de questionários de trabalhadores de 18 estufas. Destas 18 foram selecionadas 6 estufas para coleta de amostras. Os organofosforados diclorvós, metidation e metilparation foram detectados e quantificados por cromatografia gasosa/espectrometria de massas (GC-MS), operado no modo de ionização por impacto de elétrons. Os dados da avaliação qualitativa demonstraram percentual elevado de desconhecimento das práticas adequadas de saúde, higiene e segurança para a utilização destes compostos, sugerindo que há risco de exposição durante a manipulação, armazenamento e descarte dos mesmos. Os limites de quantificação (LQ) obtidos foram abaixo de 0,4 ng/mL e limites de detecção (LD) foram abaixo de 0,1 ng/mL para todos os compostos. O método mostrou boa linearidade na concentração estudada (LSQ-500 ng/mL), com coeficiente (r) maior que 0,99. A precisão foi avaliada pelo desvio padrão relativo e apresentou valores inferiores a 15% para todos os compostos estudados. Após os métodos serem validados, foi realizada a amostragem dérmica e do ar e encontraram-se concentrações variadas de praguicidas. / The state of São Paulo - Brazil, accounts for 70% of domestic production of flowers and ornamental plants. Currently, these crops occupy 7.5 thousand hectares and over 50% of this area is cultivated in greenhouses. This market has an average of 8.000 producers and generates around 200.000 jobs. Greenhouses are microcosms designed to provide environmental conditions suitable for plant growth. Infestation is controlled using pesticides that due to the closure and adjustment to the weather conditions can prolong and intensify the exposure to them. Some studies suggest that workers in greenhouses of flowers and ornamental plants are exposed to higher levels of pesticides during loading, dilution and application, as well as by continuous contact with flowers and ornamental plants, when compared to other workers who enter in contact with this class of compounds. The aim of the present was to know the work process, to survey the practices of health, hygiene and safety standards and locate possible sources of occupational exposure to pesticides in greenhouses of flowers in the Alto Tietê region (São Paulo state), and to optimize analytical methods for detection and quantification of organophosphate pesticides (dichlorvos, methyl parathion and methidathion) in patches of cotton and in XAD-2 sorbent tubes, with the aim of analyzing the occupational dermal and inhalation exposure. The evaluation process included the monitoring of the working day, interviews and questionnaires, and samples collected in greenhouses. The organophosphorus compounds were detected and quantified by gas chromatography/mass spectrometry (GC-MS) operated in electron impact ionization. The data show a high percentage of lack of appropriate health practices, hygiene and safety for pesticide use suggests that there is risk of exposure during handling, storage and disposal of them. The limits of quantification (LOQ) obtained were below 0.4 ng/mL and limits of detection (LOD) were below 0.1 ng/mL for all compounds. The method showed good linearity in the studied concentration (LOQ-500 ng/mL), with coefficient (r) greater than 0.99. The precision was evaluated by relative standard deviation and showed values below 15% for all compounds studied. After the methods are validated, the dermal and air sampling was performed and a great range of pesticide concentrations was found. So it is evident that this study is valuable for future analysis and guidance to workers about possible hazards and proper use for reducing exposure to these compounds.

Ενεργειακή βελτιστοποίηση θερμοκηπίου με χρήση συστήματος θέρμανσης με υπέρυθρη ακτινοβολία : θεώρηση της μικρού μήκους κύματος ακτινοβολίας (NIR)

Μετά την πρώτη ενεργειακή κρίση την δεκαετία του ’70 κατά την οποία τα περιορισμένα αποθέματα ενέργειας προκάλεσαν την πρώτη σημαντική αύξηση στην τιμή της ενέργειας, η χρήση ενέργειας στα θερμοκήπια έχει γίνει κύριο ερευνητικό ζήτημα. Η ανάγκη για μείωση του ενεργειακού κόστους είναι σημαντική, γιατί η ενέργεια αποτελεί σημαντικό κλάσμα του συνολικού κόστους παραγωγής. Στις Μεσογειακές χώρες έχει υπολογιστεί ότι η χρήση ενέργειας για έλεγχο των συνθηκών περιβάλλοντος και ειδικότερα για τη θέρμανση, είναι 20% - 30% του συνολικού κόστους παραγωγής, ποσοστό το οποίο στις βορειότερες χώρες αυξάνεται. Ταυτόχρονα, με το έντονο ενδιαφέρον για το παγκόσμιο φαινόμενο του θερμοκηπίου και τις κλιματικές αλλαγές, η χρήση των συμβατικών καυσίμων είναι και πάλι στην πολιτική ατζέντα. Έτσι και η βιομηχανία θερμοκηπίων είναι αντιμέτωπη με οικονομική, πολιτική και κοινωνική πίεση για μείωση της χρήσης ενέργειας και βελτίωση της ενεργειακής απόδοσης των θερμοκηπίων μέσω τεχνολογικών καινοτομιών. Η κατανάλωση ενέργειας για τη θέρμανση του θερμοκηπίου αποτελεί ένα πολύ σοβαρό πρόβλημα διότι οι απώλειες του θερμοκηπίου σε θερμότητα λόγω των λεπτών τοιχωμάτων του και της κατασκευής του, είναι πολύ μεγάλες, 6-12 φορές μεγαλύτερες από εκείνες ενός συνήθους κτίσματος ίσου όγκου. Η θερμότητα παρέχεται στο θερμοκήπιο κυρίως μέσω συμβατικών συστημάτων θέρμανσης (συστήματα σωληνώσεων θερμού νερού, συστήματα θερμού αέρα) και σε περιορισμένη έκταση με χρήση ανανεώσιμων πηγών ενέργειας (ηλιακή, γεωθερμία, βιομάζα). Για να επιτευχθεί με αυτές τις μεθόδους η απαραίτητη θερμοκρασία στο επίπεδο των φυτών, το εσωτερικό του θερμοκηπίου πρέπει να θερμανθεί στην ίδια ή υψηλότερη θερμοκρασία από την επιθυμητή θερμοκρασία των φυτών με αποτέλεσμα την δημιουργία ισοθερμοκρασιακού κλίματος σε ολόκληρο το θερμοκήπιο (όλον κλίμα). Το αποτέλεσμα αυτής της πρακτικής σε όλες τις μελέτες που διεξάγονται είναι ότι, τα θερμοκήπια καταναλώνουν απαράδεκτα υψηλά ποσά ενέργειας σε σχέση με την ενέργεια που απορροφάται από τα φυτά, για να καλύπτουν τις αυξημένες ενεργειακές απώλειες που λόγω κατασκευής παρουσιάζουν. Στην παρούσα διατριβή αναπτύσσεται μια ολοκληρωμένη πρόταση για την δημιουργία ενός "ψυχρού θερμοκηπίου" στο οποίο τα φυτά θα λαμβάνουν απευθείας την ενέργεια που χρειάζονται προκειμένου να φτάσουν και να διατηρήσουν την επιθυμητή για την ανάπτυξή τους θερμοκρασία χωρίς να υπάρχει ανάγκη για αύξηση της θερμοκρασίας του αέρα του θερμοκηπίου. Για τον σκοπό αυτό χρησιμοποιείται σύστημα θέρμανσης με μικρού μήκους υπέρυθρη ακτινοβολία (NIR). Σε αυτά τα συστήματα η θερμότητα μεταδίδεται απευθείας από την πηγή στον δέκτη, στην προκειμένη περίπτωση στα φυτά και το έδαφος και το αποτέλεσμα είναι η δημιουργία ισοθερμοκρασιακού κλίματος μόνο στην περιοχή του φυτικού θόλου (τοπικό κλίμα). Ειδικότερα η παρούσα εργασία αντιμετωπίζει τη θερμική συμπεριφορά του θερμοκηπίου λαμβάνοντας υπόψη όλα τα ουσιαστικά θερμικά φαινόμενα που συμβαίνουν κατά την διάρκεια θέρμανσης (με συμβατικό και με σύστημα θέρμανσης με ακτινοβολία) και παρουσία καλλιέργειας. Γι αυτό τον λόγο δίνεται ιδιαίτερη βαρύτητα στην ανάπτυξη ενός μαθηματικού μοντέλου που προσομοιάζει επαρκώς τις κύριες διαδικασίες μεταφοράς θερμότητας μέσα στο θερμοκήπιο, το οποίο περιλαμβάνει ένα σύνολο λειτουργικών συνθηκών, και στο οποίο καθορίζονται με σχετική ακρίβεια οι παράγοντες που επηρεάζουν την θερμική συμπεριφορά του θερμοκηπίου. Το μοντέλο προσομοίωσης δίνει την δυνατότητα αξιόπιστης περιγραφής του περιβάλλοντος του θερμοκηπίου, και σαφούς εκτίμησης των ενεργειακών αναγκών του και με τις δυο τεχνικές θέρμανσης. Ετσι προσδιορίζεται το εξοικονομούμενο ενεργειακό όφελος που προκύπτει με τη χρήση συστήματος θέρμανσης με ακτινοβολία. Ο έλεγχος της ποιότητας του μοντέλου προσομοίωσης και η περαιτέρω βελτίωσή του γίνεται με σύγκριση των θεωρητικών αποτελεσμάτων με πειραματικά δεδομένα, η συλλογή και επεξεργασία των οποίων αποτελεί τον πυρήνα της έρευνας. Τα πειραματικά δεδομένα προέρχονται από πειράματα που διεξήχθησαν σε δύο πειραματικά θερμοκήπια συζευγμένα με αυτόματο μετεωρολογικό σταθμό, στα οποία εφαρμόζεται συμβατικό σύστημα θέρμανσης και θέρμανση με υπέρυθρη ακτινοβολία αντίστοιχα. Αυτό δίνει την δυνατότητα πιστοποίησης των δυνατοτήτων της θέρμανσης με ακτινοβολία και ποσοτικοποίησης του ενεργειακού οφέλους που επιτυγχάνεται. Συμπερασματικά, η συστηματική θεωρητική και πειραματική μελέτη ενός "ψυχρού" θερμοκηπίου με χρήση μικρού μήκους υπέρυθρης ακτινοβολίας (NIR) και ενός "θερμού θερμοκηπίου" με χρήση συμβατικού συστήματος θέρμανσης αναδεικνύουν το κύριο πλεονέκτημα της υπέρυθρης ακτινοβολίας, δηλαδή η θερμότητα να μεταδίδεται απευθείας από το σύστημα ακτινοβολίας στα φυτά και το έδαφος χωρίς να παρεμβάλεται ο αέρας του εσωτερικού περιβάλλοντος του θερμοκηπίου. Αυτό οδηγεί σε ομοιομορφία θέρμανσης του φυτικού θόλου και ταυτόχρονα σε σημαντική εξοικονόμηση ενέργειας της τάξεως του 40-50%. / After the first energy crisis in the seventies during which limited energy supplies led to an important increase in energy prices, greenhouse energy consumption has again become a major research issue. The energy cost reduction need is significant, since energy forms a substantial fraction of the total production costs. In Mediterranean countries it has been estimated that energy consumption for environment conditions control and more specifically for heating, consists 20% - 30% of total production cost, a percentage that is higher in northern countries. Moreover, due to the recent pronounced interest in the global greenhouse effect and climatic change, the use of fossil fuels is once again in the political agenda. Therefore the greenhouse industry is confronted with economical, political and social pressure to reduce energy usage and improve the greenhouse energy efficiency via technological innovations. Energy consumption for greenhouse heating represents a serious concern because greenhouse heat losses due to thin covers and construction specifics are 6-12 times higher than those of a common building of equal volume. Traditionally, thermal energy is transmitted to the greenhouse mainly through conventional heating systems (either by hot water circulation through a piping system or by air heaters) and, in a limited scale, through renewable energy sources (solar, geothermal, biomass). In order for the plants to reach required temperature through these methods, the greenhouse interior has to be heated to the same or even to a slightly higher temperature than the value targeted for the plants (entire climate). This practice results in extremely increased heat losses compared to the energy absorbed by plants, because of the increased energy losses due to construction specifications. The present thesis formulates a complete proposal for the creation of a "cold greenhouse" where plants will directly receive the needed energy in order to reach and preserve the desirable growth temperature without having to increase the internal greenhouse air temperature. The near infrared radiation (NIR) is used for this purpose. In the radiation system, heating is transmitted straight from the source to the receiver, in this particular case plants and soil. The result is an isothermal climate formation in the plant canopy (local climate). Specifically this study investigates the greenhouse thermal performance taking into account all the essential thermal phenomena that take place during heating (conventional and IR heating) and cultivation. For this reason, emphasis is given to the development of a mathematical model that simulates the main heating transfer procedures inside the greenhouse, takes into account a sum of operational conditions and determines all factors influencing the greenhouse thermal performance with relevant accuracy. The simulation model allows a credible description of greenhouse environment as well as a clear estimation of its energy needs with both heating systems. Thus the resulting energy saving by IR heating usage is determined. Quality control of the simulation model and its forward improvement is done by comparing the theoretical results with experimental data. Collecting and processing these data forms the research kernel. The experimental data correspond to experiments that took place in two experimental greenhouse connected with automatic meteorological station where conventional and IR heating have been used respectively. This method makes possible the identification of IR heating potential and quantification of energy saving. Concluding, the systematic theoretical and experimental study of a "cold" greenhouse using near IR heating, and of a "warm" greenhouse using conventional heating, proves the main advantage of IR heating that is that, the heat is directly transferred from the radiation system to the plants and the soil without interference of the internal greenhouse air. This leads to uniform heating of the plant canopy and at the same time to a significant energy saving of 40-50%.

Computational study of low index surface of an anatase TiO2 doped with ruthenium (Ru) and strontium (sr) for application in Dye sensitized solar cells

MSc (Physics) / Department of Physics / Titanium dioxide (TiO2) is considered to be an ideal semiconductor for photocatalysis because of its high stability, low cost and safety towards both humans and the environment. Doping TiO2 with different elements has attracted much attention as the most important way of enhancing the visible light absorption, in order to improve the efficiency of the dye sensitized solar cells (DSSCs). In this study, first principle density functional theory was used to investigate electronic and optical properties of bulk anatase TiO2, undoped, and ruthenium (Ru) and strontium (Sr) doped anatase TiO2 (1 0 0) surface. Two different doping approaches i.e., substitutional and adsorption mechanisms were considered in this study. The results showed that absorption band edges of Ru and Sr-doped anatase TiO2 (1 0 0) surface shift to the long wavelength region compared to the bulk anatase TiO2 and undoped anatase TiO2 (1 0 0) surface. Also, the results revealed that the band gap values and the carrier mobility in the valence band, conduction band and impurity energy levels have a synergetic influence on the visible-light absorption and photocatalytic activity of the doped anatase TiO2 (1 0 0) surface. Furthermore, according to the calculated results, we propose the optical transition mechanisms of Ru and Sr-doped anatase TiO2 (1 0 0) surface. Thus, we conclude that the visible light response of TiO2 can be modulated by doping with both Ru and Sr. However, Sr-doped system shows higher photocatalytic activity than the Ru-doped system. La sélection naturelle constitue un processus clé de l’adaptation des populations à leur environnement, favorisant les variants présentant les meilleures valeurs sélectives. Les champignons présentent généralement des traits biologiques (diversité des modes de reproduction, grandes tailles de populations, fortes capacités de dispersion, entre autres) qui favorisent leur adaptation à des environnements variés. La compréhension des mécanismes qui sous-tendent l’évolution de leurs populations sous les contraintes, naturelles et anthropiques, qu’elles subissent constituent donc un enjeu majeur pour la protection des plantes, en particulier dans le contexte actuel de durabilité des méthodes de lutte. Dans cette thèse, nous avons décrit la structure et la diversité des populations Botrytis cinerea à l’aide de marqueurs neutres et sélectionnés et d’un échantillonnage emboîté, et avons proposé des mécanismes pouvant expliquer les résultats observés. Puis nous avons analysé la réponse adaptative des populations de B. cinerea en Champagne, aux applications de fongicides. Premièrement, nous avons montré que la pourriture grise était causée par un complexe de deux espèces cryptiques, vivant en sympatrie sur des hôtes communs. De plus, les populations françaises de B. cinerea sont structurées en cinq dèmes, caractérisés par le système de culture (sélection directionnelle), la plante-hôte (adaptation écologique), et dans une moindre mesure, par la géographie. Sur vigne, nous avons mis en évidence une entité dont l’isolement génétique semble lié à un isolement temporel. Par ailleurs, nous avons montré que l’application de fongicides conduit à la sélection de phénotypes résistants spécifiquement à quasiment tous les modes d’action homologués, selon des proportions variant suivant les vignobles et les usages. Plus particulièrement, la résistance aux fongicides inhibiteurs de la succinate déshydrogénase (SDHI) est causée par au moins sept mutations affectant les gènes encodant la protéine cible de ces fongicides, déterminant ainsi une grande variété de phénotypes. Enfin, nous avons montré que les fongicides ne modifiaient pas la structure neutre des populations mais qu’ils pouvaient conduire à une perte de richesse allélique dans les populations traitées ainsi qu’à un équilibre sélection-migration détectable dans certaines situations sous forme de clines au loci sous pression de sélection contemporaine tels que ceux déterminant la résistance multidrogues. La modélisation de l’évolution des fréquences de résistance hivernale a permis d’estimer le coût de la résistance pour quatre loci déterminant la résistance aux fongicides. Cette thèse a permis d’appréhender le fonctionnement des populations de B. cinerea et de comprendre et quantifier partiellement les mécanismes sélectifs opérant in natura. Ces informations seront utilisées pour raisonner des stratégies anti-résistance adaptées localement et durables. / Natural selection is the most powerful force driving population adaptation to their environment, favoring the variants with the best fitness. Fungi generally exhibit biological traits (diversity of reproduction modes, large population sizes, and intense dispersion) that favor their adaptation to changing environments. Therefore, disentangling the mechanisms that explain their evolution under natural and anthropic constraints constitute a major challenge for plant protection, especially in the actual context of agriculture sustainability. In this thesis, we described Botrytis cinerea population structure and diversity, using neutral and selected markers and a hierarchical sampling, and proposed mechanisms that may explain these observations. We then analyzed the adaptive answer of this species towards fungicide applications. First, we showed that grey mold populations were caused by a complex of two cryptic species, living sympatrically on the same hosts. Second, B. cinerea populations are divided into five demes, according to the cropping system (directional selection), the host-plant (ecological adaptation), and to a lesser extent, by geography. On grapevine, we identified a specific populations exhibiting temporal isolation, as an evidence of extreme exploration of the viticultural conditions. Moreover, fungicide applications select resistance towards all unisite modes of action, with few exceptions, but at varying proportions according to vineyards and fungicide use. More specifically, resistance to succinate dehydrogenase inhibitors (SDHIs) is caused by at least seven mutations altering the target genes of these fungicides, and determines a large variety of phenotypes in the field. At last, we showed that fungicides did not shape population structure but that they could decrease allele richness in treated areas and lead to migration-selection equilibrium, detectable in some situation and for loci under contemporary selective pressures as clines. Modeling the evolution of resistance during winter allowed estimating fitness cost of four loci involved in contemporary fungicide resistance, such as multidrug resistance. As a conclusion, this thesis helped to understand how B. cinerea populations evolve and to detect and quantify selective mechanisms at work in natura. This information will be useful to deign sustainable and locally-adapted anti-resistance strategies.

Development of a mass rearing technique for the Tasmanian brown lacewing, Micromus tasmaniae Walker

Aphids are one of the most important insect pests of greenhouse crops yet to be controlled by biological means. Broad spectrum chemical control is becoming increasingly difficult to use in integrated pest management programmes, therefore, there is a need for a suitable biocontrol agent to be mass reared and released. The Tasmanian brown lacewing, Micromus tasmaniae Walker is an aphid predator that is found commonly throughout Australasia and has suitable characteristics that make it a candidate for mass rearing. A technique for rearing M. tasmaniae was developed. Eggs of M. tasmaniae were reared in batches of 50, 100 and 200 in 20 litre clear plastic containers. The oat aphid, Rhopalosiphum padi L. was fed to the larvae. The results revealed that the highest initial egg density (200 eggs per container) produced the cheapest adults at 22 cents per adult. However, mass rearing adults was considered not practical because of the high production cost, although, mass production of eggs is considered to be economically viable. The cost of producing one egg was 0.015 cents. M tasmaniae was maintained in mass culture for six generations. Simple experiments were carried out to monitor the quality of laboratory-reared insects. The 'wild' insect was used as a quality standard and comparisons with laboratory-reared insect populations were made. The fecundity, development rates and tolerance to pirimicarb, a carbamate insecticide, were determined. Fecundity was found to decline with successive generations in mass culture. The lacewing development experiment indicated that larval stages of each generation suffered the highest mortality rate and that between 35-45% of individuals emerged as adults. The tolerance of adults to pirimicarb did not alter over five generations. Recommendations for improving the mass rearing of M. tasmaniae are discussed.

Estimation of the emissions of gases from a two landfill sites using the LandGEM and Afvalzorg models: Case study of the Weltervenden (Polokwane) and Thohoyandou landfills

MENVSC / Department of Ecology and Resource Management / Over the years it has been observed that the solid waste sector has been an increasingly major contributor to the amount of Greenhouse gases (GHGs) in the atmosphere. To some extent a great chunk of these GHGs in the atmosphere is from Landfill gas (LFG). This study employs two theoretical models (LandGEM and Afvalzorg models) to estimate the amount of LFG emitted from Weltervenden and Thohoyandou landfill sites located in Limpopo province of South Africa. Furthermore, the study investigated the appropriate technique of the LFG utilisation as a source of electricity and the number of households using electricity. LFGcost model was used to estimate the cost and benefits related to the implementation of a LFG utilisation technology. Also, the possible health and environmental impacts of the landfill emissions on the people living closer to the landfill site were determined. The LandGEM model’s simulation concludes that CH4 and CO2 peaked in the year 2020 with values of 3.323 × 103 Mg/year and 9.118 × 103 Mg/year, respectively, for the Thohoyandou landfill. Results from the Afvalzorg model indicate that the CH4 peaked in the year 2020 with value of 3.501 × 103 Mg/year. Meanwhile the total emission of CH4 from 2005-2040 by the LandGEM and Afvalzorg models are 66200 Mg/year and 69768 Mg/year, respectively. However, for the Weltervenden landfill, the total LFG peaked in the year 2023 while the CH4 peak at 4061 Mg/year and 3128 Mg/year for LandGEM and Afvalzorg models, respectively. Furthermore, results from the cost analysis and benefits for the implementation of a LFG utilisation technology in both landfills show that the implementation of such a utilisation technology will be economically feasible considering the sale of t CO2 equivalent in the carbon market. However, without considering the sales of t CO2 equivalent, not all the LFG engines are economically feasible for both landfills. This study also shows that the residents living closer to the Thohoyandou landfill are at a higher risk of environmental pollution and could suffer negative impacts from the landfill than residents living far from the landfill site. However, the Weltervenden landfill did not have lots of communities living closer to the landfill and therefore it was not included in this study. / NRF
