Effect of intrinsically disordered regions on ESR1 transcriptional activity

Approximately 70% of breast cancers are estrogen receptor positive, of which 40% have ER mutations. Some of these mutations occur in the three intrinsically disordered regions (IDRs) of estrogen receptor-α (ERα). It is not fully understood how these mutations affect the function of ERα, however they have been suggested to affect DNA and ligand binding, transcriptional activation and dimerization. In this study, we used mammalian one-hybrid (M1H) assays to determine the effect of deletions and mutations in the IDRs on the activation of ERα. We found that deletions in the N- and C-terminal IDRs and 7/11 patient mutations tested decreased the transcriptional activation of ERα in the presence of 17β-estradiol (E2). Interestingly, a 23x glycine-serine replacement of the hinge IDR replacement resulted in a 7-fold increase in activation. Our results show that IDRs are important for full transcriptional activation and regulation of ERα. / 2025-06-23T00:00:00Z

Interlaminar properties of 3D textile composites

Multilayer composite materials have a high tendency to interlaminar delamination when they are subjected to out-of-plane loading, because of their low-stiffness in the through-thickness (T-T) direction. The main aim of this research was to improve the interlaminar fracture toughness (IFT) of textile composites by using stitching as a T-T reinforcement technique. The intention was to provide greater delamination resistance and also to enhance the interlaminar fracture toughness between adjacent layers. In this research, E-glass 2x2 twill weave structure fabric layers and an epoxy resin were chosen as the base materials. Three different types of stitching; including the commonly-used modified lock-stitch and orthogonal-stitch (OS) geometries, the single-yarn orthogonal-stitch (SOS) and a newly-developed double-yarn orthogonal-stitch (DOS), as well as five different stitch densities were used to reinforce the multilayer preform lay-ups. The resin infusion moulding method was used to manufacture the E-glass/Epoxy 3D textile composites. The effect of stitched reinforcement on the Mode I-IFT mechanism was examined by performing double cantilever beam (DCB) tests and the Mode II-IFT mechanism by performing four-point bend end-notched flexure (4ENF) tests, respectively. Optical microscopy and scanning electron microscopy (SEM) imaging techniques were used to study the fracture surfaces of the stitched composite specimens, to assess the improvement in IFT mechanisms imparted by the stitched reinforcement used. The effect of stitching was analysed by comparing the various stitching geometries, stitch densities and the mechanical properties highlighted by the Mode I-IFT and Mode II-IFT results. It was found that the use of the novel double-yarn orthogonal-stitch (DOS) reinforcement allied with the use of high stitch densities gave the greatest improvement on both Mode I-IFT and Mode II-IFT tests. Moreover, in every case, the use of DOS and high stitch densities gave a significant improvement of 74.5% in Mode I-IFT and 18.3% for Mode II-IFT tests when compared with unstitched samples. It has been shown that the novel DOS stitch geometry yields significant benefits over established stitching techniques in respect of stitched reinforcement for laminated composite preforms. Besides this, the double column 5x5 mm stitch pattern reveals the highest delamination resistance performance among all the stitching formations tested for Mode I-IFT and II-IFT.

Adaptive evolution of Transcription Factors in European and wine yeast

<p>The mutability of transcription factors (<strong>TF</strong>) is thought to be of high importance for the evolutionary change of living organisms. Transcription factors, coactivators, coregulators, kinases, chromatin remodelers conditional factors and other proteins together govern the timing and level of gene expression. About 10% of the genes in the human genome are predicted to be TFs and mutational changes in these genes or in the target regulatory sequences they bind will potentially give rise to evolutionary advantages or malfunctions for the organism. Recent research has suggested that the parts of the transcription factors that are not structurally defined in solution, so called intrinsically disordered regions (IDRs), have a higher potential for evolutionary diversification than more structurally rigid regions. This suggests that these domains that earlier have been considered mostly unimportant may have an important potential for evolutionary diversification. This project aimed to further evaluate evidence supporting the hypothesis that variable-structured domains in transcription factors are of significant importance for functional diversification. This was be done by comparing the rate of synonymous and non-synonymous genetic variation in the coding regions of 12 selected TFs within a highly conserved clade of European wine yeasts and by comparing this variation to divergent phenotypic patterns within the strains. The frequency of non-synonymous mutations was much greater than for synonymous mutations indicating an important role of positive selection acting on these TFs during diversification of the different strains. No significant connections were discovered between the distribution of DNA variation and phenotypic patterns.</p>

Identification of protein targets of nevirapine reactive metabolites using click chemistry and mass spectrometry-based differential proteomics

Abstract : Adverse drug reactions (ADRs) are undesirable effects caused after administration of a single dose or prolonged administration of drug or result from the combination of two or more drugs. Idiosyncratic drug reaction (IDR) is an adverse reaction that does not occur in most patients treated with a drug and does not involve the therapeutic effect of the drug. IDRs are unpredictable and often life-threatening. Idiosyncratic reaction is dependent on drug chemical characteristics or individual immunological response. IDRs are a major problem for drug development because they are usually not detected during clinical trials. In this study we focused on IDRs of Nevirapine (NVP), which is a non-nucleoside reverse transcriptase inhibitor used for the treatment of Human Immunodeficiency Virus (HIV) infections. The use of NVP is limited by a relatively high incidence of skin rash. NVP also causes a rash in female Brown Norway (BN) rats, which we use as animal model for this study. Our hypothesis is that idiosyncratic skin reactions associated with NVP treatment are due to post-translational modifications of proteins (e.g., glutathionylation) detectable by MS. The main objective of this study was to identify the proteins that are targeted by a reactive metabolite of Nevirapine in the skin. The specific objectives derived from the general objective were as follow: 1) To implement the click chemistry approach to detect proteins modified by a reactive NVP-Alkyne (NVP-ALK) metabolite. The purpose of using NVP-ALK was to couple it with Biotin using cycloaddition Click Chemistry reaction. 2) To detect protein modification using Western blotting and Mass Spectrometry techniques, which is important to understand the mechanism of NVP induced toxicity. 3) To identify the proteins using MASCOT search engine for protein identification, by comparing obtained spectrum from Mass Spectrometry with theoretical spectrum to find a matching peptide sequence. 4) To test if the drug or drug metabolites can cause harmful effects, as the induction of oxidative stress in cells (via protein glutathionylation). Oxidative stress causes cell damage that mediates signals, which likely induces the immune response. The results showed that Nevirapine is metabolized to a reactive metabolite, which causes protein modification. The extracted protein from the treated BN rats matched 10% of keratin, which implies that keratin was the protein targeted by the NVP-ALK. / Résumé : Les effets indésirables (EI) sont les effets indésirables causés après l'administration d'une dose unique ou une administration prolongée du médicament ou le résultat de la combinaison de deux médicaments ou plus. La Réaction idiosyncratique (IDR) est une réaction indésirable qui ne se produit pas dans la plupart des patients traités avec un médicament et qui ne comporte pas l'effet thérapeutique du médicament. IDR sont imprévisibles et peuvent mettre la vie du malade en danger. Cette réaction dépend des caractéristiques chimiques du médicaments et/ou de la réponse immunitaire individuelle du patient. IDR est un problème majeur pour le développement de médicaments car ils ne sont généralement pas détectés au cours des essais cliniques. Dans cette étude, nous nous sommes concentrés sur la Réaction idiosyncratique de névirapine (NVP) qui est un inhibiteur de transcriptase inverse non nucléosidique utilisé pour le traitement du virus d'immunodéficience humaine (VIH). L'utilisation de NVP est limitée par une incidence relativement élevée d'éruption cutanée. NVP provoque également une éruption cutanée chez les rats femelles de souche Brown Norway. Notre étude vise à mieux comprendre les IDRs induites par l'administration de NVP chez l'animal. La présente étude vise à vérifier l'hypothèse que les problèmes cutanés associés à la prise de NVP soient attribuables à la modification post-traductionnelle de protéines détactable par spectrométrie de masse. Les principaux objectifs de ce projet étaient : 1) Déterminer si la Nevirapine alcynes (NVP-ALK), un analogue de la NVP peut développer la même éruption cutanée que la NVP. La NVP-ALK a été couplé avec de la biotine en utilisant la réaction chimique (click chemistry). 2) Détecter les modifications post-traductionelles des proteines par Western blot et des techniques de spectrométrie de masse, pour comprendre le mécanisme de la toxicité induite par la NVP. 3) Identifier les protéines modifiées en utilisant le moteur de recherche MASCOT pour l'identification des protéines, en comparant le les spectres de masse obtenus avec les spectres théoriques pour trouver une séquence correspondante de peptide. 4) Tester si la NVP et ses métabolites peuvent provoquer des effets nocifs, comme l'induction d'un stress oxydatif dans les cellules (par la mesure de la glutathionylation des protéines). Les résultats ont montré que la névirapine est métabolisé en métabolite réactif ce qui provoque une modification de la kératine. Ainsi nos résultats suggèrent que la kératine est la cible des métabolites de la NVP-ALK.

Diseño, construcción y evaluación de repositorios estandarizados con calidad de datos asegurada para la monitorización de la atención a la alimentación infantil

[ES] El establecimiento de la Historia Clínica Electrónica (HCE) como fuente de conocimiento en el ámbito clínico es una cuestión clave para las organizaciones sanitarias para el desarrollo de la investigación, docencia y la mejora de la calidad asistencial. El actual despliegue de los sistemas de información sanitarios supone que los datos de HCE sean una excelente fuente de datos candidata con la que generar repositorios para estos propósitos, debido a su cobertura poblacional y continuidad en el tiempo. Para garantizar la validez y fiabilidad de la reutilización de datos de HCE para investigación y monitorización de la asistencia sanitaria, es necesario evaluar y asegurar su calidad de datos. Así como utilizar estándares de información sanitaria para dar soporte a la continuidad asistencial. Esta tesis tiene como propósito diseñar un proceso de construcción de repositorios, estandarizados y con calidad de datos asegurada, con el que construir un repositorio de datos integrados (IDR) de alimentación infantil a partir de datos multi-fuente de HCE. El objetivo de este IDR es habilitar el re-uso de datos para la monitorización de la atención a la alimentación infantil hasta los dos años, centrado en la lactancia materna como uno de los principales determinantes de la salud materno-infantil. Para la gestión de la calidad de datos se ha utilizado un marco conceptual basado en dimensiones de calidad de datos para una gestión de calidad de datos total del proceso. Como estándares de información sanitaria se han utilizado UNE-EN-ISO 13940 (Contsys) y UNE-EN-ISO 13606. Para la monitorización se han utilizado los indicadores definidos por la Organización Mundial de la Salud para la evaluación de estrategias internacionales de alimentación infantil como la Iniciativa para la Humanización de la Asistencia al Nacimiento y la Lactancia (IHAN). El proceso de construcción de repositorios ha estado estrechamente relacionado con la estrategia "Hasta Que Tú Quieras (HQTQ)", desarrollada en el Área de Salud V-Altiplano del Servicio Murciano de Salud para mejorar la calidad de la atención a la alimentación al lactante. Las especificaciones y requerimientos del proceso asistencial de atención a la alimentación del lactante guiaron la definición de los modelos de información. El proceso de construcción ha sido aplicado sobre real world data multi-fuente proveniente de la HCE del Área V-Altiplano. En concreto del sistema de información hospitalario de su hospital de referencia, Hospital Virgen del Castillo y del sistema de información de Atención Primaria de los 3 centros de salud del área, durante el periodo 2009-2018. La aplicación de dimensiones de calidad de datos sobre procedimientos de evaluación y aseguramiento de calidad de datos, junto con el uso de conceptos de la norma Constys, han contribuido al diseño del proceso de construcción de repositorios estandarizados con calidad de datos asegurada. La aplicación de los métodos del proceso de construcción de repositorios sobre los datos de la HCE perinatal del hospital y del seguimiento de alimentación de los centros de atención primaria han contribuido a la creación del IDR, con calidad de datos asegurada y estandarizado según la norma Contsys; así como a la estimación robusta de indicadores de alimentación infantil y la habilitación de un sistema de monitorización poblacional y continuo a partir de datos de HCE. La experiencia de integración de los repositorios materno-infantiles del Hospital Virgen del Castillo y el Hospital 12 de Octubre de Madrid ha contribuido a la creación de un IDR estandarizado conforme a la norma UNE-EN-ISO 13606 y a una propuesta de arquetipos para integración de repositorios materno-infantiles a nivel nacional. The current deployment of health information systems means that EHR data are an excellent candidate data source with which to generate repositories for these purposes, due to their population coverage and continuity over time. To ensure the validity and reliability of the reuse of EHR data for research and healthcare monitoring, it is necessary to assess and assure its data quality. As well as using health information standards to support continuity of care. The purpose of this thesis is to design a standardized and DQ assured repository construction process to build an Integrated Data Repository (IDR) for infant feeding from multi-source EHR data. The goal of this IDR is to enable the reuse of data for monitoring infant feeding care up to two years of age, focusing on breastfeeding as one of the main determinants of maternal and child health. For DQ management, a conceptual framework based on DQ dimensions has been used for a total data quality management of the process. UNE-EN-ISO 13940 (Contsys) and UNE-EN-ISO 13606 have been used as health information standards. The "Indicators for assessing infant and young child feeding practices" (IYCF), defined by the World Health Organization were used for monitoring. This set of indicators is the current reference for the evaluation of international infant feeding strategies such as the Baby Friendly Hospital Initiative (BFHI). The construction process of repositories has been closely related to the "Hasta Que Tú Quieras (HQTQ)" strategy, developed in the Health Area V-Altiplano of the Servicio Murciano de Salud to improve the quality of care for infant feeding. The specifications and requirements of the infant feeding care process guided the definition of information models. The construction process has been applied on real world multi-source data from the EHR of the Health Area V-Altiplano. Specifically, from the hospital information system of its reference hospital, Hospital Virgen del Castillo, and from the primary care information system of the 3 health centers of the area, during the period 2009-2018. The application of DQ dimensions on DQ assessment and assurance procedures, together with the use of concepts of the Contsys standard, have contributed to the design of the process of building standardized and DQ assured repositories. The application of the methods of the repository construction process on perinatal EHR data from the hospital along with the EHR data from the area's primary care information system for monitoring infant feeding has contributed to the creation of the IDR, with DQ assured and standardized according to the Contsys standard; As well as for the robust estimation of infant feeding indicators and the implementation of a population-based and continuous monitoring system based on EHR data. The experience of integrating the mother-child repositories of the Virgen del Castillo Hospital and the 12 de Octubre Hospital in Madrid has contributed to the creation of a standardized IDR in accordance with the UNE-EN-ISO 13606 standard and a proposal of archetypes for the integration of mother-child repositories at the national level. The quality information obtained from the IDR has contributed to obtaining the Seal of Good Practices of the Ministry of Health, Social Services and Equality and to the scaling-up of the perinatal information model to the whole Servicio Murciano de Salud. / García De León Chocano, R.
