ProduÃÃo e purificaÃÃo parcial de IgY anti-lectina de sementes de Canavalia brasiliensis (ConBr) (Leguminosae) em galinhas poedeiras / Production and partial purification of IgY anti- lectin of Canavalia brasiliensis seeds (ConBr) (Fabaceae) in laying hens

nÃo hà / Lectins are a group of proteins present in various living organisms, particularly in plants and especially in Leguminosae seeds. Among this family, a species that stands out for having large amount of lectin in the seed is the Canavalia brasiliensis. One approach for the investigation of lectins on biological systems is the detection there of by techniques involving antibody. Such techniques may include from classical immunochemical assays (immunodiffusion, immunoelectrophoresis and rocketimunoeletroforese) to ELISA experiments and advanced microscopy techniques. In this sense, antibodies become biotechnological tools invaluable since they are capable of specifically recognizing epitopes of the target molecules. Immunoglobulins (Igs) are proteins present in high concentration in the blood plasma of mammals. They are vectors of humoral immunity, with the primary function to join the foreign antigens to the individual so as to neutralize them. In the case of poultry the main immunoglobulin class (IgYs) can be found not only in blood, but also in egg yolk. This fact enables fast and efficient purification of these molecules facilitating its use as biotechnological inputs. Thus, this study aimed to produce antibodies against the lectin extracted from Canavalia brasiliensis seed (ConBr) (Fabaceae) in laying hens, implementing production methodology of new biotechnological inputs for the study of lectins. For this laying hens were immunized with 200 or 400μg of lectin Canavalia brasiliensis (ConBr) for 15 weeks at 10-day intervals. Collected eggs from vaccinated birds had their isolated and precipitated yolk in 0-30% fraction with ammonium sulfate for partial purificaÃÃoo of IgYs. Confirmation of the presence of antibodies was made by immunodiffusion assays agarose gel. The fraction showed 0-30%, from the first week after the immunization, capable of recognizing ConBr IgYs. This profile appeared in all groups over the 15-week experiment. The production of specific IgYs against the lectin from Canavalia brasiliensis (ConBr) proved feasible and remained throughout the experiment, possibly in reasonable quantities. In this sense, IgYs produced in this work provide a powerful biotechnological tool to be made available for future studies. / As lectinas constituem um grupo de proteÃnas presentes em diversos organismos vivos, sobretudo em vegetais e especialmente em sementes de Leguminosae. Dentre essa famÃlia, uma espÃcie que se destaca por possuir grande quantidade de lectina na semente à a Canavalia brasiliensis. Uma das possÃveis abordagens para a investigaÃÃo de lectinas em sistemas biolÃgicos à a detecÃÃo das mesmas atravÃs de tÃcnicas que envolvam anticorpos. Tais tÃcnicas podem incluir desde ensaios clÃssicos de imunoquÃmica (imunodifusÃo, imunoeletroforese e rocketimunoeletroforese), atà experimentos de ELISA e tÃcnicas avanÃadas de microscopia. Neste sentido, os anticopos tornam-se ferramentas biotecnolÃgicas de grande valia uma vez que sÃo capazes de reconhecer especificamente epÃtopos das molÃculas alvo. As imunoglobulinas (Igs) são proteínas presentes em grande concentração no plasma sanguÃneo de mamÃferos. São os vetores da imunidade humoral, tendo como função principal unir-se aos antígenos estranhos ao indivÃduo, de modo a neutralizá-los. No caso das aves a principal classe de imunoglobulinas (IgYs) pode ser encontrada nÃo somente no sangue mas tambÃm nas gemas dos ovos. Esse fato permite purificaÃÃo rÃpida e eficiente dessas molÃculas facilitando seu uso como insumos biotecnolÃgicos. Dessa forma, este trabalho teve por objetivo produzir anticorpos contra a lectina extraÃda da semente de Canavalia brasiliensis (ConBr) (Leguminosae) em galinhas poedeiras, implementando metodologia de produÃÃo de novos insumos biotecnolÃgicos para o estudo de lectinas. Para tanto galinhas poedeiras foram imunizadas com 200 ou 400μg da lectina de Canavalia brasiliensis (ConBr) por 15 semanas em intervalos de 10 dias. Ovos coletados das aves imunizadas tiveram suas gemas isoladas e precipitadas na fraÃÃo 0-30% com sulfato de amÃnio para a purificaÃÃo parcial das IgYs. A confirmaÃÃo da presenÃa dos anticorpos foi feita por ensaios de imunodifusÃo em gel de agarose. A fraÃÃo 0-30% apresentou, a partir da primeira semana apÃs a imunizaÃÃo, as IgYs capazes de reconhecer ConBr. Esse perfil se apresentou em todos os grupos ao longo das 15 semanas de experimento. A produÃÃo das IgYs especÃficas contra a lectina de Canavalia brasiliensis (ConBr) se mostrou viÃvel e permaneceu durante todo experimento, possivelmente em quantidades razoÃveis. Neste sentido, as IgYs produzidas nesse trabalho constituem uma potente ferramenta biotecnolÃgica a ser disponibilizada para estudos futuros.

Trypanosoma evansi is a protozoan flagellate that affect cattle, sheep, goats, donkeys, cats and pigs. The disease is particularly important in horses, known as "Mal das Cadeiras", due to deficits characteristic of locomotion in animals infected. The disease has a worldwide distribution, with several reports of natural infections in different regions in Brazil. Several pharmaceutical drugs have been recommended for the prophylaxis and control of the parasite, but over the years, these drugs have lost their effectiveness and protozoan looks developing resistance. The production and use of avian antibodies has aroused great interest in the scientific community because of the diversity of diagnostic and therapeutic applications. Due to the phylogenetic distance, immune diversification mechanisms and transferability of serum immunoglobulin to the egg yolk, currently recognizes a number of advantages to using avian antibodies rather than antibodies of mammals. This study produced a specific immunoglobulin against Trypanosoma evansi from immunization of chickens using an isolate of the parasite. After extraction and purification from egg yolk, cytotoxicity and genotoxicity of these antibodies in cultures of human leukocytes were evaluated. The therapeutic efficacy was tested in Rattus norvegicus. The trial also assessed the use of avian antibody associated with imidocarb dipropionate and diaceturate of diminazene. The toxicity tests showed that immunoglobulin does not produce gene and cell injury. In vivo tests showed increased in longevity and pre-patent period, mainly in rats treated with avian immunoglobulin associated with pharmacological treatment. The immunization and the extraction protocol used in this study allowed obtaining specific antibodies against protozoan and innocuous to the host. Moreover, when administered for therapeutic purposes, strengthens the immune system and prolongs the survival of infected patients. / Trypanosoma evansi é um protozoário flagelado que acomete bovinos, ovinos, caprinos, asininos, felinos e suínos. A enfermidade tem especial importância em equinos, sendo conhecida como mal das cadeiras , devido aos déficits de locomoção característicos em animais infectados. A doença tem distribuição mundial, com vários relatos de infecções naturais em diversas regiões no Brasil. Várias drogas terapêuticas têm sido recomendadas para a profilaxia e controle do protozoário, porém ao longo dos anos estes medicamentos tem perdido a eficácia e o protozoário parece desenvolver resistência. A produção e uso de anticorpos aviários tem despertado grande interesse na comunidade científica devido à diversidade de aplicações diagnósticas e terapêuticas. Devido à distância filogenética, mecanismos de diversificação imune e capacidade de transferência das imunoglobulinas séricas para a gema do ovo, atualmente reconhece-se uma série de vantagens ao utilizar anticorpos aviários ao invés de anticorpos de mamíferos. Neste estudo foi produzido uma imunoglobulina específica contra o T. evansi a partir de imunização de galinhas usando um isolado do protozoário. Após a extração e purificação a partir da gema do ovo, foram avaliadas a citotoxicidade e genotoxicidade destes anticorpos em cultura de leucócitos humanos. A eficácia terapêutica foi testada em Rattus norvegicus. O ensaio também avaliou o uso do anticorpo aviário associado ao dipropionato de imidocarb e diaceturato de diminazene. Os testes de toxicidade mostraram que a imunoglobulina não produz lesões celulares e genéticas. Testes in vivo mostraram aumento da longevidade e período pré-patente, principalmente em ratos tratados com a imunoglobulina aviária associada ao tratamento farmacológico. O protocolo de imunização e extração usados nesta pesquisa permitiu a obtenção de anticorpos específicos contra o protozoário e inócuo para o hospedeiro. Além disso, ao ser administrado com fins terapêuticos, reforça o sistema imunológico e prolonga a sobrevida de pacientes infectados.

Ginkgo biloba ekstrakto adjuvantinis efektas, indukuojant specifinį imuninį atsaką prieš jaučio serumo albuminą vištų bei triušių modeliuose Adjuvantai – tai yra neorganiniai ir organiniai junginiai, kurie stimuliuoja imuninę sistemą ir skatina sistemos atsaką prieš pageidaujamą antigeną, kurio imunogeniškumas savaime yra menkas. Siekiant sustiprinti organizmo atsaką į pateikiamą antigeną šie junginiai dažnai įtraukiami į vakcinų sudėtį. Šiame darbe mes įvertinome augalinio Dviskiaučio ginkmedžio (DG) ekstrakto adjuvantines savybes triušių bei vištų modeliuose. Abiejų klasių gyvūnai buvo imunizuojami per gleivines jaučio serumo albuminu. Adjuvantinis efektas buvo vertinamas pagal IgA titrus seilėse, IgG ir IgM titrus gyvūnų serume ir IgY titrus vištų kiaušinio trynyje. Gyvūnuose, imunizuotuose panaudojant DG kaip adjuvantą, pasireiškė stiprus sisteminis imuninis atsakas, ypač gleivinių (IgA pagrindu) ir humoralinis (IgG ir IgM pagrindu) palyginus su imunizacijomis be adjuvanto. Taip pat paaiškėjo, kad naudojant nepilną Freundo adjuvantą, humoralinis imuninis atsakas yra stipriausias. Įvertinus IgA titrus paaiškėjo, imuninio atsako indukcija vištų modelyje didesnė, negu triušių modelyje. IgG titrai vištų ir triušių serumuose buvo panašūs, ir jų produkcija išliko stabili 8 savaičių laikotarpyje. IgY vertė buvo nustatinėjama vištų kiaušinio trynyje; IgY lygis išliko stabilus ir panašus į IgG atsaką per visą periodą. Skirtingai nuo IgA ir IgY, IgM buvo nustatyti jau po antros... They are often included in vaccines to enhance the recipient's immune response to a supplied antigen. In present study we evaluated the adjuvant capacity of a plant Ginkgo biloba (GB) extract upon the mucosal immunization of rabbits and hens with bovine serum albumin. Adjuvant efficacy was evaluated by measuring the titres of IgA in saliva, IgG and IgM immunoglobulins in serum of the animals and IgY in chicken egg yolk. Animals immunized using GB adjuvant showed strong cellular (IgA-based) and humoral (IgG and IgM) immune responses when compared to immunisations without adjuvant. It also became clear that the the immune response using Freund’s adjuvant was highest in humoral immune response. GB extract induced more important immune response in chickens when compared to rabbits as measured by IgA titres. The titres of IgG in hens and rabbits were found to be at similar levels and their production remained stable during 8 weeks. Low cost, high volume production, recognition proteins highly conserved in mammals, in addition to preventing the suffering and death of animals for experimentation, are the main advantages. The aim of our study was to develop, standardize and deploy the technology IgY for aviaries production of antibodies with aim to obtain IGY-specific forms to promastigotes of Leishmania (Leishmania) amazonensis. Initially, matching was performed by two methods of precipitation and the comparison between them, to obtain the pure IgY from the yolk of egg white obtained in trade. The method of purification employing polietienoglicol (PEG-6000) was able to extract IgY with purity comparable to commercial standard IgY (Sigma) and more than obtained by the method with 25% ammonium sulfate. Later, chickens were inoculated with three doses of inactivated antigen (promastigotes forms of L. amazonensis) mixed with incomplete Freund's adjuvant, at 0, 14 and 21 days, IM route. High yield of total and specific IgY was obtained, as determined by the methods of Bradford and ELISA, respectively. The IgY antibodies showed high specificity (indirect immunofluorescence), sensitivity and avidity (ELISA) for promastigotes forms of L. amazonensis, from the second immunization. The level of IgY-specific antibodies remained high until the end of the analysis, around 56 days. Based on the results we can conclude that the "Technology IgY" allows to obtain antibodies with high specificity and sensitivity, in ethics, can be used successfully in various fields of science. / A produ??o de anticorpo espec?fico (IgY) em galinhas (Gallus gallus domesticus) e sua purifica??o a partir da gema do ovo vem atraindo o interesse da comunidade cient?fica devido as in?meras vantagens sobre os anticorpos IgG de mam?feros. A New Immunoassay for Quantification of a Novel Cancer Antigen in Serum and Immunostaining of Carcinoma Tissues and Cultured Cells Revealing the Antigenic Cellular Location.

The purpose of this study is 1) to examine the presence of the antigen in serum by employing a newly developed ELISA immunoassay that quantifies the total antigen and bound antigen (antigen-antibody complex) using polyclonal chicken antibodies directed against an IgM-binding epitope of the new antigen, and 2) to determine the location of the antigen in carcinoma and normal cells. Sera from healthy volunteers (n = 147) and cancer patients (n = 26) were compared for both bound and total antigen concentrations using the new ELISA. Healthy volunteers were subdivided into three groups: those with a personal history of cancer (n = 13), those with no personal or family history of cancer (n = 36) and those with a family history of cancer (n = 97). Ovarian, breast, colon carcinoma tissues and their normal counterparts and cultured ovarian and prostatic carcinoma cells were subjected to immunofluorescence using IgY antibodies and goat anti-chicken fluorescent secondary antibodies. Basic imaging was performed on tissue sections while confocal microscopy was performed on cultured cells. Furthermore, immunohisto-chemical staining using an anti-chicken HRP-conjugated secondary antibody was performed on 16 normal ovarian tissues, 53 ovarian adenocarcinomas, and 3 borderline ovarian tumors. Statistical analysis revealed significant differences in cancer patients' bound and total antigen levels compared to that of healthy volunteers (p < 0.005). Bound and total antigen levels of cancer patients were also significantly higher than those of the healthy volunteers with no personal or family history of cancer and those with a family history of cancer (p < 0.01). However, no significant difference existed between the bound (p > 0.120) and total antigen levels (p > 0.076) of cancer patients and patients with a personal cancer history. Immunohistochemical staining of ovarian tissues revealed a significant difference in the lumenal staining of the carcinomas compared to that of the normal ovarian tissues. Furthermore, fluorescence imaging revealed that the antigen is localized to the cell membranes of the carcinoma cells but is absent from the normal tissues. Confocal microscopy further emphasized the antigen's association with the membrane and also revealed some filamentous cortical staining.

The illness is characterized by an acute bacteremic phase of approximately one week, followed by an immune phase in which specific antibodies are found in blood and leptospires are eliminated in urine. Clinical signs in the initial phase of leptospirosis can be confused with other feverish diseases, making laboratory diagnosis of extreme importance to start antibiotic treatment. The existing laboratory tests for early diagnosis of leptospirosis are based on IgM detection and are of low sensitivity. Thus, there is an urgent need for development of new diagnostic strategies for use in the acute phase of leptospirosis. In the present work monoclonal antibodies (MAbs) and polyclonal IgY were used in the standardization of three different antigen capture ELISA formats for direct detection of leptospires in blood during the acute phase of the disease. Detection limit of leptospires in human sera experimentally contaminated ranged from 10 5 to 10 7 cells per millilitre in the different formats. The ELISA format with the best performance was able to detect 10 5 leptospires per millilitre of human sera, using as capture antibody a MAb against LipL32, the major outer membrane protein of pathogenic leptospires, and as detection antibody biotinilated policlonal IgY against a pathogenic serovar of Leptospira interrogans. Although this format did not present the adequate sensitivity for detection of circulating leptospires at the levels existing in blood in the initial phase of the disease, it can be improved in order to do so. / A leptospirose é uma doença infecciosa causada por espiroquetas do gênero Leptospira que pode afetar diversos órgãos vitais como pulmões, fígado e rins. A doença caracteriza-se por uma fase inicial bacterêmica de aproximadamente uma semana, seguida de uma fase imune na qual anticorpos específicos são detectados no sangue e leptospiras são eliminadas na urina. Os sinais clínicos da fase inicial da leptospirose podem ser confundidos com diversas doenças febris, o que torna o diagnóstico laboratorial extremamente importante para decidir sobre o início do tratamento com antibióticos. Os testes laboratoriais existentes para o diagnóstico na fase inicial da doença são todos baseados na detecção de IgM e apresentam baixa sensibilidade. Assim, há uma necessidade urgente de desenvolvimento de novas estratégias de diagnóstico precoce da leptospirose. Neste trabalho foram utilizados anticorpos monoclonais (MAbs) e anticorpo policlonal (IgY) para padronizar três formatos de ELISA de captura de antígeno para uso na detecção direta de leptospiras no sangue durante a fase aguda da doença. O limite de detecção de leptospiras em soro humano experimentalmente contaminado variou entre 10 5 e 10 7 células por mililitro nos diferentes formatos. O formato que apresentou melhor desempenho detectou 10 5 leptospiras por mililitro de soro, e utilizou como anticorpo de captura um MAb contra LipL32, a principal proteína de membrana externa de leptospiras patogênicas, e como anticorpo de detecção IgY biotinilada, produzida contra um sorovar patogênico de Leptospira interrogans. Embora este formato não apresente ainda a sensibilidade adequada para detectar o nível de leptospiras circulantes no sangue na fase inicial da doença, ele possui potencial para ser aperfeiçoado de forma a atingir aquele nível.

Cystic fibrosis (CF) is one of the relatively common inherited diseases caused by a mutation in the gene encoding for CFTR protein, which forms a chloride channel that significantly affects ion homeostasis and the associated fluid management of the cell. This disease mainly affects the respiratory and digestive systems, being the most life-threatening in the respiratory tract. Patients with CF suffer from frequent and recurrent respiratory infections that lead to the development of chronic inflammation and gradual destruction of lung tissue. These lung infections, which are caused mostly by the opportunistic pathogen Pseudomonas aeruginosa, are the most common cause of morbidity and mortality in these patients. At present, antibiotics are used in the treatment of Pseudomonas aeruginosa infections, but new methods of antibacterial therapy need to be found to overcome the development of resistance. In addition to active immunization of CF patients against Pseudomonas aeruginosa, their passive immunization with specific chicken antibodies directed against this pathogen offers promising possibilities. Os organofosforados são agrotóxicos responsáveis por 70 por cento das intoxicações ocupacionais e atuam inibindo a enzima acetilcolinesterase por meio da fosforilação do aminoácido serina do sítio ativo. Existem diversos métodos de diagnóstico para monitorar a exposição a estes compostos, tais como: identificação e análise quantitativa de compostos livres noplasma; identificação e análise quantitativa de metabólitos na urina; determinação da atividade de colinesterases no plasma ou no sangue total. Entretanto, estas metodologias possuem algumas limitações, como por exemplo, a falta de especificidade, tecnologias de análise caras e variações interindividuais (sexo, idade, etc.). Uma tentativa de contornar estas limitações na quantificação da atividade de colinesterases é o desenvolvimento de metodologias baseadas na detecção da enzima fosforilada através de técnicas imunoquímicas. Este estudo teve como objetivo desenvolver um método de imunoensaio para o diagnóstico rápido da exposição humana aos organofosforados pela detecção deacetilcolinesterase inibida no sangue humano por anticorpos aviários produzidos contra uma sequência de aminoácidos ao redor da serina fosforilada do centro catalítico da enzima. Para isto, foram adquiridos dois decapeptídeos sintéticos correspondentes ao sítio catalítico "não fosforilado" (SOH) e outro com o resíduode serina fosforilado (SOP). Estes peptídeos foram conjugados com albumina bovina de soro (BSA), injetados em galinhas poedeiras da raça Isa-Brown e anticorpos IgY foram obtidas das gemas dos ovos das galinhas imunizadas. A especificidade dos anticorpos IgY anti-SOH e anti-SOP foi testada por meio de métodos ELISA constatando-se que estes anticorpos reconhecem os peptídeos livres, os peptídeos conjugados com BSA e a própria BSA. Além disso, ocorreu uma significativa reação cruzada, ou seja, os anticorpos IgY anti-SOH e anti-SOP não conseguiram diferenciar os peptídeo SOH e SOP. Estes resultados não permitem a indicação destes anticorpos para o desenvolvimento de métodos para diagnóstico de intoxicação. / Pesticides are chemical substances used to control plagues and diseases in plantations. Organophosphates are the pesticides involved in 70% of the occupational intoxications and act inhibiting the enzyme acetylcholinesterase by phosphorylation of the serine residue of catalytic centre. There are several methods for monitoring the exposition to these compounds, as so: identification and assay the free compounds into plasma; identification and assay metabolites into urine; cholinesterases assay into plasma or blood. However, these methods have some limitations, as for example lack of specificity, expensive analytical technologies and inter-individual variety (sex, age, etc.). One way to outline these limitations on cholinesterase assay is the development of methods based on phospho-enzyme detection by imunochemical techniques. This study have had the objective to developing an imuno-assay method for rapid diagnosis of human exposition to organophosphorus compounds by imuno-detection of inhibited acetylcholinesterase using aviary antibodies produced against a peptide sequence of the catalytic centre around phospho-serine residue. Two synthetic decapeptides were bought: one with the sequence of "not-phosphorylated" catalytic centre (SOH) and another with a serine-phosphorylated residue. These peptides were conjugated with bovine serum albumin (BSA), injected into Isa-Brown hens and antibodies IgY were isolated from egg yolk of immunized hens. The specificity of IgY anti-SOH and anti-SOP was tested using ELISA methods showing that these antibodies recognize free peptides, conjugated peptides and BSA. In addition, there was a significant cross reaction, or in other words, the antibodies IgY anti- SOH and anti-SOP failed on differentiate the peptide SOH and SOP. These results do not allow the indication of these antibodies for developing methods for intoxication diagnosis.

A prevalência da infecção tem correlação com precárias condições de higiene e saneamento. Em países em desenvolvimento, um saneamento inadequado resulta em maior transmissão desta doença, principalmente entre crianças e jovens. Atualmente, devido às melhorias das condições sanitárias, o perfil epidemiológico da doença está se deslocando para idades mais avançadas, o que facilita a ocorrência de surtos epidemiológicos. Os kits comerciais para detecção de anti-HAV total normalmente utilizam imunoglobulina G (IgG) de mamíferos no período convalescente da doença para a produção dos anticorpos de captura e do conjugado. Uma alternativa à aplicação dos anticorpos de mamíferos no diagnóstico é o uso da imunoglobulina Y (IgY), encontrada no soro e gema dos ovos de aves e répteis. Essas proteínas têm varias vantagens quando comparadas com IgG: alta resposta contra antígenos de mamíferos, redução da cor de fundo em ensaios imunoenzimáticos e de serem obtidas por um método não-invasivo (coleta da gema dos ovos). O objetivo deste trabalho foi a obtenção de anticorpos IgY anti-HAV produzidos em galinhas imunizadas contra o vírus da Hepatite A (HAV) e o desenvolvimento de um ensaio imunoenzimático para detecção de anti-HAV total utilizando IgY anti-HAV como imunoglobulinas de captura e conjugado. Cinco grupos de galinhas foram imunizadas com diferentes inóculos contendo: vacina com e sem o adjuvante CpG-ODN, HAV com adjuvante incompleto de Freund (IFA) com e sem o adjuvante CpG-ODN e um grupo controle com IFA. Os ovos foram coletados e a gema foi purificada pela precipitação com polietileno glicol. A solução purificada contendo IgY anti-HAV foi avaliada para determinação da concentração da IgY anti-HAV por espectrofotometria e sua especificidade e título foram determinados à partir de um teste imunoenzimático. Os anticorpos foram conjugados com a peroxidase e foi estabelecida a diluição ideal para os anticorpos de captura e conjugado. Para avaliar o ensaio imunoenzimático “in-house” com IgY anti-HAV, foi avaliado um painel composto de 100 amostras positivas e 100 amostras negativas para anti- HAV-total. A presença da IgY anti-HAV nas gemas dos ovos foi confirmada por SDS-PAGE e Western Blotting, e após a purificação, a média da concentração de proteínas nas gemas dos ovos foi de 8,7406 mg /mL. O grupo imunizado com HAV, IFA e CPG-ODN apresentou os maiores títulos de anticorpo. O ensaio “in-house” apresentou sensibilidade de 84%, especificidade de 79% e eficiência de 81,5%. Os métodos utilizados para a produção de IgY anti-HAV e sua conjugação com peroxidase foram eficientes e o ensaio imunoenzimático “in-house” IgY anti-HAV demonstrou uma boa sensibilidade e especificidade. A produção de anti-HAV IgY apresenta vantagens quando comparado com obtenção da IgG anti-HAV. O teste imunoenzimático “in house” com IgY anti- HAV pode ser uma alternativa a utilização da IgG nos ensaios imunoenzimáticos. / Hepatitis A is an endemic disease in Brazil and Latin America. Prevalence of this infection is related to the low degree of hygiene and sanitation. In developing countries, inadequate sanitation results in larger transmission of the disease mostly in children and young people. Nowadays, due to better sanitation conditions, the epidemiological profile of disease is changing to older ages resulting in the occurrence of outbreaks. Diagnostic kits for detection of total anti-HAV generally use mammals immunoglobulin G (IgG) in the convalescent period of disease for production of capture and conjugated antibodies. One alternative to the application of mammals antibodies in the diagnosis is the use of immunoglobulin Y (IgY), encountered in birds and reptiles. These proteins have several advantages when compared to IgG: high response against mammals antigens, reduction of the background in imunoenzymatic assays and it is obtained by a non-invasive method (harvest of the egg yolks). The objective of this work was the acquisition of anti-HAV IgY antibodies produced in immunized chickens against Hepatitis A virus and the development of an immunoenzymatic assay for total anti-HAV detection using IgY anti-HAV as capture and conjugated immunoglobulins. Five groups of chickens were immunized with different inocula containing: vaccine with and without CpG-ODN adjuvant, HAV with incomplete Freund adjuvant (IFA) with and without CpG-ODN and one control group with IFA. The eggs were harvested and the yolk was purified by precipitation with polyethylene glycol. The purified solution containing anti-HAV IgY was evaluated by espectrofotometry and their specificity and title were determined by an immunoenzymatic assay. These antibodies were conjugated with peroxidase and was estabilized the ideal dilution for capture and conjugated antibodies. For evaluation of the immunoenzymatic “in-house” assay with IgY anti-HAV, a panel composed of 100 positive samples and 100 negative samples for total anti-HAV was assessed. The presence of IgY anti-HAV in egg yolks was established by SDS-PAGE and Western Blotting, and after the purification, the average of the proteins concentrations in the egg yolks was of 8,7406 mg/mL. The group immunized with HAV, IFA and CpG-ODN demonstrate the higher titer of antibodies. The “in-house” assay showed sensibility of 84%, specificity of 79% and efficiency of 81,5%. The methods used for anti-HAV IgY production and conjugation with peroxidase were efficient and the “in-house” immunoenzymatic assay IgY anti-HAV demonstrated a good sensitivity and specificity. The production of IgY anti-HAV showed advantages when compared to the acquisition of IgG anti-HAV. The immunoenzymatic “in-house” assay IgY anti- HAV can be an alternative to the utilization of IgG in immunoenzymatic assays.
