Em algumas espécies de peixes tropicais, como o pacu (Piaractus mesopotamicus) e matrinxã (Brycon amazonicus), estudos observaram o papel da vitamina C como imunoestimulante avaliando a hematologia dos peixes como indicador das alterações imunológicas. O número de leucócitos e de trombócitos é considerado importante indicador de seu estado de saúde. Entretanto, não são conhecidos estudos que avaliem outros parâmetros imunológicos bem como os efeitos do glicano nas respostas imunológicas inatas de peixes tropicais. O pacu, por ser uma espécie onívora, de fácil cultivo e adaptada a ampla variedade de alimentos, tem grande importância na piscicultura brasileira e por esta razão foi escolhido como modelo experimental nesta pesquisa. Le clade des invertébrés cristallise en 2017 de grandes problématiques sociétales à la fois économiques et sanitaires. En effet un certain nombre des organismes présent dans ce groupe phylétique sont des vecteurs des grandes pandémies infectieuses telles que le paludisme (Anopheles sp), Zika, Chinkungunya, Fièvre jaune, etc (Aedes sp), Chagas (Triatoma sp) ou encore la bilharziose (Biomphalaria sp, Bulinus sp). La compréhension du système immunitaire de ces organismes vecteurs doit aider la communauté scientifique à proposer des solutions pour réduire la transmission de toutes ces maladies sur le terrain. Biomphalaria glabrata est le vecteur unique de la Bilharziose intestinale (Schistosomamansoni) en Amérique Latine. Depuis un premier cas de résistance induite par une première infection en 1998, de nombreux travaux ont exploré la réponse immunitaire mémoire innée de cet escargot tropical d’eau douce. Dans le cadre de ce travail de thèse, différents aspects de cette immunité (également appelé priming, résistance acquise) ont été explorés, de la mise en place phénotypiques, aux bases moléculaires et cellulaires. En premier lieu,nous avons pu démontrer qu’elle était dépendante d’une bascule phénotypique (d’une réponse cellulaire d’encapsulation à une réponse humorale) et transcriptomique qui lui permet de mieux répondre lors d’une seconde infection. La spécificité de cette réponse est portée par la production de répertoire complexe de récepteurs et d’effecteurs immunitaire spécifiques qui sont capables de différencier jusqu’aux différents stades de développement parasitaire d’une même espèce de parasite. Nous avons également pu montrer que cette interaction dépendait de microARN circulants ainsi que de Biomphalysines, des ß-PFT acquises par transferts horizontaux depuis le monde bactérien. Enfin, cette résistance semble posséder une proximité avec l’immunité mémoire entraînée des cellules immunitaires innées des vertébrés en particulier sur la base des mécanismes moléculaires sous-jacent qui seraient liés chez Biomphalaria comme chez les Vertébrés à unereprogrammation épigénétique des cellules du système immunitaire innée. / Invertebrates focus in 2017 among the major economical and societal issuesacross Earth. Some members are vectors of important infectious pandemic as malaria(Anopheles sp), Zika, Chinkungunya, Yellow fever, etc (Aedes sp), Chagas (Triatoma sp) andtrematodes (Biomphalaria sp, Bulinus sp). Comprehension of immune system of thesevectors has to help scientist to decrease transmission on endemic area. Biomphalariaexposed first failure to be reinfected following first infection as soon as 1998. In my thesiswe explore this immune priming (innate immune memory) and describe an immune shiftfrom cellular to humoral immune response both in phenotype and transcriptomic response.A specificity is handle by specific immune receptor and effector repertoire to distinguish upto different developmental stage of same parasite species. This interaction is alsodependent of mRNAs and Biomphalysin, a ß-PFT coming from bacterial kingdom. Finally,this resistance seems to look alike the trained immune memory of innate cells in vertebrates.

As well as others members of Lepidoptera order, the D. saccharalis has a fast and efficient immune innate response, characterized by primary defense barrier, cellular immune response and humoral immune response. The antimicrobial peptides (AMPs) are included in humoral immune response that show cationic and amphiphilic characteristics besides its low molecular weight, what makes them potentially therapeutic agents. In this study were analyzed proteins with low molecular weight differently expressed in the hemolymph of D. saccharalis, after the interval of six and twelve hours of humoral immune response induction with different microorganisms compared to non-challenged worms (control). The 5th instar worms were divided in groups (n= 50) and submitted to six and twelve hours of septic challenges: control (group 1), challenged by Bacillus subtilis ATCC 6623 (group 2), challenged by Escherichia coli ATCC 11229 (group 3), and challenged by Beauveria bassiana 88 strain (group 4). In each worm was inoculated a known concentration of microorganism, and after the established time, the hemolymph was collected. The low molecular weight proteins were obtained submitting each hemolymph sample to an extraction solution containing Methanol, Acetic Acid and Water. After that, the protein extracts were concentrated in columns and protein dosage was realized in 280 nm. About 500 μg of proteins were submitted to two-dimensional electrophoresis (2-DE). The spots were excised from gels, digested with Tripsin and submitted to MALDI-ToF/ToF type mass spectrometry. The identification of protein orthologs was realized using the obtained data against the data available on the MASCOT online server (Matrix science), with the database (NCBI and Swiss rot) specified for Drosophila melanogaster and an internal database of Lepidoptera. The TagIdent tool was utilized for searching proteins through their molecular weight and isoelectric point. It was possible to identify eighteen proteins, of which twelve demonstrated to be involved with immune response in D. saccharalis. The six hours septic challenge with B. subtilis was responsible for modification of expression of Peptidoglycan recognition protein (PGRP), for increase of expression of Chitin-binding protein and for induction of expression of proteins like the Attacin-A and the Serine-protease inhibitor-like. The six hours septic challenge with E. coli increased the expression of a putative defense protein. The twelve hours septic challenge using B. subtilis and E. coli were responsible for increase of expression of Lysozyme. The six hours septic challenge with B. bassiana induced of expression of a Cecropin A2 and a Drosomycin-like. On the order hand, after twelve hours of fungal challenge, there was the induction of expression of multifunctional protein Apolipophorin-3. It was concluded that the septic challenge with different microorganisms were capable of change the expression of some proteins involved in the immune response in D. saccharalis, significant for understanding this process as well as for pointing the substances with antimicrobial functions. / A Diatraea saccharalis é a praga responsável por grandes perdas econômicas à cultura da cana-de-açúcar. Assim como outros integrantes da ordem Lepidoptera, a D. saccharalis possui resposta imunológica inata rápida e eficiente, constituída por barreira primária de defesa, resposta imune celular e resposta imune humoral. Os peptídeos antimicrobianos (PAMs) são parte da resposta imune humoral que apresentam características catiônicas e anfifílicas, além de sua massa molecular baixa, o que os tornam potenciais agentes terapêuticos. Neste estudo foram analisadas proteínas com baixa massa molecular, diferencialmente expressas na hemolinfa da D. saccharalis, após os intervalos de 6 e 12 horas de indução da resposta imune humoral com diferentes microrganismos em comparação às lagartas não desafiadas (controle). As lagartas em 5º instar foram divididas em grupos (n= 50) e submetidas a desafios sépticos de 6 e 12 horas: controle (grupo 1), desafiadas por Bacillus subtilis ATCC 6623 (grupo 2), desafiadas por Escherichia coli ATCC 11229 (grupo 3) e desafiadas por Beauveria bassiana cepa 88 (grupo 4). Em cada lagarta foi inoculada uma concentração conhecida do microrganismo e, após o tempo estabelecido, a hemolinfa foi coletada. As proteínas de baixa massa molecular foram obtidas submetendo cada amostra de hemolinfa a uma solução de extração contendo Metanol, Ácido Acético e Água. Posteriormente, os extratos proteicos foram concentrados em colunas e a dosagem proteica realizada em 280 nm. Aproximadamente 500 μg de proteínas foram submetidas à eletroforese bidimensional (2-DE). Os spots foram retirados dos géis, digeridos com Tripsina, e submetidos à espectrometria de massas do tipo MALDI-ToF/ToF. A identificação das proteínas ortólogas foi realizada utilizando os dados obtidos contra os dados disponíveis no servidor online MASCOT (Matrixscience), especificando a base de dados (NCBI e Swissprot) para Drosophila melanogaster e um banco interno de Lepidoptera. A ferramenta TagIdent foi utilizada para a busca de proteínas por meio de suas massas moleculares e ponto isoelétrico. Foram identificadas dezoito proteínas, das quais doze se mostraram envolvidas com a resposta imune em D. saccharalis. O desafio séptico de 6 horas com B. subtilis foi responsável pela alteração na expressão da proteína de reconhecimento de peptideoglicano (PGRP), pelo aumento da expressão da proteína ligadora de quitina e pela indução da expressão de proteínas como a Atacina-A e a proteína inibidora de serino-protease. O desafio imunológico de 6 horas com E. coli levou ao aumento da expressão de uma provável proteína de defesa. Os desafios de 12 horas utilizando as bactérias B. subtilis e E. coli se mostraram responsáveis pelo aumento da expressão da Lisozima. O desafio de 6 horas com B. bassiana causou indução da expressão de uma Cecropina A2 e uma possível Drosomicina. Por sua vez, após o desafio fúngico de 12 horas, houve a indução da expressão da proteína multifuncional Apolipoforina-3. Concluiu-se que desafios sépticos com diferentes microrganismos foram capazes de alterar a expressão de algumas proteínas envolvidas na resposta imune em D. saccharalis, importantes para o entendimento deste processo e também para o apontamento de substâncias com funções antimicrobianas.

Diversité Génétique des Molécules de Réponse Immunitaire Innée dans les Troubles Bipolaires / Genetic Diversity of Innate Immune Response Molecules in Bipolar Disorder

Les troubles bipolaires (TB) sont des troubles chroniques, multifactoriels et multi-systémiques avec une morbidité, mortalité et fardeau socio-économique très élevée. Mieux comprendre le fond génétique et les facteurs environnementaux déclencheurs permettra d'améliorer le diagnostic et la prise en charge thérapeutique. La dysimmunité semble être parallèle à l'apparition, la progression ainsi qu'au développement de comorbidités psychiatriques et somatiques. L'inflammation chronique et l'activation de la microglie sont des mécanismes liant potentiellement l'infection et le stress pendant l'enfance avec la survenue d'un TB, en particulier la plus sévère forme d'apparition précoce. Les variants génétiques des récepteurs de reconnaissance de motifs moléculaires, première ligne de défense immunitaire, pourraient participer à la résilience/vulnérabilité aux facteurs de risque environnementaux tôt au cours de la vie. Nous avons exploré cette possibilité en analysant les associations génétiques entre des acteurs centraux de la réponse immunitaire innée, TLR2, TLR4 et NOD2 et les TB, en testant aussi l'hétérogénéité entre les sous-groupes d'apparition précoce et tardive. Étant donné que l'activation de ces voies augmente la production d'oxyde nitrique (NO) et en vue de son rôle dans l'inflammation, le stress oxydatif et la neurotransmission ainsi que son dysfonctionnement dans les TB et suicide, nous avons analysé simultanément des variants génétiques des trois isoformes de NO-synthase (NOS) chez les patients avec antécédents de conduites suicidaires. Nous avons trouvé que les génotypes TLR4 rs1927914 AA et rs11536891 TT sont significativement plus fréquents chez les patients tandis que l'allèle NOD2 rs2066842 T est significativement plus fréquent chez les témoins suggérant une vulnérabilité génétique à l'exposition aux pathogènes dans les TB. En plus, l'association avec TLR4 est restreinte au sous-groupe à début précoce et le génotype TLR2 rs3804099 TT est significativement plus fréquent chez les patients à début précoce qu'à début tardif, confortant l'hypothèse que les facteurs génétiques ont un poids plus important dans les TB d'apparition précoce. Des associations entre NOS1, NOS2 et NOS3 et TB n'ont pas été observées mais NOS3 rs1799983 T à l'état homozygote a été associé aux tentatives de suicide violentes, chez les patients à début précoce, apportant une preuve supplémentaire de l'implication de variants de NOS3 (endothéliale) dans les conduites suicidaires. En étudiant l'effet de l'interaction entre les variants génétiques de TLR2/TLR4 et les antécédents d'abus dans l'enfance mesurées par le Childhood Trauma Questionnaire (CTQ) sur l'âge de début des TB, nous avons observé un effet combiné du génotype TLR2 rs3804099 T avec antécédents d'abus sexuels sur la détermination d'un âge plus précoce d'apparition des TB. Les effets de l'abus sexuel dans l'enfance sur l'âge d'apparition des TB peuvent être accrus chez les porteurs du génotype TLR2 rs3804099 TT potentiellement médié par des voies liés aux réponses inflammatoires. Nos résultats sont en faveur d'un modèle « multiple-hit » dans lequel une vulnérabilité génétique liée au système immunitaire contribuerait à des réponses anormales aux infections périnatales baissant le seuil pour les effets adverses des stress ultérieurs. La confirmation de ces données par réplication dans des cohortes indépendantes est nécessaire. La caractérisation de la sévérité de la maladie, des phénotypes immunitaires et du type, intensité, fréquence et moment de survenue des stress doivent être prévues dans les futures études. Le développement de modèles animaux pourra aussi permettre de mieux comprendre les mécanismes impliqués à travers la manipulation expérimentale de la génétique et des conditions environnementales. Ces approches pourraient permettre l'identification de biomarqueurs et de signes cliniques prodromiques bien que des nouvelles stratégies de prévention et des cibles thérapeutiques. / Bipolar disorders (BD) are chronic, multisystem and multifactorial disorders with significant lifetime morbidity, mortality and socioeconomic burden. Understanding its genetic background and triggering environmental factors should improve diagnosis and therapeutic management. Immune dysregulation seems to parallel its onset and progression as well as the development of psychiatric and other medical comorbidities. Chronic low-grade inflammation and microglia activation are thought to be important mechanisms linking infection and childhood trauma with BD, in particular the more severe early-onset subform. Genetic variations in pattern-recognition receptors, the first line of immune defence, may thus participate in one's resilience/vulnerability to environmental exposures, particularly early in life. We explored that possibility by investigating genetic associations between central players of innate immune protection, TLR2, TLR4 and NOD2 and BD, taking into account the potential genetic heterogeneity between the early- and late-onset subgroups. Given that the activation of these pathways increases the production of nitric oxide (NO), a potent innate immune effector, by immune cells and taking into consideration the role of NO in oxidative stress and neurotransmission as well as its dysfunction in both BD and suicide, we simultaneously analysed variants of the three isoforms of nitric oxide synthase (NOS) genes with suicidal behaviour in BD. We found TLR4 rs1927914 AA and rs11536891 TT genotypes to be significantly more prevalent in patients than in controls. We also found that NOD2 rs2066842 T allele carrier state may confer some protection against BD as it was more prevalent in controls. These results suggest a genetic vulnerability to pathogen exposure in BD. We also found that the referred association with TLR4 was restricted to the early-onset subgroup and that a TLR2 genotype (rs3804099 TT) was significantly more prevalent in early- than in late-onset patients comforting the hypothesis that genetic factors are of greater importance in early-onset BD. While associations between NOS1, NOS2 and NOS3 with BD were not observed, NOS3 rs1799983 T in homozygous state was associated with violent suicide attempts, seemingly restricted to the early-onset BD bringing further evidence for the potential involvement of endothelial NOS genetic variants in the susceptibility to suicidal behaviour. By investigating the effect of potential interaction between TLR2 and TLR4 and childhood abuses as measured by the Childhood Trauma Questionnaire (CTQ) on the age at onset of BD, we observed a combined effect of TLR2 rs3804099 TT genotype and childhood sexual abuse on determining an earlier age at onset of BD by means of a Kaplan-Meier survival curve. The effects of childhood sexual abuse on age at onset of BD may be amplified in TLR2 rs3804099 risk genotype carriers through immune-mediated pathways.Altogether the present results comfort a multiple-hit model in which immune-related genetic susceptibility contributes to abnormal responses to perinatal infectious insults establishing a lower threshold for subsequent stress-triggered events. Confirmation by replication in independent BD cohorts is warranted. Characteristics of illness severity, immune phenotypes and detailed annotation of type, intensity, frequency and time of stress exposure should be documented in future studies involving large cohorts. Additionally, the development of animal models in conformity with the presented model would facilitate better understanding of the precise mechanisms involved by allowing specific manipulation of genetic and environmental conditions. These approaches may allow the identification of both biological and clinical prodromal manifestations and consequently lead to preventive strategies and novel therapeutic targets.

Modulation of the innate immune response by the oncoviruses EBV and HPV / Modulation des réponses immunitaires innées par les oncovirus EBV et HPV

Le cancer représente la deuxième cause de mortalité dans les pays industrialisés. Il a été démontré que 20% des cancers sont d'origine infectieuse. Nous nous sommes intéressés à deux oncovirus HPV (virus du papillome humain) et EBV (Epstein-Barr Virus) responsable du cancer de l'utérus et de divers lymphome B réciproquement. Les événements clés pour le développement d'un cancer viro-induit sont la persistance du virus via la dérégulation des réponses immunitaires et l'induction d'une instabilité gé¬nomique via une dérégulation du cycle cellulaire. Nous avons donc cherché si EBV était capable d'altérer la réponse immunitaire innée. Nous avons montré que EBV était capable d'inhiber TLR9 un acteur clef de la réponse immunitaire innée. Comme TLR9 est inhibé dans un certain nombre de cancers, nous nous sommes demandé si ce récepteur pouvait également, avoir un rôle dans l'oncogenèse. Nous avons montré que la réexpression de TLR9 induisait un ralentissement transitoire de la prolifération cellulaire. Nous nous sommes par la suite intéressés aux mécanismes de dérégulation du cycle cellulaire induits par E6 une oncoprotéine de HPV16. Nous avons trouvé un nouveau mécanisme d'inhibition de l'inhibiteur du cycle cellulaire, p21. HPV16E6 se lie et inhibe les fonctions de du facteur de transcription p150Sal2, ce qui induit une inhibition de p21 dans un contexte p53 indépendant / Cancer represents the second most common cause of death in industrialized countries. Epidemiological and biological studies have now conclusively proved that a variety of infectious agents constitute one of the main causes of cancer worldwide. It has been pointed out that more than 20% of cancers are from infectious origin. HPV high-risk mucosal types are associated to 98% of all cervical cancer cases. Regarding EBV, over 90% of the world’s population is infected and can give rise to malignancies such as Burkitt lymphoma or Hodgkin disease.(Young and Rickinson 2004) Keys features for oncoviruses to induce cancer are firstly to per¬sist by dampening host immune responses and to induce genomic instability in the host by altering the regulation of the cell cycle leading the infected cells to an uncontrolled proliferation. The purpose of this thesis was to find new mechanisms by which EBV and HPV can promote carcinogenesis. We have shown that EBV can alter the regulation and expression of TLRs, the key effectors molecules of the innate immune response. EBV infection of human primary B cells resulted in the inhibition of TLR9 functionality. Our study described a mechanism used by EBV to suppress the host immune response by deregulating the TLR9 transcript through LMP1-mediated NF-κB activation. As TLR was found deregulated in many cancers, we hypothesized that TLR9 may also a direct role in the process of cell cycle control and that loss of its expression may lead to transformation of the cell. Our overall objective here was to study the role of TLR9 in suppressing the events that initiates transformation of epithelial cells in the setting of cervical cancer (virus-associated) and in head and neck cancer (non–virus-associated). A third project dealt with the mechanism cell cycle deregulation by the oncoprotein E6 which expressed during infection with HPV16. We reported that HPV16E6 targets the cellular factor p150Sal2, which positively regulates p21 transcription. HPV16E6 associates with p150Sal2, inducing its functional inhibition by preventing its binding to cis elements on the p21 promoter. These data described a novel mechanism by which HPV16E6 induces cell cycle deregulation with a p53-independent pathway preventing G1/S arrest and allowing cellular proliferation and efficient viral DNA replication

Em artrópodes o sistema imune inato contribui para a adaptação de animais como os escorpiões à diferentes ambientes. Esse sistema é composto por mecanismos capazes de agir contra injúrias e a ação de microrganismos e entre esses mecanismos estão os peptídeos antimicrobianos (PAMs). O objetivo deste trabalho foi identificar PAMs presentes na hemolinfa de Tityus serrulatus. Para isso sua hemolinfa foi extraída e separados os hemócitos e plasma, em seguida fracionamos em 3 concentrações de acetonitrila em TFA 0,05% (05, 40 e 80%). Estas frações foram submetidas a uma cromatografia liquida de alta eficiência (CLAE) e os picos foram avaliados quanto a sua ação antimicrobiana e hemolítica. Foram identificadas 16 frações que apresentam atividade antimicrobiana. Uma das frações com atividade antimicrobiana, presente nos hemócitos apresentou similaridade com defensina descrita em carrapatos da espécie Ixodes scapularis. Essa fração possui aproximadamente 3486 Da, não apresenta atividade hemolítica e foi denominada como Serrulina. / In arthropods, its innate immune system contributes to the adaptation of animals like scorpions to different environments. This system consists of mechanisms that act avoiding injuries and against the action of microorganisms, among these mechanisms are antimicrobial peptides (AMPs). The aim of this study was to identify AMPs, present in the hemolymph of Tityus serrulatus. The hemolymph was extracted and then we separated hemocyte and plasma. The samples were fractionated in different concentrations of acetonitrile in TFA 0.05% (05, 40 and 80%). These fractions were subjected to high-performance liquid chromatography (HPLC) and the peaks obtained were evaluated for its antimicrobial and hemolytic action. We found sixteen fractions with antimicrobial activity. One of the fractions with antimicrobial activity, present in hemocytes, is similar with a defensin described in ticks, Ixodes scapularis. This fraction has about 3486 Da, has no hemolytic activity and was named as Serrulina.

Pathways Involved in Recognition and Induction of Trained Innate Immunity by Plasmodium falciparum

Malarial infection in naïve individuals induces a robust innate immune response, but our understanding of the mechanisms by which the innate immune system recognizes malaria and regulates its response remain incomplete. Our group previously showed that stimulation of macrophages with Plasmodium falciparum genomic DNA (gDNA) and AT-rich oligodeoxynucleotides (ODNs) derived from this gDNA induces the production of type I interferons (IFN-I) through a STING/TBK1/IRF3-dependent pathway; however, the identity of the upstream cytosolic DNA receptor remained elusive. Here, we demonstrate that this IFN-I response is dependent on cyclic GMP-AMP synthase (cGAS). cGAS produced the cyclic dinucleotide 2’3’-cGAMP in response to P. falciparum gDNA and AT-rich ODNs, inducing IRF3 phosphorylation and IFNB transcription. In the recently described model of innate immune memory, an initial stimulus primes the innate immune system to either hyperrespond (termed “training”) or hyporespond (“tolerance”) to subsequent immune challenge. Previous work in mice and humans demonstrated that infection with malaria can both serve as a priming stimulus and promote tolerance to subsequent infection. In this study, we demonstrate that initial stimulation with P. falciparum-infected red blood cells (iRBCs) or the malaria crystal hemozoin (Hz) induced human adherent peripheral blood mononuclear cells (PBMCs) to hyperrespond to subsequent Toll-like receptor (TLR) challenge. This hyperresponsiveness correlated with increased H3K4me3 at important immunometabolic promoters, and these epigenetic modifications were also seen in monocytes from Kenyan children naturally infected with malaria. However, the use of epigenetic and metabolic inhibitors indicated that malaria-induced trained immunity may occur via previously unrecognized mechanism(s).
