Varför IV? : ett fritt gymnasieval eller en följd av grundskolans brister?

Den utbildningspolitiska intentionen i Sverige har länge präglats av en stark ambition att "överbrygga" klyftan mellan de teoretiska och praktiska gymnasieutbildningarna. Flertalet utredningar och gymnasiereformer har tidigare inte lyckats åstadkomma mer än organisatoriska förändringar. 1991 års gymnasiereform genomfördes med Lpf-94 och sett ur ett utbildningspolitisktperspektiv, är genomförandet rent teoretiskt den reform som kommit närmast den utbildningspolitiska intentionen. Men frågan kvarstår vad har reformen inneburit sett ur ett individperspektiv i ett utbildningssystem med en grundskola och ett gymnasium för alla. En skola för alla betyder i praktiken en skola som skall tillgodose samtliga elevers olika behov. Det låter fantastiskt men verkligheten visar ofta att detta är ett uppdrag som är svårt att genomföra och ett samhälleligt krav som kan vara svårt att uppfylla. Hur upplever eleverna på individuella programmet att deras grundskoletid har varit?

Contract Enforcement – And Its Impact on Bilateral Trade

Today it is well known that institutions have a significant impact on growth and development. Less research has been investigating how institutions and in-stitutional quality affect trade. This thesis will specifically examine the effect contract enforcement has on bilateral trade. Secure property rights and con-tract enforcement are important for a country’s productivity and growth. The empirical analysis is based on the gravity model of trade to examine what explains the trade flows and more importantly what impact contract enforce-ment has on the bilateral trade. Instead of using one of the many existing sub-jective measurements of contract enforcement, an objective measurement called Contract-Intensive Money (CIM) is used. The results show that contract enforcement of the exporting country has a greater impact on exports than that of the importing country. As expected the institutionally dependent sector of machinery and transport equipment requires a higher level of contract enforcement than the standardized food sector. It implies that the exporting country will have a comparative advantage in export-ing complex products and import simple products. The results also indicates that the effect on exports is higher when there is development of a country’s poor contract enforcement rather than improvement in already high-quality contract enforcement in the partner country.

Identification of new virulence factors in Francisella tularensis

Francisella tularensis, the causative agent of tularemia, is a highly virulent bacterium with an infection dose of less than ten bacteria. The ability of a pathogen to cause infection relies on different virulence mechanisms, but in Francisella tularensis relatively few virulence factors are known. Two F. tularensis subspecies are virulent in humans; the highly virulent subspecies tularensis, also referred to as type A, and the less virulent subspecies holarctica, also called type B. The aim of this thesis has been to improve the knowledge regarding the ability of Francisella to cause disease, with the emphasis on surface located and membrane associated proteins and structures. In addition I have also investigated how virulence is regulated by studying the role of the small RNA chaperone, Hfq. The genome of Francisella appears to encode few regulatory genes. In my work I found that Hfq has an important role in regulation of virulence associated genes in Francisella. Similar to what has been found in other pathogens, Hfq functions in negative regulation, and this is the first time a negative regulation has been described for genes in the Francisella pathogenicity island. Another protein with a key role in virulence is a homologue to a disulphide oxidoreductase, DsbA, which was identified as an outer membrane lipoprotein in Francisella. A dsbA mutant was found to be severely attenuated for virulence and also induced protection against wild-type infections, thus making it a candidate for exploration as a new live vaccine. Additional genes with homology to known virulence determinants include a type IV pilin system. The pilin homologue, PilA, was identified to be required for full virulence in both type A and type B strains. In addition, genes involved in pili assembly and secretion, pilC and pilQ, were also found to be virulence associated in the type A strain. In summary, dsbA, hfq and type IV pili associated genes were indentified to be virulence determinants in F. tularensis. DsbA is a potential target for drug development and a dsbA mutant a candidate for a new live vaccine strain. Furthermore the identification of Hfq as a novel regulatory factor opens new insights into the virulence regulatory network in Francisella.

Design and Synthesis of Angiotensin IV Peptidomimetics Targeting the Insulin-Regulated Aminopeptidase (IRAP)

Peptidomimetics derived from the bioactive hexapeptide angiotensin IV (Ang IV, Val1-Tyr2-Ile3-His4-Pro5-Phe6) have been designed and synthesized. These peptidomimetics are aimed at inhibiting the insulin-regulated amino peptidase (IRAP), also known as the AT4 receptor. This membrane-bound zinc-metallopeptidase is currently under investigation regarding its potential as a target for cognitive enhancers. The work presented herein was based on stepwise replacement of the amino acid residues in Ang IV by natural and unnatural amino acids, non-peptidic building blocks, and also on the introduction of conformational constraints. Initially, we focused on the introduction of secondary structure mimetics and backbone mimetics. The C-terminal tripeptide His-Pro-Phe was successfully replaced by a γ-turn mimetic scaffold, 2-(aminomethyl)phenylacetic acid (AMPA), which was coupled via an amide bond to the carboxyl terminus of Val-Tyr-Ile. Substitution of Val-Tyr-Ile, Val-Tyr, Tyr-Ile and Tyr, respectively, by 4-hydroxydiphenylmethane scaffolds comprising a 1,3,5-substituted benzene ring as a central moiety unfortunately rendered peptidomimetics that were less potent than Ang IV. The subsequent approach involved the introduction of conformational constraints into Val-Tyr-Ile-AMPA by replacing Val and Ile by amino acid residues appropriate for disulfide cyclization or ring-closing metathesis. Chemically diverse structures encompassing an N-terminal 13- or 14-membered macrocyclic tripeptide and a C-terminal non-peptidic moiety were developed. Tyr2 and AMPA were modified to acquire further knowledge about the structure-activity relationships and, in addition, to improve the metabolic stability and reduce the polarity. Several of the compounds displayed a high capacity to inhibit IRAP and exhibited Ki values in the low nanomolar range. Hence, the new compounds were more than ten times more potent than the parent peptide Ang IV. Enhanced selectivity over the closely related aminopeptidase N (AP-N) was achieved, as well as improved stability against proteolysis by metallopeptidases present in the assays. However, additional investigations are required to elucidate the bioactive conformation(s) of the relatively flexible N-terminal macrocycles. The compounds presented in this thesis have provided important information on structure-activity relationships regarding the interaction of Ang IV-related pseudopeptides and peptidomimetics with IRAP. The best compounds in the series constitute important starting points for further discovery of Ang IV peptidomimetics suitable as tools in the investigation of IRAP and other potential targets for Ang IV. The literature presents strong support for the hypothesis that drug-like IRAP inhibitors would serve as a new type of future cognitive enhancers with potential use in the treatment of cognitive disorders, e.g. Alzheimer’s disease.

Psykiatriska diagnoser och genus : en undersökning av diagnoskriterier i förhållande till föreställningar om manligt och kvinnligt

Denna studie utgår från Judith Butlers genusteori om hur kön görs samt Michel Foucaults teorier om samhällsnormer och makt i förhållande till särskiljandemekanismer och vansinnesdefinitioner. Studien fokuserar på psykiatriska diagnoser ur ett genusperspektiv. Fokus har lagts på diagnoserna Aspergers syndrom och borderline personlighetsstörning då tidigare forskning visat att könsfördelningen inom dessa psykiatriska diagnoser är väldigt ojämn.   Syftet med studien är att undersöka genusstrukturers påverkan på psykiatrisk diagnostisering utifrån DSM-IV. Utifrån syftet har följande frågeställningar utformats: Går det att benämna diagnoskriterier som könsneutrala då diagnostisering i hög grad handlar om andra människors (vars egna erfarenheter och värderingar färgar neutraliteten) bedömningar? Hur kommer det sig att könsfördelningen är så ojämn inom diagnoser som Aspergers syndrom och borderline personlighetsstörning?   En enkätundersökning har utförts bland verksamma psykologer inom Östersunds kommun. Enkäten har utformats från de utvalda diagnosernas kriterier i DSM-IV. För att behandla enkätens svar har dataprogrammet SPSS använts. Undersökningens resultat visar på att det finns frågetecken kring DSM-IV:s könsneutralitet. Resultaten visar dock att det finns en medvetenhet kring den ojämna könsfördelningen inom de utvalda diagnoserna. Studien visar på psykiatriska diagnosers symbios med samhällsstrukturer och normer, däribland genus.

Stromal collagens in colorectal cancer and in colorectal liver metastases : tumour biological implications and a source for novel tumour markers

Background: Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality. About 50 % of patients with CRC will develop subsequent liver metastases (CLM). The survival for untreated CLM is only a few months and liver resection provides the only chance for a lasting cure. It is therefore essential to detect CLM early, enabling successful surgical resection and achieving a long-term cure. There are no optimal tumour markers for CRC or CLM. The best marker available is Carcinoembryonic Antigen (CEA), a marker found elevated in about 50-60% of patients with CLM, but also in many other conditions. The main focus of cancer research has been on the malignant cancer cell. However, a tumour consists of more than cancer cells. A major part of all solid tumours is made up by the stroma. The tumour stroma is defined as the non-malignant cells of a tumour such as fibroblasts, the cells of the vascular and immune systems as well as the extracellular matrix (ECM). The basement membrane (BM) is a specialized form of the ECM in which type IV collagen is the major protein component. All epithelial cells need a contact to the BM and the definition of an invasive cancer is the degradation of the BM and the spread of cancer cells beyond this structure. Different metastatic growth patterns of CLM have previously been described, namely the desmoplastic, pushing and replacement type of CLM. These differ in their stromal reaction in the border, which separates the tumour from the normal liver. In this thesis the tumour stroma of CRC and CLM is studied with a special emphasis on stromal collagens. The aim is to investigate whether stromal collagens/ circulating type IV collagen can be used as tumour markers for CRC and CLM, and to compare this to the conventional marker CEA. The circulating type IV collagen level is also measured in liver metastases from other primary tumours than CRC. Furthermore, the differences between the stroma of a primary CRC that metastasizes to the liver when compared to a CRC that never spreads are analysed. Additionally, the metastatic growth pattern of CLM is studied in relation to the primary tumour, stromal components and survival. We also sought out to find whether CRC cell lines possess the trait to produce ECM proteins endogenously, and in response to a normal liver stroma in a novel organotypic model for CLM. Methods: Expression patterns of type I, III and IV collagen were studied by immunofluorescence (IF), chemical staining and immunohistochemistry (IHC) in normal colorectal tissue, normal liver, CRC, CLM, benign liver lesions and in liver metastases of other origin than CRC. Circulating plasma levels of type IV collagen were analysed in healthy controls, patients with CRC (T stage I-III) and in patients with CLM. Samples were analysed at the time of diagnosis, during and after oncological and surgical treatment and at the time of relapsing or progressive disease. Additionally, circulating levels were analysed in patients with benign liver lesions and in liver metastases of other origin than CRC. The metastatic growth pattern of CLM was classified according to earlier descriptions. CRC cell lines were studied regarding their production of type IV collagen. The growth, invasiveness and stromal production in CRC cell lines were also investigated in a new organotypic model for CLM using human liver specimens. Results: Circulating type IV collagen levels are increased in patients with CLM and other epithelial-derived liver metastases, and is found normal in patients with primary CRC (stage I-III), with liver metastases from tumours of non-epithelial origin, benign liver lesions and in healthy controls. The type IV collagen levels in patients with CLM reflect the tumour burden in the liver, decreases in response to therapy and is found increased in progressive or relapsing disease. The combination of circulating type IV collagen and CEA increased the sensitivity and specificity for detecting CLM. Livermetastatic CRC displayed an increased stromal production when compared to non-metastatic CRC, with an increased type IV collagen expression in the direct vicinity of the CRC cells. The earlier described growth patterns of CLM were verified, with the pushing type of CLM associated with a short survival and poor outcome. Furthermore, CRC cell lines possess the trait of endogenously producing type IV collagen. The novel organotypic liver model revealed that CRC cell lines grown in the context of normal liver stroma, devoid of other cells, does not elicit a desmoplastic reaction. Conclusion: Circulating type IV collagen is a promising tumour marker for CLM, where the levels reflect the hepatic tumour burden and can detect disease relapse after liver surgery. The combination of the tumour markers CEA and type IV collagen is superior to CEA alone. The stromal composition of primary CRC predicts the risk of subsequent CLM and the metastatic growth pattern of CLM is related to survival.

Heat-transfer analysis of double-pipe heat exchangers for indirect-cycle SCW NPP

SuperCritical-Water-cooled Reactors (SCWRs) are being developed as one of the Generation-IV nuclear-reactor concepts. SuperCritical Water (SCW) Nuclear Power Plants (NPPs) are expected to have much higher operating parameters compared to current NPPs, i.e., pressure of about 25 MPa and outlet temperature up to 625 oC. This study presents the heat transfer analysis of an intermediate Heat exchanger (HX) design for indirect-cycle concepts of Pressure-Tube (PT) and Pressure-Vessel (PV) SCWRs. Thermodynamic configurations with an intermediate HX gives a possibility to have a single-reheat option for PT and PV SCWRs without introducing steam-reheat channels into a reactor. Similar to the current CANDU and Pressurized Water Reactor (PWR) NPPs, steam generators separate the primary loop from the secondary loop. In this way, the primary loop can be completely enclosed in a reactor containment building. This study analyzes the heat transfer from a SCW primary (reactor) loop to a SCW and Super-Heated Steam (SHS) secondary (turbine) loop using a double-pipe intermediate HX. The numerical model is developed with MATLAB and NIST REFPROP software. Water from the primary loop flows through the inner pipe, and water from the secondary loop flows through the annulus in the counter direction of the double-pipe HX. The analysis on the double-pipe HX shows temperature and profiles of thermophysical properties along the heated length of the HX. It was found that the pseudocritical region has a significant effect on the temperature profiles and heat-transfer area of the HX. An analysis shows the effect of variation in pressure, temperature, mass flow rate, and pipe size on the pseudocritical region and the heat-transfer area of the HX. The results from the numerical model can be used to optimize the heat-transfer area of the HX. The higher pressure difference on the hot side and higher temperature difference between the hot and cold sides reduces the pseudocritical-region length, thus decreases the heat-transfer surface area of the HX. / UOIT

Development of a heat-transfer correlation for supercritical water in supercritical water-cooled reactor applications

A large set of experimental data, obtained in Russia, was analyzed and a new heat-transfer correlation for supercritical water was developed. This experimental dataset was obtained within conditions similar to those for proposed SuperCritical Water-cooled nuclear Reactor (SCWR) concepts. Thus, this new correlation, for forced convective heat transfer in the normal heat-transfer regime, can be used for preliminary heat-transfer calculations in SCWR fuel channels. It has demonstrated a good fit for Heat Transfer Coefficient (HTC) values (±25%) and for wall temperature calculations (±15) for the analyzed dataset. This correlation can be used for supercritical water heat exchangers linked to indirectcycle concepts and the co-generation of hydrogen, for future comparisons with other independent datasets, with bundle data, as the reference case, for the verification of computer codes for SCWR core thermalhydraulics and for the verification of scaling parameters between water and modeling fluids. / UOIT

CONQUER CORROSION : Key issues of the lead-cooled fast reactor design

The lead-cooled fast reactor (LFR) is one of the concepts of the Generation IV reactorsystems. There are some issues that have to be solved before a research orcommercial LFR can be built. The objective of this thesis was to identify these keyissues and analyse them by studying results from previous research: choice of fuel,corrosion on structural materials and corrosion/erosion on pumps.The major fuel candidates for the LFR are MOX fuel (Mixed OXide fuel), metallic fuel,nitride fuel and carbide fuel. Nitride fuel has desirable properties but its production ismore difficult than for MOX fuel.Most of today’s commercial steels are not corrosion resistant at higher temperaturesbut they could possibly be used for an LFR test demonstrator with an operatingtemperature lower than 450 ºC. A new type of steel called oxide dispersionstrengthened (ODS) steel and a new ceramic material MAXTHAL both showpromising corrosion resistance even at higher temperatures.By controlling the oxygen concentration a protective oxide film is produced. Flowingliquid coolant causes erosion and wears down the oxide film. Pumps are exposed tocoolant velocities of 10-15 m/s causing both erosion and corrosion. There is nosolution today, but MAXTHAL shows promising results in tests with liquid lead of lowvelocity. There are also other issues unsolved, such as irradiation damage onstructural materials, thus more research is needed.Economic and political aspects were not covered in this study. This thesis work wasperformed at Vattenfall Research and Development AB.

An investigation of fuel cycles and material flows for a lead-cooled fast reactor using the Monte Carlo code Serpent

The Monte Carlo code Serpent has been used to model the material flows andisotope compositions for a lead cooled fast reactor. The demonstration sized trainingreactor ELECTRA was chosen for the investigation, and different fuel cycle scenarioswere studied. The scenarios differed in operation length (3 months, 1 year or 5years) and recycling technique (single and double PUREX or GANEX). The simulations gave detailed information on the changes of the isotope composition,activity and decay heat. The analysis of the generated waste also showed that thechoice of recycling method had great impact on the final storage time of the wastefrom the reprocessing. Performing double GANEX recycling, as compared to singlePUREX, reduced the storage time by a factor of about 3500. The results can be used for future work related to even more detailed studies ofmaterial flows and for designing an appropriate safeguards system.
