Filmklippning och karaktärsgestaltning : En undersökning kring vad filmklippning gör för gestaltningen av hu-vudkaraktären i filmen Swiss Army Man

Hur påverkar klippningen gestaltningen av en karaktär i en film? Syftet med denna uppsats är att undersöka om klippning påverkar gestaltningen av en filmkaraktär och vilka metoder inom filmklippning som kan användas för att uppnå detta. Undersök-ningen sker på huvudkaraktären i filmen Swiss Army Man där tre sekvenser analyse-ras med hjälp av Jens Eders modell för att analysera fiktiva karaktärer. Analysen kopplas till tidigare forskning och teorier om filmklippning. Uppsatsen kommer även att testa Eders modell för att undersöka om den är användbar som karaktärsanalysmo-dell för filmkaraktärer. Resultatet av undersökningen visar att klippning kan användas som en del av arbetet för att gestalta en filmkaraktär. Det finns klippmetoder som gör att klipparen kan på-verka gestaltningen. Eders analysmetod fungerar att koppla till filmklippning men resultatet kan variera då denna metod är subjektiv och karaktärer kan uppfattas olika av varje individ.

Efecto acoplado de la presión de confinamiento y el corte estático inicial en la resistencia ciclíca de arena de relaves

Magíster en Ciencias de la Ingeniería, Mención Ingeniería Geotécnica / Los altos niveles de producción de cobre en la actualidad, junto con la baja ley de los yacimientos, hacen que se generen cantidades importantes de desechos llamados relaves. El almacenamiento de grandes volúmenes de relaves obliga al estudio del comportamiento tanto estático como sísmico de este material, de manera de asegurar la estabilidad mecánica de las presas de relaves. En este contexto es de gran importancia estudiar la resistencia de los relaves frente al fenómeno de licuación, materia del presente trabajo. Diversos estudios han mostrado que el nivel de presión de confinamiento afecta la resistencia cíclica de arenas, proponiéndose el factor de corrección Kσ. Análogamente, se ha observado que el nivel de corte estático inicial también afecta la resistencia cíclica, a raíz de lo cual se ha introducido el factor de corrección Kα. Sin embargo, el efecto acoplado de ambas variables, que corresponde a la condición real de terreno, no ha sido debidamente investigado. Consecuentemente, mediante ensayos triaxiales cíclicos no drenados consolidados tanto isotrópicamente como anisotrópicamente, se ha estudiado el efecto conjunto de ambas variables en la resistencia cíclica de arena de relaves, para distintos niveles de densidad relativa inicial. Los resultados obtenidos del efecto acoplado de la presión de confinamiento y el corte estático en la resistencia cíclica de arena de relaves, se han resumido a través del parámetro Kσα. Este factor se presenta gráficamente con la intención de advertir donde la combinación de presión de confinamiento y corte estático inicial (en términos del parámetro α) tiene efectos positivos o negativos en la resistencia cíclica de arena de relaves. El análisis de datos indica una sobrestimación de la resistencia al usar el producto de los factores por separado (Kσ x Kα), en comparación al uso del factor acoplado Kσα, para cada una de las densidades estudiadas. Los resultados experimentales muestran que existe en definitiva dos modos de desarrollo de deformaciones. Por un lado, está el caso donde la probeta claramente exhibe deformaciones axiales cíclicas, identificándose la falla por un nivel de deformación entre los peaks de compresión y extensión. Por otro lado, está la condición de una reducida deformación cíclica, pero un marcado desarrollo de deformación acumulada hacia la zona en compresión, identificándose la falla por el nivel de deformaciones máximo acumulado. Cada uno de estos modos depende de la combinación de densidad relativa, presión confinante, corte estático y nivel de solicitación cíclica.

Transcriptome Characterization and Polymorphism Detection in Subspecies of Big Sagebrush (<em>Artemisia tridentata</em>)

Big sagebrush (Artemisia tridentata) is one of the ecologically most important shrub species in western North America. The species serves as a major source of food and habitat for the near-threatened sage grouse and various other fauna. Habitat loss due to a combination of disturbances followed by establishment of invasive plant species is considered as a serious threat to sustainability of the big sagebrush ecosystem. Because of its importance, restoration of this species is very crucial to those dependent on big sagebrush community. However, restoration of big sagebrush carried out by using diverse seed source can lead to imbalance and degradation in the native ecosystem. Therefore, restoration works aided by understanding of adaptive traits of big sagebrush using molecular markers will aid successful restoration. The major objective of this research was to create a substantial resource of nuclear sequence data and identify markers that can be used in future studies in big sagebrush. We report the development and annotation of the first expressed sequence tag (EST) collection for big sagebrush based on 454 sequencing of leaf tissue. Expressed genes of subspecies tridentata and vaseyana were sequenced using the 454 GS-FLX titanium platform, which produced 823,392 reads with an average read length of 404 bp and 702,001 reads with an average read length of 333 bp for sspp. tridentata and vaseyana, respectively. Assembly of the reads resulted in 212,102 consensus sequences in ssp. tridentata and 199,439 in ssp. vaseyana. A combined assembly of both subspecies sequences generated 29,541 contigs with an average length of 796 bp and 275,866 singletons with an average length of 370 bp. A BLASTx search against the non-redundant (NR) protein database using the contigs obtained from a combined assembly resulted in 21,436 sequences with significant blast alignments (≤ 1e-15). Gene Ontology (GO) IDs were assigned to 18,397 sequences. A total of 20,952 SNPs were detected between the two subspecies and 1,182 SNPs were confirmed in tetraploid ssp. wyomingensis. In addition, 1,003 and 507 SSRs were detected in ssp. tridentata contigs and ssp. vaseyana contigs, respectively.

System Design of an Integrated Terrestrial-Satellite Communications Network for Disaster Recovery

This thesis describes a possible integrated terrestrial-satellite network system for disaster recovery and response. The motivation of this thesis was based on the adjacent spectrum allocations between the Virginia Tech terrestrial Local Multiple Distribution Service (LMDS) system and a Ka-band satellite system, and potentially being able to provide as an additional Ka-band satellite network backbone to the Virginia Tech terrestrial LMDS system for better and faster communications deployments. The Spaceway satellite system's design parameters were adopted typically for a Ka-band satellite system. The LMDS system was assumed to use IEEE 802.16 standard protocols although it currently uses its own proprietary protocols. Four possible topologies integrating both terrestrial and satellite network were investigated. The study showed that the task was more problematic and complicated than anticipated due to incompatible network protocols, limitations of available hardware components, the high path loss at Ka-band, and the high cost of the equipment, although the adjacent frequency bands do suggest a possible integrated network. In this thesis, the final selected topology was proposed and designed. The technical characteristics of the earth station used for coupling both terrestrial and satellite networks were determined by a link budget analysis and a consideration of network implementations. The reflector antenna used by the earth station was designed. In addition, other system design concerns and engineering tradeoffs, including adjacent satellite interference, rain attenuation, antenna pointing error, noise temperature, and modulation and multiple access selection, were addressed. / Master of Science

Abstract in English, Afrikaans and Xhosa / The main aim of this study was to explore the extent to which teachers use group investigation as a teaching approach for Life Orientation in selected secondary schools in the Northern Cape Province in South Africa. Group investigation (GI) as a cooperative learning teaching approach is based on the philosophical principles of John Dewey who believed that schools had the responsibility of capturing learners’ interests, to expand and develop their horizons and to assist them in responding appropriately to new ideas and influences. Group investigation, a particular cooperative learning strategy, is based on the pragmatic paradigm that suggests that learners construct and apply knowledge as they interact with one another in a teacher-learner environment. This study was adopted in a concurrent triangulation mixed methods research design to be conducted concurrently. Quantitative and qualitative data were collected and analysed separately but simultaneously, the findings converged in the conclusion in order to respond to the research question. Convenient and purposive sampling was employed to select teachers and learners as participants in this study. Data were collected in the social setting where teachers employed group investigation as a teaching strategy with learners in cooperative working groups participating in learner-centred activities. The researcher used face-to-face interviews, focus group interviews, classroom observations as well as document analysis to collect data. The findings of this study revealed that teachers tend to display adequate content and pedagogical knowledge for the teaching of Life Orientation. However, the findings established that there is a need for novice teacher development in the interpretation and application of the content. Furthermore, group investigation has indicated that learners developed group skills easily, learn to work cooperatively in groups and that learners can depend on each other for the groups’ success. This study recommends that group investigation as a teaching approach be incorporated in the Life Orientation curriculum and that this needs to be shown in the lesson planning of teachers. The importance in the body of knowledge regarding teaching approaches and the academic value of the subject, Life Orientation as a fundamental subject, is indispensable within the FET curriculum. / Die hoofdoel van hierdie studie was om te bepaal in watter mate onderwysers groepondersoek as ʼn onderrigbenadering vir Lewensoriëntering in uitgesoekte sekondêre skole in die Noord-Kaapprovinsie gebruik. Groepondersoek as ʼn koöperatiewe leer- en onderrigbenadering is gegrond op die filosofiese beginsels van John Dewey, wat geglo het dat skole die verantwoordelikheid het om leerders se belange te akkommodeer, om hul horisonne te verbreed en te ontwikkel en om hulle by te staan om gepas op nuwe idees en invloede te reageer. Groepondersoek, ʼn spesifieke koöperatieweleerstrategie, is gegrond op die sosiaal-interpretivistiese-konstruktivistiese paradigma wat suggereer dat leerders kennis konstrueer en toepas wanneer hulle met mekaar interaksie het in ʼn onderwyser-leerder-omgewing. Die bevindings van die studie het getoon dat onderwysers geneig is om voldoende inhoud en pedagogiese kennis vir die onderrig van Lewensoriëntering te toon, terwyl daar ʼn behoefte aan die ontwikkeling van nuweling-onderwysers se interpretasie en toepassing van die inhoud is. Daarbenewens het die groepondersoeke ook gewys dat leerders maklik groepvaardighede ontwikkel, leer om koöperatief in groepe te werk, en dat hulle op mekaar kan staatmaak vir die sukses van die groep. Hierdie studie beveel aan dat groepondersoeke as ‘n onderrigstrategie in die Lewensorientering kurrikulum geïnkorporeer moet word en dat dit in die lesbeplanning van die onderwysers aangedui moet word. Rôle des récepteurs glutamatergiques dans l'activité épileptiforme des interneurones inhibiteurs de l'hippocampe

Les patients atteints d'épilepsie du lobe temporal (TLE) ainsi que les rats injectés à l'acide kaïnique (KA) exhibent des patrons pathophysiologiques similaires de crises, de sclérose de l'hippocampe et de perte de certains types neuronaux. Parmi les cellules atteintes dans le modèle KA du TLE on retrouve certains interneurones inhibiteurs du CA1. En effet, certains interneurones des couches oriens et alveus (O/A-IN) meurent suite à une injection de KA chez le rat, contrairement aux interneurones à la bordure des couches radiatum et lacunosum/moleculare (R/LM-IN) de la même région. Bien que cette perte soit empêchée par des antagonistes des récepteurs glutamatergiques métabotropes de groupe I (mGluR1/5), la cause de cette perte sélective des O/A-INs reste à être précisée. Au cours des travaux de cette thèse, nous avons effectué des enregistrements de patch-clamp en configuration cellule-entière en modes courant- et voltage-imposé couplés à l'imagerie calcique pour étudier les causes de la vulnérabilité sélective des O/A-INs dans ce modèle. Dans un premier temps, nous avons évalué les effets d'une application aiguë de KA sur les propriétés membranaires et calciques pour voir s'il y avait des différences entre les O/A-INs et R/LM-INs qui pourraient expliquer la vulnérabilité. Nos résultats montrent que les dépolarisations et variations de résistance d'entrée ainsi que les augmentations de calcium intracellulaire, dépendantes principalement des récepteurs -amino-3-hydroxy-5-methyl-4-isoxasole propionic acid (AMPA), sont similaires entre les deux types d'interneurones suite à des applications aigües de KA. Ceci indique que l'effet aigu du KA sur les interneurones ne serait pas la cause de la vulnérabilité des O/A-INs. Dans un second temps nous avons comparé l'implication des sous-types de récepteurs mGluR1 et 5 dans l'activité épileptiforme des deux types d'interneurones évoquée dans un modèle de tranche désinhibée. Dans ce cas, nos données montrent un rôle important des mGluR1 et 5 activés synaptiquement lors des décharges épileptiformes et ce, de manière spécifique aux O/A-INs. Les courants synaptiques sous-tendant ces décharges impliquent des récepteurs ionotropes et métabotropes du glutamate. En présence d'antagonistes des récepteurs ionotropes glutamatergiques, les courants synaptiques sont biphasiques et formés de composantes rapide et lente. Les récepteurs mGluR1 et 5 sont différemment impliqués dans ces composantes: les mGluR5 étant impliqués dans les composantes rapide et lente, et les mGluR1 que dans la composante lente. Ces résultats indiquent que les mGluR1 et 5 contribuent différemment à l'activité épileptiforme, et spécifiquement dans les O/A-INs, et pourraient donc être impliqués dans la vulnérabilité sélective de ces interneurones dans le modèle KA. / Temporal lobe epilepsy (TLE) patients, as well as kainic acid (KA)-treated rodents, display similar pathophysiological patterns of behavioural seizures, hippocampal sclerosis and loss of certain neuronal types in the hippocampus. Among the cell types selectively vulnerable in the experimental KA model of TLE are certain inhibitory interneurons of the CA1 hippocampal region. Specifically, interneurons located in the oriens and alveus layers (O/A-IN) are lost following KA injections, whereas interneurons found in the radiatum/lacunosum-moleculare layers (R/LM-IN) are resistant. Although it has been shown that the group I metabotropic glutamate receptor (mGluR1/5) inhibitors can block this cell loss seen in the KA model, the precise cause of the selective O/A-IN vulnerability remains to be clarified. In this thesis, we have performed whole-cell patch-clamp recordings with simultaneous calcium imaging in an effort to elucidate the cause of the selective vulnerability of O/A-INs. We first determined the effects of acute KA applications on membrane properties and intracellular calcium rises in hippocampal slices to see if they might be different between O/A-INs and R/LM-INs. Our results reveal similar -amino-3-hydroxy-5-methyl-4-isoxasole propionic acid (AMPA) receptor dependent membrane depolarizations, input resistance variations and calcium reponses in these cells following KA applications, suggesting that acute KA actions may not cause the selective vulnerability of O/A-INs. Furthermore, we evaluated the contribution of mGluR1/5 to epileptiform discharges evoked in a disinhibited slice model, comparing responses between O/A-INs and R/LM-INs. Our data show an important role of synaptically activated mGluR1/5 during epileptiform discharges specifically in O/A-INs. In addition we show that the synaptic currents underlying these discharges involve ionotropic and metabotropic glutamate receptors. In the presence of antagonists of ionotropic glutamate receptors, synaptic currents are biphasic and composed of fast and slow components. mGluR1 and mGluR5 are involved differently in these components with mGluR5 implicated in fast and slow components and mGluR1 in the slow component only. Our findings therefore suggest that mGluR1 and 5 contribute differently to epileptiform discharges, and do so specifically in O/A-INs, suggesting that their activation may contribute to the selective vulnerability of these interneurons in the KA model of TLE.

A partir de uma análise comparativa de duas obras do romance moçambicano, Os sobreviventes da noite (2008), de Ungulani Ba Ka Khosa, e Neighbours (1995), de Lilia Momplé, é possível visualizar certo tipo de estruturação da narrativa em que o tempo presente parece afetado por engastes narrativos que interceptam constantemente o avanço da ação. A esses engastes narrativos correspondem inúmeras camadas de passado em que as histórias pessoais de cada personagem ganham vez na economia da obra e se sobrepõem ao tempo que corresponde à ação no presente. Essa característica estética, aqui chamada de presente dependente, é analisada à luz de uma crítica materialista que tenta conectar o dado estético à dimensão histórica do período em que essas obras foram escritas e a que ambas fazem referência. O enredo dos dois romances situa-se no conflito armado que se seguiu à independência de Moçambique. Nesse sentido, a investigação avança sobre as hipóteses que, entre a literatura e a história, motivam o fato de duas obras tratarem de um mesmo momento histórico através de estruturas estéticas aproximáveis. / Starting from a comparative analysis of two novels pertaining to the literature from Mozambique, Os sobreviventes da noite (2008), by Ungulani Ba Ka Khosa, and Neighbours (1995), by Lilia Momplé, its possible to visualize a certain kind of narrative structuring in which the present seems affected by narrative insertions that constantly intercept the progress of the action. These narrative insertions are the countless layers of a past in which the personal stories of each character have the turn in the economy of the novel and put themselves on the top of the present, when the action is happening. This esthetic characteristic, here nominated as dependant present, is analyzed illuminated by a materialist criticism that tries to connect the esthetic fact to the historic dimension of the period when these novels were written and both make reference to. The plot of the two novels is situated in the armed conflict that emerged after the independence of Mozambique. In this meaning, the investigation advances in the hypothesis that, among literature and history, motivates the fact that both the novels treat about the same historic moment using esthetic structurings that, possibly, are close one to the other.

A crítica consagrou formas de observar a prática de gêneros literários no surgimento e desenvolvimento da literatura moçambicana: se, por um lado, tornou-se comum a percepção de que, durante o período colonial, esta literatura se consolida por meio de textos poéticos, por outro lado, tornou-se assente que, com a independência do país em 1975, a prosa passa a ser predominante, ao menos após a opção de expedientes romanescos no final da década de 1980. Esta tese se debruça a investigar porque a emancipação política e construção do Estado independente é um catalisador do fenômeno literário e em que medida a prática da poesia e a adoção de expedientes romanescos se relacionam com dinâmicas históricas e sociais que propiciam a emergência das articulações estéticas facultadas por essas opções genéricas. Tal dinâmica pode ser observada por meio da análise e interpretação histórica dos textos literários das coletâneas Poesia de combate (1977, 1979 e 1983) e da obra Ualalapi (1987), de Ungulani Ba Ka Khosa. / The literary criticism establishes some ways to observe the practice of literary genres in the emergence and development of Mozambican literature: on the one hand, it became common the perception that, during the colonial period, this literature is consolidated through poetic texts, on the other hand it became accordance that, with the independence of the country in 1975, prose became predominant, especially after the option of romanesque expedients in the late 1980s. This thesis focuses on investigating why the political emancipation and construction of the independent state is a catalyst for the literary phenomenon and how the practice of poetry and romanesque expedients adoption are related to historical and social dynamics that foster the emergence of the aesthetic articulations provided by these options generics. These dynamics can be observed through the analysis and historical interpretation of the literary texts of the compilations Poesia de combate (1977, 1979 and 1983) and Ualalapi (1987), by Ungulani Ba Ka Khosa.
