Synthèse et caractérisation microstructurale de poudres nanométriques à base de Bi2Te3 et Sb2Te3 : contribution à l'état de l'art des nanocomposites thermoélectriques / Synthesis and characterization of nanometric powders based on Bi2Te3 and Sb2Te3 : contribution to the state-of-the-art of thermoelectric nanocomposites

L’étude de matériaux thermoélectriques nanocomposites et nanostructurés est en plein essor suite à l’intérêt de multiplier le nombre d’interfaces et de diminuer la taille des objets pour améliorer les performances. Nous avons mis au point une nouvelle méthode de préparation de nanopoudres thermoélectriques de type n (Bi0.95Sb0.05)2(Te0.95Se0.05)3 et de type p (Bi0.2Sb0.8)2Te3, à partir de la fracturation laser en milieu aqueux de poudres de taille micrométrique. La cellule de préparation développée permet d’obtenir par jour environ 200 mg de poudres nanométriques cristallisés présentant la structure cristallographique des poudres initiales et dont la taille moyenne est comprise entre 7 et 12 nm. Les mécanismes mis en jeu dans l’obtention des nanoparticules ont été abordés. Ils dépendent fortement de la densité d’énergie du faisceau laser. Les nanopoudres ont ensuite été mélangées mécaniquement aux poudres micrométriques de même nature et ont été compactées à froid. Les propriétés thermoélectriques (résistivité électrique, pouvoir thermoélectrique, conductivité thermique) des nanocomposites ont été évaluées à température ambiante. Les premiers résultats montrent que même si le pouvoir thermoélectrique est maintenu dans les matériaux nanostructurés et nanocomposites et que la conductivité thermique totale peut, de manière tout à fait exceptionnelle, être diminuée d’un facteur deux, la résistivité électrique obtenue est jusqu’alors trop élevée pour conduire à de bonnes performances en terme de facteur de mérite adimensionnel, par rapport à un matériau massif conventionnel de même composition / The study of thermoelectric nanostructured and nanocomposite materials is expanding because of the interest to multiply the number of interfaces and to decrease the size of the objects in order to improve the thermoelectric performance. We developed a new method to prepare thermoelectric n type (Bi0.95Sb0.05)2(Te0.95Se0.05)3 and p type (Bi0.2Sb0.8)2Te3 nanopowders, from the laser fracture in a liquid medium of powders of micrometric size. The developed cell preparation makes it possible to obtain per day approximately 200 mg of crystallized nanometric powders having the crystallographic structure of the initial powders and whose mean size lies between 7 and 12 nm. The mechanisms concerned in obtaining the nanoparticules were approached. They strongly depend on the density of energy of the laser beam. The nanopowders then were mechanically mixed with the micrometric powders of comparable nature and were cold pressed. The thermoelectric properties (electrical resistivity, thermoelectric power, thermal conductivity) of the nanocomposites were evaluated at room temperature. The first results show that even if the thermoelectric power is maintained in nanostructured and nanocomposite materials and that the total thermal conductivity can, in a completely exceptional way, being decreased by a factor two, the electrical resistivity obtained is hitherto too high to lead to high values of the dimensionless thermoelectric figure of merit, with regard to conventional bulk materials of same composition

Enriquecimento seletivo para pesquisa de Mycobacterium bovis em leite e queijo / Selective enrichment for research of Mycobacterium bovis in milk and cheese

O Mycobacterium bovis causa a tuberculose bovina, doença zoonótica que propaga, provavelmente com baixa prevalência, em todo o território nacional e pode ser transmitida pelo leite. A adoção sistemática da pasteurização do leite contribuiu para a redução dos casos humanos da doença, mas em alguns países, como o Brasil, é comum o consumo de leite cru e de seus derivados, o que pode contribuir para ocorrência de casos humanos. A participação desse agente nos atuais índices de tuberculose humana, cujo principal agente é o M. tuberculosis, é pouco investigada, mas acredita-se que seja maior do que parece. As razões que contribuem para isso incluem o fato do tratamento humano ser o mesmo independentemente do agente,e as dificuldades e alto custo de distinguir as espécies. O método de diagnóstico \"gold standard\" em laboratório para Mycobacterium bovis em amostras clínicas é o isolamento do agente em meios como Stonebrink-Leslie ou similares. A riqueza em nutrientes dos meios, mais o lento metabolismo deste agente e o alto grau de contaminantes das amostras torna imprescindível o uso de descontaminantes que, via de regra, são tóxicos para o agente, dificultando o isolamento em amostras com níveis baixos do patógeno; cenário provável no caso do leite devido à mistura com o leite de animais sadios. Mas, a despeito de constituir-se uma importante zoonose transmitida pelo leite, não existe uma metodologia oficial para detecção desse agente em alimentos lácteos. Esses fatos justificam um estudo para avaliar o desempenho de meios líquidos, já utilizados em caros sistemas automatizados para detecção de micobactérias, como alternativa para um enriquecimento seletivo do M. bovis em amostras lácteas. Assim, amostras de leite integral esterilizado e de queijo tipo parmesão foram contaminadas com 10 a 100 UFC/ml ou g de M. bovis AN5 e enriquecidas em dois meios líquidos seletivos (MGIT e MGIT modificado), foram analisados nos dias 0, 7, 14, 21, 24, 28 e 32, mantidas em estufa a 37°C. Nessas datas, uma alíquota foi semeada em meio Stonebrink-Leslie e incubada a 37°C por 60 dias. No leite, houve crescimento exacerbado do M. bovis principalmente no MGIT modificado. No queijo, não foi possível isolar M. bovis de nenhuma amostra em MGIT modificado, devido à contaminação excessiva que deteriorou o meio de cultura. O MGIT modificado foi mais eficaz como enriquecimento para o Mycobacterium bovis, mas foi menos seletivo que o MGIT. Estudos futuros devem concentrar a investigação da curva de crescimento do agente na primeira semana de enriquecimento. / Mycobacterium bovis causes bovine tuberculosis, zoonotic disease raging, probably with low prevalence, throughout the national territory and can be transmitted through the milk. On that date, an aliquot was plated on Stonebrink half- Leslie and incubated at 37 ° C for 60 days. In milk, there was overgrowth of M. bovis mainly in MGIT modified. Cheese, it was not possible to isolate M. bovis no sample in MGIT modified due to excessive contamination it deteriorated the culture medium. The modified MGIT was more effective as enrichment for Mycobacterium bovis, but was less selective than the MGIT. Future studies should focus on investigating the growth curve of the agent in the first week of enrichment.

Due to the high market demand, depredation of habitat, low rate of seed germination in natural environment and slow growth, tissue culture has been presented as an important tool for the production of seedlings of orchids on a large scale, in a reduced period of time and with greater seedling quality over conventional methods. The polysaccharide chitosan has been tested as an additive in the culture medium of some species as it stand out as a growth promoter and by acting as an antioxidant. This study aimed to evaluate the influence of dietary supplementation of chitosan to the nutrient medium in vitro cultivation of Dendrobium Phalaenopsis and Cattleya labiata in liquid and solid medium. Shoots from in vitro seedlings of both species were inoculated into vials containing 30 ml of MS medium with half the ionic strength. The experiment consisted of the combination of three chitosan concentrations (0, 15 and 20 mg.L-1) with nutrient medium in solid and liquid state. Each treatment had four replicates, each with two plants. The experimental design was completely randomized with a factorial scheme of 3x2 (chitosan concentration x physical state of the medium). For 50 days were evaluated the total fresh weight, number of shoots and roots, and were determined activities of superoxide dismutase enzyme (SOD), ascorbate peroxidase (APX) and catalase (CAT). In cultivation in liquid medium, D. phalaenopsis fresh weight showed higher treatment with 20 mg.L-1 of chitosan as the highest number of shoots was observed in both media, at the same concentration. C. labiata also exhibited higher fresh weight and number of shoots in liquid medium, but at the concentration of 15 mg.L-1 of chitosan. The solid medium favored the emission of roots in both species, but inhibited the formation of shoots of C. labiata regardless of the chitosan concentration. The best development of seedlings of both species in liquid medium coincided with increased activity of SOD directly proportional to the chitosan. This activation of SOD was not accompanied by an increase in the activity of APX or CAT, showing the action of chitosan in the dismutation of superoxide without forming hydrogen peroxide. On solid medium, the action of chitosan on the antioxidative mechanism is also provided by the activation of the APX and CAT. Verified the prevalence of the liquid medium for micropropagation of the species studied and confirmed the combination of chitosan in controlling levels of reactive oxygen species in joint action with the antioxidant enzyme system action. / As orquídeas destacam-se entre as ornamentais tropicais, devido à beleza exótica das flores, exuberância de cores e formas. Em virtude da alta demanda do mercado, depredação do habitat, baixa taxa de germinação das sementes em ambiente natural e o lento crescimento, a cultura de tecidos vem se apresentando como uma importante ferramenta para a produção de mudas de orquídeas em larga escala, em período reduzido de tempo e com maior qualidade das mudas em relação aos métodos convencionais. O polissacarídeo quitosana tem sido testado como aditivo no meio de cultura de algumas espécies por se destacar como promotor de crescimento e por sua ação como antioxidante. O presente trabalho teve por objetivo avaliar a influência da suplementação de quitosana ao meio nutritivo no cultivo in vitro de Dendrobium phalaenopsis e Cattleya labiata, em meio líquido e sólido. Brotações provenientes de mudas in vitro de ambas as espécies foram inoculadas em frascos com 30 ml de meio MS com metade da força iônica. O experimento constou da combinação de três concentrações de quitosana (0, 15 e 20 mg L-1 de quitosana) em meio nutritivo em estado sólido e líquido. Cada tratamento contou com quatro repetições, cada uma com duas plantas. O delineamento experimental foi inteiramente casualizado em esquema fatorial de 3x2 (concentrações de quitosana x estado físico do meio). Aos 50 dias avaliaram-se o peso fresco total, número de brotos e de raízes, e foram determinadas as atividades das enzimas superóxido dismutase (SOD), ascorbato peroxidase (APX) e catalase (CAT). No cultivo em meio líquido, D. phalaenopsis apresentou maior peso fresco no tratamento com 20 mg.L-1 de quitosana, enquanto o maior número de brotos foi observado em ambos os meios, nessa mesma concentração. C. labiata também exibiu maior peso fresco e número de brotos em meio líquido, porém na concentração de 15 mg.L-1 de quitosana. O meio sólido favoreceu a emissão de raízes das duas espécies, mas inibiu a formação de brotos de C. labiata, independente das doses de quitosana. O melhor desenvolvimento das mudas das duas espécies em meio líquido coincidiu com aumento da atividade de SOD diretamente proporcional ao da concentração de quitosana. Essa ativação da SOD não foi acompanhada pelo aumento na atividade da APX nem da CAT, evidenciando a ação da quitosana na dismutação do superóxido sem formação de peróxido de hidrogênio. Em meio sólido, a ação da quitosana no mecanismo antioxidativo se deu pela ativação da APX e CAT. Constatou-se a prevalência do meu líquido para a micropropagação das espécies estudadas e confirmou-se a ação combinada da quitosana no controle dos níveis de espécies reativas de oxigênio, em ação conjunta com o sistema enzimático antioxidativo.

A Ghost Fluid Method for Modelling Liquid Jet Atomization

Liquid jet atomisation has a wide variety of application in areas such as injectors in automobile and launch vehicle combustors, spray painting, ink jet printing etc. Understanding physical mechanisms involved in the primary regime of atomisation in combustors is extremely challenging due to the lack of experimental techniques that can reliably provide measurements of gas and liquid velocity fields in this region. Experimental studies have so far been mostly restricted to conditions at atmospheric conditions rather than technically relevant operating pressures. We present a computational fluid dynamics based modelling approach that can capture the evolution of the flow field in the dense primary atomization region of the spray as part of the present thesis work. A fully compressible 3D flow solver is coupled with an interface tracking solver based on level set method. A generalised mathematical formulation for thermodynamic models is implemented in flow solver enabling easy switching between various equations of states. Solvers are parallelised to run on large number of processors and are shown to have good scalability. A modification to the level set method which greatly reduces mass conservation inaccuracies when compared with existing state-of-art baseline schemes has been developed during this work. The Ghost uid Method is used for applying matching conditions at the Interface. The liquid and gas phases are modelled using the perfect gas and Tait equations of state respectively. Several validation studies have been carried out to ensure quantitative accuracy of the solver implemented. Results from canonical Rayleigh Taylor instability simulations shows good agreement with reported results in literature. Finally, results for unsteady evolution of a water-air jet at a liquid to gas density ratio of 10 are shown. Physical mechanisms causing the initial droplet formation are discussed in detail. Droplet feedback is identified as one of the important mechanisms in triggering liquid core instabilities. Comparisons between droplet size distributions obtained from computations are carried out. Vorticity dynamics is used to understand hole and ligament formation from liquid core. Effect of numerical droplets on the simulation results is also looked at in detail.

Multi-scale Modelling of Lamellar Mesophases

Surfactants are amphiphilic molecules which self-assemble at the interface in oil-water-surfactant mixtures such that the hydrophobic part, called tail, stays in oil and the remaining part, called head, resides in hydrophilic en-vironment. Depending upon concentration of individual components, these mixtures form several microphases, such as bilayers, micelles, columnar and lamellar phases. A lamellar phase, at equilibrium, is made up of alternat-ing layers of water and oil separated by surfactants, or of alternate layers of water and surfactant bilayers such that the hydrophilic heads are in contact with water. This equilibrium state is rarely achieved in macroscopic samples due to thermodynamic and kinetic constraints; instead, a lamellar fluid is usually disordered with a large number of defects. These defects have significant effect on the flow behaviour of the lamellar mesophase systems. They are known to alter the flow field, resulting stresses and in turn could get distorted or annihilated by the flow. In present work, we analyse this two way coupling between lamellar structure and flow field. The structural and rheological evolution of an initially disordered lamellar phase system under a shear flow is examined using a mesoscale model based on a free energy functional for the concentration field, which is the scaled difference in the concentration between the hydrophilic and hydrophobic components. Two distinct modes of structural evolution are observed depending only on Peclet number, which ratio of inertial forces to mass diffusivity, in-dependent of system size. At low Peclet number, local domains are formed which are then rotated and stretched by shear. A balance between defect creation and annihilation is reached due to which the system never reaches the equilibrium layer configuration. In the opposite limit, partially formed layers break and reform so as to form a nearly aligned lamellar phase con-figuration with residual defects. Viscosity of lamellar phase system increases with layer moduli, differences in viscosity of individual components, fluidity of the lamellae due to shear banding and defect pinning. These factors however, do not have any effect on alignment mechanism.
