Within the thesis the technology of airconditioning units, the programmable logic controller FA-M3 and its programming tool Widefield are described.

The main aim of this thesis is to design a communication and control card for an electron microscope, eventually for other equipments of the company Delong Instruments a.s., which is dealing with its development and production. This card should replace existing communication card because of its low computational performance. Also, it should replace control and measure card manned with A/D and D/A converters. Thus, the new card will be providing not only the communication with a superior PC and other electronic systems, but also the control of other subsystems of the electron microscope, the determining of processional states and the measuring of internal physical quantities. At the beginning the requirements were determined and the main concept was made with the inclusion of the simple block diagram. It was very important to choose a suitable microcontroller, which will be an intelligence of the whole card and so it will be the most important component. The accent was set mainly on the sufficient computational performance, high modularity of its periphery, good vision of the future innovation and support and, of course, the price. The next requirement was the implementation of the Ethernet, which will be used for the communication between the card and the superior PC. The microcontroller which was chosen have the core architecture ARM Cortex-M3 and it is described in separate chapter. The next part of this thesis is dealing with the main communication standard of the card, which is the Ethernet. After the general description, the Ethernet was mainly discussed in context of the chosen microcontroller. The possibilities of the application of higher layer TCP/IP protocols were also discussed. Implied part of the thesis was the selection of other important components, especially A/D and D/A converters. Its characteristics will have a big effect on the characteristics of the card. In the last and the most important phase the electrical scheme was designed and it was described in detail. This scheme is the main result of this thesis and it is the main document for the future realization. Designed card will be able to process and control 16 differential analogue input signals, 16 differential analogue output signals, 8 digital inputs, 8 simple digital outputs and 4 digital outputs triggered by optocouplers. It will be able to communicate with the superior PC over the Ethernet with the maximum bit rate of 100 Mbit/s, with other electronic cards over the serial line UART through the optical fibers and with other internal and external equipments over the RS-485 and RS-232.

Wider relationships The plot is located south of the historic center of Brno in the city of Brno- Thorny. It is called the "New South Center" along with the land in the northern part behind Opuštěná Street and the eastern lands on which the Central Bus Station is located today The bell ringer. Significant buildings near the solved area is the area of the shopping gallery Vaňkovka s FAIT Gallery and Trinity Palace, then the Šlapanice Municipal Office and the aforementioned bus station. The land has not been built up for a long time due to the existence of floodplains, so it is not contaminated in any way. It is also related to the very long discussion about the relocation of the main train station, the location of which already was decided in 2018. The station should therefore be built by the river. Related to this is the development of the South centers, construction of mixed urban development with functions of housing, services, trade, office and rentable space. This action should expand the center by a busy "boulevard" connecting the visual axis of the railway station with the historical center of Brno. It would then be at the end of the boulevard main train station and integrated main bus station. The existence of the river is very important and the park, which enter this area and give it the unmistakable character of a city-wide importance. Definition and purpose of construction The aim of the work is to the given block urban structure (territorial study of the so - called South Quarter - Trnitá KAM Brno 2019) to design a multifunctional house. Students then choose the location of the object in the area or objects, establish and justify the construction program and process the work to the extent specified below. Proposal will be developed freely according to the regulations set out in the above-mentioned territorial study. Designed by me the building combines the functions of housing with spaces for services in the street parterre of the building. Urban concept The uniqueness of the whole area is direct contact with the historic center, the new main train station and the very close presence of the Svratka River, which is of direct contact in this area borders of my parcel. These facts bring important ideas to this situation and that urban traffic and recreation by the river. It is evident that the river does not have the potential in this area and only so it will have no analogues in Brno, precisely because of the expected high concentration of people traveling on the train, so thanks to the new inhabitants, which should be up to 10,000. The new waterfront would be used by both residents district, as well as people from the area, or visitors commuting by train. That's why I design the whole street parterre buildings as public. I am creating small spaces for a café with a possible use of the anteroom in front of the building itself to sit. The upper floors then serve purely as living areas. The embankment of the Svratka River separates the greenery and the adjoining cycle path to the already very popular track along whole river. Transport solutions The building is transported for cars from the west side of the river to the service two-way calmed communication. At the ends of the building is the entrance to the elevator for passengers cars leading to underground parking garages. Supply of commercial space on.

Purification, serology and pathogenic role of the 110 kilodalton rtx hemolysins of Actinobacillus pleuropneumoniae

<i>Actinobacillus pleuropneumoniae</i> is the etiological agent of contagious swine pleuropneumonia, an economically important disease of the swine industry worldwide. Improved control of this disease requires enhanced understanding of the factors contributing to pathogenesis. The objectives of this study were to investigate the immune response and virulence properties of the 110-kilodalton (110-KDa) hemolysins [hemolysin I (HlyI) and hemolysin II (HlyII)] of <i>A. pleuropneumoniae</i>. Several monoclonal antibodies (MAb) to the hemolysins were developed. An IgGl. MAb (8C2) specific for HlyII, as determined by immunoblotting, was cross-linked to Protein A-Sepharose, and HlyII was purified from serotypes 1 and 5 by immunoaffinity chromatography. An indirect enzyme-linked immunosorbent assay (ELISA) using MAb 8C2, or affinitypurified rabbit IgG to both hemolysins, was developed for detection of swine antibody to one or both hemolysins, respectively. In comparison with the complement fixation test, the ELISA was highly sensitive and specific, and was able to identify animals infected with or exposed to most, if not all, serotypes of <i>A. pleuropneumoniae</i>. Several nonhemolytic mutants of <i>A. pleuropneumoniae</i> serotype 5 were isolated following electroporation of the parent with an hemolysin gene whose open-reading-frame was disrupted with a kanamycin resistance gene. One mutant was characterized for phenotypic and pathogenic properties. Biochemical profiles, growth rate, capsule content, and lipopolysaccharide and whole cell protein electrophoretic profiles of the parent and one of the mutants were similar. The nonhemolytic mutant lacked both HlyI and HlyII proteins in culture supernatant and in whole cell lysates as determined by immunoblot analysis; extracellular and intracellular hemolytic and cytotoxic activity was also absent. The mutant was avirulent in mice and pigs at doses greater than 10 times the lethal dose of the parent. Unlike the parent, the nonhemolytic mutant failed to confer protection against lethal challenge in mice following immunization. Thus, one or both hemolysins are essential for virulence and immunoprotection in <i>A. pleuropneumoniae</i> serotype 5. / Ph. D.

Darstellung und Charakterisierung neuartiger, chiraler, basischer Benzilsäureester mit anticholinerger Wirkung

Basische Benzilsäureester stellen mit ihrer ausgeprägten anticholinergen Wirksamkeit potenzielle Arzneistoffe zur Behandlung der Harninkontinenz, der Ulkuserkrankung und des Morbus Parkinson dar. Von besonderem Interesse sind Benzilsäurevertreter, die neben anticholinergen auch dopaminerge Effekte aufweisen. Wegen ihrer dualistischen Wirkung könnten sie eine neue Klasse von Antiparkinsonica begründen. Aufgrund der vielfältigen Funktionen von Muscarinrezeptoren treten bei wenig selektiv wirksamen Arzneistoffen atropinartige Nebenwirkungen auf. Mit der Entwicklung von Verbindungen, die eine erhöhte muscarinerge Subtypenselektivität besitzen, lassen sich Nebenwirkungen reduzieren. Ziel der Arbeit war eine Wirkungsoptimierung chiraler N-Methyl-4-piperidyl benzilate durch Variation von stereochemischen Parametern und Einführen elektronisch verschiedenartiger Substituenten in die aromatischen Ringe. In Radioligand-Bindungsstudien an M1- bis M3-Rezeptoren wurden die Auswirkungen der sterischen und strukturellen Variationen untersucht. Die Ergebnisse der Bindungsstudien zeigen, dass sich Affinität und Subtypenselektivität durch die absolute Konfiguration des stereogenen Zentrums und die Art der Kernsubstitution modifizieren lassen. Mit Hilfe von Molecular Modelling ist es gelungen, auf Basis der experimentellen Bindungsdaten ein aussagekräftiges Rezeptormodell für N-Methyl-4-piperidyl benzilate zu entwickeln. Sowohl die Affinitätsunterschiede enantiomerer Benzilate als auch die Unterschiede der Rezeptorsubtypenselektivität werden durch das Rezeptormodell umfassend erklärt. / Basic substituted benzilic esters with distinctive anticholinergic effects are potential drugs for the treatment of urinary incontinence, duodenal and gastric ulcers and Parkinson disease. Derivatives of benzilic esters, exhibiting a combination of anticholinergic and dopaminergic effects, are of special interest because, as a consequence of their dualistic effect, they are in a position to form a new class of Antiparkinson drugs. As muscarinic receptor subtypes possess a large variety of functional properties, drugs which show less selectivity on muscarinic receptors exhibit atropine-like side effects. A reduction of these side effects may be achieved by the development of more selective anticholinergic compounds. The objective was to optimise the effect of N-Methyl-4-piperidyl benzilates through a variation of sterical parameters and the introduction of electronically differentiated substituents within the aromatic rings. The effect of sterical and structural variations was investigated in radioligand binding studies on muscarinic receptors (M1 – M3). The results of these binding studies reveal that a modification of affinity and selectivity can be achieved by varying the absolute configuration of the stereogenic center and the properties of the substitution of the aromatic system. The development of a relevant model of the receptor ligand complex for N-Methyl-4-piperidyl benzilates was achieved by molecular modelling on the basis of experimental binding studies. Both the diverse affinity of enantiomeric benzilic esters and the subtype selectivity on muscarinic receptors are comprehensively explained by this model.

Generation problems for finite groups

It can be deduced from the Burnside Basis Theorem that if G is a finite p-group with d(G)=r then given any generating set A for G there exists a subset of A of size r that generates G. We have denoted this property B. A group is said to have the basis property if all subgroups have property B. This thesis is a study into the nature of these two properties. Note all groups are finite unless stated otherwise. We begin this thesis by providing examples of groups with and without property B and several results on the structure of groups with property B, showing that under certain conditions property B is inherited by quotients. This culminates with a result which shows that groups with property B that can be expressed as direct products are exactly those arising from the Burnside Basis Theorem. We also seek to create a class of groups which have property B. We provide a method for constructing groups with property B and trivial Frattini subgroup using finite fields. We then classify all groups G where the quotient of G by the Frattini subgroup is isomorphic to this construction. We finally note that groups arising from this construction do not in general have the basis property. Finally we look at groups with the basis property. We prove that groups with the basis property are soluble and consist only of elements of prime-power order. We then exploit the classification of all such groups by Higman to provide a complete classification of groups with the basis property.

A inserção do nióbio (Nb) como elemento de liga nos aços rápidos iniciou-se com o objetivo de substituir elementos de liga como o vanádio (V) e o tungstênio (W). A sinterização com fase líquida é uma das técnicas de fabricação de aços rápidos obtidos pela metalurgia do pó. O objetivo deste trabalho é a obtenção dos aços rápidos AISI M2 e M3:2 com e sem a adição de carboneto de nióbio (NbC) por compactação uniaxial a frio e sinterização a vácuo com fase líquida. Os pós dos aços rápidos AISI M2 e M3:2 foram processados por moagem de alta energia (MAE) com e sem adição de carboneto de nióbio e compactado uniaxialmente a frio a 700 MPa e sinterizados nas temperaturas de 1240 °C, 1270 °C, para os aços sem a adição de NbC e 1250 °C, para os aços com a adição de 6% de NbC. Os pós dos aços rápidos AISI M2 e M3:2 com a adição de carboneto de nióbio (NbC) processados por uma técnica de moagem de alta energia (MAE) foram caracterizados por difração de raios-X e Microscopia eletrônica de varredura (MEV) e espectroscopia de baixa energia dispersiva de raios-X (EDS) para avaliar a incorporação do reforço do carboneto de nióbio (NbC) aos pós elementares dos aços rápidos. Amostras dos aços rápidos AISI M2 e M3:2 sinterizadas a vácuo com e sem a adição de carboneto de nióbio (NbC) tiveram suas densidades avaliadas e as microestruturas foram caracterizadas por microscopia óptica (MO) para avaliar as microestruturas (porosidades, tamanho e distribuição de carbonetos). Os aços rápidos AISI M2 e M3:2 objetos desse trabalho de pesquisa foram avaliados ainda no que concerne a propriedade mecânica de dureza através de ensaios de dureza Vickers (HV).
