Resource allocation and NFV placement in resource constrained MEC-enabled 5G-NetworksFedrizzi, Riccardo 29 June 2023 (has links)
The fifth-generation (5G) of mobile communication networks are expected to support a large number of vertical industries requiring services with diverging requirements. To accommodate this, mobile networks are undergoing a significant transformation to enable a variety of services to coexist on the same infrastructure through network slicing. Additionally, the introduction of distributed user-plane and multi-access edge computing (MEC) technology allows the deployment of virtualised applications close to the network edge. The first part of this dissertation focuses on end-to-end network slice provisioning for various vertical industries with different service requirements. Two slice provisioning strategies are explored, by formulating a mixed integer linear programming (MILP) problem. Further, a genetic algorithm (GA)-based approach is proposed with the aim to improve search-space exploration. Simulation results show that the proposed approach is effective in providing near-optimal solutions while drastically reducing computational complexity. In a later stage, the study focuses on building a measurement-based digital twin (DT) for the highly heterogeneous MEC ecosystem. The DT operates as an intermediate and collaborative layer, enabling the orchestration layer to better understand network behavior before making changes to the physical network. Assisted by proper AI/ML solutions, the DT is envisioned to play a crucial role in automated network management. The study utilizes an emulated and physical test-bed to gather network key performance indicators (KPIs) and demonstrates the potential of graph neural network (GNN) in enabling closed loop automation with the help of DT. These findings offer a foundation for future research in the area of DT models and carbon footprint-aware orchestration.

A Educação a Distância tem significativa importância no contexto educacional brasileiro, sendo uma das apostas do Ministério da Educação para democratizar o acesso ao ensino superior, pela possibilidade de contribuir sensivelmente com a ampliação e, sobretudo, com a interiorização da oferta de educação em nosso país. A graduação a distância registrou em 2007, conforme último censo, crescimento de 80,7% em relação a 2006, contra 5,4% da educação presencial no mesmo período. É crescente o número de organizações educacionais interessadas em obter credenciamento para a oferta de ensino superior nesta modalidade. No entanto, esses números não garantem sobrevivência a todas as organizações envolvidas neste setor. Como em qualquer outro segmento, as instituições de ensino superior também sofrem com as mudanças que ocorrem no ambiente e precisam enfrentá-las desenvolvendo novas competências organizacionais que lhes confiram vantagens competitivas. Assim, torna-se essencial para as organizações educacionais o desenvolvimento e o aprimoramento de competências próprias que as diferenciem no processo concorrencial, possibilitando que elas se posicionem estrategicamente em diferentes momentos de sua trajetória. Para tanto, a necessidade de aprendizagem torna-se vital como um diferencial competitivo, frente à dinâmica movimentação concorrencial. O processo de aprendizagem, nessa realidade, significa o repensar das ações estratégicas da organização. No presente estudo, foram escolhidas instituições de ensino superior públicas e privadas que passaram por um processo de mudança, tendo em vista a oferta de ensino superior na modalidade a distância. Previous issue date: 2018-04-27 / Conselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPq / This dissertation aims to analyze and discuss the news published in the newspaper, O Estado de São Paulo (OESP) from 1962 to 1973, relating to bilateral technical and financial 'help' established between Brazil and United States of America, in order to reform and revamp the country's education in all degrees of education. Through the signing of agreements between the Ministry of Education and Culture and Agency for International Development (USAID), known as, historically, MEC-USAID agreement. To this end, investigates the agreements and representations, as such idea was organizing in terms of speeches produced by OESP. It has been by chance that the journal played the role of a trainer of public consensus making the necessary arrangements for the so stage of national education. For the development of this research, were analyzed one hundred and eighty-six (186) national editions of OESP, considering the following criteria from observation of information from a database: a) categorization of news by main themes; b) analysis of news and editorials, considering the position of OESP newspaper, before and after the civil-military coup (1964). Were also analyzed legal and official documents on the theme. It was notice that OESP centralized discussions around the agreements formulated for Advise and Planning of Higher Education, and fluctuated between criticism and criminalization of movements opposed to the establishment of such agreements, particularly in relation to the National Union of Students (UNE) / Esta dissertação tem o objetivo de analisar e problematizar as matérias publicadas no jornal, O Estado de S. Paulo (OESP) de 1962 a 1973, relativas aos acordos bilaterais de “ajuda” técnica e financeira estabelecidos entre Brasil e Estados Unidos da América, visando à reforma e a reformulação da educação do país em todos os graus de ensino, por meio da assinatura de acordos firmados entre o Ministério da Educação e Cultura e Agency for International Development (USAID), denominados, historicamente, como “Acordos MEC-USAID”. Para tal, investiga-se as representações lançadas sobre os acordos e como tal ideia foi organizada no plano dos discursos produzidos pelo jornal OESP. Tem-se por hipótese que o periódico desempenhou o papel de formador de consenso público tornando os acordos imprescindíveis para o então estágio da educação nacional. Para o desenvolvimento desta pesquisa foram analisadas cento e oitenta e seis (186) edições nacionais de OESP, pensando os seguintes critérios, a partir de observação de informações de um banco de dados: a) categorização das matérias por temas principais; b) análise das matérias e editoriais, considerando o posicionamento do jornal OESP, antes e depois do golpe civil-militar (1964). Também foram analisados documentos oficiais de ordem jurídica e relativa ao tema. Percebeu-se que OESP centralizou os debates em torno dos acordos formulados para Assessoria e Planejamento do Ensino Superior, e oscilou entre a crítica e criminalização dos movimentos contrários ao estabelecimento dos referidos acordos, principalmente em relação ao movimento estudantil universitário liderado pela União Nacional dos Estudantes (UNE)

Previous issue date: 2011-07-29 / The growing concern with the solid residues management, observed in the last
decade, due to its huge amount and impact, has motivated the search for recycling
processes, where these residues can be reprocessed to generate new products,
enlarging the cycle of materials and energy which are present. Among the polymeric
residues, there is poly (ethylene terephthalate) (PET). PET is used in food
packaging, preferably in the bottling of carbonated beverages. The reintegration of
post-consumer PET in half can be considered a productive action mitigation of
environmental impacts caused by these wastes and it is done through the
preparation of several different products at the origin, i.e. food packaging, with
recycling rates increasing to each year. This work focused on the development and
characterization mechanical, thermal, thermo-mechanical, dynamic mechanical
thermal and morphology of the pure recycled PET and recycled PET composites with
glass flakes in the weight fraction of 5%, 10% and 20% processed in a single screw
extruder,
using the following analytical techniques: thermogravimetry (TG), differential
scanning calorimetry (DSC), tensile, Izod impact, Rockwell hardness, Vicat softening
temperature, melt flow rate, burn rate, dynamic mechanical thermal analysis (DMTA)
and scanning electron microscopy (SEM). The results of thermal analysis and
mechanical properties leading to a positive evaluation, because in the thermograms
the addition of glass flakes showed increasing behavior in the initial temperatures of
thermal decomposition and melting crystalline, Furthermore was observed growing
behavior in the mechanical performance of polymer composites, whose
morphological structure was observed by SEM, verifying a good distribution of glass
flakes, showing difference orientation in the center and in the surface layer of test
body of composites with 10 and 20% of glass flakes. The results of DMTA Tg values
of the composites obtained from the peak of tan ? showed little reductions due to
poor interfacial adhesion between PET and recycled glass flakes. / A crescente preocupa??o com a gest?o de res?duos s?lidos, observada na ?ltima
d?cada, em virtude de sua enorme quantidade e impacto ambiental, tem motivado a
busca por processos de reciclagem, onde estes res?duos podem ser reprocessados
de modo a gerarem novos produtos, ampliando o ciclo dos materiais e energia neles
contidos. Dentre os res?duos polim?ricos, destaca-se o poli(tereftalato de etileno)
(PET). O PET ? utilizado em embalagens de alimentos, preferencialmente, no
envase de bebidas carbonatadas. A reintegra??o do PET p?s-consumo ao meio
produtivo pode ser considerada uma a??o mitigat?ria dos impactos ambientais
causados por estes res?duos e j? ? realizada por meio da obten??o de diversos
produtos diferentes ao da origem, ou seja, embalagens para alimento, com taxas de
reciclagem crescente a cada ano. Este trabalho focou o desenvolvimento e a
caracteriza??o mec?nica, t?rmica, termo-mec?nica, termo-din?mico-mec?nica e
morfol?gica do PET reciclado processado e dos comp?sitos de PET reciclado com
flocos de vidro na fra??o em peso de 5%, 10% e 20% processados em uma
extrusora rosca simples, utilizando as seguintes t?cnicas de an?lise:
termogravimetria (TG), calorimetria explorat?ria diferencial (DSC), tra??o uniaxial,
impacto Izod, dureza Rockwel, temperatura de amolecimento Vicat, ?ndice de fluidez,
taxa de queima, an?lise termo-din?mico-mec?nica (DMTA) e microscopia eletr?nica
de varredura (MEV). Os resultados das an?lises t?rmicas e das propriedades
mec?nicas conduziram a uma avalia??o positiva, pois nos termogramas as adi??es
dos flocos de vidro mostraram comportamento crescente nas temperaturas inicial de
decomposi??o t?rmica e de fus?o cristalina, al?m disso, observou-se
comportamento crescente no desempenho mec?nico dos comp?sitos polim?ricos,
cuja estrutura morfol?gica foi observada por MEV, verificando uma boa distribui??o
dos flocos de vidro, apresentando diferen?a na orienta??o no centro e na camada
superficial do corpo de prova dos comp?sitos com 10 e 20% de flocos de vidro. Nos
resultados de DMTA os valores de Tg dos comp?sitos obtidos a partir do pico de tan
?, apresentaram pequenas redu??es, devido a pobre ades?o interfacial entre o PET
reciclado e os flocos de vidro.

Previous issue date: 2013-12-09 / Coordena??o de Aperfei?oamento de Pessoal de N?vel Superior / As most current studies, reinforced plastics have been, in recent years, a viable alternative in building structural elements of medium and large, since the lightness accompanied by high performance possible. The design of hybrid polymer composites (combination of different types of reinforcements) may enable structural applications thereof, facing the most severe service conditions. Within this class of composite materials, reinforced the underlying tissues hybrid high performance are taking space when your application requires high load bearing and high rigidity. The objective of this research work is to study the challenges in designing these fabrics bring these materials as to its mechanical characterization and fracture mechanisms involved. Some parameters associated with the process and / or form of hybridization stand out as influential factors in the final performance of the material such as the presence of anisotropy, so the fabric weave, the process of making the same, normative geometry of the specimens, among others. This sense, four laminates were developed based hybrid reinforcement fabrics involving AS4 carbon fiber, kevlar and glass 49-E as the matrix epoxy vinyl ester resin (DERAKANE 411-350). All laminates were formed each with four layers of reinforcements. Depending on the hybrid fabric, all the influencing factors mentioned above have been studied for laminates. All laminates were manufactured industrially used being the lamination process manual (hand-lay-up). All mechanical characterization and study of the mechanism of fracture (fracture mechanics) was developed for laminates subjected to uniaxial tensile test, bending in three and uniaxial compression. The analysis of fracture mechanisms were held involving the macroscopic, optical microscopy and scanning electron microscopy / Conforme estudos mais atuais, os pl?sticos refor?ados v?m sendo, nos ?ltimos anos, uma alternativa vi?vel na constru??o de elementos estruturais de m?dio e grande porte, desde que a leveza acompanhada de alto desempenho seja poss?vel. its behavior related to many conditions of loading and service, as well as under several ways
of connections involved in mechanisms of structural projects. Within these project conditions
are highlighted the presence of geometrical discontinuities in the area of cross and
longitudinal sections of structural elements and environmental conditions of work like UV
radiation, moisture, heat, leading to a decrease in final mechanical response of the material. In
this sense, this thesis aims to develop studies detailed (experimental and semi-empirical
models) the effects caused by the presence of geometric discontinuity, more specifically, a
central hole in the longitudinal section (with reduced cross section) and the influence of
accelerated environmental aging on the mechanical properties and fracture mechanism of
FGRP composite laminates under the action of uniaxial tensile loads. Studies on
morphological behavior and structural degradation of composite laminates are performed by
macroscopic and microscopic analysis of affected surfaces, in addition to evaluation by the
Measurement technique for mass variation (TMVM). The accelerated environmental aging
conditions are simulated by aging chamber. To study the simultaneous influence of
aging/geometric discontinuity in the mechanical properties of composite laminates, a semiempirical
model is proposed and called IE/FCPM Model. For the stress concentration due to
the central hole, an analisys by failures criteria were performed by Average-Stress Criterion
(ASC) and Point-Stress Criterion (PSC). Two polymeric composite laminates, manufactured
industrially were studied: the first is only reinforced by short mats of fiberglass-E (LM) and
the second where the reinforced by glass fiber/E comes in the form of bidirectional fabric
(LT). In the conception configurations of laminates the anisotropy is crucial to the final
mechanical response of the same. Finally, a comparative study of all parameters was
performed for a better understanding of the results. How conclusive study, the characteristics
of the final fracture of the laminate under all conditions that they were subjected, were
analyzed. These analyzes were made at the macroscopic level (scanner) microscope (optical
and scanning electron). At the end of the analyzes, it was observed that the degradation
process occurs similarly for each composite researched, however, the LM composite
compared to composite LT (configurations LT 0/90? and LT ?45?) proved to be more
susceptible to loss of mechanical properties in both regarding with the central hole as well to
accelerated environmental aging / A crescente demanda na utiliza??o dos materiais comp?sitos torna imprescind?vel uma melhor

afetadas, al?m da avalia??o mediante a T?cnica de Media??o de Varia??o de Massa (TMVM).
As condi??es de envelhecimento ambiental acelerado ser?o simuladas atrav?s da c?mara de
envelhecimento. Para avalia??o da influ?ncia simult?nea do envelhecimento/descontinuidade
geom?trica nas propriedades mec?nicas dos laminados comp?sitos, um modelo semi-empirico
? proposto e denominado de Modelo IE/FCPM. Para o caso da presen?a da concentra??o de
tens?o devido ao furo conc?ntrico, a mesma ? avaliada a partir do uso dos crit?rios de falhas
da Tens?o M?dia (Average-Stress Criterion) e da Tens?o Pontual (Point-Stress Criterion).
Dois laminados comp?sitos polim?ricos, confeccionados industrialmente, foram estudados: o
primeiro ? unicamente refor?ado com mantas de fibras curtas de vidro/E (LM) e o segundo
onde o refor?o de fibras de vidro/E vem na forma de tecido bidirecional (LT). Na concep??o
das configura??es dos laminados a presen?a da anisotropia ? fundamental para a resposta
mec?nica final dos mesmos. Por fim, um estudo comparativo entre todos os par?metros foi

Previous issue date: 2017-03-24 / This work presents a historical background of the Basic Curriculum for the Municipal Public School of AMOP and the origin of this Association. To understand what were the influences to produce this material and trying to find a solution the following problem: how and why an association of Municipalities, as a representative of the state, capitalist social development, published a curriculum with a dialectical conception of education? We started with information from the document itself, which was created under the coordination of AMOP Pedagogical Department (2003), created after the extinction of ASSOESTE (2002), an entity created by the MEC / OAS Special Multinational Project in 1980. Our hypothesis was that the influence in the creation of education present in the Curriculum has its origin, largely, in the work accomplished by ASSOESTE. Therefore, the research sought historical antecedents of this referential, in order to know the structure of the West Region and the events that implied in the transformation of the area, analyzing the impacts caused in education; To know the origin of AMOP, analyzing the historical and political context of its creation and its role in regional development; To know the MEC / OAS Special Multinational Project, and analyzing the historical and political context in which it was drawn up and executed, as well as some of its contributions to the education of the Western Region; To know the history of ASSOESTE, the context of their foundation and some of their contributions to the education of the West, its process of extinction and consequent creation of the Pedagogical Department of AMOP. To the completion of this dissertation, readings of historical texts, bibliographies, minutes, statutes, and laws were made, and this dissertation was constituted by exploratory and investigative research, based on the assumptions that reality is dynamic, in constant development, and that men make history, and do it according to the physical conditions at their disposal. The results obtained with this work indicate that the creation of AMOP occurred for strategic military government reasons, and it played a fundamental role in consolidation of the conservative modernization of agriculture that caused great changes in the region, as well as the construction of Itaipu. To promoting search for solutions to the problems caused by these two phenomena, the Special Project MEC / OAS was created and implemented, which had great contributions to education, which one of them being the creation of ASSOESTE. The research on ASSOESTE allowed us to prove our hypothesis that was the work carried out by ASSOESTE that influenced the beginning of the education of the AMOP’s curriculum. It also allowed us to understand that its extinction was not only due to financial issues, but also to the political ideology assumed by its Pedagogical Department. / Este trabalho apresenta antecedentes históricos do Currículo Básico para a Escola Pública Municipal, da AMOP, e a gênese desta Associação, a fim de conhecermos quais foram as influências para a produção desse material, procurando responder ao seguinte problema: como e por que uma associação de municípios, enquanto representação do Estado, de formação social capitalista, publicou um currículo com uma concepção dialética de educação? Partimos de informações trazidas no próprio documento, engendrado sob a coordenação do Departamento Pedagógico da AMOP (2003), criado após a extinção da ASSOESTE (2002), entidade criada pelo Projeto Especial Multinacional MEC/OEA, em 1980. Nossa hipótese era de que a influência da concepção de educação presente no Currículo tem origem, em grande medida, no trabalho realizado pela ASSOESTE. Então, a pesquisa buscou antecedentes históricos desse referencial, a fim de: conhecer a formação da Região Oeste e os eventos que implicaram na transformação do espaço, analisando os impactos causados na educação; conhecer a origem da AMOP, analisando o contexto histórico e político de sua criação, e o seu papel no desenvolvimento regional; conhecer o Projeto Especial Multinacional MEC/OEA, analisando, igualmente, o contexto histórico e político em que fora elaborado e executado, bem como algumas de suas contribuições para a educação da Região Oeste; conhecer a história da ASSOESTE, o contexto de sua fundação, algumas de suas contribuições para a educação do Oeste, seu processo de extinção e consequente criação do Departamento Pedagógico da AMOP. Para tanto, foram realizadas leituras de textos históricos, bibliográficos, atas, estatutos, leis, constituindo-se essa dissertação pela pesquisa exploratória e investigativa, partindo dos pressupostos de que a realidade é dinâmica, estando em constante devir, e, ainda, de que os homens fazem a história, e a fazem de acordo com as condições materiais de que dispõem. Os resultados alcançados com esse trabalho apontam que a criação da AMOP ocorreu por motivos estratégicos do governo militar, tendo ela papel fundamental para a consolidação da modernização conservadora da agricultura que provocou grandes mudanças na região, assim como a construção da Itaipu. Para promover a busca de soluções para os problemas causados por esses dois fenômenos, foi elaborado e executado o Projeto Especial MEC/OEA, o qual teve grandes contribuições para a educação, sendo uma delas a criação da ASSOESTE. A pesquisa sobre a ASSOESTE nos possibilitou comprovar nossa hipótese, de que o trabalho realizado por ela influenciou a concepção de educação do Currículo da AMOP. Nos proporcionou, ainda, compreender que sua extinção não se deu apenas por questões financeiras, mas também pela ideologia política assumida por seu Departamento Pedagógico.

User equipment based-computation offloading for real-time applications in the context of Cloud and edge networks / Délestage de calcul pour des applications temps-réel dans le contexte du Cloud et du edgeMessaoudi, Farouk 16 April 2018 (has links)
Le délestage de calcul ou de code est une technique qui permet à un appareil mobile avec une contrainte de ressources d'exécuter à distance, entièrement ou partiellement, une application intensive en calcul dans un environnement Cloud avec des ressources suffisantes. Le délestage de code est effectué principalement pour économiser de l'énergie, améliorer les performances, ou en raison de l'incapacité des appareils mobiles à traiter des calculs intensifs. Plusieurs approches et systèmes ont été proposés pour délester du code dans le Cloud tels que CloneCloud, MAUI et Cyber Foraging. La plupart de ces systèmes offrent une solution complète qui traite différents objectifs. Bien que ces systèmes présentent en général de bonnes performances, un problème commun entre eux est qu'ils ne sont pas adaptés aux applications temps réel telles que les jeux vidéo, la réalité augmentée et la réalité virtuelle, qui nécessitent un traitement particulier. Le délestage de code a connu un récent engouement avec l'avènement du MEC et son évolution vers le edge à multiple accès qui élargit son applicabilité à des réseaux hétérogènes comprenant le WiFi et les technologies d'accès fixe. Combiné avec l'accès mobile 5G, une pléthore de nouveaux services mobiles apparaîtront, notamment des service type URLLC et eV2X. De tels types de services nécessitent une faible latence pour accéder aux données et des capacités de ressources suffisantes pour les exécuter. Pour mieux trouver sa position dans une architecture 5G et entre les services 5G proposés, le délestage de code doit surmonter plusieurs défis; la latence réseau élevée, hétérogénéité des ressources, interopérabilité des applications et leur portabilité, la consommation d'énergie, la sécurité, et la mobilité, pour citer quelques uns. Dans cette thèse, nous étudions le paradigme du délestage de code pour des applications a temps réel, par exemple; les jeux vidéo sur équipements mobiles et le traitement d'images. L'accent sera mis sur la latence réseau, la consommation de ressources, et les performances accomplies. Les contributions de la thèse sont organisées sous les axes suivants : Étudier le comportement des moteurs de jeu sur différentes plateformes en termes de consommation de ressources (CPU / GPU) par image et par module de jeu ; Étudier la possibilité de distribuer les modules du moteur de jeu en fonction de la consommation de ressources, de la latence réseau, et de la dépendance du code ; Proposer une stratégie de déploiement pour les fournisseurs de jeux dans le Cloud, afin de mieux exploiter les ressources, en fonction de la demande variable en ressource par des moteurs de jeu et de la QoE du joueur ; Proposer une solution de délestage statique de code pour les moteurs de jeu en divisant la scène 3D en différents objets du jeu. Certains de ces objets sont distribués en fonction de la consommation de ressources, de la latence réseau et de la dépendance du code ; Proposer une solution de délestage dynamique de code pour les moteurs de jeu basée sur une heuristique qui calcule pour chaque objet du jeu, le gain du délestage. En fonction de ce gain, un objet peut être distribué ou non ; Proposer une nouvelle approche pour le délestage de code vers le MEC en déployant une application sur la bordure du réseau (edge) responsable de la décision de délestage au niveau du terminal et proposer deux algorithmes pour prendre la meilleure décision concernant les tâches à distribuer entre le terminal et le serveur hébergé dans le MEC. / Computation offloading is a technique that allows resource-constrained mobile devices to fully or partially offload a computation-intensive application to a resourceful Cloud environment. Computation offloading is performed mostly to save energy, improve performance, or due to the inability of mobile devices to process a computation heavy task. There have been a numerous approaches and systems on offloading tasks in the classical Mobile Cloud Computing (MCC) environments such as, CloneCloud, MAUI, and Cyber Foraging. Most of these systems are offering a complete solution that deal with different objectives. Although these systems present in general good performance, one common issue between them is that they are not adapted to real-time applications such as mobile gaming, augmented reality, and virtual reality, which need a particular treatment. Computation offloading is widely promoted especially with the advent of Mobile Edge Computing (MEC) and its evolution toward Multi-access Edge Computing which broaden its applicability to heterogeneous networks including WiFi and fixed access technologies. Combined with 5G mobile access, a plethora of novel mobile services will appear that include Ultra-Reliable Low-latency Communications (URLLC) and enhanced Vehicle-toeverything (eV2X). Such type of services requires low latency to access data and high resource capabilities to compute their behaviour. To better find its position inside a 5G architecture and between the offered 5G services, computation offloading needs to overcome several challenges; the high network latency, resources heterogeneity, applications interoperability and portability, offloading frameworks overhead, power consumption, security, and mobility, to name a few. In this thesis, we study the computation offloading paradigm for real-time applications including mobile gaming and image processing. The focus will be on the network latency, resource consumption, and accomplished performance. The contributions of the thesis are organized on the following axes : Study game engines behaviour on different platforms regarding resource consumption (CPU/GPU) per frame and per game module; study the possibility to offload game engine modules based on resource consumption, network latency, and code dependency ; propose a deployment strategy for Cloud gaming providers to better exploit their resources based on the variability of the resource demand of game engines and the QoE ; propose a static computation offloading-based solution for game engines by splitting 3D world scene into different game objects. Some of these objects are offloaded based on resource consumption, network latency, and code dependency ; propose a dynamic offloading solution for game engines based on an heuristic that compute for each game object, the offloading gain. Based on that gain, an object may be offloaded or not ; propose a novel approach to offload computation to MEC by deploying a mobile edge application that is responsible for driving the UE decision for offloading, as well as propose two algorithms to make best decision regarding offloading tasks on UE to a server hosted on the MEC.

Molecular characterisation of methicillin-resistant Staphylococcus aureus (MRSA) from South AfricaOosthuysen, Wilhelm Frederick 03 June 2008 (has links)
ABSTRACT
Few antibiotics are left that are effective against methicillin-resistant Staphylococcus
aureus (MRSA) and even strains resistant to these agents have been isolated. Previous
studies have identified five distinct MRSA clonotypes, which are present globally. No
comprehensive national study has previously been undertaken to investigate the MRSA
types in South Africa, and this study was aimed at elucidating the genotypic population
structure of South African MRSA isolates. SmaI digested genomic DNA, separated by
pulsed-field gel electrophoresis, was used to characterise 349 S. aureus isolates, obtained
from various state and private diagnostic laboratories. PFGE results were complemented
with those of spa typing and staphylococcal cassette chromosome mec (SCCmec) typing
results. Two-hundred-and-five different PFGE patterns were identified, which were
grouped into twenty-four clusters. Three were major lineages, containing more than 20%
of the isolates with a similarity cut-off of 70%. Only thirty-seven spa types were identified
(fourteen novel spa types), which clustered into six spa-Clonal Complexes after BURP
analysis. SCCmec types I-IV were identified, including variants of each type. Data
suggest that the Archaic clone (RSA05), oldest of the epidemic clones, represents one of
the major clones in South Africa. Strains that were part of this complex (n=98 (28.2%);
t064; SCCmec type I-pls) clustered together with strain E2125/ATCC BAA-38 (t051;
SCCmec type I). Another major complex, RSA16 (n=90 (25.7%); t012; SCCmec type
II/IIB) possessed a single-locus variant (SLV) spa type and the same or a SLV SCCmec
types as EMRSA-16 (t018; SCCmec type II). The third major complex, RSA03 (n=74
(21.2%); t037; SCCmec type III/IIIE), had similar spa and SCCmec types to control strainANS46 (t037; SCCmec type III). One MRSA and twelve MSSA isolates were also
identified as carrying genes for the toxin Panton-Valentine leukocidin, which was
confirmed by DNA nucleotide sequencing.
