A Anatomia comparada de mamíferos é tema de grande interesse em pesquisas e tem como objetivo buscar conhecimentos que possam auxiliar o entendimento do binômio unidadevariedade. O conhecimento da Anatomia de qualquer animal silvestre é de importância capital para a sua preservação e proteção (RIBEIRO, 2002). Os primatas do novo mundo estão distribuídos na Região Neotropical das Américas Central e do Sul, (RYLANDS; MITTERMEIER; RODRÍGUEZ-LUNA, 1997). O Cebus apella, animal das matas do continente sul-americano, geogra. camente distribuído por quase todo o Brasil, adapta-se bem à vida em cativeiro, onde se reproduz com facilidade (HILL, 1960). O estudo da macro- e micro-anatomia dos órgãos genitais masculinos do macaco Cebus apella é de suma importância para estabelecer os parâmetros morfológicos comparativos com outros primatas. Neste estudo, foram utilizados oito exemplares de macacos Cebus apella, machos, adultos (de aproximadamente sete anos de idade). Estabeleceu-se um padrão anatômico da espécie, e os resultados então obtidos permitem discutir a sua correlação morfo-funcional. Os dados do Cebus apella foram comparados aos de outros primatas já estudados (chimpanzé, gorila, babuíno, rhesus e outros). / Comparative Anatomy of mammals is theme of relevant interest in researches and has the aim to achieve knowledge which could help the understanding of the unit-variety binomial. The knowledge of the Anatomy of any wild animal is of capital importance for his preservation and protection (RIBEIRO, 2002). New world primates are distributed in the Neotropical Region of the Central and South America (RYLANDS; MITTERMEIER; RODRÍGUEZ-LUNA, 1997). The Cebus apella, animal of the South American continent forests, geographically distributed for almost the whole Brazil, is well adapted to the captive life, where reproduces with easiness (HILL, 1960). The macro and micro-anatomical study of the male genital organs of the monkey Cebus apella is of great importance to establish the comparative morphologic parameters with other primates. In this study, eight adult male Cebus apella monkeys (about seven years old) were used and an anatomical pattern was established for the specie. The obtained data allow discussing his morfofunctional correlation. Results for the Cebus apella monkey were compared with that ones of other primates already studied (chimpanzee, gorilla, baboon, rhesus and others).

A Bioacoustica oferece ferramentas úteis para diversas ciências e práticas na utilização acadêmica, científica, aplicada e conservação da vida animal. Nossa hipótese é que as chamadas naturais, como o rugido de longo alcance do bugios preto, atuam como elemento de enriquecimento ambiental, se a sua execução aumenta a exibição de um comportamento natural em cativeiro. O bugio faz um uso diário do rugido e sua emissão é conhecida por evocar respostas de outros bandos de bugio em vida livre. Para testar a eficácia dos rugidos em melhorar o bem-estar foi realizado um experimento no Bosque e Zoológico Fábio Barreto, no município de Ribeirão Preto, onde treze indivíduos (6 fêmeas e 7 machos) viviam em cinco recintos adjacentes. Eles tiveram contato visual e acústico entre eles e com outras duas espécies de primatas e outros grupos de vida livre de bugio preto. Adotoamos o delineamento experimental ABABA, em que fases controle A foram intercaladas com o tratamento (playback de rugido) nas fases B, duas vezes ao dia (manhã e tarde). Na primeira fase B, o estímulo foi sempre tocado no mesmo horário (previsível) e, na segunda fase tratamento, horários aleatórios foram adotados (imprevisível). Nós avaliamos as alterações na duração relativa de 6 comportamentos entre as fases A e B, entre sexo e em ambos os períodos, com o teste ANOVA de medidas repetidas. Nós notamos um aumento no comportamento natural da espécie em fases de tratamento, como alerta, atenção dirigida, e interações sociais, juntamente com a diminuição de comportamentos estereotipados, sugerindo melhoria do bem-estar. As emissões naturais de rugidos também aumentaram. Concluímos que ambos os tratamentos, previsível e imprevisível, melhoraram o bem-estar e propomos a utilização da reprodução de chamadas específicas de especies como uma nova categoria de enriquecimento ambiental. A resposta dos outros primatas nos levou a concluir que houve um efeito sobre animais vizinhos que pode ser benéfico ou prejudicial de acordo com a espécie e a condição psicológica dos animais. / The Bioacoustics offers useful tools for diverse academic, scientific and applied sciences and practices in use and conservation of animal life. We hypothesized that natural call, as the loud roar of the black-howler monkey, would act as environmental enrichment element if its execution increased the display of natural behavior in captivity. The howler-monkey makes an everyday use of the roar and its emission is known to evoke free-living howler-monkeys to call back in response. To test the effectiveness of roars in improving welfare we conducted an experiment in Bosque e Zoológico Fábio Barreto, in the municipality of Ribeirão Preto where thirteen individuals (6 females and 7 males) were living in five adjacent enclosures. They had visual and acoustic contact between them and with two other primate species and a free-living black-howler monkey group. We adopted the ABABA experimental design, in which control phases A were interspersed with treatment (roar playback) phases B, two times a day (morning and afternoon). In the first B phase, the stimulus was always played at the same time (predictable) and, in the other different random schedules were adopted (unpredictable). We evaluate changes in relative duration of 6 behaviors from A to B phases, between sex and in both periods with a repetitive factorial ANOVA. We reported increase in natural behavior of the species in treatment phases, as alert, oriented attention, and social interactions, coupled with the decrease of stereotyped behaviors, suggesting welfare improvement. Thus, natural emissions of roars also increased. We concluded that both predictable and unpredictable routines improved welfare and we propose the use of playback of specie-specific calls as a new category of environmental enrichment. The answer of the other primates led us to conclude that there was a neighborhood effect that can be beneficial or harmful according to the species and the psychological condition of the animals.

Notre environnement est complexe et changeant, ce qui apporte de l'incertitude dans les décisions de tous les jours. La capacité de détecter et résoudre l'incertitude est cruciale pour un comportement flexible et adapté. Notre hypothèse est que l'efficacité et la flexibilité comportementale en situation d'incertitude dépendent de la façon dont l'individu a appris à apprendre. Dans une 1ère étude, trois singes ont acquis un learning set pour une tâche aux règles stochastiques et changeantes. Leur réactivité aux évènements inattendus a augmenté lors de l'apprentissage, suivant l'évolution du degré d'incertitude environnementale. Cela a permis un transfert sans coût à une tâche plus complexe partageant la même structure, suggérant que les singes ont appris à apprendre la structure statistique de l'environnement. Nous avons ensuite étudié les mécanismes cérébraux sous-jacents à ce comportement flexible. Deux animaux ont reçu un implant d'électrocorticographie, sur les aires frontales et pariétales. Nous montrons d'abord, avec les données d'un animal, que des potentiels évoqués au feedback sont sensibles à la valence et au degré de surprise du feedback, et prédisent la stratégie à venir. Ensuite, nous présentons des résultats préliminaires montrant que des oscillations dans les bandes beta et thêta sont présentes au moment du feedback et de la décision, et que leur puissance est modulée de manière différente par les facteurs de la tâche. Ces résultats contribuent à révéler la complexité du réseau frontal pour la flexibilité comportementale, et ouvrent la voie à de nouvelles expériences pour comprendre comment ces mécanismes sont façonnés au cours du processus d'apprendre à apprendre / Our environment is both complex and changing, which triggers uncertainty in every decision we make. The ability to detect and solve the resulting uncertainty is crucial for adapted and flexible behavior. Our hypothesis is that behavioral efficiency and flexibility in an uncertain environment depend on the way the agent has learnt to learn. In a first study, 3 macaque monkeys developed a learning-set for a task with stochastic and changing rules. Monkey’s reactivity to unexpected feedback increased across learning and paralleled the evolution of the degree of environmental uncertainty. This enabled them to transfer, without cost, to a more complex task with the same structure, suggesting that they learned to learn the statistical structure of the environment. We then studied the cerebral mechanisms underpinning this flexible behavior. Two animals were implanted with an electrocorticography implant over the frontal and parietal areas. We first showed, using data from one animal, that feedback related potentials were sensitive to feedback valence and unexpectedness, and predictive of the upcoming behavioral strategy. Then, we present preliminary results showing that oscillations in the beta and theta bands can be recorded at the time of feedback and at the time of decision, and that their power is modulated differently depending on the various task factors. These results contribute to reveal the complexity of the frontal cortical network enabling behavioral flexibility and open new horizons for future research to understand how these mechanisms are shaped throughout the learning to learn process

Dispersão e estrutura social de macacos-prego (Sapajus nigritus) do Parque Estadual Carlos Botelho, São Paulo / Dispersal and social structure in black capuchin monkeys (Sapajus nigritus) of Carlos Botelho State Park, São PauloTokuda, Marcos 20 February 2013 (has links)
Padrões de assimetria sexual na dispersão e relações de parentesco são fatores intimamente relacionados e considerados fundamentais para a compreensão da estrutura social dos primatas. Apesar da relevância desses dois assuntos, pouco se sabe sobre como eles afetam o comportamento individual e a estrutura social nos primatas neotropicais. Assim, essa pesquisa teve como objetivos: determinar o padrão de dispersão de uma população selvagem de macacosprego (Sapajus nigritus) por meio de análises genéticas e examinar o efeito do parentesco sobre a estrutura social dos grupos de S. nigritus. Esta pesquisa foi realizada no Parque Estadual Carlos Botelho, localizado no município de São Miguel Arcanjo/SP. Todo material genético foi obtido através de amostras fecais dos indivíduos adultos e subadultos de três grupos sociais. Após a extração do DNA, parte dele foi amplificada através da técnica da Reação em Cadeia da Polimerase. Para as análises genéticas utilizamos marcadores moleculares do tipo microssatélites. Os métodos de amostragem por varredura instantânea e ad libitum foram utilizados para o registro do comportamento dos indivíduos adultos de dois grupos. Os dados genéticos obtidos nesta tese indicaram que ambos os sexos dispersam, pois: 1) o grau de parentesco intragupo entre machos não diferiu do grau de parentesco entre fêmeas, 2) não houve diferenciação genética ao analisar a distribuição das frêquencias alélicas de machos e de fêmeas, e 3) não houve diferença entre machos e fêmeas quanto à probabilidade de terem nascido dentro dos grupos sociais nos quais foram amostrados. Os resultados de associações espaciais e interações sociais indicaram que as fêmeas adultas não formam fortes relações sociais entre si, sendo pouco afiliativas. Os machos adultos estabelecem fracas relações sociais, que podem ser classificadas como tolerantes e caracterizadas pela ausência de catação e pela baixa frequência de interações agonísticas. As relações sociais mais fortes observadas dentro de cada grupo social foram entre machos e fêmeas. Além de machos e fêmeas associarem-se espacialmente e catarem-se mais do que as díades compostas somente por fêmeas ou por machos, eles também interagiram de maneira agonística numa menor frequência. As díades consideradas como aparentadas não demonstraram ser mais afiliativas ou manter maior proximidadade espacial quando comparadas com as díades não aparenteadas. O baixo grau de parentesco/familiaridade parece ser um fator importante como uma explicação geral para as fracas relações sociais de fêmeas no PECB. No entanto, em termos individuais, aparentemente, esse fator exerceu pouca influência sobre as relações sociais entre as fêmeas e entre os machos. Os resultados apresentados nesta tese ampliam o conhecimento sobre relações sociais em primatas neotropicais e os possíveis fatores que as afetam. O padrão de dispersão e a estrutura social são elementos do sistema social de primatas neotropicais altamente flexíveis, variando entre populações da mesma espécie ou mesmo entre grupos de uma mesma população. Além disso, a alegada assimetria sexual na dispersão talvez não seja passível de generalização entre as populações de Sapajus, e talvez dispersão de ambos os sexos nesse gênero possa ser mais comum do que previamente se considerava / Sex-biased dispersal patterns and kinship are related factors extremely important to understand the social structure of primates. In spite of their importance, little is known about how these factors affect individual behavior and the social structure of neotropical primates. Therefore, the aims of this research were: to determine the dispersal pattern of a wild population of black capuchin monkeys (Sapajus nigritus) through genetic analyses, and to verify the effect of kinship on the social structure of groups of S. nigritus. This research was conducted at Carlos Botelho State Park, in the municipality of São Miguel Arcanjo/SP. We used DNA from fecal samples of adult and subadult members from three wild social groups. DNA was amplified by Polymerase Chain Reaction and microsatellite molecular markers were used. Behavioral data were collected systematically for two groups, and we used scan sampling and ad libitum methods to record the behavior of adults. Genetic data indicated that both sexes disperse from their natal groups, since: 1) relatedness between males was not statistically different from relatedness between females, 2) there was no difference between males and females in population genetic differentiation, and 3) there was no difference between males and females in the probability of being born in the group from which they were sampled. The results of the spatial association and social interactions analysis indicate that females are little affiliative and do not form strong relationships among themselves. Adult males established weak relationships, being classified as tolerant, and characterized by no grooming interactions and by low frequency of agonism. The strongest social relationships observed within each group were between males and females. They associated and groomed more than female dyads and male dyads. Moreover, there were fewer agonistic interactions between males and females as compared to same sex dyads. Related and unrelated dyads showed similar rates of association and affiliative behavior. The low level of relatedness/familiarity might be an important factor contributing to the weak social relationship among females in the PECB. However, at the individual level, kinship had low influence on male and female social relationships. The results presented here extended our knowledge about social relationships of neotropical primates and about the factors that influence the relationships. The dispersal pattern and the social structure are flexible elements of social systems, and can vary among populations of the same species or among groups of the same population. In addition, male-biased dispersal is not a general characteristic for all populations of Sapajus, and dispersal by both sexes might be more commom than previously thought

Habituation and Desensitization as Methods for Reducing Fearful Behavior in Singly-Housed Rhesus MacaquesClay, Andrea Wolstenholme 20 July 2007 (has links)
Operant conditioning using positive reinforcement techniques has been used extensively in the management of nonhuman primates in both zoological and laboratory settings. Based on a large body of previous research that demonstrates the utility of such techniques in reducing stress, abnormal behavior, and aggression, this research project was intended to develop and test the usefulness of habituation and counter-conditioning techniques in reducing the fear-responses of singly-housed male rhesus macaques living in the laboratory environment. Additionally, we investigated the variable of temperament as it relates to the reduction of fear-responsivity and overall training success. Based on a Wilcoxon Matched-Pairs Sign Test, we found that animals exposed to desensitization training were significantly likely to show a reduction in the rate at which they engaged in cringing toward humans (exact significance = .016, one-tailed, N ties = 6), cringing in general (exact significance = .016, one-tailed, N ties = 6), and in stress-related behaviors (exact significance = .016, one-tailed, N ties = 6). Animals exposed to basic husbandry training or exposed to no training at all were not significantly likely to show a reduction in the rates of these behaviors. When these same behaviors were analyzed in terms of duration of behavior, desensitization-exposed animals were significantly likely to show reduction in the amount of time spent cringing toward humans (exact significance = .016, one-tailed, N ties = 6), but not in cringing behaviors in general or in stress-related behaviors. Neither the husbandry-exposed group nor the group exposed to no training showed a significant number of subjects exhibiting a reduction in duration of any of these behaviors. Additionally, initial temperament assessments were found to significantly predict the relative ability of subjects exposed to training to acquire trained behaviors such that animals generally ranked as more inhibited in terms of temperament also ranked as slower learners based on a Wilcoxon Matched-Pairs Signed-Ranks test, z = -.316, p = .752 (two-tailed). Results of this study could enhance both laboratory animal welfare and laboratory animal research, and could be a first step in developing techniques for reducing fearful behavior in rhesus monkeys in the laboratory environment.

In this thesis, I problematize the notion of democracy¡Vthe ordained rights of human beings to pursuit liberty, happiness and prosperity¡Vby articulating the idea of the American Dream with African Americans¡¦ experiences of self-realization in a so-called democratic society. The purpose is to discuss whether or not African Americans can reverse and utilize their marginalized position as a critical stance for self-articulation to undo the racists¡¦ misnaming on African American people. With a special emphasis on Milkman¡¦s improvisation of the meanings of his family name, Dead, I discuss how the African Americans¡¦ distinctive way of double-talk can facilitate them to negotiate the apparent dualism to inscribe their hybridized identity and how this kind of creativity can help them produce an alternative narrative of their traumatizing as well as truncated history. Also, I intend to analyze both the limitation and liability of conventional psychoanalytic paradigm which is blind to the specificity of African American manhood and the problems peculiar to African American family. Though it is an undeniable historical fact that the African Americans do suffer from the aftermath of plantation slavery, they should be able to empower themselves by re-imagining a collective ancestry as a strategy to formulate an applicable identification politics. While narrating an inspiring genealogy for her people, Toni Morrison wraps up this novel with an open ending. This arrangement suggests to her people that the significations of their cultural identities be opened to further contestation and re-definition.

Effets de l’alcoolisme fœtal sur le développement du corps genouillé latéral du singePapia, Marc 11 1900 (has links)
L’exposition du fœtus à l’éthanol est reconnue comme étant la principale cause de maladies évitables lors du développement. Une forte exposition à l’alcool durant la gestation peut occasionner des dysmorphies cranio-faciales et des retards mentaux, ainsi que des troubles d’apprentissages et du comportement. Le développement du système visuel est également perturbé chez une grande majorité d’enfants qui ont été exposés à l’alcool. Lorsque les doses prises sont élevées, le système visuel peut présenter une panoplie de symptômes comme une augmentation de la tortuosité des vaisseaux rétiniens, de la myopie, de l’hypermétropie, du strabisme et une hypoplasie du nerf optique. Cependant, très peu d’études se sont penchées sur les effets de plus faibles doses sur le développement du système visuel du primate. Le singe est un excellent modèle pour étudier le système visuel car il possède plusieurs similitudes avec l’humain tant au niveau développemental qu’au niveau structurel. De plus, le singe utilisé, le Chlrocebus aethiops sabeus, possède l’avantage que des individus de cette espèce ont une consommation naturelle et volontaire à l’alcool. Une étude (Clarren et al., 1990) a suggéré qu’une faible exposition à l’alcool du fœtus du primate non humain occasionnait une diminution du nombre de cellules ganglionnaires de la rétine (CGRs). Étant donné que le corps genouillé latéral dorsal (CGLd) reçoit la plupart de ses intrants de la rétine, il est raisonnable d’assumer que les couches rétino-récipientes du CGLd devraient être aussi affectées. Nous avons alors émis l’hypothèse que le CGLd devrait également subir une diminution du nombre de neurones. Pour la première fois, nous avons utilisé une méthode stéréologique pour quantifier le nombre de cellules dans les couches parvo- (P) et magnocellulaires (M) du CGLd. Contrairement à notre hypothèse de départ, nous n’avons pas observé de diminution dans le nombre global de neurones dans le CGLd des animaux exposés à l’alcool par rapport à des sujets contrôles, ni une diminution de son volume. Nous avons toutefois observé une diminution de la taille du corps cellulaire seulement dans la population M du CGLd. Ces résultats suggèrent que le système visuel est affecté par une faible exposition à l’alcool durant son développement qui devrait se traduire sur le comportement par des déficits dans les fonctions de la voie M. / An alcohol exposition during the gestation is recognized as the leading causes of preventable developmental disorders. A heavy exposure to ethanol can lead to a litany of symptoms ranging from cranio-facial dysmorphology and mental retardation to developmental learning and behavioural disorders. The visual system development is also the target for a high percentage of children exposed prenatally to alcohol. When the concentrations are relatively high, the visual system can show signs of increased tortuosity of the retina vessels, myopia, hyperopia, strabismus and hypoplasia of the optic nerve. Few studies have examined the effects of a moderate exposure to alcohol on the development of the primate visual system. The monkey is an excellent model to study the visual system because this model owns a lot of similarities with human at the developmental and structural levels. Moreover, the monkey used for this study, Chlrocebus sabeus, naturally and voluntarily consumes alcohol. One study (Clarren et al., 1990) suggested that a low dose exposure of alcohol induce a diminution of neurons in the ganglion retinal cell population. But, no information is given about the numbers and the cell population of neurons. We hypothesized that the lateral geniculate nucleus (LGN) should show a decrease in the number of neurons after moderate prenatal exposure to alcohol. The LGN is a key region because it is situated between the region that receives visual information, the retina, and the first visual region that analyzes the visual information, the primary visual cortex (V1). We utilized a stereological method to study the effects of moderate doses of alcohol on the neuronal population of the LGN in the non-human primate. In opposition to our hypothesis, we have not observed any diminution in the number of neurons or the volume of the LGN. However, we have found a diminution in the neuronal volume of only the magnocellular (M) region. These results suggest that the visual system is affected by a low dose of alcohol that should translate into deficits in the functions of the M pathway.

Thus far, the novel Naked Lunch has not been discussed from the aspect of critical animal studies, nor has it been connected to the theories of Michel Foucault. This essay however, argues that these diverse fields could be connected through the use of the simians that are frequently employed in Naked Lunch. By analyzing the metaphorical role of the simian, the structure of the normalization process can be revealed. Therefore the simian’s metaphorical role becomes to reveal the different stages character goes through in that process and ultimately revealing its negative effects. They also prove to employ the role of abnormality which normalization wants to subtract from the human in order to render her docile. By applying the power mechanisms such as signals, the concept of panopticism and the theory of the docile body to specific passages where simians are highly prominent, the claim of this paper can be demonstrated. Besides Foucault, the theories of Robin Lydenberg are also used consistently throughout the essay due to her valuable observations such as the struggle between body and mind.

Non-human primates have constituted an important group among animals subjected
to various studies, such as ethological and paleontological studies who have
revealed changes in anatomical structures linked to the evolution of primates and the
detailed knowledge on their anatomy may represent an important factor for their
preservation and protection. Aimed to identify the distribution of the femoral,
saphenous, obturator and sciatic nerve of the pelvic limb of Cebus libidinosus (C.l.).
In general, the enervation model of the four primates, men, baboons, chimpanzees
and C.l. is identical, but in specific terms, the differences in enervations indicate
evolution divergence closer to the branch of baboons in the evolutionary tree, and
these data corroborate other comparative studies in relation to the same primates to
vessels, muscles and nerves. In conclusion, the sciatic nerve sends branches to the
biceps femoris muscle, semitendinous, semimebranosus and accessory and to the
ischiofemoral muscle, in the distal third, this nerve is divided into tibial, sural
cutaneous medial and common fibular nerves; the femoral nerve sends branches to
the sartorius muscle, vastus intermedius muscle, vastus medialis muscle, to the
vastus lateralis muscle and branches to the rectus femoris muscle and sends the
saphenous nerve. The obturator nerve provides branches to the pectineus muscles,
adductor brevis muscles, adductor magnus muscles, adductor longus muscle and
glacilis muscle. / Os primatas não-humanos têm constituído um grupo importante dentre os animais
submetidos a diversos estudos, tais como etológicos e paleontológicos que têm
revelado mudanças nas estruturas anatômicas associadas à evolução dos primatas
e o conhecimento detalhado sobre a sua anatomia pode representar um fator
importante para a sua preservação e proteção. Objetivou-se verificar a distribuição

Le suivi des changements dans les populations de cellules immunitaires tels que les lymphocytes, monocytes et cellules dendritiques (DC) au cours de maladies infectieuses comme le virus de l'immunodéficience humaine (VIH) chez l’homme ou son équivalent chez le singe (VIS) est crucial. Grâce aux récentes avancées technologiques en cytométrie en flux, il est maintenant possible de mesurer et d’analyser simultanément jusqu'à 14 paramètres individuels à l’échelon cellulaire. L'objectif de ce travail consiste en la mise au point de 2 panels multicouleurs de 12 anticorps permettant d'analyser simultanément les principales populations de cellules immunitaires, respectivement chez l’humain et le macaque rhésus. Au terme de ce travail, il est maintenant possible de mesurer précisément tous les principaux acteurs du système immunitaire, à savoir les lymphocytes T CD4+ et T CD8+, les lymphocytes B, les cellules NK et NKT, les sous-populations de monocytes, et toutes les sous-populations de cellules dendritiques connues à ce jour, en utilisant une approche multiparamétrique de cytométrie en flux. Ce protocole d’analyse est réalisé sur du sang total, il est rapide, il n’implique pas de technique d’isolation cellulaire, et requiert une quantité minimum de sang. De plus, l’analyse de chaque population cellulaire est plus précise grâce à une contamination minimum entre les populations séparées. L’intérêt de ce travail est d’étudier les interactions entre les différentes populations de cellules immunitaires durant l’infection par VIH chez l’homme, ou VIS chez le singe ou potentiellement d‘autres maladies, et en particulier de mieux comprendre le rôle important que les cellules dendritiques jouent dans la progression de ces maladies. / Monitoring changes in immune cell populations such as lymphocytes, monocytes and dendritic cells (DC) during infectious diseases like human immunodeficiency virus (HIV) or its counterpart in rhesus monkeys (SIV) is crucial. Thanks to recent technological advances in flow cytometry, it is now possible to measure and analyze simultaneously up to 14 individual parameters at the single cell level.The goal of this work is to develop 2 multicolor flow cytometry panels comprising of 12 antibodies, allowing measuring simultaneously the main immune cells population, respectively in humans and rhesus monkeys. After 2 years of development and optimization, we can now measure precisely all the main actors of the immune system, that is CD4+ and CD8+ T lymphocytes, B lymphocytes, NK and NKT cells, the 3 monocyte subsets, and all the dendritic cell subsets known today, by using a multicolor flow cytometry approach. This assay is done on whole blood, it is rapid to do, it does not involve a cell isolation technique, and it requires only a minimum amount of blood. Moreover, the analysis of each population is much more precise because of a minimum contamination between different cell populations. The advantage of this work is to study interactions between different cell populations of immune cells during HIV infection in humans, or SIV infection in monkeys, or potentially other diseases, and in particular to better understand the important role that dendritic cells might play in disease progression.
