Mathematical modeling of oncogenesis control in mature T-cell populations

T-cell receptor (TCR) polyclonal mature T cells are surprisingly resistant to oncogenic transformation after retroviral insertion of T-cell oncogenes. In a mouse model, it has been shown that mature T-cell lymphoma/leukemia (MTCLL) is not induced upon transplantation of mature, TCR polyclonal wild-type (WT) T cells, transduced with gammaretroviral vectors encoding potent T-cell oncogenes, into RAG1-deficient recipients. However, further studies demonstrated that quasi-monoclonal T cells treated with the same protocol readily induced MTCLL in the recipient mice. It has been hypothesized that in the TCR polyclonal situation, outgrowth of preleukemic cells and subsequent conversion to overt malignancy is suppressed through regulation of clonal abundances on a per-clone basis due to interactions between TCRs and self-peptide-MHC-complexes (spMHCs), while these mechanisms fail in the quasi-monoclonal situation. To quantitatively study this hypothesis, we applied a mathematical modeling approach. In particular, we developed a novel ordinary differential equation model of T-cell homeostasis, in which T-cell fate depends on spMHC-TCR-interaction-triggered stimulatory signals from antigen-presenting cells (APCs). Based on our mathematical modeling approach, we identified parameter configurations of our model, which consistently explain the observed phenomena. Our results suggest that the preleukemic cells are less competent than healthy competitor cells in acquiring survival stimuli from APCs, but that proliferation of these preleukemic cells is less dependent on survival stimuli from APCs. These predictions now call for experimental validation.

Les diasporas comme ressources d'intégration dans l'économie mondiale. / Diasporas as resources of integration into the world economy

L'objectif de cette thèse est de livrer des éclaircissements sur la contribution que les diasporas apportent au développement de leurs pays d'origine et par conséquent à une meilleure intégration dans l'économie mondiale. Nous avons fait comme hypothèse principale, que toutes les diasporas peuvent jouer un rôle considérable dans le processus de développement des pays de départ en raison des diverses ressources qu'elles envoient vers les territoires d'origine. Cette problématique est le fil conducteur du travail de recherche que nous avons réalisé pour trouver des réponses aux questions portant sur la définition du terme « diaspora » (Sheffer, 1986 ; Safran, 1991 ; Cohen, 1997 ; Ma Mung, 1992), sur les éléments constitutifs du groupe diasporique (Massey et al.,1993 ; Prévélakis, 2005 ; Bruneau, 2004), sur les mécanismes de production des effets positifs (Saxenian, 1999), sur les ressources générées par la communauté diasporique et leur impact sur le pays de départ (Meyer, 2002 ; Kapur et McHale, 2005 ; Orozco, 2005 ; Kuznetsov, 2006), sur les déterminants de l'ampleur de l'impact produit par la diaspora sur le pays de départ. Les études existantes sur les diasporas sont peu nombreuses dans la littérature économique et les effets qu'elles produisent sur les pays de départ restent peu développés étant limités généralement à l'analyse de l'impact des remises de fonds sur les ménages bénéficiaires. Les rares travaux portant sur les ressources non financières sont le plus souvent des études de cas. Dans le cadre de cette thèse, nous nous sommes proposés d'aller plus loin, en participant à la construction d'une théorie générale et en tenant à mettre en évidence les ressources non financières d'une diaspora. Nous montrons que la diaspora n'est pas uniquement un fournisseur de ressources financières pour le pays d'origine avec des effets positifs à court terme mais elle est aussi le pourvoyeur d'autres ressources qui sont susceptibles de produire des effets bénéfiques plus durables et générer des changements plus profonds. Par le biais d'une grille de lecture des ressources de la diaspora, on fait d'une part, un état des lieux des effets engendrés par les envois de fonds et d'autre part, on analyse les effets produits par la diffusion de cinq types de ressources non financières (cognitives, relationnelles, institutionnelles, organisationnelles et symboliques) générées par la diaspora. L'introduction de la notion d'intentionnalité nous permet de montrer que les effets induits sur le pays de départ par la diaspora à travers l'envoi de différentes ressources sont plus complexes et dépassent largement le cadre des effets recherchés. La problématique des diasporas traitée dans cette thèse, est analysée selon trois axes principaux. Le premier est défini par la nécessité d'une conceptualisation générale de la diaspora dans le contexte de la globalisation, comme ressource de convergence économique entre pays, et comme ressource d'intégration dans l'économie mondiale. Le deuxième axe de notre étude présente l'importance des groupes diasporiques à l'heure de l'économie de la connaissance comme porteurs de ressources non financières qui peuvent être mobilisées en faveur des pays d'origine. Enfin, le dernier axe d'analyse de la problématique renvoie à la nécessité de vérifier, empiriquement, le rôle de la diaspora dans le développement du pays d'origine, notamment à travers les effets des ressources non financières (RNF). Cet objectif nous a conduit à choisir trois diasporas émergentes post-communistes : la diaspora roumaine, la diaspora moldave et la diaspora kirghize comme terrain propice d'étude de ces effets. Notre choix s'explique par le fait que ces diasporas sont essentiellement composées de personnes qualifiées ayant un haut niveau d'éducation. Malgré leur existence récente, ces groupes nationaux se caractérisent par un activisme diasporique très favorable aux effets des RNF qui nous a permis de répondre aux questions posées au cours de cette thèse. Les questions soulevées durant ce travail couvrent trois domaines : l'existence et la nature des diasporas, la diaspora et le développement économique du pays d'origine et les déterminants de l'impact des ressources non financières des diasporas sur les pays de départ. Les réponses à ces questions nous ont permis de montrer le rôle des diasporas dans le développement de leurs pays d'origine à travers les ressources qu'elles procurent aux territoires de départ, notamment les ressources non financières qui ont un effet accélérateur plus important en matière de rattrapage économique que les transferts de fonds. Cette thèse aide à comprendre le fonctionnement des diasporas comme vecteurs de développement, d'une part, à travers les caractéristiques générales qui permettent la distinction d'autres groupes de migrants et d'autre part, à travers les spécificités de chaque diaspora qui expliquent l'émergence des phénomènes particuliers comme dans le cas de la diaspora chinoise ou les diasporas post-soviétiques. / The objective of this thesis is to underline the contribution of Diasporas to the development of origin countries and to their integration into the world economy. The central hypothesis points out that all Diasporas can play a valuable role in the development process of source countries through the wide range of resources they send to homelands. The thesis investigates: the clarification of the concept of Diaspora (Sheffer, 1986 ; Safran, 1991 ; Cohen, 1997 ; Ma Mung, 1992), the infrastructure of diasporic communities (Massey et al.,1993 ; Prévélakis, 2005 ; Bruneau, 2004), the mechanisms of positive effects induced by Diaspora on origin society (Saxenian, 1999), the types of resources created by expatriates and their impact on the country of origin (Meyer, 2002 ; Kapur and McHale, 2005 ; Orozco, 2005 ; Kuznetsov, 2006), as well as the determinants of the impacts of Diasporas on their home countries. In the current economic literature, the studies on Diasporas are rare and the analysis of their effects on countries of origin is limited to the role of financial resources (remittances). This work goes much beyond by participating in the elaboration of a general theory which focuses on the importance of non financial resources generated by a Diaspora. The expatriates settled abroad are not merely purveyors of financial resources for countries of origin producing only positive effects in a short term perspective but they generate other resources which are likely to produce more profound and durable positive effects. In this thesis we identify five non financial resources created by a Diaspora: cognitive, relational, institutional, organizational and symbolic resources. The use of the concept of intentionality helps emphasizing the complex effects of resources transfers from migrants showing their intentional and non-intentional sides. The research problem of this dissertation is organized around three main issues. The first one is related to the need of a Diaspora general conceptualization in the era of globalization as resource of economic convergence between sending and receiving countries, and as resource of integration into the world economy. The second direction of our argumentation is guided by the Diasporas' place in the context of knowledge economy as generator of non financial resources which can be mobilized in favour of countries of origin. Finally, the last element of the research problem is the empirical verification of the Diaspora's role in the development of the homeland, namely through the effects of non financial resources (NFR). This objective determined the selection of three emerging post-Communist Diasporas: Romanian, Moldovan and Kyrgyz Diaspora as a very interesting ground of investigation of these effects. Our choice is explained by the composition of these diasporic groups characterized by a strong presence of skilled migrants with a high level of education. Despite their recent creation, these Diasporas defined by an important “diasporic activism” which spurs the production of NFR positive effects helped answering the questions that arose during this work. These questions covered three domains: Diasporas existence and their nature, Diaspora and economic development of country of origin and the determinants of the non financial resources impact on the source country.

Nesse contexto, as mudanças recentes na indústria petrolífera brasileira, decorrentes da promulgação da Lei do Petróleo de 1997, fizeram com que fosse necessário reavaliar a situação desses campos e seu potencial de produzir impactos econômicos positivos nos municípios em que estão localizados. O objetivo principal da análise quantitativa foi o de verificar se i) a produção de petróleo e gás natural, e ii) o recebimento de royalties têm algum impacto sobre o crescimento econômico dos grupos de municípios baianos em que isso ocorre. Para isso, foi realizada uma análise de dados utilizando a metodologia de Painel Dinâmico Generalized Method of Moments (GMM), utilizando tanto o estimador Arellano-Bond (GMM-DIF) quanto o Arellano-Bover (GMM-SYS), e comparando seus resultados com outras metodologias de Painel Estático. Foi analisado o desempenho de municípios baianos produtores e não produtores de petróleo e gás, e de recebedores e não recebedores de royalties, bem como se há um efeito multiplicativo dos valores recebidos por esses municípios no período de 2005 a 2010, para os impactos dos royalties, e de 2007 a 2010, para os impactos da produção. Os resultados encontrados apontam que, em conformidade com a literatura sobre a “maldição dos recursos naturais”, há uma relação inversa entre arrecadação com royalties pelos municípios que recebem essa renda petrolífera e seu nível de renda, ainda que essa relação seja pouco relevante em termos econômicos. No entanto, as estimações realizadas apenas para os municípios produtores mostram que essa relação se inverte, tornando-se positiva, mas também de baixa magnitude. O mesmo ocorre quando se adiciona um termo interativo entre produção e royalties recebidos, que também apresenta um impacto positivo mas cuja magnitude é pouco relevante. Dessa forma, é plausível afirmar que, ainda que haja um impacto positivo no caso dos municípios produtores, há indícios de que esse impacto pode ser ainda maior, caso sejam adotados incentivos para o aumento da atividade petrolífera na região e políticas públicas que busquem atrelar as rendas petrolíferas recebidas por esses municípios a despesas como, por exemplo, saúde e/ou educação, além de exigir desses a adoção e divulgação de indicadores de governança mais robustos. / This study aims to evaluate the economic impacts of the oil and gas industry – production and royalties received – in municipalities in the state of Bahia between the years of 2005 and 2010. In this context, recent changes in the Brazilian oil and gas industry, resulting from the passing of the Petroleum Law of 1997, made it necessary to reevaluate the situation of these wells and their potential to generate positive economic impacts for the municipalities where they are located. The main objective of the quantitative analysis was to verify whether i) oil and gas production, and ii) receiving royalties had any impact on economic growth of the groups of municipalities where these occur. In order to accomplish that, an analysis of the available data was carried out with the methodology Generalized Method of Moments (GMM), using both the Arellano-Bond (GMM-DIF) and Arellano-Bover (GMM-SYS) estimators and comparing their results to those of other methodologies, namely Static Panel ones. The economic performance of producing and non-producing municipalities in Bahia, and of those that received and did not receive royalties was analyzed. Moreover, it was also evaluated whether or not there is a multiplicative effect of the royalty figures received by these municipalities during the period analyzed. Results show that, in accordance with the literature on the Curse of Natural Resources, there is an inverse relation between the collection of royalties and income levels for municipalities in Bahia, but this impact is of little economic relevance. However, estimations carried out solely for producing municipalities point to an inverse relationship in that sense, being positive for this specific group, but also of little economic relevance. A similar impact is observed when an interactive term including a dummy for production and the amount of royalties received is added to the model, also resulting in a positive but low economic impact. Thus, it is plausible to claim that even if there is a positive impact in producing municipalities steming from this activity, there is evidence that this impact could be even greater if incentives for the increase of production are provided for this region. In addition to that, public policies searching to bond oil incomes received by these municipalities to expenses in areas such as education and health, as well as more robust governance indicators, are also desirable in that sense.

This work aims to develop methodologies based on comparative statistical processes of hydrogeochemical analysis of production zones in order to minimize types of high-cost interventions to perform identification test fluids - TIF. For the study, 27 samples were collected from five different production zones were measured a total of 50 chemical species. After the chemical analysis was applied the statistical data, using the R Statistical Software, version 2.11.1. Statistical analysis was performed in three steps. In the first stage, the objective was to investigate the behavior of chemical species under study in each area of production through the descriptive graphical analysis. The second step was to identify a function that classify production zones from each sample, using discriminant analysis. In the training stage, the rate of correct classification function of discriminant analysis was 85.19%. The next stage of processing of the data used for Principal Component Analysis, by reducing the number of variables obtained from the linear combination of chemical species, try to improve the discriminant function obtained in the second stage and increase the discrimination power of the data, but the result was not satisfactory. In Profile Analysis curves were obtained for each production area, based on the characteristics of the chemical species present in each zone. With this study it was possible to develop a method using hydrochemistry and statistical analysis that can be used to distinguish the water produced in mature fields of oil, so that it is possible to identify the zone of production that is contributing to the excessive elevation of the water volume. / A ?gua produzida caracteriza-se como um dos res?duos mais comuns gerados durante as fases de explora??o e produ??o de petr?leo. Este trabalho tem como objetivo desenvolver metodologias baseadas em processos estat?stico-comparativos de an?lises hidrogeoqu?micas de zonas de produ??o com o objetivo de minimizar tipos de interven??es de altos custos para realiza??o de teste de identifica??o de fluidos TIF. Para o estudo, foram coletadas 27 amostras de cinco zonas de produ??o diferentes e foram medidos um total de 50 esp?cies qu?micas. Ap?s as an?lises qu?micas foi aplicado o tratamento estat?stico aos dados, utilizando o Software Estat?stico R, vers?o 2.11.1. O tratamento estat?stico foi realizado em tr?s etapas. Na primeira etapa, o objetivo foi verificar o comportamento das esp?cies qu?micas em estudo em cada zona de produ??o atrav?s da an?lise gr?fica descritiva. A segunda etapa consistiu em identificar uma fun??o que classificasse as zonas de produ??o de cada amostra, utilizando An?lise Discriminante. Na etapa de treinamento, a taxa de acerto de classifica??o da fun??o da an?lise discriminante foi de 85,19%. This deep change in the legal regime of oil sector brought the possibility of entrance to small and medium size producers in this industry, especially through the production activities developed in mature and marginal fields of oil, which are located mainly in northeast region of Brazil. Considering that the intervention of state over the economy finds its guidelines and limits in Federal Constitution disposals, the present work investigated in which way states regulation, mainly through taxation rules, has obeyed the constitutional regime in force, and specially, the reduction of regional inequalities principle. By mean, firstly, of an analysis of central concepts (mature fields, marginal fields, small and medium size producers) we observed that the imprecision over the conceptual aspect has constituted an obstacle to a specific states‟ regulation, directed to this newborn class of producers, whose growth has been pursuit by the state. That is verified in the case of concession procedures, and also, concerning the taxation system applied to small and medium size producers. Examining the main constitutional principles related to this universe which are the legality, equality, privileged treatment to small enterprises, contributive capacity, and reduction of regional inequalities we conclude that it is legally possible, a truly specific regulation, including a special taxation regime, to the small and medium size producers whose activities are concentrate over mature-marginal fields, aiming the concretization of the Brazilian state main goals / No contexto de desestatiza??o que permeou a economia brasileira na d?cada de 1990, surgiu a Emenda Constitucional 9/95, que deu contornos mais flex?veis ao monop?lio da Uni?o sobre o petr?leo. Tal modifica??o no regime jur?dico do setor petrol?fero trouxe a possibilidade de inser??o de pequenos e m?dios produtores, sobretudo no ?mbito da produ??o em campos maduros e marginais de petr?leo, os quais se situam majoritariamente na regi?o Nordeste. Considerando que a interven??o do Estado sobre o dom?nio econ?mico encontra diretrizes e limites postos pela Constitui??o Federal, o presente trabalho investiga de que forma a atividade regulat?ria do Estado, principalmente por interm?dio de normas tribut?rias, tem se conformado ao regime constitucional vigente e, em especial, ao princ?pio da redu??o das desigualdades regionais. This study focuses on three theoretical areas: project management, internationalization of companies and the resource-based view (RBV). Thus defined the following research question: "How the project management maturity contributes to internationalization of Brazilian companies from the perspective of resource-based view?” Particularly analyzed the contributions of mature project management for internationalization features in international Brazilian companies from the RBV-VRIO analysis, ie, examined whether the mature nature of the management of projects adopted by the selected companies are considered strategic resources, with implications competitive, sustainable competitive advantage for the company. The research strategy adopted was a multiple case study of descriptive exploratory approach within-case analysis and cross-case method, in internationalized Brazilian companies from different business segments. As a result, the study found that the project management features: sustainability of the project, involvement of stakeholders, defining objectives and strategies, change management and risk management, have the greatest contribution to the internationalization of selected companies, with implication of competitive advantage sustainable, considering the RBV-VRIO analysis. Thus, the research presents contributions for companies that are or intend to initiate a process of internationalization, so the process is clearer and driven to a better use of market opportunities. / As empresas enfrentam desafios de diversas naturezas em diferentes ambientes competitivos, dependendo do mercado-alvo, do modelo do negócio e das estratégias corporativas. Atuar em mercados internacionais é um desafio constante na pauta das empresas no ambiente globalizado ao iniciar seu processo de internacionalização. São desafios desde a capitalização de recursos, conquista de novos mercados e clientes, desenvolvimento de competências e capacidades, instruídas para a formação estratégica de internacionalização da empresa, entre outros. Este estudo concentra-se em três eixos teóricos: a gestão de projetos, a internacionalização de empresas e a visão baseada em recursos (RBV). Assim, definiu-se a seguinte questão de pesquisa: “Como a maturidade em gestão de projetos contribui para internacionalização de empresas brasileiras sob a ótica da visão baseada em recursos?”. Particularmente foram analisadas as contribuições das características da gestão madura de projetos para internacionalização em empresas brasileiras internacionais a partir da análise RBV-VRIO, ou seja, analisou se as características maduras da gestão de projetos adotados pelas empresas selecionadas são consideradas como recursos estratégicos, com implicação competitiva, de uma vantagem competitiva sustentável para empresa. A estratégia de pesquisa adotada foi de estudo de múltiplos casos, de abordagem exploratória descritiva com método de análise inter-caso e intra-caso, em empresas brasileiras internacionalizadas de diferentes segmentos empresariais. Como resultado, o estudo identificou que as características da gestão de projetos: sustentabilidade do projeto, envolvimento dos stakeholders, definição de objetivos e estratégias, gerenciamento da mudança e gerenciamento de riscos, possuem alta contribuição para internacionalização das empresas selecionadas, com implicação de vantagem competitiva sustentável, considerando a análise RBV-VRIO. Com isso, a pesquisa apresenta contribuições para as empresas que estão ou pretendem iniciar um processo de internacionalização, de forma que o processa seja mais claro e orientado para um melhor aproveitamento das oportunidades de mercado.

Объектом исследования явилась личностная тревожность. Предметом исследования стала связь личностной тревожности с мотивацией достижения, прокрастинацией и копинг-стратегиями у мужчин и женщин зрелого возраста. Магистерская диссертация состоит из введения, двух глав, заключения, списка литературы (121 источник) и приложения, включающего в себя бланки применявшихся методик. Объем магистерской диссертации 120 страниц, на которых размещены 7 рисунков и 12 таблиц. Во введении раскрывается актуальность проблемы исследования, разработанность проблематики, ставятся цель и задачи исследования, определяются объект и предмет исследования, формулируются гипотезы, указываются методы и эмпирическая база, а также научная новизна исследования. Первая глава включает в себя обзор иностранной и отечественной литературы по теме тревожности, мотивации достижения, прокрастинации и копинг-стратегий. Представлены разделы, посвященные исследованию связи личностной тревожности с мотивацией достижения, прокрастинацией и копинг-стратегиями у мужчин и женщин зрелого возраста. Выводы по первой главе представляют собой итоги по изучению теоретического материала. Вторая глава посвящена эмпирической части исследования. В ней представлено описание организации и методов проведенного исследования и результатов, полученных по всем использованным методикам: тесту Шкалы личностной тревожности Спилбергера-Ханина (State-Trate Anxiety Inventory, STAI), опроснику «Способы совладающего поведения» Р. Лазаруса и С. Фолкман, опроснику А. А. Реана «Мотивация успеха и боязнь неудачи», тесту Шкалы общей прокрастинации С. H. Lay. Также в главе представлены сравнительный и корреляционный анализ результатов исследования. Выводы по главе 2 включают в себя основные результаты эмпирического исследования. В заключении в обобщенном виде изложены результаты теоретической и эмпирической частей работы, а также выводы по выдвинутым гипотезам, обоснована практическая значимость исследования и описаны возможные перспективы дальнейшей разработки данной проблематики. / The object of the study was personal anxiety. The subject of the study was the relationship between personal anxiety and achievement motivation, procrastination and coping strategies in men and women of mature age. The master's thesis consists of an introduction, two chapters, a conclusion, a list of references (121 sources) and an appendix that includes the forms of the applied methods. The volume of the master's thesis is 120 pages, which contain 7 figures and 12 tables. The introduction reveals the relevance of the research problem, the elaboration of the problematics, the goal and objectives of the research are set, the object and subject of the research are determined, hypotheses are formulated, the methods and empirical base, as well as the scientific novelty of the research, are indicated. The first chapter includes a review of foreign and domestic literature on the topic of anxiety, achievement motivation, procrastination, and coping strategies. The sections devoted to the study of the relationship between personal anxiety and achievement motivation, procrastination and coping strategies in men and women of mature age are presented. Conclusions for the first chapter represent the results of the study of theoretical material. The second chapter is devoted to the empirical part of the study. It presents a description of the organization and methods of the study and the results obtained by all the methods used: the Spielberger-Hanin Personal Anxiety Scale test (State-Trate Anxiety Inventory, STAI), the Coping Behavior Methods questionnaire R. Lazarus and S. Folkman, A. A. Rean's questionnaire "Motivation for success and fear of failure", the test of the General Procrastination Scale S. H. Lay. The chapter also presents a comparative and correlation analysis of the research results. Conclusions from Chapter 2 include the main findings of the empirical study. In the conclusion, in a generalized form, the results of the theoretical and empirical parts of the work are presented, as well as conclusions on the hypotheses put forward, the practical significance of the study is substantiated and possible prospects for the further development of this problem are described.

5 Abstract Tobacco mature pollen rehydrates in vivo on a stigma tissue, and develops into the rapidly-growing pollen tube. This rehydration process is accompanied by the de-repression of stored mRNA transcripts, resulting in the synthesis of novel proteins. Furthermore, such metabolic switch is also likely to be regulated on the level of post-translational modifications of the already-present proteins, namely via phosphorylation, since it was shown to play a significant regulatory role in numerous cellular processes. Since only a minor part of proteins is phosphorylated in a cell at a time, the employment of various enrichment techniques is usually of key importance. In this diploma project, metal oxide/hydroxide affinity chromatography (MOAC) with aluminium hydroxide matrix was applied in order to enrich phosphoproteins from the mature pollen and the 30-minute in vitro activated pollen crude protein extracts. The enriched fraction was separated by both 2D-GE and gel-free liquid chromatography (LC) approaches with subsequent mass spectrometric analyses. Collectively, 139 phosphoprotein candidates were identified.
