Estudo da sensibilização de cães com dermatite atópica na região central do Rio Grande do Sul / Sensitization study of the dogs with atopic dermatitis in central region of Rio Grande do Sul

Canine atopic dermatitis (CAD) is a common dermatosis, defined as a genetic-based disease, which predisposes to cutaneous inflammation and pruritus, mediated by class IgE immunoglobulins directed against specific antigens in most cases. Clinical diagnosis may be later complemented by skin allergic and/or serological tests (ELISA). The aim of these tests is to identify possible allergens in order to enable the clinicians to select candidate antigens for allergen specific immunotherapy. This work aimed to identify the sensitization profile of 58 dogs with atopic dermatitis diagnosis. All animals were submitted to intradermic test (IDT) and screened for the presence of antibodies against different allergens using a serologic test. House dust mites are described as the most frequent allergens in all continents. However, the positivity to C. dactylon is not commonly described and may be characteristic for the region. With this work it was possible to identify the main allergens involved in the immunologic response of atopic dogs residing in Rio Grande do Sul, pointing to the importance to include C. dactylon in screening tests for allergy. / A dermatite atópica canina (DAC) é uma dermatose comum, definida como uma doença de cunho genético que predispõe à inflamação e ao prurido cutâneo, mediada por imunoglobulinas da classe IgE dirigidas contra antígenos específicos na maior parte dos casos. O diagnóstico da DAC é clínico e pode ser posteriormente complementado por testes alérgicos cutâneos e/ou sorológicos. O objetivo desses testes é identificar possíveis alérgenos e, com isso, possibilitar ao clínico a seleção de antígenos candidatos para a imunoterapia alérgeno-específica. No presente trabalho buscou-se identificar o perfil de sensibilização de 58 cães diagnosticados com dermatite atópica. Todos os animais foram submetidos ao teste intradérmico (TID) e à detecção de anticorpos específicos para diferentes alérgenos através de teste sorológico (ELISA). Os ácaros domiciliares são descritos como os alérgenos mais frequentes em todos os continentes. Entretanto, a positividade ao C. dactylon não é usualmente descrita e pode ser característica da região. Com esse trabalho foi possível identificar os principais alérgenos envolvidos na resposta imunológica de cães atópicos residentes no Rio Grande do Sul, ressaltando-se a importância da inclusão do extrato de C. dactylon em testes alérgicos.

The cohort Astigmatina is divided in two major groups: Psoroptidia, composed mainly by feather and fur mites, and Non-psoroptidia, a dominant component of the acarofauna in ephemeral habitats. In these environments Astigmatina usually are saprophages or feed on fungi or bacteria. Astigmatina protonymphs undergo a complete reorganization of the body structure leading to the production of heteromorphic deutonymphs, generally specialized for dispersion through phoresy using arthropods and vertebrates as phoronts. Although most Astigmatina occur in natural environments, some species live in anthropic environments, such as food deposits, where some of them became pests; some Astigmatina infest subterraneous plant organs. Despite their economic and ecological importance, studies on the diversity and taxonomy of Astigmatina in Brazil have been rare over the last decades. The general objective of this thesis was to collaborate to the knowledge of the diversity and to evaluate the potential practical uses of these mites in Brazil. For this, new genera and species were described, method for rearing dust mites was studied and the efficiency of Astigmatina as prey for edaphic predators was evaluated. A new species of Thyreophagus (Astigmatina: Acaridae) was described based on specimens collected in Brazil, the association of three other species of this genus with stored food was reviewed and a key to all species of this genus was prepared. The genus Neotropacarus (Astigmatina: Acaridae), commonly found on plant leaves, was reviewed with the redescription of two species and description of new species collected in Brazil and from the Philippines. Two new genera and seven new species of Acaridae associated with the bee family Apidae was described and a key to Acaridae genera in subfamily Horstiinae was prepared. Several species of Astigmatina were evaluated as prey for predatory mites Stratiolaelaps scimitus (Womersley) (Mesostigmata: Laelapidae) and Protogamasellopsis zaheri Abo-Shnaf, Castilho and Moraes (Mesostigmata: Rhodacaridae), which oviposited on all evaluated astigmatids, with Tyrophagus putrescentiae (Schrank) and Aleuroglyphus ovatus (Tropeau) (Acaridae) being the most suitable prey. Seven foods and two development period, 30 and 60 days, after the introduction of 400 females of two important dust mite species, Blomia tropicalis van Bronswijk, de Cock e Oshima and Dermatophagoides pteronyssinus (Trouessart) were evaluate. With the most suitable foods, the population growth were higher than 20.2 and 15.3 for B. tropicalis and D. pteronyssinus, respectively. / A coorte Astigmatina é dividida em dois grandes grupos: Psoroptidia, composto majoritariamente por ácaros de pena e pelos, e Não-Psoroptidia, componente dominante da acarofauna de habitats efêmeros. Protoninfas de Astigmatina passam por uma completa reorganização da estrutura corporal levando à produção de deutoninfas heteromórficas geralmente especializadas para dispersão por forese utilizando artrópodes e vertebrados como forontes. Apesar de a maioria dos Astigmatina ocorrer em ambientes naturais, algumas espécies passaram a ocupar ambientes antrópicos, como depósitos de alimentos, onde algumas se tornaram pragas; alguns Astigmatina infestam órgãos subterrâneos de plantas. A despeito de sua importância econômica e ecológica, estudos sobre a diversidade e taxonomia dos Astigmatina no Brasil têm sido raros nas últimas décadas. O objetivo geral desta tese foi colaborar para o conhecimento da diversidade e avaliar o potencial de uso prático de espécies Astigmatina no Brasil. Para isso, novos gêneros e espécies foram descritos, métodos para criação de ácaros de poeira foram estudados e a eficiência de Astigmatina como presas para ácaros predadores edáficos foi avaliada. Uma nova espécie de Thyreophagus (Astigmatina: Acaridae) foi descrita com base em espécimes coletados no Brasil, uma revisão sobre três outras espécies deste gênero associadas com alimentos armazenados foi realizada e uma chave para todas as espécies deste gênero foi elaborada. O gênero Neotropacarus (Astigmatina: Acaridae), comumente associado a folhas de plantas, foi revisado, com redescrição de duas espécies e descrição de novas espécies coletadas no Brasil e nas Filipinas. Dois novos gêneros e sete novas espécies de Acaridae associados à família de abelha Apidae foram descritos e uma chave para os gêneros da subfamília Horstiinae foi elaborada. Diversas espécies de Astigmatina foram avaliadas como presas para os ácaros predadores Stratiolaelaps scimitus (Womersley) (Mesostigmata: Laelapidae) e Protogamasellopsis zaheri Abo-Shnaf, Castilho e Moraes (Mesostigmata: Rhodacaridae), que ovipositaram em todas os Astigmatina avaliados, sendo Tyrophagus putrescentiae (Schrank) e Aleuroglyphus ovatus (Tropeau) (Acaridae) as presas mais promissoras. Sete alimentos e dois períodos de desenvolvimento, 30 e 60 dias, após inoculação de 400 fêmeas de duas espécies importantes na poeira residencial, Blomia tropicalis van Bronswijk, de Cock e Oshima e Dermatophagoides pteronyssinus (Trouessart) foram avaliados. Com os alimentos mais adequados, o crescimento populacional nas colônias foram maiores que 20.2 e 15.3 para B. tropicalis e D. pteronyssinus, respectivamente.

A systematic appraisal of the Eriophyoidea (Acari Prostigmata)

The diversity of the Eriophyoidea is largely unknown and their systematic study mostly entails alpha-taxonomy which is critically important for these mites. Eriophyoid morphology is almost exclusively studied on slide-mounted specimens, and truly permanent specimen slides cannot be prepared and are eventually lost. Shortcomings in taxon descriptions are persistent, and too few morphological characters are available for systematic use, particularly for phylogenetic studies. The fragile, simplified and minute eriophyoid bodies, and the inadequacy of study methods and technology, including preparation and light microscopy, contribute to these problems. The present eriophyoid classification is widely accepted, relatively stable and useful. The major part of the classification, however, is probably artificial, and some taxon delimitations and identifications are becoming increasingly difficult. Scanning electron microscopy (SEM) is only sporadically used to supplement conventional descriptions of eriophyoid mites, and their phylogeny has hardly been studied. In the present study some aspects of eriophyoid systematics and its improvements by incorporating SEM for morphological study and phylogenetic analyses for testing and improving the naturalness of the present eriophyoid classification, are used and appraised. The morphology of about 64 species, mostly from South Africa, was studied with low-temperature (cryo) SEM. The specimens remained turgid and the shape of the mites largely unaltered. A general overview of the contribution of the SEM study towards systematic morphology of the Eriophyoidea is presented. Discrepancies between species descriptions from slide-mounted specimens and the SEM images were found. These include body form, interpretation of structures, resolution and information on minute morphology, and the presence of secretions. Some of these differences were caused by artefacts introduced with slide-mounting of specimens. The SEM study includes a comparative morphological study of the gnathosoma, including a review and appraisal of characters presently used in eriophyoid systematics. New morphological information was found, including new characters that may be of systematic use. Morphology studied with SEM should be routinely incorporated into eriophyoid descriptions, which is not presently the case. The phylogeny of the Eriophyoidea was studied at genus level, using morphological data, to test the monophyly of the present suprageneric taxa. Three data matrices with 66, 60 and 27 informative characters of 316 (including most Diptilomiopus spp.), 64 and 17 eriophyoid ingroup species respectively were analyzed with parsimony analyses, and trees were searched under different parameters. This was done to find different hypotheses regarding the taxon relationships, to roughly assess the robustness of the tree groups, and to use different approaches: a very comprehensive taxon sample, but with low ratio of characters to taxa; an exemplar species sample to improve the ratio between characters to taxa; and a very small taxon sample with a good ratio between characters and taxa, but very little inclusion of variation found in the Eriophyoidea. Most groups found were supported only by homoplasy, but many made biological sense and various potentially monophyletic groups, additional to taxa in the present classification, are proposed for further study. The robustness and convergence of these groups on monophyly are discussed. The Phytoptidae was found to be polyphyletic. Part of the Nalepellinae is probably positioned outside the remainder of the Eriophyoidea, while the rest of the Phytoptidae were positioned in smaller subgroups among the Eriophyidae. The Phytoptinae and Sierraphytoptinae, including Pentasetacus, may group together. The Eriophyidae never grouped together with much support, and the family is both polyphyletic and paraphyletic. The Diptilomiopidae was largely found to be monophyletic, with a relatively strong phylogenetic structure. The Rhyncaphytoptinae is mainly paraphyletic, and the Diptilomiopinae polyphyletic, but part of the Diptilomiopinae may be monophyletic. Three new Diptilomiopus spp. from South Africa are described as part of the study: D. faurius sp. nov. from Faurea rochetiana (A. Rich.) Pic. Serm. (Proteaceae); D. apobrevus sp. nov. and D. apolongus sp. nov. from Apodytes dimidiata E. Mey. ex Arn. (Icacinaceae). Por esta razão, a busca por formulações com ação acaricida tem sido preocupação constante das indústrias farmacêuticas veterinárias. No entanto, a grande maioria dos produtos atualmente comercializados apresenta alta toxicidade aos hospedeiros dos ectoparasitas e ao meio ambiente, induzindo, inclusive, resistência nos carrapatos. Métodos alternativos de controle, como o uso de extratos de plantas, por exemplo, têm se tornado cada vez mais interessantes, já que apresentam menores custos e baixa toxicidade. Neste sentido, destaca-se a espécie Azadirachta indica (neem), cujos extratos de folhas e sementes apresentam reconhecido potencial repelente, inseticida e acaricida. Desta forma, este estudo teve como objetivo avaliar as alterações morfológicas observadas no ovário e nas glândulas salivares de fêmeas semiingurgitadas de carrapatos R. sanguineus, submetidas à aplicação tópica do óleo extraído das sementes de neem, nas concentrações de 20, 40 e 60%, utilizando-se etanol aquoso a 10% como solvente. Os órgãos foram coletados, fixados, e analisados por meio de técnicas de microscopia de luz convencional, microscopia confocal de varredura a laser e microscopia eletrônica de transmissão. De forma geral, a metodologia proposta neste trabalho permitiu a simulação de um tratamento tópico dos animais infestados, tornando os resultados obtidos mais próximos de uma aplicação prática em campo. Os ovócitos e os ácinos glandulares de carrapatos R. sanguineus exibiram alterações morfológicas semelhantes, tais como sinais evidentes de desorganização citoplasmática,... / Abstract: Rhipicephalus sanguineus ticks have attracted, in recent years, the attention of researchers as well as public health managers, since they have become major urban pests, transmitters of diseases to humans and dogs. For this reason, the search for formulations with acaricide action has been constant concern of veterinary pharmaceutical industries. However, the vast majority of products currently commercialized exhibits high toxicity to the hosts of these ectoparasites and to the environment, even inducing resistance in ticks. Alternative control methods such as the use of plant extracts, for example, have become increasingly interesting, since they have reduced costs and low toxicity. In this sense, Azadirachta indica (neem) species stands out, as its leaf and seed extracts have recognized repellent, insecticide and acaricide potential. Thus, this study aimed to evaluate the morphological alterations resulting from the action of neem in the ovary and salivary glands of semi-engorged R. sanguineus female ticks, subjected to the topical application of the oil extracted from neem seeds, at concentrations of 20, 40 and 60%, using 10% aqueous ethanol as solvent. The organs were collected and fixed, and analyzed by means of techniques in conventional light microscopy, confocal laser scanning microscopy and transmission electron microscopy. In general, the methodology proposed in this work allowed the simulation of a topical treatment of the infested animals, making the results closer to a practical application in field. In both analyzed organs, neem oil clearly demonstrated a dose-dependent effect. Oocytes and glandular acini of R. sanguineus ticks exhibited similar morphological changes, such as evident signs of cytoplasmic disorganization, cellular vacuolation, nuclear or nucleolar irregularities, dilation of mitochondrial cristae, as well as dilation in rough endoplasmic reticulum lumen. Characterisation of mites and peniciccium species associated with apple core rot diseases

Thesis (MSc (Plant Pathology))--University of Stellenbosch, 2009. / ENGLISH ABSTRACT: Dry core rot (DCR) and wet core rot (WCR) are among some of the most important postharvest diseases of apples in South Africa. Mouldy core (MC) is also a symptom associated with the core region of apples, but it is not of economical importance since apple tissue surrounding the core region is not affected as is the case with DCR and WCR. The incidence of core rots in harvested fruits can be as high as 12%, but in general ranges from 3 to 8%. Infections and losses can also occur during fruit handling in pack houses and during storage. Additionally, yield losses also occur prior to harvest within orchards due to premature fruit drop of core rot affected fruits. The incidence of core rot diseases in apples differ among apple cultivars, with most Red Delicious varieties being susceptible to the development of core rots, whereas core rots have rarely been reported in other cultivars such as Granny Smith. The etiology and epidemiology of WCR and DCR are poorly understood. Although many fungal genera have been associated with the diseases, small-spored Alternaria species are mainly associated with DCR, whereas Penicillium species including P. roquefortii, P. expansum and P. funiculosum have mainly been associated with WCR. Dry core rot infections have long been known to occur pre-harvest, whereas WCR is primarily known as a post-harvest disease where infections take place during fruit handling in pack houses. Recently, Tarsonemus mites have also been indicated as being a potential role player in the etiology of core rot diseases. The mites have been hypothesised to carry pathogen spores into the core region of apples, and they may also possibly cause small wounds that facilitate pathogen entry. In South Africa, apple growers have recently reported WCR as being present prior to harvest, which has not been reported previously. Therefore, the first aim of the study was to investigate the incidence, as well as the causal agent/s of pre-harvest WCR. The incidence of WCR ranged from 0% to 1.7% in eleven orchards, and was in general lower than that of DCR (0.4% to 6%). Isolation studies from eight internal positions in WCR apples showed that Penicillium was the predominant fungal genus in most of the positions, including the lesion area. Morphological and molecular characterisation of Penicillium isolates from WCR showed that P. 2 ramulosum prov. nom. was the main species isolated from lesions, as well as other isolation positions. However, this species was also the main species isolated from DCR, MC and asymptomatic apples. Penicillium expansum was only isolated at low frequencies from WCR and DCR apples. Other Pencillium species that were occasionally isolated included P. glabrum, P. chloroloma, P. chermisinum and a putative new species with closest affinity to P. dendriticum (P. species aff. dendriticum) on a DNA nucleotide sequence basis. Pathogenicity and virulence studies using three different inoculation methods showed that P. expansum was the most virulent species, followed by P. species aff. dendriticum. The P. ramulosum prov. nom. isolates varied in their virulence, but were all considered to have low virulence. The role of Tarsonemus mites in the etiology and epidemiology of core rot diseases is poorly understood, and therefore the second aim of the study was to investigate some of these aspects. The specific aims of the study were to (1) investigate the ecology of Tarsonemus mites in Red Delicious and Granny Smith orchards during different apple developmental stages, (2) determine if there is a significant association of Tarsonemus mites with diseased (WCR and DCR) fruits and (3) determine if potential core rot pathogenic fungi are associated with the mites. Tarsonemus mites were found in all of the investigated apple developmental stages (buds, blossoms, 4cm diameter fruit, mature fruit and mummies), having the highest incidence in mummies and mature fruits from Red Delicious and Granny Smith orchards. In Red Delicious fruits the Tarsonemus mites were found within the core and/or calyx tube, whereas in Granny Smith fruits the mites were restricted to the calyx tube. In Red Delicious fruits there was a significant association between dry core rot as well as total core rot (wet- and dry-core rot) with the presence of mites in the core, as well as total mites (mites in core and calyx tubes). Fungal isolation studies from the Tarsonemus mites showed that they carried potential core rot fungal pathogens within the genera Penicillium and Alternaria. The Penicillium species isolated from the mites included two of the most virulent WCR species, P. expansum and P. species aff. dendriticum. / AFRIKAANSE OPSOMMING: Droë kernvrot and nat kernvrot is van die belangrikste na-oes siektes van appels in Suid- Afrika. Die voorkoms van nat kernvrot was tussen 0 en 1.7% in elf boorde en was oor die algemeen laer as die voorkoms van droë kernvrot (0.4 tot 6%). Isolasiestudies uit agt interne posisies van nat kernvrot appels het getoon dat Penicillium die dominante swamgenus in die meeste posisies was, insluitend die letsels. Morfologiese en molekulêre karakterisering van 4 Penicillium isolate uit nat kernvrot letsels het aangedui dat P. ramulosum prov. nom. die spesie is wat die meeste geïsoleer is vanuit die letsels, asook ander isolasie posisies. Dié spesie was egter ook die mees algemene spesie wat uit nat- en droë kernvrot, asimptomatiese appels en appels wat slegs swamgroei in die kern gehad het, geïsoleer is. Penicillium expansum was ook in lae getalle uit nat- en droë kernvrotletsels geïsoleer. Ander Penicillium spesies wat ook soms geïsoleer is, sluit P. glabrum, P. chloroloma, P. chermisinum, asook ‘n moontlik nuwe spesie wat op DNA volgorde basis die naaste aan P. dendriticum (P. spesie aff. dendriticum) is. Studies wat patogenesiteit en virulensie van die isolate ondersoek het, is ook uitgevoer deur gebruik te maak van drie verskillende inokulasie metodes. Die studies het aangedui dat P. expansum die mees virulente spesie is, gevolg deur P. spesie aff. dendriticum. Die spesifieke doelwitte was (1) om die ekologie van die Tarsonemus myte in “Red Delicious” en Granny Smith boorde tydens verskillende ontwikkelingstadiums van die appels te ondersoek, (2) om te bepaal of daar ‘n betekenisvolle assosiasie van Tarsonemus myte met siek (nat- en droë kernvrot) vrugte bestaan en (3) om te bepaal of potensiële kernvrot patogeniese swamme geassosieer is met die myte. Tarsonemus myte is gevind in al die ontwikkelingstadiums (knoppies, bloeisels, 4 sentimeter deursnee vrugte, volwasse vrugte en mummies) van appels wat ondersoek is. Die hoogste voorkoms van myte was in die mummies en volwasse vrugte van “Red Delicious”, asook Granny Smith kultivars gevind. In “Red Delicious” vrugte is myte in die kern en/of kaliksbuis gevind, maar in die Granny Smith vrugte was die myte tot die kaliksbuis beperk. In “Red Delicious” vrugte was daar ‘n betekenisvolle assosiasie tussen droë kernvrot, asook totale kernvrot (nat en droë kernvrot) met die teenwoordigheid van myte in die kern, asook totale myte (myte in die kern en kaliksbuis). Swam isolasiestudies vanaf die Tarsonemus myte het aangetoon dat potensiële kernvrot swampatogene in die genera Penicillium en Alternaria wel by die myte teenwoordig was. Die Penicillium spesies wat vanaf die myte geïsoleer is het twee van die mees virulente nat kernvrot spesies ingesluit, nl. P. expansum en P. spesie aff. dendriticum.

Temperature effects on the biology of Tetranychus cinnabarinus (Boisduval) and Typhlodromus occidentalis (Nesbitt) (Acarina: Phytoseiidae) were studied in the laboratory. Temperature had a definite effect on the developmental time, longevity and survival of adult females, and duration and rate of oviposition in both T. occidentalis and T. cinnabarinus. Increases in temperature significantly decrease developmental time, pre-oviposition periods, oviposition duration and rate, and the longevity and survival rates of both adult females. However, the daily consumption rate of T. occidentalis protonymph, deutonymph and adult gravid female increased significantly with an increase in temperature. Mean generation times decreased for both mites with an increase in temperature. The intrinsic rate of natural increase rose with each temperature increase. However, the r(m) of T. occidentalis was less than that for T. cinnabarinus at all temperature regimes, and it decreased from the 26.6° C regime to the 30.5° C regime. Of the three temperatures studied, 22.7°, 26.6°, and 3.5° C, 22.7° C proved to be the optimum for the predator to maintain control over the prey.

O conhecimento das interações entre Brevipalpus phoenicis (Geijskes), outros ácaros fitófagos ou predadores e as plantas de cafeeiro ainda são insuficientes para se poder elaborar um programa adequado de manejo da cultura, para tentar solucionar os problemas causados pela mancha anular. Apesar da importância do problema, pouco se sabe sobre a diversidade de ácaros, assim como a dinâmica populacional do ácaro B. phoenicis e seus inimigos naturais na cultura cafeeira, bem como nas diversas cultivares comercialmente exploradas. As informações sobre o impacto de agroquímicos nas populações de ácaros em cafeeiro são praticamente inexistentes. Contudo, sabe-se que alguns inseticidas ou fungicidas podem afetar a população de ácaros predadores podendo favorecer o aumento populacional de ácaros pragas. Ao caracterizar a diversidade de ácaros em duas importantes regiões produtoras (Jeriquara e Garça), constatou-se que em Jeriquara a diversidade foi superior ao encontrado em Garça. Foram coletados no total 13.052 ácaros nos dois locais estudados, sendo 7.155 em Jeriquara e 5.897 em Garça. De um total de 108 espécies encontradas, 45 espécies foram observadas em ambos os locais avaliados, que apresentaram similaridade de 56%. Os predadores mais freqüentes nas folhas foram Euseius citrifolius Denmark & Muma, E. concordis (Chant) e Agistemus brasiliensis Matioli, Ueckermann & Oliveira. Foram observadas correlações significativas a 0,05% (Pearson) entre as populações E. concordis e B. phoenicis; Zetzellia malvinae Matioli, Ueckermann & Oliveira e B. phoenicis; E. concordis e A. brasiliensis; entre outras. Em relação à diversidade em diferentes cultivares de cafeeiro (Coffea canephora cv Apoatã e de C. arabica cultivares Mundo Novo, Icatu Vermelho, Icatu Amarelo e Catuaí Amarelo) realizado em Garça, verificou-se que a maior riqueza de espécies e o maior número de indivíduos na superfície das folhas foram observados para Apoatã. A cultivar Icatu Vermelho foi a que apresentou maior uniformidade na distribuição das espécies de ácaros e Apoatã a que apresentou menor uniformidade. Em relação à preferência hospedeira, B. phoenicis foi encontrado em maior abundância na cultivar Apoatã, representando 61% de todos os indivíduos. E. citrifolius ocorreu em maior número na cultivar Mundo Novo e E. concordis na Apoatã. A. brasiliensis ocorreu em maior quantidade na cultivar Icatu Vermelho e Z. malvinae ocorreu sem diferença estatística em todas as cultivares. Em relação aos efeitos de pesticidas sobre a diversidade de ácaros, constatou-se que no tratamento com triadimenol + disulfoton apresentou a menor , enquanto que no deltametrina + triazophos a maior. B. phoenicis apresentou redução no número de indivíduos no tratamento com aldicarb e um aumento de cerca de duas vezes nos tratamentos com triadimenol + disulfoton e no thiamethoxam. Os tratamentos que apresentaram as maiores semelhanças na composição das espécies foram: testemunha e cartap, e thiamethoxam e ethion; e os tratamentos de menor similiaridade foram: testemunha e aldicarb, aldicarb e cartap, e aldicarb e deltametrina + triazophos. Reduções significativas na população de A. brasiliensis nos tratamentos com aldicarb e thiamethoxam e de E. citrifolius no tratamento com cartap foram detectadas. / The knowledge on the interactions among Brevipalpus phoenicis (Geijskes), other phytophagous or predaceous mites and coffee plants is still insufficient to establish an adequate management program for this crop, in order to minimize the problems caused by coffee ringspot virus (CoRSV), transmitted by B. phoenicis. In spite of the problem, little is known on the diversity of mites, as well as on the population dynamics of B. phoenicis and its natural enemies on coffee plants of several important commercial cultivars. Information on the impact of agrochemicals on the population of mites on coffee plantations are practically inexistent. However, it is known that some insecticides and fungicides can affect the population of predaceous mites inducing pest mite population increases. Characterizing the diversity of mites on two important coffee-producing areas of the state of São Paulo (Jeriquara and Garça), it was observed that the diversity of mites was higher in Jeriquara county than in Garça. A total of 13,052 mites was collected in both studied areas, of which 7,155 in Jeriquara and 5,897 in Garça. Of the total of 108 species identified in this study, 45 species were observed in both localities (Garça an Jeriquara), which presented 56% of similarity. The most frequent predaceous mites on leaves were Euseius concordis (Chant), E. citrifolius Denmark & Muma and Agistemus brasiliensis Matioli, Ueckermann & Oliveira, among others. The study on the diversity of mites in different coffee cultivars (Coffea canephora cv. ‘Apoat㒠and C. arabica cv. ‘Mundo Novo’, ‘Icatu Vermelho’, ‘Icatu Amarelo’ and ‘Catuai Amarelo’), carried out in Garça, showed the highest species richness and the highest number of specimens for the leaf surface of ‘Apoatã’. ‘Icatu Vermelho’ was the cultivar which presented the highest uniformity of distribution of mite species and ‘Apoat㒠was the cultivar with the lowest uniformity. The species B. phoenicis was found in higher abundance in ‘Apoatã’, representing 61% of all specimens collected. E. citrifolius occurred in higher number on the cultivar ‘Mundo Novo’ and E. concordis in ‘Apoatã’. The stigmaeid mite A. brasiliensis occurred in higher number on the cultivar ‘Icatu Vermelho’ and Z. malvinae was present in similar populations on all coffee cultivars. The study on the effect of pesticides on the mite diversity, showed the lowest diversity for the treatment with triadimenol + dissulfoton, and the highest diversity was observed for treatment with deltamethrin + triazophos. B. phoenicis presented population reduction for the treatments aldicarb, but an increase of around two times for treatments with triadimenol + disulfoton and with thiamethoxam. The treatments with the highest similarities in species composition were: cartap and control (without pesticide), and thiamethoxam and ethion; and the treatments with the lowest similarities were: control and aldicarb, aldicarb and cartap, and aldicarb and deltamethrin + triazophos. A significant reduction in A. brasiliensis population was detected for the treatments with aldicarb and thiamethoxam. E. citrifolius population was affected significantly by cartap.

Uma das principais espécies de ácaro praga é o ácaro rajado, Tetranychus urticae. Um dos inimigos naturais mais importantes deste ácaro é o patógeno Neozygites floridana. Para viabilizar o uso deste patógeno através de liberações inoculativas, ainda são necessárias informações básicas para subsidiar o desenvolvimento de uma metodologia de produção em escala e para liberações do fungo em campo. Para tanto, foi determinado em laboratório, a exigência de umidade e temperatura para a esporulação do fungo, colocando-se ácaros mumificados em folhas de feijão-de-porco. A 80 e 85% de umidade, em nenhuma das temperaturas (13, 17, 21 e 25°C) ocorreu esporulação. O fungo esporulou pouco a 90% UR em todas as temperaturas. As melhores condições para formação de capiloconídios foram a 95% e 100% UR a 21°C e 25°C. Ácaros mumificados foram colocados para esporular em feijão-de-porco em telados em diferentes épocas do ano e a temperatura e umidade monitorada. Notou-se que acima de 13°C o fungo conseguiu produzir um número considerável de conídios primários (>130) mesmo a 90% UR. Entretanto, nesta UR somente a 25°C a produção de capiloconídios foi significativa (>389). Determinou-se o efeito da exposição de duas intensidades luminosas (2.960 Lux e 15.392 Lux), três fotoperíodos (24h de luz, 12:12h(L:E) e 24h de escuro) e duas temperaturas (18°C e 23°C), na produção de conídios primários e de capiloconídios de isolados da Noruega e do Brasil. Ácaros mumificados foram expostos aos diferentes tratamentos em BOD. Não foram encontradas diferenças entre os dois isolados nas duas temperaturas testadas. Os resultados para o fotoperíodo de 12:12h(L:E) foram similares aos encontrados para 24h de escuro. A inibição da luz na formação de capiloconídios foi maior no isolado Norueguês. Baixa intensidade de luz por 24h teve efeito moderado na esporulação dos dois isolados, mas inibiu fortemente a formação de capiloconídios no isolado Norueguês. Foram realizados estudos para avaliar o efeito de agrotóxicos utilizados na produção de morango no Brasil e na Noruega sobre a esporulação e produção de capiloconídios de N. floridana em laboratório. Os resultados revelaram que Tebuconazol, Fenpropatrina na concentração recomendada e Abamectina na metade da concentração recomendada, foram os menos agressivos a N. floridana. Foram também realizadas tentativas de produção em escala deste fungo. Através de inoculações experimentais em telado foi possível induzir epizootias em populações de T. urticae em plantas de feijão de porco em vasos nos telados. Liberações de quatro ácaros mumificados por vaso foi a forma mais prática de inoculação. Maiores quantidades de ácaros mumificados foram produzidos quando as condições climáticas foram caracterizadas por alta UR e 20°C, sendo que durante o inverno a produção não foi eficiente. O uso de lâmpadas e ventiladores durante o período noturno ao final da epizootia, foi eficiente em inibir a esporulação e garantir a qualidade dos ácaros mumificados para o armazenamento. Embora tenha sido possível produzir um grande número de ácaros mumificados, vários estudos ainda são necessários para desenvolver uma metodologia de produção em escala para que seja economicamente viável a sua utilização em campo. / One of the main pest mite species is the Twospotted Spider Mite, Tetranychus urticae. The most important natural enemy of this mite is the pathogen Neozygites floridana. To enabling the use of this pathogen through inoculative releases, basic information is still needed to support the large scale methodology development for fungus production and release in the field. It was determined in the laboratory, the humidity and temperature requirements for fungus sporulation, by placing mummified mites on Jack-bean leaves. There was no sporulation at any of the temperatures tested (13, 17, 21 and 25°C) at 80% and 85% RH. The fungus had low sporulation at 90% RH at all temperatures. The best conditions for the capilloconidia development were 95% and 100%RH at 21°C and 25°C. Mummified mites were placed to sporulate in Jack-bean plants inside screenhouses at different times of year and temperature and humidity were monitored. It was noted that above 13°C the fungus can produce a considerable number of primary conidia (> 130) even at 90%RH. However, only at 90%RH and 25°C the capilloconidia production is significant (> 389). It was determined the effect of exposure of two light intensities (2.960 Lux and 15.392 Lux), three photoperiods (24h of light, 12:12h(L:D) and 24h of darkness) and two temperatures (18°C and 23°C), on the production of primary conidia and capilloconidia of N. floridana isolates from Norway and Brazil. Fungus-killed cadavers by each of the isolates were exposed to different treatments in BOD. No differences were found between the two isolates in the two temperatures tested. The results for the 12:12h(L:D) were similar to those found for 24h of darkness. Light inhibition of capilloconidia formation was greater on Norwegian isolate. Low light intensity for 24h had a mild effect on sporulation of the two isolates, but strongly inhibited the formation of capilloconidia of the Norwegian isolate. Studies were carried out to evaluate the effect of pesticides used in production of strawberries in both countries on sporulation and capilloconidia production of N. floridana in the laboratory. The results showed that Tebuconazole and Fenpropatrin in the recommended concentration and Abamectin in half of the recommended concentration were the less aggressive to N. floridana. Attempts to scale production of this fungus were also made. Through experimental inoculations it was possible to induce epizootics of the fungus in T. urticae populations on Jack-beans, planted in pots in screenhouses. The most convenient way for inoculation of the fungus was to release four mummified mites per pot. Larger quantities of mummified mites by N. floridana were produced when climatic conditions were characterized by high RH and 20°C, during the winter production was inefficient. The use of light and fans at night during the end of the epizootic phase, was effective in inhibiting the sporulation of the fungus and ensure the quality of mummified mites for storage. Although it was possible to produce a large number of mummified mites, several studies are still needed to develop an economically viable methodology for large scale production for field use.
