Identification and Characterization of GAS2L3 as a Novel Mitotic Regulator in Human Cells / Die Identifizierung und Charakterisierung von GAS2L3 als neuer Regulator der Mitose in humanen Zellen

Precise control of mitotic progression is vital for the maintenance of genomic integrity. Since the loss of genomic integrity is known to promote tumorigenesis, the identification of knew G2/M regulatory genes attracts great attention. LINC, a human multiprotein complex, is a transcriptional activator of a set of G2/M specific genes. By depleting LIN9 in MEFs, a core subunit of LINC, Gas2l3 was identified as a novel LINC target gene. The so far uncharacterized Gas2l3 gene encodes for a member of the family of growth arrest specific 2 (GAS2) proteins, which share a highly conserved putative actin binding CH and a putative microtubule binding GAS2 domain. In the present study GAS2L3 was identified as a LINC target gene also in human cells. Gene expression analysis revealed that GAS2L3 transcription, in contrast to all other GAS2 family members, is highly regulated during the cell cycle with highest expression in G2/M. The GAS2L3 protein showed a specific localization pattern during the M phase: In metaphase, GAS2L3 localized to the mitotic spindle, relocated to the spindle midzone microtubules in late anaphase and concentrated at the midbody in telophase where it persisted until the end of cytokinesis. Overexpression of a set of different GAS2L3 deletion mutants demonstrated that the localization to the mitotic microtubule network is dependent on the C-terminus, whereas the midbody localization is dependent on full length GAS2L3 protein. Additionally, exclusive overexpression of the CH domain induced the formation of actin stress fibers, suggesting that the CH domain is an actin binding domain. In contrast, the GAS2 domain was neither needed nor sufficient for microtubule binding, indicating that there must be an additional so far unknown microtubule binding domain in the C-terminus. Interestingly, immunoblot analysis also identified the C-terminus as the domain responsible for GAS2L3 protein instability, partially dependent on proteasomal degradation. Consistent with its specific localization pattern, GAS2L3 depletion by RNAi demonstrated its responsibility for proper mitosis and cytokinesis. GAS2L3 depletion in HeLa cells resulted in the accumulation of multinucleated cells, an indicator for chromosome mis-segregation during mitosis. Also the amount of cells in cytokinesis was enriched, indicating failures in completing the last step of cytokinesis, the abscission. Strikingly, treatment with microtubule poisons that lead to the activation of the spindle assembly checkpoint (SAC) indicated that the SAC was weakened in GAS2L3 depleted cells. Although the exact molecular mechanism is still unknown, fist experiments support the hypothesis that GAS2L3 might be a regulator of the SAC master kinase BUBR1. In conclusion, this study provides first evidence for GAS2L3 as a novel regulator of mitosis and cytokinesis and it might therefore be an important guardian against tumorigenesis. / Der korrekte Verlauf durch die Mitose des Zellzyklus trägt entscheidend zur Aufrechterhaltung der genomischen Integrität bei. Da ein Verlust der genomischen Integrität die Tumorentstehung begünstigt, ist die Identifizierung neuer G2/M regulatorischer Gene ein Forschungsbereich, der großes Interesse weckt. Der humane Multiproteinkomplex LINC ist für die transkriptionelle Aktivierung einer Vielzahl G2/M spezifischer Gene verantwortlich. Durch die Depletion von LIN9 in MEFs, einer Kernkomponente von LINC, wurde Gas2l3 als ein neues Zielgen von LINC identifiziert. Das bisher uncharakterisierte Gas2l3 Gen codiert für ein der GAS2 (growth arrest specific 2) Familie zugehöriges Protein, deren Mitglieder sich durch eine hoch konservierte putative Aktin-bindende Domäne (CH) und eine putative Mikrotubuli-bindende Domäne (GAS2) auszeichnen. In der vorliegenden Arbeit konnte gezeigt werden, dass GAS2L3 auch in humanen Zellen ein Zielgen von LINC ist. Die Transkription von GAS2L3 wies, im Gegensatz zu allen anderen GAS2 Familienmitgliedern, eine starke Regulation während des Zellzyklus auf, wobei die höchste Genexpression in der G2/M Phase vorlag. Das GAS2L3 Protein zeigte eine spezifische Lokalisation während der M Phase: In der Metaphase findet sich GAS2L3 an der mitotischen Spindel, wandert von dort an die Mikrotubuli der zentralen Spindel der Anaphase und konzentriert sich in der Telophase am Midbody, wo es bis zum Ende der Zytokinese verweilt. Der Einsatz unterschiedlicher Deletionsmutanten demonstrierte, dass die Lokalisation an die mitotischen Mikrotubuli vom C-Terminus abhängig ist, wohingegen die Lokalisation am Midbody von der gesamten Proteinsequenz abhängt. Die Ausbildung von Aktin-Streß-Filamenten nach alleiniger Überexpression der CH Domäne deutete darauf hin, dass die CH Domäne eine Aktin-bindende Domäne ist. Die GAS2 Domäne hingegen wurde weder für die Interaktion mit Mikrotubuli gebraucht, noch war sie alleine für diese ausreichend. Alle Daten weisen darauf hin, dass GAS2L3 eine bisher unbekannte Mikrotubuli-bindende Domäne im C-Terminus trägt. Interessanterweise ist der C-Terminus auch für die hohe Instabilität des GAS2L3 Proteins, die teilweise durch den Abbau im Proteasom verursacht wird, verantwortlich. Entsprechend der spezifischen Lokalisation zeigte die Depletion von GAS2L3 durch siRNA Transfektion dessen Wichtigkeit für den korrekten Verlauf der M Phase. GAS2L3 depletierte HeLa Zellen zeigten eine Anreicherung von multinukleären Zellen, welche ein Indikator für die fehlerhafte Verteilung der Chromosomen in der Mitose sind. Ein Hinweis auf Probleme im Beenden der Zytokinese stellte die erhöhte Anzahl von Zellen dar, die sich in der Zytokinese befanden. Eines der auffallendsten Merkmale war ein geschwächter mitotischer Spindelkontrollpunkt, den GAS2L3 depletierte Zellen nach der Behandlung mit den Kontrollpunkt aktivierenden Mikrotubuli-Giften aufwiesen. Auch wenn der exakte molekulare Mechanismus hierbei noch unbekannt ist, deuten erste Experimente darauf hin, dass GAS2L3 die Aktivität von BUBR1, einer essentiellen Kinase des mitotischen Spindelkontrollpunkts, beeinflusst. Alle Daten dieser Arbeit verdeutlichen die Wichtigkeit von GAS2L3 als einen neuen Regulator der Mitose und Zytokinese. Somit ist anzunehmen, dass die korrekte Funktion von GAS2L3 entscheidend zum Schutz vor Tumorentstehung beiträgt.

Die Bedeutung von LIN9 für die Regulation der Genexpression, die genomische Stabilität und die Tumorsuppression / The significance of LIN9 for gene regulation, genomic stability and tumor suppression

Pocket proteins and E2F transcription factors regulate the expression of cell cycle associated genes and play a central role in the coordination of cell division, differentiation, and apoptosis. Disorders of these pathways contribute to the development of various human tumor entities. Despite intensive research in the field of cell cycle regulation many details are not yet understood. The LIN complex (LINC / DREAM) is a recently discovered human multiprotein complex, which dynamically interacts with pocket proteins and E2F transcription factors. An essential component of the LIN complex is the LIN9 protein. In order to obtain a better insight into the function of this protein in cell cycle regulation and tumorigenesis, a conditional Lin9 knockout mouse model was established in our laboratory. The primary objective of this study was the phenotypic characterization of embryonic fibroblasts (MEFs) from these mice. Shortly after inactivation of Lin9 cell proliferation was massively impaired. Multiple types of mitotic defects such as structural abnormalities of the spindle apparatus, aberrant nuclei, failed nuclear segregation and cytokinesis failure have been observed in Lin9-depleted cells leading to a dramatic increase in polyploid and aneuploid cells. Ultimately these serious aberrations result in premature cellular senescence. If the senescence of Lin9-deficient cells is overcome by the Large T antigen the cells can adhere to the loss of Lin9, but show severe genomic instability and grow anchorage-independently in soft-agar as a sign of oncogenic transformation. In the second part of the thesis the gene expression of Lin9-deficient cells was assessed by quantitative real time PCR analyses to determine, whether the mitotic abnormalities are caused by transcriptional defects. Here a significant reduction of mitotic gene expression was observed in Lin9-depleted cells. Additionally chromatin immunoprecipitation experiments were performed to clarify the underlying molecular mechanisms. Compared to control cells epigenetic alterations at the promoters of mitotic target genes with regard to activating histone modifications were found in Lin9-deficient MEFs. In the last section of this study, the effects of Lin9 heterozygosity were analyzed. Lin9 heterozygous MEFs showed normal proliferation, although expression of different mitotic genes was slightly reduced. It appeared, however, that the mitotic spindle checkpoint of Lin9 heterozygous MEFs is weakened and thus over several cell generations an increase in polyploid cells was observed. Soft-agar assays showed that Lin9 heterozygosity contributes to oncogenic transformation. Taken together, these results document a crucial role of LIN9 in the regulation of cell cycle-associated gene expression. LIN9 is an essential factor for cell proliferation on one hand, while at the same time it functions as a tumor suppressor. / Pocket-Proteine und E2F-Transkriptionsfaktoren regulieren die Expression von Zellzyklus-assoziierten Genen und spielen eine zentrale Rolle bei der Koordination der Zellteilung, Differenzierung und Apoptose. Störungen dieser Signalwege tragen zur Entstehung zahlreicher Tumorentitäten beim Menschen bei. Trotz der intensiven Untersuchung der Zellzyklusregulation sind viele Details noch unverstanden. Der LIN-Komplex (LINC / DREAM) ist ein kürzlich entdeckter humaner Multiprotein-komplex, welcher dynamisch mit Pocket-Proteinen und E2F-Transkriptionsfaktoren interagiert. Eine essentielle Komponente des LIN-Komplexes ist das LIN9-Protein. Um die Funktion dieses Proteins bei der Zellzyklusregulation und Tumorentstehung genauer untersuchen zu können, wurde in unserer Arbeitsgruppe ein konditionelles Lin9-Knockout-Mausmodell etabliert. Primäres Ziel der Arbeit war es, den Phänotyp embryonaler Fibroblasten (MEFs) aus diesen Mäusen zu charakterisieren. Bereits kurz nach Inaktivierung von Lin9 konnte ein stark verlangsamtes Zellwachstums beobachtet werden. In Lin9-depletierten MEFs wurden multiple mitotische Defekte detektiert, die u. a. strukturelle Auffälligkeiten des Spindelapparates, aberrante Zellkerne, Störungen der Chromosomensegregation sowie zytokinetische Defekte umfassen und in einer dramatischen Zunahme polyploider und aneuploider Zellen resultieren. Im Langzeitverlauf führen diese erheblichen Aberrationen zu einer vorzeitigen zellulären Seneszenz. Wird diese durch das Large T-Protoonkogen durchbrochen, können sich MEFs an den Verlust von Lin9 adaptieren, zeigen dann jedoch eine hochgradige genomische Instabilität und Substrat-unabhängiges Wachstum im Weichagar als Zeichen onkogener Transformation. Im zweiten Abschnitt der vorliegenden Arbeit wurde die Genexpression in Lin9-defizienten MEFs mittels quantitativer Real Time-PCR untersucht um zu klären, ob die beschriebenen Defekte auf Veränderungen der transkriptionellen Aktivität zurück-zuführen sind. Dabei wurde eine erhebliche Reduktion der Expressionslevel mitotischer Gene nach Verlust von Lin9 beobachtet. Des Weiteren wurden zur Klärung der zu Grunde liegenden molekularen Mechanismen Chromatin-Immunpräzipitations-Experimente (ChIP) durchgeführt. Im Vergleich zu Kontrollzellen wurden dabei in Lin9-defizienten Zellen signifikante epigenetische Veränderungen bezüglich aktivierender Histon-Modifikationen an den Promotoren mitotischer Lin9-Zielgene festgestellt. Im letzten Abschnitt der Arbeit sollten die Auswirkungen des heterozygoten Verlustes von Lin9 analysiert werden. Dabei zeigte sich, dass Lin9-haploinsuffiziente Zellen normal proliferieren, obwohl die Expression verschiedener G2/M-Gene leicht vermindert war. Es wurde jedoch eine Schwächung des mitotischen Spindelkontrollpunktes und in der Folge über mehrere Zellgenerationen eine Zunahme polyploider Zellen beobachtet. Mit Weichagar-Assays konnte gezeigt werden, dass bereits der heterozygote Verlust des Lin9-Gens zur onkogenen Transformation beiträgt. Zusammengenommen dokumentieren diese Studien, dass LIN9 eine entscheidende Bedeutung bei der Regulation von Zellzyklus-assoziierten Genen spielt und sowohl einen essentiellen Faktor für die Zellproliferation darstellt als auch durch die Gewährleistung genomischer Stabilität tumorsuppressive Eigenschaften aufweist.

Análise de novos agentes quimioterápicos em linhagens celulares de câncer gástrico humano

O câncer gástrico é a terceira causa de morte por câncer no mundo e a quimioterapia combinatória é um dos principais tratamentos para esta doença. Contudo, a principal causa de falha do tratamento é a quimiorresistência. Considerando este obstáculo juntamente com a descoberta de que muitas proteínas envolvidas na mitose podem estar associadas à tumorigênese, vários agentes antimitóticos estão sendo desenvolvidos e testados. As proteínas mitóticas AURKB e NDC80 foram recentemente identificadas como proteínas potenciais a serem inibidas no câncer gástrico. Portanto, o objetivo deste estudo foi analisar os efeitos da inibição de AURKB e NDC80 pelos fármacos experimentais ZM447439 e INH1, respectivamente. Analisamos os efeitos em duas linhagens de adenocarcinoma gástrico avaliando expressão gênica, ciclo celular, morte celular, análise morfométrica nuclear, capacidade de migração e presença de células-tronco tumorais. É importante enfatizar que essas duas pequenas moléculas não foram testadas na quimioterapia do câncer gástrico até o momento. Além disso, após uma revisão da literatura, concluímos que os melhores resultados dos ensaios clínicos foram alcançados quando antimitóticos foram combinados com a terapia convencional. Dessa forma, avaliamos também os efeitos da associação de 5- FU com ZM447439. Nossos resultados demonstraram que a monoterapia com 5-FU, ZM447439 e INH1 induziu a expressão gênica diferencial nas linhagens de câncer gástrico (ACP02 e ACP03) de pelo menos 22 de um total de 82 genes envolvidos em várias vias, como apoptose, ciclo celular, senescência e transição epitélio-mesenquimal. Observamos também que ZM447439 e sua combinação com 5-FU induziram apoptose, catástrofe mitótica, senescência e ciclo celular alterado nas linhagens de câncer gástrico. Outro dado interessante foi a expressão dos marcadores de células-tronco tumorais gástricas, LGR5 e CD24, e a capacidade de formar esferas, indicando que ambas ACP02 e ACP03 podem conter células tronco tumorais. Além disso, a monoterapia com ZM447439 ou 5-FU foi capaz de inibir a formação de esferas. Por outro lado, o INH1 mostrou menor atividade antitumoral, uma vez que apresentou o maior valor de IC50 e foi capaz somente de induzir apoptose e reduzir a migração celular. Interessantemente, a combinação do 5-FU com ZM447439 permitiu o uso de doses menores dos compostos, além de ser capaz de induzir a apoptose num tempo mais curto quando comparado com o tratamento com os fármacos isolados. Em síntese, os resultados observados sugerem o inibidor de AURKB, ZM447439, e sua associação com o agente quimioterápico 5-FU, como tratamentos promissores do câncer gástrico com base em sua elevada atividade antitumoral in vitro. / Gastric cancer is the third cause of cancer death worldwide and combinatorial chemotherapy is one of the main treatments for this disease. However, the major cause of treatment failure is chemoresistance. Considering this obstacle together with the discovery that many proteins involved in mitosis might be associated to tumorigenesis, several new anti-mitotics are being developed and tested. The mitotic proteins AURKB and NDC80 were recently shown as potential proteins to be inhibited in gastric cancer. Therefore, the aim of this study was to analyze the effects of AURKB and NDC80 inhibition by the experimental drugs ZM447439 and INH1, respectively. We analyzed the effects in two gastric adenocarcinoma cell lines evaluating gene expression, cell cycle, cell death, nuclear morphometric analysis, migration ability and presence of gastric cancer stem cells. It is important to emphasize these two small molecules have not been tested in gastric cancer chemotherapy until now. Moreover, after reviewing the literature, we concluded the best results of clinical trials were achieved when anti-mitotics were combined with conventional therapy. Thus, we evaluated the effects of 5-FU and ZM447439 combination as well. Our results demonstrated that monotherapy with 5-FU, ZM447439 and INH1 induced differential gene expression in gastric cancer cell lines (ACP02 and ACP03) of at least 22 from 82 genes involved in several pathways such as apoptosis, cell cycle, senescence and epithelial mesenchymal transition. We also observed that ZM447439 and its combination with 5-FU induced apoptosis, mitotic catastrophe, senescence and altered cell cycle in gastric cancer cell lines. Another interesting data was the expression of the gastric cancer stem cell markers LGR5 and CD24, and the ability to form spheres, indicating that both ACP02 and ACP03 may be composed by cancer stem cells. Furthermore, monotherapy with either ZM447439 or 5-FU were capable of inhibiting the formation of spheres. On the other hand, INH1 have shown lower antitumoral activity since it presented the highest IC50 value and was only capable of inducing apoptosis and reducing cell migration. Interestingly, the combination of 5-FU with ZM447439 allowed the use of smaller doses of the compounds, in addition to being able to induce apoptosis in a shorter time when compared to the treatment with the isolated drugs. Taken together, our findings suggest that due to the high antitumoral activity shown by ZM447439 and mainly its association with 5-FU, these might be promising gastric cancer therapies.

Les microtubules sont des composants hautement dynamiques du cytosquelette. La tubuline gamma est localisée au centrosome. Elle y forme le complexe de nucléation des microtubules, le gamma-TuRC, en association avec les protéines GCPs 2-6. Les GCPs 2-6 forment une famille de protéines caractérisée par deux domaines conservés appelés GRIP1 et 2. De par sa structure, le gamma-TuRC sert de moule pour la nucléation des microtubules. Le gamma-TuRC est constitué de plusieurs sous-complexes : les gamma-TuSCs qui sont composés d'une GCP2 et d'une GCP3 qui interagissent entre elle par leur domaine amino-terminal, chacune liant une tubuline gamma via leur domaine carboxy-terminal. Les gamma-TuSCs s'assemblent latéralement pour former une structure à un tour d'hélice, les deux extrémités de l'hélice se recouvrant. La structure atomique de GCP4 s'intègre particulièrement bien dans structure du gamma-TuSC de levure, obtenue en microscopie électronique, à la place de GCP2 et 3 suggérant une forte conservation structurale entre les GCPs. GCP4, 5 et 6 pourraient donc être partie intégrante de l'hélice. Durant ma thèse j'ai étudié la position relative des GCP4, 5 et 6 au sein du gamma-TuRC. Pour cela j'ai développé des approches d'échange de domaines et de mutagénèse. J'ai également mis en place des stratégies de FLIM-FRET et d'immunoprécipitation. J'ai ainsi montré que c'est le domaine N-terminal des GCPs qui définit leur identité, les domaines C-terminaux étant échangeables. J'ai également mis en évidence, au sein du gamma-TuRC, des interactions latérales entre GCP4 et GCP5 semblables à celles établies par GCP2 et GCP3 dans les gamma-TuSC. J'ai également pu isoler un complexe contenant GCP4, 5, 6 et la tubuline gamma indépendamment du gamma-TuRC. J'apporte ainsi les premières preuves expérimentales soutenant l'idée que GCP4, 5 et 6 sont partie intégrante de l'hélice du gamma-TuRC et qu'elles y forment un sous complexe qui occupe une position bien définie. / Microtubules are highly dynamic components of the cytoskeleton. gammatubulin is found at the centrosome where it forms a microtubule nucleation complex together with GCPs 2-6, the gamma-TuRC. GCPs 2-6 form a conserved family of proteins characterised by two conserved domains called GRIP1 and 2. The gamma-TuRC functions as a structural template for microtubule nucleation. The gamma-TuRC is composed of smaller subcomplexes called gamma-TuSC. Each gamma-TuSC is composed by one GCP2, one GCP3 and two gamma?tubulins. GCP2 and GCP3 interact via their N-terminal domain and bind gamma tubulin through their C-terminal domain. Several gamma-TuSCs can assemble laterally to form a one-turn helix with the two ends overlapping. The atomic structure of GCP4 fits almost perfectly in the place of GCP2 and GCP3 within the gamma-TuSC envelope obtained by electron microscopy suggesting a strong structural conservation among GCPs. Hence, GCP4, 5 and 6 may be part of the helix. During the course of my thesis, I studied the relative position of GCPs 4, 5, 6 within the gamma-TuRC. To this aim, I developed a domain swapping and mutagenesis approaches. I also combined FLIM-FRET and immunoprecipitation strategies. I have been able to show that the N-terminal domains of GCPs define their identity while the C-terminal domains can be swapped. My results also indicate that GCP4 and GCP5 establish gamma-TuSC like interactions within the gamma-TuRC. I also isolated a complex containing GCP4, 5, 6 and gamma tubulin independently of the gamma-TuRC. My thesis provides the first experimental evidence supporting the model where GCP4, 5 and 6 are part of the gamma-TuRC helix where they form a sub-complex localised at a defined position.

Rôle du Rho-GEF Trio dans la division cellulaire / Role of Rho-GEF Trio in the cell division

Durant la division cellulaire, la cellule subit des changements importants dans sa forme et son adhésion qui dépendent de l'efficacité du remodelage du cytosquelette d'actine. Ce processus est localement et temporellement régulé pour assurer le bon déroulement de la cytokinèse, l'étape finale de la division cellulaire. Il est contrôlé par les GTPases de la famille Rho via le remodelage du cytosquelette d'actine. Les Rho-GTPases fonctionnent comme des interrupteurs moléculaires, passant d'une forme au repos (liée au GDP) à une forme active (liée au GTP). La forme au repos interagit avec des facteurs d'échange, les GEFs (Guanine nucleotide Exchange Factors) qui déplacent le GDP et permet la fixation du GTP. Le retour à la forme inactive se fait par hydrolyse du GTP en GDP, stimulée par les protéines GAPs (GTPase Activating Proteins). RhoA est un régulateur positif de la cytokinèse, activée spécifiquement à l'équateur de la cellule, et qui promeut l'assemblage et la constriction de l'anneau d'actomyosine. En contraste, Rac1 a été proposée pour réguler négativement ce processus et doit être inactivée spécifiquement à l'équateur de la cellule pour le bon déroulement de la cytokinèse. Ainsi, une GAP de Rac1, MgcRacGAP, qui est localisé sur le fuseau central de microtubules, inactive Rac1 à l'équateur de la cellule. La déplétion de MgcRacGAP induit des défauts de cytokinèse qui peuvent être sauvés en co-déplétant Rac1. Cependant, le Rho-GEF activant Rac1 durant la division cellulaire n'a pas encore été identifié. Pour identifier un GEF régulant l'activité de Rac1 dans les cellules en division, nous avons réalisé une approche de « screening » par siRNA dans les cellules HeLa. Les Rac-GEFs sont déplétés par siRNA seul ou en combinaison avec un siRNA ciblant MgcRacGAP, dans le but d'identifier lesquels sont capables de sauver le nombre de cellules multinuclées induit par la déplétion de MgcRacGAP. De façon intéressante, la co-déplétion de MgcRacGAP et du Rho-GEF Trio, un GEF caractérisé principalement pour son rôle dans la croissance et le guidage axonal, entraîne une forte diminution du nombre de cellules multinuclées. Par la suite, nous démontrons que ce sauvetage du phénotype passe par la voie Trio-Rac1 en utilisant des mutants GEFs inactifs de Trio et un inhibiteur spécifique de l'activation de Rac1 par Trio. Ces résultats et le rôle de MgcRacGAP dans l'inactivation de Rac1 en cytokinèse, suggèrent que la déplétion de Trio pourrait sauver les défauts de cytokinèse induits par la déplétion de MgcRacGAP en diminuant l'activité de Rac1. Cela suggère aussi que Trio pourrait être un GEF de Rac1 dans les cellules en division. Pour directement tester si Trio pouvait fonctionner comme un GEF de Rac1 dans les cellules en division, la quantité de Rac1 a été mesurée par « pull-down assay » dans des cellules synchronisées en mitose. Comparé aux cellules traitées avec un siRNA contrôle, la déplétion de Trio réduit de moitié la quantité de Rac1 activée dans les cellules en mitose, démontrant que Trio active Rac1 en mitose. De plus, la déplétion de Trio induit des défauts de remodelage du cytosquelette d'actine dans les cellules en anaphase. De façon intéressante, la déplétion de Trio phénocopie la déplétion de Rac1 et de son effecteur Arp2/3, en accord avec un rôle de la voie Trio-Rac1 dans le contrôle du remodelage du cytosquelette d'actine dans les cellules en division. L'ensemble de ce travail a permis d'identifier pour la première fois un GEF contrôlant l'activité de Rac1 dans les cellules en division dont l'activité s'oppose à la fonction de MgcRacGAP en cytokinèse. Nous proposons ainsi un modèle dans lequel Trio contrôle l'activation de Rac1 et le remodelage du cytosquelette d'actine au cortex cellulaire dans les cellules en division. Dans notre modèle, MgcRacGAP s'oppose à l'action de Trio en inhibant localement et temporellement l'activation de Rac1 au plan de division, assurant ainsi le bon déroulement de la cytokinèse. / During cell division, cells undergo dramatic changes in shape and adhesion that depend on efficient actin cytoskeleton remodeling. This process has to be locally and temporally regulated to accurately ensure cytokinesis, the final stage of cell division. The small GTPases Rac1 and RhoA play an essential role in this process by controlling F-actin cytoskeleton remodeling. GTPases oscillate between an inactive, GDP-bound state and an active, GTP-bound state. They are activated by Guanine-nucleotide Exchange Factors (GEFs), which stimulate the GDP-to-GTP exchange, while they are turned off by GTPase-Activating Proteins (GAPs) which catalyse the hydrolysis of GTP. RhoA is a positive regulator of cytokinesis specifically activated at the division plane, which promotes the assembly and constriction of the actomyosin network. In contrast, Rac1 has been proposed to negatively regulate this process and has to be inactivated at the division plane for cytokinesis to occur properly. A central spindle localized GAP, MgcRacGAP, component of the centralspindlin complex, controls Rac1 inactivation at the cleavage plane. Depletion of Rac1 can suppress the cytokinesis failure induced by MgcRacGAP depletion. However, the Rho-GEF that activates Rac1 during cell division has not been identified yet. To identify a GEF regulating Rac1 activity in dividing cells, we performed a siRNA screening approach in HeLa cells. Rac-GEFs were depleted by siRNA alone or in combination with MgcRacGAP siRNAs, in order to identify the ones able to rescue the multinucleated cells induced by MgcRacGAP depletion. Importantly, co-depletion of MgcRacGAP and Rho-GEF Trio, a GEF characterized primarily for its role in axon outgrowth and guidance resulted in a strong decrease in the number of multinucleated cells. Then, we demonstrate that this rescue is mediated by the Trio-Rac1 pathway, using GEF dead mutants of Trio and a specific inhibitor of Rac1 activation by Trio. These data and the fact that MgcRacGAP was recently described to be essential for Rac1 inactivation in cytokinesis, suggest that Trio depletion could rescue the cytokinesis failure induced by MgcRacGAP depletion by decreasing Rac1 activity. It therefore suggests that Trio could be a GEF of Rac1 in dividing cells. To directly test if Trio could function as a GEF of Rac1 in dividing cells, the amount of activated Rac1 was monitored by pull down assay in synchronized mitotic cells. Compared to control siRNA-treated cells, Trio depletion reduced by half the amount of activated Rac1 in mitotic cells, showing that Trio activates Rac1 in mitosis. Strikingly, Trio depletion led to defects in F-actin cytoskeleton remodeling in anaphase cells. Indeed, the F-actin staining at the cortex was significantly reduced in Trio-depleted cells compared to control cells. Interestingly, Trio depletion phenocopied the depletion of Rac1, consistent with a role for the Trio-Rac1 pathway in controlling F-actin remodeling in dividing cells.Overall, this work identifies for the first time a GEF controlling Rac1 activation in dividing cells that counteracts MgcRacGAP function in cytokinesis. Based on these observations, we propose a model in which Trio functions as a GEF of Rac1 during cell division. Trio, which is expressed throughout the cell cycle, activates Rac1 to control F-actin cytoskeleton remodeling at the cell cortex of dividing cells. MgcRacGAP therefore counteracts the action of Trio by locally and temporally inhibiting Rac1 activation at the division plane, subsequently ensuring accurate cytokinesis.

Caractérisation du rôle des protéines phosphatases impliquées dans la déphosphorylation de la protéine kinase Greatwall lors de la sortie de mitose / Characterization of phosphatases involved in mitosis exit and its regulation mechanisms

Chez la drolosophile, des mutants de Greatwall présentent des défauts de condensation des chromosomes lors de la mitose. Plus tard, la même équipe a montré que chez le Xénope, Greatwall est nécessaire pour entrer en mitose. L'idée consistant à penser que puisque Greatwall ne permet plus l'entrée en mitose, il joue un rôle dans la boucle qui conduit à l'auto-amplification de MPF. En 2009, notre équipe a montré que Greatwall est réellement impliquée dans l'entrée en mitose, mais de façon indirecte par rapport à la boucle d'amplification de MPF, et cela en contrôlant l'activité de la phosphatase PP2A. Notre équipe a montré que lorsque l'on enlève PP2A, on peut sauver le phénotype de l'absence de Greatwall. Plus tard, il a été montré que la phosphorylation de Greatwall est nécessaire pour l'entrée en mitose. La phosphorylation de Greatwall sur la partie C-terminale est nécessaire pour activer Greatwall. Par conséquent, Greatwall doit être phosphorylé pour être actif. Une fois activé, Greatwall est capable de phosphoryler Arpp19 qui lie la phosphatase PP2AB55, et qui l'inhibe permettant ainsi de maintenir les phosphorylations des substrats mitotiques. Si cette voie de signalisation n'est pas fonctionnelle, la phosphatase PP2A va déphosphoryler tous les substrats mitotiques et la cellule n'entrera jamais en mitose. Greatwall doit être phosphorylé pour s'activer et pour entrer en mitose, mais on observe aussi qu'au moment de la sortie de mitose, il est déphosphorylé, et il doit être déphosphorylé pour s'inactiver. (On ne sait pas s'il est requisse pour sortir). Mon projet consiste à chercher la/les phosphatase(s) qui pourrait contrôler l'activité ou l'inactivation de Greatwall. Les questions que l'on se pose : Comment et par quelle(s) phosphatase(s) Greatwall est déphosphorylé, comment ces phosphoatases sont activées, quel est l'ordre d'activation de ces phosphatases ? Pour étudier comment Greatwall est déphosphorylé, il y a 2 sites majors : T194 et S875. Ces 2 sites sont nécessaires pour l'activité de Greatwall. Nous avons réalisé les 2 mutants T194A et S875A, et les traduit dans l'extrait interphasique d'œufs de Xénope, pour mesurer l'activité de kinase Greatwall. Pour déphosphoryler ces 2 sites, il y a 4 phosphatases principales comme candidats : Calcineurine, Fcp1, PP1, PP2A. / The establishment of mitosis requires phosphorylaton of several substrates induced by kinases. Cdk1-cyclin B and Greatwall kinases are both necessary for the entry into mitosis. Cdk1-cyclin B complex phosphorylates many substrates and at the same time Greatwall phosphorylates Arpp19 which binds PP2AB55 phosphatase and inhibits it. PP2AB55 has an important role in the dephosphorylation of Cdk1-cyclin B mitotic substrates.In my laboratory, we found that after Greatwall depletion, either in Xenopus egg extracts or in human cells, PP2A is no longer inhibited and cells exit mitosis. Since activation of Greatwall requires its phosphorylation in the c-terminal part and in the T-loop site, we suppose that mitosis exit require dephosphorylation of Greatwall. So these dephosphorylations could be involved for Greatwall inactivation. Several phosphatases are candidates for this process: Fcp1, PP1, PP2A and Calcineurin. My project proposes to determine the involvement of these four phosphatases in Xenopus egg extracts after depletion and overexpression of these four proteins.

La chromatine est le support de l'information génétique tout le long du cycle cellulaire. Elle est soumise à des modifications diverses et rigoureusement coordonnées par les complexes CDK-Cyclines, sous le contrôle de mécanismes de surveillance. En mitose, la kinase Aurora est un acteur clé qui contrôle la ségrégation correcte des chromosomes. Aurora participe à la bi-orientation des centromères, à la condensation des chromosomes et à la cytocinèse. Le dysfonctionnement de cette kinase conduit alors à une instabilité chromosomique et à l'aneuploïdie, un phénotype observé dans la majorité des cancers solides. Les travaux réalisés au cours de cette thèse démontrent un nouveau rôle pour cette kinase dans la dispersion et la disjonction des télomères en mitose chez la levure S. Pombe. La dispersion des télomères s'accompagne en métaphase de la dissociation aux télomères des protéines Swi6/HP1 et cohésine Rad21. Tandis que la disjonction a lieu en anaphase après la phosphorylation de la sous-unité de condensine Cnd2. L'inhibition d'Aurora induit la formation de ponts chromosomiques anaphasiques qui révèle des défauts de séparation des télomères. La délétion d'une protéine spécifique aux télomères, Ccq1, protège la cellule de la formation de ces ponts chromosomiques en favorisant le chargement de la condensine en anaphase malgré l'inhibition d'Aurora. / Chromatin is the support of the genetic information throughout the cell cycle. It is subject to various modifications that occur with precise coordination. This coordination is led by CDK-cyclins under the control of cell cycle checkpoints. In mitosis, correct chromosome segregation is ensured by Aurora kinases. Aurora participates to centromere bi-orientation, chromosome condensation and cytokinesis. A dysfunction in the activity of this kinase leads to chromosomal instability and aneuploidy, phenotypes frequently observed in cancer. The results obtained during this thesis reveal a new function for fission yeast Aurora kinase during mitosis in telomere dispersion and disjunction. Telomere dispersion is triggered in metaphase by the dissociation of Swi6/HP1 and cohesion Rad21 from telomeres. Then, during anaphase, the phosphorylation of the condensin subunit Cnd2 is required for telomere disjunction. Aurora inhibition leads to anaphase chromosome bridges with unseparated telomeres. Deletion of a specific telomeric protein, Ccq1, prevents the formation of anaphase chromosome bridges by favoring condensin loading despite Aurora inhibition.

Les microtubules (MT) sont des polymères dynamiques ancrés par leurs extrémités moins aux centres de nucléation alors que leurs extrémités plus, explorent le cytoplasme, jusqu’à être stabilisées. Cette capture des extrémités permet l’organisation du réseau des MT. Les +TIP sont un groupe de protéines qui s’associent aux bouts plus des MT. EB1 est une protéine centrale dans le réseau des +TIP qui régule la dynamique des MT et leur interaction avec les structures d’ancrage des extrémités plus. Par protéomique ciblée, nous avons caractérisé l’interactome d’EB1, et mis en évidence un groupe de protéines, précédemment associées aux centres de nucléation incluant AKAP9, une protéine échafaudage pour les protéines kinases A (PKA), la protéine de la matrice péricentriolaire CDK5RAP2, et une isoforme courte de la myomégaline que nous avons appelé SMYLE (Short MYomegalin Like EB1 binding protein). La cartographie moléculaire a permis de montrer que ces protéines formaient un complexe organisé de manière hiérarchique. Nous avons observé que l’association transitoire deLa protéine SMYLE (Short MYomegalin Like EB1 binding protein )dans l'organisation d'un complexe centrosomal, la régulation de la nucléation et la stabilisation des microtubules : conséquences sur la migration et la division des cellules cancéreuses avec les MT néo-nucléés au centrosome favorisait la nucléation et l’acétylation des MT. De manière notable, la déplétion de SMYLE aboutissait à un défaut de nucléation, mais aussi de la capture corticale des MT. Ces défauts dans l’organisation des MT étaient associés à une baisse notable de la migration des cellules de carcinome mammaire et à des anomalies mitotiques. Nos résultats nous permettent de proposer que SMYLE fait partie d’un complexe centrosomale, qui favorise l’assemblage ou la stabilité des microtubules néo-nucléés, contribuant ainsi à des processus majeurs pour le développement tumoral. / Microtubules (MT) are dynamic polymers anchored by their minus ends at the MT organizing centers while their highly dynamic plus end explores the cytoplasm until it get stabilized. This plus end capture allows the organization of the MT network. +TIPs are a group of proteins that share the commonality to associate either directly or indirectly to MT plus ends. EB1 is a central protein of the +TIP network that regulates MT dynamics and their interactions with plus end anchoring structures. Using targeted proteomics, we have characterized the EB1 interactome and revealed a set of protein previously shown to associate with the nucleating centers that included AKAP9 an anchoring protein for protein kinase A (PKA), the pericentriolar matrix protein CDK5RAP2 and a short Myomegalin isoform that we named SMYLE (Short MYomegalin Like EB1 binding protein). Molecular mapping revealed that the proteins formed a hierarchically organized complex. We have observed that the transient association of SMYLE to the newly nucleated MTs at the centrosome favored the nucleation and acetylation. Interestingly, SMYLE depletion led to MT nucleation defects, but also a disruption of cortical MT capture. These defects in the MT network were associated with a steep fall in the migratory potential of breast cancer cells and mitotic abnormalities. Our results allow proposing that SMYLE belongs to centrosomal supramolecular complex that favors the assembly and stability of newly nucleated MTs, thus contributing to major processes in tumor development.

La protéine Kinase Haspine comme nouvelle cible thérapeutique : analyse de ses fonctions et caractérisation d'inhibiteurs spécifiques / The protein kinase Haspin as new therapeutic target : analysis of function and characterization of specific inhibitors

Depuis sa découverte en 1994, la protéine kinase Haspine fait l’objet d’un intérêt scientifique croissant en raison de son rôle clé dans la mitose. Elle est impliquée dans localisation et l’activation spatio-temporel d’Aurora B en créant un site d’ancrage (phosphorylation de l’Histone H3 sur la Thr3) sur les chromosomes et notamment aux centromères en première partie de mitose. Une perte d’activité de l’Haspine s’accompagne irrémédiablement d’erreurs dans l'alignement des chromosomes, la cohésion centromérique et l'intégrité des fuseaux mitotiques. Ces fonctions en fait une cible thérapeutique potentielle contre le cancer. Les objectifs de cette thèse ont été de mieux comprendre les fonctions de cette protéine dans la cellule en mitose, et parallèlement, de caractériser de nouveaux inhibiteurs spécifiques de cette kinase. Nous avons montré que l'intégrité des centrosomes et du fuseau mitotique dépend de l'activité kinase de l’Haspine de façon indépendante de l’activité d’Aurora B. De plus, nous montrons que l’Haspine agit comme un régulateur négatif de la nucléation des microtubules aux centrosomes ainsi que sur les chromosomes. Pour mieux comprendre le rôle de Haspine dans la nucléation des microtubules, nous avons cherché de nouveaux substrats à l'aide d'une puce protéique. Nous avons identifié plusieurs candidats parmi lesquels l’effecteur de nucléation, la kinase Nima Nek9. Nous avons confirmé que Nek9 est un substrat de l’Haspine in vitro. De plus, nos résultats ont montré que la déplétion de Nek9 sauve en partie le phénotype de déplétion de l’Haspine, ce qui suggère que l’Haspine a un rôle antagoniste de la fonction centrosomale de Nek9. L’ensemble de nos résultats démontre une nouvelle fonction de l’Haspine de son implication dans la régulation des voies de signalisation de la nucléation des microtubules. En parallèle, nous avons caractérisé une nouvelle série de petites molécules inhibitrices de Haspine, des imidazopyridines dérivées du CHR-6494. Nos composés hits montrent une bonne activité inhibitrice de l’Haspine et une sélectivité accrue. Ils ont l’avantage de ne pas provoquer un arrêt du cycle cellulaire en G2/M comme le CHR-6494 et n’inhibent pas la CDK1. Ils s’avèrent être de précieux outils d’études des fonctions de l’Haspine et une base structurale pour la synthèse d’outils thérapeutiques potentiels. / Since its discovery in 1994, Haspin protein kinase has been of growing scientific interest due to its key role in mitosis. It is involved in spatio-temporal localization and activation of Aurora B kinase by creating a specific anchoring site (phosphorylation of Histone H3 on Thr3) on chromosomes and specifically at centromers during early mitosis. Loss of Haspin activity is irremediably accompanied by chromosome alignment errors, centromeric cohesion and mitotic spindle defects. Its essential mitotic functions make it a potential therapeutic target for cancer. The objectives of this thesis were to better understand the functions of Haspin in mitosis, and at the same time, to characterize new specific inhibitors. We have shown that centrosome and mitotic spindle integrity depends on Haspin kinase activity independently of Aurora B activity. In addition, we show that Haspin acts as a negative regulator microtubule nucleation both at centrosomes and on chromosomes. To better understand Haspin's role in microtubule nucleation we looked for new substrates using protein chips. We have identified several candidates including the Nima kinase nucleation effector, Nek9. We confirmed that Nek9 is an in vitro Haspin substrate. In addition, our results showed that Nek9 depletion partly saves the Haspin depletion phenotype, suggesting that Haspin antagonizes Nek9 nucleation function. All of our results demonstrate a new Haspin function in the regulation of microtubule nucleation signaling pathway. At the same time, we have characterized a new series of small inhibitory molecules of Haspin, imidazopyridines derived from CHR-6494. Our hit compounds showed good Haspin inhibitory activity and increased selectivity. Unlike CHR-6494, they have the advantages of not causing cell cycle arrest in G2/M through CDK1 inhibition. They prove to be valuable tools for Haspin function studies and form a strong structural basis for the development of potential therapeutic drugs.
