An Integrated Model of Eating Disorder Risk and Protective Factors: Implications for Research and Treatment

Body dissatisfaction and disordered eating behaviors are significant public health concerns, especially among college-age women. Despite extensive research, the complex nature of disordered eating and its etiology has prevented a satisfactory predictive model from being developed and validated and has therefore hindered the development of effective prevention and intervention strategies. This study aimed to integrate four of the most common etiological theories into a model to predict disordered eating behaviors. Bivariate correlations and moderation analyses were conducted to determine the strength of relationships between variables and to determine whether a protective factor (i.e., self-compassion) moderates the effects of risk factors (i.e., media exposure, thin-ideal internalization, negative affect) on body dissatisfaction, drive for thinness, and bulimic behaviors. Results indicated that there were correlations between negative affect, thin-ideal internalization, and disordered eating behaviors. Contrary to hypotheses, media exposure had significant relationships with both bulimia and body dissatisfaction but was not significantly associated with thin-ideal internalization. Broadly, negative affect and thin-ideal internalization predicted disordered eating behaviors. Finally, self-compassion did not emerge as a consistent moderator of the relationship between the three risk factors and disordered eating behaviors – except with drive for thinness. Findings have potential implications for mental and physical healthcare interventions to reduce symptom experiences and increase well-being.

Effects of PTEN Loss and Activated KRAS Overexpression on Viscoelasticity, Adhesion, and Mechanosensitivity of Breast Epithelial Cells

Therapeutics targeting the PI3K (phosphatidylinositol 3-kinase) and the Ras/MAPK (mitogen-activated protein kinases) pathways have potential as non-toxic treatments for triple-negative breast cancer due to their frequent over-activation in several forms of cancer. Interestingly, the PI3K and Ras/MAPK pathways have been shown to incite cancer dormancy behavior individually and tumorigenic behavior in unison when induced in healthy breast epithelial cells (MCF-10A) in vivo. Tumorigenesis and metastasis are heavily reliant on the specific mechanical and adhesive properties of cells, including decreased stiffness, increased mechanosensitivity, and decreased adhesion. However, the describe cellular behaviors are poorly understood for dormant cancer phenotypes. Understanding the mechanical and adhesive behaviors of MCF-10A cells as a function of PI3K and/or Ras/MAPK pathway over-activation further explores the cross-talk enabling unique dormant and tumorigenic characteristics. Cellular viscoelasticity and adhesion were measured for MCF-10A cells with PTEN (phosphatase and tensin homolog) knockout and activated KRAS (Kristen rat sarcoma viral oncogene homolog) overexpression to activate the PI3K and Ras/MAPK pathways respectively with atomic force microscopy. PTEN knockout alone has no observable influence on cell adhesion but resulted in softer cells with less organized cytoskeleton. Activated KRAS overexpression increased cell stiffness and cell adhesion regardless of PTEN expression level. Moreover, the overexpression of activated KRAS enhanced the sensitivity of cells to the substrate stiffness. The findings suggest that the cancer-associated pathways PI3K and Ras/MAPK regulate cell adhesion and mechanics to promote tumor formation and metastasis. More importantly, the results that signify mutations of different molecular pathways associated with cancer dormancy regulate cell mechanics differently suggests that cell stiffness is a biomarker that detects and differentiates different types of dormant cancers.

Western countries are facing an increasing poverty among seniors related to the demographic revolution and lengthening of life span. The so-called over-indebtedness is one of the "modern" causes and also consequences of poverty. This thesis discusses the relationship between over-indebtedness and poverty with emphasis on senior population (65+). An empiric analysis is employed to explore causes and consequences of over-indebtedness. The increasing over-indebtedness is already being targeted by public policies, however their effectiveness is low. Sufficient material and personal resources are lacking as well as not being optimally utilized. There is only limited cooperation between civil and public-law sectors. Communication with the private sector in a way of common goal setting is also deficient. In a fierce market-driven economy, elderly people become a weak link and find themselves in situations conflicting with morals, human and civil rights, and adequate quality of life in a modern society. This state of affairs threatens social integrity and demands a solution. Three participating groups were surveyed: the elderly, the employees of advisory centers and the non- bank financial institutions. Various approaches to solving the problem are discussed.
