The investigation is theoretically grounded in the Phenomenological Sociology (Alfred Sch?tz), the Sociology of Knowledge (Peter Berger, Thomas Luckmann), and the Sociology of Processes and Figurations (Norbert Elias). The biographical narrative interview method of the German tradition (Frits Sch?tze, Gabriele Rosenthal) is used in the reconstruction of an empirical case, which encompasses the analysis of a Haitian migrant?s life and of its family in Haiti, Ecuador, Peru and Brazil. Therefore empirically grounded knowledge on the constitution of feelings of belonging in the life trajectory of a Haitian migrant in Brazil is possible. Based on the analysis of the empirical case, a few remarks that put in question traditionally used categories in the migration field of studies arise from the case reconstruction. The research points towards the existence of a type of return migrant that is uncommon in migration studies, for his return is planned taking into account the country for which the migrant developed his sense of belonging. These feelings are expressed through his will to become president of his nation in order to help the people of his homeland. This type of migrant has traits of national leadership, whose sense of belonging was forged through its displacement across international frontiers. / O trabalho consiste em uma investiga??o a respeito da constru??o do pertencimento na vida de um migrante. A pesquisa fundamenta-se teoricamente na tradi??o da Sociologia de orienta??o fenomenol?gica (Alfred Sch?tz), da Sociologia do conhecimento (Peter Berger, Thomas Luckmann) e da Sociologia dos processos e figura??es (Norbert Elias). O m?todo de entrevista narrativa biogr?fica de matriz alem? (Fritz Sch?tze e Gabriele Rosenthal) ? utilizado para a reconstru??o de um caso emp?rico, que compreende uma an?lise da vida de um migrante haitiano e de sua fam?lia no Haiti, Equador, Peru e Brasil. Com isso, procura-se oferecer conhecimento empiricamente fundamentado a respeito da constitui??o dos sentimentos de perten?a na trajet?ria de vida de um imigrante haitiano no Brasil, a partir do qual ? proposto um questionamento de categorias difundidas nos estudos correntes sobre a migra??o. As a first issue, it aspired to understand the way the TV is thought by those who produce it daily. Electing to field the backstage of A Praça é Nossa, the oldest of the Brazilian comedy TV still on display, the search ends up embarking on other reflections, in which the realized ethnography is used to support the structural analysis of comedic sketches presented by the program. / O trabalho que ora se apresenta tinha como proposta original fazer uma etnografia da produção do entretenimento televisivo. Como problema primeiro, aspirava-se compreender o modo pelo qual a Televisão é pensada por aqueles que cotidianamente a produzem. Elegendo como campo os bastidores de A Praça é Nossa, o mais antigo humorístico da TV Brasileira ainda em exibição, a pesquisa acaba enveredando por reflexões outras, nas quais a etnografia realizada é utilizada como suporte da análise estrutural dos sketches cômicos apresentados pelo programa.

It has been found that a decrease in their physiological levels may be indicative of several diseases, standing out as an important biomarker. Several techniques are used to detect glutathione, as in the case of liquid chromatography. However, the electrochemical sensors method has been of increasing interest. Recently, the contribution of nanomaterials in the manufacture of these devices has been translated into an increase in the reproducibility and reach of low detection limits, highlighting themselves as promising tools for clinical diagnosis. In this work, two different sensors based on nanomaterials were developed for the detection of the GSH. The first sensor was composed of a gold electrode modified with a film based on the metal-organic framework copper (II) -benzene-1,3,5-tricarboxylate (CuBTC) and the acrylamide sodium acrylate copolymer (CuBTC/EAu). Electrochemical measurements were performed using the cyclic voltammetry method in an electrochemical cell, using silver/silver chloride (Ag/AgCl(KClsat)) as a reference electrode, platinum wire as an auxiliary electrode, and the CuBTC/EAu sensor as the working electrode. According to the results, the developed sensor exhibited a good electrocatalytic activity for the glutathione oxidation, in a potential of 0,41 V, with a low detection limit (0,03 μmol L-1), and a linear range operating range of 0,05 – 2 μmol L-1 of GSH and a response time of 15 minutes. The second sensor deals with the electrosynthesis of a nanocomposite based on CuBTC and gold nanoparticles stabilized with PVP, which was denominated Au@CuBTC. The morphological and structural characteristics of the new compound were investigated by scanning electron microscopy (SEM), thermal gravimetric analysis (TGA), diffraction Xray powder (XRPD) and absorption spectroscopy in the infrared region (FTIR) and its application as an electrochemical sensor for GSH quantification was evaluated by square wave voltammetry. In accordance with the results, it was observed that the use of nanoparticles in the structure of the metal-organic framework significantly increased the sensitivity and the response time of the electrode. The analytical curve of the sensor showed a linear response range between 10 - 40 μmol L-1 of GSH, and a detection limit of 0,3 μmol L-1, with response time of 60 seconds. The proposed sensors are presented as an innovative technology and open paths for applications of metal organic frameworks in electrocatalysis and the construction of attractive sensors for the detection of glutathione in biological fluids. / A glutationa (GSH) é o principal tiol de baixo peso molecular encontrado nas células humanas, cuja função está relacionada à biotransformação e eliminação de xenobióticos e a defesa das células contra o estresse oxidativo. Tem sido estabelecido que uma redução nos seus níveis fisiológicos pode ser indicativa de diversas doenças, destacando-se como um importante biomarcador. Várias técnicas são utilizadas para detecção da gluatationa, a exemplo da cromatografia líquida, entretanto, o método dos sensores eletroquímicos tem despertado interesse crescente. Recentemente, a contribuição de nanomateriais na fabricação desses dispositivos, tem se traduzido no aumento na reprodutibilidade e alcance de baixos limites de detecção, destacando-se como ferramentas promissoras para diagnóstico clínico. Neste trabalho foram desenvolvidos dois sensores a base de nanomaterias para a detecção de GSH. O primeiro sensor foi constituído por um eletrodo de ouro modificado com um filme baseado na rede metal-orgânica cobre(II)–benzeno-1,3,5–tricarboxilato (CuBTC) e o copolímero acrilamida acrilato de sódio (CuBTC/EAu). As medidas eletroquímicas foram realizadas pelo método da voltametria cíclica numa célula eletroquímica, utilizando-se como eletrodo de referência prata/cloreto de prata (Ag/AgCl(KClsat)), um eletrodo auxiliar de fio de platina e como eletrodo de trabalho o sensor CuBTC/EAu. De acordo com os resultados, o sensor desenvolvido obteve uma boa atividade eletrocatalítica para a oxidação da glutationa, no potencial de 0,41 V, com baixo limite de detecção (0,03 μmol L-1), faixa de trabalho linear de 0,05 - 2 μmol L-1 de GSH e um tempo de resposta de 15 minutos. O segundo sensor aborda a eletrossíntese de um nanocompósito baseado no CuBTC e nanopartículas de ouro estabilizadas com PVP, o qual foi denominado Au@CuBTC. As características morfológicas e estruturais do novo composto foram investigadas por microscopia eletrônica de varredura (MEV), análise termogravimétrica (TGA), difração de raios-X de pó (XRPD) e espectroscopia de absorção na região do infravermelho com transformada de Fourier (FTIR) e sua aplicação como sensor eletroquímico para quantificação da GSH foi avaliada pela voltametria de onda quadrada. De acordo com os resultados obtidos, foi observado que o uso das nanopartículas na estrutura da rede metal-orgânica aumentou significativamente a sensibilidade e o tempo de resposta do eletrodo. A curva analítica do sensor mostrou uma faixa de resposta linear entre 10 - 40 μmol L-1 de GSH e um limite de detecção de 0,3 μmol L-1, com tempo de resposta de 60 segundos. Os sensores propostos apresentam-se como tecnologias inovadoras e abre caminhos para aplicações das redes metal orgânicas na eletrocatálise e na construção de sensores atrativos para a detecção de glutationa em fluidos biológicos.

Existing regeneration protocols are limited to a few responsive varieties, such as Coker, commonly used as recipients during genetic transformation. One of the factors that may be related to recalcitrance is the inefficiency in the activation of different genes during in vitro culture that may be associated with the acquisition of embryogenic competence. Among the genes mentioned in the literature, the Somatic Embryogenesis Receptor-Like Kinase (SERK) is shown to be responsible for the transition from the somatic cell state to the embryogenic in different plant species. The potential of this gene as a marker of somatic embryogenesis has been investigated. In this work, the relationship of the levels of GhSERK1 expression with the acquisition of embryogenic competence in cotton genotypes was analyzed. For this, a short sequence (186 bp) of this gene was selected from the conserved region between cotton and five other dicotyledons (Theobroma cacao, Citrus sinensis, Vitis vinifera, Glycine max and Phaseolus vulgaris) in order to allow the probe can be applied in the selection of other cultures. Initially, the RT-qPCR gene expression tests were performed in six genotypes with known embryogenic capacity (embryogenic - Coker 312, BRS Rubi and BRS Seridó and non-embryogenic - BRS 201, CNPA Precoce 1 and BRS Topázio), from RNA extracted from meristematic zygotic tissues. The aforementioned genotypes were induced to somatic embryogenesis in medium supplemented with naphthaleneacetic acid and kinetin. Corroborating with the result obtained via RT-qPCR, somatic embryos were formed only in Coker 312, BRS Seridó and BRS Rubi. In the latter, the percentage of fertile plants obtained was higher (50%) than Coker 312 (41.6%). The selective applicability of GhSERK1 was validated in four genotypes top lines (CNPA 286, CNPA BA 139, CNPA BA 1366 and CNPA BA 2247) with unknown in vitro behavior. Simultaneously, we performed induction tests on somatic embryogenesis to verify the expression results. In the genotypes in which the relative expression was higher (Coker 312 and CNPA BA 139), somatic embryos were formed. A nonradioactive probe was then grown from the 186 bp fragment and attested for its efficiency via Southern Blot in the top line genotypes, robust spots were observed in Coker 312 and CNPA BA 139. Based on these results, the latter was selected as the recipient genotype in future transformation studies. In addition, the sequence used allowed to identify genotypes responsive to somatic embryogenesis from zygotic tissues, optimizing time and costs. This work provides collaborations for the understanding of factors that may be involved in the acquisition of embryogenic competence generating perspectives of new studies to strengthen the programs of genetic improvement of the culture. / O lançamento de cultivares transgênicas de espécies recalcitrantes como o algodoeiro pode ser retardado devido à dificuldade em regenerar o tecido transformado. Os protocolos de regeneração existentes limitam-se a poucas variedades responsivas, como a Coker, normalmente utilizadas como receptoras durante a transformação genética. Um dos fatores que podem estar ligados à recalcitrância, é a ineficiência na ativação de diferentes genes durante o cultivo in vitro que podem estar associados à aquisição de competência embriogênica. Dentre os genes citados na literatura, destaca-se o Somatic Embryogenesis Receptor-Like Kinase (SERK) retratado como responsável pela transição do estado celular somático para embriogênico em diferentes espécies de plantas. A potencialidade deste gene como marcador de embriogênese somática tem sido investigada. Neste trabalho, foram analisadas a relação dos níveis de expressão de GhSERK1 com a aquisição de competência embriogênica em genótipos de algodoeiro. Para isto, uma sequência curta (186 pb) deste gene foi selecionada a partir de região conservada entre o algodão e cinco outras dicotiledôneas (Theobroma cacao, Citrus sinensis, Vitis vinifera, Glycine max e Phaseolus vulgaris) de modo a permitir que a sonda também possa ser aplicada na seleção de outras culturas. Inicialmente, os testes de expressão gênica via RT-qPCR foram realizados em seis genótipos com capacidade embriogênica conhecida (embriogênicos – Coker 312, BRS Rubi e BRS Seridó e não embriogênicos – BRS 201, CNPA Precoce 1 e BRS Topázio), a partir de RNA extraído de tecidos zigóticos meristemáticos. Os genótipos supracitados foram induzidos à embriogênese somática em meio suplementado com ácido naftalenoacético e kinetina. Corroborando com o resultado obtido via RT-qPCR, embriões somáticos formaram-se apenas na Coker 312, BRS Seridó e BRS Rubi. Nesta última, o percentual de plantas férteis obtidas foi maior (50%) do que o Coker 312 (41,6%). A aplicabilidade seletiva do GhSERK1 foi validada em quatro genótipos top lines (CNPA 286, CNPA BA 139, CNPA BA 1366 e CNPA BA 2247) com comportamento in vitro desconhecido. Simultaneamente, realizamos ensaios de indução à embriogênese somática para comprovar os resultados de expressão. Nos genótipos em que a expressão relativa foi maior (Coker 312 e CNPA BA 139) houve a formação de embriões somáticos. Uma sonda não radioativa foi então desenvolvida a partir do fragmento de 186 pb e atestada sua eficiência via Southern Blot nos genótipos top lines, manchas robustas foram observadas em Coker 312 e CNPA BA 139. Com base nestes resultados, este último foi selecionado como genótipo receptor em futuros estudos de transformação. Além disso, a sequência utilizada permitiu identificar os genótipos responsivos à embriogênese somática a partir de tecidos zigóticos, otimizando o tempo e os custos. Este trabalho fornece colaborações para a compreensão de fatores que podem estar envolvidos na aquisição de competência embriogênica gerando perspectivas de novos estudos para fortalecer os programas de melhoramento genético da cultura.

Urban sociology has advanced in the studies on urban public spaces between the environment of practices, behaviors, cultures and policies. Thus, urban space presents itself as a relation of power of socio-cultural dynamics between the values and meanings of public life. Understanding these cultural phenomena that emerge in the spaces of the cities are fundamental to advance in ethnographic data that exploring the cultures between and through the places where the activities and the values prospect insurgencies in the urban spaces. In order to explain the formation of the cultural space in the city, it is necessary to analyze the urban space of the city, with its uses and counter-uses, to present the significant processes of influence of globalization in the planning of urban public spaces. The urban experience by means of the skateboarding in the spaces of the cities demands to find its rhythm on the conception between the public and the private, its urban structure put in tension to its space of use. In this approach, skateboarding reveals contemporary urban experience as a continuous process of contesting watertight social values, the democratization of urban public space, the destruction of value systems and the continuous formation of plural itineraries, whether as leisure, communication or social interaction . This urban experience reveals new ways for the social sciences to reflect critically on city spaces and their continuous practices, identifying the formative processes present in contemporary urban cultures and locating the genesis of spaces in cities. / A cidade e o espa?o urbano contempor?neo est?o sofrendo significativas influ?ncias de novas rela??es culturais. E em especial, analisar suas rela??es sociais com funcion?rios, pacientes, familiares e refletir sobre a import?ncia do Estado laico para a pluralidade de grupos religiosos nos hospitais. Kongo and Angola. For this, we seek to situate Bantu culture in the context of restructuring processes of the African religiosity in Brazil, as well as in the scenario of social changes that influence the construction of identities. The first chapter introduces the peoples of Africa, Bantu and Sudanese, through historical data and sociological, about the time of slavery, from cultural and religious. The next chapter deals with the "candombl?" as a research object, from the point of view of researchers who assess methodologically the experiences of initiation into the cult, and my positioning, as person divinized, and also a researcher. The third section shows some nuances that give authenticity to the cult of matrix Kongo and Angola, comparing them with the cult to "Orix?s". In addition, describe, briefly, the main deities in ?candombl?? Bantu and "nag?". The fourth and fifth chapters describe the exercises for field in the state of S?o Paulo. A narrative brings the participation of the researcher in an international meeting to discuss the identity of the ?candombl?? Angola; another shows a schematic of the ethnographic experience of identity within a temple of Bantu "candombl?". Both activities provide reflections and comparison of data. The sixth chapter is a review of the literature on the notions of identity developed by contemporary theorists in the Social Sciences, even though the concept discussed across disciplines. In addition, references on the movements of Africanization of the "candombl?" reinforce the employment of a perspective anthropological research on the topic. Finally, the last two chapters are devoted to the examination of the empirical exercises, where they sought to highlight the importance of Bantu culture and religious identity, from which they were extracted some considerations which point to the need for continuity of research on other aspects that comprise the African religions. / Este estudo tem como objetivo analisar a viv?ncia de identidades do ?povo do santo?, com foco em experi?ncias no candombl? de matriz Kongo e Angola. Para isto, busca situar a cultura banta no contexto dos processos de reestrutura??o da religiosidade africana no Brasil, bem como no cen?rio de mudan?as sociais, que influenciam a constru??o de identidades. O primeiro cap?tulo apresenta os povos origin?rios africanos, bantos e sudaneses, atrav?s de dados hist?ricos e sociol?gicos sobre a ?poca da escravid?o, a partir de aspectos culturais e religiosos. O cap?tulo seguinte trata do candombl? como objeto de pesquisa, sob o ponto de vista de pesquisadores que avaliam metodologicamente as experi?ncias de inicia??o no culto, e de meu posicionamento, como iniciado e, tamb?m, pesquisador. Efeito da estrutura da paisagem sobre a riqueza e Diversidade de árvores em grupos funcionais reprodutivos

Estudos recentes tem documentado diminuicao na riqueza e abundancia de especies arboreas em determinados grupos funcionais. Estes estudos investigaram a influencia dos efeitos de borda e algumas metricas dos fragmentos sobre alguns grupos funcionais. No entanto, nao ha estudos na literatura que avaliaram como a estrutura da paisagem afeta diretamente diferentes grupos funcionais reprodutivos, que e o objetivo deste estudo. Este estudo foi realizado em oito paisagens (buffer de 1000 m de raio) ao redor de fragmentos florestais. Para cada paisagem nos calculamos a area do fragmento, conectividade, o percentual de cobertura florestal e percentagens de matrizes de pasto, cafe e cana de acucar. Todas as especies (195) e individuos (2031) com DAP . 3 cm amostrados nos fragmentos foram classificados em grupos funcionais reprodutivos (GFRs), dentro dos 8 agrupamentos funcionais. A influencia dos parametros estruturais da paisagem sobre a diversidade de agrupamentos funcionais e sobre a riqueza de especies e individuos nos GFRs foi verificada por Modelos Lineares Generalizados. A qualidade destes modelos foi avaliada atraves de valores de Akaike corrigidos. A diversdade de agrupamentos funcionais e a riqueza de individuos nos GFRs foram negativamente influenciadas pela porcentagem de matriz de pasto e cana matriz e positivamente pela area do fragmento, conectividade e porcentagem de matriz de cafe. Para a manutencao de todos os grupos funcionais reprodutivos originais em paisagens fragmentadas a combinacao destes parametros da paisagem deve ser considerada na conservacao e nas politica de gestao. / Recent studies have documented decreased richness and abundance of tree species from particular functional groups. These studies investigated the influence of edge effects and some within-patches aspects. However, there are no studies in literature that evaluated how landscape structure affects directly different reproductive functional groups, which is the aim of this study. This study was carried out in 8 landscapes (1000m buffer radius) centered in a forest fragment. For each landscape we calculate the area, connectivity, percent of forest cover and percentages of coffee, pasture and sugarcane matrices. All species (195) and individuals (2031) with DBH ≥3 cm sampled in fragments were classified in reproductive functional groups (RFGs), within functional associations. The influence of landscape structural parameters on diversity of functional association was verified by Generalized Linear Models. These models quality was assessed by Akaike corrected values. Functional associations and RFGs were negative influenced by percentage of pasture and sugarcane matrix and positive influenced by fragment area, connectivity and percentage of coffee matrix.
