Para a identificação da data de término do teste foram realizadas contagens diárias do número de plântulas normais e para cada lote, tratamento de superação de dormência e temperatura, foi ajustada uma curva de crescimento para a avaliação da germinação. As sementes remanescentes do teste de germinação foram seccionadas longitudinalmente e medianamente através do embrião para a realização do teste de tetrazólio. As duas metades da semente foram imersas em uma solução de tetrazólio a 0,1% e mantidas em câmara escura, à 37ºC, por um período de 3 horas e após esse período as sementes foram lavadas e a leitura feita imediatamente, classificando-se as sementes em viáveis e não viáveis (mortas). In that contest, this study aimed to evaluation the morphological composition, structural characteristics and nutritional value of the Panicum maximum cv. Massai forage, cultivated under urea sources (common and coated Policote?) and N doses (200, 400 e 600 kg ha-1 year-1), during autumn, winter and spring 2014, summer and autumn 2015 seasons. The experiment was conducted in UFRRJ, Serop?dica/RJ, under a complete block randomized design with four replications in a factorial arrangement (3x2) + 1. The variance analysis of data was performed using the PROC MIXED of the SAS? (9.2 version), a 5% probability. The means of treatments were compared using PDIFF (p<0.05), and the quantitative effects using a PROC REG of SAS? (probability<0.05). There was interaction (p<0.05) between doses, sources of urea, and seasons for the forage mass (FM), percentages of leaf blades (LBDMP), stem (SDMP) and dead material (DMDMP) dry matter, leaves blades: steams ratio (LBSR), forage accumulation rate (FAR), nitrogen use efficiency (NUE), and for dry matter (DM) and crude protein (CP) content of the forage. Since the canopy height (CH), tiller population density (TPD), weight per tiller (WT) and forage bulk density (FBD), mineral matter (MM) and insoluble neutral detergent fiber (NDF) contents of the forage varied (p<0.05) due to N rates and seasons interaction. There was a linear increased in the FM with increasing of N doses until to the 600 kg ha-1 yr-1 level in the winter and spring seasons by both urea sources. The increasing N doses promoted higher LBDMP and lower SDMP and DMDMP of the FM, besides providing increased TPD, FBD and FAR of the massai grass forage, during all seasons studied. There was a quadratic positive effect for NUE during autumn 1, spring, summer and autumn 2 for common and coated urea, with higher maximum points for coated urea. And during the winter there was a positive linear effect only for coated urea. Higher average values of the CP were verified on the spring (11.76%), and lower on the autumn/winter (9.15%), and the use of coated urea providing higher CP levels of the forage than common urea in all seasons (10.40% and 9.41% general average, respectively). The CP content increased linearly with the increase of N levels in all seasons, while the NDF reduced linearly with increasing of these levels, except for the fall season 2 (there was not effect for N dose - p>0.05). Intensification of nitrogen fertilization favors the leaf blades participation in the forage mass, benefiting the structural characteristics, the forage accumulation, and the nutritional value of Massai grass. Coated urea promotes greater NUE during all seasons of the year. / A busca pela melhor efici?ncia da aduba??o nitrogenada justifica a necessidade da avalia??o de fontes e doses de nitrog?nio (N) que promovam menores perdas para o ambiente, al?m de maior produtividade e qualidade da forragem produzida. Nesse contexto, objetivou-se avaliar a composi??o morfol?gica, as caracter?sticas estruturais e o valor nutricional da forragem do Panicum maximum cv. Massai, cultivado sob fontes de ureia (comum e revestida por Policote?) e doses de N (200, 400 e 600 kg ha-1 ano-1), durante as esta??es de outono, inverno e primavera de 2014, ver?o e outono de 2015. Houve efeito quadr?tico positivo para EUN durante o outono 1, primavera, ver?o e outono 2 para ureia comum e revestida, com maiores pontos de m?ximo para ureia revestida, e durante o inverno houve efeito linear positivo somente para ureia revestida. Maior valor m?dio de PB foi verificado na primavera (11,76%), e menor no outono/inverno (9,15%), e o uso da ureia revestida promoveu maiores teores de PB na forragem que a ureia comum em todas as esta??es (m?dias gerais de 10,40% e de 9,41%, respectivamente). O teor PB aumentou linearmente com o incremento de doses de N em todas as esta??es, enquanto que os teores de FDN reduziram linearmente com aumento dessas doses, exceto para a esta??o outono 2 (n?o houve efeito de dose de N ? p>0,05). Responses of C3 and C4 Panicum grasses to CO2 enrichment

This project aims at investigating the effect of CO2 enrichment on the growth and gas exchange of C3, C3-C4 and C4 Panicum grasses. Potted plants were grown in soil under well watered conditions, in artificially lit environmentally controlled cabinets or naturally lit greenhouses at varying levels of CO2 enrichment. CO2 enrichment enhanced the dry weight of C3 and C4 Panicum species under optimal light and N supplies, but had no effect on the total leaf N or TNC concentrations. The high-CO2 induced photosynthetic reaction in the C3 species was accompanied by a reduced Rubisco concentration and was related to the conservation of the relative growth rate of the plant. Elevated CO2 had no effect on the photosynthetic capacity of the C4 species, but enhanced its CO2 assimilation rates under high light and N supplies. The effect of elevated CO2 on the leaf and stem anatomy reflected increased carbon supply at high CO2 in the C3 grass, and reduced transpiratory demand at high CO2 in C4 grasses. Consequently, it is clear that both C3 and C4 grasses are likely to be more productive under rising atmospheric CO2 concentrations. / Doctor of Philosophy (PhD)

Virus vector gene inserts are stabilized in the presence of satellite panicum mosaic virus coat protein

The coat protein of satellite panicum mosaic virus (SPMV) was used to stabilize viral vector gene inserts in planta. A Potato virus X (PVX) vector carrying the SPMV capsid protein (CP) gene was successfully stabilized through three serial passages in Nicotiana benthamiana from the upper non-inoculated leaves following rub inoculation. The presence of SPMV CP expression from the PVX vector was confirmed by necrotic lesions that occur only when SPMV CP is present and by western blot and reversetranscription PCR analyses. In addition, PVX-SPCP was co-inoculated onto N. benthamiana with a Tomato bushy stunt virus vector carrying a green fluorescent protein gene, which normally does not yield GFP expression in upper tissue due to loss of the insert. However, upon co-inoculation with PVX-SPCP, upper non-inoculated leaves exhibited GFP accumulation based on green fluorescence by UV illumination at 488 nm and western blot analysis. GFP expression was more abundant in upper non-inoculated N. benthamiana leaves as well as systemic tissues when the co-inoculation experiments were performed at 20°C compared to 25°C. These results suggest that SPMV CP is a viable molecular tool for stabilizing viral vector gene inserts in planta.

Evaluation of switchgrass as an energy feedstock : economic feasibility, and carbon dioxide accounting

Energy availability and environmental issues are of growing concern; nations are striving to use energy more efficiently while at the same time decreasing the negative impacts on the environment. / The objectives of this study are to: (1) establish a supply price for ethanol derived from switchgrass, (2) establish an accounting budget for carbon dioxide during feedstock production and processing into ethanol, and (3) determine the cost/tonne of CO$ sb2$ using this strategy. / Total cost of production of the feedstock for cycles of 5, 10, 15, and 20 years are $357.06/ha ( $32.73/ODT), $337.81/ha ( $30.96/ODT), $331.52/ha ( $30.39/ODT), and $328.47/ha ( $30.11/ODT) respectively. Thus, the approximate cost of producing one litre of ethanol from switchgrass is $0.47/litre. / Overall, the switchgrass-ethanol system is a net carbon sink for all four cycles, and each hectare of swithgrass sequesters between 1.9 and 6.8 tonne of CO$ sb2$ per year. However, this process is not indefinite, and will stop once the soil organic matter reaches an equilibrium. / The current price for gas is $0.24/litre (excluding taxes and profit margins). According to the price differential existing between ethanol and gasoline and the CO$ sb2$ emissions difference between both systems, the cost of sequestering CO$ sb2$ ranges between $83/tonne and $129/tonne, with the adoption of this specific strategy. At the macro level, Quebec and Canada's total carbon dioxide emissions reached 70 million tonnes and 461 million tonnes respectively, in 1991. Thus, displacing fossil-based energy sources (gasoline) with a renewable energy source (ethanol from switchgrass) to reduce those emissions by 20 % by the year 2010, incurs a cost ranging between $1.16 billion and $1.8 billion for Quebec, and between $7.7 billion and $11.9 billion for Canada. SMALL MAMMAL POPULATIONS IN SWITCHGRASS STANDS MANAGED FOR BIOMASS PRODUCTION COMPARED TO HAY AND CORN FIELDS IN KENTUCKY

Switchgrass (Panicum virgatum), a native warm-season grass, has been investigated as a renewable energy crop that may provide viable wildlife habitat. This study investigated small mammal populations in switchgrass, hay, and corn to assess the relative habitat quality. Four, three-night trapping sessions were conducted at four locations in Kentucky using Sherman livetraps. Trapping occurred in spring (before first hay harvest), summer, fall (before switchgrass and corn harvest), and winter (post-harvest). Relative abundance of small mammals, calculated using a capture per unit effort index (per 100 trapnights), and mean taxonomic richness were used to compare habitats. Switchgrass had a significantly greater mean taxonomic richness than hay but not corn; however, four genera were captured in switchgrass and only two in corn. Switchgrass had a greater relative abundance of small mammals than hay during the summer, and corn and hay during the fall. Vegetative cover was positively correlated with relative abundance of small mammals. No-till corn and three year old switchgrass had a greater relative abundance of small mammals than conventionally tilled corn and two year old switchgrass, respectively. In conclusion, switchgrass stands managed as a renewable energy crop has the potential to be viable wildlife habitat for some small mammal species.

Carbon storage in switchgrass (Panicum virgatum L.) and short-rotation willow (Salix alba x glatfelteri L.) plantations in southwestern Québec

Carbon storage was compared between two perennial biomass energy systems, namely switchgrass (Panicum virgatum L.) and short-rotation willow Salix alba x glaffelteri L.) at 2 adjacent sites, and further compared with a corn cultivation, a 20-year-old abandoned field, and a mature hardwood forest, in southwestern Quebec. Aboveground carbon results indicated that switchgrass and corn had significantly greater carbon levels than willow at the less fertile site, but no significant differences were detected at the more fertile site. Root carbon results indicated that corn had significantly lower carbon levels than both perennial systems to a depth of 30 cm at both sites. However, switchgrass had significantly greater root carbon levels beyond 30 cm compared with willow and corn, and beyond 45 cm compared with the forest and abandoned field. These findings indicate that deep-rooted perennial grasses such as switchgrass have the potential to sequester carbon at deeper soil layers. Soil carbon results showed that at the more fertile site, willow was associated with significantly greater soil carbon levels than switchgrass. Moreover, both perennial crops had soil carbon levels that were greater than for corn, the abandoned field, and the forest. In contrast, at the less fertile site, no significant differences in soil carbon were detected between the various plant systems examined. The results of this study suggest that the perennial energy crops used, when grown on relatively fertile soils, have the potential to substantially increase soil carbon levels compared with conventional agricultural and/or forest systems. Consequently, when these crops are grown on less fertile soils, their added advantage of increasing carbon storage is lost.

O delineamento experimental foi inteiramente casualizado, com quatro repeti??es, sendo o per?odo experimental de 90 dias, em ambos os experimentos. Determinou-se a massa seca e os teores de Cd na parte a?rea, colmo e ra?zes das forrageiras. Os conte?dos de Cd foram calculados com base nos teores e nas produ??es de massa seca em cada parte da planta. No caso do experimento em solo, para avalia??o do teor de Cd foram utilizados os extratores Mehlich-1, DTPA pH 7,3, teor semitotal - USEPA 3051 e teor total - USEPA 3052. O crescimento relativo das forrageiras foi reduzido pelas doses de Cd, tanto na solu??o nutritiva quanto no solo, configurando a fitotoxidade do Cd nessas plantas. Por?m a sequ?ncia de suscetibilidade foi diferenciada para os experimentos, sendo a ordem decrescente de suscetibilidade na solu??o nutritiva a seguinte: Aruana > Tanz?nia > Xara?s > Basilisk > Marandu, enquanto no solo a sequ?ncia foi: Marandu > Xara?s > Aruana > Tanz?nia > Basilisk. Essa diferen?a de suscetibilidade ? atribu?da ?s diferentes intera??es das forrageiras com o solo. Em ambos os experimentos o teor de Cd nas forrageiras aumentou em fun??o das doses Cd. Por?m a presen?a de Cd na solu??o nutritiva fez com que o ac?mulo e ?ndice de transloca??o apresentassem resposta diferenciada para as forrageiras. Os extratores Mehlich-1 e DTPA foram eficientes para expressar o crescimento da planta. De toda forma, as forrageiras foram incapazes de limitar a absor??o e transloca??o de Cd, acarretando em toxicidade e redu??o de crescimento e altos teores de Cd em todas as partes da planta, n?o proporcionando barreira ? entrada do Cd na cadeia alimentar. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Produ??o Vegetal, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2012. / ABSTRACT This work was carried out in order to evaluate the forage grasses tolerance and the absorption, accumulation and bioavailability of cadmium (Cd) on soil. The experiments were settled under conditions of greenhouse on the Department of Agronomy of the College of Agricultural Sciences of the Universidade Federal dos Vales do Jequitinhonha e Mucuri, Diamantina (MG), Brazil. The forages grasses evaluated were: Panicum maximum cv. Aruana and cv. Tanz?nia and Brachiaria decumbens cv. Basilisk, Brachiaria brizantha cv. Xara?s and cv. Marandu and the four doses of Cd were: 0, 5, 10, and 20 mg l-1 and the nutrients in solution 0, 2, 4 and 12 mg kg-1 for soil, respectively. The experimental design was completely randomized with four replications, and the trial period of 90 days, in both experiments. The dry weight and concentration of Cd in shoots, stems and roots of forages were determined. The Cd content had been calculated based on the dry matter yield and content in each part of the plant. In the soil experiment, to measure the levels of Cd, there had been used the extractors Mehlich-1, DTPA pH 7.3, semi total content - USEPA 3051 and total content - USEPA 3052. The relative growth of the forage grasses was reduced with doses of Cd applied, both in the nutrient solution and on the soil, setting the phytotoxicity of Cd in these plants. However the sequence of susceptibility was different t tohe experiments, the decreasing order of susceptibility in the nutrient solution was: Aruana> Tanzania> Xara?s> Basilisk > Marandu, while in soil the sequence was: Marandu> Xara?s> Aruana> Tanzania> Basilisk. This difference in susceptibility is attributed to the different interactions of forage with soil. In both experiments (soil and nutrient solution) the Cd content in the forage increased due of increasing doses of Cd. Nevertheless the presence of Cd in the nutritive solution has made the accumulation and translocation index differently for the forages. Mehlich-1 and DTPA showed high positive correlation with relative growth of forages. The forages were unable to limit the absorption and translocation of Cd, resulting in toxicity and declining growth and high levels of Cd in all parts of the plant, providing no barrier to entry of Cd into the food chain.

As forrageiras estudadas reduziram o crescimento com aumento das doses de Pb aplicadas, tanto na solu??o nutritiva quanto no solo, configurando a fitotoxidade do Pb nessas plantas. Por?m, a suscetibilidade foi diferenciada entre os experimentos, sendo que, na solu??o nutritiva, as cultivares Marandu e Basilisk foram mais promissoras para a fitorremedia??o, devido a menor influ?ncia do Pb em seus crescimentos relativos, enquanto no solo a cv. The objective of this project was to evaluate in forage grasses the absorption, accumulation and tolerance to lead (Pb) in dystrophic Red-Yellow Latosol soil and nutrient solution, and to evaluate the availability of Pb through extraction methods Mehlich 1, DTPA pH 7.3, USEPA 3051 and 3052 in samples soil. The experiments were settled under conditions of a greenhouse in the Department of Agronomy of the College of Agricultural Sciences of the Universidade Federal dos Vales do Jequitinhonha e Mucuri, (UFVJM), Diamantina (MG). The forages Panicum maximum cv. Aruana and cv. Tanz?nia and Brachiaria decumbens cv. Basilisk, Brachiaria brizantha cv. Xara?s and cv. Marandu and four rates of Pb in nutrient solution of 0, 40, 120, and 360 mg L-1 and in soil of 0, 45, 90 e 270 mg kg-1, have been evaluated. The dry weight and concentration of Pb in the shoot, in the stem base and roots of forages were assessed. The contents of Pb were calculated based on the amounts and on the production of dry mass in each part of the plant. In the case of the soil experiment, to measure the levels of Pb were used Mehlich 1, DTPA pH 7,3 for available content; semi total content - USEPA 3051 and total content - USEPA 3052. The forages studied reduced growth when increasing rates of Pb were applied, both in the nutrient solution and on the soil, setting the phytotoxicity of Pb in these plants. However the susceptibility was different for the experiments, because in the nutrient solution, the Marandu and Basilisk forages proved to be promising for phytoremediation because of the lowering influence of Pb for the last one mentioned and the higher accumulation of metal for the first one, while on the soil the forage Basilisk did better than the other, proving to be more tolerant to the toxic element. In both experiments (soil and nutrient solution) the Pb content in the forage increased due to increasing rates of Pb. However, the presence of Pb in the cultivation conditions has made the accumulation and translocation index in different parts of the plants to respond differently to the forages. There was not correlation between the forage growth with the admeasurement of Pb in soil by extraction methods DTPA pH 7,3 e Mehlich 1, proving the inefficiency of the same for the phytoavailability of Pb.
