Dentition is a key vertebrate innovation showing not only great morphological diversity, but also different maintenance or replacement of functional teeth. Most extant vertebrates replace their dentition through addition of new teeth from deeply invaginated epithelium, i.e. the successional dental lamina, due to presence of dental stem cells. However, in some early branching lineages of ray-finned fishes (Actinopterygii), new tooth germs arise from the adjacent superficial epithelium without the presence of the successional dental lamina. Whether the two types of dental development in vertebrates are equivalent and whether comparable dental stem cells play role in tooth replacement is currently not satisfactorily evaluated. This Master thesis aims at describing the development of palatal and pharyngeal dentition of a member of an early branching lineage of ray-finned fishes, the sterlet sturgeon (Acipenser ruthenus). The sterlet dentition is fairly dynamic. The teeth are replaced without the successional dental lamina, however, this replacement shows characteristics similar to those described in vertebrates with the successional dental lamina. Phylum Tardigrada

A sister group of the Arthropoda, the Tardigrada are micrometazoans that occupy a diversity of niches in freshwater, marine, and terrestrial habitats. Commonly called water bears because of their slow, lumbering gait, these molting lobopods have four pairs of legs, usually terminating in claws. Most are less than 1 mm in length, with a complete digestive tract, a dorsal gonad with one or two gonoducts, and a dorsal lobed brain with a ventral nerve cord and four bilobed ganglia, one per leg-bearing metamere. The body cavity (hemocoel) functions in respiration and circulation. Over 1200 species have been described based primarily on the morphology of the claws and buccal-pharyngeal apparatus. Individuals may be either gonochoric, unisexual, or hermaphroditic, with fertilized or unfertilized eggs deposited either freely or within the shed exuvium. Parthenogenesis occurs frequently in limnic and terrestrial tardigrades, allowing them to colonize new territories by passive dispersal of a single individual. Cryptobiosis (anhydrobiosis, anoxybiosis, cryobiosis, and osmobiosis) and diapause (encystment and resting eggs) occur during the life history. Active adults (surrounded by water) and cryptobiotic adults and eggs are primarily dispersed passively, but some active dispersal can also occur. Due to the characteristic patchy distributions of tardigrade populations, little is known about their population dynamics and trophic relationships. Improved methods for collection, microscopy, culturing, and molecular analyses have been have contributed much to our knowledge of tardigrades.

Facteurs de risque professionnels des cancers des voies aérodigestives supérieures chez les femmes : analyse des données de l’étude Icare. / Occupational risk factors for head and neck cancer in French women : a population based case-control study in France, Icare

Contexte : Peu d’études ont recherché le rôle des facteurs de risque professionnels dans la survenue des cancers des voies aérodigestives supérieures (VADS). Ces études ont été conduites principalement chez des hommes.Objectif : L’objectif de cette thèse était d’étudier les associations entre les cancers des VADS et les expositions professionnelles chez les femmes.Méthodes : Icare est une étude cas-témoins en population générale incluant 296 cas féminins de cancers épidermoïdes des VADS et 775 femmes témoins. Les historiques de carrières recueillis ont été codés et croisés avec les matrices emplois-expositions du programme Matgéné. Outre les intitulés d’emplois, les expositions à 5 solvants chlorés (chloroforme, chlorure de méthylène, perchloréthylène, trichloréthylène, tétrachlorure de carbone), 5 solvants oxygénés (éthylène glycol, tétrahydrofurane, éther éthylique, cétones, alcools), 5 solvants pétroliers (essences carburants, essences spéciales, gazole, benzène, white-spirit) et à 7 poussières et fibres (amiante, farine, cuir, fibres céramiques réfractaires, ciment, laines minérales, silice) ont été étudiées. Les odds-ratios et leurs intervalles de confiance à 95% ajustés sur l’âge, le département, les consommations de tabac et d’alcool ont été estimés par régressions logistiques non conditionnelles.Résultats :Plusieurs professions et secteurs d’activité associés à un risque élevé de cancer des VADS ont été identifiés. Certaines professions (ouvrières de l’alimentation et des boissons, monteuses en appareillage électrique ou électronique, soudeuses) peuvent être à l’origine d’expositions professionnelles aux solvants, aux métaux, aux fumées de soudage et à diverses poussières. Les analyses par nuisance ont mis en évidence des associations significatives entre le risque de cancer des VADS et l’exposition au perchloréthylène et au au trichloréthylène. Aucune association claire n’est observée avec les solvants pétroliers et oxygénés, certains largement utilisés par les femmes L’exposition aux poussières de farine augmente significativement le risque de cancer des VADS. Une exposition probable à l’amiante est associée à une augmentation modérée et non significative du risque. Les analyses par localisation de cancer (cavité orale, pharynx, larynx), limitées par des effectifs faibles, ne mettent pas en évidence d’association spécifique.Conclusion : Nos résultats suggèrent un rôle des expositions professionnelles au trichloréthylène, au perchloréthylène et aux poussières de farine dans la survenue des cancers des VADS chez les femmes. / Background : Few occupational studies have addressed head and neck cancer, and these studies have been predominantly conducted in men. Objective : Our objective was to investigate the associations between head and neck cancer and occupational exposures in women Population and methods : ICARE, a French population-based case–control study, included 296 squamous cell carcinomas of the head and neck (HNSCC) in women and 775 female controls. Lifelong occupational history was collected. Job-exposure matrices were used to assess exposure to five chlorinated solvents (carbon tetrachloride; chloroform; methylene chloride; perchloroethylene; trichloroethylene), 5 petroleum solvents (benzene; special petroleum product; gasoline; white-spirits and other light aromatic mixtures; diesel, fuels and kerosene), 5 oxygenated solvents (alcohols; ketones and esters; ethylene glycol; diethyl ether; tetrahydrofuran) and 7 fibers and dusts (asbestos, flour dust, leather dust, refractory ceramic fibers, cement dust, mineral wools and silica) . An analysis by job title was conducted, and then associations with specific occupational exposures were investigated.Odds ratio (ORs) and 95% confidence intervals (CI), adjusted for smoking, alcohol drinking, age and residence area, were estimated with logistic models. Results : Significantly increased HNSCC risks were found for several jobs and industries. Some of these occupations (food and beverage processors, electrical and electronic equipment assemblers, welders and flame cutters) may entail exposure to agents such as solvents, metals, welding fumes and various dusts. Analyses for specific occupational exposures showed a significantly elevated risk of HNSCC associated with exposure to trichloroethylene and perchloroethylene. There is no clear evidence that petroleum or oxygenated solvents, some of them commonly used by women, are risk factors for HNSCC. Exposure to flour dust increased significantly HNSCC risk. Probable exposure to asbestos was associated with a moderate, non-significant elevation in risk. Analyses by cancer site (oral cavity, pharynx, larynx) were hampered by small numbers and did to reveal any specific association.Conclusion : These findings suggest that occupational exposure to perchloroethylene, trichloroethylene and flour dust may increase the risk of HNSCC in women.

Normal sväljning hos friska vuxna

Bakgrund Sväljning bedöms utifrån två aspekter - hur säker och hur effektiv den är. Tidigare studier har visat att vid stigande ålder blir sväljningen mindre effektiv och att förekomst av penetration ökar. Åldrande sväljning har i tidigare studier inte studerats specifikt utifrån säkerhet och effektivitet. Tidigare forskning med avseende på sväljning har enbart fokuserat på äldre individer respektive snävare åldersgrupper.  Syfte Aktuella studiens syfte är att undersöka om det ses en försämring i normal sväljförmåga vid stigande ålder. Studien syftar  även till att undersöka om det finns könsskillnader i hur sväljförmåga förändras vid ökande ålder, men även oavsett ålder. Bedömning av sväljfunktion, sväljningssäkerhet och sväljningseffektivitet genomfördes med Flexibel videoendoskopisk Undersökning av Sväljning (FUS).  Metod Totalt inkluderades 65 deltagare (33 st kvinnor och 32 st män), i åldrarna 21-91 år (median 44 år). FUS genomfördes för att undersöka sväljningen. Vid FUS sväljer deltagarna föda i olika konsistenser. Fynden från FUS bedömdes utifrån standardiserade skalor: The Dysphagia Outcome and Severity Scale (DOSS), Penetration-Aspiration Scale (PAS) och The Yale Pharyngeal Residue Severity Rating Scale (YPRSRS).  Resultat Majoriteten av alla deltagare bedömdes ha funktionell sväljning. Kvinnor uppvisade en bättre sväljfunktion överlag, men inga könsskillander kunde påvisas gällande säkerhet och effektivitet. Deltagare med en högre uppnådd ålder uppvisade en sämre sväljningssäkerhet och sväljningseffektivitet. Ålder kan således vara en påverkande faktor för sväljningssäkerhet och sväljningseffektivitet.   Slutsatser Denna studie visade att sväljfunktionen hos äldre deltagare var sämre än hos yngre deltagare. Sväljningens säkerhet och effektivitet var inte försämrad hos friska äldre deltagare, jämfört med friska yngre deltagare. Det fanns könsskillnader i sväljförmåga avseende sväljfunktion, kvinnor har en högre förekomst av normal sväljning än männen. Könsskillnader gällande sväljningssäkerhet kunde inte konstateras. Könsskillnad för sväljningseffektivitet kunde ses för sinus piriformis, men inte vallecula. Männen uppvisade genomgående en mindre effektiv sväljning, jämfört med kvinnorna.

Validação da medida da pressão crítica de fechamento da faringe durante o sono induzido / Validation of the pharyngeal critical closing pressure during induced sleep

Introdução: A pressão crítica de fechamento da faringe (PCrit) é a pressão nasal em que há colapso da faringe. Conceitualmente a Pcrit reflete a contribuição anatômica na gênese da apnéia obstrutiva do sono (AOS). No entanto, a relação entre a PCrit e a anatomia das vias aéreas superiores (VAS) tem sido pouco estudada. A PCrit determinada durante o sono requer pesquisadores experientes durante a noite. A indução do sono com midazolam é usada na prática clínica para a realização de procedimentos ambulatoriais e poderia ser uma alternativa mais conveniente para se determinar a Pcrit. Porém, o midazolam pode provocar sedação além de simples indução do sono, reduzir a atividade muscular das VAS e aumentar a colapsabilidade quando comparado com o sono normal. Objetivos: 1. validar a determinação da PCrit durante o dia após a indução do sono com midazolam; 2. comparar a arquitetura do sono induzido com baixa dose de midazolam com o sono natural; 3. correlacionar a PCrit com a anatomia das VAS. Métodos: Homens com graus variados de sintomas sugestivos de AOS foram submetidos a polissonografia completa noturna, determinação da PCrit durante o sono natural e após a indução do sono com midazolam bem como tomografia computadorizada de cabeça e pescoço para avaliação das VAS. Resultados: Foram estudados 15 sujeitos com idade (média±DP) de 54 ± 10 anos, índice de massa corporal de 29,9 ± 3,9 kg/m2 e índice de apnéiahipopnéia (IAH) de 38 ± 22 (variação: 8-66 eventos/h). A indução do sono foi obtida em todos os sujeitos, utilizando doses mínimas de midazolam (mediana [intervalo interquartil]) (2,4 [2,0-4,4] mg). A PCrit durante o sono natural e induzido foram semelhantes (-0,82 ± 3,44 e -0,97 ± 3,21 cmH2O, P = 0,663) e se associaram (coeficiente de correlação intraclasse=0,92 (IC 95% 0,78-0,97 P<0,001). A distribuição das fases do sono durante sono natural e induzido foi similar, com excessão da fase 1 (10,5 ± 5,1% vs. 20,6 ± 8,1, respectivamente; P=0,001). A Pcrit determinada durante o sono natural e induzido se correlacionaram com o IAH (r=0,592, P=0,020 e r=0,576, P=0,025, respectivamente). Além disso tanto a Pcrit determinada por sono natural e induzido se correlacionaram com diversas variáveis tomográficas de VAS, incluindo a posição do osso hióide, ângulo da base do crânio e as áreas seccionais da velofaringe e hipofaringe (r variando de 0,577 a 0,686, P<0,05). A regressão linear múltipla revelou que o IAH foi independentemente associado com a Pcrit durante sono induzido, circunferência da cintura e idade (r2 = 0,785, P = 0,001). Sleep induction was obtained with minimum doses of midazolam (median[interquartile range] (2.4 [2.0-4.4] mg). Sleep phase distribution during natural and induced sleep was similar, except for stage 1 (10.5 ± 5.1% vs. 20.6 ± 8.1, respectively; P=0.001). Natural and induced sleep Pcrit were similar (-0.82 ± 3.44 and -0.97 ± 3.21 cmH2O, P=0.663) and closely associated (intraclass correlation coefficient=0.92 (95%CI 0.78-0.97, P<0.001). Natural and sleep induced Pcrit correlated with AHI (r=0.592, P=0.020; r=0.576, P=0.025, respectively). Pcrit determined both during natural and induced sleep were significantly associated with several tomographic variables, including hyoid position, cranial base angle and cross sectional areas of the velopharynx and hypopharynx (r range: 0.577 to 0.686, P<0.05). An endoscopic and immunopathological study of respiratory tract disorders in thoroughbred racehorses

Much of the impetus for this research can be attributed to Kenneth W. Hinchcliff, who has studied exercise-induced pulmonary haemorrhage (EIPH) extensively. This thesis focused on EIPH in Thoroughbred racehorses competing in South Africa. Using tracheobronchoscopy, the prevalence and severity of EIPH and the association with racing performance was determined. Thereafter, the prevalence of other respiratory tract disorders and their association with racing performance is reported. This is followed by a study assessing interobserver variability using grading systems in the detection of respiratory tract disorders. Finally, there is a report on the immunopathogenesis of EIPH. Using tracheobronchoscopy after racing, the prevalence and severity of EIPH was assessed in 1,005 racehorses competing at high altitude (> 1,400 meters above sea level) and at sea level in a racing jurisdiction that does not allow the use of furosemide and nasal dilator strips. The prevalence and severity of EIPH was affected by altitude as racing at sea level was associated with a higher prevalence and greater severity of EIPH. Results also suggested that EIPH was associated with superior performance in South African Thoroughbred racehorses. Upper and lower respiratory tract disorders identified following tracheobronchoscopic examination included left arytenoid asymmetry, left laryngeal hemiplegia, epiglottic deformity, epiglottic entrapment, subepiglottic cysts, dorsal displacement of the soft palate, pharyngeal lymphoid hyperplasia (PLH), laryngeal and tracheal dirt, tracheal mucous (TM), tracheal stenosis and tracheal cartilage ring spikes in Thoroughbred racehorses after racing. Overall, there was a low prevalence of grade 2 and 3 arytenoid cartilage asymmetry, left laryngeal hemiplegia, epiglottic entrapment, subepiglottic cysts and epiglottic deformity, while more severe grades of PLH, laryngeal debris, tracheal debris, TM and tracheal cartilage ring spikes had a higher prevalence. An association with sex was identified as tracheal cartilage ring spikes occurred more often in male racehorses. Superior racing performance was identified in racehorses with grade 3 tracheal mucous and tracheal cartilage ring spikes. Endoscopic grading of EIPH, PLH, arytenoid cartilage movement (ACM), and TM was performed by 3 observers that were blinded to each racehorse’s identity and race day performance using previously established grading criteria. Excellent interobserver reliability was seen using the EIPH grading system, while the weighted kappa for PLH, ACM and TM was lower. The study demonstrated sufficient reliability for the use of the EIPH, PLH, ACM and TM grading systems in racehorses competing in South Africa. The study concluded that tracheobronchoscopy seemed to be a practical screening technique that may have prognosticative validity and clinical dependability and that would allow safe and quick assessment of the respiratory tract of a large number of racehorses in field conditions. Venous blood was collected from 10 horses in each EIPH grade classification (grade 0 to 4) following tracheobronchoscopic examinations for the determination of the presence and severity of EIPH. Following RNA isolation and cDNA synthesis, real-time PCR was used to detect equine cytokine-specific mRNA for interleukin (IL) -1, -6, -10, interferon (INF) -ã, and tumor necrosis factor (TNF) -á. Results of this study indicated that increased IL-6, and -10 mRNA production was associated with more severe forms of EIPH. Also, there was greater expression of IL-6 mRNA at sea level and TNF-á mRNA at high altitude. This study concluded that although it was unclear whether the inflammatory response observed in the study was due to pre-existing pulmonary inflammation or as a direct consequence of pulmonary bleeding, the study demonstrated a systemic correlation to pulmonary inflammation. The research reported in this thesis has contributed substantially to the determination of the prevalence, severity and affect on racing performance of respiratory tract disorders in Thoroughbred racehorses competing in South Africa. Also, determination of an association between EIPH and inflammation at a molecular level may assist future researchers in anti-cytokine therapies which may help reduce the prevalence and severity of EIPH. / Thesis (PHD)--University of Pretoria, 2007. / Companion Animal Clinical Studies / unrestricted
