Studies in this area allow you to develop pigments with more advanced properties and qualities to be used in the industrial context. Studies on synthesis and characterization of cobalt aluminate has been widely researched, cobalt aluminate behavior at different temperatures of calcinations, highlighting especially the temperatures of 700, 800 and 900? C that served as a basis in the development of this study, using the method of polymerization of complex (CPM), economic, and this method applied in ceramic pigment synthesis. The procedure was developed from a fractional factorial design 2 (5-2) in order to optimize the process of realization of the cobalt aluminate (CoAl2O4), having as response surfaces the batch analysis data of Uv-vis spectroscopy conducted from the statistic software 7.0, for this were chosen five factors as input variables: citric acid (stoichiometric manner), puff or pyrolysis time (h), temperature (? C), and calcinations (? C/min), at levels determined for this study. By applying statistics in the process of obtaining the CoAl2O4 is possible the study of these factors and which may have greater influence in getting the synthesis. The pigments characterized TG/DSC analyses, and x-ray diffraction (XRD) and scanning electron microscope (SEM/EDS) in order to establish the structural and morphological aspects of pigment CoAl2O4, among the factors studied it were found to statically with increasing calcinations temperature 700?< 800 <900 ?C, the bands of Uv-vis decrease with increasing intensity of absorbance and that with increasing time of puff or pyrolysis (h) there is an increase in bands of Uv-vis proportionally, the generated model set for the conditions proposed in this study because the coefficient of determination can explain about 99.9% of the variance (R?), response surfaces generated were satisfactory, so it s possible applicability in the ceramics industry of pigments / Na ind?stria cer?mica utilizam-se cada vez mais pigmentos de natureza predominantemente inorg?nica. Os estudos nessa ?rea permitem desenvolver pigmentos com qualidades e propriedades mais avan?adas para serem empregados em ?mbito industrial. Estudos de s?ntese e caracteriza??o do aluminato de cobalto t?m sido amplamente pesquisados, o comportamento do aluminato de cobalto em diferentes temperaturas de calcina??es, destacando principalmente as temperaturas de 700, 800 e 900?C utilizando o m?todo de polimeriza??o de complexos (MPC), m?todo este, econ?mico e aplicado em s?ntese de pigmentos cer?micos. Atualmente, a maioria desses grãos vem sendo descartada, o que gera a necessidade de aproveitamento desses resíduos, inclusive com estudos no sentido de incorporá-los à alimentação humana. Ante o exposto, objetivouse secar grãos residuais de urucum e farinhas de grãos residuais de urucum por meio convencional (secagem em estufa) e utilizando energia solar. Os grãos e as farinhas foram secados em estufa com circulação forçada de ar, nas temperaturas de 40, 50, 60 e 70 °C. Nas secagens por meio de energia solar foram utilizados dois secadores, um para secagem diurna, denominado secador solar, e o outro para secagem noturna, denominado Secador Acumulador de Calor para Secagem Noturna (ACSN). O secador ACSN é composto por dois circuitos de circulação de água independentes. No período diurno funcionava o circuito coletor solar/reservatório, o qual era interrompido durante a noite e aberto o circuito reservatório/câmara de secagem. Na secagem por exposição direta ao sol as amostras eram colocadas em bandejas em base de concreto recoberta com lona plástica (polietileno) preta. Para a representação da cinética de secagem dos grãos e das farinhas de urucum desidratados em estufa, secadores solar e noturno e por exposição direta ao sol foram empregados os modelos matemáticos de Aproximação da Difusão, Dois Termos, Midilli, Page e Thompson. Nas secagens convencionais, o aumento da temperatura de desidratação promoveu reduções nos tempos de secagem. A utilização do secador ACSN no período notumo evitou a absorção de água pelas amostras e promoveu sua secagem, o que possibilitou a obtenção de produto com menor teor de água. Para as amostras cuja secagem se iniciou no período notumo, a utilização do secador ACSN no período notumo possibilitou maiores taxas de secagem, em comparação com a testemunha, com obtenção de produto com menor teor de água. As secagens solares apresentaram tempos de desidratação menores que as observadas nas secagens convencionais. Os modelos de Aproximação da Difusão, Dois Termos, Midilli, Page e Thompson propiciaram bons ajustes aos dados experimentais da cinética de secagem das amostras de grãos e farinhas de grãos residuais de urucum com e sem óleo pelos métodos convencional e solar. / Residual annatto seeds are a subproduct derived from the extraction of bixin and norbixin pigments in the food industry. The fact that most of these seeds are typically discarded demonstrates the need for making better use of these residues, through studies aimed at incorporating them into the human diet. The objective of this study was to dry residual annatto seeds and annatto seed powder using conventional means (oven-drying) and solar energy. The seeds and powder were oven dried with forced air circulation at temperatures of 40, 50, 60 and 70 °C. Two dryers were used in the solar energy method, one for daytime drying, called a solar dryer, and the other for nighttime drying, denominated a heat accumulator for nighttime drying (ACSN). The ACSN dryer is composed of two independent water circulation circuits. The solar collector circuit/reservoir was in operation in the daytime. This was interrupted during the night, whereupon the reservoir circuit/drying chamber was opened. In solar drying, samples were collected in concrete trays covered with a black plastic tarp (polyethylene). Diffusion approximation, two term, Midilli, Page and Thompson mathematical models were used to represent the drying kinetics of annatto seeds and powder dried in an oven and solar and nighttime dryers, as well as by direct exposure to the sun. In conventional drying, an increase in temperature promoted reductions in drying time. Using the ACSN dryer at night avoided water absorption by the samples, promoting their drying and resulting in a product with lower water content. For samples dried at night, employing the ACSN dryer led to greater drying rates compared to the control sample, generating a product with lower water content. Solar dryers exhibited shorter drying times than those observed in conventional dryers. Diffusion Approximation, Two Term, Midilli, Page and Thompson models showed a good fit to experimental data derived from the drying kinetics of annatto seed samples and residual powder, with and without oil, using conventional and solar methods.

Conselho Nacional de Desenvolvimento Científico e Tecnológico / Phosphorus (P) is one of the macronutrient that most limits crop production, especially of that potato, a crop that is highly responsive to soil fertility. The deficiency of this nutrient occurs in more than half of the world's arable land. This work aimed to study the effect of two contrasting levels of P in potato genotypes through physiological (growth, chlorophyll a fluorescence, photosynthetic pigments) and biochemical parameters (acid phosphatase activity, phosphorus content in tissues), as well as determinate the efficiency and response index to P. The experiment was conducted hydroponically in a greenhouse, using sand as substrate. The treatments consisted of four potato genotypes SMIC148-A (C), SMIF212-2 (M), SMIG145-1 (O) and SMIJ319-7 (S) and two levels of phosphorus (P), 5% P and 50% (2,32 e 23,23 mg L-1) P of the standard solution. Evaluations were performed at 18, 39 and 62 days after transplanting (DAT). The low level of P was responsible for reductions in tuber yield and production of shoot and root, as decreases in the number of leaves and fresh and dry weight of these parts of the plant were found. The activity of the acid phosphatase enzyme was higher on 5% of P in root and lower in the fourth leaf at initial period of cultivation. The concentration of soluble phosphorus (Pi) and total P content in the tissue, were generally lower at the lower phosphorus level that at the highest level. The concentration of the photosynthetic pigments has been generally kept similar between levels. The Fv/Fm and Fv/Fo ratios as well as a transport rate of electrons (ETR1500) for some genotypes had reductions on 5% P. On the basis of P use and response efficiency indexes, genotypes SMIC148-A (C) and SMIF212-2 (M) were classified into NENR and genotypes SMIJ319-7(S) and SMIG145-1 (O) ER. A deficiência deste nutriente ocorre em mais da metade dos solos agricultáveis do mundo. Este estudo teve como objetivo estudar o efeito de níveis contrastantes de P em genótipos de batata através de parâmetros fisiológicos (crescimento, fluorescência da clorofila a, pigmentos fotossintéticos) e bioquímicos (atividade da fosfatase ácida, teor de fósforo nos tecidos), bem como a determinação de índices de eficiência e resposta ao P. O experimento foi conduzido em casa de vegetação, em sistema hidropônico em areia. Os tratamentos utilizados constaram de quatro genótipos de batata, SMIC148-A (C), SMIF212-2 (M), SMIG145-1 (O) e SMIJ319-7 (S) e dois níveis de fósforo (P), 5% de P e 50% (2,32 e 23,23 mg L-1, respectivamente) de P da solução padrão. As avaliações foram realizadas aos 18, 39 e 62 dias após o transplantio (DAT). O baixo nível de P foi responsável por reduções na produção de tubérculos bem como na produção de parte aérea e raiz, pois se verificou decréscimos no número de folhas e na massa fresca e seca destas partes da planta. A atividade da enzima fosfatase ácida foi maior em 5% de P em raiz e menor na quarta folha em período inicial de cultivo. A concentração de fósforo solúvel (Pi) bem como o conteúdo de P total no tecido, foram em geral menores no menor nível de P que no maior nível. A concentração de pigmentos fotossintéticos de maneira geral foi mantida em baixo P. As razões Fv/Fm e Fv/Fo, bem como a taxa de transporte de elétrons (ETR1500) para alguns genótipos tiveram reduções em 5% de P. A partir de índices de eficiência de uso e resposta ao P o genótipos SMIC148-A (C) e SMIF212-2 (M) foram classificados em NENR e os genótipos SMIJ319-7(S) e SMIG145-1 (O) em ER.

No. of bitstreams: 0 / A irradiação de alimentos é um meio de preservação de alimentos onde se utiliza uma técnica de processamento que expõe os alimentos a uma radiação ionizante controlada de alta energia. O tratamento realizado com o uso das radiações ionizantes em alimentos tem muitas aplicações tecnologicamente e tecnicamente viáveis incluindo a capacidade de melhorar a segurança microbiológica, reduzir os níveis de bactérias patogénicas, inibir a germinação de tubérculos, aplicação fitossanitária, conservar alimentos armazenados ou a estabilidade de armazenamento e também é utilizada para aumentar a vida de prateleira de determinados produtos devido à redução da contaminação por microrganismos. Em função do aumento do comércio internacional de alimentos e das crescentes exigências regulatórias dos mercados consumidores, cada vez mais países importadores e exportadores têm demonstrando interesse na irradiação de alimentos e desenvolvido pesquisas na aplicação prática desta tecnologia e de métodos de detecção do tratamento. Inúmeras pesquisas foram realizadas mundialmente, resultando em eficientes protocolos para identificar quais alimentos foram ou não irradiados. Até então, o \"mito\" que alimentos irradiados não poderiam ser detectados e que não eram formados quaisquer produtos únicos da radiação foi substituído pelo conhecimento que muitas mudanças podem ocorrer nos alimentos irradiados e essas mudanças poderiam ser utilizadas como ferramentas para identificar essa tecnologia. O processamento por radiação resulta em formações de padrões característicos de hidrocarbonetos saturados, aldeídos, metil e etil ésteres e 2-alcilciclobutanonas, dependendo da composição do ácido graxo do lipídio que compõe o alimento. Essa espécie é endêmica dos afloramentos rochosos do município do Rio de Janeiro, RJ, e vem sofrendo com o pisoteio de alpinistas, queimadas, invasão de gramíneas exóticas e extrativismo vegetal. A micropropagação pode ser utilizada como alternativa às condições de risco em que as populações dessa espécie se encontram submetidas, visando à recomposição de populações ameaçadas em ambiente natural, assim como o abastecimento do mercado consumidor. A etapa final da micropropagação é a aclimatização, período em que as plantas ficam mais susceptíveis e sofrem com o estresse oxidativo devido às mudanças nas condições ambientais. No presente trabalho foram avaliadas as atividades enzimáticas antioxidativas da CAT, SOD, POD, PPO e o conteúdo de prolina, além dos teores de pigmentos fotossintéticos em plantas de Pitcairnia albiflos cultivados in vitro, em meios de cultura contendo duas concentrações de sacarose (15 ou 30 g L-1), tampas não vedadas que permitiam trocas gasosas e frascos vedados com tampas e filme plástico de PVC, que impediam a ventilação. Sob essas condições, as plantas foram cultivadas em meios contendo GA3 ou ANA. Após o período de crescimento in vitro, as plantas foram transferidas para condições ex vitro em casa de vegetação. As análises supra-citadas e a determinação dos teores de carboidratos solúveis totais, sacarose, amido, açúcares redutores, conteúdo relativo de água e suculência também foram realizadas nas plantas previamente cultivadas in vitro com 15 ou 30 g L-1 de sacarose e GA3 em tubos com tampas vedadas, após submissão das mesmas a estresse hídrico durante 24, 38 ou 52 dias. Após o período de estresse hídrico, as plantas foram reidratadas durante 34 dias sob irrigação periódica em casa de vegetação. Nos tecidos cultivados in vitro percebeu-se o surgimento de características de hiperidricidade nas plantas cultivadas com 15 g L-1 de sacarose, GA3 e tubos com tampas vedadas, o que foi evidenciado pelo menor acúmulo de prolina, aumento das atividades das enzimas antioxidantes e menor acúmulo de pigmentos fotossintetizantes. Na condição ex vitro, as plantas cultivadas anteriormente em meio de cultura contendo 15 g L-1 de sacarose apresentaram maior atividade das enzimas antioxidantes não havendo, em alguns casos, diferenças significativas em comparação com a concentração mais elevada de sacarose. Nessa condição o acúmulo de prolina foi menor, o que é indicativo de maior estresse oxidativo nessas plantas durante a aclimatização. Durante o estresse hídrico houve queda na atividade de todas as enzimas estudadas, embora essa queda tenha sido mais acentuada para as plantas que inicialmente foram cultivadas com 15 g L-1 de vi sacarose. O acúmulo de prolina aumentou com o prolongamento do estresse hídrico, sendo maior nas plantas que foram cultivadas in vitro com 30 g L-1 de sacarose. Não houve diferenças significativas no conteúdo de pigmentos fotossintetizantes e nas suas relações para nenhuma das concentrações de sacarose, exceto para os carotenóides totais, que apresentaram aumento significativo ao longo do período de estresse hídrico para as plantas previamente cultivadas com a menor concentração de sacarose. Os conteúdos de carboidratos solúveis totais e sacarose aumentaram com o prolongamento do estresse, sendo mais acentuados na concentração de 30 g L-1 de sacarose. Os conteúdos de amido, o conteúdo relativo de água e a suculência apresentaram redução com o aumento do estresse hídrico. Após a reidratação, todas as plantas mostraram capacidade de recuperação, apresentando valores próximos aos dos controles para todas as variáveis analisadas. Ressalta-se, todavia, que as plantas tratadas com 30 g L-1 de sacarose tiveram melhor recuperação quando comparadas com aquelas que foram tratadas com 15 g L-1 de sacarose. Em função dos resultados obtidos, é possível concluir que a concentração de sacarose utilizada in vitro apresenta influência no processo de aclimatização ex vitro e também, posteriormente, no campo, na capacidade de recuperação das plantas à seca quando elas são submetidas a estresse hídrico. As plantas cultivadas in vitro com 15 g L-1 de sacarose se mostraram mais sensíveis à seca e, possivelmente, não sobreviveriam caso fossem transferidas dos tubos de ensaio diretamente para o campo. As plantas cultivadas in vitro com 30 g L-1 de sacarose aparentemente eram mais resistentes ao processo de aclimatização ex vitro, apresentando maiores chances de sobrevivência em campo, maior tolerância à seca e maior capacidade de recuperação após períodos prolongados de estresse hídrico. / Pitcairnia albiflos Herb. (Bromeliaceae) is currently in the list of endangered species. This species is endemic of the inselbergs of the city of Rio de Janeiro, RJ, and has been suffering with the mountaineer’s trampling, wildfires, invasion of exotic grasses and plant extraction. The micropropagation can be used as an alternative to the risk conditions under which populations of this species are submitted, aiming at the recomposition of endangered populations in the natural environment, as well as the supply of the consumer market. The final stage of the micropropagation is the acclimatization, period in which the plants become more susceptible and suffer from oxidative stress due to the changes in the environmental conditions. In the present study it were evaluated the antioxidative enzymatic activities of CAT, SOD, POD, PPO and proline content, besides the levels of photosynthetic pigments in plants of Pitcairnia albiflos grown in vitro in culture mediums containing two sucrose concentrations (15 or 30 g L-1). Part was covered with unsealed lids that allowed gas exchanges and part was kept in sealed flasks with lids and PVC plastic film, that didn’t allow the ventilation. Under these conditions, the plants were cultivated in culture mediums containing GA3 or NAA. After the in vitro growth period, the plants were transferred to ex vitro conditions at a greenhouse. The above mentioned analyses and the determination of total soluble carbohydrate levels, sucrose, starch, reducer sugars, relative water content and succulence were also performed on the plants previously grown in vitro with 15 or 30 g L-1 of sucrose and GA3 in tubes with sealed lids, after the submission of these to water stress during 24, 38 or 52 days. After the water stress period, the plants were rehydrated for 34 days under regular irrigation at the greenhouse. In the in vitro cultivated tissues it was noted the emergence of hyperhydricity characteristics in the plants grown with 15 g L-1 of sucrose, GA3 and tubes with sealed lids, which was evidenced by the lowest proline accumulation, the increased in the antioxidative enzymatic activities and the lowest accumulation of photosynthetic pigments. In the ex vitro condition, the plants previously grown in culture mediums containing 15 g L-1 of sucrose presented larger antioxidative enzymatic activities, which did not show, in some cases, significant differences compared with the largest concentration of sucrose. In this condition, the proline accumulation was lower, which is an indicative of larger oxidative stress in these plants during acclimatization. During the water stress, there was a fall in the activity of all studied enzymes, although that fall had been more evident in the plants that were initially cultivated with 15 g L-1 of sucrose. The proline accumulation increased with the extension of the water stress, being larger in the plants grown in vitro with 30 g L-1 of sucrose. There were no viii significant differences in the content of photosynthetic pigments and in their relation with any sucrose concentrations, except for total carotenoids, which significantly increased over the period of water stress for the plants previously grown with the lowest concentration of sucrose. The contents of total soluble carbohydrates and sucrose increased with the extension of the stress, being more accentuated in the 30 g L-1 of sucrose concentration. The contents of starch, the relative content of water and succulence presented a reduction with the increase of water stress. After the rehydration, all plants showed recovery capacity, presenting values close to those from the control groups for all the analyzed variables. It should be noted, however, that the plants treated with 30 g L-1 of sucrose had better recovery compared with those that were treated with 15 g L-1 of sucrose. According to the obtained results, it is possible to conclude that the concentration of sucrose used in vitro presents influence on the process of ex vitro acclimatization and also, later in the field, in the recovery capacity of the plants to drought when they are submitted to water stress. The plants grown in vitro with 15 g L-1 of sucrose were more sensitive to drought and, possibly, would not survive if they were transferred from the test tubes directly to the field. The plants grown in vitro with 30 g L-1 of sucrose were apparently more resistant to the ex vitro acclimatization process, presenting greater survival chances in the field, larger drought tolerance and larger recovery capacity after extended periods of water stress.

Extrakce anthokyanových barviv z bezinkových výlisků / Extraction of anthocyanin pigments from elderberries pomace

This Diploma thesis deals with the production of the concentrate from the Elderberry marc extract (Sambucus nigra L.). The basic chemical characteristics in this concentrate were determined. The theoretical part of the thesis deal with the botanic characteristic of the Elderberry and its berries. The characteristic of the phenolic compounds, mainly anhocyanins was summarized. The use of the anthocyanin pigments in the food industry, the possibilities of their extraction and the concentrate production was discussed. Thereafter the methods of the anthocyanin content and the phenolic content determination were described. In the experimental part of study, the production of the anthocyanin rich extract from Elderberry marc was optimized. The extraction of the marc by the 1:1 ethanol/water (V/V) solvent with the ratio marc/solvent 1:2 (w/V) showed the most convenient. The production of the concentrate followed. During the concentration on the rotary evaporator the pigment degradation was investigated. The degradation decreased with the decrease of the temperature. The final concentrate was prepared from the extract on the rotary evaporator by 45 °C for 15 minutes. This product had the 7times lower volume compared to the initial extract. In the final concentrate come of the chemical characteristic were determined. By liquid chromatography (HPLC) the major pigments cyanidin-3-sambubiosid and cyanidin-3-glucosid were determined. Their total content was determined to 34,9 gl-1. The total anthocyanin content was determined by pH differential method to 7,1 gl-1 (expressed as cyanidin-3-glucoside equivalent). The total phenolic content was determined to 27,1 gl-1 (expressed as gallic acid equivalent) by Folin-Ciocalteu method. By electron paramagnetic resonance the increase in antioxidant activity with the increase in concentration was found. Up to 42 aromatic compounds was determined in Elderberry marc, its extract and the final concentrate.

This diploma thesis is focused on the study of the influence of the of size cosmetics pigments on the properties of the final product. This effect was studied on samples of make-up given by industry partner. Make-up samples were measured by the rheology method and the yield stress of all samples was determined. Scanning electron microscopy was used to determine the size and shape of the pigment particles. Particle size was also measured by dynamic light scattering, but the results were influenced by forming aggregates. To easier determination of the types of pigments in the sample was determined elemental composition of the pigments (Ti, Fe, O, Al) by EDS detector. Due to the unknown composition of make-up, the information was supplemented by X-ray powder diffraction (XRD) analysis, and it was possible to determine the compounds TiO2 (anatase and rutile), FeOOH and Fe2O3. The results obtained by these methods showed what pigments can be found in make-up samples, what their sizes and shapes are. It has been shown that the smaller the pigments are, the higher the viscosity and yield stress of the final product. The higher these quantities, the harder it is to handle the product.

Organická elektronika a bioelektronika zažívá v moderním věku obrovskou snahu o vývoj nových organických materiálů s vlastnostmi, které běžná elektronika na bázi křemíku obvykle nedosahuje. Tato práce se zabývá syntézou a charakterizací nových organických materiálů pro organickou elektroniku. Zaobírá se přípravou nových pentafluorsulfanylových heterocyklických stavebních bloků, alkylovaných vysokoúčinných organických pigmentů a bioinspirovaných organických -konjugovaných materiálů s modifikovatelnými fyzikálně-chemickými vlastnostmi a racionálními syntetickými přístupy k jejich přípravě. Pentafluorsulfanylová skupina (SF5) je ceněna pro svou vysokou elektronegativitu, lipofilitu, tepelnou a chemickou stabilitu. Pozitivně ovlivňuje optické a elektronické vlastnosti, rozpustnost a stabilitu v často lepší míře než u zavedených CF3-analogů. Šesti a čtyřkrokovou syntézou byly připraveny dva typy derivátů 3-SF5-substituovaných pyrrolidinů, které mohou sloužit jako všestranné stavební jednotky pro začlenění do pokročilých aromatických a heteroaromatických -konjugovaných systémů prostřednictvím atomů dusíku jako fluorované terminální skupiny. Modelový derivát byl zaveden jako terminální skupina do elektronově chudého heteroaromatického systému nukleofilní substitucí. Vodíkovými můstky vázané vysokovýkonné organické pigmenty přitahují velkou pozornost díky svým působivým polovodivým vlastnostem, silné 2D molekulární asociaci, vysoké tepelné, chemické a fotochemické stabilitě a nízké toxicitě. Přesto jsou pouze omezeně rozpustné a zpracovatelné, což lze vyřešit zavedením solubilizačních skupin. Je známo, že obzvláště objemné rigidní skupiny nesoucí adamantyl zlepšují uspořádání molekulární struktury, tepelnou stabilitu a výsledné vlastnosti díky samoorganizační schopnosti adamantanu. Adamantylmethylová a adamantylethylová skupina byla zavedena do vybraných barviv a pigmentů ze skupiny karbonylových azaacenů, rylen-diimidů a indigoidů pomocí nukleofilní substituce se zaměřením na zvýšení rozpustnosti a zpracovatelnosti při zachování tepelné stability a efektivní molekulární organizaci v pevné fázi. Fyzikálně-chemické studie série derivátů quinacridonu a epindolidionu ukázaly srovnatelnou, nebo vyšší tepelnou stabilitu než u nesubstituovaných derivátů, dobrou rozpustnost v organických rozpouštědlech, silnou fluorescenci v pevné fázi a roztoku v oblasti VIS a unikátní krystalovou strukturu pozorovanou z RTG analýzy. Flaviny jsou všudypřítomné bioinspirované organické materiály s nepostradatelnými biologickými funkcemi, výhodnými fyzikálně-chemickými vlastnostmi, chemickou a aplikační všestranností. Cílem práce byla modifikace optických, elektronických, elektrochemických, tepelných a dalších vlastností rozšířením -konjugovaného systému syntetizovaných materiálů. V první fázi byly navrženy dva syntetické přístupy pro přípravu série NH-nesubstituovaných flavinů, u kterých byly provedeny komplexní fyzikálně-chemické studie. Poté byly navrženy dva přístupy k syntéze N,N'-dialkylovaných flavinů s inkorporovaným butylovým, adamantylethylovým a triethylenglykolovým substituentem. Alkylací byla zvýšena rozpustnost v organických rozpouštědlech a vodném prostředí, zesílena fluorescence v pevné fázi a v roztocích a modifikovány tepelné vlastnosti v závislosti na zavedeném alkylovém substituentu.

The purpose of this thesis is to compare the contents anthocyans in different varieties of European elderberry (Sambucus nigra L.). It was also studied the relationship between the content anthocyans in selected samples of blue grape varieties (Vitis vinifera L.) and position of the track, where the grapes were grown. The theoretical part describes elderberries and grapes in terms of botanical, chemical, and in terms of their using in the food industry. The dominant part of this chapter is to describe anthocyans and possibilities for their determination in the elderberry and grapes. Emphasis has been placed on the pH-differential method and determination by high performance liquid chromatography. The experiments are divided into two parts. The aim of the first experimental part is comparison of the total anthocyanins content in fifteen varieties of elderberry. The performed analysis shows that in terms of anthocyanins content for cultivation of elderberry in the Czech Republic the most suitable varieties are Mammut, Samyl, Reis aus Vossloch and Haschberg. In the second part of the experiments were determined total anthocyanins content in nine varieties of blue grape grown in the Mikulov region. For determination were used HPLC and pH-differential methods, as well as in the first part. The highest content of pigments was found in varieties Alibernet and Neronet. The present study focused on the suitability of the location pointed to vineyard south to southeast orientation plantings. At the end of the experimental part is a comparison of both methods of determination. Method using high performance liquid chromatography to provide very accurate results compared to less demanding pH-differential method of determination. This is recommended only for tentative determination of anthocyanins content in real samples.

Removal of cationic and anionic dyes from aqueous solution using a clay-based nanocomposite.

PhDENV / Department of Ecology and Resource Management / Some industries such as textiles, ceramics, paper and printing are known to use significant amounts of dye to colour their products and during the colouring process, certain quantities of dyes are absorbed by the products, and some of them end up in wastewater. Depending on their application, some synthetic dyes are designed to be chemically or biologically resistant and their presence in the environment can cause severe environmental problems because of their colour impartation to water bodies. Therefore, proper treatment is required to remove these pollutants from wastewater before discharge into the environment. In this thesis, the potential of dye removal from wastewater by calcined magnesite, halloysite nanoclay and calcined magnesite - halloysite nanoclay composite was evaluated. To this end, the study was subdivided to four segments. The first segment of the study focused on evaluating the efficiency of using calcined magnesite to remove Methylene Blue (MB), Direct Red 81 (DR81), Methyl Orange (MO) and Crystal Violet (CV) dyes from aqueous systems using a batch study. To achieve that, several operational factors like residence time, adsorbent dosage, dye concentration and temperature were appraised. The adsorbent was subjected to different kinds of physicochemical characterization to determine the various characteristics that would assist in the dye uptake process. Characterization results showed that the adsorbent material was highly crystalline with magnesite, periclase, dolomite, and quartz as some of the crystalline phases. The batch study proved that calcined magnesite is effective in the treatment of dye contaminated water and moreover it performed well in terms of colour removal, though exceptional results were recorded for CV removal with complete decolourisation occurring in first few minutes of contact. In terms of experimental adsorption capacity, the performance of calcined magnesite was in the order CV (14.99 mg/g) > DR81 (12.56 mg/g) > MO (0.64 mg/g) > MB (0.39 mg/g). Mechanisms of adsorption where explained by fitting the experimental data into adsorption isotherms, kinetics, and thermodynamic parameters. Neither, the Langmuir, nor the Freundlich nor the Dubinin Radushkevich, nor the Temkin model could perfectly describe the adsorption of the four dyes onto calcined magnesite, however adsorption kinetics obeyed the pseudo second order model, implying that, the dye removal process was primarily a chemical process. In accordance with the results of this study, it can be concluded that calcined magnesite can be used effectively for the removal of dyes in aqueous solution and thus can be applied to treat wastewater containing dyes. The second segment of the study focused on the removal of MB, DR81, MO and CV dyes by halloysite nanoclay. Physicochemical characterisation revealed that the nanoclay has a surface area of 42 m²/g and its ABSTRACT iv morphology is predominated by tubular structures, which exhibit some hollow rod like structures. Various important parameters namely contact time, initial concentration of dyes, dosage, solution temperature and solution pH were optimized to achieve maximum adsorption capacity and it was observed that the effect of initial pH and temperature of the aqueous solution was neglibible on removal of the four dyes. The experimental adsorption capacities towards 40 mg/L of MB, DR81, MO and CV dyes were 17.51, 14.11, 0.38, and 4.75 mg/g respectively. The results indicate that natural halloysite nanoclay is an efficient material for the removal of the selected dyes. Due to its low cost and non-toxicity, halloysite nanoclay can be considered a good replacement option of other high cost materials used to treat coloured wastewater especially in developing countries like South Africa. Having observed the performance of calcined magnesite and halloysite nanoclay individually in the removal of selected dyes, a third study was designed with the aim of preparing a nanocomposite adsorbent from the aforementioned adsorbent materials and then evaluating the synergistic influence of the mechanochemical modification by a ball miller on the removal of MB, DR81, MO and CV dyes. Physicochemical characterization was carried out to get an insight of pre- and -post adsorption characteristics of the nanocomposite material and results showed major changes which could be an indication of dye uptake by the nanocomposite material. According to the results, the nanocomposite material outcompeted its component individual constituent materials i.e (calcined magnesite and halloysite nanoclay material) in the removal of DR81, which in turn was the highest removal efficiency observed for the whole batch adsorption study recording a maximum adsorption capacity and percentage removal of 19.89 mg/g and 99.40% respectively. Experimental results fitted the Langmuir and pseudo-second order models perfectly hence demonstrating that adsorption took place on a homogenous adsorbent layer via chemisorption. In overall, the results suggested that the nanocomposite is a suitable adsorbent for decolourising industrial wastewater. Based on the overall performance of the adsorbents in removing the four dyes, it was observed that the nanocomposite material had a high affinity for DR81 dye hence it was chosen as the model dye for further application in column studies. The effect of flow rate, bed height and initial dye concentration on the removal of DR81 was investigated. Maximum bed capacity and equilibrium dye uptake were determined and break through curves were plotted. Percentage dye removal increased with decrease in flow rate and increase in bed height. The maximum capacity of column was found to be about 51.73 mg DR81 per gram of the nanocomposite adsorbent for a flow rate of 3 mL/min, initial concentration of 10 mg/L and 4 cm bed height. Data from column studies was fitted to the Thomas model and Adams-Bohart models. The comparison of the R2 values obtained from both models showed a better fit for the nanocomposite material than the individual halloysite nanoclay and calcined magnesite materials. The study revealed the applicability of calcined magnesite- halloysite nanoclay composite in fixed bed column for the removal of DR81 dye from aqueous solution. v The reuse of an adsorbent is essential in order to make the adsorption process economic and environmentally friendly. To recover the adsorbate and renew the adsorbent for further use, a chemical method of regeneration was applied by using 0.1 M NaOH as the desorbent. Regeneration with 0.1 M NaOH proved very efficient for some dyes and less efficient for others depending on the adsorbent material used at the time. The general observation was that the adsorption capacity of the adsorbent materials decreased with successive adsorption – desorption cycles. Furthermore, regeneration with NaOH, favoured the acidic dyes (DR81 and MO) more than the basic dyes (MB and CV) possibly due to electrostatic interactions between oppositely charged molecules allowing for reversible reactions to take place. The three tested adsorbents namely calcined magnesite, halloysite nanoclay and their nanocomposite thereof were applied for the treatment of real wastewater effluent from a printing and ink industry. The adsorbents performed very well in terms of colour removal as recommended by the South African standards of wastewater discharge, However, in terms of pH, calcined magnesite and the nanocomposite produced a highly alkaline solution hence wastewater neutralisation by an acid is recommended before discharge. These findings show that the two natural clay-based materials (calcined magnesite and halloysite nanoclay) and their nanocomposite thereof have a great potential for application in dye wastewater remediation since the materials used in the process are inexpensive, abundant and require minimal modifications. / NRF
